Activation of stress reactions in the dinophyte microalga Prorocentrum cordatum as a consequence of the toxic effect of ZnO nanoparticles and zinc sulfate

According to the results of the experimental study, the main regularities of changes in morphological, structural-functional and fluorescent indices of P. cordatum were established when zinc oxide nanoparticles ZnO NPs (0.3–6.4 mg L−1) and Zn in form of salt (0.09–0.4 mg L−1) were added to the medium. The studied pollutants have cytotoxic (growth inhibition, development of oxidative stress, destruction of cytoplasmic organelles, disorganization of mitochondria) and genotoxic (changes in the morphology of nuclei, chromatin condensation) effects on microalgae, affecting almost all aspects of cell functioning. Despite the similar mechanism of action of zinc sulfate and ZnO NPs on P. cordatum cells, the negative effect of ZnO NPs is also due to the inhibition of photosynthetic activity of cells (significant decrease in the maximum quantum yield of photosynthesis and electron transport rate), reduction of chlorophyll concentration from 3.5 to 1.8 pg cell−1, as well as mechanical effect on cells: deformation and damage of cell membranes, aggregation of NPs on the cell surface. Apoptosis-like signs of cell death upon exposure to zinc sulfate and ZnO NPs were identified by flow cytometry and laser scanning confocal microscopy methods: changes in cell morphology, cytoplasm retraction, development of oxidative stress, deformation of nuclei, and disorganization of mitochondria. It was shown that the first signs of cell apoptosis appear at 0.02 mg L−1 Zn and 0.6 mg L−1 ZnO NPs after 72 h of exposure. At higher concentrations of pollutants, a dose-dependent decrease in algal enzymatic activity (up to 5 times relative to control) and mitochondrial membrane potential (up to 4 times relative to control), and an increase in the production of reactive oxygen species (up to 4–5 times relative to control) were observed. The results of the presented study contribute to the disclosure of fundamental mechanisms of toxic effects of pollutants and prediction of ways of phototrophic microorganisms reaction to this impact.

Effect of butachlor on Microcystis aeruginosa: Cellular and molecular mechanisms of toxicity

The rapid development of agriculture increases the release of butachlor into aquatic environments. As a dominant species causing cyanobacterial blooms, Microcystis aeruginosa (M. aeruginosa) can produce microcystin and poses threats to aquatic ecosystems and human health. However, the impact of butachlor on M. aeruginosa remains unclarified. Therefore, the physiochemical responses of M. aeruginosa to butachlor were investigated, and the relevant underlying molecular mechanism was highlighted. There were no significant changes (P > 0.05) in the growth and physiology of M. aeruginosa at the low concentrations of butachlor (0–0.1 mg/L), which evidenced a high level of butachlor tolerance in Microcystis aeruginosa. For the high concentrations of butachlor (4–30 mg/L), the inhibition of photosynthetic activity, disruption of cell ultrastructure, and oxidative stress were dominant toxic effects on M. aeruginosa. Additionally, the impaired cellular integrity and lipid peroxidation may be attributed to the substantial elevations of extracellular microcystin-LR concentration. Downregulation of genes associated with photosynthesis, energy metabolism, and oxidative stress was inferred to be responsible for the growth suppression of M. aeruginosa in 30 mg/L butachlor treatment. The upregulation of gene sets involved in nitrogen metabolism may illustrate the specific effort to sustain the steady concentration of intracellular microcystin-LR. These findings dissect the response mechanism of M. aeruginosa to butachlor toxicity and provide valuable reference for the evaluation of potential risk caused by butachlor in aquatic environments.

Impact of heat priming on heat shock responses in Origanum vulgare: Enhanced foliage photosynthetic tolerance and biphasic emissions of volatiles

