Kinetics of antibacterial activities of cellulose nanocrystals and their silver-zinc oxide nanocomposites: Application as potential disinfectants

Cellulose nanocrystals (CNCs) were produced from corncob and used as reducing agent in the formation of ZnO/CNCs, Ag/CNCs, and ZnO-Ag/CNCs composites and applied as antibacterial agents for both grampositive and gram-negative bacteria and the kinetics of microbial growth inhibition studied. The CNCs and composites were characterized by UV-visible and Fourier transform-infrared (FTIR) spectroscopy. The characterization results revealed that the functional groups of CNCs were affected by their interactions with Ag+ and Zn2+ ions. The composites at 100 mg/mL displayed activities against Gram-positive bacteria - Bacillus subtilis, Staphylococcus aureus, Staphylococcus epidermis, Bacillus coagulans and Enterococcus faecalis and Gram-negative bacteria - Escherichia coli, Klebsiella pneumonia, Salmonella typhimurium, Pseudomonas aeruginosa, Enterococcus faecium and Acinetobacter baumanii) bacteria with inhibitory zones ranged between 9-46 mm. The kinetics of inhibition showed that the composites treated water had 4 to 28 times lower rate constants compared to the untreated water. This implied that the composites inhibit growth rates and have the potential of disinfecting for between 4 to 28 h depending on the bacteria. The materials showed better growth inhibition rate on gram-positive bacteria than on gram-negative. These composites may have potential applications as disinfectants in personal care products and serve as more eco-friendly alternative disinfectants to chlorophenols.

Low-biofouling membrane bioreactor: Effects of cis-2-Decenoic acid addition on EPS and biofouling mitigation

Biofouling is inevitable in the membrane process, particularly in membrane bioreactors (MBR) combined with activated sludge processes. Regulating microbial signaling systems with diffusible signal factors such as cis-2-Decenoic acid (CDA) can control biofilm formation without microbial death or growth inhibition. This study assessed the effectiveness of CDA in controlling biofouling in membrane bioreactors (MBRs), essential for wastewater treatment. By modulating microbial signaling, CDA mitigated biofilm formation without hindering microbial growth. Analysis using Confocal Laser Scanning Microscopy (CLSM) revealed structural alterations in the biofilm, reducing biomass and thickness upon CDA application. Moreover, examination of extracellular polymeric substances (EPS) highlighted a decrease in total EPS, particularly effective polysaccharides. In addition, the possibility of shifting from high molecular weight EPS to low molecular weight EPS was revealed through the change in dispersion activity. The 56% extension of MBR operational lifespan resulting from the reduction in EPS is anticipated to offer potential cost savings and improved performance. Despite these results, further investigation is crucial to validate any potential environmental risks associated with CDA and to comprehend its long-term effects at various conditions.

Enhancing Dental Material Performance: Tung Oil-Infused Polyurea Microcapsule Coatings for Self-Healing and Antimicrobial Applications.

Within the realm of dental material innovation, this study pioneers the incorporation of tung oil into polyurea coatings, setting a new precedent for enhancing self-healing functionality and durability. Originating from an ancient practice, tung oil is distinguished by its outstanding water resistance and microbial barrier efficacy. By synergizing it with polyurea, we developed coatings that unite mechanical strength with biological compatibility. The study notably quantifies self-healing efficiency, highlighting the coatings' exceptional capacity to mend physical damages and thwart microbial incursions. Findings confirm that tung oil markedly enhances the self-repair capabilities of polyurea, leading to improved wear resistance and the inhibition of microbial growth, particularly against Streptococcus mutans, a principal dental caries pathogen. These advancements not only signify a leap forward in dental material science but also suggest a potential redefinition of dental restorative practices aimed at prolonging the lifespan of restorations and optimizing patient outcomes. Although this study lays a substantial foundation for the utilization of natural oils in the development of medical-grade materials, it also identifies the critical need for comprehensive cytotoxicity assays. Such evaluations are essential to thoroughly assess the biocompatibility and the safety profile of these innovative materials for clinical application. Future research will concentrate on this aspect, ensuring that the safety and efficacy of the materials align with clinical expectations for dental restorations.

