Estimands for clinical endpoints in tuberculosis treatment randomized controlled trials: a retrospective application in a completed trial

BackgroundRandomized trials for the treatment of tuberculosis (TB) rely on a composite primary outcome to capture unfavorable treatment responses. However, variability between trials in the outcome definition and estimation methods complicates across-trial comparisons and hinders the advancement of treatment guidelines. The International Council for Harmonization (ICH) provides international regulatory standards for clinical trials. The estimand framework outlined in the recent ICH E9(R1) addendum offers a timely opportunity for randomized trials of TB treatment to adopt broadly standardized outcome definitions and analytic approaches. We previously proposed and defined four estimands for use in this context. Our objective was to evaluate how the use of these estimands and choice of estimation method impacts results and interpretation of a large phase III TB trial.MethodsWe reanalyzed participant-level data from the REMoxTB trial. We applied four estimands and various methods of estimation to assess non-inferiority of both novel 4-month treatment regimens against standard of care.ResultsWith each of the four estimands, we reached the same conclusion as the original trial analysis that the novel regimens were not non-inferior to standard of care. Each estimand and method of estimation gave similar estimates of the treatment effect with fluctuations in variance and differences driven by the methods applied for handling intercurrent events.ConclusionsOur application of estimands defined by the ICH E9 (R1) addendum offers a formalized framework for addressing the primary TB treatment trial objective and can promote uniformity in future trials by limiting heterogeneity in trial outcome definitions. We demonstrated the utility of our proposal using data from the REMoxTB randomized trial. We outlined methods for estimating each estimand and found consistent conclusions across estimands. We recommend future late-phase TB treatment trials to implement some or all of our estimands to promote rigorous outcome definitions and reduce variability between trials.Trial registrationClinicalTrials.gov NCT00864383. Financial market regulation and private law: a new frontier of transnational commercial law

Abstract Global financial markets are among the most extensively regulated markets in the world economy. This poses significant challenges from a private law perspective when regulatory rules interfere with private rights and obligations. This article examines the potential of transnational commercial law (TCL) to provide for a private law framework suited to the regulatory reality in global financial markets. The article finds that, while regulatory aspects are generally excluded from TCL instruments, notable exceptions to this general rule occur. Based on an analysis of the approaches undertaken in the Unidroit Convention of Substantive Rules for Intermediated Securities (Geneva Convention), including the accompanying Legislative Guide, the Unidroit Principles on the Operation of Close-Out Netting Provisions and the UNCITRAL Model Law on Secured Transactions, the article maps out three methods that allow for a reconciliation of private and regulatory rules: (i) the adoption of a flexible private law system including default and conflict rules; (ii) the use of opening clauses to allow deviations from TCL rules in accordance with international regulatory standards; and (iii) a codificatory approach through the development of a comprehensive and systematic legal framework consisting of both private law and regulatory rules. Finally, the article addresses potential obstacles and objections that future TCL instruments will face when addressing regulatory aspects. Overall, the article argues that an efficient financial market requires a foreseeable set of private law rules that—considering the continuous expansion of regulatory law—must address the pressing issues deriving from the interrelation of regulation and private law. However, the adaption of TCL instruments should be limited to common-sense financial market regulation that, even if differing from State to State, generally follows globally shared regulatory goals.

Effect of phthalate-free plasticizer addition on thermal, mechanical, and aging properties of nitrile rubber (NBR)

