Pediococcus pentosaceus RC007 and Saccharomyces boulardii RC009 as antibiotic alternatives for gut health in post-weaning pigs.

The aim of the present study was to evaluate a novel probiotic Pediococcus pentosaceus RC007 used alone and convined with Saccharomyces cerevisiae var. boulardii RC009, as in-feed additives to substitute the non-therapeutic use of antibiotics, and evaluate the different structural characteristics of intestinal bacterial populations between groups, correlated with pig production performance. The in vivo study was conducted on post-weaning pigs, from 21 to 56 days-old. Three dietary treatments were included: T1- basal diet (BD - Control group); T2- BD with P. pentosaceus RC007; and T3- BD with a mix of P. pentosaceus RC007 and S. boulardii RC009. The weight gain increase of pigs consuming non-therapeutic antibiotics was similar to those that did not consume antibiotics during the study (p=0.0234), but had better health indicators. The use of a probiotic combination increased carcass weight and significantly reduced the lumbar fat thickness. In terms of taxonomic composition, there was a tendency to modify the abundance of Proteobacteria, Cyanobacteria, Enterobacteriaceae and Lactobacillaceae in pigs that consumed the additives. The genus Butyricicoccus, Collinsella and Ruminococcus tended to be more abundant in the microbiota of pigs at T3. For the first time, the results of the present study indicate that P. pentosaceus RC007 and S. boulardii RC009, a probiotic combination, could be a good substitute for antibiotics in improving pig production performance, while also contributing to a healthier gut microbiota, especially with the reduced abundance of Proteobacteria and Cyanobacteria.

Modelling the influence of vitamin D and probiotic supplementation on the microbiome and immune response

Abstract The intestinal microbiota play a critical role in human health and disease, maintaining metabolic and immune/inflammatory health, synthesizing essential vitamins and amino acids and maintaining intestinal barrier integrity. The aim of this paper is to develop a mathematical model to describe the complex interactions between the microbiota, vitamin D/vitamin D receptor (VDR) pathway, epithelial barrier and immune response in order to understand better the effects of supplementation with probiotics and vitamin D. This is motivated by emerging data indicating the beneficial effects of vitamin D and probiotics individually and when combined. We propose a system of ordinary differential equations determining the time evolution of intestinal bacterial populations, concentration of the VDR:1,25(OH)$_{2}$D complex in epithelial and immune cells, the epithelial barrier and the immune response. The model shows that administration of probiotics and/or vitamin D upregulates the VDR complex, which enhances barrier function and protects against intestinal inflammation. The model also suggests co-supplementation to be superior to individual supplements. We explore the effects of inflammation on the populations of commensal and pathogenic bacteria and the vitamin D/VDR pathway and discuss the value of gathering additional experimental data motivated by the modelling insights.

Evaluating the potential prebiotic effects of umbu-cajá (Spondias spp.) fruit processing by-product flour on the human intestinal microbiota

This study evaluated the potential prebiotic properties of umbu-cajá (Spondias spp.) fruit processing by-product flour (UCF) on the human intestinal microbiota by monitoring the relative abundance of specific intestinal bacterial populations, microbial metabolic behavior, and antioxidant activity during 48 h of in vitro colonic fermentation using fecal inoculum donated from healthy adult individuals. UCF had a high insoluble fiber content and flavonols, especially quercetin-3-O-glucoside, as the most prevalent phenolic compounds, besides showing antioxidant activity. Medium with UCF increased the relative abundance of Lactobacillus spp./Enterococcus spp., Bifidobacterium spp., and Clostridium histolyticum during the colonic fermentation and decreased the relative abundance of Bacteroides spp./Prevotella spp. and Eubacterium rectale/Clostridium coccoides. Medium with UCF maintained a higher relative abundance of Ruminococcus albus/R. flavefaciens than negative control during the colonic fermentation. UCF stimulated intense metabolic activity of the intestinal microbiota during the colonic fermentation, evidenced by decreased pH values, sugar consumption, and production of several metabolites linked to host health promotion, concomitant with high antioxidant activity. The results characterize UCF as a candidate for prebiotic use and a novel value-added circular ingredient to functionalize foods and nutraceuticals.

The Microbiome in Inflammatory Bowel Disease.