Climate change enhances the frequency of heatwaves that negatively affect photosynthesis and can alter constitutive volatile emissions and elicit emissions of stress volatiles, but how pre-exposure to mildly warmer temperatures affects plant physiological responses to subsequent severe heat episodes remains unclear, especially for aromatic plants with high and complex volatile defenses. We studied the impact of heat shock (45 °C/5 min) applied alone and after exposure to moderate heat stress (35 °C/1 h, priming) on foliage photosynthesis and volatile emissions in the aromatic plant Origanum vulgare through 72 h recovery period. Heat stress decreased photosynthesis rates and stomatal conductance, whereas the reductions in photosynthesis were primarily due to non-stomatal factors. In non-primed plants, heat shock-induced reductions in photosynthetic activity were the greatest, but photosynthetic activity completely recovered by the end of the experiment. In primed plants, a certain inhibition of photosynthetic activity remained, suggesting a sustained priming effect. Heat shock enhanced the emissions of volatiles including lipoxygenase pathway volatiles, long-chained fatty acid-derived compounds, mono- and sesquiterpenes, geranylgeranyl diphosphate pathway volatiles, and benzenoids, whereas different heat treatments resulted in unique emission blends. In non-primed plants, stress-elicited emissions recovered at 72 h. In primed plants, volatile emissions were multiphasic, the first phase, between 0.5 and 10 h, reflected the primary stress response, whereas the secondary rise, between 24 and 72 h, indicated activations of different defense metabolic pathways. Our results demonstrate that exposure to mild heat leads to a sustained physiological stress memory that enhances plant resistance to subsequent severe heat stress episodes.

Toxic responses of freshwater microalgae Chlorella sorokiniana due to exposure of flame retardants

Flame retardants, such as Tetrabromobisphenol A (TBBPA), Tris(1,3-dichloro-2-propyl) phosphate (TDCPP) and tributyl phosphate (TBP), are frequently detected in surface water. However, the effects of FRs exposure on aquatic organisms especially freshwater microalgae are still unclear. In this study, the toxicities of TBBPA, TDCPP and TBP to microalgae Chlorella sorokiniana, in terms of growth inhibition, photosynthetic activity inhibition and oxidative damage, were investigated, and according ecological risks were assessed. The results showed that TBBPA, TDCPP and TBP had inhibitory effects on C. sorokiniana, with 96 h EC50 (concentration for 50% of maximal effect) values of 7.606, 41.794 and 49.996 mg/L, respectively. Fv/Fm decreased as the increase of exposure time under 15 mg/L TBBPA. Under 50 mg/L TDCPP and 80 mg/L TBP exposure, Fv/Fm decreased significantly after 24 h. However, Fv/Fm rose after 96 h, indicating that the damaged photosynthetic activity was reversible. The content of chlorophyll a decreased, as the increase of TBBPA concentration from 3 to 15 mg/L. However, chlorophyll a increased first and then decreased, as the increase of TDCPP and TBP concentrations from 0 to 50 mg/L and 0–80 mg/L, respectively. Results indicated that C. sorokiniana could use the phosphorus of TDCPP and TBP to ensure the production of chlorophyll a. The risen content of reactive oxygen species, malondialdehyde as well as superoxide dismutase activity indicated that exposure to FRs induced oxidative stress. Additionally, the risk quotients showed that tested FRs had ecological risks in natural waters or wastewaters. This study provides insights into the toxicological mechanisms of different FRs toward freshwater microalgae for better understanding of according environmental risks.

Elucidating the mechanisms underlying the cytotoxic effects of nano-/micro-sized graphene oxide on the microalgae by comparing the physiological and morphological changes in different trophic modes

Understanding the cytotoxic mechanisms of environmental contaminants is important to estimate their environmental impacts and prepare guidelines for pollution control. Many studies have assessed the cytotoxic mechanisms of graphene oxide (GO), an emerging aquatic contaminant. However, in many cases, the effect of GO size and putative trophic modes of microalgae on cytotoxicity has been neglected, hindering complete understanding of the cytotoxic mechanisms of GO. In this study, the microalga Euglena gracilis cultivated under light (phototrophic) or dark (heterotrophic) conditions was exposed to two sizes of GO [nano-sized (N) and micro-sized (M)] for assessing the effect of GO on microalgal growth. The cytotoxic effect of GO was higher under phototrophic conditions than under heterotrophic conditions, suggesting that a major cytotoxic mechanism of GO is related to photosynthetic activity inhibition. Moreover, N-GO showed higher toxicity than M-GO. The morphological and physiological changes in N-GO- and M-GO-exposed E. gracilis were assessed to further elucidate the cytotoxic mechanisms. N-GO internalized the cells via endocytic activity/piercing, whereas M-GO partially attached to the cell surface and did not enter the cells. Moreover, N-GO and M-GO negatively affected the cells by inducing oxidative stress; the oxidative stress parameters were higher in N-GO-exposed cells than in M-GO-exposed cells.

