Antidiarrheal activities of methanolic crude extract and solvent fractions of the root of Verbascum sinaiticum Benth. (Scrophularaceae) in mice

BackgroundIn Ethiopian traditional medicine, V. sinaiticum is one of the most often utilized medicinal herbs for the treatment of diarrhea. Therefore, this study was conducted to validate the use of the plant for the treatment of diarrhea in the traditional medical practice of Ethiopia. MethodsCastor oil-induced diarrhea, enteropooling, and intestinal motility test models in mice were used to evaluate the antidiarrheal properties of the 80% methanol crude extract and the solvent fractions of the root component of V. sinaiticum. The effects of the crude extract and the fractions on time for onset, frequency, weight, and water content of diarrheal feces, intestinal fluid accumulation, and intestinal transit of charcoal meal were evaluated and compared with the corresponding results in the negative control. ResultsThe crude extract (CE), aqueous fraction (AQF), and ethyl acetate fraction (EAF) at 400 mg/kg (p < 0.001) significantly delayed the onset of diarrhea. Besides, the CE and AQF at 200 and 400 mg/kg (p < 0.001) of the doses, and EAF at 200 (p < 0.01) and 400 mg/kg (p < 0.001) significantly decreased the frequency of diarrheal stools. Furthermore, CE, AQF, and EAF at their three serial doses (p < 0.001), significantly reduced the weights of the fresh diarrheal stools as compared to the negative control. The CE and AQF at 100 (p < 0.01), and 200 and 400 mg/kg (p < 0.001) of their doses and EAF at 200 (p < 0.01) and 400 mg/kg (p < 0.001) significantly decreased the fluid contents of diarrheal stools compared to the negative control. In the enteropooling test, the CE at 100 (p < 0.05), and 200 and 400 mg/kg (p < 0.001), AQF at 200 (P < 0.05) and 400 mg/kg (p < 0.01), and EAF at 200 (p < 0.01) and 400 mg/kg (p < 0.001) significantly decreased the weights of intestinal contents compared to the negative control. Additionally, the CE at 100 and 200 (p < 0.05) and 400 mg/kg (p < 0.001), AQF at 100 (p < 0.05), 200 (p < 0.01), and 400 mg/kg (p < 0.001) of the doses, and EAF at 400 mg/kg (p < 0.05), produced significant reductions in the volumes of intestinal contents. In the intestinal motility test model, the CE, AQF, and EAF at all their serial doses (p < 0.001), significantly suppressed the intestinal transit of charcoal meal and peristaltic index compared to the negative control. ConclusionOverall, the results of this study showed that the crude extract and the solvent fractions of the root parts of V. sinaiticum had considerable in vivo antidiarrheal activities. Besides, the crude extract, especially at 400 mg/kg, produced the highest effect followed by the aqueous fraction at the same dose. This might indicate that the bioactive compounds responsible for the effects are more of hydrophilic in nature. Moreover, the antidiarrheal index values were increased with the doses of the extract and the fractions, suggesting that the treatments might have dose-dependent antidiarrheal effects. Additionally, the extract was shown to be free of observable acute toxic effects. Thus, this study corroborates the use the root parts of V. sinaiticum to treat diarrhea in the traditional settings. Furthermore, the findings of this study are encouraging and may be used as the basis to conduct further studies in the area including chemical characterization and molecular based mechanism of actions of the plant for its confirmed antidiarrheal effects.