The management of patients with inflammatory bowel disease (IBD) aims to control inflammation through the use of immunosuppressive treatments that target various points in the inflammatory cascade. However, the efficacy of these therapies in the long term is limited, and they often are associated with severe side effects. Although the pathophysiology of the disease is not completely understood, IBD is regarded as a multifactorial disease that occurs due to an inappropriate immune response in genetically susceptible individuals. The gut microbiome is considered one of the main actors in the development of IBD. Gut dysbiosis, characterised by significant changes in the composition and functionality of the gut microbiota, often leads to a reduction in bacterial diversity and anti-inflammatory anaerobic bacteria. At the same time, bacteria with pro-inflammatory potential increase. Although changes in microbiome composition upon biological agent usage have been observed, their role as biomarkers is still unclear. While most studies on IBD focus on the intestinal bacterial population, recent studies have highlighted the importance of other microbial populations, such as viruses and fungi, in gut dysbiosis. In order to modulate the aberrant immune response in patients with IBD, researchers have developed therapies that target different players in the gut microbiome. These innovative approaches hold promise for the future of IBD treatment, although safety concerns are the main limitations, as their effects on humans remain unknown.

Comparative effects of dietary zinc nanoparticle and conventional zinc supplementation on broiler chickens: A meta-analysis.

Zinc (Zn) is important for various physiological processes in broiler chickens, including protein and carbohydrate metabolism, growth, and reproduction. The gastrointestinal absorption of Zn in broiler chickens was notably low. One approach that has been explored for enhancing the bioavailability of Zn is the development of Zn nanoparticles (NPs). Zn is required for various physiological processes in broiler chickens, including protein and carbohydrate metabolism, growth, and reproduction. Therefore, this study aimed to assess the impact of conventional Zn and Zn NPs on broiler chickens using a meta-analysis methodology. A database was built from published literature to evaluate the effects of the addition of Zn NPs and conventional Zn on broiler chicken responses, including the following parameters: production performance; carcass cuts; visceral organ weight; lymphoid organ weight; nutrient digestibility; intestinal villi; mineral Zn, calcium, and phosphorus concentrations; hematology; blood parameters; immunoglobulin; and intestinal bacterial population. Various scientific platforms, including Scopus, Web of Science, PubMed Central, and Google Scholar, were used to search for peer-reviewed articles. A database was created from 25 studies that met the inclusion criteria. The data were then processed for a meta-analysis using a mixed-model methodology. Different types of Zn (NPs versus conventional) were considered fixed effects, different studies were treated as random effects, and p-values were used as model statistics. Across the parameters observed in this study, the use of Zn NPs was more efficient in Zn utilization than conventional Zn, as evidenced by the average dose of Zn NPs being much lower than that of conventional Zn (79.44 vs. 242.76 mg/kg) yet providing similar (p > 0.05) or even significantly better effects (p < 0.05) compared to conventional Zn usage. This investigation revealed the beneficial influence of Zn NPs in broiler chickens compared to the conventional utilization of Zn through an all-encompassing meta-analysis. Moreover, Zn NPs have proven to be more effective in Zn utilization when juxtaposed with conventional Zn, as demonstrated by the significantly lower quantity of Zn NPs administered compared to conventional Zn, while yielding comparable or even superior outcomes compared to the traditional utilization of Zn. A limitation of this study is that the Zn NPs used were sourced from inorganic Zn NPs. Therefore, future research should focus on evaluating the efficiency of organic Zn NPs in broiler chicken feed.

Moderate cryptoxanthin intake correlates with maintenance of a proper PINT index in a general Japanese population