Potency of Dimethyl Dicarbonate on the Microbial Inhibition Growth Kinetics, and Quality of Passion Fruit (Passiflora edulis) Juice during Refrigerated Storage.

This study aimed to investigate the effectiveness of dimethyl dicarbonate (DMDC) at various concentrations (0-250 ppm) in inhibiting the growth of Escherichia coli TISTR 117 and spoilage microbes in passion fruit juice (PFJ) and its impact on the physicochemical and antioxidant quality of the juice during refrigerated storage. The highest log reduction in the total viable count, yeast/molds and E. coli was attained in PFJ samples with 250 ppm of DMDC (p ≤ 0.05) added. Microbial growth inhibition by DMDC followed the first-order kinetic model with a coefficient of determination (R2) and inhibition constants (k) ranging from 0.98 to 0.99 and 0.022 to 0.042, respectively. DMDC at 0-250 ppm showed an insignificant effect on pH, °Brix, color (L*, a*, b*), ascorbic acid, total phenolic compound (TPC), total flavonoid content, and antioxidant activity (DPPH, FRAP) (p > 0.05). Control (untreated PFJ), DMDC-250 ppm, and pasteurized (15 s at 72 °C) samples were subjected to 27 days of cold storage at 4 °C. A decreasing trend in pH, total soluble solid, ascorbic acid content, DPPH and FRAP values were observed in all the samples during refrigerated storage. However, the DMDC-250 ppm sample showed a better prospect in physicochemical quality changes compared to the pasteurized and untreated control PFJ samples. ΔE values showed marked changes in the control sample than the DMDC-250 ppm and pasteurized samples at 27 days of storage. Additionally, the total viable count and yeast/mold count were augmented during storage, and an estimated shelf-life of the control, DMDC-250 ppm, and pasteurized samples was approximately 3, 24 and 18 days, respectively. In conclusion, DMDC at 250 ppm could ensure microbial safety without affecting the quality attributes of PFJ during 24 days of storage at 4 °C.

Mango Peels as an Industrial By-Product: A Sustainable Source of Compounds with Antioxidant, Enzymatic, and Antimicrobial Activity.

Plant waste materials are important sources of bioactive compounds with remarkable health-promoting benefits. In particular, industrial by-products such as mango peels are sustainable sources of bioactive substances, with antioxidant, enzymatic, and antimicrobial activity. Appropriate processing is essential to obtain highly bioactive compounds for further use in generating value-added products for the food industry. The objective of the study was to investigate and compare the biological activity of compounds from fresh and dried mango peels obtained by different conventional methods and unconventional extraction methods using supercritical fluids (SFE). The highest total phenolic content (25.0 mg GAE/g DW) and the total content of eight phenolic compounds (829.92 µg/g DW) determined by LC-MS/MS were detected in dried mango peel extract obtained by the Soxhlet process (SE). SFE gave the highest content of proanthocyanidins (0.4 mg PAC/g DW). The ethanolic ultrasonic process (UAE) provided the highest antioxidant activity of the product (82.4%) using DPPH radical scavenging activity and total protein content (2.95 mg protein/g DW). Overall, the dried mango peels were richer in bioactive compounds (caffeic acid, chlorogenic acid, gallic acid, catechin, and hesperidin/neohesperidin), indicating successful preservation during air drying. Furthermore, outstanding polyphenol oxidase, superoxide dismutase (SOD), and lipase activities were detected in mango peel extracts. This is the first study in which remarkable antibacterial activities against the growth of Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa) and Gram-positive bacteria (Bacillus cereus and Staphylococcus aureus) were evaluated by determining the microbial growth inhibition rate after 12 and 24 h incubation periods for mango peel extracts obtained by different methods. Ethanolic SE and UAE extracts from dried mango peels resulted in the lowest minimum inhibitory concentrations (MIC90) for all bacterial species tested. Mango peels are remarkable waste products that could contribute to the sustainable development of exceptional products with high-added value for various applications, especially as dietary supplements.

