CsCl as an auxiliary reagent for the analysis of phosphatidylcholine mixtures by matrix-assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI-TOF MS)

Matrix-assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI-TOF MS) is mainly used for protein and peptide analysis. However, there is growing evidence that also phospholipids like phosphatidylcholines (PC) can be easily analyzed by MALDI-TOF MS. In MALDI-TOF methodology, the sample is cationized by the addition of inorganic ions. This process is strongly dependent on the corresponding ion concentration. In biological samples various cations are present (mainly H +, Na + and K +) and, therefore, a mixture of different adducts is formed. Since phospholipids exhibit a wide distribution of different fatty acid residues a considerable peak overlap may occur. This is a major problem since the peak assignment in a mixture will be often unclear. In this paper we demonstrate that this problem can be easily overcome by mixing the analyte with caesium chloride (CsCl). This yields naturally non-occurring Cs + adducts that are apparent due to the large shift of the molecular mass. The proposed method facilitates the clear assignment of most peaks. Besides that, we will show that CsCl can also be used for the determination of the relative fatty acid composition of a given PC sample. For this purpose naturally occurring mixtures of PCs as well as organic extracts of human lipoproteins—that are mainly composed of PC and sphingomyeline—are used.

Lipid analysis of human HDL and LDL by MALDI-TOF mass spectrometry and 31P-NMR

The analysis of HDL and LDL is important for the further understanding of atherosclerosis because changes of the protein and lipid moieties occur under pathological conditions. Because destruction of lipids leads to the formation of well-defined products such as lysophospholipids or chlorohydrins, methods that allow their fast and reliable determination would be useful. In this study, matrix-assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI-TOF MS) was applied for the analysis of the lipid composition of human lipoproteins. These data were compared with high resolution 31P-NMR spectroscopy. Differences between LDL and HDL in sphingomyelin and phosphatidylcholine content could be monitored by NMR and mass spectrometry, and differences with respect to the extraction efficiency were found by MALDI-TOF MS. Additionally, treatment of LDL with hypochlorite and phospholipase A2 resulted in marked changes (formation of chlorohydrines and lysolipids). Lysophosphatidylcholines were detectable by both methods, whereas MALDI-TOF MS failed to detect chlorohydrines of phospholipids. We conclude that MALDI-TOF MS provides rapidly a reliable lipid profile of lipoproteins. However, a previous lipid separation must be performed to detect lipid oxidation products. Resonant infrared pulsed-laser deposition of polymer films using a free-electron laser

Thin films of polyethylene glycol (MW 1500) have been prepared by pulsed-laser deposition (PLD) using both a tunable infrared (λ=2.9 μm, 3.4 μm) and an ultraviolet laser (λ=193 nm). A comparison of the physicochemical properties of the films by means of Fourier transform infrared spectroscopy, electrospray ionization mass spectrometry, and matrix-assisted laser desorption and ionization shows that when the IR laser is tuned to a resonant absorption in the polymer, the IR PLD thin films are identical to the starting material, whereas the UV PLD show significant structural modification. These results are important for several biomedical applications of organic and polymeric thin films.

Decreased brain levels of 2',3'-cyclic nucleotide-3'-phosphodiesterase in Down syndrome and Alzheimer's disease.

