Why is Pain not Characteristic of Inflammation of the Lung Tissue?

The article presents materials that make it possible to understand the reason for the absence of one of the classic signs of inflammatory processes in patients with acute pneumonia. The peculiarities of the functional significance of the lungs for the body are the reason that in the case of inflammation in the tissues of the organ, nature has provided for the presence of a more important adaptive mechanism instead of pain as a signal sign. Understanding the causes of the absence of pain in pneumonia in the initial period, which is most responsible for timely and effective care for these patients, allows us to look at the pathogenesis of the disease from a new point of view, which is of fundamental importance for the correction and selection of pathogenetic means of care.

Platelet-associated NETosis in monitoring inflammation in rheumatoid arthritis and systemic lupus erythematosus

Objective. To assess the possibility of using platelet-associated NETosis parameters to monitor inflammatory activity in rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE).Materials and methods. 50 patients with RA and 29 patients with SLE were examined. The parameters of early and late platelet-associated NETosis were assessed in autologous mixed cultures containing a leukocyte suspension and intact and ADP-stimulated platelets.Results. In patients with RA and SLE in the acute stage, rates of platelet-associated NETosis were increased compared to healthy individuals (p = 0,00001-0,001). During the period of clinical remission, NETosis parameters decreased in comparison with the exacerbation stage (NET-Рst: p=0,017-0,04), in some cases reaching control values. Based on regression analysis, threshold values for platelet-associated NETosis parameters were determined to assess the activity of the inflammatory process in patients with RA and SLE.Conclusion. Parameters of platelet-associated NETosis are informative laboratory markers of the inflammatory process in patients with RA and SLE.

Haemocytometric profile and plasma levels of selected cytokines in patients at various stages of cervical cancer

Introduction and aim. Reports have shown that there is alteration in haematological and inflammatory processes in patients with cervical cancer. However, there is the dearth of information on the pattern of alteration in Nigerian patients with cervical cancer at various stages of the disease. Therefore, haemocytometric profile and plasma levels of interleukin-6 (IL-6) and IL-12 were determined in Nigerian patients with cervical cancer at various stages of the disease. Material and methods. Eighty-nine adults consisting of 49 patients with cervical cancer and 40 apparently healthy controls were enrolled into this study. Haemocytometric profile was determined using automated haematology analyzer while the plasma levels of interleukin-6 (IL-6) and IL-12 were determined using ELISA. Results. Of the participants with cervical cancer, 6.12%, 24.49%, 53.06% and 16.33% were in stages I, II, III and IV respectively. The mean plasma IL-6 level was significantly higher in patients at stage IV of the cancer compared with those in stages I, II and III. No significant differences were observed in the mean plasma IL-12 level, and the haemocytometric profile when patients in different stages of the cancer were compared with one another. Plasma IL-6 had significant positive correlation with the lymphocytes count and cancer stage but had significant negative correlation with packed cell volume (PCV), haemoglobin and total white blood cells count (WBC) in patients with cervical cancer. Conclusion. Interleukin-6 appears to play an important role in the progression of cervical cancer and could be involved in cervical cancer-associated alteration in haemocytometric profile.

Increased Thyroid DPP4 Expression Is Associated With Inflammatory Process in Patients With Hashimoto Thyroiditis.

Dipeptidyl peptidase-4 (DPP4) is originally described as a surface protein in lymphocytes. Lymphocyte infiltration and subsequent destruction of thyroid tissue have been considered as the central pathological mechanism in Hashimoto thyroiditis (HT). The present study aimed to investigate DPP4 expression in peripheral blood and thyroid tissue in HT patients, and explore the role of DPP4 in the pathophysiological process of HT. This case-control study recruited 40 drug-naive HT patients and 81 control individuals. Peripheral blood and thyroid specimens were collected for assessing the expression and activity of DPP4. Moreover, single-cell RNA sequencing (scRNA-seq) analysis of 6 "para-tumor tissues" samples from scRNA-seq data set GSE184362 and in vitro cell experiments were also conducted. The HT patients had similar DPP4 serum concentration and activity as the controls. However, the expression and activity of DPP4 was significantly increased in the thyroid of the HT group than in the control group. The scRNA-seq analysis showed that DPP4 expression was significantly increased in the HT group, and mainly expressed in T cells. Further in vitro studies showed that inhibition of lymphocyte DPP4 activity with sitagliptin downregulated the production of inflammatory factors in co-cultured thyroid cells. DPP4 expression was significantly increased in the thyroid of the HT group compared with the control group, and was mainly localized in the lymphocytes. Inhibition of lymphocyte DPP4 activity reduced the production of inflammatory factors in co-cultured thyroid cells. Therefore, inhibition of DPP4 may have a beneficial effect by alleviating inflammatory reactions in HT patients.