Ethanol leaf extract of Dialium guineense produces anti-diarrhoeal and gastrointestinal motility slowing activities in Wistar rats

Dialium guineense leaves have been traditionally used for the treatment of diarrhoeal diseases in Nigeria. The study was conducted to evaluate the anti-diarrhoeal and gastrointestinal motility slowing effects of the ethanol leaf extract of D. guineense in Wistar rats. The anti-diarrhoeal and gastrointestinal motility slowing effects of the ethanol leaf extract of D. guineense were evaluated using castor oil-induced diarrhoeal model, charcoal meal, and anti-enteropooling tests in Wistar rats. The test groups received various doses (100, 200, and 400 mg/kg) of the extract whereas positive controls received Loperamide (4 mg/kg) or Atropine (5 mg/kg) and negative controls received Normal Saline (10 ml/kg). The phytochemical screening as well as the acute toxicity test of the extract was also performed. The extract produced a dose-dependent significant reduction in the watery nature and frequency of fecal droppings in the castor oil-induced diarrhea. On gastrointestinal transit time and enteropooling, the extract also dose-dependently reduced the small intestinal transit of charcoal meal and intestinal fluid volume in a manner comparable to 5 mg/kg of atropine sulphate and 4 mg/kg of loperamide. The acute toxicity study on the extract revealed an oral LD50 value greater than 5000 mg/kg in mice. The phytochemical constituents detected in the extract were tannins, phenols, resin, alkaloids, saponins, terpenoids, and glycosides. The findings from this study showed that the ethanol leaf extract of D. guineense possesses anti-diarrhoeal properties and thus supports the traditional application of the leaf extract in the treatment of diarrhea in Nigeria.

Neuropharmacological and Antidiarrheal Potentials of Duabanga grandiflora (DC.) Walp. Stem Bark and Prospective Ligand-Receptor Interactions of Its Bioactive Lead Molecules.

Duabanga grandiflora (DC.) Walp. is an ethnomedicinally significant plant used to treat various illnesses, but there is little scientific evidence to support its use. This study explored the pharmacological activities of methanol extract of D. grandiflora stem barks (MEDG) through in vivo approaches in Swiss albino mice and a computer-aided molecular approach. The forced swimming test (FST), tail suspension test (TST), elevated plus maze (EPM), and hole board test (HBT) were used to determine anti-depressant and anxiolytic activity in experimental mice. In addition, anti-diarrheal studies were performed using castor oil-induced diarrhea, castor oil-induced enter pooling, and the charcoal-induced gastrointestinal motility test. MEDG showed substantial depletions in the immobility times in both FST and TST after treatment with the MEDG extract, whereas moderate anxiolytic activity was manifested at a higher dose (400 mg/kg) compared with the control. Correspondingly, MEDG extract revealed a significant reduction in wet feces and decreased the small intestinal transit of charcoal meal in castor oil-induced diarrhea and charcoal-induced gastrointestinal motility test. In the computer-aided molecular approaches, vanillin displayed a promising binding score for both anxiolytic and anti-diarrheal activities, while duabanganal C showed a promising score for the anti-depressant activity. The present experimental findings along with a computer-aided model conclude that MEDG could be a possible Phyto therapeutic agent with potential anti-depressant, anxiolytic and anti-diarrheal activity.

Evaluation of spasmolytic effects of naringenin on ileum contraction and intestinal charcoal meal transit: Involvement of ATP-sensitive K+ channels

Introduction: Naringenin is a flavonoid constituent of many herbal plants, including citreous fruits. Biological studies have suggested various therapeutic effects for naringenin, including protective effects on gastrointestinal (GI) motility. The present study was performed to investigate the involvement of ATP-sensitive K+ channels on the effect of naringenin in rat ileum motility. Methods: Ileum contractions were induced by either KCl or acetylcholine (ACh) in vitro. Inhibitory concentration-response curves were constructed for naringenin and diazoxide after exposure of rat isolated ileum to KCl (20mM) or ACh (500nM). The relaxant effects of naringenin and diazoxide were also examined in the presence of glibenclamide. Furthermore, oral effects of diazoxide (25 mg/kg) and naringenin (25, 50 mg/kg) were also assessed on the intestinal charcoal meal transit in mice (n=10) in the absence and presence of glibenclamide (50 mg/kg). Results: Diazoxide and naringenin in a concentration-dependent manner inhibited ileum contractions induced by low bath concentration of KCl (20mM). However, both drugs had no effect on contractions induced by a high concentration of KCl (160mM). The inhibitory effects of diazoxide and naringenin were blocked by glibenclamide. Oral administration of diazoxide and naringenin significantly reduced the intestinal transit of charcoal meal. The delay in the intestinal transit was blocked by the oral dose of glibenclamide. The effect of naringenin on the rat intestinal strip pre-contracted with the KCl was relatively similar to that of ATP-sensitive K+ channel opener (diazoxide). Conclusion: This research supports that ATP-sensitive K+ channels are involved in the rat small intestinal smooth muscles relaxation induced by naringenin.

