Nanosafety: a Perspective on Nano-Bio Interactions.

Engineered nanomaterials offer numerous benefits to society ranging from environmental remediation to biomedical applications such as drug or vaccine delivery as well as clean and cost-effective energy production and storage, and the promise of a more sustainable way of life. However, as nanomaterials of increasing sophistication enter the market, close attention to potential adverse effects on human health and the environment is needed. Here a critical perspective on nanotoxicological research is provided; the authors argue that it is time to leverage the knowledge regarding the biological interactions of nanomaterials to achieve a more comprehensive understanding of the human health and environmental impacts of these materials. Moreover, it is posited that nanomaterials behave like biological entities and that they should be regulated as such.

Effects of superparamagnetic iron oxide nanoparticles (SPIONS) testicular injection on Leydig cell function and sperm production in a murine model

In the domain of medical advancement, nanotechnology plays a pivotal role, especially in the synthesis of biocompatible materials for therapeutic use. Superparamagnetic Iron Oxide Nanoparticles (SPIONs), known for their magnetic properties and low toxicity, stand at the forefront of this innovation. This study explored the reproductive toxicological effects of Sodium Citrate-functionalized SPIONs (Cit_SPIONs) in adult male mice, an area of research that holds significant potential yet remains largely unknown. Our findings reveal that Cit_SPIONs induce notable morphological changes in interstitial cells and the seminiferous epithelium when introduced via intratesticular injection. This observation is critical in understanding the interactions of nanomaterials within reproductive biological systems. A striking feature of this study is the rapid localization of Cit_SPIONs in Leydig cells post-injection, a factor that appears to be closely linked with the observed decrease in steroidogenic activity and testosterone levels. This data suggests a possible application in developing nanostructured therapies targeting androgen-related processes. Over 56 days, these nanoparticles exhibited remarkable biological distribution in testis parenchyma, infiltrating various cells within the tubular and intertubular compartments. While the duration of spermatogenesis remained unchanged, there were many Tunel-positive germ cells, a notable reduction in daily sperm production, and reduced progressive sperm motility in the treated group. These insights not only shed light on the intricate mechanisms of Cit_SPIONs interaction with the male reproductive system but also highlight the potential of nanotechnology in developing advanced biomedical applications.

A critical review on the effect of graphene oxide in fiber reinforced cementitious systems

The most widely used building materials nowadays are aggregates and cementitious material composites. Recently, the Graphene Oxide (GO) nanomaterial has grabbed a lot of attention in the material science of building materials. These graphene oxide-based cementitious composites have been studied in terms of their effect on microstructure, hydration mechanisms, mechanical and durable properties, as well as the performance of GO-cement composites, which transform ordinary cement materials into intelligent, sturdy, and long-lasting composites. An overview of the contemporary state-of-the-art effect of cementitious composites containing GO with fiber addition is presented in this paper. The main purpose of this paper is to critically review GO's performance with fibers in cementitious composites. In this review, we examined the properties like Workability, mechanical properties, and durability of cement-based materials along with the microstructural characterization. The tailored property of cementitious composite made with graphene oxide and fibers controls not only the secondary hydration mechanism but also improves the material’s mechanical performance, robustness and other multifunctional properties. The interaction of GO nanomaterial with fiber results in a remarkable and unique performance, such as the improvement of gel pores, the decrease of bigger capillary pores, the bridging of microcracks and the resistance to their spread, and the overall performance improvement of cementitious composites. Thus, this article aims to provide a substantial critical review of the impact of GO and fibers in cementitious composites.

Mechanistic Insight into Binding of Graphene Towards Multiple Therapeutic Target of SARS-CoV-2 by Molecular Docking and Dynamics Studies