ABSTRACT Introduction Small fibre neuropathy (SFN) is an early manifestation of diabetic polyneuropathy. Although oxidative stress, inflammation and change of intestinal bacterial population are assumed to be their pathogenesis, the effects of dietary nutrition have not been evaluated. The relationship between dietary nutrition intake and pain sensation was evaluated in the Japanese population. Methods We conducted the Iwaki project, a population-based study recruiting 1,028 individuals, in 2018. The relationships between the pain threshold from intraepidermal electrical stimulation (PINT) and the amount of dietary nutrition evaluated by a brief-type self-administered diet history questionnaire were examined. The odds ratio was further explored after categorizing subjects based on low (< 63.7 μg/day), intermediate (63.7-159.2 μg/day), and high cryptoxanthin levels (> 159.2 μg/day). Results Univariate linear regression analyses showed significant correlations between PINT and cryptoxanthin intake even after adjustments for other nutritional intakes (ß = 0.107, p < 0.01). Multivariate logistic regression analysis revealed low and high cryptoxanthin intake as significant risk factors for abnormal PINT (≥ 0.20 mA). Multivariate linear regression analyses showed significant correlations between PINT and cryptoxanthin intake levels after adjustment for other clinically PINT-related factors (ß = 0.09, p < 0.01). Conclusions Adequate intake of cryptoxanthin is recommended to maintain the pain threshold in the Japanese population.

Organic arabic coffee husk: Antioxidant and cytoprotective properties and potential impacts on selected human intestinal bacterial populations of individuals with diabetes

Coffee husks are the main by-product of the coffee industry and have been traditionally discarded in the environment or used as fertilizers. However, recent studies have shown that coffee husks have bioactive compounds, such as phenolics and fiber-bound macro antioxidants, offering a range of potential health benefits. This study evaluated the antioxidant capacity, cytoprotective/cytotoxic properties, and stimulatory effects on the relative abundance of selected intestinal bacterial populations of individuals with diabetes of organic coffee husks. Organic coffee husk had good antioxidant capacity, maintained under simulated gastric conditions, with more than 50% of antioxidant capacity remaining. Organic coffee husk exerted cytoprotective properties in Caco-2 cells, indicating that cellular functions were not disturbed, besides not inducing oxidation. Overall, organic coffee husk promoted positive effects on the abundance of distinct intestinal bacterial groups of individuals with diabetes during in vitro colonic fermentation, with a higher relative abundance of Bifidobacterium spp., indicating the availability of components able to reach the colon to be fermented by intestinal microbiota. Organic coffee husk could be a circular material to develop new safe and pesticide-free functional ingredients with antioxidant and potential beneficial effects on human intestinal microbiota.

Effects of Pea (Pisum sativum) Prebiotics on Intestinal Iron-Related Proteins and Microbial Populations In Vivo (Gallus gallus).

Iron deficiency remains a public health challenge globally. Prebiotics have the potential to improve iron bioavailability by modulating intestinal bacterial population, increasing SCFA production, and stimulating expression of brush border membrane (BBM) iron transport proteins among iron-deficient populations. This study intended to investigate the potential effects of soluble extracts from the cotyledon and seed coat of three pea (Pisum sativum) varieties (CDC Striker, CDC Dakota, and CDC Meadow) on the expression of BBM iron-related proteins (DCYTB and DMT1) and populations of beneficial intestinal bacteria in vivo using the Gallus gallus model by oral gavage (one day old chicks) with 1 mL of 50 mg/mL pea soluble extract solutions. The seed coat treatment groups increased the relative abundance of Bifidobacterium compared to the cotyledon treatment groups, with CDC Dakota seed coat (dark brown pigmented) recording the highest relative abundance of Bifidobacterium. In contrast, CDC Striker Cotyledon (dark-green-pigmented) significantly increased the relative abundance of Lactobacillus (p < 0.05). Subsequently, the two dark-pigmented treatment groups (CDC Striker Cotyledon and CDC Dakota seed coats) recorded the highest expression of DCYTB. Our study suggests that soluble extracts from the pea seed coat and dark-pigmented pea cotyledon may improve iron bioavailability by affecting intestinal bacterial populations.

Efficacy of a capsicum extract on growth, blood parameters and intestinal bacterial population of broilers