In vivo anti-diarrheal activity of jujube honey on castor oil-induced diarrhea in mice.

To evaluate the antidiarrheal effect of jujube honey on experimentally castor oil-induced diarrhea in mice by using different testing models (diarrhea, enteropooling and gastrointestinal motility). The mice intragastric administration castor oil was post-treated after 30 min with jujube honey, diluted jujube honey and loperamide or vehicles in different experimental groups. The onset and number of wet defecation on the absorbent paper was recorded for each animal for 4 h. Plasma was examined for C-reactive protein (CRP) and nitric oxide (NO) for clinical inflammation evaluation. The oxidative stress was investigated by superoxide dismutase (SOD) and catalase levels. The diluted jujube honey exhibited an important antidiarrheal activity manifested by significant delay in onset of diarrhea (P < 0.05), loss in number of wet stools (P < 0.001), total number of stools (P < 0.001) and total stool weight of fecal output (P < 0.001) in 4 h in castor oil-induced diarrheal groups. The inhibition of intestinal transit of charcoal meal (P < 0.01) is a most likely mechanism that may account for antidiarrheal effect of jujube honey. It also significantly increased SOD and catalase levels (P < 0.001) and significantly decreased CRP and NO in plasma diarrheic mice (P < 0.01). The results of this research reveal that the jujube honey contains pharmacologically active compounds with antidiarrheal properties. However, diluted honey is more effective in the treatment of diarrhea and imbalance of intestinal motility. In conclusion, these findings illustrated the antidiarrheal activity effect of jujube honey and which has the strongest evidence supporting its use in the treatment of diarrhea in traditional medicine.

Effect of Aqueous Ethanol Stem Extract of Entada africana Guill. Et Perrott. on Castor Oil and Magnesium Sulphate-Induced Diarrhoea Models in Mice

This study investigated the phytochemical, elemental analysis, acute toxicity study of aqueous ethanol stem bark extract of Entada africana, as well as its antidiarrhoeal activity in mice and its effects on isolated rabbit jejunum. The preliminary phytochemical screening revealed the presence of alkaloids, flavonoids, cardiac glycosides, phenols, saponins, steroids, tannins and terpenoids. Elemental analysis of the extract showed the presence of magnesium (Mg), manganese (Mn), iron (Fe), copper (Cu), lead (Pb), zinc (Zn) and sodium (Na) while acute toxicity study revealed intraperitoneal median lethal dose (LD50) values for the extract to be 774.6 mg/kg body weight. The antidiarrheal effect of the extract was studied using castor-oil and magnesium sulphate induced diarrhoeal models (dropping test) and gastrointestinal transit test in mice. The result showed that the extract produced a dose-dependent protection against diarrhoea induced by castor oil and magnesium sulfate, with the highest protection (80 and 100%), obtained at 100 and 200 mg/kg. The extract significantly (p≤0.01) reduced the small intestinal transit of charcoal meal in mice at all doses tested. The extract (0.4-3.2 mg/ml) produced a concentration dependent relaxation of the rabbit jejunum, and the effects were blocked by propranolol (0.04 and 0.64 μg/ml). The results of this study showed that the extract contain pharmacologically active substance with antidiarrhoeal properties mediated through inhibition of hyper secretion and reduced gastrointestinal motility. These properties may explain the rationale for use of it’s stem bark as antidiarrhoeal remedy in traditional medicine.

