Mepiquat chloride priming confers the ability of cotton seed to tolerate salt by promoting ABA-operated GABA signaling control of the ascorbate–glutathione cycle

BackgroundEnsuring that seeds germinate and emerge normally is a prerequisite for cotton production, esp. in areas with salinized soil. Priming with mepiquat chloride (MC) can promote seed germination and root growth under salt stress, but its mechanism has not been fully elucidated. In this study, physiological and biochemical experiments revealed that MC-priming promotes the tolerance of cotton seeds to salt stress by increasing the ability of antioxidant enzymes related to the ascorbate–glutathione (AsA-GSH) cycle to scavenge reactive oxygen species (ROS).ResultsResults revealed that treatment with inhibitors of abscisic acid (ABA) and γ-aminobutyric acid (GABA) biosynthesis reduced the positive effects of MC-priming. Similarly, MC-priming increased the contents of ABA and GABA under salt stress by stimulating the expression levels of GhNCED2 and GhGAD4 and the activity of calmodulin-binding (CML) glutamate decarboxylase (GAD). Further analysis showed that an inhibitor of ABA synthesis reduced the positive impacts of MC-priming on the content of GABA under salt stress, but the content of ABA was not affected by the GABA synthesis inhibitor. Furthermore, a multi-omics analysis revealed that MC-priming increased the abundance and phosphorylation levels of the proteins related to ABA signaling, CML, and Ca2+ channels/transporters in the MC-primed treatments, which resulted in increased oscillations in Ca2+ in the MC-primed cotton seeds under salt stress.ConclusionIn summary, these results demonstrate that MC-mediated ABA signaling operates upstream of the GABA synthesis generated by GAD by activating the oscillations of Ca2+ and then enhancing activity of the AsA-GSH cycle, which ensures that cotton seeds are tolerant to salt stress.

R3-MYB proteins OsTCL1 and OsTCL2 modulate seed germination via dual pathways in rice

Salt and drought stress are common abiotic factors that exert a detrimental influence on seed germination, potentially leading to significantly impaired growth and production in rice. Gaining a comprehensive understanding of the molecular response of seeds to abiotic stress during the germination is of paramount importance. In the present study, we identified two R3-MYB genes in rice, namely OsTCL1 and OsTCL2, and characterized their roles in regulating seed germination under salt and drought stress. Plants with tcl1 and tcl2 mutant alleles exhibited delayed seed germination, particularly under stress conditions. The tcl1 tcl2 double mutant showed an even more pronounced reduction in germination during initial stages of germination, thereby indicating a redundant regulatory function of OsTCL1 and OsTCL2 in seed germination under abiotic stresses. Furthermore, we demonstrated that the transcript levels of several phospholipase D (PLD) genes were downregulated in the tcl1 tcl2 mutant, resulting in a decreased level of the phosphatidic acid (PA) product. Application of 1-butanol, a competitive substrate inhibitor of PLD-dependent production of PA, attenuated the stress response of the tcl1 tcl2 mutant. This suggests that OsTCL1 and OsTCL2 partially modulate seed germination through the PLD-PA signaling pathway. Moreover, there were alterations in the expression of genes involved in abscisic acid (ABA) biosynthesis, metabolism and signaling transduction in the double mutant. These changes affected the endogenous ABA level and ABA response, thereby influencing seed germination. Application of both 1-butanol and ABA synthesis inhibitor sodium tungstate (Na2WO4) nearly eliminated the differences in stress response between wild type and the tcl1 tcl2 mutant. This indicates that OsTCL1 and OsTCL2 synergistically coordinate seed germination under abiotic stresses through both the PLD-PA signaling and ABA-mediated pathways.