The association between systemic immune-inflammation index and in vitro fertilization outcomes in women with polycystic ovary syndrome: a cohort study

BackgroundAs a novel prognostic and inflammatory marker, the systemic immune-inflammation index (SII) has come to the foreground in recent years. SII may be used as an indicator reflecting the progressive inflammatory process in patients with polycystic ovary syndrome (PCOS). This study aimed to evaluate the correlation between SII and assisted reproductive outcomes in PCOS patients.ResultsA total of 966 women undergoing in vitro fertilization (IVF) procedure with PCOS were included in the study. The SII was calculated as platelet count (/L) × neutrophil count (/L)/lymphocyte count (/L). Participants were divided into four groups according to SII quartiles calculated at baseline, and the differences of clinical and laboratory outcomes between these four groups were compared. Moreover, a univariate linear regression model was used to evaluate the associations between SII and the outcomes. Patients in the highest SII quartile (Q4) had lower antral follicle count (AFC), estradiol (E2), and progesterone (P) levels on the day of human chorionic gonadotropin (HCG) start compared with the lower three SII quartiles (Q1-Q3). Moreover, our analysis demonstrated that women in the lower SII quartiles had a higher rate of available embryos and blastocyst formation compared with those in the highest SII quartile. Logarithm of SII correlated negatively with available embryo rate, but not with number of available embryos. Additionally, the results of our multivariate logistic regression analyses indicated that the highest SII quartile was negatively associated with biochemical pregnancy rate (BPR), clinical pregnancy rate (CPR), live birth rate (LBR), and implantation rate (IR). A non-linear relationship between the SII and number of available embryos, with a negative relationship seen to the right of the inflection point was also found.ConclusionsThe interplay among thrombocytosis, inflammation, and immunity could influence assisted reproductive outcomes in PCOS patients. In this regard, SII may serve as a valuable marker for exploring potential correlations.

Human macrophage immunometabolism regulator (MACIR) in patients with periodontitis

ObjectivePeriodontitis is a local inflammatory reaction caused by bacterial infection in which immune cells, including macrophages, are involved. Recent studies have shown that an important regulator of macrophage function is the human macrophage immunometabolism regulator (MACIR). This gene has been shown to play a key role in modulating the immune response by affecting the activity of fibroblasts and macrophages.In this study, we investigated the expression of MACIR in the gingival tissues of patients with periodontal disease, as well as the effect of IL-1β and TNF-α on the expression of MACIR gene and protein in human gingival fibroblasts. MethodsMACIR mRNA expression in gingival tissue samples was determined using Real-time PCR. Expression of MACIR protein was determined using immunofluorescent staining and western blotting. ResultsThe MACIR mRNA expression in gingival tissue samples in patients with periodontitis was statistically significantly lower than in gingival tissue samples from healthy controls (p = 0.009). The stimulation of human gingival fibroblasts with IL-1β and TNF-α resulted in a statistically significant decrease of MACIR gene mRNA expression. In western blotting and immunofluorescent analysis, we confirmed that the stimulation of the primary culture of human gingival fibroblasts by both IL-1β and TNF-α decreases the expression of MACIR protein. ConclusionThe results of the study suggest that MACIR is an important regulator of the inflammatory process in patients with periodontitis. Decreased expression of the MACIR gene may activate macrophages to secrete mediators that increase inflammation and cause periodontal tissue destruction.