Context The ban on antibiotics in broiler feed has led to the need for alternative additives that can be used in poultry production, particularly in tropical climates where animals experience high levels of stress. A capsicum extract (CE), derived from chili peppers, is a potential feed additive that may enhance the performance of broiler chickens. Aims This experiment investigated the effects of CE on the growth performance, blood parameters, and intestinal bacterial population of broilers raised under high stocking density in a tropical region. Methods Four hundred and thirty-two 1-day-old Ross-308 male broiler chicks were divided into six treatments, each with six replicate pens of 12 birds. The experiment followed a randomised complete-block design. Feeding was conducted for 38 days in the following three phases on the basis of breeder recommendations: starter, grower, and finisher. CE was supplemented in the feed at concentrations of 80, 160, 240, and 320 mg/kg. All feeds were pelleted to different sizes for each phase. Growth performance was assessed at the end of each phase. Blood samples were collected to determine the complete blood count (CBC) and serum growth-hormone concentration. Chicks were euthanised using CO2 inhalation, and samples from the jejunum were collected for histological analysis. The caecocolonic contents were sampled for microbiological testing. Key results CE supplementation at concentrations of 160 and 240 mg/kg improved the performance of broilers equivalent to enramycin supplementation at 250 mg/kg, with additional benefits observed in water intake when supplemented at 240 mg/kg. The slight improvement in villous height may have contributed to the enhanced growth performance. Both the lowest (80 mg/kg) and highest (320 mg/kg) supplemented concentrations of CE slightly improved broiler performance compared with the control group. The lowest dose of CE also had a beneficial effect in reducing the E. coli:Lactobacillus ratio. Conclusions It is concluded that CE supplementation may improve performance of broilers under moderate heat-stress conditions by increasing feed and water intake with an enhanced bodyweight gain. Implications A capsicum extract can be used as an antibiotic alternative in broiler production in hot tropical environments.

Effects of Bacillus subtilis on the immune parameters, intestinal morphology and mucin gene expression in broilers exposed to Salmonella enterica challenge

The objective of this study was to evaluate the effect of the probiotic Bacillus subtilis supplementation on bacterial population, morphometry and mucin gene expression of intestine, and on immune response in Salmonella challenged broilers. Treatments were: 1) negative control (no probiotic- no challenging); 2) probiotic treated (no challenging); 3) positive control (no probiotic-salmonella challenging), and 4) Salmonella challenged chicks fed probiotic. Salmonella infection resulted in significant decrease in the relative weight of thymus. In challenged birds, dietary addition of probiotic increased (P&lt;0.0.5) the relative weights of bursa and thymus. Population of lactic acid bacteria was significantly higher in probiotic treated groups as compared to negative control or challenged chickens. Challenged chickens had the lowest count of goblet cells and those received probiotic had the highest goblet cells count. The expression of mucin 2 gene was higher in group received dietary probiotic as compared with other treatments. Difference for gene expression was not found in challenged chickens and the negative control (P&gt;0.05). In conclusion, the dietary inclusion of Bacillus subtilis could alleviate the negative effects of Salmonella infection on intestinal cells and bacterial population and could improve the growth and development of immune organs and function in infected broilers.&#x0D;

Fermented Vegetables: Health Benefits, Defects, and Current Technological Solutions.

This review summarizes current studies on fermented vegetables, analyzing the changes in nutritional components during pickling, the health benefits of fermented vegetables, and their safety concerns. Additionally, the review provides an overview of the applications of emergent non-thermal technologies for addressing these safety concerns during the production and processing of fermented vegetables. It was found that vitamin C would commonly be lost, the soluble protein would degrade into free amino acids, new nutrient compositions would be produced, and the flavor correlated with the chemical changes. These changes would be influenced by the variety/location of raw materials, the original bacterial population, starter cultures, fermentation conditions, seasoning additions, and post-fermentation processing. Consuming fermented vegetables benefits human health, including antibacterial effects, regulating intestinal bacterial populations, and promoting health (anti-cancer effects, anti-diabetes effects, and immune regulation). However, fermented vegetables have chemical and biological safety concerns, such as biogenic amines and the formation of nitrites, as well as the existence of pathogenic microorganisms. To reduce hazardous components and control the quality of fermented vegetables, unique starter cultures, high pressure, ultrasound, cold plasma, photodynamic, and other technologies can be used to solve these problems.