The influence of ABA on the photosynthesis of the rare and endangered Emmenopterys henryi under salt stress

Emmenopterys henryi Oliv., a monotypic genus in family Rubiaceae with high scientific and application value, is currently endangered. Adaptability of E. henryi to environment is important for its cultivation, management, and application. In this paper, the effect of abscisic acid (ABA) on the photosynthesis of E. henryi under salt stress was discussed. Results showed that 0.3 % (m/v) NaCl led to reduction of net photosynthetic rate (PN), instantaneous carboxylation efficiency (ICE), and Rubisco activity but the increase of intercellular CO2 concentration (ci), which were remarkable relieved by ABA pretreatment. NaCl also induced reduction of the maximal efficiency of PS II photochemistry (Fv/Fm), the photochemical quenching (qP), the non-photochemical quenching (NPQ), the actual PS II efficiency [Y(II)], the regulated-energy dissipation [Y(NPQ)], whereas the increase of the unregulated energy dissipation [Y(NO)]. ABA application remarkably mitigated the depression of Fv/Fm, qP, Y(II) and the increase of Y(NO), but further promoted NPQ and Y(NPQ) in E. henryi due to high salinity. In addition, decrease of chlorophyll content, accumulation of MDA, and ion leakage in E. henryi due to high salinity were significantly alleviated by ABA pretreatment. Fluridone, an inhibitor of endogenous ABA synthesis, partially or completely reversed ABA mediated effects. These results suggested that ABA might improve the photosynthesis of E. henryi under salt stress via allocating more energy to photochemical reactions, or/and mitigating the decrease of Rubisco activity, or/and protecting photosynthetic apparatus by reinforcement of heat dissipation and alleviating oxidative damage.

FASCICLIN-LIKE 18 Is a New Player Regulating Root Elongation in Arabidopsis thaliana.

The plasticity of root development represents a key trait that enables plants to adapt to diverse environmental cues. The pattern of cell wall deposition, alongside other parameters, affects the extent, and direction of root growth. In this study, we report that FASCICLIN-LIKE ARABINOGALACTAN PROTEIN 18 (FLA18) plays a role during root elongation in Arabidopsis thaliana. Using root-specific co-expression analysis, we identified FLA18 to be co-expressed with a sub-set of genes required for root elongation. FLA18 encodes for a putative extra-cellular arabinogalactan protein from the FLA-gene family. Two independent T-DNA insertion lines, named fla18-1 and fla18-2, display short and swollen lateral roots (LRs) when grown on sensitizing condition of high-sucrose containing medium. Unlike fla4/salt overly sensitive 5 (sos5), previously shown to display short and swollen primary root (PR) and LRs under these conditions, the PR of the fla18 mutants is slightly longer compared to the wild-type. Overexpression of the FLA18 CDS complemented the fla18 root phenotype. Genetic interaction between either of the fla18 alleles and sos5 reveals a more severe perturbation of anisotropic growth in both PR and LRs, as compared to the single mutants and the wild-type under restrictive conditions of high sucrose or high-salt containing medium. Additionally, under salt-stress conditions, fla18sos5 had a small, chlorotic shoot phenotype, that was not observed in any of the single mutants or the wild type. As previously shown for sos5, the fla18-1 and fla18-1sos5 root-elongation phenotype is suppressed by abscisic acid (ABA) and display hypersensitivity to the ABA synthesis inhibitor, Fluridon. Last, similar to other cell wall mutants, fla18 root elongation is hypersensitive to the cellulose synthase inhibitor, Isoxaben. Altogether, the presented data assign a new role for FLA18 in the regulation of root elongation. Future studies of the unique vs. redundant roles of FLA proteins during root elongation is anticipated to shed a new light on the regulation of root architecture during plant adaptation to different growth conditions.

Abscisic Acid Promotes γ-Aminobutyric Acid Accumulation in Soybean Germinating Under Hypoxia-NaCl Stress

γ-aminobutyric acid is a nonprotein amino acid that accumulates in plants under stress. Abscisic acid is important for stress regulation via modulation of γ-aminobutyric acid function. Our results show that the expression of glutamate decarboxylase but not diamine oxidase gene is upregulated in germinating soybean treating treated with exogenous abscisic acid. There was a concomitant increase in glutamate decarboxylase and diamine oxidase activities and putrescine and spermine contents with a decrease in glutamate. These changes were abrogated by fluridone, an inhibitor of abscisic acid synthesis. In conclusion, abscisic acid treatment increases γ-aminobutyric acid accumulation by upregulating diamine oxidase gene expression and activation of glutamate decarboxylase and diamine oxidase activity in germinating soybean under hypoxia-salt stress.

