Claustrum of the short-tailed fruit bat, Carollia perspicillata: Alignment of cellular orientation and functional connectivity.

The claustrum is a gray-matter structure that underlies neocortex and reciprocates connections with cortical and subcortical targets. In lower mammals, the claustrum is directly adjacent to neocortex, making the definition of claustral boundaries challenging. Latexin, an endogenous inhibitor of metallocarboxypeptidases, localizes to claustral cells, enabling a clear delineation of claustrum. Given its proportionately large claustrum, we hypothesized that the short-tailed fruit bat, Carollia perspicillata, can be a useful model for claustral structure-function relations. We used latexin immunohistochemistry to identify claustral boundaries and intrinsic structure and multielectrode recordings from brain slices to explore intrinsic excitatory connectivity of the claustrum. Carollia's claustrum contains cells whose intrinsic connectivity and alignment permit the generation of spontaneous, synchronous population events and mirror their pattern of spread in disinhibited brain slices over millimeters. Carollia shows cellular alignment and spontaneous population-activity spread along both horizontal and dorsoventral axes. Carollia claustrum possesses intrinsic excitatory connectivity sufficient to: 1) generate single, spontaneous, synchronized burst discharges, 2) support activity spread along axes where claustral cells are aligned, and 3), because of multiple axes for cell alignment, support activity spread along both rostrocaudal and dorsoventral axes. The smaller event sizes in bat claustrum compared with rat claustrum are consistent with events occurring in population subsets rather than the full claustral cell population. The overall size of claustrum, its pronounced vascularity, and its more complex intrinsic connectivity than rat suggest that the bat is an animal model for claustral structure and function that will permit unique access to claustrum's processing capabilities. Claustrum: a case for directional, excitatory, intrinsic connectivity in the rat.

Claustrum, a gray matter structure that underlies the neocortex, is reciprocally connected with many neocortical and limbic cortical areas. This connectivity positions claustrum ideally for the integration or coordination of widespread cortical activity. In anatomical studies using multiple planes of section, claustrum has distinct subregions based on latexin immunohistochemistry, and an approximately rostro-caudal alignment of fusiform cells supporting a laminar intrinsic organization. Physiological studies of claustral connectivity in disinhibited brain slices demonstrate (1) intrinsic connectivity sufficient to generate spontaneous synchronized burst discharges, (2) activity spread within the oblique laminae that contained the principal cellular axis, and (3) segregation of activity as evidenced by the absence of spread within coronal planes. Activity spread depended on glutamatergic synaptic transmission, and activity restrictions did not depend on inhibitory circuits. We conclude that the claustrum has an intrinsic excitatory connectivity that is constrained in approximately rostro-caudal laminae, with minimal cross-communication between laminae. Further, claustrum has the intrinsic capability of generating synchronized population activity and facilitating its spread within laminae, a feature that may contribute to seizure generation and spread.

Local axon collaterals of area CA1 support spread of epileptiform discharges within CA1, but propagation is unidirectional

CA3 and subiculum are hippocampal formation regions that can initiate seizure activity because each has a substantial intrinsic excitatory connectivity. We studied the intrinsic connectivity of area CA1 by exploring the spread of synchronous population discharges in ventral hippocampal slices from rats using a recording chamber that permitted multiple simultaneous extracellular recordings along all laminae of CA1. Brief single stimulus pulses were applied to stratum oriens (SO) or stratum radiatum (SR) on the CA3 side or the subicular side of CA1. In disinhibited slices, events triggered with SO or SR stimulation on the CA3-side propagated over the proximo-distal extent of CA1 with a maximal conduction velocity of 0.4 m/s, comparable with antidromic conduction velocities within CA1. By contrast, SO or SR stimuli applied on the subicular side of CA1 triggered events that did not spread "backward" toward CA3. These events are rapidly decremented in amplitude and duration. Whereas antidromic responses were largest when stimuli were applied on the subicular side of CA1, such responses were not sufficient to trigger epileptiform discharges when excitatory transmission was intact. We conclude that the unidirectional spread of epileptiform activity in area CA1 is the result of an intrinsic axon collateral system where each pyramidal cell has a proportionally larger projection toward subiculum. Although this collateral system is sparse compared with other hippocampal formation regions, its unidirectionality protects against re-entrant activation of CA3 and may be physiologically significant as a relay from proximal CA1 to distal CA1.

Intrinsic excitatory connections in the prefrontal cortex and the pathophysiology of schizophrenia

Working memory, a fundamental cognitive process that is disturbed in schizophrenia, appears to depend upon the sustained activity of specific populations of neurons in the prefrontal cortex. Understanding the neural mechanism(s) that may contribute to the sustained activity of these neurons represents a critical step in predicting the types of alterations in prefrontal circuitry that may be present in schizophrenia, and in determining how such alterations may contribute to the cognitive symptoms of this disorder. This article reviews recent findings which suggest that intrinsic horizontal connections among pyramidal neurons in layer 3 of the dorsolateral prefrontal cortex may provide a critical anatomical substrate for working memory processes, and that alterations in these connections may account for the observations of disturbed working memory, adolescence-related onset of clinical features, and certain pathological changes in the prefrontal cortex of subjects with schizophrenia.

Cholinergic modulation of cortical oscillatory dynamics.