Comparative Antibacterial Activity of Bharangyadi Yoga Variants w.s.r. to Salmonella Typhi

Salmonella typhi, the causative organism of typhoid fever, remains a major global public health concern. Despite the availability of conventional antibiotics, the emergence of antibiotic-resistant strains of Salmonella typhi poses a serious challenge in combating this infectious pathogen. As a result, the exploration of alternative therapeutic options, such as traditional herbal remedies, has gained prominence. Bharangyadi Yoga is a combination of specific herbal ingredients, used in traditional medicine system to address various types of fever and related ailments as mentioned in Sahasrayogam, has to be prepared and administered in the form of Kashaya. In vitro antibacterial activity of Bharangyadi Kashaya and Bharangyadi Arka was carried out by well diffusion method with different concentrations against Salmonella typhi and the area of inhibition of microbial growth is measured. Moderate zone of inhibition was observed in Arka, while no zone of inhibition was seen in any of the concentration of Kashaya. The antibacterial effectiveness of Bharangyadi Arka can be attributed to its volatile and other aqueous extract content, enhanced solubility and release of volatile active constituents. In this study, Arka is established to have better antibacterial activity compare to Kashaya as a new dosage form. However, Negative antibacterial result of Kashaya does not indicate lack of therapeutic effectiveness.

Synthesis of Novel Nanocomposite Materials with Enhanced Antimicrobial Activity based on Poly(Ethylene Glycol Methacrylate)s with Ag, TiO2 or ZnO Nanoparticles

The aim of this investigation was to prepare novel hybrid materials with enhanced antimicrobial properties to be used in food preservation and packaging applications. Therefore, nanocomposite materials were synthesized based on two stimuli-responsive oligo(ethylene glycol methacrylate)s, namely PEGMA and PEGMEMA, the first bearing hydroxyl side groups with three different metal nanoparticles, i.e., Ag, TiO2 and ZnO. The in situ radical polymerization technique was employed to ensure good dispersion of the nanoparticles in the polymer matrix. FTIR spectra identified the successful preparation of the corresponding polymers and XRD scans revealed the presence of the nanoparticles in the polymer matrix. In the polymer bearing hydroxyl groups, the presence of Ag-NPs led to slightly lower thermal stability as measured by TGA, whereas both ZnO and TiO2 led to nanomaterials with better thermal stability. The antimicrobial activity of all materials was determined against the Gram-negative bacteria E. coli and the Gram-positive S. aureus, B. subtilis and B. cereus. PEGMEMA nanocomposites had much better antimicrobial activity compared to PEGMA. Ag NPs exhibited the best inhibition of microbial growth in both polymers with all four bacteria. Nanocomposites with TiO2 showed a very good inhibition percentage when used in PEGMEMA-based materials, while in PEGMA material, high antimicrobial activity was observed only against E. coli and B. subtilis, with moderate activity against B. cereus and almost absent activity against S. aureus. The presence of ZnO showed antimicrobial activity only in the case of PEGMEMA-based materials. Differences observed in the antibacterial activity of the polymers with the different nanoparticles could be attributed to the different structure of the polymers and possibly the more efficient release of the NPs.

Recent Highlights in Sustainable Bio-Based Edible Films and Coatings for Fruit and Vegetable Applications.

The present review paper focuses on recent developments in edible films and coatings made of base compounds from biological sources, namely plants, animals, algae, and microorganisms. These sources include by-products, residues, and wastes from agro-food industries and sea products that contribute to sustainability concerns. Chitosan, derived from animal biological sources, such as crustacean exoskeletons, has been the most studied base compound over the past three years. Polysaccharides typically constitute no more than 3-5% of the film/coating base solution, with some exceptions, like Arabic gum. Proteins and lipids may be present in higher concentrations, such as zein and beeswax. This review also discusses the enrichment of these bio-based films and coatings with various functional and/or bioactive compounds to confer or enhance their functionalities, such as antimicrobial, antioxidant, and anti-enzymatic properties, as well as physical properties. Whenever possible, a comparative analysis among different formulations was performed. The results of the applications of these edible films and coatings to fruit and vegetable products are also described, including shelf life extension, inhibition of microbial growth, and prevention of oxidation. This review also explores novel types of packaging, such as active and intelligent packaging. The potential health benefits of edible films and coatings, as well as the biodegradability of films, are also discussed. Finally, this review addresses recent innovations in the edible films and coatings industry, including the use of nanotechnologies, aerogels, and probiotics, and provides future perspectives and the challenges that the sector is facing.

