Microscopic observation and transcriptome analysis provide insights into mechanisms of hybrid incompatibility in Rhododendron

Hybridization across species has been widely utilized to improve agriculturally important traits in Rhododendron. These interspecific hybridizations are often inhibited by reproductive barriers at the prezygotic and postzygotic stages. Thus, in order to character the barriers and reveal the underlying regulation mechanism of interspecific hybrid incompatibility in Rhododendron, we compared the pollen germination and pollen tube growth, fruit growth, seed number and germination rate between intraspecific (Rhododendron ‘XXL’ × R. ‘XXL’) and interspecific (R. ‘XXL’ × R. delavayi) hybridizations. After pollination, the pollen germination rate and pollen tube growth rate of interspecific hybridization (Inter) were always higher than that of intraspecific hybridization (Intra) during the whole observation period. Conversely, the fruits and ovules of Intra grew faster than that of Inter after 20 DAP. The ratio of shrunken embryos of Inter was 73.27 ± 10.19 %, which was significantly higher than that of Intra with 28.41 ± 8.33 % at 40 DAP. Moreover, the fruit setting rate, the average number of seeds per fruit and seed germination rate of Intra were significantly higher than that of Inter at 150 DAP. These results indicated that the barriers of interspecific incompatibility between R. ‘XXL’ × R. delavayi could be mainly ascribed to post-fertilization isolation. Consequently, RNA-Seq analysis on ovaries at three developmental stages from two hybridizations indicated that plant hormone signal transduction, flavonoid biosynthesis, MAPK signaling pathway and plant-pathogen interaction pathways might play important roles in regulation of hybrid incompatibility between R. ‘XXL’ × R. delavayi. The results can provide a basis for the study of the hybrid incompatibility mechanism of Rhododendron, and provide useful information for the breeding and conservation of genetic resources of Rhododendron.

A pollen selection system links self and interspecific incompatibility in the Brassicaceae.

Self-incompatibility and recurrent transitions to self-compatibility have shaped the extant mating systems underlying the nonrandom mating critical for speciation in angiosperms. Linkage between self-incompatibility and speciation is illustrated by the shared pollen rejection pathway between self-incompatibility and interspecific unilateral incompatibility (UI) in the Brassicaceae. However, the pollen discrimination system that activates this shared pathway for heterospecific pollen rejection remains unknown. Here we show that Stigma UI3.1, the genetically identified stigma determinant of UI in Arabidopsis lyrata × Arabidopsis arenosa crosses, encodes the S-locus-related glycoprotein 1 (SLR1). Heterologous expression of A. lyrata or Capsella grandiflora SLR1 confers on some Arabidopsis thaliana accessions the ability to discriminate against heterospecific pollen. Acquisition of this ability also requires a functional S-locus receptor kinase (SRK), whose ligand-induced dimerization activates the self-pollen rejection pathway in the stigma. SLR1 interacts with SRK and interferes with SRK homomer formation. We propose a pollen discrimination system based on competition between basal or ligand-induced SLR1-SRK and SRK-SRK complex formation. The resulting SRK homomer levels would be sensed by the common pollen rejection pathway, allowing discrimination among conspecific self- and cross-pollen as well as heterospecific pollen. Our results establish a mechanistic link at the pollen recognition phase between self-incompatibility and interspecific incompatibility.

Dysfunction of duplicated pair rice histone acetyltransferases causes segregation distortion and an interspecific reproductive barrier

Postzygotic reproductive isolation, which results in the irreversible divergence of species, is commonly accompanied by hybrid sterility, necrosis/weakness, or lethality in the F1 or other offspring generations. Here we show that the loss of function of HWS1 and HWS2, a couple of duplicated paralogs, together confer complete interspecific incompatibility between Asian and African rice. Both of these non-Mendelian determinants encode the putative Esa1-associated factor 6 (EAF6) protein, which functions as a characteristic subunit of the histone H4 acetyltransferase complex regulating transcriptional activation via genome-wide histone modification. The proliferating tapetum and inappropriate polar nuclei arrangement cause defective pollen and seeds in F2 hybrid offspring due to the recombinant HWS1/2-mediated misregulation of vitamin (biotin and thiamine) metabolism and lipid synthesis. Evolutionary analysis of HWS1/2 suggests that this gene pair has undergone incomplete lineage sorting (ILS) and multiple gene duplication events during speciation. Our findings have not only uncovered a pair of speciation genes that control hybrid breakdown but also illustrate a passive mechanism that could be scaled up and used in the guidance and optimization of hybrid breeding applications for distant hybridization.