Nitrile rubber (NBR) is widely used in the automotive, aerospace, and chemical sectors due to its resistance to fuels, oil, thermal aging, and fatigue. A typical NBR compound is prepared with several components that are added to obtain a product to meet specific industry requirements. Plasticizer is one of the main components used in NBR compounds. Typically, phthalates like DBP (dibutyl phthalate) or DOP (dioctyl phthalate) are used for NBR plasticization. However, these phthalates-based plasticizers are associated with toxicity and often prohibited by international regulatory standards. In this way, in the pursuit of a non-toxic NBR compound, the use of phthalate-free plasticizer, namely dioctyl terephthalate (DOTP), has been proposed. In this work, NBR compounds with DOTP, DBP, or DOP were processed in an open mill and vulcanized in hydraulic press. The unvulcanized samples were tested in a moving die rheometer (MDR). Subsequently, thermal, mechanical, and aging analyses were conducted for the vulcanized samples. The lowest torque observed in the rheometric curve, the displacement value of glass transition temperature (Tg), the smaller Shore A hardness, and the high elongation at break indicate that the addition of DOTP results in higher plasticization. The better-aging properties indicate lower migration of the DOTP plasticizer. Thus, the use of DOTP as a plasticizer holds promise as a viable alternative to replace the phthalates-based plasticizer in NBR compounds.

Assets of foreignness in a regulated industry

Purpose Foreign subsidiaries incur substantial institutional conformity costs because they have to respond to host-country institutional pressures (Slangen & Hennart, 2008). The purpose of this paper is to study this type of cost from institutional and regulatory perspectives. The authors argue that these costs decrease when the host country adopts concepts of international regulations that multinationals may be familiar with due to their own home country regulation experience. This prior regulatory experience gives foreign subsidiaries an advantage of foreignness (AoF), which can offset their liability of foreignness (LoF). Design/methodology/approach This study compared the returns on assets of 35 domestic firms with those of foreign subsidiaries in the Brazilian energy industry between 2002 and 2021, using regression dynamic panel data. Findings The existence of a relationship between the international regulatory norm and the Brazilian regulator has transformed the LoF into an advantage of foreignness to compete with local energy firms. The results also suggest that the better the regulatory quality of the subsidiary’s country of origin, the better its performance in Brazil, as it can reduce compliance costs. Finally, the greater the psychic distance between Brazil and the foreign subsidiary’s home country, the worse its performance. Second, prior regulatory experience of multinational enterprise in similar environment can affect its foreign affiliate performance (Perkins, 2014). Third, this study confirms current literature that argues that knowledge and ability to operate in an institutionalized country can be transferred from parent to affiliate. In the end, this study investigates whether AoF persists when host governments improve the governance of their industries.

Regulatory “Reliance” in Global Trade Governance

AbstractOne of the most significant recent trends in global trade governance has been the increasing use of regulatory “reliance” arrangements as a significant element of trade alliances. Against this backdrop, an important set of questions are raised about how existing institutions of global trade governance – especially the World Trade Organization and international regulatory standards organisations – should respond. To what extent, and how, should such institutions facilitate reliance arrangements? And what role can they usefully play in overseeing and guiding their use? This paper begins to answer these questions through a focused case study of regulatory reliance in the agrifood sector. Four challenges are identified regarding the implementation of such arrangements: the high costs of establishment and maintenance; the lack of agreed and reliable assessment methodologies; the potential for arbitrary discrimination between trade partners; and the difficulties of dealing with regulatory change over time. In light of these challenges, the paper assesses the work of existing international organisations in governing reliance arrangements in the agrifood sector. The paper concludes with a number of preliminary suggestions as to how this architecture of global governance might be supplemented or harnessed to address some of the challenges posed by reliance arrangements.

A dynamic framework to align company climate reporting and action with global climate targets

AbstractThere are global aspirations to reach net zero emissions, which triggered the development of standards, guidance and tools to measure and manage climate action across countries, sectors and companies. However, carbon accounting inaccuracies, ambiguous emissions disclosures and unambitious climate targets are hampering these aspirations. This paper reports on the disparity between high‐level guidance and practical implementation of carbon accounting and reporting at company level. By conducting a systematic literature analysis and focussing on the English Water sector, we utilise case study data from Northumbrian Water Limited (NWL), to identify limitations in the current guidance and frameworks. The results indicate the necessity for enhanced alignment in high‐level guidance, particularly regarding the Greenhouse Gas Protocol and the Science‐Based Target Initiative. It is evident that clarity and consistency from high‐level resources are essential for climate mitigation. This paper shows, depending on the sector and company types, that different benefits are gained from using the available resources. We propose a carbon measurement and management process for the English Water sector (and beyond) to reach net zero targets and make recommendations for decision makers. This helps to understand best practices of carbon accounting and reporting and to make effective investment decisions. Consequently, we advocate for policy interventions to improve the standardisation of carbon accounting models. Harmonising international regulatory frameworks and standards is needed, which will empower organisations to effectively assess, manage and reduce their carbon footprints.