The chemical composition, in vitro, and in silico studies of Lavandula mairei essential oil

Lavandula mairei is an aromatic, and medicinal plant endemic to Morocco, mainly found in the southeastern region of the Kingdom, and used in traditional medicine for its many benefits. The purpose of the study was to identify the chemical composition of Lavandula mairei essential oil using Gas Chromatography - Tandem Mass Spectrometry, to evaluate the antimicrobial activity of the plant against bacterial strains (Staphylococcus aureus, Staphylococcus aureus MRSA, Enterococcus faecalis, Escherichia coli ATCC 25922, Escherichia coli ESBL, Klebsiella pneumoniae ATCC 700603), and two fungal strains (Aspergillus niger ATCC 10231, Candida albicans ATCC 16404) in order to measure the microbial growth inhibition zone diameter, and to determine the minimum inhibitory, bactericidal, and fungicidal concentration of the essential oil (MIC, MBC, and MFC). Concerning antioxidant activity, six assays (6DPPH, and 1ABTS Free Radical Scavenging Assay, Total Antioxidant Capacity, Hydroxyl Radical, Reducing Power, and β-Carotene Bleaching Inhibition) were performed to determine the antioxidant potential of the essential oil. For the in silico aspect, molecular docking studies were performed to explore the potential interactions of some compounds with five microbial targets (i) Dihydropteroate synthase (1AJ0), ii) The elongation factor EF-Tu (1OB2), iii) d-alanine ligase (2I80), iv) DNA gyrase (2XCT), v) The cytochrome P450 monooxygenase (5V5Z), and 2ADMET prediction analysis was completed on a compound designated as dehydroabietinol. Carvacrol was identified as the major compound (36.38%) by analysis of the chemical composition, E. coli ATCC 25922 (MIC = 1.87 mg/mL ± 0.00, MBC = 15.00 mg/mL ± 0.00), and Enterococcus faecalis (MIC = 0.23 mg/mL ± 0.00, MBC = 5.53 mg/mL ± 2.77) were the most sensitive to the antibacterial treatment, and Candida albicans was resistant to the essential oil. The highest antioxidant potential of the plant was observed in the β-carotene bleaching inhibition test (IC50 = 1.55 mg/mL ± 0.009 and 90 % of inhibition). In the in silico study, the compound dehydroabietinol provided relevant evidence for molecular docking, and this was confirmed by the ADMET prediction analysis. The results of the study were significant, and further experiments will be necessary to find out more about the biological properties of Lavandula mairei.

Green design and synthesis of some novel thiazolidinone appended benzothiazole-triazole hybrids as antimicrobial agents.

The global increase in bacterial resistance poses a significant threat, jeopardizing the effectiveness of antibiotics. Therefore, the development of new and efficient antimicrobial agents is pre-dominant. Taking this into consideration, in the present study, we designed and reported the facile synthesis of two novel series benzothiazole-triazole and thiazolidinone-appended benzothiazole-triazole hybrids using a molecular hybridization approach in a one-pot process. The synthesized compounds were tested for microbial growth inhibition against bacterial and fungal strains. Among all the synthetics, compounds derived from methoxyphenyl and heteroaromatic ring substitutions exhibited promising inhibitory activity. The current investigation has emphasized that benzothiazole-triazole hybrids incorporating thiazolidinone can be a promising and potent category of molecules with potential biological activities. This sustainable and eco-friendly protocol involves the metal-free, catalyst-free synthesis of bioactive scaffolds, which merges broad tolerance for functional groups with a short reaction time, resulting in good to excellent yields.