Cross compatibility and endogenous phytohormone profiles in interspecific hybridization between Iris tectorum and Iris germanica

In order to introduce new valuable trait to other species, the interspecific hybridization is used in ornamental breeding. The success of interspecific hybridization is hindered by incompatibility occurring at various stages of hybridization. To character the barriers of interspecific hybridization between Iris tectorum and Iris germanica cultivars, this study investigated the compatibility of pre-fertilization and post-fertilization, and profiles of endogenous phytohormones in ovary of self-pollination and interspecific hybridization. After pollination, pollen tubes germinated, grew through style, and entered ovules in both self-pollination and interspecific hybridization. Thus, the interspecific incompatibility between I. tectorum and I. germanica cultivars were not caused by pre-fertilization barrier. The observations of embryo and endosperm development revealed that incompatibility of interspecific-hybridization occurred at post-fertilization stage during the development of the embryo. The degeneration of endosperms was observed at 15 days after pollination and no embryo was observed in the interspecific hybridization. The abnormal development of embryos and endosperms indicates that reproductive barriers between I. tectorum and I. germanica cultivars happened at post-fertilization stage. To character the profiles of endogenous phytohormones in the ovary of self-pollination and interspecific hybridization, concentration of auxins, cytokinins (CKs), gibberellins (GAs), abscisic acid (ABA), Jasmonate (JA), salicylic acid (SA), and their derivatives were analyzed by UPLC-ESI-MS/MS. The results showed concentrations of indole-3-acetic acid (IAA) and bioactive free based of CKs were highest in ovary of I. tectorum self-pollination (IT-Self), which has normally developed embryos and endosperms. Four bioactive forms of GAs were identified in this study, while the patterns of their concentration changes were not constant among different materials. Besides, the concentration of ABA was highest in unfertilized ovary of I. tectorum (IT-CK). According to the profiles of endogenous phytohormones, the abnormal development of embryos in interspecific hybridization might be caused by the low concentration of IAA and CKs, proper concentration of these phytohormone could be added into in vitro culture medium to rescue embryos of interspecific hybridization. The investigation of cross compatibility and endogenous phytohormone profiles in this study will provide insight for overcoming the barrier of interspecific hybridization and embryo rescue in Iris genus.

Intra- and inter-specific reproductive barriers in the tomato clade.

Tomato (Solanum lycopersicum L.) domestication and later introduction into Europe resulted in a genetic bottleneck that reduced genetic variation. Crosses with other wild tomato species from the Lycopersicon clade can be used to increase genetic diversity and improve important agronomic traits such as stress tolerance. However, many species in the Lycopersicon clade have intraspecific and interspecific incompatibility, such as gametophytic self-incompatibility and unilateral incompatibility. In this review, we provide an overview of the known incompatibility barriers in Lycopersicon. We begin by addressing the general mechanisms self-incompatibility, as well as more specific mechanisms in the Rosaceae, Papaveraceae, and Solanaceae. Incompatibility in the Lycopersicon clade is discussed, including loss of self-incompatibility, species exhibiting only self-incompatibility and species presenting both self-compatibility and self-incompatibility. We summarize unilateral incompatibility in general and specifically in Lycopersicon, with details on the 'self-compatible x self-incompatible' rule, implications of self-incompatibility in unilateral incompatibility and self-incompatibility-independent pathways of unilateral incompatibility. Finally, we discuss advances in the understanding of compatibility barriers and their implications for tomato breeding.

Addition of Trichoderma consortia to Chilean endemic flora compost teas strongly enhances in vitro and in vivo biocontrol of phytopathogenic fungi.