Background: Even after more than three years of emergence of coronavirus diseases 2019 (COVID-19), it remains incurable, despite the use of emergency-approved vaccines. The impact of the emergency-approved vaccines against it is not highly certain. The entire world may face surge of new wave of the infection, and therefore, its reoccurrence cannot be denied. Causalities and economic losses may happen in the future. In this situation, it is need of the hour to explore some suitable therapeutic agents against this highly infectious and challenging disease. Organic molecules, inactivated virus etc were largely focused to derive the vaccines, however, nanomaterials still remain untested for it. Interaction of nanomaterials like graphene against proteins of the coronavirus to inhibit its activity is yet to be tested computationally. Method: In this computational research work, graphene is tested for interaction with the viral proteins, as the dimension of the novel coronavirus also belongs to nano-scale (diameter 65-125 nm). A detailed study ondocking structures and molecular dynamics between viral proteins and graphene, is undertaken. Autodock and Discovery Studio packages were used for docking calculations and interaction analysis, respectively. Molecular dynamics simulation was performed using NAMD software and obtained results were visualized using VMD software. Results: Range of proteins of the coronavirus including spike proteins (both open and closed form), Main Protease (MPRO), Receptor-Binding Domain (RBD), RNA-dependent RNA polymerase (RdRp), and Angiotensin Converting Enzyme 2 (ACE2) have been individually evaluated for their docking and dynamics with graphene, so as to inactivate the virus. Details of interacting amino acid residues of coronavirus with graphene is found in the study. Conclusion: Computationally, it is observed that most of these proteins were found interactive through their amino acid residues with graphene. Number of interacting amino acid residues is considerable enough to collectively acquire high binding energy. It can be envisaged that cumulative perturbations of all the interactions of each protein caused by graphene can lower down or inactivate the virus.

Interaction of Carbon Nanotubes, Capped Carbon Nanotubes, CNT2–5, C60, C70, HO-C60, [C60]2, and [C60]3 Fullerenes with Virulence Factors of Gram-Negative and Gram-Positive Bacteria: Potential Applications for 3D-Printed Scaffolds

The antimicrobial application of carbon nanomaterials, such as carbon nanotubes (CNTs), capped CNTs, CNT2–5, C60, C70, HO-C60, [C60]2, and [C60]3 fullerenes, is increasing, owing to their low cytotoxicity properties compared to other nanomaterials such as metallic nanoparticles. Enhanced mechanical properties and antibacterial activity can be caused by the incorporation of CNTs in 3-dimensional (3D) printed nanocomposites (NCs). The interruption of the bacterial membrane resulting from the cylindrical shape and high aspect ratio properties has been found to be the most prominent antibacterial mechanism of CNTs. However, the unraveling interaction of CNTs, capped CNTs, CNT2–5, C60, C70, HO-C60, [C60]2, and [C60]3 fullerenes with virulence factors of the main bacterial pathogenesis has not yet been understood. Therefore, in the present study, interactions of these carbon-based nanomaterials with the eight virulence factors, including protein kinase A and (ESX)-secreted protein B of Mycobacterium tuberculosis, pseudomonas elastase and exotoxin A of Pseudomonas aeruginosa, alpha-hemolysin and penicillin-binding protein 2a of Staphylococcus aureus, and shiga toxin 2a and heat-labile enterotoxin of Escherichia coli, were evaluated with the molecular docking method of AutoDock Vina. This study disclosed that the binding affinity was highest for CNT2–5 and [C60]3 toward alpha-hemolysin, with binding energies of −32.7 and −26.6 kcal/mol, respectively. The stability of the CNT2–5–alpha-hemolysin complex at different times was obtained according to the normal mode analysis of ElNémo and iMOD servers.

Green chemistry routed sugar press mud for (2D) ZnO nanostructure fabrication, mineral fortification, and climate-resilient wheat crop productivity

Nanotechnology appears to be a promising tool to redefine crop nutrition in the coming decades. However, the crucial interactions of nanomaterials with abiotic components of the environment like soil organic matter (SOM) and carbon‒sequestration may hold the key to sustainable crop nutrition, fortification, and climate change. Here, we investigated the use of sugar press mud (PM) mediated ZnO nanosynthesis for soil amendment and nutrient mobilisation under moderately alkaline conditions. The positively charged (+ 7.61 mv) ZnO sheet-like nanoparticles (~ 17 nm) from zinc sulphate at the optimum dose of (75 mg/kg blended with PM (1.4% w/w) were used in reinforcing the soil matrix for wheat growth. The results demonstrated improved agronomic parameters with (~ 24%) and (~ 19%) relative increases in yield and plant Zn content. Also, the soil solution phase interactions of the ZnO nanoparticles with the PM-induced soil colloidal carbon (− 27.9 mv and diameter 0.4864 μm) along with its other components have influenced the soil nutrient dynamics and mineral ecology at large. Interestingly, one such interaction seems to have reversed the known Zn-P interaction from negative to positive. Thus, the study offers a fresh insight into the possible correlations between nutrient interactions and soil carbon sequestration for climate-resilient crop productivity.