Stimulatory effect of ZnO nanoparticles as a nanofertilizer in seed priming of pearl millet (Pennisetum glaucum) and their bioactivity studies

The nutrient-dense cereal crop pearl millet (Pennisetum glaucum) is well known for its exceptional tolerance to extreme heat and water deprivation. Proposing to designate 2023 the International Year of Millets (IYM), the Indian government has received approval from the United Nations General Assembly (UNGA). Nano-fertilizers have emerged as a very promising approach to increase productivity and address the escalating problems in agriculture. Zinc nitrate and Calotropis procera dry leaf extract were utilized in the current experiment to create ZnO NPs. The produced nanoparticles were examined using UV-visible spectroscopy, EDX, SEM, FTIR, and XRD techniques. Using ZnSO4, ZnO NPs, and hydro priming, hybrid pearl millet "HHB 67 improved" was seed primed during the current study. The effects of ZnO NPs were compared with both ZnSO4 and hydro primed seeds. 150 ppm ZnO NPs improved 20 % seed germination and 51 % vigour index in pearl millet under laboratory conditions. Further, the experiment was set in green house, and the agronomic traits of pearl millet like fresh weight, chlorophyll, leaf area, plant height, and dry weight contents (DWCs) etc. were evaluated. Besides, leaves from the treated plant were also tested for DPPH radical scavenging activity and reveled 78.11 % inhibition, which is higher than from ZnSO4 and control treatment. Owing to their significant surface reactivity, zinc oxide nanoparticles (ZnO NPs) were found to be highly effective antibacterial agents, with a MIC of 15 µg/ml against E. coli and S. aureus. This research project's main goal was to evaluate ZnO NPs' enhanced agronomic traits, inhibitory potential, and antioxidative qualities. The aim of this extensive study was to clarify their diverse functions, which include improved agricultural characteristics, suppression of microbial growth, and antioxidant properties. This will facilitate a deeper comprehension of their uses in different fields.

Functional validation of Lobularia maritima thioredoxin-h2 protein for its ability to combat bacterial and fungal infections

Thioredoxins (Trxs) belong to a family of multifunctional redox proteins that is critical for maintaining and regulating the cellular redox environment during plant cell growth. Also, they are important for the development of plant's response to biotic and abiotic stress; however, the specific biological functions of h-type thioredoxins (Trxhs) in plants have not been fully elucidated. Herein, we investigated the role of LmTrxh2, a specific member of the Trxh family, in response to various biotic stress simulants, including mechanical wounding, exposure to jasmonic acid (JA), picolinic acid (PA), salicylic acid (SA), ethephon (ETP), and hydrogen peroxide (H2O2). We observed that LmTrxh2 transcripts were significantly upregulated upon exposure to these stress simulants. The characterization of enzymatic activity revealed that the recombinant LmTrxh2 protein functions as a disulfide reductase. While the role of Trx proteins in redox regulation is well known, their involvement in antimicrobial activity is still unexplored. Therefore, we assessed the antimicrobial effect of LmTrxh2 towards various microorganisms and observed a concentration-dependent inhibition of microbial growth. The minimum inhibitory and minimum bactericidal concentrations and the diameters of the inhibition zones were 40–1250 μg/mL, 40–1250 μg/mL, and 12.5–32.5 mm, respectively. In addition, we used previously developed LmTrxh2-transgenic tobacco lines and found that they showed enhanced resistance to fungal infections triggered by Fusarium graminearum and Aspergillus niger. This resistance was associated with an upregulation of known defense-related genes. Overall, our findings suggest that LmTrxh2 is responsive to multiple biotic stress simulants and plays a critical role in the basal resistance of plants to pathogen infections. These results highlight the potential of LmTrxh2 in the development of strategies to protect crops from various stress factors and emphasize its importance in the adaptation of plants to different stress conditions.