To evaluate the in vitro and in vivo antifungal capability of diverse compost teas of endemic Chilean flora inoculated with a consortium of fungal strains of Trichoderma spp. (biocontrol agent) against three important phytopathogens: Botrytis cinerea, Fusarium oxysporum and Lasiodiplodia theobromae. Compost teas were obtained from endemic flora of Chile (Azara celastrina, Citronella mucronate, Cryptocarya alba, Peumus boldus and Quillaja saponaria). Eleven Trichoderma strains were isolated and antagonism tests were performed to develop fungal consortia with biocontrol properties. The biocontrol effect of compost teas inoculated with Trichoderma consortia was also analyzed. The results showed that the teas possess antifungal activity against B. cinerea and F. oxysporum and, to a lower degree against L. theobromae. In vitro tests showed that Trichoderma consortia improved the suppressive effect against B. cinerea (94%-97%), F. oxysporum (89%-92%) and L. theobromae (51%-73%). P. boldus tea showed the highest suppressive effect against the plant pathogen L. theobromae. In addition, the in vivo assay showed that tomato plants treated only with Trichoderma or compost tea, did not show differences in height with regard to control plants. However, when these two treatments were combined, the best performance in plant height and protection against pathogens was observed. This study indicates that the addition of a consortium of Trichoderma strains with intra- and interspecific incompatibilities, significantly improves the inhibitory effect of compost teas in in vitro tests against the plant pathogenic fungi, while in vivo, enhances tomato plant growth and reduce plant disease symptoms.

Ontogenes and Their Role in Cellular Construction

The genes referred to as ontogenes are responsible for conditional mutations. Based on the results of the research of conditional mutations in D. melanogaster, we attempt to figure out the biological role of ontogenes. We conclude that ontogenes in the process of individual development control the construction of the living organisms of cells (cellular construction), which comprises the induction of cell division, determination of division plane, and the location of daughter cells after the division is completed. The process of morphogenesis consists of cellular construction and protein synthesis. Protein synthesis is controlled by protein-coding (Mendelian) genes. Mendelian genes are switched on by ontogenes. In terms of the two-component genome composed of Mendelian genes and ontogenes, we consider 1) the concept of biological character; 2) interspecific incompatibility; 3) ontogenesis; 4) phylogenesis; and 5) mutagenesis. Ontogenes, which control cellular construction, possess the specific features unusual for Mendelian genes, namely, 1) the activity in germ line tissue; 2) remote interaction; and 3) activity in a compacted state (heterochromatization). These specific features of ontogenes suggest that unlike the Mendelian genes with their chemical activity, ontogenes possess another type of activity (biophysical) involving induction of an electromagnetic field.

Rapid evolution of hybrid breakdown following recent divergence with gene flow in Senecio species on Mount Etna, Sicily

How do nascent species evolve reproductive isolation during speciation with on-going gene flow? How do hybrid lineages become stabilised hybrid species? While commonly used genomic approaches provide an indirect way to identify species incompatibility factors, synthetic hybrids generated from interspecific crosses allow direct pinpointing of phenotypic traits involved in incompatibilities and the traits that are potentially adaptive in hybrid species. Here we report the analysis of phenotypic variation and hybrid breakdown in crosses between closely-related Senecio aethnensis and S. chrysanthemifolius, and their homoploid hybrid species, S. squalidus. The two former species represent a likely case of recent (<200 ky) speciation with gene flow driven by adaptation to contrasting conditions of high- and low-elevations on Mount Etna, Sicily. As these species form viable and fertile hybrids, it remains unclear whether they have started to evolve reproductive incompatibility. Our analysis represents the first study of phenotypic variation and hybrid breakdown involving multiple Senecio hybrid families. It revealed wide range of variation in multiple traits, including the traits previously unrecorded in synthetic hybrids. Leaf shape, highly distinct between S. aethnensis and S. chrysanthemifolius, was extremely variable in F2 hybrids, but more consistent in S. squalidus. Our study demonstrates that interspecific incompatibilities can evolve rapidly despite on-going gene flow between the species. Further work is necessary to understand the genetic bases of these incompatibilities and their role in speciation with gene flow.