Influence of Particle Size of CeO2 Nanospheres Encapsulated in SBA-15 Mesopores on SO2 Tolerance during NH3-SCR Reaction

Ce-based selective catalytic reductions with an NH3 (NH3-SCR) catalyst have emerged as a focal point in denitrification catalyst research. However, the correlation between the structural characteristics of Ce-based catalysts and the influence of CeO2 nanoparticle size on SO2 resistance remains unclear. CeO2 nanospheres with different sizes of less than 10 nm were synthesized, and a series of supported CeO2/SBA-15 catalysts were prepared according to the 10 nm pore size of SBA-15. These catalysts were used to explore the influence of the size of the CeO2 nanospheres on these catalysts, specifically on their SO2 resistance in NH3-SCR reactions. With the increase in size, their SO2 resistance became stronger. The results of NH3-TPD, H2-TPR, and XPS indicated that the catalyst with the largest particle size had the lowest adsorption of SO2, which was attributed to more acid sites and a mutual effect between Si and Ce, resulting in the best SO2 resistance. It was also observed that there was less sulfate deposition on the catalyst by thermogravimetric analysis. In situ DRIFTs revealed that after SO2 poisoning, the NH3-SCR reaction on the catalyst predominantly follows the E-R mechanism. This study offers recommendations for the development of Ce-based SO2-resistant NH3-SCR catalysts, specifically focusing on the synthesis and interaction of nanomaterials.

Cellular Alterations Due to Direct and Indirect Interaction of Nanomaterials with Nucleic Acids.

Deoxyribonucleic acid (DNA) represents the main reservoir of genetic information in the cells, which is why it is protected in the nucleus. Entry into the nucleus is, in general, difficult, as the nuclear membrane is a selective barrier to molecules longer than 40 kDa. However, in some cases, the size of certain nanoparticles (NPs) allows their internalization into the nucleus, thus causing a direct effect on the DNA structure. NPs can also induce indirect effects on DNA through reactive oxygen species (ROS) generation. In this context, nanomaterials are emerging as a disruptive tool for the development of novel therapies in a broad range of biomedical fields; although their effect on cell viability is commonly studied, further interactions with DNA or indirect alterations triggered by the internalization of these materials are not always clarified, since the small size of these materials makes them perfectly suitable for interaction with subcellular structures, such as the nucleus. In this context, and using as a reference the predicted interactions presented in a computational model, we describe and discuss the observed direct and indirect effects of the implicated nanomaterials on DNA.

PH-tailored monodispersed nanopolystyrene: environmental impact on PGPR and nitrogen cycling in agricultural soils

ABSTRACT Emulsion polymerization was optimized to synthesize polystyrene nanoparticles (NPSt) with tailored sizes by adjusting pH and azobisisobutyronitrile (AIBN) content. An NPSt sample prepared with 20 mg AIBN at pH 8 showed the smallest size (88 nm) and narrowest distribution (24 nm peak width). XRD patterns indicated semi-crystalline nature for this optimized sample. The involvement of synthesized NPSt in the nitrogen (N) cycle enhanced our understanding of nitrogen transformations and the response of microbial populations to polystyrene nanoparticles in the environment. In vitro experiments revealed compatibility of NPSt with tested PGPR strains, with no inhibition zones observed. Furthermore, carrot growth in soil containing 400 mg/kg NPSt showed no significant changes in biomass or shoot height compared to control soil. This demonstrates the NPSt samples had no negative effects on PGPR or soil nitrogen transformations and bacterial communities. Overall, the study optimized the synthesis of NPSt, evaluated environmental impacts on soil nitrogen cycling and PGPR, and found no toxicity of NPSt on PGPR or carrot growth up to 400 mg/kg concentration. The insights contribute to understanding nanomaterial interactions in agricultural systems, highlighting the need to consider the environmental impacts of nanoparticles.