Unrevealing the in vitro impacts of Cereus jacamaru DC. cladodes flour on potentially probiotic strains, selected bacterial populations, and metabolic activity of human intestinal microbiota

This study investigated the potential impacts of the flour from Cereus jamacaru cactus cladodes (CJF), a cactus native to the Brazilian Caatinga biome, on the growth and metabolism of different potentially probiotic strains, as well as on the abundance of selected intestinal bacterial populations and microbial metabolic activity during in vitro colonic fermentation with a pooled human fecal inoculum. Cultivation of the probiotics in a medium with C. jamacaru cladodes flour (20 g/L) resulted in viable cell counts of up to 9.8 log CFU/mL, positive prebiotic activity scores (0.73–0.91), decreased pH and sugar contents, and increased lactic, acetic, and propionic acid production over time, indicating enhanced probiotic growth and metabolic activity. CJF overall increased the relative abundance of Lactobacillus spp./Enterococcus spp. (2.12–3.29%) and Bifidobacterium spp. (4.08–4.32%) and decreased the relative abundance of Bacteroides spp./Prevotella spp. (8.35–6.81%), Clostridium histolyticum (6.91–3.59%), and Eubacterium rectale/Clostridium coccoides (7.70–3.95%) during 48 h of an in vitro colonic fermentation using a pooled human fecal inoculum. CJF stimulated the microbial metabolic activity, with decreased pH, sugar consumption, lactic and short-chain fatty acid production, alterations in overall metabolic profiling and phenolic compound contents, and maintenance of high antioxidant capacity during colonic fermentation. These results show that CJF stimulated the growth and metabolic activity of distinct potential probiotics, increased the relative abundance of beneficial intestinal bacterial groups, and stimulated microbial metabolism during in vitro colonic fermentation. Further studies using advanced molecular technologies and in vivo experimental models could forward the investigation of the potential prebiotic properties of CJF.

Assessment of the safety of Levilactobacillus brevis CNCM I-5321, a probiotic candidate strain isolated from pulque with anti-proliferative activities.

Gut dysbiosis has been strongly correlated with colorectal cancer (CRC) development and the use of probiotics to modulate this imbalance represents a potential and promising therapy to prevent and treat CRC. For this reason, the identification of novel probiotic strains from diverse origins has widely increased in recent years, including traditional fermented foods. In this work we describe a new strain previously isolated from pulque (a traditional Mexican beverage), Levilactobacillus brevis CNCM I-5321, which may represent an interesting probiotic candidate to prevent and treat cancer. Indeed, our results show that CNCM I-5321 displays significant and specific antiproliferative capacities in human intestinal cancer cell lines (HT-29, HTC-116 and Caco-2 cells), but not in normal cells (FH cells). In addition, CNCM I-5321 is able to induce: (1) a pro-inflammatory immune response through stimulation of interleukin (IL)-2, IL-6, IL-12 and IL-17 cytokines and (2) apoptosis via activation of caspase 8. On the other hand, a minimum inhibitory concentration (MIC) assay revealed phenotypic resistance of this strain to ampicillin and chloramphenicol. However, no known transferable determinants were found in the genome of CNCM I-5321, thus this probiotic candidate presents no risk of horizontal transfer to the intestinal bacterial population. Finally, the safety status of CNCM I-5321 was evaluated using an innovative model of chicken embryo chorioallantoic membrane (CAM) to assess undesirable and/or toxic effects. Overall, our results support that CNCM I-5321 strain is non-pathogenic and safe for potential use as an anti-cancer candidate in human and animal medicine.

Chia Phenolic Extract Appear to Improve Small Intestinal Functionality, Morphology, Bacterial Populations, and Inflammation Biomarkers In Vivo (Gallus gallus).

Phenolic compounds can act as a substrate for colonic resident microbiota. Once the metabolites are absorbed and distributed throughout the body, they can have diverse effects on the gut. The objective of this study was to evaluate the effects of the intra-amniotic administration of a chia phenolic extract on intestinal inflammation, intestinal barrier, brush border membrane functionality, intestinal microbiota, and morphology in vivo (Gallus gallus model). Cornish-cross fertile broiler eggs, at 17 days of embryonic incubation, were separated into groups as follows: non-injected (NI; this group did not receive an injection); 18 MΩ H2O (H2O; injected with ultrapure water), and 10 mg/mL (1%) chia phenolic extract (CPE; injected with phenolic extract diluted in ultrapure water). Immediately after hatch (21 days), chickens were euthanized and their small intestine, cecum, and cecum content were collected and analyzed. The chia phenolic extract reduced the tumor necrosis factor-alpha (TNF-α) and increased the sucrose isomaltase (SI) gene expression, reduced the Bifidobacterium and E. coli populations, reduced the Paneth cell diameter, increased depth crypt, and maintained villus height compared to the non-injected control group. Chia phenolic extract may be a promising beneficial compound for improving intestinal health, demonstrating positive changes in intestinal inflammation, functionality, microbiota, and morphology.