Comparisons among populations and individuals to evaluate pollen-pistil interaction as a mechanism of reproductive interference in Taraxacum.

Reproductive interference (RI), an interspecific mating interaction that reduces the fitness of at least one of the species involved, can lead to exclusive distributions in closely related species. A hypothesis previously proposed is that RI in plants may occur by ovule usurpation, in which pistils lack interspecific incompatibility and mistakenly accept heterospecific pollen, thereby losing an opportunity for conspecific pollen fertilization. However, few comparative studies have evaluated the consistency of the inferred mechanism within and among individuals and populations. We conducted hand-pollination experiments in six populations of three native Taraxacum species that suffered from different levels of RI from an alien congener, T. officinale, and compared pollen-pistil interactions among populations. We also investigated the interactions for eight individual T. japonicum plants whose response to heterospecific pollen deposition had been previously measured. Our results revealed that pollen tubes often penetrated native ovaries following heterospecific pollination in populations suffering from strong RI, whereas they seldom did in populations suffering from marginal RI. However, the relative frequency of the pollen tube penetration was not significantly related to the strength of alien RI. Not all pistils on an individual plant showed the same pollen receptivity following heterospecific pollination; rather, some accepted and some refused the pollen tubes. The relationship between pollen tube penetration following heterospecific pollination and the strength of the alien RI was also not significant among individuals. Our present results generally support the ovule usurpation hypothesis, but suggest that other factors, such as competition for pollinator services, variation in the effects of heterospecific pollen donors, and condition of the native inflorescences, might also affect the observed RI strength.

Rapid Macrosatellite Evolution Promotes X-Linked Hybrid Male Sterility in a Feline Interspecies Cross.

The sterility or inviability of hybrid offspring produced from an interspecific mating result from incompatibilities between parental genotypes that are thought to result from divergence of loci involved in epistatic interactions. However, attributes contributing to the rapid evolution of these regions also complicates their assembly, thus discovery of candidate hybrid sterility loci is difficult and has been restricted to a small number of model systems. Here we reported rapid interspecific divergence at the DXZ4 macrosatellite locus in an interspecific cross between two closely related mammalian species: the domestic cat (Felis silvestris catus) and the Jungle cat (Felis chaus). DXZ4 is an interesting candidate due to its structural complexity, copy number variability, and described role in the critical yet complex biological process of X-chromosome inactivation. However, the full structure of DXZ4 was absent or incomplete in nearly every available mammalian genome assembly given its repetitive complexity. We compared highly continuous genomes for three cat species, each containing a complete DXZ4 locus, and discovered that the felid DXZ4 locus differs substantially from the human ortholog, and that it varies in copy number between cat species. Additionally, we reported expression, methylation, and structural conformation profiles of DXZ4 and the X chromosome during stages of spermatogenesis that have been previously associated with hybrid male sterility. Collectively, these findings suggest a new role for DXZ4 in male meiosis and a mechanism for feline interspecific incompatibility through rapid satellite divergence.

Identification and chromosome doubling of Fragaria mandshurica and F. nilgerrensis

Genetic factors that control various excellent agronomic traits are present in wild strawberries; however, barriers such as ploidy differences and interspecific incompatibility prevent the transmission of these genetic factors to cultivated strawberries. Using the colchicine method to induce doubling, the autotetraploids of Fragaria mandshurica and F. nilgerrensis were obtained, and the biological characteristics of the autotetraploids and the compatibility status distribution in different cross-pollination combinations relating to the two species were further examined. The autotetraploids showed thicker and stronger morphological characteristics than those of their corresponding diploids, especially the F. mandshurica species. The change in ploidy can cause the appearance of new excellent agronomic traits: the autotetraploid of F. nilgerrensis is more resistant to anthracnose disease, caused by Colletotrichum gloeosporioides, than the diploid, and the flowering period is significantly longer, with an increase in flower quantity. There is unilateral incompatibility between the two self-compatible species. The autotetraploids are similar to the corresponding diploid in pollination compatibility; however, due to the decline in pollen viability, the related hybridisation combinations show slightly lower compatibility. The closer the ploidy is, the easier it is to obtain hybrid offspring; the tetraploids of both species crossed with cultivated strawberries, showing possible increases in compatibility after chromosome doubling.