Growth, ROS accumulation site, and photosynthesis inhibition mechanism of Chlorella vulgaris by triclosan.

Although the addition of triclosan (TCS) in consumer products has been strictly restricted, its continuous applications in hospitals and other medical facilities and its numerous residues still pose a potential risk to aquatic organisms and aquatic ecosystems. In this study, we investigated the growth, biochemical alterations, and physiological responses of Chlorella vulgaris exposed to different concentrations of TCS. The potential toxicity mechanisms associated with excessive production of reactive oxygen species (ROS) and disruption of photosynthetic system II (PSII) were also analyzed. The results indicated that the growth, cellular ultrastructure, and physiology of C. vulgaris were severely affected by TCS in a dose-effect dependent manner. TCS inhibited the growth of C. vulgaris, leading to mitochondria enlargement, the disordering of the arrangement of thylakoids, cell wall rupture, organelles loss, and the cytoplasm lysis. TCS induced severe oxidative damage characterized by ROS accumulation, elevated malondialdehyde (MDA), and up-regulation of antioxidant enzyme activities. Moreover, in TCS-induced algal cells, the main sites of ROS accumulation were chloroplasts, mitochondria, and cell membranes, with ROS accumulating most in the mitochondria. In addition, TCS caused damage to the reaction center (RC inactivation), donor side (OEC damage), and accepted side (electron transport from QA to QB) of PSII in C. vulgaris, leading to inhibition of photosynthetic activity. These results could provide novel insights into the mechanisms of TCS-induced ROS accumulation and photosynthetic inhibition in C. vulgaris, which would contribute to a deep understanding of TCS toxicity on algae.

The humic acid-like substances released from Microcystis aeruginosa contribute to defending against smaller-sized microplastics.

Microplastics (MPs) are ubiquitous in freshwater ecosystems, but knowledge of their effects on extracellular polymeric substance (EPS) produced by algae is poorly understood. The components in specific EPS fractions of Microcystis respond when exposed to MPs is also still unclear. In this study, the responses of Microcystis aeruginosa under polystyrene (PS) microplastic exposure were studied over 17 days of cultivation, using 0.1μm and 1.0μm sized PS at three concentration gradients (1, 10 and 100mg/L). Results indicate that algal growth significantly increased using the 0.1 and 1.0μmPS at a high concentration (100mg/L) on day 17, with growth rates of 74.71%±0.94% and 35.87%±1.23%, respectively. All tested PS had a maximum inhibitory effect on the photosynthesis on day 5, but the inhibition of photosynthetic activity by 0.1μmPS alleviated after 13 days of exposure, indicating recovery of microalgae from the toxic environment. The two PS sizes at 100mg/L concentration triggered EPS release in the latter stage of the experiment; meanwhile, fluorescence EEM analysis showed that smaller-sized PS (0.1μm) at various doses noticeably increased humic acid-like substances in tightly bound EPS (TB-EPS) fractions on day 17. Our findings showed that EPS release and humic acid-like substances secretion of Microcystis likely can resist MPs exposure. The results provide new insights into the toxicity mechanism of MPs on freshwater microalgae, as well as understanding the ecological risks of microplastics.

Organic matter production and recycling in marine biofilm developing on common and new plastics

To face the recent pandemic and comply with international legislation, new plastic objects (surgical masks, nitrile gloves, compostable plastics) have been produced, with a significant increase of their input into the marine environment, together with other common plastics. Given that floating plastic provides a suitable surface for settlement of micro-organism, biofilm accretion was studied in laboratory experiments. The characteristics of biofilm in terms of organic matter production and recycling were evaluated under natural and forced conditions, some of them resembling anthropogenic-affected states (eutrophication) and others environmental variability (darkness and oligotrophy). Under natural conditions, the different plastics, due to their structure and composition, hosted different biofilms. Thicker biofilm was observed on surgical mask and compostable plastic (organic carbon maxima of 35.0 ± 4.7 μg cm−2 and 4.3 ± 0.8 μg cm−2, respectively). Compostable plastic hosted a higher carbohydrate quantity than polyethylene terephthalate, polystyrene and nitrile (on average 8.0 ± 0.8 μg cm−2 vs 3.6 ± 1.6 μg cm−2 for the others). The multi-layer structure of masks and the composition of compostable plastic were the main factors responsible for these differences. Polystyrene and nitrile hosted a higher photoautotrophic biomass, with chlorophyll-a maxima higher than 50 μg cm−2 vs values lower than 10 μg cm−2 for compostable plastic. Inhibition of photosynthetic activity (darkness) allowed a greater biofilm mass, which in natural aphotic zone, may enhance the sinking of plastics. The large availability of carbon (eutrophication) allowed thicker biofilms, providing seawater of additional organic matter load. These biofilms could protect pathogenic organisms, especially on disposable protection equipment, allowing a larger spreading.