Why are international standards not set? Explaining “weak” cases in shadow banking regulation

AbstractWhy are international regulatory standards not set? While most of the literature focuses on explaining positive cases of standard-setting where international rules are agreed upon, weak or negative cases remain prevalent and yet surprisingly under-explored. To explain these cases in the area of financial services, we integrate an inter-state explanation, which focuses on competition between major jurisdictions, with a transgovernmental explanation, which relates to conflict between different regulatory bodies at the international level. We also consider how these dimensions interact with financial industry lobbying. This allows us to construct a typology differentiating between distinct types of cases concerning international standard-setting: (1) absent standards, (2) non-agreed standards, (3) symbolic standards, and (4) agreed standards. The explanatory leverage of our approach is illustrated through a systematic structured focused comparison of four post-crisis cases related to “shadow banking.” The article generates novel insights into regulatory conflicts and the scope conditions for international agreement.

EVALUASI PADA WEBSITE SRAGENKAB.GO.ID MENGGUNAKAN METODE WEB USABILITY EVALUATION (WEBUSE) DAN WEB CONTENT ACCESSIBILITY GUIDELINES (WCAG) 2.1

The sragenkab.go.id website serves as a vital platform supporting the operations of the Sragen district government, disseminating accurate and prompt information to the public. However, various issues hamper its functionality, such as interface display problems, unclear layout, non-functional links, excessively simple design, and slow response times. To address these concerns, this study aims to evaluate the sragenkab.go.id website's usability and accessibility aspects using the Website Usability Evaluation (WEBUSE) method and the Web Content Accessibility Guideline (WCAG) 2.1 method, respectively. Using the WEBUSE method, a survey questionnaire was distributed to 100 respondents from Sragen to assess the usability level of the website. The obtained score of 0.65 indicated that the website's usability is categorized as "good" and has been accepted by the users. On the other hand, the accessibility evaluation using the WCAG 2.1 method, with the assistance of the WAVE tool and accessibilitychecker, revealed a score below 75%, indicating a significant risk of non-compliance with international accessibility standards. In conclusion, the sragenkab.go.id website exhibits commendable usability; however, it falls short in terms of accessibility. The findings emphasize the importance of optimizing the website's accessibility to adhere to international regulatory standards, ensuring equitable access to information and services for all users. Future improvement efforts should focus on rectifying accessibility issues to enhance the overall user experience and inclusivity of the website.

High acidity of bracatinga honeydew honey: A new regulatory limit proposal

The free acidity of bracatinga honeydew honey (BHH) was monthly monitored over short-term storage (four months) until all the samples exceeded 50 mEq kg−1 – the maximum value allowed by the international regulatory honey standards. In addition, BHH quality was also investigated through moisture content, water activity, electrical conductivity, pH, 5-hydroxymethylfurfural, and aliphatic organic acids (AOA) analyses. According to our results, most of the parameters investigated presented significant differences during the short storage period studied; however, the quality parameters (except acidity) did not exceed the limits established by the international regulatory honey standards. Therefore, the high free acidity observed in the BHH samples did not affect its quality. Moreover, the total AOA concentration decreased as the free acidity increased, indicating that the high acidity is not related to postharvest fermentation. Since all BHH samples exceeded the established limit of 50 mEq kg−1 after four months of storage (up to 62.7 mEq kg−1), this data corroborates that this type of honey does not comply with the regulatory honey standards, which represents an obstacle to its commercialization. Therefore, our data reinforce the need for a future reassessment of the international regulatory honey standards regarding the free acidity limit for BHH. In this sense, taking together all the studies developed by our research group since 2014, a new free acidity value of 65 mEq kg−1 is proposed, which may discourage fraud practices and negative impacts on the BHH beekeeping chain.

