Neural oscillations guiding action during effects imagery

Goal-directed acting requires the integration of sensory information but can also be performed without direct sensory input. Examples of this can be found in sports and can be conceptualized by feedforward processes. There is, however, still a lack of understanding of the temporal neural dynamics and neuroanatomical structures involved in such processes. In the current study, we used EEG beamforming methods and examined 37 healthy participants in two well-controlled experiments varying the necessity of anticipatory processes during goal-directed action. We found that alpha and beta activity in the medial and posterior cingulate cortex enabled feedforward predictions about the position of an object based on the latest sensorimotor state. On this basis, theta band activity seems more related to sensorimotor representations, while beta band activity would be more involved in setting up the structure of the neural representations themselves. Alpha band activity in sensory cortices reflects an intensified gating of the anticipated perceptual consequences of the to-be-executed action. Together, the findings indicate that goal-directed acting through the anticipation of the predicted state of an effector is based on accompanying processes in multiple frequency bands in midcingulate and sensory brain regions.

Somatostatin interneurons control the timing of developmental desynchronization in cortical networks

Synchronous neuronal activity is a hallmark of the developing brain. In the mouse cerebral cortex, activity decorrelates during the second week of postnatal development, progressively acquiring the characteristic sparse pattern underlying the integration of sensory information. The maturation of inhibition seems critical for this process, but the interneurons involved in this crucial transition of network activity in the developing cortex remain unknown. Using invivo longitudinal two-photon calcium imaging during the period that precedes the change from highly synchronous to decorrelated activity, we identify somatostatin-expressing (SST+) interneurons as critical modulators of this switch in mice. Modulation of the activity of SST+ cells accelerates or delays the decorrelation of cortical network activity, a process that involves regulating the maturation of parvalbumin-expressing (PV+) interneurons. SST+ cells critically link sensory inputs with local circuits, controlling the neural dynamics in the developing cortex while modulating the integration of other interneurons into nascent cortical circuits.

Neuronal Control of Posture in Blind Individuals.

The control of posture is guided by the integration of sensory information. Because blind individuals cannot apply visual information to control posture as sighted individuals do they must compensate by the remaining senses. We therefore hypothesize that blind individuals alter their brain activation in the sensorimotor cortex during postural control to compensate for balance control without vision by the increased integration of somatosensory information. Ten blind and ten sighted (matched) individuals controlled posture during conditions with (I) eyes closed / open, and (II) stable / unstable surface conditions. Postural sway was recorded by applying a pressure distribution measuring plate. Brain activation was collected by functional Near InfraRed Spectroscopy (fNIRS) above motor-sensory cortices of the right and left hemispheres. Blind individuals showed significantly increased postural sway when balancing with open eyes on an unstable surface and when compared to sighted individuals. Whereas blind individuals showed significantly increased brain activation when balancing with open eyes on stable and unstable surface conditions, sighted individuals increased their brain oxygenation only during closed eyes and unstable surface conditions. Overall conditions, blind individuals presented significantly increased brain activation in two channels of the left and right hemispheric motor-sensory cortex when compared to sighted individuals. We therefore conclude that sighted individuals increase their brain oxygenation in the sensorimotor cortex during postural control tasks that demand sensory integration processes. Blind individuals are characterized by increased brain activation overall conditions indicating additional sensory integration during postural control. Thus, the sensorimotor cortex of blind individuals adapts to control posture without vision.

Vestibular dysfunction leads to cognitive impairments: Stateofknowledge in the field and clinical perspectives (Review).

The vestibular system may have a critical role in the integration of sensory information and the maintenance of cognitive function. A dysfunction in the vestibular system has a significant impact on quality of life. Recent research has provided evidence of a connection between vestibular information and cognitive functions, such as spatial memory, navigation and attention. Although the exact mechanisms linking the vestibular system to cognition remain elusive, researchers have identified various pathways. Vestibular dysfunction may lead to the degeneration of cortical vestibular network regions and adversely affect synaptic plasticity and neurogenesis in the hippocampus, ultimately contributing to neuronal atrophy and cell death, resulting in memory and visuospatial deficits. Furthermore, the extent of cognitive impairment varies depending on the specific type of vestibular disease. In the present study, the current literature was reviewed, potential causal relationships between vestibular dysfunction and cognitive performance were discussed and directions for future research were proposed.