The effect of Lignova sorbent to the concentration of interleukin-6 in the blood and bile in patients with mechanical jaundice of tumors etiology

The OBJECTIVE was to study changes of IL-6 concentration in the blood and bile in patients with MJ of tumor genesis by means of biliary sorption.METHODS AND MATERIALS. The investigation was conducted to study changes in the concentration of IL-6 in the blood and bile against the background of bile sorption in 105 patients with mechanical jaundice (MJ), the cause of obturation was a tumor of the periampullary zone. The patients are divided into two groups. The first group consisted of 51 patients, the second group included 54 patients. All patients of the main group underwent percutaneous perhepatic cholangiostomy for the purpose of decompression; bile sorption was performed using the domestic Lignov enterosorbent. The level of IL-6 in blood serum and bile was determined by enzyme immunoassay.RESULTS. The use of bile sorption in the treatment complex significantly changed the concentration of IL-6 in blood serum (P<0.01) and bile. Determination of IL-6 in blood and bile makes it possible to more objectively assess the degree of cytokine-induced endogenous intoxication in response to ongoing therapeutic measures for tumors of the periampullary zone complicated by mechanical jaundice. The use of bile sorption contributes to a decrease in the concentration of the pro-inflammatory cytokine IL-6 in the blood and bile as an inducer of endogenous intoxication and a cascade of inflammatory processes in patients with tumors of the periampullary zone complicated by mechanical jaundice.CONCLUSION. Bile sorption application helps to decrease proinflammatory cytokine IL-6 concentration in blood and bile as an inductor of endogenous intoxication and inflammatory processes cascade.

Alterations in <I>IL</I>-17 expression in pediatric patients with psoriatic arthritis

Introduction. Psoriatic arthritis is a common inflammatory disease affecting the joints and it is usually accompanied by plaque psoriasis. The pathogenetic link between psoriasis and psoriatic arthritis well reflects the mechanistic hypotheses of disease pathogenesis. Psoriatic arthritis is characterized by chronic inflammation which results in bone erosion and bone loss, as well as new bone formation around the affected joints. The exaggerated inflammatory response leads to enthesitis with the crucial contribution of IL-17 producing T cells and entheseal resident cells, expressing IL-23R. Studying the IL-17 gene expression patterns can help choose a therapy for patients with psoriatic arthritis.Aim. To study alterations in IL-17 gene expression in immune cells of paediatric patients with psoriatic arthritis.Materials and methods. Mono nuclear cells were isolated from the peripheral blood of 45 patients with psoriatic arthritis and 20 healthy controls. The IL-17 gene expression was analysed using a real-time PCR.Results and discussion. Mononuclear cells were isolated from whole peripheral blood for subsequent analysis of IL-17 gene expression by quantitative RT-PCR. The comparative analysis of the expression levels of patients with psoriatic arthritis and healthy volunteers showed that the expression level of IL-17 gene in patients with psoriatic arthritis was 345 times higher than the expression level in healthy volunteers.Conclusion. Patients with psoriatic arthritis are characterized by a very high level of IL-17 gene expression in immune blood cells. The high IL-17 gene expression level confirms its significant role in the inflammatory process in patients with psoriatic arthritis.