Cardiotoxic and neurobehavioral effects of sucralose and acesulfame in Daphnia: Toward understanding ecological impacts of artificial sweeteners

Artificial sweeteners are widely used in food and pharmaceuticals, but their stability and persistence raise concerns about their impact on aquatic life. Although standard toxicity tests do not reveal lethal effects, recent studies suggest a potential neurotoxic mode of action. Using environmentally relevant concentrations, we assessed the effects of sucralose and acesulfame, common sugar substitutes, on Daphnia magna focusing on biochemical (acetylcholinesterase activity; AChE), physiological (heart rate), and behavioural (swimming) endpoints. We found dose-dependent increases in AChE and inhibitory effects on heart rate and behaviour for both substances. Moreover, acesulfame induced a biphasic response in AChE activity, inhibiting it at lower concentrations and stimulating at higher ones. For all endpoints, the EC50 values were lower for acesulfame than for sucralose. Additionally, the relationship between acetylcholinesterase and heart rate differed depending on the substance, suggesting possible differences in the mode of action between sucralose and acesulfame. All observed EC50 values were at μg/l levels, i.e., within the levels reported for wastewater, with adverse effects observed at as low as 0.1 μg acesulfame /l. Our findings emphasise the need to re-evaluate risk assessment thresholds for artificial sweeteners and provide evidence for the neurotoxic effects of artificial sweeteners in the environment, informing international regulatory standards.

Public health implementation of pathogen genomics: the role for accreditation and application of ISO standards.

Pathogen genomics has transitioned rapidly from the research setting into a powerful tool now routinely used in public health microbiology, for surveillance, outbreak investigations and disease control. As these investigations can have significant public health, treatment and legal impacts, we must ensure the accuracy of these results through validation of testing processes. For laboratories working in this space, it is important to approach this work with a quality and accreditation framework in mind, working towards implementation of quality systems and test validation that meet international regulatory standards. Here we outline the key international standards and processes that lead toward accreditation for pathogen genomics.

Development and validation of HPLC-MS/MS method for simultaneous analysis of B vitamins present naturally or after fortification in fruit juices

In this study, a multivitamin analysis method with a run time of 20 min was developed for nine major B vitamins in fruit juices using HPLC coupled with a triple quadrupole mass spectrometer. A reverse phase column with ammonium formate buffers in methanol and water as mobile phases under gradient elution mode were used to accomplish chromatographic separation. A simple dilute and shoot method was used for sample preparation. A calibration and accuracy study for method validation was conducted. The accuracy profiles were created using measurement uncertainty estimated from the validation data according to the ISO/DTS 21748. The proportional bias due to the matrix effect were ranging between 80% and 115% in the matrices investigated in this study. The measurement uncertainty and the corresponding coefficient of variation were within the acceptable limits defined in the literature and international regulatory standards i.e. EU regulation. The limits of quantitation were low enough to prove fitness for purpose for all vitamins except cyanocobalamin. The study concludes with a method with the potential to be used in quality control laboratories for high throughput routine vitamin analysis.

Characterization of MSW incineration bottom ash for use as structural fill in reinforced soil structures: Geoenvironmental, geotechnical and economical assessment

The study presents the geoenvironmental and geotechnical characterization of MSW incineration bottom ash (IBA) and examines its reuse as structural fill in reinforced soil structures (RSS).The suitability of reuse has been assessed with regard to international regulatory standards. The prime focus of the work remains on evaluating the pullout response of geosynthetic reinforcements through IBA fill to determine the interaction coefficient, which has never been addressed in the literature. The economic viability of using IBA instead of locally available river sand for a 12 m high MSE wall is also established.The column leaching test results confirm that IBA can be utilized in RSS with suitable design measures. The geotechnical investigation shows that IBA is a well-graded, non-plastic lightweight material with adequate drainage and high shear strength. The pullout test results demonstrate that the interaction coefficient of polymeric strips and geogrid in IBA (0.73–1.53 and 0.79–1.91, respectively) is comparable or higher to materials conventionally used as structural fill in RSS, indicating adequate bondage between IBA and geosynthetic reinforcement. Further, it is estimated that using IBA as a substitute for available river sand in the vicinity can potentially reduce the overall RSS project cost by 15–20%, even if IBA has to be transported 50 km away from the project site.