In Down syndrome (DS) as well as in Alzheimer's disease (AD) oligodendroglial and myelin alterations have been reported. 2',3'-cyclic nucleotide-3'-phosphodiesterase (CNPase) and carbonic anhydrase II (CA II) are widely accepted as markers for oligodendroglia and myelin. However, only data on CNPase activity have been available in AD and DS brains so far. In our study we determined the protein levels of CNPase and CA II in DS, AD and in control post mortem brain samples in order to assess oligodendroglia and myelin alterations in both diseases. We used two dimensional electrophoresis to separate brain proteins that were subsequently identified by matrix assisted laser desorption and ionization mass-spectroscopy (MALDI-MS). Seven brain areas were investigated (frontal, temporal, occipital and parietal cortex, cerebellum, thalamus and caudate nucleus). In comparison to control brains we detected significantly decreased CNPase protein levels in frontal and temporal cortex of DS patients. The level of CA II protein in DS was unchanged in comparison to controls. In AD brains levels of CNPase were decreased in frontal cortex only. The level of CA II in all brain areas in AD group was comparable to controls. Changes of CNPase protein levels in DS and AD are in agreement with the previous finding of decreased CNPase activity in DS and AD brain. They probably reflect decreased oligodendroglial density and/or reduced myelination. These can be secondary to disturbances in axon/oligodendroglial communication due to neuronal loss present in both diseases. Alternatively, reduced CNPase levels in DS brains may be caused by impairment of glucose metabolism and/or alterations of thyroid functions.

Limits for the detection of (poly-)phosphoinositides by matrix-assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI-TOF MS)

Matrix-assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI-TOF MS) has been recently established as a powerful tool for the analysis of biomolecules. Here, MALDI-TOF MS was used for the detection of (poly-)phosphoinositides (PPI). PPI possess higher molecular weights than other phospholipids and a high phosphorylation-dependent negative charge. Both features affect the MALDI detection limits expressed as the minimum of analyte on the sample plate resulting in a signal-to-noise-ratio of S/ N=5. Using 2,5-dihydroxybenzoic acid (DHB) as matrix the detection limit for phosphatidylinositol (PI) is seven times higher than for phosphatidylcholine (PC) and further increases with increasing phosphorylation or in mixtures with other well-detectable phospholipids. For phosphatidylinositol-tris-phosphate (PIP 3) in a mixture with PC, the limit is about 20 times higher than for PI. The consequences for the experimental conditions are discussed. It is advisable to pre-separate PPI from biological lipid mixtures prior to the application of MALDI-TOF MS.

Recent progress in matrix-assisted laser desorption ionization postsource decay

In matrix-assisted laser desorption and ionization mass spectrometry, using a time-of-flight analyzer, decomposition of analytes by postsource decay can be examined by making use of an electrostatic ion reflector, thus recording tandem mass spectra. In recent years reasonable progress has been made using different instrument configurations and different types of reflectors. Also, the resolution of the first mass spectrum, i.e. the resolution of the precursor ion selector and the speed and possibility of automation of the acquisition and interpretation of the data has been improved. This article reviews the work of different groups working in this field and discusses the advantages of various arrangements.

Lipid analysis of human spermatozoa and seminal plasma by MALDI-TOF mass spectrometry and NMR spectroscopy — effects of freezing and thawing

In the present study, the applicability of proton NMR spectroscopy and matrix-assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI-TOF MS) to the analysis of the lipid composition of human spermatozoa and seminal fluids as well as changes after cryopreservation of human spermatozoa was investigated. Whereas NMR spectra primarily indicated a high content of double bonds within the spermatozoa but no marked differences upon cryopreservation, MS detected intense peaks which could be assigned to phosphatidylcholines containing one docosahexaenoic and one palmitic or stearic acid residue ( m/ z=806 and 834). In contrast, the seminal plasma contained more saturated fatty acids and especially more sphingomyelin (SM). A freezing/thawing cycle markedly influences the lipid composition of spermatozoa. There was a diminution of phosphatidylcholines (16:0, 22:6 and 18:0, 22:6) and SM (16:0) and the appearance of lysophosphatidylcholines (16:0 and 18:0) and ceramide (16:0). These data demonstrate the release or activation of both phospholipase A 2 and sphingomyelinase in human spermatozoa due to the freezing/thawing cycle. These results were finally confirmed by experiments on the action of phospholipases on lipids containing docosahexaenoic acid.

Major phosphorylation site (Ser55) of neurofilament L by cyclic AMP-dependent protein kinase in rat primary neuronal culture.