Comparing the effects of different circular vection stimuli on upright stance

BackgroundUpright quiet stance is maintained through the complex integration of sensory information from the visual, vestibular, and somatosensory systems [1]. Virtual reality (VR) is a well-established tool that has been used to study sensory contributions to balance and induce visual perturbations. Previous assessments of virtual environments have suggested that VR can be used to create various visual stimuli that affect balance [2]; however, there is limited work examining which dynamic visual stimulus, in the form of circular vection (CV), is the most effective at inducing whole body lean. Research questionTherefore, this study assessed the effects of two visual stimuli using VR to better understand their effects on postural control. Methods33 healthy young adults between the ages of 18–40, free of neurological impairments, stood quietly on a force plate for 30 s while wearing a head-mounted display. Participants were exposed to a field of random white dots (DOTS) or a black and white striped tunnel (TUNNEL) that rotated in the roll plane at 60°/s clockwise or counterclockwise. Amplitude was calculated from head orientation data recorded from a head-mounted display, and centre of pressure (COP). ResultsIndependent of visual stimuli, postural lean was in the same direction as the stimulus. The DOTS stimulus increased Head orientation and COP position compared to the TUNNEL stimulus. There was no significant main effect or interaction with direction for Head or COP data. SignificanceWhen comparing the effect of stimulus design on postural sway, a DOTS stimulus was most effective at inducing direction-modulated postural sway This study builds on our understanding of the VR-related destabilizing effects on postural control and shows evidence that a DOTS stimulus has a stronger effect than a TUNNEL stimulus. Overall, it is important to consider the design of visual stimuli when examining VR effects on upright stance.

Cognition and saccadic eye movement performance are impaired in chronic rhinosinusitis.

Patients with chronic rhinosinusitis (CRS) can experience cognitive dysfunction. The literature on this topic mostly reflects patient-reported measurements. Our goal was to assess cognitive function in patients with CRS using objective measures, including saccadic eye movements-a behavioral response reflecting cognitive and sensory information integration that is often compromised in conditions with impaired cognition. Participants (N=24 with CRS, N=23 non-CRS healthy controls) enrolled from rhinology clinic underwent sinonasal evaluation, quality of life assessment (Sino-nasal Outcome Test 22 [SNOT-22]), and cognitive assessment with the Neuro-QOL Cognitive Function-Short Form, the Montreal Cognitive Assessment (MoCA), and recording of eye movements using video-oculography. Participants with CRS were more likely to report cognitive dysfunction (Neuro-QOL; 45.8%vs. 8.7%; p=0.005) and demonstrate mild or greater cognitive impairment (MoCA; 41.7%vs. 8.7%; p=0.005) than controls. Additionally, participants with CRS performed worse on the MoCA overall and within the executive functioning and memory domains (all p<0.05) and on the anti-saccade (p=0.014) and delay saccade (p=0.044) eye movement tasks. Poorer performance on the MoCA (r=-0.422; p=0.003) and the anti-saccade (r=-0.347; p=0.017) and delay saccade (r=-0.419; p=0.004) eye movement tasks correlated with worse CRS severity according to SNOT-22 scores. This study is the first to utilize objective eye movement assessments in addition to researcher-administered cognitive testing in patients with CRS. These patients demonstrated a high prevalence of cognitive dysfunction, most notably within executive functioning and memory domains, with the degree of dysfunction correlating with the severity of CRS.

Insufficiencies in sensory systems reweighting is associated with walking impairment severity in chronic stroke: an observational cohort study.

Walking and balance impairment are common sequelae of stroke and significantly impact functional independence, morbidity, and mortality. Adequate postural stability is needed for walking, which requires sufficient integration of sensory information between the visual, somatosensory, and vestibular centers. "Sensory reweighting" describes the normal physiologic response needed to maintain postural stability in the absence of sufficient visual or somatosensory information and is believed to play a critical role in preserving postural stability after stroke. However, the extent to which sensory reweighting successfully maintains postural stability in the chronic stages of stroke and its potential impact on walking function remains understudied. In this cross-sectional study, fifty-eight community-dwelling ambulatory chronic stroke survivors underwent baseline postural stability testing during quiet stance using the modified Clinical test of Sensory Interaction in Balance (mCTSIB) and assessment of spatiotemporal gait parameters. Seventy-six percent (45/58) of participants showed sufficient sensory reweighting with visual and somatosensory deprivation for maintaining postural stability, albeit with greater postural sway velocity indices than normative data. In contrast, survivors with insufficient reweighting demonstrated markedly slower overground walking speeds, greater spatiotemporal asymmetry, and limited acceleration potential. Adequate sensory system reweighting is essential for chronic stroke survivors' postural stability and walking independence. Greater emphasis should be placed on rehabilitation strategies incorporating multisensory system integration testing and strengthening as part of walking rehabilitation protocols. Given its potential impact on outcomes, walking rehabilitation trials may benefit from incorporating formal postural stability testing in design and group stratification.