Biomimicry: How the environment serves as a guide for technological advancements

AbstractMultiple ecological tactics, like the ones described here, can be imitated to strongly affect these two components in the construction sector. This article provides an in‐depth examination of the literature on biomimicry‐based strategies intended to enhance building designs. This analysis is motivated by the rising need for energy legislation to achieve future local goals and provide a framework for use in Panama. These biomimicry‐based methods have undergone more review and evaluation, and the incorporation of organism‐based design for three of Panama's climate types has been proposed. Therefore, a SWOT analysis assisted in realizing the full potential that biomimicry‐based techniques might have in raising the likelihood of satisfying local and international regulatory standards. In order to design constructions using biomimicry, multidisciplinary collaboration is required. This gives a competitive edge for better‐trained human resources and expands horizons in the conventional construction industry. Finally, the analysis provided here might act as the preliminary step for the additional technical assessment using numerical and experimental procedures.

Characterisation of soil-like material restored from landfill mining activities in Indian cities

ABSTRACT This paper demonstrates the characteristics of soil-like material extracted from the landfill mining activity at the Ariyamangalam dump yard, located in Tiruchirappalli city of southern India. The landfill mining waste was collected and characterised for contaminant potential, thereby its suitability for reuse applications offsite. Soil-like materials of size fractions finer than 2 mm have been assessed for their physico-chemical characteristics and compared the acceptability criterion with the drinking standard IS 10500: 2012. Total soluble solids and closely associated parameters have been determined in the water extract from soil-like materials and local soils and compared with the international regulatory standards for reuse acceptability. From the total heavy metal concentration of soil-like material and local soil, three different pollution indices, namely contamination ratio, geo-accumulation index, and enrichment factor, have been identified and categorised soil-like material as low contaminated for lead; low to moderately contaminated for chromium, nickel, and iron; moderately contaminated for manganese; and moderately to strongly contaminated for copper and zinc. The organic and moisture content of soil-like material was reported as 22 and 19.6%, whereas the local soil showed 2.4 and 1.5%, respectively. Due to its enormous organic content, soil-like materials pose as less efficient in utilising offsite applications without primary treatment.

Quality assessment and chemometrics application on physicochemical characteristics, antioxidant properties, and 5‐HMF content of Malaysian stingless bee honey from different topographical origins

The popularity of Malaysian stingless bee honey is rising among health-conscious individuals; thus, chemical and physical evaluations of Malaysian stingless bee honey are vital to ensure the honey has achieved the optimum limits set by Malaysian and international regulatory standards so that it can be commercialized locally and internationally. Therefore, in the present study, the physicochemical characteristics (moisture content, total dissolved solids, pH, free acidity, electrical conductivity, and ash content), antioxidant properties (total phenolic and flavonoid contents), and 5-hydroxymethylfurfural (5-HMF) of Heterotrigona itama (H. itama) honey from different sites in Peninsular Malaysia were investigated. Subsequently, the correlation between these chemical and physical parameters was studied using Spearman correlation coefficients. The significant difference between H. itama honey from different topographical origins was studied using univariate analysis (one-way ANOVA followed by post hoc Tukey's test). The discrimination pattern of 45 honey samples based on their topographical origins was evaluated using cluster analysis (heatmap and dendrogram) and chemometrics analysis (partial least squares-discriminant analysis). Results showed that some samples of certain parameters (electrical conductivity, free acidity, and moisture content) have exceeded the limit set by the international regulatory standard. However, the 5-HMF content of all samples was within the allowed range. A statistically significant difference (p<0.05) has been observed for all the parameters except electrical conductivity and ash content in terms of inter-topographical origins. Although the profiles of H. itama honey from different origins were close, most of them were separated according to their topographical origins and were validated using a permutation test.