Introduction. Sepsis, severe sepsis and septic shock are major public health problems worldwide. The consequences of sepsis are especially unfavorable for people with weakened immunity. Biomarkers of inflammation play an important role in the diagnosis of sepsis: C-reactive protein and procalcitonin, the indices of which increase significantly in this pathology. The aim of the study. To investigate quantitative indices of cytokines and biomarkers of inflammation in patients with peritoneal sepsis with different degrees of severity. Materials and methods. A group under examination included 101 patients. The esteemed laboratory indices included interleukins-1β, -2, -6, -10; tumor necrosis factor-α; C-reactive protein and procalcitonin concentration. Results. In patients with sepsis, the interleukin-1β index exceeded control values by 2.9 times. In the severe sepsis group this same index increased by 1.2 times, and in septic shock – by 1.4 times, compared to patients with sepsis. The concentration of tumor necrosis factor-α in the blood of patients with peritoneal sepsis, severe sepsis, and septic shock was 1.1, 9.2, and 2.9 times higher than control values, respectively. While measuring the concentration of interleukin-6, it was detected this index enhancement in all three groups of septic patients. Also it was documented certain decrease in the interleukin-2 index in all patients under investigation, compared to control. In patients with severe sepsis and septic shock, an increase in the level of interleukin-10 in comparison to control values was estimated up to 4.9 and 5.1 times. An increase in C-reactive protein and procalcitonin registered in all groups of septic patients, as well as the observed cytokine imbalance apparently reflect disability of the immune system to respond adequately and resist pathogenic microorganisms due to the deepening of the sepsis severity. Conclusions. In patients with peritoneal sepsis, the level of interleukin-1β exceeded control values by 2.9 times, in patients with severe sepsis – 3.7 times, and with septic shock – 4.2 times. The concentration of tumor necrosis factor-α in the blood of patients with peritoneal sepsis, severe sepsis, and septic shock was 1.1, 9.2, and 2.9 times higher than control values, respectively. Concentration of interleukin-6 in patients with peritoneal sepsis exceeded by 8.4 times, in the severe sepsis – by 18.8 times, and in septic shock – by 17.4 times control values. In patients with sepsis and severe sepsis of peritoneal genesis, the level of interleukin-2 decreased by 1.85 times, and in the septic shock group – by 1.6 times compared to the control values. Interleukin-10 indices in patients with sepsis increased by 1.28 times, in severe sepsis – by 4.9 times, and in septic shock – by 5.1 times in comparison with control values. An increase in C-reactive protein compared to its control values was detected in all patients under investigation: in the sepsis group – by 77.8 times, in severe sepsis – by 128.1 times, and in patients with septic shock – by 95.7 times. Similar enhancement of procalcitonin levels was observed in all patients under investigation with these indices correlation to the severity of the disease. Indices of pro-inflammatory and anti-inflammatory cytokines, as well as C-reactive protein and procalcitonin can be recommended as a reliable markers of the severity of the inflammatory process in patients with peritoneal sepsis. Digital values of cytokines and inflammatory biomarkers can serve as a valuable additional criteria for the assessment severity of the pathological process (sepsis, severe sepsis, and septic shock) of peritoneal origin. Association between Polyunsaturated Fatty Acid Profile and Bronchial Inflammation in Bronchiolitis Obliterans.

Bronchiolitis obliterans (BO) is a chronic lung disease, which occurs after an insult to the lower airways, in particular after airway infections or after stem cell transplantation, and which results in persistent inflammation. N-3 and n-6 polyunsaturated fatty acids (PUFA) have been shown to influence the inflammatory processes in chronic inflammatory conditions. Since BO is maintained by persistent pulmonary inflammation, a disbalanced n-6/n-3 fatty acid profile could support the inflammatory process in patients with BO and therefore, could become an approach to new therapeutic options. Twenty-five patients with BO (age: 13; 7-39) and 26 healthy controls (age: 19; 7-31) participated in the study. Lung function (forced viral capacity (FVC), forced expiratory volume 1 (FEV1), residual volume (RV)), and lung clearance index (LCI) were measured. Induced sputum was analyzed for cytology and cytokine levels (IL-1ß, IL-6, IL-8, TNF-α) using cytometric bead array (CBA). The PUFA profile was determined in the serum and induced sputum by gas chromatography. Patients presented with significantly lower FVC and FEV1 as well as higher RV and LCI measurements compared to the control group. Apart from a massive airway inflammation indicated by elevated numbers of total cells and neutrophils, the CBA analysis showed significantly increased levels of IL-1β, IL-6, and IL-8. The analysis of PUFA in sputum and serum revealed a significant difference in the ratio between the n-6 PUFA arachidonic acid (AA) and the n-3 PUFA docosahexaenoic acid (DHA) (AA : DHA). Furthermore, the AA : DHA ratio significantly correlated with the inflammatory cytokines in induced sputum. Lung function in BO is significantly impaired and associated with uncontrolled neutrophil-dominated airway inflammation. Furthermore, the imbalance in the AA/DHA ratio in favor of n-6 PUFA demonstrates a pro-inflammatory microenvironment in the cell membrane, which correlates with the inflammatory cytokines in induced sputum and might be an option for an anti-inflammatory therapy in BO.