1. The effect of cholinergic modulation on cortical oscillatory dynamics was studied in a computational model of the piriform (olfactory) cortex. The model included the cholinergic suppression of neuronal adaptation, the cholinergic suppression of intrinsic fiber synaptic transmission, the cholinergic enhancement of interneuron activity, and the cholinergic suppression of inhibitory synaptic transmission. 2. Electroencephalographic (EEG) recordings and field potential recordings from the piriform cortex were modeled with a simplified network in which cortical pyramidal cells were represented by excitatory input/output functions with gain parameters dependent on previous activity. The model incorporated distributed excitatory afferent input and excitatory connections between units. In addition, the model contained two sets of inhibitory units mediating inhibition with different time constants and different reversal potentials. This model can match effectively the patterns of cortical EEG and field potentials, showing oscillatory dynamics in both the gamma (30-80 Hz) and theta (3-10 Hz) frequency range. 3. Cholinergic suppression of neuronal adaptation was modeled by reducing the change in gain associated with previous activity. This caused an increased number of oscillations within the network in response to shock stimulation of the lateral olfactory tract, effectively replicating the effect of carbachol on the field potential response in physiological experiments. 4. Cholinergic suppression of intrinsic excitatory synaptic transmission decreased the prominence of gamma oscillations within the network, allowing theta oscillations to predominate. Coupled with the cholinergic suppression of neuronal adaptation, this caused the network to shift from a nonoscillatory state into an oscillatory state of predominant theta oscillations. This replicates the longer term effect of carbachol in experimental preparations on the EEG potential recorded from the cortex in vivo and from brain-slice preparations of the hippocampus in vitro. Analysis of the model suggests that these oscillations depend upon the time constant of neuronal adaptation rather than the time constant of inhibition or the activity of bursting neurons. 5. Cholinergic modulation may be involved in switching the dynamics of this cortical region between those appropriate for learning and those appropriate for recall. During recall, the spread of activity along intrinsic excitatory connections allows associative memory function, whereas neuronal adaptation prevents the spread of activity between different patterns. During learning, the recall of previously stored patterns is prevented by suppression of intrinsic excitatory connections, whereas the response to the new patterns is enhanced by suppression of neuronal adaptation.

Intrinsic synaptic organization of the motor cortex.

Recent anatomical and electrophysiological studies of the intrinsic synaptic circuitry of the motor cortex are revealing novel aspects related to the functional organization of this cortical area. These studies demonstrate that the motor cortex is composed of modules consisting of columnar aggregates of neurons related to different aspects of the same movement. Excitatory and inhibitory connections of intracortical origin link neurons within each of these cortical modules, and also mediate interactions between different cortical modules located as far as 2-3 mm from each other. The intrinsic excitatory connections utilize glutamate, and act via both NMDA and non-NMDA postsynaptic receptors. The intrinsic inhibitory connections are GABAergic. Intrinsic cortical circuits are involved in the temporal coordination of different cortical modules for the execution of complex movement patterns. In addition, the intrinsic inhibitory and excitatory connections are involved in the plasticity of representation zones in the motor cortex, a phenomenon that occurs following nerve damage or during the acquisition of novel motor skills.

Receptive-field properties and neuronal connectivity in striate and parastriate cortex of contour-deprived cats.

An attempt was made to relate the alterations of cortical receptive fields as they result from binocular visual deprivation to changes in afferent, intrinsic, and efferent connections of the striate and parastriate cortex. The experiments were performed in cats aged at least 1 jr with their eyelids sutured closed from birth. The results of the receptive-field analysis in A17 confirmed the reduction of light-responsive cells, the occasional incongruity of receptive-field properties in the two eyes, and to some extent also the loss of orientation and direction selectivity as reported previously. Other properties common to numerous deprived receptive fields were the lack of sharp inhibitory sidebands and the sometimes exceedingly large size of the receptive fields. Qualitatively as well as quantitatively, similar alterations were observed in area 18. A rather high percentage of cells in both areas had, however, preserved at least some orientation preference, and a few receptive fields had tuning properties comparable to those in normal cats. The ability of area 18 cells in normal cats to respond to much higher stimulus velocities than area 17 cells was not influenced by deprivation. The results obtained with electrical stimulation suggest two main deprivation effects: 1) A marked decrease in the safety factor of retinothalamic and thalamocortical transmission. 2) A clear decrease in efficiency of intracortical inhibition. But the electrical stimulation data also show that none of the basic principles of afferent, intrinsic, and efferent connectivity is lost or changed by deprivation. The conduction velocities in the subcortical afferents and the differentiation of the afferents to areas 17 and 18 into slow- and fast-conducting projection systems remain unaltered. Intrinsic excitatory connections remain functional; this is also true for the disynaptic inhibitory pathways activated preferentially by the fast-conducting thalamocortical projection. The laminar distribution of cells with monosynaptic versus polsynaptic excitatory connections is similar to that in normal cats. Neurons with corticofugal axons remain functionally connected and show the same connectivity pattern as those in normal cats. The nonspecific activation system from the mesencephalic reticular formation also remains functioning both at the thalamic and the cortical level. We conclude from these and several other observations that most, if not all, afferent, intrinsic, and efferent connections of areas 17 and 18 are specified from birth and depend only little on visual experience. This predetermined structural plan, however, allows for some freedom in the domain of orientation tuning, binocular correspondence, and retinotopy which is specified only when visual experience is possible.