Transport of Nanoparticles into Plants and Their Detection Methods.

Additionally, this article delves into the importance of detection methods for studying nanoparticle localization and quantification within plant tissues. Various techniques are presented as valuable tools for comprehensively understanding nanoparticle-plant interactions. The reliance on multiple detection methods for data validation is emphasized to enhance the reliability of the research findings. The future outlooks of this field are explored, including the potential use of alternative introduction sites, such as stems, and the continued development of nanoparticle formulations that improve adhesion and penetration. By addressing these challenges and fostering multidisciplinary research, the field of nanoparticle transport in plants is poised to make significant contributions to sustainable agriculture and environmental management.

Silica-coated LiYF4:Yb3+, Tm3+ upconverting nanoparticles are non-toxic and activate minor stress responses in mammalian cells.

Lanthanide-doped upconverting nanoparticles (UCNPs) are ideal candidates for use in biomedicine. The interaction of nanomaterials with biological systems determines whether they are suitable for use in living cells. In-depth knowledge of the nano-bio interactions is therefore a pre-requisite for the development of biomedical applications. The current study evaluates fundamental aspects of the NP-cell interface for square bipyramidal UCNPs containing a LiYF4:Yb3+, Tm3+ core and two different silica surface coatings. Given their importance for mammalian physiology, fibroblast and renal proximal tubule epithelial cells were selected as cellular model systems. We have assessed the toxicity of the UCNPs and measured their impact on the homeostasis of living non-malignant cells. Rigorous analyses were conducted to identify possible toxic and sub-lethal effects of the UCNPs. To this end, we examined biomarkers that reveal if UCNPs induce cell killing or stress. Quantitative measurements demonstrate that short-term exposure to the UCNPs had no profound effects on cell viability, cell size or morphology. Indicators of oxidative, endoplasmic reticulum, or nucleolar stress, and the production of molecular chaperones varied with the surface modification of the UCNPs and the cell type analyzed. These differences emphasize the importance of evaluating cells of diverse origin that are relevant to the intended use of the nanomaterials. Taken together, we established that short-term, our square bipyramidal UCNPs are not toxic to non-malignant fibroblast and proximal renal epithelial cells. Compared with established inducers of cellular stress, these UCNPs have minor effects on cellular homeostasis. Our results build the foundation to explore square bipyramidal UCNPs for future in vivo applications.

Interaction of a Porphyrin Aluminum Metal-Organic Framework with Volatile Organic Sulfur Compound Diethyl Sulfide Studied via In Situ and Ex Situ Experiments and DFT Computations.

The study presents complementary experiments and quantum chemical DFT computations to reveal the molecular-level interactions of an advanced nanomaterial, porphyrin aluminum metal-organic framework (compound 2), with the volatile organic sulfur compound diethyl sulfide (DES). First, the intermolecular host-guest interactions during the sorption of DES were explored under dynamic conditions, using the vapor of DES in flowing air. The in situ time-dependent ATR-FTIR spectroscopy in a controlled atmosphere was significantly improved though the use of a new facilely built spectroscopic mini-chamber. The binding site of DES in compound 2 involves the μ(O-H) and COO- groups of the linker of the sorbent. Further, the chemical kinetics of the sorption of DES was investigated, and it follows the Langmuir adsorption kinetic model. That is, depending on the time interval, the process obeys either the pseudo-first- or pseudo-second-order rate law. For the Langmuir adsorption of the pseudo-first order, the rate constant is robs = 0.165 ± 0.017 min-1. Next, the interaction of compound 2 with the saturated vapor of DES yields the adsorption complex compound 3 [Al-MOF-TCPPH2]2(DES)7. The adsorbed amount of DES is very large at 36.5 wt.% or 365 mg/g sorbent, one of the highest values reported on any sorbent. The molecular modes of bonding of DES in the complex were investigated through quantum chemical DFT computations. The adsorption complex was facilely regenerated by gentle heating. The advanced functional material in this work has significant potential in the environmental remediation of diethyl sulfide and related volatile organic sulfur compounds in air, and it is an interesting target of mechanistic studies of sorption.