Development of a highly tolerant bacterial consortium for asphaltene biodegradation in soils

Asphaltenes are the most polar and heavy fraction of petroleum, and their complex structure and toxicity make them resistant to biodegradation. The ability to tolerate high asphaltene concentrations is crucial to reducing the toxicity-related inhibition of microbial growth and improving their capacity for adaptation, survival, and biodegradation in soils highly contaminated with asphaltenes. This study developed a highly tolerant consortium for efficient asphaltene biodegradation in soils from 22 bacterial isolates obtained from heavy-crude oil-contaminated soils. Isolates corresponded to the Rhodococcus, Bacillus, Stutzerimonas, Cellulosimicrobium, Pseudomonas, and Paenibacillus genera, among others, and used pure asphaltenes and heavy crude oil as the only carbon sources. Surface plate assays were used to evaluate the tolerance of individual isolates to asphaltenes, and the results showed variations in the extension and inhibition rates with maximum tolerance levels at 60,000 mg asphaltenes l−1. Inhibition assays were used to select non-antagonistic bacterial isolates among those showing the highest tolerance levels to asphaltenes. A consortium made up of the five most tolerant and non-antagonistic bacterial isolates was able to degrade up to 83 wt.% out of 10,000 mg asphaltenes kg−1 in the soil after 52 days. Due to its biological compatibility, high asphaltene tolerance, and ability to utilise it as a source of energy, the degrading consortium developed in this work has shown a high potential for soil bioremediation and is a promising candidate for the treatment of aged soil areas contaminated with heavy and extra-heavy crude oil. This would be the first research to assess and consider extreme bacterial tolerance and microbial antagonism between individual degrading microbes, leading to the development of an improved consortium capable of efficiently degrading high amounts of asphaltenes in soil.

A bidirectional kinetic reaction model to predict uranium distribution in permeable reactive bio-barrier with high-sulfate environment

The use of permeable reactive bio-barriers (Bio-PRBs) is a developing method for remediation of uranium groundwater pollution. However, some remediation effects are difficult to estimate when because of the subsurface environment. Advanced knowledge of uranium migration and reactions in Bio-PRBs is crucial for successful practical application. In this study, a bidirectional kinetic reaction model was developed for predicting uranium reduction in a Bio-PRB system. The research demonstrates that the model is able to predict the uranium migration and rapidly evaluate the Bio-PRBs performance. The results show that contact period and microbial growth are the key factors that affect the remediation performance of Bio-PRBs. Microbial growth could lead to a decrease in hydraulic conductivity (K). The hydraulic conductivity loss in the free microorganisms (FM) group was 0.8–2.3 m/d, which was significantly smaller than the immobilized microorganism (IM) group (0.8–5.2 m/d). Compared to the IM group, the simulation results reveal that longer contact reaction period improves the remediation performance of SO42− and uranium by 32.6% and 21.7%, respectively. The bidirectional reaction between microorganisms and pollutants leads to a decrease in the remediation efficiency. In addition, the model can be used to design standard Bio-PRBs for real field of uranium contanminated groundwater. To meet the remediation goal of groundwater, the width of IM group needs to be increased to 250 cm while 500 cm for FM group. Therefore, IM-PRBs save costs significantly. The model has successfully optimized Bio-PRBs and predicted uranium contaminant-plume evolution and microbial growth inhibition in different Bio-PRBs.

Bioactive compounds from the leaves of Maytenus ilicifolia Mart. ex Reissek: Inhibition of LDL oxidation, glycation, lipid peroxidation, target enzymes, and microbial growth