Antidiarrheal Activity of Hydromethanolic Root Extract and Solvent Fractions of Clutia abyssinica Jaub. & Spach. (Euphorbiaceae) in Mice.

Introduction Diarrheal diseases are associated with an estimated 1.3 million deaths annually, with most occurring in resource-limited countries; up to 25% of deaths in young children living in Africa and southeast Asia are attributable to acute gastroenteritis. Due to limitations associated with various treatments available, the need for developing newer drugs is imperative. Objective This study was aimed to evaluate the antidiarrheal activity of root extract and fractions of C. abyssinica Jaub. & Spach. (Euphorbiaceae) in mice. Methods After plant extraction and subsequent fractionation of the crude extract, the antidiarrheal activity was screened in castor oil induced diarrhea, castor oil induced enteropooling, and gastrointestinal motility test models accordingly. Result The root extract of C. abyssinica produced neither visible signs of toxicity nor death at a single dose of 2000 mg/kg, suggesting the LD50 > 2000 mg/kg. In the castor oil induced diarrheal model, the highest dose of the extract (400 mg/kg) showed a maximal inhibition in the onset (158.00 ± 14.64, p < 0.01, in minutes) of wet feces as compared to the negative control. In the enteropooling model, 400 mg/kg treated mice showed a significant reduction in volume (0.47 ± 0.02 ml, p < 0.01) and weight (0.50 ± 0.02 g, p < 0.05) of intestinal content as compared to the vehicle treated group. In the gastrointestinal motility test, the hydromethanolic root extract of C. abyssinica significantly inhibited the intestinal transit of charcoal meal at 400 mg/kg. In addition, chloroform and n-butanol fractions significantly reduced the distance moved by charcoal at doses of 200 mg/kg and 400 mg/kg, whereas aqueous fraction showed a significant effect at all test doses. The highest antidiarrheal index was observed at the maximal dose of extract and each fraction. Conclusion The results obtained showed that the findings provide scientific support for the folkloric repute of C. abyssinica roots as treatment of diarrhea.

Antidiarrheal activity of some selected Nigerian plants used in traditional medicine

Context: Herbal preparations of the various parts of Vitellaria paradoxa, Neorautanenia mitis, Senna surattensis, and Hydnora abyssinica have been used in the Nigerian traditional medical practice to treat the symptoms of diarrhea in humans and animals. Aims: This study aims to validate claims of the traditional use of these plants in the treatment of diarrhea and provide a scientific basis for further studies. Materials and Methods: The median lethal dose (LD50) values of the extracts were obtained using the Limit test. Castor oil induced diarrhea and intestinal transit (motility) models in albino rats were used to determine the antidiarrheal activity. Graded doses of the extracts were administered to 3 test groups, while the positive control groups were given standard drugs (atropine and loperamide) and the negative control groups received distilled water per os. Results: The LD50was considered higher than 2000 mg/Kg for all the extracts. In the castor oil-induced diarrhea model, the highest percentage inhibition of defecation was observed in the test groups treated with the extracts of H. abyssinica (82%) followed by S. surattensis (81%), N. mitis (66%), and V. paradoxa (32%). H. abyssinica extract significantly decreased the intestinal transit of charcoal meal compared to the other extracts. Conclusion: The antidiarrheal activities of all the extracts give credence to their traditional use. H. abyssinica comparatively had the best antidiarrheal activity and has the potential as an antidiarrheal agent. Thus, the need for further studies of this extract to investigate active fractions, isolate and characterize active compounds, and determine their activities and safety.

Hot-water extract of the branches of Hovenia dulcis Thunb. (Rhamnaceae) ameliorates low-fiber diet-induced constipation in rats.