Ser55 of neurofilament L (NF-L) is reported to be partly phosphorylated in neurons and to be phosphorylated by cyclic AMP-dependent protein kinase (PKA). Bovine NF-L was phosphorylated by PKA in a low concentration of MgCl2 (0.3 mM) and digested by trypsin. Trypsin-digested fragments were assigned by MALDI/ TOF (matrix-assisted laser desorption and ionization/ time-of-flight) mass spectrometry. Phosphorylation sites were found at Ser41, Ser55, and Ser62 in the head region, with Ser55 considered the preferred site. A site-specific phosphorylation-dependent antibody against Ser55 rendered NF-L phosphorylated at Ser55 detectable in primary cultured rat neurons. One-hour treatment with 20 nM okadaic acid increased the phosphorylation level of Ser55, and co-treatment with 10 microM forskolin enhanced it. However, forskolin alone did not elevate the phosphorylation level. As a consequence, NF-L may be phosphorylated at Ser55 by PKA or by a PKA-like kinase in vivo; however, the phosphorylation level of Ser55 may be modulated by certain phosphatases sensitive to okadaic acid.

Ionization in matrix-assisted laser desorption/ionization: singly charged molecular ions are the lucky survivors.

A new model for the ionization processes in UV matrix-assisted laser desorption/ionization (MALDI) which accounts for the major phenomena observed is presented and discussed. The model retains elements of earlier approaches, such as photoionization and photochemical reactions, but it redefines these in the light of new working questions, most importantly why only singly charged ions are detected. Based on experimental evidence, the formation of singly and multiply charged clusters by a deficiency/excess of ions and also by photoionization and subsequent photochemical processes is pointed out to be the major ionization processes, which typically occur in parallel. The generation of electrons and their partial loss into the surrounding vacuum and solid, on the one hand, results in a positively charged ion-neutral plume facilitating a high overall ionization yield. On the other hand, these electrons, and also the large excess of protonated matrix ions in the negative ion mode, induce effective ion reneutralization in the plume. These neutralization processes are most effective for the highly charged cluster ions initially formed. Their fragmentation behaviour is evidenced in fast metastable fragmentation characteristics and agrees well with an electron capture dissociation mechanism and the enthalpy transfer upon neutralization forms the rationale for the prominent fragmentation and intense chemical noise accompanying successful MALDI. Within the course of the paper, cross-correlations with other desorption/ionization techniques and with earlier discussions on their mechanisms are drawn.

Matrix-Assisted Infrared-Laser Desorption-Ionization Mass Spectrometry of Organic Molecules on NaNO3

We demonstrate sensitive mass identification of model organic compounds directly from sodium nitrate by infrared matrix-assisted laser desorption and ionization mass spectrometry. Sensitivity limits of order 10−6 are achieved for resonant desorption, with a linear calibration verified for crown ether molecules over almost three decades in concentration. We also have observed that crown ether molecules can abstract atomic sodium from the sample when irradiated at the resonant frequency of the NO3 stretching vibrations around 7.1 μm. A model for this process is proposed which suggests great promise for this technique on untreated tank waste.

The Use of a Porous Graphitic Carbon Column for Desalting Hydrophilic Peptides prior to Matrix-Assisted Laser Desorption/ Ionization Time-of-Flight Mass Spectrometry

Reversed-phase high-performance liquid chromatography is the standard method for separating peptides generated from proteolytic digests of proteins. Typically, the small hydrophilic peptides from a proteolytic digest are recovered in the flowthrough fraction along with nonvolatile buffers and salts. Unfortunately, the presence of these salts can interfere with subsequent mass spectrometric analysis or Edman sequencing. To overcome this limitation, and thus enable these small peptides to be identified and characterized, we have investigated a porous graphitic carbon (PGC) column for desalting the peptides found in the unretained fraction. Using a volatile mobile phase combined with a PGC column (Hypercarb), we demonstrate that small hydrophilic peptides at the picomole level can be desalted and characterized by matrix-assisted laser desorption and ionization–time-of-flight–mass spectrometry. Even after desalting, we show that the choice of matrix still plays a significant role in distinguishing the small peptides from the matrix background. The utility of this approach is demonstrated with the flowthrough fraction of an endoproteinase Lys-C digest of a recombinant immunoglobulin. In addition, we demonstrate that a PGC column offers an alternative approach for the separation of hydrophilic, phosphorylated peptides from their unphosphorylated counterparts.