The role of ABA in the responses of wild-type and abscisic acid mutants of Arabidopsis thaliana to excess zinc

To explore the function of endogenous abscisic acid (ABA) in answer to excess zinc (Zn), calli from Arabidopsis (Arabidopsis thaliana) mutant lines, abi1-1 (ABA insensitive) and aba1-1 (with reduced level of ABA), and their associated wild-type (Landsberg erecta, Ler) were used. Excess Zn resulted in oxidative damage characterized by ion leakage, malondialdehyde (MDA), and hydrogen peroxide (H2O2) accumulation, and relative cell viability decrease in Arabidopsis calli. ABA-deficient mutant (aba-1) callus suffered more serious oxidative damage in comparison with wild-type (Ler) callus. Sodium tungstate, an ABA synthesis inhibitor, aggravated excess Zn-induced ion leakage, H2O2 and MDA accumulation in wild-type and aba1-1 mutant calli. Also, sodium tungstate obviously suppressed proline production and ascorbate peroxidase (APX) activity whereas promoted lipoxygenase (LOX) activity in wild-type and aba1-1 mutant calli in the presence of excess Zn. Instead, exogenous ABA treatment significantly elevated proline concentration and APX activity and inhibited LOX activity and thus alleviated excess Zn-induced ion leakage, H2O2 and MDA accumulation, and relative cell viability decrease in wild-type and aba-1 mutant calli. Moreover, no significant effects of sodium tungstate or exogenous ABA application on calli derived from abi1-1 mutant were observed under excess Zn. Endogenous ABA might function as a signal in exciting antioxidases and proline accumulation and thus protect against toxicity induced by excess Zn.

Constitutive expression of a group 3 LEA protein from Medicago falcata (MfLEA3) increases cold and drought tolerance in transgenic tobacco.

MfLEA3 is involved in protection of catalase activity and confers multiple abiotic stress tolerance. Late embryogenesis abundant (LEA) proteins are involved in plant growth, development and abiotic stress tolerance. A member of group 3 LEA proteins from Medicago sativa subsp. falcata (L.) Arcang, MfLEA3, was investigated in the study. MfLEA3 transcript was induced in response to cold, dehydration, and abscisic acid (ABA), while the cold-induced transcript of MfLEA3 was blocked by pretreatment with inhibitor of ABA synthesis. Constitutive expression of MfLEA3 led to enhanced tolerance to cold, drought, and high-light stress in transgenic tobacco plants. Compared to accumulated reactive oxygen species (ROS) in the wild-type in response to treatments with low temperature, drought, and high light, ROS were not accumulated in transgenic plants. Superoxide dismutase, catalase (CAT), and ascorbate-peroxidase activities were increased in all plants after treatments with the above stresses, while higher CAT activity was maintained in transgenic plants compared with wild-type. However, transcript level of CAT-encoding genes including CAT1, CAT2, and CAT3 showed no significant difference between transgenicplants and wild-type, indicating that the higher CAT activity was not associated with its gene expression. ABA sensitivity and transcripts of several ABA and stress-responsive genes showed no difference between transgenic plant and wild-type, indicating that ABA signaling was not affected by constitutive expression of MfLEA3. The results suggest that MfLEA3 may be involved in the protection of CAT activity and confers multiple abiotic stress tolerance.