The Microflora Structure in the Digestive Tract, Culture Water, and Feed of Hybrid Grouper (Epinephelus fuscoguttatus♀ × E. polyphekadion♂) Cultured in an Outdoor Pond Based on a High-Throughput Sequencing Technique

Nutrition, disease, and general wellbeing can be affected by the microbial communities associated with the digestive tracts of aquaculture species. Different sections of aquaculture species’ digestive tracts have distinctive surfaces and structures, which can change microbial communities. The present study examined the composition and distribution of bacterial species in the intestine of hybrid grouper (Epinephelus fuscoguttatus E. polyphekadion) and its aquaculture environment. Using high-throughput pyrosequencing, a 16S rRNA sequence analysis was performed on hybrid grouper foregut, midgut, and hindgut, as well as cultured water and feed. There were 610,452 sequences obtained from five components (foregut, midgut, hindgut, water, and feed). Among operational taxa (OTUs), 506 of them were detected in the foregut, 605 in the midgut, 510 in the hindgut, and 573 in aquaculture water and feed samples. A total of 113 were detected in 5 samples. A species annotation revealed that hybrid grouper intestinal tracts were dominated by Proteobacteria (67.3%–73.7%), Firmicutes (8.4%–14.0%), and Actinobacteria (6.9%–10.5%). In aquaculture culture water, Proteobacteria were predominant (36.3%), Actinobacteria (30.0%), and Planctomycetes (14.0%). Acinetobacter (1.4%–17.9%) and Photobacterium (32.0%–57.5%) dominated the intestine. Photobacterium (3.6%) and Mycobacterium (7.1%) dominated the water bacteria. The water and intestine contained five potentially pathogenic bacteria: Pseudomonas, Flavobacterium, Escherichia coli, Aeromonas bacteria, and Vibrio. The highest proportion of Vibrio was found in the water (1.7%), while Pseudomonas dominated the midgut (2.6%). Six potential probiotics were detected in the aquaculture water and intestine (Lactococcus, Streptococcus, Bdellovibrio, Lactobacillus, Bacillus, and Bacteroides). Aquaculture water and intestines contained Bacillus, Bacteroides, and Lactobacillus. According to the findings, the intestinal flora of hybrid grouper is closely correlated with its pond culture environment. Results from the study provide an experimental basis for the controlled breeding of hybrid groupers and the regulation of their microecological processes in the breeding environment deepen our understanding of the intestinal bacterial population of healthy hybrid groupers.

Effects of Intra-Amniotic Administration of the Hydrolyzed Protein of Chia (Salvia hispanica L.) and Lacticaseibacillus paracasei on Intestinal Functionality, Morphology, and Bacterial Populations, In Vivo (Gallus gallus).

As a protein source, chia contains high concentrations of bioactive peptides. Probiotics support a healthy digestive tract and immune system. Our study evaluated the effects of the intra-amniotic administration of the hydrolyzed chia protein and the probiotic Lacticaseibacillus paracasei on intestinal bacterial populations, the intestinal barrier, the inflammatory response, and brush border membrane functionality in ovo (Gallus gallus). Fertile broiler (Gallus gallus) eggs (n = 9/group) were divided into 5 groups: (NI) non-injected; (H2O) 18 MΩ H2O; (CP) 10 mg/mL hydrolyzed chia protein; (CPP) 10 mg/mL hydrolyzed chia protein + 106 colony-forming unit (CFU) L. paracasei; (P) 106 CFU L. paracasei. The intra-amniotic administration was performed on day 17 of incubation. At hatching (day 21), the animals were euthanized, and the duodenum and cecum content were collected. The probiotic downregulated the gene expression of NF-κβ, increased Lactobacillus and E. coli, and reduced Clostridium populations. The hydrolyzed chia protein downregulated the gene expression of TNF-α, increased OCLN, MUC2, and aminopeptidase, reduced Bifidobacterium, and increased Lactobacillus. The three experimental groups improved in terms of intestinal morphology. The current results suggest that the intra-amniotic administration of the hydrolyzed chia protein or a probiotic promoted positive changes in terms of the intestinal inflammation, barrier, and morphology, improving intestinal health.