CRM-SYSTEMS IN MARKETING RISKS MANAGEMENT OF BANK`S RISK PROTECTION PROJECTS

The article examines the theoretical, methodological and practical aspects of the formation of information and analytical support for marketing risk management of risk protection projects of a commercial bank in the conditions of modern challenges of the uncertainty of the marketing environment and global digital transformations. Requirements for an effective bank risk management system based on modern technological tools and innovative methodological approaches are formulated. The directions of the transformation of risk management paradigms were analyzed, the expediency of a systemic multidimensional approach to the implementation of integrated risk management based on modern international regulatory standards was clarified. The need for the convergence of project management methods in risk management for better prioritization of risks, consistency with the strategic goal of activity, and planning of steps to optimally achieve the goal is substantiated. The marketing orientation of bank risk protection projects is revealed. It is proposed to expand the list of banks' risks and to transform the weighting coefficients of the existing ones in the conditions of innovative technological transformations in order to strengthen socio-ecological responsibility to society. Particular attention is paid to determining the place of marketing risks in the management system of banks, as a critical element of an effective risk management model. The direction of improving the information and analytical support of the bank's risk protection projects through the implementation of CRM systems as an information communication tool and an information source on marketing risk management is proposed.

The role of regulatory zones in mitigating the consequences of nuclear accidents

This paper addresses the New Alamein City and its proximity to El Dabaa nuclear power plant. It is mainly concerned with the 32 km radius of low population zone around the plant. It questions the erection of the new city and its future extension and their proximity to El Dabaa plant because nuclear safety regulations require the presence of a low population zone (100–194 inhabitants/square kilometer) around nuclear power plants. International and national regulatory standards have been revised for this purpose. Egyptian regulations for population are derived from the IAEA guides, the USNRC regulation and the Russian regulations regarding population protection zones around nuclear power plants. The research is status defining and data based, and the methodology used is the analysis of maps and documents, measurements and calculations. Data about the New Alamein City are reviewed and distances between El Dabaa nuclear power plant and the new city are measured in Google maps. Population numbers are collected, estimated, and areas and densities are calculated. Wind directions from the plant in the direction of the city are outlined. The distance between the Dabaa site and the west border of New Alamein City is 27.4kms, 4.6 km inside the recommended 32kms radius of low population zone around the plant. The intersection between that part of the city and the low population zone covers an area of 16.4 sq km. The population density of that part is 9900 inhabitants/sq km, which means that around 162,630 inhabitants may inhabit that area. The downwind from the plant reaches the lower half of the city. Recommendations are made to reduce the population density in that part of the new city lying in the downwind of the nuclear power plant and omission of future plans of extending it to the west direction. No new areas should be populated, as the density recommended for low population zone has already been exceeded. The only extension to New Alamein City can be in the east direction away from the plant. When a site is brilliant but is near highly populated areas, feasible emergency plan is key. Yet, the process of evacuation is not an easy job as the Fukushima accident has proved, even though Japan is a highly developed country, as many elderly people have died from the evacuation and fear of it, rather than from the radiation by the accident.

During our research, the project conducted a review of the scientific literature and reviewed the methodology for assessing and monitoring the level of poverty. The differentiation of the methodology and measurement methodology used by the World Bank in countries with developed economies and developing economies was carried out, including the specifics of the Kazakh methodology, the methodology for assessing poverty according to international regulatory standards (World Bank, etc.). Theoretical studies of poverty, social inequality and the social structure of society in modern sociological approaches. The approaches to understanding society in the issues of poverty, wealth and inequality, as well as subjectivity, data sets and methodological approaches to understanding society, on the compliance of the current policy of poverty eradication in developed countries in comparison with developing countries are considered.