Differentiation of workers into soldiers is associated with a size reduction of higher-order brain centers in the neotropical termite Procornitermes araujoi

Comparing the size of functionally distinct brain regions across individuals with remarkable differences in sensory processing and cognitive demands provides important insights into the selective forces shaping animal nervous systems. We took advantage of the complex system of worker-to-soldier differentiation in the termitid Procornitermes araujoi, to investigate how a profound modification of body morphology followed by an irreversible shift in task performance are translated in terms of brain structure and size. This behavioural shift is characterised by a reduction of the once wide and complex behavioural repertoire of workers to one exclusively dedicated to nest defence (soldiers). In accordance with soldier’s reduced cognitive and sensory demands, we show here that differentiation of workers into soldiers is associated with a size reduction of the mushroom body (MB) compartments, higher-order brain regions responsible for multimodal processing and integration of sensory information, as well as learning, memory, and decision-making. Moreover, in soldiers, we found an apparent fusion of the medial and lateral MB calyces likely associated with its volume reduction. These results illustrate a functional neuroplasticity of the MB associated with division of labour, supporting the link between MB size and behavioural flexibility in social insect workers.

Transcriptional regulation in the development and dysfunction of neocortical projection neurons.

Glutamatergic projection neurons generate sophisticated excitatory circuits to integrate and transmit information among different cortical areas, and between the neocortex and other regions of the brain and spinal cord. Appropriate development of cortical projection neurons is regulated by certain essential events such as neural fate determination, proliferation, specification, differentiation, migration, survival, axonogenesis, and synaptogenesis. These processes are precisely regulated in a tempo-spatial manner by intrinsic factors, extrinsic signals, and neural activities. The generation of correct subtypes and precise connections of projection neurons is imperative not only to support the basic cortical functions (such as sensory information integration, motor coordination, and cognition) but also to prevent the onset and progression of neurodevelopmental disorders (such as intellectual disability, autism spectrum disorders, anxiety, and depression). This review mainly focuses on the recent progress of transcriptional regulations on the development and diversity of neocortical projection neurons and the clinical relevance of the failure of transcriptional modulations.

Perception-Action Integration Is Altered in Functional Movement Disorders.

Although functional neurological movement disorders (FMD) are characterized by motor symptoms, sensory processing has also been shown to be disturbed. However, how the integration of perception and motor processes, essential for the control of goal-directed behavior, is altered in patients with FMD is less clear. A detailed investigation of these processes is crucial to foster a better understanding of the pathophysiology of FMD and can systematically be achieved in the framework of the theory of event coding (TEC). The aim was to investigate perception-action integration processes on a behavioral and neurophysiological level in patients with FMD. A total of 21 patients and 21 controls were investigated with a TEC-related task, including concomitant electroencephalogram (EEG) recording. We focused on EEG correlates established to reflect perception-action integration processes. Temporal decomposition allowed to distinguish between EEG codes reflecting sensory (S-cluster), motor (R-cluster), and integrated sensory-motor processing (C-cluster). We also applied source localization analyses. Behaviorally, patients revealed stronger binding between perception and action, as evidenced by difficulties in reconfiguring previously established stimulus-response associations. Such hyperbinding was paralleled by a modulation of neuronal activity clusters, including reduced C-cluster modulations of the inferior parietal cortex and altered R-cluster modulations in the inferior frontal gyrus. Correlations of these modulations with symptom severity were also evident. Our study shows that FMD is characterized by altered integration of sensory information with motor processes. Relations between clinical severity and both behavioral performance and neurophysiological abnormalities indicate that perception-action integration processes are central and a promising concept for the understanding of FMD. © 2023 The Authors. Behavioral and Electrocortical Response to a Sensorimotor Conflict in Individuals with Fibromyalgia.