Food-Grade Metal Oxide Nanoparticles Exposure Alters Intestinal Microbial Populations, Brush Border Membrane Functionality and Morphology, In Vivo (Gallus gallus).

Among food additive metal oxide nanoparticles (NP), titanium dioxide (TiO₂) and silicon dioxide (SiO₂) are commonly used as food coloring or anti-caking agents, while zinc oxide (ZnO) and iron oxide (Fe₂O₃) are added as antimicrobials and coloring agents, respectively, and can be used as micronutrient supplements. To elucidate potential perturbations associated with NP consumption on gastrointestinal health and development, this in vivo study utilized the Gallus gallus (broiler chicken) intraamniotic administration to assess the effects of physiologically relevant concentrations of food-grade metal oxide NP on brush border membrane (BBM) functionality, intestinal morphology and intestinal microbial populations in vivo. Six groups with 1 mL injection of the following treatments were utilized: non-injected, 18 MΩ DI H2O; 1.4 × 10-6 mg TiO2 NP/mL, 2.0 × 10-5 mg SiO2 NP/mL, 9.7 × 10-6 mg ZnO NP/mL, and 3.8 × 10-4 mg Fe2O3 NP/mL (n = 10 per group). Upon hatch, blood, cecum, and duodenum were collected to assess mineral (iron and zinc) metabolism, BBM functional, and pro-inflammatory-related protein gene expression, BBM morphometric analysis, and the relative abundance of intestinal microflora. Food additive NP altered mineral transporter, BBM functionality, and pro-inflammatory cytokine gene expression, affected intestinal BBM development and led to compositional shifts in intestinal bacterial populations. Our results suggest that food-grade TiO₂ and SiO₂ NP have the potential to negatively affect intestinal functionality; food-grade ZnO NP exposure effects were associated with supporting intestinal development or compensatory mechanisms due to intestinal damage, and food-grade Fe₂O₃ NP was found to be a possible option for iron fortification, though with potential alterations in intestinal functionality and health.

The Use of Unripe Banana Flour as a Functional Feed Ingredient on Blood Profile, Serum Biochemical and Intestinal Indices of Broilers

Background: The study evaluated the effect of unripe banana flour (UBF) given individually or in combination with probiotics or multienzymes on blood profile, serum biochemical parameters and intestinal indices of broilers. Methods: About 392 broiler chicks were assigned to four groups (each with seven replicates) including CONT (birds provided with control feed), UBF (birds provided with 5% UBF in feed), UBFPRO (5% UBF plus 0.05% probiotics) and UBFZYM (5% UBF plus 0.05% multienzyme). Blood and intestinal segment and digesta were collected on days 21 and 38. Results: On day 21, the mean corpuscular volume (MCV) value in UBF was lower (p &lt; 0.05) than UBFPRO. At day 38, haemoglobin levels in UBFZYM were higher (p &lt; 0.05) than in CONT and UBF. Mean corpuscular haemoglobin (MCH) and mean corpuscular haemoglobin concentration (MCHC) were higher (p &lt; 0.05) in UBFZYM than in the other groups. UBFZYM had higher (p &lt; 0.05) leukocyte counts than CONT. Uric acid levels in UBFZYM were lower (p &lt; 0.05) than in CONT on day 21. On day 38, UBFPRO chicks had lower (p = 0.05) pH values in the duodenum than other groups. UBFZYM had higher (p = 0.05) pH values in the ileum, while UBFPRO had higher (p = 0.05) pH values in the caecum than the CONT groups. Lactic acid bacteria were found in higher (p &lt; 0.05) concentrations in the ileum of UBFPRO and UBFZYM than in the CONT at day 38. The lactic acid bacteria to coliform ratio in UBFPRO and UBFZYM ileums was also higher (p = 0.07) than in CONT. On day 38, the crypt depth of the duodenum in UBFZYM was lower (p &lt; 0.05) than in CONT. The duodenum villus height to crypt depth ratio was higher (p &lt; 0.05) in UBFZYM than in the other groups. The villus height to crypt depth ratio in the UBF ileum was higher (p &lt; 0.05) than in the CONT. When compared to CONT broilers, UBF, UBFPRO, and UBFZYM broilers had higher (p &lt; 0.05) daily weight gain and feed efficiency. Conclusion: Broiler growth performance was improved when UBF was given alone or in combination with probiotics or multienzymes. The use of probiotics or multienzymes in combination with UBF improved the intestinal bacterial population, while multienzymes improved broiler physiology and intestinal morphology.