Strategies to combat the problem of yam anthracnose disease: Status and prospects

Yam (Dioscorea spp.) anthracnose, caused by Colletotrichum alatae, is the most devastating fungal disease of yam in West Africa, leading to 50%–90% of tuber yield losses in severe cases. In some instances, plants die without producing any tubers or each shoot may produce several small tubers before it dies if the disease strikes early. C. alatae affects all parts of the yam plant at all stages of development, including leaves, stems, tubers, and seeds of yams, and it is highly prevalent in the yam belt region and other yam‐producing countries in the world. Traditional methods adopted by farmers to control the disease have not been very successful. Fungicides have also failed to provide long‐lasting control. Although conventional breeding and genomics‐assisted breeding have been used to develop some level of resistance to anthracnose in Dioscorea alata, the appearance of new and more virulent strains makes the development of improved varieties with broad‐spectrum and durable resistance critical. These shortcomings, coupled with interspecific incompatibility, dioecy, polyploidy, poor flowering, and the long breeding cycle of the crop, have prompted researchers to explore biotechnological techniques to complement conventional breeding to speed up crop improvement. Modern biotechnological tools have the potential of producing fungus‐resistant cultivars, thereby bypassing the natural bottlenecks of traditional breeding. This article reviews the existing biotechnological strategies and proposes several approaches that could be adopted to develop anthracnose‐resistant yam varieties for improved food security in West Africa.

Restoring Fertility for Novel Interspecific Hybrids between Kalanchoe garambiensis and K. nyikae Using Colchicine Treatment.

Interspecific hybridization is an effective strategy in Kalanchoe breeding programs for the introduction of new traits. Wild species within the Kalanchoe genus are valuable genetic resources for providing new horticulture traits and to improve environmental adaptations. However, reproductive barriers associated with fertilization and hybrid sterility must be overcome to produce fertile hybrid progenies. To approach the breeding objectives for Kalanchoe cut flower cultivars with long stem traits and adaptation to tropical/subtropical regions, a tropical species endemic to Taiwan, Kalanchoe garambiensis Kudo, was used as a parent to cross with other long stem Kalanchoe species. Reciprocal crossing was effective in overcoming interspecific unilateral incompatibility in our crossed pairs. One superior hybrid, ‘103-1’, produced capsules without seeds by selfing and backcrossing with pollens from either parent. Other than the seedless trait, failure of pollen releasing from anther, pollen aggregation and no pollen germination in ‘103-1’ suggested its F1 sterility. Colchicine treatments on apical buds of ‘103-1’ successfully overcame potential meiotic abnormalities by doubling ploidy. For the first time, fertile interspecific hybrids of K. garambiensis and K. nyikae Engler were generated. The fertile hybrid has further produced progeny populations by crossing with K. nyikae or K. blossfeldiana von Poelln, ‘Ida’.

The caffeoyl-CoA O-methyltransferase gene SNP replacement in Russet Burbank potato variety enhances late blight resistance through cell wall reinforcement.

Metabolic pathway gene editing in tetraploid potato enhanced resistance to late blight. Multiallelic mutation correction of a caffeoyl-CoA O-methyltransferase gene increased accumulation of resistance metabolites in Russet Burbank potato. Late blight of potato is a devastating disease worldwide and requires weekly applications of fungicides to manage. Genetic improvement is the best option, but the self-incompatibility and inter-specific incompatibility makes potato breeding very challenging. Immune receptor gene stacking has increased resistance, but its durability is limited. Quantitative resistance is durable, and it mainly involves secondary cell wall thickening due to several metabolites and their conjugates. Deleterious mutations in biosynthetic genes can hinder resistance metabolite biosynthesis. Here a probable resistance role of the StCCoAOMT gene was first confirmed by an in-planta transient overexpression of the functional StCCoAOMT allele in late blight susceptible Russet Burbank (RB) genotype. Following this, a precise single nucleotide polymorphism (SNP) mutation correction of the StCCoAOMT gene in RB potato was carried out using CRISPR-Cas9 mediated homology directed repair (HDR). The StCCoAOMT gene editing increased the transcript abundance of downstream biosynthetic resistance genes. Following pathogen inoculation, several phenylpropanoid pathway genes were highly expressed in the edited RB plants, as compared to the non-edited. The disease severity (fold change = 3.76) and pathogen biomass in inoculated stems of gene-edited RB significantly reduced (FC = 21.14), relative to non-edited control. The metabolic profiling revealed a significant increase in the accumulation of resistance-related metabolites in StCCoAOMT edited RB plants. Most of these metabolites are involved in suberization and lignification. The StCCoAOMT gene, if mutated, can be edited in other potato cultivars to enhance resistance to late blight, provided it is associated with other functional genes in the metabolic pathway network.