A Predictive Model for Monolayer‐Selective Metal‐Mediated MoS2 Exfoliation Incorporating Electrostatics

AbstractThe metal‐mediated exfoliation (MME) method enables monolayer‐selective exfoliation of van der Waals (vdW) crystals, improving the efficacy of large monolayer production. Previous physical models explaining monolayer‐selective MME propose that the main contributors to monolayer‐selectivity are vdW crystal/metal surface binding energy and/or vdW crystal layer strain resulting from lattice mismatch. However, the performance of some metals for MME is inconsistent with these models. Here, a new model is proposed using MoS2 as a representative vdW crystal. The model explains how the MoS2/metal interface electrostatics, in combination with strain, determines monolayer‐selectivity of MME by modulating the MoS2 interlayer energy. Monolayer MoS2/metal interfaces are characterized using in situ Raman spectroscopy and density functional theory calculations to estimate the electrostatics and strain of MoS2 in contact with different metals. The model successfully demonstrates the dependence of MME monolayer‐selectivity on the MoS2/metal interface electrostatics and highlights the significance of electrostatics in nanomaterial vdW interactions.

A review on toxicity of nanomaterials in agriculture: Current scenario and future prospects.

Phytonanotechnology plays a crucial part in the production of good quality and high-yield food. It can also alter the plant's production systems, hence permitting the efficient, controlled and stable release of agrochemicals such as fertilizers and pesticides. An advanced understanding of nanomaterials interaction with plant responses like localization and uptake, etc. could transfigure the production of crops with high disease resistance and efficient nutrients utilization. In agriculture, the use of nanomaterials has gained acceptance due to their wide-range applications. However, their toxicity and bioavailability are the major hurdles for their massive employment. Undoubtedly, nanoparticles positively influence seeds germination, growth and development, stress management and post-harvest handling of vegetables and fruits. These nanoparticles may also cause toxicity in plants through oxidative stress by generation of excessive reactive oxygen species thus affecting the cellular biomolecules and targeting different channels. Nanoparticles have shown to exert various effects on plants that are mainly affected by various attributes such as physicochemical features of nanomaterials, coating materials for nanoparticles, type of plant, growth stages and growth medium for plants. This article discusses the interaction, accretion and toxicity of nanomaterials in plants. The factors inducing nanotoxicity and the mechanisms followed by nanomaterials causing toxicity are also instructed. At the end, detoxification mechanism of plant is also presented.

The intrinsic and extrinsic mechanisms regulated by functional carbon nanodots for the phytoremediation of multi-metal pollution in soils

Functional carbon nanodots (FCNs) were currently demonstrated to regulate plant behavior in the agricultural and environmental areas. However, their regulation mechanisms on the interactions of plant-soil system during phytoremediation remain unrevealed. Here, Solanum nigrum L. was employed to explore the intrinsic and extrinsic mechanisms regulated by FCNs in the phytoremediation of Cd-Pb co-contaminated soils. The mediation of FCNs on metal removal and plant growth showed a hormesis manner, wherein the maximum induction effect was contributed by 15 mg kg-1 FCNs. Cd/Pb removal were enhanced by 8.5% and 31.6%, respectively. Moreover, FCNs reallocate metal distribution in plant by immobilized metals in roots and suppressed metal translocation to leaves. Improving plant growth (by 82.8% for root), stimulating plant hormesis, and activating plant detoxification pathways are the intrinsic mechanism for the phytoremediation smartly regulated by FCNs. Notably, FCNs induced soil enzyme activities that associated with soil nutrients recycling, up-regulated the microbial diversity and the soil immune system, and regulated S. nigrum L. to recruit beneficial microbials in the rhizosphere. The above-mentioned comprehensive improvement of soil micro-environment is the extrinsic mechanism regulated by FCNs. This study provides new insights to evaluate the interactions of nanomaterials with plant-soil system under soil contamination.