People with fibromyalgia have been shown to experience more somatosensory disturbances than pain-free controls during sensorimotor conflicts (i.e., incongruence between visual and somatosensory feedback). Sensorimotor conflicts are known to disturb the integration of sensory information. This study aimed to assess the cerebral response and motor performance during a sensorimotor conflict in people with fibromyalgia. Twenty participants with fibromyalgia and twenty-three pain-free controls performed a drawing task including visual feedback that was either congruent with actual movement (and thus with somatosensory information) or incongruent with actual movement (i.e., conflict). Motor performance was measured according to tracing error, and electrocortical activity was recorded using electroencephalography. Motor performance was degraded during conflict for all participants but did not differ between groups. Time-frequency analysis showed that the conflict was associated with an increase in theta power (4-8 Hz) at conflict onset over the left posterior parietal cortex in participants with fibromyalgia but not in controls. This increase in theta suggests a stronger detection of conflict in participants with fibromyalgia, which was not accompanied by differences in motor performance in comparison to controls. This points to dissociation in individuals with fibromyalgia between an altered perception of action and a seemingly unaltered control of action.

Structural and functional differences of the thalamus between drug-naïve Parkinson's disease motor subtypes.

The thalamus is an integrative hub of motor circuits in Parkinson's disease (PD). This study aimed to investigate the alterations of structure and functional connectivity (FC) of the thalamic subregions in the tremor-dominant (TD) subtype and the postural instability and gait difficulty (PIGD) subtype in PD. A total of 59 drug-naïve patients (24 TD and 35 PIGD) and 37 healthy controls were recruited. The volumes of the thalamus and the thalamic subregions were calculated using FreeSurfer. Functional connectivity (FC) analysis of the resting-state functional MRI (rsfMRI) was conducted on the thalamic subregions. Finally, the altered structure and FC were used for correlation analysis with clinical motor scores and for further motor subtypes differentiation. The volumes of the left posterior parietal thalamus (PPtha) in TD patients were significantly lower than those of PIGD patients. Compared with PIGD patients, TD patients exhibited higher FC between the thalamic subregions, the left middle temporal gyrus (MTG), the right dorsolateral superior frontal gyrus (SFGdl), the left middle occipital gyrus (MOG), and the right superior temporal gyrus (STG). Compared with HCs, TD patients showed higher FC between the thalamic subregions and the right SFGdl, as well as the left MOG. Compared with HCs, PIGD patients showed lower FC between the thalamic subregions and the left MTG. In addition, the altered FC was closely related to clinical symptoms and performed high-discriminative power in differentiating the motor subtypes. Increased FC between the thalamic subregions and the sensory cortices in TD patients may indicate a better compensatory capacity for impairment of sensory information integration than that in PIGD patients. The altered FC between the thalamus and the MTG was a potential biomarker for the distinction of the PD motor subtypes.

White Matter Correlates of Reactive Stepping in People with Multiple Sclerosis

Background and Purpose Reactive steps are rapid protective responses after balance challenges. People with MS (PwMS) demonstrate impaired stepping, which can increase fall-risk, and these deficits may be caused by white matter (WM) loss. However, the specific neural mechanisms contributing to reactive stepping in PwMS are poorly understood. We aimed to determine WM correlates of reactive stepping and responsiveness to step training. Methods 14 PwMS participated in an 18-week multiple-baseline study. Participants attended 2 baseline assessments (B1 and B2) before training, a 2-week, 6-session training protocol, and a post-training assessment (P1). Each assessment consisted of 3 backward reactive step trials. Training consisted of 32 stepping trials. Outcomes included the anterior-posterior margin of stability (MOS), step length, and step latency. Tract-Based Spatial Statistics (TBSS) was performed to investigate the WM microstructure by correlating fractional anisotropy (FA), mean diffusivity (MD), and radial diffusivity (RD) with baseline reactive step performance (B1) and immediate responsiveness to training (P1). Results The FA of the right superior longitudinal fasciculus (R-SLF) was associated with baseline step length (p=0.048) and MOS (p=0.048). Corpus Callosum (p=0.047) and right anterior corona radiata (R-ACR) (p=0.047) FA were also related to baseline MOS during backward stepping. FA of the corticospinal tract was also associated with baseline step length (p=0.048). The FA (p=0.047) and the MD (trending- p=0.082) of the L-ACR were also related to step length. No significant associations were found between WM and training responsiveness, but a trend showed an association between the right uncinate fasciculus and a change in step latency (p=0.072). Discussion We identified several WM loci associated with baseline stepping involved in the rapid integration of sensory information and the execution of motor responses. Local brain oscillations and interregional connectivity differentially serve sensory and expectation effects on pain