Hovenia dulcis Thunb. (Rhamnaceae), also known as oriental raisin tree, is used in traditional herbal medicine. Its extracts have been reported to show various pharmacological effects such as hepatoprotection, antitumor, antiatopic dermatitis, antilipid peroxidation, anti-steatotic, anti-inflammatory, and antiallergic activities. However, there have been no reports on the effect of H. dulcis extracts in relieving constipation so far. The aim of this study was to investigate the effects of a hot-water extract of the branches of H. dulcis (WEHD) on low-fiber diet-induced constipation in Sprague Dawley rats. The in vivo laxative activity of WEHD was assessed by measuring the intestinal transit of charcoal meal and stool parameters. Furthermore, the in vitro spasmogenic activity of WEHD was evaluated by monitoring the temporal profiles of contraction of rat colon in the absence or presence of WEHD. In addition, constituent profiling was conducted using high-performance liquid chromatography analysis. Pretreatment with WEHD significantly enhanced the intestinal transit of charcoal meal and increased the frequency and weight of stools in rats. In addition, the frequency and amplitude of contractile responses of isolated rat colon were markedly enhanced by WEHD. Two organic phenolic acids, ferulic and vanillic acids, were identified in WEHD, of which vanillic acid exhibited spasmogenic activity. To the best of our knowledge, this is the first study to report the laxative and spasmogenic activities of H. dulcis and its constituents, suggesting that WEHD can serve as a complementary and/or alternative laxative in alleviating chronic constipation.

Antidiarrheal activity of 80% methanol extract of the aerial part of Ajuga remota Benth (Lamiaceae) in mice.

BackgroundIn the Ethiopian traditional medicine, the aerial part of Ajuga remota Benth is used in the treatment of diarrhea. There are different mechanisms by which Ajuga remota may have antidiarrheal effect. Some of the possible mechanisms are through its anthelmintic and antibacterial activity. The present study aimed to evaluate whether the antidiarrheal effect of the plant also include antimotility and antisecretory effect using 80 % methanol extract of A. remota (MEAR).MethodsThe MEAR was administered at doses of 200, 400, 600 and 800 mg/kg to four groups of mice (six animals per group) orally in castor oil diarrhea model. The effect of the extract on enteropooling and gastrointestinal transit model was also evaluated using the same grouping and dosing. Two other groups, one as control and the other as standard (loperamide 5 mg/kg) were used for comparison with the treatment groups.ResultsThe extract at the doses of 400, 600, and 800 mg/kg produced a dose-dependent and significant inhibition both on the frequency and onset of diarrhea. The percentage purging frequency was 53.4, 66.7, 79.6, and 66.7 % (p < 0.001) at three doses of MEAR (400, 600, and 800 mg/kg) and with loperamide (5 mg/kg), respectively. The percentage inhibition in intestinal fluid accumulation was 42.5, 62.1, and 74.2 % (p < 0.001) at the doses of 400, 600 and 800 mg/kg of MEAR, respectively. The MEAR also inhibited the intestinal transit of charcoal meal in a dose dependent manner both in the normal and castor oil induced intestinal transit.ConclusionThis study has shown that the 80 % methanol extract of A. remota contains pharmacologically active substances with significant antimotility and antisecretory effect contributing for its antidiarrheal activity.

Investigation On Antidiarrhoeal Activity Of &lt;i&gt;Aristolochia Indica&lt;/i&gt; Linn. Root Extracts In Mice

The present study aimed at investigating the effect of ethanolic extract (EtAI), and aqueous extract (AqAI) of Aristolochia indica Linn roots on castor oil-induced diarrhoea and study on small intestinal transit. Phytochemical analysis of extracts was performed as per standard procedure. The oral toxicity study using Swiss albino mice was performed in accordance with OECD guidelines. The EtAI and AqAI extracts of Aristolochia indica Linn were studied for antidiarrhoeal property using castor oil-induced diarrhoeal model and charcoal-induced gastrointestinal motility test in Swiss albino mice. Among the tested doses of 200 and 400 mg/kg body weight, the extracts reduced the frequency and severity of diarrhoea in test animals throughout the study period. At the same doses, the extract delayed the intestinal transit of charcoal meal in test animals as compared to the control and the results were statistically significant. Experimental findings showed that ethanol extract of Aristolochia indica Linn root possess significant antidiarrheal activity and may be a potent source of anti-diarrhoeal drug in future.