Asymmetric Fitness of Second-Generation Interspecific Hybrids Between Ciona robusta and Ciona intestinalis.

Reproductive isolation is central to speciation, but interspecific crosses between two closely related species can produce viable and fertile hybrids. Two different species of tunicates in the same ascidian genus, Ciona robusta and Ciona intestinalis, can produce hybrids. However, wild sympatric populations display limited gene flow, suggesting the existence of obstacles to interspecific reproduction that remain unknown. Here, we took advantage of a closed culture system to cross C. robusta with C. intestinalis and established F1 and F2 hybrids. We monitored post-embryonic development, survival, and sexual maturation to characterize the genetic basis of simple traits, and further probe the physiological mechanisms underlying reproductive isolation. Partial viability of first and second generation hybrids suggested that both pre- and postzygotic mechanisms contributed to genomic incompatibilities in hybrids. We observed asymmetric fitness, whereby the C. intestinalis maternal lines fared more poorly in our system, pointing to maternal origins of species-specific sensitivity. We discuss the possibility that asymmetrical second generation inviability and infertility emerge from interspecific incompatibilities between the nuclear and mitochondrial genomes, or other maternal effect genes. This work paves the way to quantitative genetic approaches to study the mechanisms underlying genomic incompatibilities and other complex traits in the genome-enabled Ciona model.

Geography is essential for reproductive isolation between florally diversified morning glory species from Amazon canga savannahs

The variety, relative importance and eco-evolutionary stability of reproductive barriers are critical to understanding the processes of speciation and species persistence. Here we evaluated the strength of the biotic prezygotic and postzygotic isolation barriers between closely related morning glory species from Amazon canga savannahs. The flower geometry and flower visitor assemblage analyses supported pollination by the bees in lavender-flowered Ipomoea marabaensis and recruitment of hummingbirds as pollinators in red-flowered Ipomoea cavalcantei. Nevertheless, native bee species and alien honeybees foraged on flowers of both species. Real-time interspecific hybridization underscored functionality of the overlap in flower visitor assemblages, questioning the strength of prezygotic isolation underpinned by diversification in flower colour and geometry. Interspecific hybrids were fertile and produced offspring in nature. No significant asymmetry in interspecific hybridization and hybrid incompatibilities among offspring were found, indicating weak postmating and postzygotic isolation. The results suggested that despite floral diversification, the insular-type geographic isolation remains a major barrier to gene flow. Findings set a framework for the future analysis of contemporary evolution of plant-pollinator networks at the population, community, and ecosystem levels in tropical ecosystems that are known to be distinct from the more familiar temperate climate models.

The stigma has tightly regulated recognition mechanisms at several levels to prevent unwanted pollen from achieving fertilization. Knowledge about barriers controlling interspecific incompatibility is scarce. New evidence reveals a novel gene involved in regulating interspecies incompatibility in self-compatible Arabidopsis thaliana.