Pain emerges from the integration of sensory information about threats and contextual information such as an individual's expectations. However, how sensory and contextual effects on pain are served by the brain is not fully understood so far. To address this question, we applied brief painful stimuli to 40 healthy human participants and independently varied stimulus intensity and expectations. Concurrently, we recorded electroencephalography. We assessed local oscillatory brain activity and interregional functional connectivity in a network of six brain regions playing key roles in the processing of pain. We found that sensory information predominantly influenced local brain oscillations. In contrast, expectations exclusively influenced interregional connectivity. Specifically, expectations altered connectivity at alpha (8 to 12 hertz) frequencies from prefrontal to somatosensory cortex. Moreover, discrepancies between sensory information and expectations, i.e., prediction errors, influenced connectivity at gamma (60 to 100 hertz) frequencies. These findings reveal how fundamentally different brain mechanisms serve sensory and contextual effects on pain.

Conceptual Associations Generate Sensory Predictions.

A crucial ability of the human brain is to learn and exploit probabilistic associations between stimuli to facilitate perception and behavior by predicting future events. Although studies have shown how perceptual relationships are used to predict sensory inputs, relational knowledge is often between concepts rather than percepts (e.g., we learned to associate cats with dogs, rather than specific images of cats and dogs). Here, we asked if and how sensory responses to visual input may be modulated by predictions derived from conceptual associations. To this end we exposed participants of both sexes to arbitrary word-word pairs (e.g., car-dog) repeatedly, creating an expectation of the second word, conditional on the occurrence of the first. In a subsequent session, we exposed participants to novel word-picture pairs, while measuring fMRI BOLD responses. All word-picture pairs were equally likely, but half of the pairs conformed to the previously formed conceptual (word-word) associations, whereas the other half violated this association. Results showed suppressed sensory responses throughout the ventral visual stream, including early visual cortex, to pictures that corresponded to the previously expected words compared with unexpected words. This suggests that the learned conceptual associations were used to generate sensory predictions that modulated processing of the picture stimuli. Moreover, these modulations were tuning specific, selectively suppressing neural populations tuned toward the expected input. Combined, our results suggest that recently acquired conceptual priors are generalized across domains and used by the sensory brain to generate category-specific predictions, facilitating processing of expected visual input.SIGNIFICANCE STATEMENT Perceptual predictions play a crucial role in facilitating perception and the integration of sensory information. However, little is known about whether and how the brain uses more abstract, conceptual priors to form sensory predictions. In our preregistered study, we show that priors derived from recently acquired arbitrary conceptual associations result in category-specific predictions that modulate perceptual processing throughout the ventral visual hierarchy, including early visual cortex. These results suggest that the predictive brain uses prior knowledge across various domains to modulate perception, thereby extending our understanding of the extensive role predictions play in perception.

Reshaping the full body illusion through visuo-electro-tactile sensations.

The physical boundaries of our body do not define what we perceive as self. This malleable representation arises from the neural integration of sensory information coming from the environment. Manipulating the visual and haptic cues produces changes in body perception, inducing the Full Body Illusion (FBI), a vastly used approach to exploring humans' perception. After pioneering FBI demonstrations, issues arose regarding its setup, using experimenter-based touch and pre-recorded videos. Moreover, its outcome measures are based mainly on subjective reports, leading to biased results, or on heterogeneous objective ones giving poor consensus on their validity. To address these limitations, we developed and tested a multisensory platform allowing highly controlled experimental conditions, thanks to the leveraged use of innovative technologies: Virtual Reality (VR) and Transcutaneous Electrical Nerve Stimulation (TENS). This enabled a high spatial and temporal precision of the visual and haptic cues, efficiently eliciting FBI. While it matched the classic approach in subjective measures, our setup resulted also in significant results for all objective measurements. Importantly, FBI was elicited when all 4 limbs were multimodally stimulated but also in a single limb condition. Our results behoove the adoption of a comprehensive set of measures, introducing a new neuroscientific platform to investigate body representations.