Empire Apple (Malus domestica) Juice, Pomace, and Pulp Modulate Intestinal Functionality, Morphology, and Bacterial Populations In Vivo (Gallus gallus).

Approximately $20 billion of apple sales are generated annually in the United States. With an estimated 5 million tons produced yearly in the U.S. within the last decade, apple consumption is considered ubiquitous. Apples are comprised of bioactive constituents such as phytochemicals and prebiotics that may potentiate intestinal health and the gut microbiome. This study aimed to evaluate the effects of Empire apple juice, pomace, and pulp soluble extracts on intestinal functionality, morphology, and the microbiome in vivo (Gallus gallus). There were five treatment groups: non-injected (NI); 18 MΩ H2O (H2O); 6% apple juice (AJ); 6% apple pomace (APo); 6% apple pulp (APu). The eggs were treated by intra-amniotic administration of the samples on day 17 of incubation. After hatching, the blood, tissue, and cecum samples were collected for further analyses—including duodenal histomorphology, hepatic and duodenal mRNA expression, and cecal bacterial populations. Crypt depth was significantly (p < 0.5) shortest in AJ when compared to APo and APu. APo and APu soluble extracts significantly improved villi surface area compared to NI and H2O control groups. The highest count of Paneth cells per crypt was observed in APo as compared to all groups. In addition, the expression of brush border membrane micronutrient metabolism and functional proteins varied between treatments. Lastly, Lactobacillus cecal microbial populations increased significantly in the AJ group, while AJ, APu, and APu increased the abundance of Clostridium (p < 0.5). Ultimately, these results indicate the potential of Empire apple pomace to improve host intestinal health and the gut microbiome.

Effect of Salmonella Typhimurium Colonization on Microbiota Maturation and Blood Leukocyte Populations in Broiler Chickens.

The intestinal tract of chickens is home to a diverse population of bacteria that serve as one of the first lines of defense against pathogenic microbes. Therefore, we sought to understand changes in the intestinal bacterial populations following Salmonella infection. Our results showed a clear change in the intestinal bacterial structure after Salmonella colonization. An inverse relationship between Salmonella and Lactobacillus and Escherichia was observed as well as an increase in Bacteroides. Additionally, we provide insight into the timing of the host immune response with monocytes and thrombocytes acting early following colonization followed by heterophils. In total, this data demonstrates the ability of Salmonella to change intestinal microbiota composition and highlights genera that may be useful as probiotics to fight Salmonella colonization.Reducing Salmonella in commercial chickens is vital to decreasing human salmonellosis infections resulting from contact with contaminated poultry and poultry products. As the intestinal microbiota plays an important role in preventing pathogen colonization, we sought to understand the relationship between Salmonella infection and the cecal microbiota and the host immune system. Day-of-hatch broiler chicks were assigned to three treatments: control, artificial (SA), and natural (SN) Salmonella infection. At seven days of age, control and SA birds were inoculated with PBS or Salmonella Typhimurium, respectively. Five SA birds were transferred to SN cages to facilitate natural infection. Cecal content and blood samples were collected at 0, 8, 14, and 21 days of age for microbiota and leukocyte analysis, respectively. A significant change in microbiota composition was observed in both groups as noted by a decrease in Lactobacillus and Escherichia and an increase in Bacteroides. Leukocyte analysis revealed a decrease in the percentage of circulating monocytes at 7 days post-infection while a decrease in thrombocyte and an increase in heterophil percentages were seen at 14 days post-infection. Taken together, these results demonstrate the ability of Salmonella to modulate the intestinal microbiota to facilitate colonization. Additionally, results indicated an early role of monocytes and thrombocytes during colonization, followed by heterophils.