Study of blood microcirculation in the area of purulent inflammatory process in patients with phlegmon of the maxillofacial region with the analyzer “LASMA PF”

The article is devoted to the assessment of the state of the microcirculatory bed of the skin in the purulent-inflammatory process in patients with phlegmon of the maxillofacial region who were treated in the maxillofacial department of the Moscow Pirogov State Clinical Hospital. Microcirculation parameters were recorded by laser Doppler flowmetry (LDF) with simultaneous temperature recording directly from the skin in the area of purulent-inflammatory infiltrate, by applying the apparatus of the analyzer «LASMA PF» to the patient’s skin. The diagnosis by the analyzer «LAZMA PF» is based on the simultaneous assessment of the activity of the tissue coenzyme of the reduced nicotinamide adenine dinucleotide (NADH) by fluorescence spectroscopy and indicators of microcirculation of blood flow by laser Doppler flowmetry. It is concluded that the differences obtained during the study between microcirculatory disorders in the purulent-inflammatory process and the healthy side indicate the need for drug therapy that regulates blood rheology and the state of the microcirculatory bed.

Morphological and ultrastructural changes in the common bile duct wall in stenting

Aim . To carry out a comprehensive morphological study into pathological changes in the common bile duct wall following various periods of plastic and nitinol stent implantation; to examine telocytes in the common bile duct wall. Materials and methods. The study involves the material of 91 patients, who underwent surgery for periampullary carcinoma from 2014 to 2020. Plastic stents were inserted in 56 cases, nitinol stents – in 35. The study of morphological structure of the common bile duct wall took into account the type of stent and period of its implantation (14–30 days, 31–60 days, ≥ 61 days). Results . Inflammatory processes in patients with nitinol stent implantation have been revealed to prevail over fibroplastic processes, leading to hyperplasia of the common bile duct mucosa. Following plastic stents implantation, fibroplastic and atrophic changes prevailed. It was found that cases with plastic stent implantation are more often subject to damage to the telocyte ultrastructure and loss of intercellular connections. Conclusion . Structural changes in telocytes can reflect the degree of fibrosis in the common bile duct wall depending on the type of stent.

ANALISIS D-DIMER DAN CRP PADA PASIEN COVID-19 DENGAN KOMORBID DIABETES MELLITUS

Severe Acute Respiratory Syndrome Coronavirus 2 (SARS CoV-2) is a virus that attacks and causes respiratory problems with general symptoms of fever accompanied by a dry cough, shortness of breath, and in severe cases. Patient with comorbid diabetes mellitus are in the high-risk category if they get infected with Covid-19, with a 50% risk of death compared to those without diabetes mellitus. In Covid-19 patients, coagulation disorders are often found, and to assess the continuity of the coagulation process, a D-Dimer examination is carried out. Disturbances in coagulation are often associated with inflammatory processes in patients with Covid-19, and to find out, CRP levels are checked. This study aims to determine the relationship between D-dimer and CRP levels in confirmed Covid-19 patients with comorbid diabetes mellitus at the Jakarta Hajj Hospital. The research was conducted in June 2022 with a cross-sectional research design. Data was taken from medical records of patients aged over 25 years who were treated with confirmed Covid-19 with comorbid diabetes mellitus at the Jakarta Hajj Hospital for the period June 2021-September 2021 and obtained data for 80 people. Analysis of D-Dimer levels in this study with confirmed Covid-19 patients with comorbid Diabetes Mellitus were high (0.5 ng/mL) in 50 people (63%) and normal (≤0.5 ng/dL) in 30 people (37%). Serum Levels of 8-Iso-Prostaglandin F2α and Raftlin in Patients with Generalized Anxiety Disorder.

Although there are neurobiological studies of patients with generalized anxiety disorder (GAD), the topic is still open to research. Lipid peroxidation can generate new molecular signal sequences by altering protein amounts and activity. 8-Iso-Prostaglandin F2α (8-iso-PGF2α) is known to be an important lipid peroxidation marker. Raftlin, which is defined as a major lipid raft protein, is important for the regulation of signal transduction and inflammatory processes. Our aim in this study was to compare the 8-iso-PGF2α and Raftlin levels of forty patients diagnosed with GAD and 40 healthy controls (age-sex and body mass index-matched). In the present study, increased serum 8-iso-PGF2α and Raftlin levels were found in patients with GAD compared to healthy controls. To our knowledge, this is the first study to examine 8-iso-PGF2α and Raftlin levels in patients with GAD. These results expand our knowledge of oxidative stress and inflammatory processes in patients with GAD. Our study should be considered preliminary and further studies should be performed with larger sample groups comparing values before and after treatment.