Effects of root abscisic acid on Na+ transport and photosystem 2 in Helianthus tuberosus under salt stress

The effects of root abscisic acid (ABA) signal on Na+ transport and photosystem 2 (PS2) in Jerusalem artichoke (Helianthus tuberosus) under salt stress (150 mmol·L-1 NaCl) were examined by applying ABA synthesis inhibitor sodium tungstate to roots. Sodium tungstate inhibited ABA synthesis in roots, reduced root Na+ efflux, and increased the efficiency of Na+ transport from roots to leaves under salt stress. Salt stress increased leaf Na+ content and did not affect leaf membrane lipid peroxidation, PS2 reaction center protein and PS2 maximum photochemical efficiency (Fv/Fm ). The inhibition on root ABA synthesis significantly increased leaf Na+ accumulation, aggravated leaf membrane lipid peroxidation, impaired PS2 reaction center protein, decreased Fv/Fm, and induced PS2 photoinhibition. In conclusion, root ABA signal was beneficial to reducing leaf Na+ accumulation and preventing PS2 oxidative damage by inducing root Na+ efflux and inhibiting Na+ transport to the aerial part in H. tuberosus under salt stress.

Hydrogen-induced tolerance against osmotic stress in alfalfa seedlings involves ABA signaling

This study investigated the detailed mechanism underlying the alleviation of osmotic stress by exogenous hydrogen (H2) in Medicago sativa. By using biochemical and molecular approaches, the experiments were performed with the analyses of biomass, relative water content (RWC), lipid peroxidation, abscisic acid (ABA) content, antioxidant activities, and related gene expression profiles. H2 application stimulated ABA production, which was accompanied by the regulation of ABA biosynthesis and deactivation/activation genes. Elevated H2-induced ABA synthesis was sensitive to tungstate, an inhibitor of ABA synthesis. Meanwhile, H2-alleviated osmotic stress, which was supported by the increases in biomass and RWC, and the reduction of lipid peroxidation, was impaired by the inhibition of ABA synthesis. Consistently, tungstate blocked H2-induced antioxidant defense. Molecular evidence revealed that miR528 was down-regulated by H2, showing a negative correlation with its target gene POD2. When tungstate was added together, the decreased miR528 and increased POD2 transcripts were respectively blocked. Transcriptional factor genes involved in ABA signaling, including MYB102, MYC2, and ABF/AREB2, were differentially upregulated by H2, but further impaired by the co-incubation with tungstate. Collectively, our results suggested the possible role of ABA signaling in exogenous H2-mediated tolerance against osmotic stress in alfalfa.

Overexpression of the UGT76E12 gene modulates seed germination, growth, and response to NaCl, mannitol, and abscisic acid

Seed germination and following seedling growth are largely affected by environmental conditions. However, the genes involved in adaptations to these conditions are largely unknown. In this study, we cloned and characterized an Arabidopsis uridine diphosphate glycosyltransferase gene UGT76E12 and investigated its function in seed germination and plant growth under adverse environments. We found that UGT76E12 gene expression was induced by NaCl, mannitol, and abscisic acid (ABA) treatments. Under these treatments, the UGT76E12 overexpression lines exhibited a delayed seed germination and cotyledon growth compared with a wild type, and this delayed growth could be restored by treatment with sodium tungstate, an ABA synthesis inhibitor. Further, real-time quantitative PCR analysis reveals that the stress-induced expressions of genes involved in ABA biosynthesis were greatly enhanced in the UGT76E12 overexpression lines. Therefore, our data suggest that the UGT76E12 gene plays an important role in regulation of seed germination and growth under adverse environmental conditions by affecting ABA biosynthesis.

Root Abscisic Acid Contributes to Defending Photoinibition in Jerusalem Artichoke (Helianthus tuberosus L.) under Salt Stress.