Effects of Glyphosate-Based Herbicide on Primary Production and Physiological Fitness of the Macroalgae Ulva lactuca

The use of glyphosate-based herbicides (GBHs) worldwide has increased exponentially over the last two decades increasing the environmental risk to marine and coastal habitats. The present study investigated the effects of GBHs at environmentally relevant concentrations (0, 10, 50, 100, 250, and 500 μg·L−1) on the physiology and biochemistry (photosynthesis, pigment, and lipid composition, antioxidative systems and energy balance) of Ulva lactuca, a cosmopolitan marine macroalgae species. Although GBHs cause deleterious effects such as the inhibition of photosynthetic activity, particularly at 250 μg·L−1, due to the impairment of the electron transport in the chloroplasts, these changes are almost completely reverted at the highest concentration (500 μg·L−1). This could be related to the induction of tolerance mechanisms at a certain threshold or tipping point. While no changes occurred in the energy balance, an increase in the pigment antheraxanthin is observed jointly with an increase in ascorbate peroxidase activity. These mechanisms might have contributed to protecting thylakoids against excess radiation and the increase in reactive oxygen species, associated with stress conditions, as no increase in lipid peroxidation products was observed. Furthermore, changes in the fatty acids profile, usually attributed to the induction of plant stress response mechanisms, demonstrated the high resilience of this macroalgae. Notably, the application of bio-optical tools in ecotoxicology, such as pulse amplitude modulated (PAM) fluorometry and laser-induced fluorescence (LIF), allowed separation of the control samples and those treated by GBHs in different concentrations with a high degree of accuracy, with PAM more accurate in identifying the different treatments.

Effect of Cameraria ohridella feeding on Aesculus hippocastanum photosynthesis

The complex of invasive species of phytophagous insects that can adapt to novel living conditions is constantly increasing. The ecological effect of their vital activity in the new environment is almost impossible to predict. As a result, invasions undesirable in the economic sense are often observed. The horse -chestnut leaf miner (Cameraria ohridella Deschka & Dimič, 1986, Gracillariidae) is one of these problematic invader species in the introduced range of the horse chestnut (Aesculus hippocastanum (Linnaeus, 1753), Sapindaceae). We studied the effect of C. ohridella on the state of the photosynthetic apparatus in Ae. hippocastanum leaves. Photosynthesis is the one of the processes most vulnerable to stress factors, so information about the state of photosynthetic apparatus in a plant under the influence of phytophage feeding obtained with fluorescence analysis can be significant. The feeding effect of C. ohridella caterpillars on the functional state and activity of Ae. hippocastanum photosynthetic apparatus was studied. We studied critical parameters of chlorophyll fluorescence induction reflecting the effect of a single C. ohridella generation (feeding the caterpillar for the five stages of this species’ development) on the functional links of the photosystem II in Ae. hippocastanum leaves. The data obtained show a decrease in PS II quantum efficiency (inhibition of photosynthetic activity) in the leaves from different parts of the crown; it suggests the destructive effect of C. ohridella caterpillar feeding on Ae. hippocastanum photosynthetic apparatus. Values of all key parameters of chlorophyll fluorescence indication evidence inhibition of photophysical and photochemical processes of photosynthesis and impaired coherence of Calvin cycle reactions. Study of the feeding effect of C. ohridella caterpillars on the efficiency of the main enzyme in the Calvin cycle (which closely correlates with the coefficient of fluorescence induction that characterizes the efficiency of dark photosynthetic processes) showed a significant decrease in its activity in the leaves of both the illuminated and shaded parts of the crown. Our study has shown that the method of chlorophyll fluorescence induction allows one to determine the general state of a plant in an express regime by evaluating the main process of plant life as photosynthesis. Analysis of chlorophyll fluorescence parameters is a powerful and effective tool for determining the effect of phytophages on the plant body. The obtained data allow us to apply the method of analyzing chlorophyll fluorescence induction in practice to establish the physiological state of tree flora in forests and garden farms.