Some pharmacological studies on the methanolic extract of &amp;lt;i&amp;gt;Inula graveolense&amp;lt;/i&amp;gt; L.

Inula graveolens L. is widely used in Iraq for the treatment of rheumatic fever, infant convulsions, toothache, blood sugar, and also to dissolve internal blood clots, and to aid digestion. However, the efficacy and mechanisms of action of the plant remain unclear. Therefore, the objective of the present study was to investigate the pharmacological effects of the methanolic extract (MEIG) of this plant belonging to compositae family. Anti-diarrheal and antipyretic activities of the extract were examined in rats. Anti-inflammatory and antinociceptive were studied in mice. At the doses of 200 (P < 0.05) and 400 mg/kg body weight (P < 0.01), the extract displayed remarkable anti-diarrheal activity, evidence by a reduction in the rate of defecation as well as by retardation of intestinal transit of charcoal meal compared to normal saline control group, dose dependently similar to loperamide (5 mg/kg). The methanolic extract (400 mg/kg) showed a significant (P < 0.01) dose dependent anti-pyretic effect in yeast induced elevation of body temperature in experimental rats. The methanolic extract showed significant anti-inflammatory and antinociceptive activity at the dose of 400 mg/kg (P < 0.01) as compared to standard drug diclofenac sodium (50 mg/kg). The extract inhibited paw and ear edema in a dose-related manner. A dose-dependent analgesic action was obtained against chemical (writhing test) and thermal (hot-plate test) stimuli indicated that antinociceptive activity may involve inhibition of pain by peripheral and central mechanisms. Again, the methanolic extract (MEIG) was subjected for in vitro protein anti-denaturation using Bovine serum albumin and anti-platelet aggregation of human blood activity. It was observed that the extract showed greater percentage of inhibition of BSA (P < 0.01) at the highest concentration (400 μg/ml). The extract also showed potential platelet aggregation inhibitory activity in adose-dependent manner. The maximum inhibition was observed at the dose 400 μg/ml (P < 0.01) compared to standard drug commercial heparin (20 μg/ml).

Studies on the Antiulcer and Gastrointestinal Effects of Stem Bark Extract of Bridelia ferruginea

The antiulcer and gastrointestinal effects of methanol stem bark extract (BFME) of Bridelia ferruginea Benth. (Euphorbiaceae) and its solvent fractions-dichloromethane (DCMF) and methanol (MF)-were studied using indomethacin- and ethanol-induced ulcers in rats, small intestinal transit of charcoal meal in mice, and the effects on acetylcholine-induced contractions of the isolated guinea pig ileum. The extract and fractions significantly (P<0.05) protected the rats against ethanol and indomethacin-induced ulcers and inhibited small intestinal propulsion in the order of magnitude: DCMF>MF>BFME. On the guinea pig ileum, MF (0.05 - 6.40 mg/ml) elicited no inhibition, DCMF (5 - 40 μg/ml) antagonized acetylcholine-induced contractions of the guinea pig ileum with IC50 of 10.47μg/ml, while BFME (0.05 - 12.80 mg/ml) contracted the guinea pig ileum with EC50 of 1 mg/ml. Oral LD50 of BFME in mice was estimated to be 2,154 mg/kg. Phytochemistry tests revealed the presence of tannins, saponins, steroids, terpenoids, flavonoids and resins in BFME, MF tested positive for tannins, saponins, steroids, terpenoids, and flavonoids, while DCMF gave positive reactions for flavonoids, steroids, terpenoids and resins. These findings suggest that constituents of the stem bark of B. ferruginea possess antiulcer properties. In addition, some non-polar constituents possess spasmolytic activity while spasmogenic activity is likely associated with some polar constituents.