Imaging of Vapor Plumes Produced by Matrix Assisted Laser Desorption: A Plume Sharpening Effect

The first gated imaging of both light matrix molecule and heavy biomolecule vapor expansion during matrix assisted laser desorption and ionization is reported, revealing a plume sharpening effect. Laser induced fluorescence imaging of dye-tagged DNasethinspthinspI proteins shows that these heavy molecules (30thinsp000thinspthinspDa) propagate within a very narrow angular distribution compared to that of the 3-HPA matrix (139thinspthinspDa). A special solution of the gas dynamic equations is developed to describe the 3D coexpansion of heavy and light plume components. {copyright} {ital 1999} {ital The American Physical Society}

Experiments towards quantification of saturated and polyunsaturated diacylglycerols by matrix-assisted laser desorption and ionization time-of-flight mass spectrometry

As recently shown, different physiologically relevant lipid classes can easily be analyzed by matrix-assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI–TOF MS). In the present study the first application of MALDI–TOF for the quantitative analysis of diacylglycerols is described. It is shown that the use of a suitable reference sample enables the quantification of diacylglycerols up to the picomolar range. The best reproducibility of quantitative results for diacylglycerols was obtained using a matrix of 2,5-dihydroxybenzoic acid in ethylacetate and incorporation of an internal standard of the same lipid class. A moderate laser power was used, resulting in a very low extent of fragmentation, allowing a quantification by using solely the highest signal arising from sodium adduct formation of diacylglycerols. A linear correlation between peak intensity and lipid concentration over one order of magnitude was found. The applicability of this new technique for the analysis of other lipids like phosphatidylcholines is also discussed.

Lipid Analysis by Matrix-Assisted Laser Desorption and Ionization Mass Spectrometry: A Methodological Approach

Whereas matrix-assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI-TOF-MS) has gained high importance in the field of protein analysis, surprisingly few studies were published about the use of MALDI for lipid analysis. Lipids, however, are well-suited for MALDI since all experiments can be performed in a sole organic phase and, thus, extremely homogeneous matrix/analyte mixtures are formed. We report here for the first time the application of MALDI-TOF-MS for the analysis of diacylglycerols, phosphatidylcholines, and (poly)phosphoinositides. It is shown that in a matrix of 2,5-dihydroxybenzoic acid the molecular ions (M + 1) of phosphatidylcholines as well as the corresponding adducts of different phosphoinositides are easily detected even in complex mixtures, and thus, detailed data on the fatty acid composition are provided. In contrast, diacylglycerols are mainly detected as the corresponding sodium or potassium adducts, but not as the protonated forms. Fragmentation reactions of fatty acids on the double bonds and on the polar lipid head group are observed to a minor extent in the spectra of all investigated lipids. Generally, choline derivatives are most sensitive toward further fragmentation reactions. Due to its very high sensitivity (up to picomolar concentrations) MALDI-TOF-MS can be used for the direct investigation of biologically relevant lipid mixtures occurring, e.g., in cell membranes. The analysis of the lipid composition of neutrophilic granulocytes is given as a representative example for future applications.