Ultraviolet radiation stress response of haploid and diploid spores of Mazzaella laminarioides: Do bio-optical traits matter?

Early stages of macroalgae are known to be more vulnerable to changes in the abiotic environment than their adult phases. However, in algal groups attaining intricate life-cycles, the ecophysiological responses of haploid and diploid propagules have not been sufficiently studied. In the present study, stress responses of haploid tetraspores and diploid carpospores of sub-Antarctic red alga Mazzaella laminarioides exposed to ultraviolet (UV) radiation were studied. We tested the hypothesis that UV stress responses of two types of spores attaining similar size are determined by light use characteristics by comparing the photobiological and bio-optical traits. Fluorescence-based photosynthetic efficiency was measured during a 5- h exposure and after a 5- h recovery. Germination and growth of spores exposed to UV treatments were also assessed. Diploid carpospores exhibited higher chlorophyll a content and higher absorption in the UV band than tetraspores. Although the maximal quantum yield was higher in carpospores than tetraspores, no differences in light demands were detected. Exposure to photosynthetically active radiation (PAR) caused inhibition of photosynthetic activity only in carpospores (43%), while exposure to a combination of PAR + UV reduced the maximal quantum yield significantly in tetraspores (36%) and carpospores (60%). In both types of spores, a recovery from UV stress was observed, allowing their germination and further development. The different responses to radiation found in haploid and diploid propagules highlight the ecophysiological versatility of different life-cycle phases to cope with light stress factors ensuring finally successful recruitment and survival.

Glyphosate-Based Herbicide Toxicophenomics in Marine Diatoms: Impacts on Primary Production and Physiological Fitness

Glyphosate is the main active component of the commercial formulation Roundup®, the most widely used chemical herbicide worldwide. However, its potential high toxicity to the environment and throughout trophic webs has come under increasing scrutiny. The present study aims to investigate the application of bio-optical techniques and their correlation to physiological and biochemical processes, including primary productivity, oxidative stress, energy balance, and alterations in pigment and lipid composition in Phaeodactylum tricornutum, a representative species of marine diatoms, using the case study of its response to the herbicide glyphosate-based Roundup® formulation, at environmentally relevant concentrations. Cultures were exposed to the herbicide formulation representing effective glyphosate concentrations of 0, 10, 50, 100, 250, and 500 μg L−1. Results showed that high concentrations decreased cell density; furthermore, the inhibition of photosynthetic activity was not only caused by the impairment of electron transport in the thylakoids, but also by a decrease of antioxidant capacity and increased lipid peroxidation. Nevertheless, concentrations of one of the plastidial marker fatty acids had a positive correlation with the highest concentration as well as an increase in total protein. Cell energy allocation also increased with concentration, relative to control and the lowest concentration, although culture growth was inhibited. Pigment composition and fatty acid profiles proved to be efficient biomarkers for the highest glyphosate-based herbicide concentrations, while bio-optical data separated controls from intermediate concentrations and high concentrations.

Development of Microalgae Biosensor Chip by Incorporating Microarray Oxygen Sensor for Pesticides Sensing.

A microalgae (Pseudokirchneriella subcapitata) biosensor chip for pesticide sensing has been developed by attaching the immobilized microalgae biofilm pon the microarray dye spots (size 100 μm and pitch 200 μm). The dye spots (ruthenium complex) were printed upon SO3-modified glass slides using a polydimethylsiloxane (PDMS) stamp and a microcontact printer (μCP). Emitted fluorescence intensity (FI) variance due to photosynthetic activity (O2 production) of microalgae was monitored by an inverted fluorescent microscope and inhibition of the oxygen generation rate was calculated based on the FI responses both before and after injection of pesticide sample. The calibration curves, as the inhibition of oxygen generation rate (%) due to photosynthetic activity inhibition by the pesticides, depicted that among the 6 tested pesticides, the biosensor showed good sensitivity for 4 pesticides (diuron, simetryn, simazine, and atrazine) but was insensitive for mefenacet and pendimethalin. The detection limits were 1 ppb for diuron and 10 ppb for simetryn, simazine, and atrazine. The simple and low-cost nature of sensing of the developed biosensor sensor chip has apparently created opportunities for regular water quality monitoring, where pesticides are an important concern.