Antidiarrheal Action of Hydroalcoholic Extract of Pycnocycla spinosa in Comparison with Loperamide and Dicyclomine.

Pycnocycla spinosa Decne. ex Boiss. var. spinosa (Fam. Umbelliferae) is an essential oil-containing wild plant growing in central part of Iran. Hydroalcoholic extract of Pycnocycla spinosa has antispasmodic and antidiarrheal activity. The aim of this study was to further investigate antidiarrheal and small intestinal transit effect of P. spinosa extract for a comparison with loperamide and dicyclomine. Male mice fasted over night with free access to water, were treated with P. spinosa extract, loperamide, dicyclomine or vehicle (p.o.). After thirty min, castor oil was given orally to the animals. In separate groups, magnesium sulphate was given in the beginning and 30 min after the extract or drugs were administered. The onset and number of wet defecation on the absorbent paper was recorded for each animal for 2.5 h. In another group of mice, intestinal transit of charcoal meal after the administration of extract, loperamide or dicyclomine was determined and compared with the control group. P. spinosa extract sharply reduced castor oil and magnesium sulphate induced diarrhea. The extract, in dose of 1 mg/Kg, had antidiarrheal effect similar to loperamide (2 mg/Kg) and with dose of 0.5 mg/Kg, its antidiarrheal action was greater than that of dicyclomine (5 mg/Kg). Unlike dicyclomine, P. spinosa extract significantly reduced the small intestinal transit of charcoal meal. However, its inhibitory effect on intestinal transit was less than loperamide. This study shows that anti-diarrhea l effect of P. spinosa extract is similar to loperamide. The inhibition of intestinal propulsion is a most likely mechanism that may account for anti-diarrhea l activity of the extract.

Species differences in the prokinetic effects of ginger

This study describes the prokinetic actions of the aqueous extract of ginger (Zingiber officinale). Ginger extract (Zo.Cr), which tested positive for saponins, terpenes, phenols, flavonoids and alkaloids, showed a spasmogenic effect in isolated guinea-pig ileum with 8–50 times more potency than in rabbit jejunum and ileum and rat stomach fundus and ileum. Spasmogenicity in all the gut preparations except in guinea-pig ileum was atropine-sensitive. Zo.Cr exhibited a stimulant effect in vivo in mice and enhanced the intestinal transit of charcoal meal. A spasmolytic effect, mediated via Ca2 + antagonist activity, was also exhibited by Zo.Cr, reflected in terms of inhibition of spontaneous contractions, K+ (80 mM)-induced contractions and displacement of Ca2 + dose–response curves. The ginger pure compounds (6-shogaol, 6-gingerol, 8-gingerol and 10-gingerol) also exhibited a spasmolytic activity, which reduced with the increasing size of the side chain in their chemical structures. The study showed that the aqueous extract of ginger exhibits species-specific spasmogenicity in gut tissues of rabbit and rat (muscarinic-type) while through an uncharacterized pathway in guinea-pig ileum, along with a dormant relaxant effect, mediated via the blockade of voltage-dependent Ca2 + channels.

Anti-Inflammatory, Antipyretic And Antispasmodic Properties Of Chromolaena Odorata

A methanol extract of the leaves of Chromolaena odorata was evaluated for anti-inflammatory effects in the carrageenan-induced rat paw edema model as well as for antipyretic activity in mice. The effects of the extract on intestinal transit of charcoal meal and castor oil-induced diarrhoea were also investigated. The extract (50-200 mg/kg) inhibited paw edema in rats and produced significant (p <0.05) reduction in rectal temperature of mice rendered hyperthermic by yeast suspension. Antimotility and antidiarrhoeal effects were produced by the extract in intact mice. This study establishes the out-inflammatory, antipyretic, and anti-spasmodic properties of C. odorata.