Changes in both top-down and bottom-up effective connectivity drive visual hallucinations in Parkinson's disease.

Visual hallucinations are common in Parkinson's disease and are associated with a poorer quality of life and a higher risk of dementia. An important and influential model that is widely accepted as an explanation for the mechanism of visual hallucinations in Parkinson's disease and other Lewy body diseases is that these arise due to aberrant hierarchical processing, with impaired bottom-up integration of sensory information and overweighting of top-down perceptual priors within the visual system. This hypothesis has been driven by behavioural data and supported indirectly by observations derived from regional activation and correlational measures using neuroimaging. However, until now, there was no evidence from neuroimaging for differences in causal influences between brain regions measured in patients with Parkinson's hallucinations. This is in part because previous resting-state studies focused on functional connectivity, which is inherently undirected in nature and cannot test hypotheses about the directionality of connectivity. Spectral dynamic causal modelling is a Bayesian framework that allows the inference of effective connectivity-defined as the directed (causal) influence that one region exerts on another region-from resting-state functional MRI data. In the current study, we utilize spectral dynamic causal modelling to estimate effective connectivity within the resting-state visual network in our cohort of 15 Parkinson's disease visual hallucinators and 75 Parkinson's disease non-visual hallucinators. We find that visual hallucinators display decreased bottom-up effective connectivity from the lateral geniculate nucleus to primary visual cortex and increased top-down effective connectivity from the left prefrontal cortex to primary visual cortex and the medial thalamus, as compared with non-visual hallucinators. Importantly, we find that the pattern of effective connectivity is predictive of the presence of visual hallucinations and associated with their severity within the hallucinating group. This is the first study to provide evidence, using resting-state effective connectivity, to support a model of aberrant hierarchical predictive processing as the mechanism for visual hallucinations in Parkinson's disease.

Efficient sensory encoding predicts robust averaging

Not every item in a stimulus ensemble equally contributes to the perceived ensemble average. Rather, items with feature values close to the ensemble mean (inlying items) contribute stronger compared to those items whose feature values are further away from the mean (outlying items). This nonuniform weighting process, named robust averaging, has been interpreted as evidence against an optimal integration of sensory information. Here, however, we show that robust averaging naturally emerges from an optimal integration process when sensory encoding is efficiently adapted to the ensemble statistics in the experiment. We demonstrate that such a model can accurately fit several existing datasets showing robust perceptual averaging in discriminating low-level stimulus features such as orientation. Across various feature domains, our model accurately predicts subjects’ decision accuracy and nonuniform weighting profile, and both their dependency on the specific stimulus distribution in the experiments. Our results suggest that the human visual system forms efficient sensory representations on short time-scales to improve overall decision performance.

Atypical sensory perception has been recognized in autistic individuals since its earliest descriptions and is now considered a key characteristic of autism. Although the integration of sensory information (multisensory integration; MSI) has been demonstrated to be altered in autism, less is known about how this perceptual process differs with age. This study aimed to assess the integration of audiovisual information across autistic children and adolescents. MSI was measured using a non-social, simultaneity judgment task. Variation in temporal sensitivity was evaluated via Gaussian curve fitting procedures, allowing us to compare the width of temporal binding windows (TBWs), where wider TBWs indicate less sensitivity to temporal alignment. We compared TBWs in age and IQ matched groups of autistic (n= 32) and neurotypical (NT; n= 73) children and adolescents. The sensory profile of all participants was also measured. Across all ages assessed (i.e., 6 through 18 years), TBWs were negatively correlated with age in the autistic group. A significant correlation was not found in the NT group. When compared as a function of child (6-12 years) and adolescent (13-18 years) age groups, a significant interaction of group (autism vs NT) by age group was found, whereby TBWs became narrower with age in the autistic, but not neurotypical group. We also found a significant main effect of age and no significant main effect of group. Results suggest that TBW differences between autistic and neurotypical groups diminishes with increasing age, indicating an atypical developmental profile of MSI in autism which ameliorates across development.