Inhibition of Photosynthetic Activity in Wastewater-Borne Microalgal–Bacterial Consortia under Various Light Conditions

Microalgal–bacterial consortia are considered an alternative method to conventional wastewater treatment processes with several benefits, such as low oxygen production cost and reduced emission of carbon dioxide resulting from photosynthetic activity. Besides, microalgae effectively remove various emerging contaminants and heavy metals that are hardly removed by conventional wastewater treatment processes. The purpose of this study is finding optimal operation conditions (e.g., light wavelengths, light intensity, microalgal–bacterial consortia biomass) when applying microalgae in wastewater treatment system. Firstly, reduced transmittance was monitored at four different wavelengths (i.e., blue, green, red, and white light) and at various concentrations of microalgal–bacterial consortia. Light transmittance rates were rapidly reduced as the biomass increased, where the highest transmittance was observed in green light. Secondly, the reduction of oxygen production over time, by the inhibition of the photosynthetic activity, was tested as the light intensity increased at four different wavelengths and at low (100 mg L−1) and high (500 mg L−1) concentrations of microalgal–bacterial consortia. The observations and subsequent statistical analyses verify that microalgal–bacterial consortia show the strongest resistance to the inhibition of the photosynthetic activity in green light, with white coming next, when the intensity of light is increased.

Linkage between leaf development and photosynthetic response at hyperosmotic salinity in the C-4 grass Panicum antidotale

This study aimed at elucidating the impact of salinity on the differential responses of the photosynthetic apparatus at different stages of leaf development in the C4 perennial grass Panicum antidotale. Plants were grown for a period of 15 days at hyper-osmotic salinity (up to 400 mM NaCl) or at control conditions (no addition of NaCl to the culture medium). Each day plants were harvested and salt stress effect on biomass production (fresh and dry weight) was recorded. In parallel leaves were divided from leaf tip to leaf base in five segments for analysis of stress effects on the following parameters: chlorophyll fluorescence parameters (Fv/Fm, ΦPSII, ETR), MDA (an indicator of peroxidation of membrane lipids), and ion leakage.A direct correlation between membrane lipid peroxidation and (i) membrane integrity as well as (ii) inhibition of PSII function was found under salinity. In monocots the leaf meristem is located at the base and mature parenchymatic cells preferentially are found at the leaf tip. This may explain our finding that the degree of salt mediated inhibition of photosynthetic activity (ETRcontrol/ETRstressed) was 20-fold higher at leaf tip than at leaf base. Thus, we discuss observed variance in apparent salt resistance in terms of heterogeneity of the photosynthetic apparatus (due to difference in spatial and temporal development) along the leaf lamina.

Multistep approach to control microbial fouling of historic building materials by aerial phototrophs

The scientific multistep approach described herein is a result of two years of research into a control method against microbial fouling and biodeterioration of historic building materials by phototrophs. A series of tests were conducted to select the best antifouling agent for eliminating ‘green’ coatings and protecting surfaces against biofouling. Of the seven active compounds, two with the best penetration abilities were subjected to a photosynthetic activity inhibition test using confocal microscopy. Of the two, a quaternary ammonium salt (QAC) – didecyldimethylammonium chloride (DDAC) – was found to be the most effective. Ten biocides containing QACs at different concentrations were then tested against ‘green’ coatings on wood, brick and plaster, with the best four being selected for further research in model conditions. As a result, biocides containing >14% (v v–1) DDAC were found to be successful antifouling agents for protecting historical materials against biodeterioration by phototrophs.

Changes in soil properties after remediation influence the performance and survival of soil algae and earthworm

Previous research on soil remediation focused on soil properties and not on its effects on soil ecosystems. The present study investigated the adverse effects of soil physicochemical changes due to remediation on the biological indicators Chlorococcum infusionum and Chlamydomonas reinhardtii (algae) and Eisenia andrei (earthworm). Soil physicochemical properties, concentrations of total, bioavailable, and water-soluble heavy metals in soil were measured before and after remediation. Changes in soil pH, electrical conductivity, total nitrogen, and total phosphorous immediately after soil remediation were the primary causes of the biomass and photosynthetic activity inhibition observed in C. infusionum and C. reinhardtii, and the survival, normality, and burrowing behavior decrease observed in E. andrei in remediated soils showing dramatic changes in those properties. These findings suggest that remediated soils need some time to recover before restoring their functions, although heavy metals are no longer contaminating the soil.