A comparison of reproductive isolation between two closely related oak species in zones of recent and ancient secondary contact

BackgroundMuch of the debate over the evolutionary consequences of hybridization on genetic divergence and speciation results from the breakdown or reinforcement of reproductive barriers in secondary hybrid zones. Among hybrid populations established for different lengths of time following secondary contact, stronger reproductive barriers are generally expected to occur in zones with longer contact. However, in plants no detailed investigation of recent and ancient zones of secondary contact has been conducted despite the importance of such a comparative study. Here, we compare pre- and postzygotic reproductive barriers between two closely related oak species, Quercus mongolica and Q. liaotungensis, in such a situation.ResultsThe recorded flowering times of both species overlapped in both contact zones. The fruit set at 10 and 30 days after interspecific hand pollination was not significantly lower than that after intraspecific pollination whenever Q. mongolica or Q. liaotungensis comprised the maternal parents in both populations. These results indicated that neither prezygotic phenological barriers nor interspecific incompatibility could have resulted in the reproductive isolation between the two species in both hybrid zones. However, the proportion of hybrid seeds produced by both species in the ancient zone was significantly lower than that recorded in the recent zone of secondary contact. In addition, the proportion of hybrid seeds simulated to form, assuming both random mating and an absence of postpollination barriers, was significantly higher than that detected in the ancient contact zone but not in the recent contact zone. These results suggest stronger early-acting postzygotic isolation between the two oak species in the ancient relative to the recent contact zone.ConclusionsOur comparative study demonstrated that postzygotic barriers during seed maturity were the main contributing factor to total reproductive isolation, particularly in the ancient contact zone, which aided species delimitation. In the recently formed secondary contact zone, pre- and postzygotic barriers were not well developed, and a high frequency of natural hybridization was evident. To our knowledge this study provides the first comparison of reproductive isolation between the ancient and recent secondary contact zones in plants and helps to clarify the evolutionary consequences of hybridization in a temporal context.

Correlated evolution of self and interspecific incompatibility across the range of a Texas wildflower.

Selection to prevent interspecific mating can cause an increase or a decrease in self-pollination in sympatric populations. Characterizing the geographical variation in self and interspecific incompatibilities within a species can reveal if and how the evolution of self and interspecific mate choice are linked. We used controlled pollinations to characterize the variation in self and interspecific incompatibility across 29 populations of Phlox drummondii. We evaluated seed set from these pollinations and described the developmental timing of variation in pollen-pistil compatibility. There is extensive quantitative variation in self-incompatibility and interspecific-incompatibility with its close congener P.cuspidata. Phlox drummondii populations that co-occur and hybridize with P.cuspidata have significantly higher interspecific incompatibility and self-incompatibility than geographically isolated P.drummondii populations. The strength of self and interspecific incompatibility is significantly correlated among individuals and the strength of both incompatibilities is explained by the success of pollen adhesion to the stigma. The correlated strength of self and interspecific incompatibility across the range of P.drummondii and the concurrent developmental timing of the pollen-pistil interaction, suggests these incompatibilities have an overlapping molecular mechanism. The geographical distribution of variation in incompatibilities indicates that this mechanistic link between incompatibilities may affect the evolution of mate choice in plants.

Introgression of Gossypium barbadense L. into Upland cotton germplasm RMBUP-C4S1

Gossypium barbadense L. cotton has significantly better fiber quality than Upland cotton (G. hirsutum L.); however, yield and environmental adaptation of G. barbadense is not as wide as Upland. Most cotton in the world is planted to Upland cultivars. Many attempts have been made, over a considerable number of years, to introgress fiber quality alleles from G. barbadense into Upland. However, introgression barriers, primarily in the form of interspecific incompatibility, have limited these traditional approaches. The use of chromosome substitution lines (CSL) as a bridge should provide a more efficient way to introgress alleles from G. barbadense into Upland. We crossed 18 G. barbadense CSL to three cultivars and developed a random mated population. After five cycles of random mating followed by one generation of self-pollination to increase the seed supply, we grew the random mated population and used 139 G. barbadense chromosome specific SSR markers to assess a random sample of 96 plants for introgression. We recovered 121 of 139 marker loci among the 96 plants. The distribution of the G. barbadense alleles ranged from 10 to 28 alleles in each plant. Among the 96 plants we found individual plants with marker loci from 6 to 14 chromosomes or chromosome arms. Identity by descent showed little relatedness among plants and no population structure was indicated by a heat map. Using CSL we were able to develop a mostly Upland random mated population with considerable introgression of G. barbadense alleles which should be useful for breeding.