The aim of the study was to examine the role of root abscisic acid (ABA) in protecting photosystems and photosynthesis in Jerusalem artichoke against salt stress. Potted plants were pretreated by a specific ABA synthesis inhibitor sodium tungstate and then subjected to salt stress (150 mM NaCl). Tungstate did not directly affect root ABA content and photosynthetic parameters, whereas it inhibited root ABA accumulation and induced a greater decrease in photosynthetic rate under salt stress. The maximal photochemical efficiency of PSII (Fv/Fm) significantly declined in tungstate-pretreated plants under salt stress, suggesting photosystem II (PSII) photoinhibition appeared. PSII photoinhibition did not prevent PSI photoinhibition by restricting electron donation, as the maximal photochemical efficiency of PSI (ΔMR/MR0) was lowered. In line with photoinhibition, elevated H2O2 concentration and lipid peroxidation corroborated salt-induced oxidative stress in tungstate-pretreated plants. Less decrease in ΔMR/MR0 and Fv/Fm indicated that PSII and PSI in non-pretreated plants could maintain better performance than tungstate-pretreated plants under salt stress. Consistently, greater reduction in PSII and PSI reaction center protein abundance confirmed the elevated vulnerability of photosystems to salt stress in tungstate-pretreated plants. Overall, the root ABA signal participated in defending the photosystem’s photoinhibition and protecting photosynthesis in Jerusalem artichoke under salt stress.

Phytohormones, Isoprenoids, and Role of the Apicoplast in Recovery from Dihydroartemisinin-Induced Dormancy of Plasmodium falciparum.

Many organisms undergo dormancy as a stress response to survive under unfavorable conditions that might impede development. This is observed in seeds and buds of plants and has been proposed as a mechanism of drug evasion and resistance formation in Plasmodium falciparum We explored the effects of the phytohormones abscisic acid (ABA) and gibberellic acid (GA) on dihydroartemisinin (DHA)-induced dormant erythrocytic stages of P. falciparum parasites. Dormant ring stages exposed to ABA and GA recovered from dormancy up to 48 h earlier than parasites exposed to DHA alone. Conversely, fluridone, an herbicide inhibitor of ABA synthesis, blocked emergence from dormancy. Additionally, the role of the apicoplast was assessed in dormant parasite recovery. Apicoplast-deficient P. falciparum remained viable for up to 8 days without the organelle and recrudesced only when supplemented with isopentyl pyrophosphate (IPP). IPP was not required for survival in the dormant state. Fosmidomycin inhibition of isoprenoid biosynthesis did not prevent dormancy release from occurring in parasites with an intact apicoplast, but IPP or geranylgeranyl pyrophosphate was needed for complete recrudescence. In addition, the apicoplast and specifically the isoprenoids it produces are essential for recovery of dormant parasites. In summary, ABA and GA have significant effects on dormant parasites, and the phenotypes produced by these phytohormones and the herbicide fluridone also provide a means to explore the mechanism(s) underlying dormancy and the regulatory network that promotes cell cycle arrest in P. falciparum.

Nitric oxide acts downstream of abscisic acid in molybdenum-induced oxidative tolerance in wheat.

Our study first reveals that Mo mediates oxidative tolerance through ABA signaling. Moreover, NO acts downstream of ABA signaling in Mo-induced oxidative tolerance in wheat under drought stress. Nitric oxide (NO) is related to the improvement of molybdenum (Mo)-induced oxidative tolerance. While the function of Mo in abscisic acid (ABA) synthesis and in mediating oxidative tolerance by the interaction of ABA and NO remain to be studied. The -Mo and +Mo treatment-cultivated wheat was separated and subsequently was pretreated with AO inhibitor, ABA synthesis inhibitor, exogenous ABA, NO scavenger, NO donor or their combinations under polyethylene glycol 6000 (PEG)-stimulated drought stress (PSD). The AO activity and ABA content were increased by Mo in wheat under PSD, however, AO inhibitor decreased AO activity, correspondingly reduced ABA accumulation, suggesting that AO involves in the regulation of Mo-induced ABA synthesis. Mo enhanced activities andexpressions of antioxidant enzyme, while these effects of Mo were reversed by AO inhibitor and ABA synthesis inhibitor due to the decrease of ABA content, but regained by exogenous ABA, indicating that Mo induces oxidative tolerance through ABA. Moreover, NO scavenger inhibited activities of antioxidant enzyme caused by Mo and exogenous ABA, but the inhibitions were eliminated by NO donor, indicating that NO is involved in ABApathway in the regulation of Mo-induced oxidative tolerance in wheat under PSD. Finally, we proposed a scheme for the mechanism of Mo-induced oxidative tolerance.

