Altered profile of basket cell afferent synapses in hyper‐excitable dentate gyrus revealed by optogenetic and two‐pathway stimulations

Cholecystokinin (CCK-) positive basket cells form a distinct class of inhibitory GABAergic interneurons, proposed to act as fine-tuning devices of hippocampal gamma-frequency (30-90 Hz) oscillations, which can convert into higher frequency seizure activity. Therefore, CCK-basket cells may play an important role in regulation of hyper-excitability and seizures in the hippocampus. In normal conditions, the endogenous excitability regulator neuropeptide Y (NPY) has been shown to modulate afferent inputs onto dentate gyrus CCK-basket cells, providing a possible novel mechanism for excitability control in the hippocampus. Using GAD65-GFP mice for CCK-basket cell identification, and whole-cell patch-clamp recordings, we explored whether the effect of NPY on afferent synapses to CCK-basket cells is modified in the hyper-excitable dentate gyrus. To induce a hyper-excitable state, recurrent seizures were evoked by electrical stimulation of the hippocampus using the well-characterized rapid kindling protocol. The frequency of spontaneous and miniature excitatory and inhibitory post-synaptic currents recorded in CCK-basket cells was decreased by NPY. The excitatory post-synaptic currents evoked in CCK-basket cells by optogenetic activation of principal neurons were also decreased in amplitude. Interestingly, we observed an increased proportion of spontaneous inhibitory post-synaptic currents with slower rise times, indicating that NPY may inhibit gamma aminobutyric acid release preferentially in peri-somatic synapses. These findings indicate that increased levels and release of NPY observed after seizures can modulate afferent inputs to CCK-basket cells, and therefore alter their impact on the oscillatory network activity and excitability in the hippocampus.

Processing cardiovascular information in the vlPAG during electroacupuncture in rats: roles of endocannabinoids and GABA.

A long-loop pathway, involving the hypothalamic arcuate nucleus (ARC), ventrolateral periaqueductal gray (vlPAG), and the rostral ventrolateral medulla (rVLM), is essential in electroacupuncture (EA) attenuation of sympathoexcitatory cardiovascular reflex responses. The ARC provides excitatory input to the vlPAG, which, in turn, inhibits neuronal activity in the rVLM. Although previous studies have shown that endocannabinoid CB(1) receptor activation modulates gamma-aminobutyric acid (GABA)-ergic and glutamatergic neurotransmission in the dorsolateral PAG in stress-induced analgesia, an important role for endocannabinoids in the vlPAG has not yet been observed. We recently have shown (Fu LW, Longhurst JC. J Appl Physiol; doi:10.1152/japplphysiol.91648.2008) that EA reduces the local vlPAG concentration of GABA, but not glutamate, as measured with high-performance liquid chromatography from extracellular samples collected by microdialysis. We, therefore, hypothesized that, during EA, endocannabinoids, acting through CB(1) receptors, presynaptically inhibit GABA release to disinhibit the vlPAG and ultimately modulate excitatory reflex blood pressure responses. Rats were anesthetized, ventilated, and instrumented to measure heart rate and blood pressure. Gastric distention-induced blood pressure responses of 18 +/- 5 mmHg were reduced to 6 +/- 1 mmHg by 30 min of low-current, low-frequency EA applied bilaterally at pericardial P 5-6 acupoints overlying the median nerves. Like EA, microinjection of the fatty acid amide hydrolase inhibitor URB597 (0.1 nmol, 50 nl) into the vlPAG to increase endocannabinoids locally reduced the gastric distention cardiovascular reflex response from 21 +/- 5 to 3 +/- 4 mmHg. This inhibition was reversed by pretreatment with the GABA(A) antagonist gabazine (27 mM, 50 nl), suggesting that endocannabinoids exert their action through a GABAergic receptor mechanism in the vlPAG. The EA-related inhibition from 18 +/- 3 to 8 +/- 2 mmHg was reversed to 14 +/- 2 mmHg by microinjection of the CB(1) receptor antagonist AM251 (2 nmol, 50 nl) into the vlPAG. Pretreatment with gabazine eliminated reversal following CB(1)-receptor blockade. Thus EA releases endocannabinoids and activates presynaptic CB(1) receptors to inhibit GABA release in the vlPAG. Reduction of GABA release disinhibits vlPAG cells, which, in turn, modulate the activity of rVLM neurons to attenuate the sympathoexcitatory reflex responses.

Glutamate spillover suppresses inhibition by activating presynaptic mGluRs.