Ethnopharmacological relevanceMaytenus ilicifolia Mart. ex Reissek, a medicinal plant used for treating gastritis, ulcers, and gastric disorders, possesses therapeutic properties attributed to diverse leaf compounds—terpenoids, alkaloids, flavonoids, phenols, and tannins, reflecting the ethnopharmacological knowledge of traditional users. Aims of the studyWe aimed to assess the antioxidant and antiglycant capacities of Maytenus ilicifolia's ethanolic extract and organic fractions, identify bioactive compounds through HPLC-MS/MS analysis, and conduct phytochemical assessments. We also assessed their potential to inhibit digestive and cholinesterase enzymes, mitigate oxidation of human LDL and rat hepatic tissue, and examine their antimicrobial and cytotoxic properties. Materials and methodsOrganic fractions (hexane - HF-Mi, dichloromethane - DMF-Mi, ethyl acetate - EAF-Mi, n-butanol - BF-Mi, and hydromethanolic - HMF-Mi) were obtained via liquid-liquid partitioning. Antioxidant (DPPH, FRAP, ORAC) and antiglycant (BSA/FRU, BSA/MGO, ARG/MGO/LDL/MGO models) capacities were tested. Phytochemical analysis employed HPLC-MS/MS. We also studied the inhibitory effects on α-amylase, acetylcholinesterase, butyrylcholinesterase, human LDL and rat hepatic tissue oxidation, antimicrobial activity, and cytotoxicity against RAW 264.7 macrophages. ResultsHPLC-ESI-MS/MS identified antioxidant compounds such as catechin, quercetin, and kaempferol derivatives. Ethanolic extract (EE-Mi) and organic fractions demonstrated robust antioxidant and antiglycant activity. EAF-Mi and BF-Mi inhibited α-amylase (2.42 μg/mL and 7.95 μg/mL) compared to acarbose (0.144 μg/mL). Most organic fractions exhibited ∼50% inhibition of acetylcholinesterase and butyrylcholinesterase, rivaling galantamine and rivastigmine. EAF-Mi, BF-Mi, and EE-Mi excelled in inhibiting lipid peroxidation. All fractions, except HMF-Mi, effectively countered LDL oxidation, evidenced by the area under the curve. These fractions protected LDL against lipid peroxidation. ConclusionThis study unveils Maytenus ilicifolia's ethanolic extract and organic fractions properties. Through rigorous analysis, we identify bioactive compounds and highlight their antioxidant, antiglycant, enzyme inhibition, and protective properties against oxidative damage. These findings underline its significance in modern pharmacology and its potential applications in healthcare.

Formulation and Characterization of Hesperidin-Loaded Transethosomal Gel for Dermal Delivery to Enhance Antibacterial Activity: Comprehension of In Vitro, Ex Vivo, and Dermatokinetic Analysis.

In this study, hesperidin was loaded into a transethosome and was developed employing the rotary evaporator method. The formulation was optimized using the Box-Behnken design (BBD). The optimized HSD-TE formulation has a spherical shape, vesicle size, polydispersity index, entrapment efficiency, and zeta potential within the range of 178.98 nm; the PDI was 0.259 with a zeta potential of -31.14 mV and % EE of 89.51%, respectively. The in vitro drug release shows that HSD-TE exhibited the release of 81.124 ± 3.45% in comparison to HSD suspension. The ex vivo skin permeation showed a 2-fold increase in HSD-TE gel permeation. The antioxidant activity of HSD-TE was found to be 79.20 ± 1.77% higher than that of the HSD solution. The formulation showed 2-fold deeper HSD-TE penetration across excised rat skin membranes in confocal laser microscopy scanning, indicating promising in vivo prospects. In a dermatokinetic study, HSD-TE gel was compared to HSD conventional gel where TE significantly boosted HSD transport in the epidermis and dermal layers. The formulation showed greater efficacy than free HSD in the inhibition of microbial growth, as evidenced by antibacterial activity on the Gram-negative and positive bacteria. These investigations found that the HSD-TE formulation could enhance the topical application in the management of cutaneous bacterial infections.

Rational design and characterization of cell-selective antimicrobial peptides based on a bioactive peptide from Crocodylus siamensis hemoglobin