The genetic characteristics in cytology and plant physiology of two wheat (Triticum aestivum) near isogenic lines with different freezing tolerances.

Freezing tolerance in taft plants relied more upon an ABA-independent- than an ABA-dependent antifreeze signaling pathway. Two wheat (Triticum aestivum) near isogenic lines (NIL) named tafs (freezing sensitivity) and taft (freezing tolerance) were isolated in the laboratory and their various cytological and physiological characteristics under freezing conditions were studied. Proplastid, cell membrane, and mitochondrial ultrastructure were less damaged by freezing treatment in taft than tafs plants. Chlorophyll, ATP, and thylakoid membrane protein contents were significantly higher, but malondialdehyde content was significantly lower in taft than tafs plants under freezing condition. Antioxidant capacity, as indicated by reactive oxygen species accumulation and antioxidant enzyme activity, and the relative gene expression were significantly greater in taft than tafs plants. Soluble sugars and abscisic acid (ABA) contents were significantly higher in taft plants than in tafs plants under both normal and freezing conditions. The upregulated expression levels of certain freezing tolerance-related genes were greater in taft than tafs plants under freezing treatment. The addition of sodium tungstate, an ABA synthesis inhibitor, led to only partial freezing tolerance inhibition in taft plants and the down-regulated expression of some ABA-dependent genes. Thus, both ABA-dependent and ABA-independent signaling pathways are involved in the freezing tolerance of taft plants. At the same time, freezing tolerance in taft plants relied more upon an ABA-independent- than an ABA-dependent antifreeze signaling pathway.

The Arabidopsis heat‐intolerant 5 (hit5)/enhanced response to aba 1 (era1) mutant reveals the crucial role of protein farnesylation in plant responses to heat stress

Protein farnesylation is a post-translational modification known to regulate abscisic acid (ABA)-mediated drought tolerance in plants. However, it is unclear whether and to what extent protein farnesylation affects plant tolerance to high-temperature conditions. The Arabidopsis heat-intolerant 5 (hit5) mutant was isolated because it was thermosensitive to prolonged heat incubation at 37°C for 4d but thermotolerant to sudden heat shock at 44°C for 40min. Map-based cloning revealed that HIT5 encodes the β-subunit of the protein farnesyltransferase. hit5 was crossed with the aba-insensitive 3 (abi3) mutant, the aba-deficient 3 (aba3) mutant, and the heat shock protein 101 (hsp101) mutant, to characterize the HIT5-mediated heat stress response. hit5/abi3 and hit5/aba3 double mutants had the same temperature-dependent phenotypes as hit5. Additionally, exogenous supplementation of neither ABA nor the ABA synthesis inhibitor fluridone altered the temperature-dependent phenotypes of hit5. The hit5/hsp101 double mutant was still sensitive to prolonged heat incubation, yet its ability to tolerate sudden heat shock was lost. The results suggest that protein farnesylation either positively or negatively affects the ability of plants to survive heat stress, depending on the intensity and duration of high-temperature exposure, in an ABA-independent manner. HSP101 is involved in the hit5-derived heat shock tolerance phenotype.

Overexpression of MfPIP2-7 from Medicago falcata promotes cold tolerance and growth under NO3 (-) deficiency in transgenic tobacco plants.