Mediterranean Diet Improves Plasma Biomarkers Related to Oxidative Stress and Inflammatory Process in Patients with Non-Alcoholic Fatty Liver Disease.

Non-alcoholic fatty liver disease (NAFLD) shows liver fat depots without alcohol consumption. NAFLD does not have specific drug therapies, with a healthy lifestyle and weight loss being the main approaches to prevent and treat NAFLD. The aim was to assess the antioxidant and pro-inflammatory state in patients with NAFLD after 12-month-lifestyle intervention depending on the change in adherence to a Mediterranean diet (AMD). Antioxidant and inflammatory biomarkers were measured in 67 adults (aged 40-60 years old) diagnosed with NAFLD. Anthropometric parameters and dietary intake were measured by a validated semi-quantitative 143-item food frequency questionnaire. The nutritional intervention improved anthropometric and biochemical parameters after a 12-month follow-up. However, decreases in alanine aminotransferase (ALT) and C reactive protein (CRP) were higher in participants with high AMD, which also showed higher improvement in physical fitness (Chester step test) and intrahepatic fat contents. The intervention reduced plasma levels of malondialdehyde, myeloperoxidase, zonulin, and omentin, and increased resolvin D1 (RvD1), whereas the decrease in leptin, ectodysplasin-A (EDA), cytokeratin-18 (CK-18), interleukin-1ra (IL-1ra) and endotoxin was only significant in participants with higher AMD. The current study showed that a one-year nutritional intervention improved main NAFLD features such as body mass index, IFC, liver enzymes, and prooxidant and proinflammatory status. There was also a decrease in the concentration of plasmatic endotoxin, suggesting an improvement in intestinal permeability. These health benefits were more evident in participants that improved AMD to a greater extent. The trial was registered at ClinicalTrials.gov with registry number NCT04442620.

Sputum Interleukin-32 in childhood asthma: correlation with IL-1β

_Background:_ Asthma is an airway disorder where inflammatory cytokines are partly responsible for exacerbating the disease. Interleukin (IL)-32, generally referred to as natural killer cell (NK4) transcript 4, is described as an immunoregulator involved in the stimulation of anti-/pro-inflammatory cytokines. The abnormal presence of IL-32 has been observed during inflammatory diseases, particularly in asthma. This study aims to characterize IL-32 in the inflammatory process in patients with severe asthma. _Methods:_ IL-32 and IL-1β levels in the supernatant of induced sputum obtained from 59 asthma patients (mild: 20 patients; moderate: 20 patients; severe: 19 patients) were measured using ELISA. Sputum IL-32 mRNA expression was measured by RT- PCR. _Results:_ Sputum IL-32 was significantly elevated at the protein (_p= 0.00019_) and mRNA expression (_p < 0.0001_) in asthmatics compared to non-asthmatic controls. IL-32 was elevated in severe asthmatic patients (_p < 0.0001_) compared to mild-moderate asthma. Severe asthmatics allergic to house dust mites expressed higher sputum IL-32 levels than severe asthmatics without allergy (_p = 0.0001_). A significant association was found between sputum IL-32 and IL-32 mRNA (r =0.531_; p = 0.0159_) in severe asthma. In the same way, sputum IL-1β and IL-32 were significantly correlated (r = 0.476; _p = 0.0337_). _Conclusion:_ The level of IL-32 in induced sputum may be associated with asthma severity.