Neisseria gonorrhoeaeThat Infect Men Have Lipooligosaccharides with Terminal N-Acetyllactosamine Repeats

Infectious Neisseria gonorrhoeae make relatively large lipooligosaccharides (LOS) that structurally resemble human glycosphingolipids. MS11mkC is an LOS variant of N. gonorrhoeae strain MS11 which was isolated from men at the onset of dysuria (Schneider, H., Griffiss, J. M., Boslego, J. W., Hitchcock, P. J., Zahos, K. M., and Apicella, M. A. (1991) J. Exp. Med. 174, 1601-1605). Delayed extraction matrix-assisted laser desorption and ionization and electrospray ionization mass spectrometry of O-deacylated MS11mkC LOS produced ions consistent with known LOS which have lacto-N-neotetraose (Galbeta1-->4GlcNAcbeta1-->3Galbeta1-->4Glc; paraglobosyl; monoclonal antibodies (mAbs) 1B2(+) and 06B4(+)) and GalNAc-->lacto-N-neotetraose (gangliosyl; mAb 1-1-M+) oligosaccharides. Ion peaks for a larger LOS which also bound mAb 1B2 indicated the addition of a hexose (+162 Da) to gangliosyl LOS or the addition of a hexose and a N-acetylhexosamine (+365 Da) to paraglobosyl LOS. Analysis of HF-treated and O-deacylated LOS revealed three major components present in a phosphoethanolamine (PEA)0 and a PEA1 series. Digestion of MS11mkC LOS by beta-N-acetylhexosaminidase and beta-galactosidase, alone and sequentially, combined with mAb binding patterns, confirmed the presence of a nonreducing terminal repeating LacNAc ((Galbeta1-->4GlcNAc)2) on the largest LOS, rather than a parallel oligosaccharide structure.

Analysis of phospho- and glycopolypeptides with infrared matrix-assisted laser desorption and ionization.

The analytical characteristics of infrared (IR) matrix-assisted laser desorption and ionization (MALDI) were investigated for the analysis of phosphopeptides, a phosphopolypeptide, and glycopeptides. Two commercially available instruments, a high-resolution delayed extraction (DE) reflectron time-of-flight (RETOF) mass spectrometer and a high-power pulsed Er:YAG laser, were interfaced to produce a high-resolution MALDI-DE-RETOF instrument that is easy to use and can be switched between UV- and IR-MALDI mode within seconds. In the interface design, particular attention was paid to maintaining the same professional operating environment for the new IR-MALDI mode as exists for the commercial UV-MALDI mode. This instrument configuration facilitates comparative observation and investigation of the relative analytical merits of IR- and UV-MALDI. The results of studies of the tryptic alpha-casein phosphopeptides, RP1 (a Thr45-monophosphorylated congener of the recombinant protein hirudin variant 1), and fetuin Asn81 tryptic glycopeptides are presented. The elimination of labile substituents such as phosphoric acid and sialic acid is suppressed in IR-MALDI-RETOF mass spectrometry, with concomitant higher analyte ion yields. These results reflect the advantages that accrue from deposition of significantly less internal energy in the case of IR-MALDI.

An algorithm for the identification of proteins using peptides with ragged N- or C-termini generated by sequential endo- and exopeptidase digestions.

We have developed an algorithm (MassDynSearch) for identifying proteins using a combination of peptide masses with small associated sequences (tags). Unlike the approach developed by Matthias Mann, 'Tag searching', in which the sequence tags are generated by gas phase fragmentation of peptides in a mass spectrometer, 'Rag Tag' searching uses peptide tags which are generated enzymatically or chemically. The protein is digested either chemically or with an endopeptidase and the resultant mixture is then subjected to partial exopeptidase degradation. The mixture is analyzed by matrix assisted laser desorption and ionization time of flight mass spectrometry and a list of intact peptide masses is generated, each associated with a set of degradation product masses which serve as unique tags. These 'tagged masses' are used as the input to an algorithm we have written, MassDynSearch, which searches protein and DNA databases for proteins which contain similar tagged motifs. The method is simple, rapid and can be fully automated. The main advantage of this approach is that the specificity of the initial digestion is unimportant since multiple peptides with tags are used to search the database. This is especially useful for proteins like membrane, cytoskeletal, and other proteins where specific endopeptidases are less efficient and lower specificity proteases such as chymotrypsin, pepsin, and elastase must be used.