Metabotropic glutamate receptors (mGluRs) found on synaptic terminals throughout the brain are thought to be important in modulating neurotransmission. Activation of mGluRs by synaptically released glutamate depresses glutamate release from excitatory terminals but the physiological role of mGluRs on inhibitory terminals is unclear. We have investigated activation of mGluRs on inhibitory terminals within the cerebellar glomerulus, a structure in which GABA (gamma-aminobutyric acid)-releasing inhibitory terminals and glutamatergic excitatory terminals are in close apposition and make axo-dendritic synapses onto granule cells. Here we show that 'spillover' of glutamate, which is released from excitatory mossy fibres, inhibits GABA release from Golgi cell terminals by activating presynaptic mGluRs under physiological conditions. The magnitude of the depression of the inhibitory postsynaptic current is dependent on the frequency of mossy fibre stimulation, reaching 50% at 100 Hz. Furthermore, the duration of inhibitory postsynaptic current depression mirrors the time course of mossy fibre activity. Our results establish that mGluRs on inhibitory interneuron axons sense the activity of neighbouring excitatory synapses. This heterosynaptic mechanism is likely to boost the efficacy of active excitatory fibres by locally reducing the level of inhibition.

Valproate effect on gamma-aminobutyric acid release in pars reticulata of substantia nigra: combination of push-pull perfusion and fluorescence histochemistry.

To study further the previously demonstrated suppressive in vivo effect of sodium valproate (VPA) on gamma-aminobutyric acid (GABA) release in the preoptic area, we examined GABA neurotransmission in substantia nigra (SN), using the push-pull cannula technique in freely moving ovariectomized rats. To clarify whether the area in the substantia nigra that is actually perfused is pars reticulata, known to receive rich GABAergic input from striatum, we used the retrograde fluorescence tracer fast blue (FB) after each perfusion experiment, applying the tracer through the push-pull cannula. Nigral perfusion with VPA caused significant suppression of local GABA release. This effect was more marked in a subgroup of animals showing retrograde labeled cell bodies in striatum, i.e., animals with a tracer application site and therefore also a perfusion site precisely in pars reticulata. Our data suggest that VPA inhibits GABA release in rat SN as it does in the preoptic area, which may be in agreement with our hypothesis of enhanced GABAergic neurotransmission by VPA, causing suppression of presynaptic GABA release through negative feedback actions on the GABA autoreceptor complex. Furthermore, the combination of push-pull cannula technique and retrograde fluorescence tracer application appears to be an important tool to prove afferent connections of the area actually perfused in neuronal networks.

Neural mechanisms of chromatic adaptation in L-type cone horizontal cells of the carp retina.

When a background light is delivered, the responsiveness of horizontal cells to light stimulus initially lowers but subsequently recovers gradually as shown by the increase in response amplitude to test light. These changes of responsiveness are observed when white or close wavelengths are used for both the background and test lights. However, the response amplitude to blue-green test light was initially enhanced but decreased and reached a steady state after the onset of red background illumination. The mechanism causing such a change of test responses was studied in the luminosity-type cone horizontal cells. The initial response enhancement was accompanied by an increase of the slope of hyperpolarizing phase, while the subsequent decrease of response amplitude was caused by the advancement of the recovery phase. The advancement of the recovery phase was eliminated by gamma-aminobutyric acid (GABA) or Co2+ (50 microM), which blocks GABA-induced currents in cone terminals. Dopamine, which inhibits GABA release from horizontal cells, stimulated the advancement of the recovery phase. The time course of the hyperpolarizing phase was not affected by these agents. The enhancement and the subsequent decrease of test responses were also observed in 6-hydroxydopamine-treated retinae. The results suggest that the GABAergic feedback pathway from horizontal cells to cones advances the recovery phase of response to test light. It appears, therefore, that the feedback modifies the responsiveness of horizontal cells to light stimulus under a red background illumination. The mechanism enhancing response amplitude will be also discussed.

Interactions between enkephalin and GABA in avian retina.

In addition to conventional neurotransmitters such as acetylcholine, dopamine, glycine and gamma-aminobutyric acid (GABA), a number of peptide-immunoreactive substances have recently been localized in the vertebrate retina. The functional roles of these retinal peptides and their interactions with conventional neurotransmitters are largely unknown. We have previously shown that exogenous opiates affect both the release of GABA and the firing patterns of ganglion cells in the goldfish retina, and we have now begun a systematic characterization of the opioid pathways in the chicken retina, because, among vertebrate retinas, avian retinas contain the highest concentration of enkephalins. Monoclonal antibodies specific for enkephalin have been used to demonstrate that a subpopulation of enkephalin-containing amacrine cells exists in the chicken retina. This retina also synthesizes Met-enkephalin and releases it on cell depolarization. The enkephalin-induced inhibition of GABA release in goldfish retina led us to examine whether similar interactions occur in chicken, and if so, whether enkephalins and GABA coexist in the same amacrine cells. Our results, presented here, indicate that exogenous enkephalins do indeed inhibit GABA release in the chicken retina. Surprisingly, we found that although some amacrine cells contain both enkephalin and GABA, others contain only one or the other.