The Mechanism of Drug Carryover in Feed Manufacturing as a Function of Drug Properties and Equipment Design—A Brief Review

This paper thoroughly reviews the mechanism of veterinary drug carryover in feed manufacturing facilities, factors resulting in varying concentrations of drug carryover in processing equipment, the impact of chemical and physical properties of drugs, and the effect of equipment type and design. The Google Scholar database (from 1998 to 2023) was searched with words and phrases such as drug carryover, feed manufacturing, equipment cleaning and validation, food allergen control, sources of drug carryover, and process parameters in drug carryover. Some papers were from the Iowa State University Library database and PubMed. Drug carryover is a function of ingredients, nature of drugs, equipment type, process parameters, and cleaning procedures. The gaps are the lack of commercial feed mills data on the role and interaction of nanomaterials, molasses, equipment type, and process parameters in drug carryover in animal feed. Modification of process parameters, e.g., airflow in bucket elevators and the interaction of feed ingredients, composition, equipment type, and design, need to be investigated in the commercial setting to address drug carryover. Rhetorically, can big data facilitate the standardization of cleaning procedures at feed mills? The findings can result in drug carryover prevention/control in animal feed and animal-based human food.

Moderate binding of villin headpiece protein to C3N3 nanosheet reveals the suitable biocompatibility of this nanomaterial

Since its recent successful synthesis and due to its promising physical and chemical properties, the carbon nitrite nanomaterial, C3N3, has attracted considerable attention in various scientific areas. However, thus far, little effort has been devoted to investigating the structural influence of the direct interaction of this 2D nanomaterial and biomolecules, including proteins and biomembranes so as to understand the physical origin of its bio-effect, particularly from the molecular landscape. Such information is fundamental to correlate to the potential nanotoxicology of the C3N3 nanomaterial. In this work, we explored the potential structural influence of a C3N3 nanosheet on the prototypical globular protein, villin headpiece (HP35) using all-atom molecular dynamics (MD) simulations. We found that HP35 could maintain its native conformations upon adsorption onto the C3N3 nanosheet regardless of the diversity in the binding sites, implying the potential advantage of C3N3 in protecting the biomolecular structure. The adsorption was mediated primarily by vdW interactions. Moreover, once adsorbed on the C3N3 surface, HP35 remains relatively fixed on the nanostructure without a distinct lateral translation, which may aid in keeping the structural integrity of the protein. In addition, the porous topological structure of C3N3 and the special water layer present on the C3N3 holes conjointly contributed to the restricted motion of HP35 via the formation of a high free energy barrier and a steric hindrance to prevent the surface displacement. This work revealed for the first time the potential influence of the 2D C3N3 nanomaterial in the protein structure and provided the corresponding in-depth molecular-level mechanism, which is valuable for future applications of C3N3 in bionanomedicine.

Emerging Trends in Nanomaterial-Based Biomedical Aspects

Comprehending the interfacial interaction of nanomaterials (NMs) and biological systems is a significant research interest. NMs comprise various nanoparticles (NPs) like carbon nanotubes, graphene oxides, carbon dots, graphite nanopowders, etc. These NPs show a variety of interactions with biological interfaces via organic layers, therapeutic molecules, proteins, DNA, and cellular matrices. A number of biophysical and colloidal forces act at the morphological surface to regulate the biological responses of bio-nanoconjugates, imparting distinct physical properties to the NMs. The design of future-generation nano-tools is primarily based on the basic properties of NMs, such as shape, size, compositional, functionality, etc., with studies being carried out extensively. Understanding their properties promotes research in the medical and biological sciences and improves their applicability in the health management sector. In this review article, in-depth and critical analysis of the theoretical and experimental aspects involving nanoscale material, which have inspired various biological systems, is the area of focus. The present review article focuses on recent emerging trends in the synthesis and applications of nanomaterials with respect to various biomedical applications. This article provides value to the literature as it summarizes the state-of-the-art nanomaterials reported, especially within the health sector. It has been observed that nanomaterial applications in drug design, diagnosis, testing, and in the research arena, as well as many fatal disease conditions like cancer and sepsis, have explored alongwith drug therapies and other options for the delivery of nanomaterials. Even the day-to-day life of the synthesis and purification of these materials is changing to provide us with a simplified process. This review article can be useful in the research sector as a single platform wherein all types of nanomaterials for biomedical aspects can be understood in detail.