Antimicrobial resistance is a growing health concern. Antimicrobial peptides are a potential solution because they bypass conventional drug resistance mechanisms. Previously, we isolated a peptide from Crocodylus siamensis hemoglobin hydrolysate, which has antimicrobial activity and identified the main peptide from this mixture (QL17). The objective of this work was to evaluate and rationally modify QL17 in order to: (1) control its mechanism of action through bacterial membrane disruption; (2) improve its antimicrobial activity; and (3) ensure it has low cytotoxicity against normal eukaryotic cells. QL17 was rationally designed using physicochemical and template-based methods. These new peptide variants were assessed for: (1) their in vitro inhibition of microbial growth, (2) their cytotoxicity against normal cells, (3) their selectivity for microbes, and (4) the mode of action against bacteria using scanning electron microscopy (SEM), transmission electron microscopy (TEM) and confocal microscopy. The results indicate that all designed peptides have more potent antimicrobial efficacy than QL17 and IL15 peptides. However, only the most rationally modified peptides showed strong antimicrobial activity and minimal toxicity against normal cells. In particular, IL15.3 (hydrophobicity of 47% and net charge of + 6) was a potent antimicrobial agent (MIC = 4–12 μg/mL; MBC = 6–25 μg/mL) and displayed excellent selectivity for microbes (cf. human cells) via FACS assays. Microscopy confirmed that IL15.3 acts against bacteria by disrupting the cell membrane integrity and penetrating into the membrane. This causes the release of intracellular content into the outer environment leading to the death of bacteria. Moreover, IL15.3 can also interact with DNA suggesting it could have dual mode of action. Overall, a novel variant of QL17 is described that increases antimicrobial activity by over 1000-fold (~ 5 μg/mL MIC) and has minimal cytotoxicity. It may have applications in clinical use to treat and safeguard against bacteria.

Green synthesis of Multifunctional Zinc oxide Nanoparticles from Cordia myxa gum; and their Catalytic Reduction of Nitrophenol, Anticancer and Antimicrobial Activity

In situ exfoliated natural polysaccharide Cordia myxa (CMX) is used to promote the utilization of zinc-oxide nanoparticles for eco-friendly catalytic hydrogenation of p-nitrophenol (p-NP) and microbial growth inhibition. Polysaccharide-mediated biosynthetic nanocomposite materials are interesting because they are cheap, green, and environmentally friendly. This study uses CMX gum as a bioreduction to produce multifunctional, environmentally friendly zinc-oxide nanocomposites (ZnO NPs). The process involves a low reaction time and temperature and utilizes CMX as a reducing and stabilizing agent. The structural, morphological, and optical properties of the CMX-ZnO nanocomposite were characterized. The biosynthetic CMX-ZnO NPs exhibited robust catalytic activity and recycling capacity for rapidly oxidizing hazardous p-NPs. The complete reduction of 4-NP to CMX-ZnO NPs in excess NaBH4 was achieved within 15 min, with recyclability and pseudo-first-order kinetics with a rate constant of 0.2571 min−1. Additionally, human colon cancer (HCT116) and 3T3L1 cell lines were remarkably sensitive to the cytotoxic effects of ZnO nanoparticles. CMX-ZnO NPs exhibited potent antibacterial properties against human pathogenic gram-positive and gram-negative bacteria (Bacillus, Salmonella, E. coli, and Pseudomonas aeruginosa) based on the zone of inhibition measured by the disc-diffusion method. The significant antibacterial activity of CMX-ZnO NPs can overcome the current limitations associated with removing water-soluble organic pollutants and microbiological contaminants for long-term environmental sustainability.

A novel formulation of triethyl orthoformate mediated durable, smart and antibacterial chitosan cross-linked cellulose fabrics

Antibacterial, durable and smart cotton fabrics was developed using chitosan-based formulation. The cellulose was covalently cross-linked with chitosan using TEOF. The antibacterial activity of prepared smart fabrics and CS was studied against S. aureus and E. coli strains. The FTIR, SEM and XRD were employed to confirm the linkage of CS molecules with cellulose in cotton fabrics. The CS of 160 KDa extracted from shrimp shell showed the optimum antibacterial activity. The prominent asymmetric, symmetric alkyl CH peaks of CS were shifted to 2930 and 2845 (cm−1), respectively. Moreover, the shifted peaks at 1590 and 1400 (cm−1) indicate the CO stretching and NH2 bending bands of CS, respectively. This confirm the existence of new imine functional group that was generated after cross-linking of NH2 groups of CS. The SEM results showed more uniform morphology of TEOF cross-linked fabrics versus CS coated fabrics, which revealed a promising microbial growth inhibition activity. The TEOF as a cross-linker has been unveiled, showcasing the effectiveness of this innovative crosslinking approach. The fabric treated with cross-linked CS exhibited remarkable antibacterial properties that endured even after undergoing 30 washing cycles. These antibacterial textiles possess substantial commercial potential across a diverse range of industries.