Potassium Phosphite and Water Deficit: Physiological Response of Eucalyptus Using Multivariate Analysis

This study aimed to evaluate the physiological response of Eucalyptus citriodora Hook seedlings subjected to foliar application of potassium phosphite and water deficit with the aid of multivariate statistical analysis, using MANOVA and canonical discriminant analysis. The experiment was conducted in a completely randomized design with five treatments (0.0 L c.p. ha-1 of potassium phosphite with irrigation; 0.0; 1.25; 2.50 and 5.00 L c.p. ha-1 of potassium phosphite without irrigation) and six replicates, in a greenhouse located in the municipality of Cruz das Almas, Bahia, Brazil. Treatments with potassium phosphite were applied by foliar spray, using the commercial product Reforce from Agrichem® (25.0% K2O + 35.0% P2O5 p/v). Irrigation was suspended seven days after application of the product. The correlation (0.6603) between the evaluated variables indicated that the use of multivariate analysis techniques was adequate to analyze this data set. Eucalyptus plants of the control treatment without irrigation responded to the water deficit conditions with inhibition of photosynthetic activity and increase of free proline content in the leaves. On the other hand, plants which received foliar application of potassium phosphite at highest concentrations (2.50 and 5.00 L c.p. ha-1), even under water deficit conditions, preserved the photosynthetic activity and proline content in the leaves with values equal to those observed in the irrigated control treatment. From this result it is possible to infer about the role of potassium phosphite as an attenuating effect of the water deficit in Eucalyptus citriodora Hook.

Regulation of PSII function in Cyanothece sp. ATCC 51142 during a light-dark cycle.

Cyanobacteria, as well as green algae and higher plants, have highly conserved photosynthetic machinery. Cyanothece sp. ATCC 51142 is a unicellular, aerobic, diazotrophic cyanobacterium that fixes N2 in the dark. In Cyanothece, the psbA gene family is composed of five members, encoding different isoforms of the D1 protein. A new D1 protein has been postulated in the literature, which blocks PSII during the night and allows the fixation of nitrogen. We present data showing changes in PSII function in cells grown in cycles alternating between 12h of light and dark, respectively, at Cyanothece sp. ATCC 51142. Cyanothece sp. ATCC 51142 uses intrinsic mechanisms to protect its nitrogenase activity in a two-stage process. In Stage I, immediately after the onset of darkness, the cells lose photosynthetic activity in a reversible process, probably by dissociation of water oxidation complex from photosystem II via a mechanism that does not require de novo protein synthesis. In Stage II, a more severe disruption of photosystem II function occurs is in part protein synthesis dependent and it could be a functional signature of the presence of sentinel D1 in a limited number of reaction centers still active or not yet inactivated by the mechanism described in Stage I. This process of inhibition uses light as a triggering signal for both the inhibition of photosynthetic activity and recovery when light returns. The intrinsic mechanism of photosynthetic inactivation during darkness with the interplay of the two mechanisms requires further studies.

Effect of Mercury Chloride on the Chlorophyl a and Pheophytin Content in Marine Microalgae: Measuring the Flow of Autotrophic Phytoplankton Using Sediment Traps Data

The present work aimed to study the influence of mercuric chloride (HgCl2) on pigment content (chlorophyll a and pheophytin) in sediment traps on the basis of experimental data. The marine microalgal cultures of Conticribra weissflogii (Mediophyceae), Tetraselmis viridis (Chlorodendrophyceae), Amphidinium carterae (Dinophyceae), Nannochloropsis sp. (Eustigmatophyceae) were used. The obtained results revealed that the influence of mercury on pigment content as well as on photosynthetic activity was different in different species. The considerable decrease in phytopigments content was observed in Amphidinium carterae and Conticribra weissflogii, while the proportion of pheophytin increased. The pigment content of Tetraselmis viridis was not different from the control line, while for Nannochloropsis sp. the increase in pigment concentration was noticed. The complete immediate inhibition of photosynthetic activity was observed in Conticribra weissflogii, Tetraselmis viridis, Amphidinium carterae, while Nannochloropsis sp. responded only after 24 h of exposition. The pigment degradation rate was almost the same for diatoms and dinoflagellates. The equation for estimating the phytopigments content in sediment traps, that takes into consideration the exposition time, is proposed.