BackgroundPlasma membrane intrinsic proteins (PIPs), which belong to aquaporins (AQPs) superfamily, are subdivided into two groups, PIP1 and PIP2, based on sequence similarity. Several PIP2s function as water channels, while PIP1s have low or no water channel activity, but have a role in water permeability through interacting with PIP2. A cold responsive PIP2 named as MfPIP2-7 was isolated from Medicago falcata (hereafter falcata), a forage legume with great cold tolerance, and transgenic tobacco plants overexpressing MfPIP2-7 were analyzed in tolerance to multiple stresses including freezing, chilling, and nitrate reduction in this study.ResultsMfPIP2-7 transcript was induced by 4 to 12 h of cold treatment and 2 h of abscisic acid (ABA) treatment. Pretreatment with inhibitor of ABA synthesis blocked the cold induced MfPIP2-7 transcript, indicating that ABA was involved in cold induced transcription of MfPIP2-7 in falcata. Overexpression of MfPIP2-7 resulted in enhanced tolerance to freezing, chilling and NO3− deficiency in transgenic tobacco (Nicotiana tabacum L.) plants as compared with the wild type. Moreover, MfPIP2-7 was demonstrated to facilitate H2O2 diffusion in yeast. Higher transcript levels of several stress responsive genes, such as NtERD10B, NtERD10C, NtDREB1, and 2, and nitrate reductase (NR) encoding genes (NtNIA1, and NtNIA2) were observed in transgenic plants as compared with the wild type with dependence upon H2O2. In addition, NR activity was increased in transgenic plants, which led to alterations in free amino acid components and concentrations.ConclusionsThe results suggest that MfPIP2-7 plays an important role in plant tolerance to freezing, chilling, and NO3− deficiency by promoted H2O2 diffusion that in turn up-regulates expression of NIAs and multiple stress responsive genes.Electronic supplementary materialThe online version of this article (doi:10.1186/s12870-016-0814-4) contains supplementary material, which is available to authorized users.

Overexpression of a NF-YC transcription factor from bermudagrass confers tolerance to drought and salinity in transgenic rice.

Nuclear factor Y (NF-Y) is a ubiquitous transcription factor formed by three distinct subunits, namely NF-YA, NF-YB and NF-YC. A stress-responsive cDNA of NF-YC (Cdt-NF-YC1) was isolated from triploid bermudagrass (Cynodon dactylon×Cynodon transvaalensis), and its role in abiotic stress tolerance was investigated in this study. Cdt-NF-YC1 transcript was detected in all vegetative tissues with higher levels being observed in roots. Transcription of Cdt-NF-YC1 in leaves was induced by dehydration, salinity, and treatments with abscisic acid (ABA), hydrogen peroxide (H2 O2 ) or nitric oxide (NO), but not altered by cold. The dehydration- or salt-induced transcription of Cdt-NF-YC1 was blocked by inhibitor of ABA synthesis and scavenger of H2 O2 or NO, indicating that ABA, H2 O2 and NO were involved in the dehydration- and salt-induced transcription of Cdt-NF-YC1. Overexpression of Cdt-NF-YC1 resulted in elevated tolerance to drought and salt stress and increased sensitivity to ABA in transgenic rice. Transcript levels of stress/ABA responsive genes (OsLEA3, OsRAB16A, OsLIP9 and OsP5CS1), ABA synthesis and signalling genes (OsNCED3 and OsABI2), and ABA-independent genes (OsDREB1A, OsDREB1B and OsDREB2A) were substantially higher in transgenic rice than in wild-type plants. The results suggested that that Cdt-NF-YC1 is a good candidate gene to increase drought and salinity tolerance in transgenic rice through modulating gene regulation in both ABA-dependent and ABA-independent pathways.

NFYA1 Is Involved in Regulation of Postgermination Growth Arrest Under Salt Stress in Arabidopsis

The nuclear factor Y (NF-Y), which is a ubiquitous transcription factor found in eukaryotes, is composed of three distinct subunits, namely, NF-YA, NF-YB, and NF-YC. Here, we firstly characterized the detailed function of the Arabidopsis NFYA1 factor. It is found that the 35S::AtNFYA1-overexpressed lines were hypersensitive to salt stress and Abscisic acid (ABA) during the early-postgermination growth stages. The transgenic lines exhibited a severe postgermination growth arrest compared with the wild-type (WT) under salt stress and ABA treatment. Interestingly, sodium tungstate, which is an ABA synthesis inhibitor, restored the salt-sensitive phenotype of the 35S::AtNFYA1 lines. Results of the qRT-PCR analysis showed that the mRNA levels of ABI3 and ABI5, as well as their downstream genes AtEM1 and AtEM6, were more greatly upregulated under salt stress during seed germination in the transgenic lines compared with those in WT. On the other hand, the NFYA1-RNAi lines were found to be insensitive to salt stress and exhibited decreased levels of ABI3, ABI5, EM1, and EM6 transcripts. Our results provide clear evidence supporting a role of AtNFYA1 in regulating postgermination growth arrest under salt stress.

