Light-Driven Electron Uptake from Nonfermentative Organic Matter to Expedite Nitrogen Dissimilation by Chemolithotrophic Anammox Consortia.

Nonphotosynthetic microorganisms are typically unable to directly utilize light energy, but light might change the metabolic pathway of these bacteria indirectly by forming intermediates such as reactive oxygen species (ROS). This work investigated the role of light on nitrogen conversion by anaerobic ammonium oxidation (anammox) consortia. The results showed that high intensity light (>20000 lx) caused ca. 50% inhibition of anammox activity, and total ROS reached 167% at 60,000 lx. Surprisingly, 200 lx light was found to induce unexpected promotion of the nitrogen conversion rate, and ultraviolet light (<420 nm) was identified as the main contributor. Metagenomic and metatranscriptomic analyses revealed that the gene encoding cytochrome c peroxidase was highly expressed only under 200 lx light. 15N isotope tracing, gene abundance quantification, and external H2O2 addition experiments showed that photoinduced trace H2O2 triggered cytochrome c peroxidase expression to take up electrons from extracellular nonfermentative organics to synthesize NADH and ATP, thereby expediting nitrogen dissimulation of anammox consortia. External supplying reduced humic acid into a low-intensity light exposure system would result in a maximal 1.7-fold increase in the nitrogen conversion rate. These interesting findings may provide insight into the niche differentiation and widespread nature of anammox bacteria in natural ecotopes.

Responses of anammox process to elevated Fe(III) stress: Reactor performance, microbial community and functional genes

The aim of present study was to re-evaluate the impacts of elevated Fe(III) stress on anaerobic ammonium oxidation (anammox) process. The results indicated that long-term low concentration Fe(III) (5 and 10 mg/L) exposure significantly improved the nitrogen removal efficiency of anammox process, while high concentration Fe(III) (50 and 100 mg/L) significantly deteriorated the reactor performance. Batch assays showed that the specific anammox activity, heme c content and hydrazine dehydrogenase activity were significantly increased and decreased under low and high concentration Fe(III) exposure, respectively, indicating an enhancement and inhibition of anammox activity. Moreover, the presence of high concentration Fe(III) significantly shifted the anammox community structure. Ca. Brocadia was the predominant anammox genus, whose abundance decreased from 14.26% to 8.13% as Fe(III) concentration increased from 0 to 100 mg/L. In comparison, the abundance of denitrifiers progressively increased from 3.70% to 6.68% with increasing Fe(III) concentration. These suggested that different functional bacteria differed in their responses to Fe(III) stress. Furthermore, long-term Fe(III) exposure significantly up-regulated the abundances of genes associated with nitrogen metabolism and Fe(III) reduction. Overall, the obtained findings are expected to advances our understanding of the responses of anammox process to elevated Fe(III) stress.

Transcriptomics Uncovers the Response of Anammox Bacteria to Dissolved Oxygen Inhibition and the Subsequent Recovery Mechanism.

Understanding the recovery of anaerobic ammonium-oxidizing (anammox) bacteria after inhibition by dissolved oxygen (DO) is critical for the successful applications of anammox-based processes. Therefore, the effects of oxygen exposure (2 mg L-1 DO for 90 min) and subsequent recovery treatments [N2 purging or nano zero-valent iron (nZVI) addition] on the activity and gene expression in a Kuenenia stuttgartiensis enrichment culture were examined. Combining the self-organizing map clustering and enrichment analysis, we proposed the oxidative stress response of anammox bacteria based on the existing concepts of oxidative stress in microbes: the DO exposure triggered a stringent response in K. stuttgartiensis, which downregulated the transcription levels of genes involved in the central metabolism and diverted energy to a flagellar assembly and metal transport modules; these changes possibly promoted survival during the inhibition of anammox activity. According to the cotranscription with central catabolism genes, putative reactive oxygen species (ROS) scavenger genes (kat and sod) were presumed to detoxify the anammox intermediates rather than ROS. In addition, both activity and mRNA profiles with appropriate amount of nZVI addition (5 and 25 mg L-1) were close to that of control, which proved the effectiveness of nZVI addition in anammox recovery. These results would be relevant to the physio-biochemistry development of anammox bacteria and further enhancement of nitrogen removal in wastewater treatment.

Build the expressway for the salt-tolerant anammox process: Acclimation strategy tells the story

The wide existence of saline wastewater has attracted public attention due to its environmental destructiveness. The potential of anammox to treat saline wastewater was systematically evaluated in this study. Three bioreactors with different salinity increasing strategies (R1: inhibition kinetic; R2: gradient increasing; R3: pulsed increasing) were operated to identify the optimal acclimation mode. The results showed that the inhibition of anammox activity by salinity was mainly caused by the loss of enzyme activity. Under the condition of 25.0 g NaCl L−1, the highest nitrogen removal rate of R3 (2.36 ± 0.14 kg N m−3 d−1) indicated that the pulsed strategy might be optimal. Changes in microbial community might be the primary reason lead to different acclimatization results. The relative abundance of anammox bacteria increased by 37.19% in R1 and by 46.81% in R3, but remained stable in R2 with increasing salinity. Dynamic varations in bacterial interactions, proteins, and functional genes revealed the potential resistance mechanisms of bacteria to salinity. The present work provides a novel approach and guidance for the treatment of nitrogen-rich saline wastewater by the anammox process.

Inhibition of anammox activity by phenol: Suppression effect, community analysis and mechanism simulation

Anammox is a distinctive bacterium that is extremely sensitive to and inhibited by organic matter. This weakness becomes a limiting factor for the application of anammox to treat wastewater especially that the underlying mechanisms in the inhibition of phenol remain unclear. In this study, phenol was chosen to determine the possible factors leading to the inhibition of anammox through community variation and molecular simulations. Results of the short-time batch experiment showed that 400 mg L−1 or 1000 mg L−1 of phenol immediately inhibited the anammox activity. Profiles from 16S rRNA sequences showed that the community structures remained unchanged during the first four hours. Molecular simulation between phenol and the key enzyme hydrazine synthase (HZS) showed that phenol combined with HZS and even had the ability to competitively bind with HZSα-haem αI and HZSγ-haem γI sites.