This study aims to characterize IL-32 in the inflammatory process in patients with severe asthma. _Methods:_ IL-32 and IL-1β levels in the supernatant of induced sputum obtained from 59 asthma patients (mild: 20 patients; moderate: 20 patients; severe: 19 patients) were measured using ELISA. Sputum IL-32 mRNA expression was measured by RT- PCR. _Results:_ Sputum IL-32 was significantly elevated at the protein (_p= 0.00019_) and mRNA expression (_p < 0.0001_) in asthmatics compared to non-asthmatic controls. IL-32 was elevated in severe asthmatic patients (_p < 0.0001_) compared to mild-moderate asthma. Severe asthmatics allergic to house dust mites expressed higher sputum IL-32 level than severe asthmatics without allergy (_p = 0.0001_). A significant association was found between sputum IL-32 and IL-32 mRNA (r =0.531_; p = 0.0159_) in severe asthma. In the same way, sputum IL-1β and IL-32 were significantly correlated (r = 0.476; _p = 0.0337_). _Conclusion:_ The level of IL-32 in induced sputum may be associated with asthma severity.

Sputum Interleukin-32 in childhood asthma: correlation with IL-1β

Interleukin (IL)-32, generally referred to as natural killer cell (NK4) transcript 4, is described as an immunoregulator involved in the stimulation of anti-/pro-inflammatory cytokines. The abnormal presence of IL-32 has been observed during inflammatory diseases, particularly in asthma. Objective: The definite cause of overactive bladder (OAB), which negatively affects the quality of life of patients, is unknown. This study aims to provide evidence for the inflammatory process in patients with OAB whose etiology has not been fully elucidated. Material and Methods: The study included 154 people with OAB diagnosis and 131 people as controls. This study was conducted retrospectively, cross-sectionally between April 2015 and April 2020. The data were recorded at the first meeting by an expert on this subject. Groups were compared in terms of CRP and ESR measured in serum. Statistical analysis used the chi-square test, independent samples t-test, Mann-Whitney U test, and Pearson and Spearman correlation analyses. P<0.05 was accepted as statistical significance. Results: There were no differences between the groups in terms of comorbid diseases. There were significant differences identified in terms of age and BMI between the groups (p<0.005). When groups are compared according to CRP levels, values were 0.28 [0.54] for the OAB group and 0.17 [0.22] mg/dl for the control group (p=0.047). The distribution according to ESR in the groups was 19 [30.5] and 12.5 [13] mm/h, respectively (p=0.004). Conclusion: This study provides new evidence to the literature showing the relationship between OAB and an inflammatory process. It was determined that CRP and ESR levels were increased in patients with OAB compared to the control group. These results show us that there is an inflammatory process at the onset of this disease. The production of pro-inflammatory cytokines and the state of cellular immunity in patients with pulmonary tuberculosis with and without relapse

The purpose of the study was to conduct a comparative study of the synthesis of pro-inflammatory cytokines, the state of cellular immunity, free radical oxidation reactions and antioxidant protection in patients with pulmonary tuberculosis with and without relapse. Material and methods. The study was carried out on two groups of patients: the first group basic which included 39 patients with relapsed pulmonary tuberculosis (TBR); 2nd control group 39 patients with non-relapsed pulmonary tuberculosis (TB). Pro-inflammatory cytokines (TNF-α, IL-2 IL-6), CD-3 lymphocyte content, lymphocyte blast transformation response to phytohemagglutinin and tuberculin, NBT-test, AOPP, SOD, and catalase were determined in patients of both groups and healthy people. Results. In patients with tuberculosis with relapses, compared with patients without relapses, complaints of cough and dyspnea were more often presented, a longer duration of hospitalization, late abacillation. In patients with relapses, a pronounced suppression of the functional and specific activity of lymphocytes and neutrophils was noted, which is confirmed by high levels of lipid peroxidation (Advanced Oxidation Protein Products AORP) and low levels of the antioxidant system (SOD and catalase). The high content of pro-inflammatory cytokines in patients with relapses indicates a higher activity of the inflammatory process in these patients and corresponds to a low blast formation rate for both phytohemagglutinin and tuberculin. Conclusion. Сhanges in the content of pro-inflammatory cytokines, the state of cellular immunity and the characteristics of reactions of free radical oxidation and antioxidant protection indicate a higher activity of the inflammatory process in patients with pulmonary tuberculosis with relapses compared to patients without relapses, which led to a longer duration of hospitalization, late abacillation.