Opening of Iris flowers is regulated by endogenous auxins

Flower opening in Iris (Iris×hollandica) requires elongation of the pedicel and ovary. This moves the floral bud upwards, thereby allowing the tepals to move laterally. Flower opening is requires with elongation of the pedicel and ovary. In cv. Blue Magic, we investigated the possible role of hormones other than ethylene in pedicel and ovary elongation and flower opening. Exogenous salicylic acid (SA) and the cytokinins benzyladenine (N6-benzyladenine, BA) and zeatin did not affect opening. Jasmonic acid (JA) and abscisic acid (ABA) were slightly inhibitory, but an inhibitor of ABA synthesis (norflurazon) was without effect. Flower opening was promoted by gibberellic acid (GA3), but two inhibitors of gibberellin synthesis (4-hydroxy-5-isopropyl-2-methylphenyltrimethyl ammonium chloride-1-piperidine carboxylate, AMO-1618; ancymidol) did not change opening. The auxins indoleacetic acid (IAA) and naphthaleneacetic acid (NAA) strongly promoted elongation and opening. An inhibitor of auxin transport (2,3,5-triodobenzoic acid, TIBA) and an inhibitor of auxin effects [α-(p-chlorophenoxy)-isobutyric acid; PCIB] inhibited elongation and opening. The data suggest that endogenous auxins are among the regulators of the pedicel and ovary elongation and thus of flower opening in Iris.

Delay of Iris flower senescence by cytokinins and jasmonates

It is not known whether tepal senescence in Iris flowers is regulated by hormones. We applied hormones and hormone inhibitors to cut flowers and isolated tepals of Iris × hollandica cv. Blue Magic. Treatments with ethylene or ethylene antagonists indicated lack of ethylene involvement. Auxins or auxin inhibitors also did not change the time to senescence. Abscisic acid (ABA) hastened senescence, but an inhibitor of ABA synthesis (norflurazon) had no effect. Gibberellic acid (GA3 ) slightly delayed senescence in some experiments, but in other experiments it was without effect, and gibberellin inhibitors [ancymidol or 4-hydroxy-5-isopropyl-2-methylphenyltrimethyl ammonium chloride-1-piperidine carboxylate (AMO-1618)] were ineffective as well. Salicylic acid (SA) also had no effect. Ethylene, auxins, GA3 and SA affected flower opening, therefore did reach the flower cells. Jasmonates delayed senescence by about 2.0 days. Similarly, cytokinins delayed senescence by about 1.5-2.0 days. Antagonists of the phosphatidylinositol signal transduction pathway (lithium), calcium channels (niguldipine and verapamil), calmodulin action [fluphenazine, trifluoroperazine, phenoxybenzamide and N-(6-aminohexyl)-5-chloro-1-naphtalenesulfonamide hydrochloride (W-7)] or protein kinase activity [1-(5-isoquinolinesulfonyl)-2-methylpiperazine hydrochloride (H-7), N-[2-(methylamino)ethyl]-5-isoquinolinesulfonamide hydrochloride (H-8) and N-(2-aminoethyl)-5-isoquinolinesulfonamide dihydrochloride (H-9)] had no effect on senescence, indicating no role of a few common signal transduction pathways relating to hormone effects on senescence. The results indicate that tepal senescence in Iris cv. Blue Magic is not regulated by endogenous ethylene, auxin, gibberellins or SA. A role of ABA can at present not be excluded. The data suggest the hypothesis that cytokinins and jasmonates are among the natural regulators.