Identification of the binding surface on beta-lactamase for GroEL by limited proteolysis and MALDI-mass spectrometry.

Escherichia coli beta-lactamase, alone or as a complex with GroEL at 48 degreesC, was partially digested with trypsin, endoproteinase Glu-C, or thermolysin. Peptides were analyzed by matrix-assisted laser desorption and ionization mass spectrometry and aligned with the known sequence. From the protease cleavage sites which become protected upon binding and those which become newly accessible, a model of the complex is proposed in which the carboxy-terminal helix has melted, two loops form the binding interface and the large beta-sheet become partially uncovered by the slight dislocation of other structural elements. This explains how hydrophobic surface on the substrate protein can become accessible while scarcely disrupting the hydrogen bond network of the native structure. An analysis of the GroEL-bound peptides bound after digestion of the beta-lactamase showed no obvious sequence motifs, indicating that binding is provided by hydrophobic patches in the three-dimensional structure.

Matrix-assisted laser desorption and ionization in the OH and CO absorption bands of aliphatic and aromatic matrices: dependence on laser wavelength and temporal beam profile

A tunable free-electron laser (FEL) was used to initiate infrared (IR) matrix-assisted laser desorption and ionization (MALDI) of small proteins in aliphatic and aromatic matrices. The laser wavelength was scanned from 2.65 to 4.2 μm and from 5.5 to 6.5 μm, covering the absorption bands of the OH and CO stretching vibrations found in such commonly used IR matrices as succinic, fumaric and nicotinic acids. The temporal profile of the laser pulse was also varied using a broadband electro-optic switch (Pockels cell) to study the effects of fluence and irradiance. Although there are absorption peaks at 3.3 μm for succinic acid and fumaric acid, and at 4.1 μm for nicotinic acid, the lowest threshold-fluence for IR MALDI in this region was around 2.94 μm for all matrices. Moreover, the threshold-fluence increased with increasing absorption up to a value five times that of the 2.94 μm value. This result raises questions about the relative contributions of the different sample constitutents to the absorption and the role of resonant absorption in IR MALDI. The threshold-fluences are typically one order of magnitude higher than those for ultraviolet (UV) MALDI, while extinction coefficients of the IR matrices are 100–1000 times smaller than for UV matrices. Therefore, the absorbed energies per unit volume at the MALDI threshold are 10–100 times smaller than in UV MALDI. All these facts clearly indicate that a different desorption/ionization process must be operative in IR MALDI. Variations in temporal profile of the FEL pulse also revealed that ion desorption depends on laser irradiance rather than laser fluence, a result which cannot be explained simply by energy loss due to heat conduction. Two possible models for IR desorption are suggested based on these observations.

Ionization Modelling of Matrix Molecules in Ultraviolet Matrix‐assisted Laser Desorption/Ionization

A scheme of matrix ionization in UV matrix-assisted laser desorption/ionization is presented. Excitation to an upper lying electronic singlet state by the absorption of two photons and subsequent thermionic emission by vibronic coupling is proposed as the primary route for the formation of unprotonated matrix ions within the desorbed gas-phase plume. Under conditions where matrix/matrix collisions are sufficiently frequent to establish a thermal distribution of energy amongst the matrix vibrational states, thermionic emission is assumed to proceed as a unimolecular reaction that draws upon the vibrational energy reservoir of individual molecules. The dominant free-electron loss mechanism is assumed to be electron/neutral attachment, thus providing an equal number of positive and negative matrix ions whilst maintaining the overall ionization level. Predictions based on excited-state thermionic emission and a photoionization model shows that the former provides a considerably better agreement with experimentally determined ion yield vs. fluence characteristics of matrices. For example, for a 3 ns (full width at half maximum) Gaussian N2 laser (337 nm) incident fluence (F) of 15 mJ cm−2, the predicted fractional ion yield is ∼9.8 × 10−4 scaling as F8.5. © 1997 John Wiley & Sons, Ltd.
