Exogenous IAA enhances low potassium tolerance of sweetpotato by regulating root response strategy

ABSTRACT Plant roots are sensitive to potassium (K+) deficiency signals. Therefore, regulating root growth by exogenous methods is a vital strategy to improve low K+ tolerance of sweetpotato. We studied the effects of exogenous indole-3-acetic acid (IAA) on growth, K+ absorption, and root characteristics in sweetpotato exposed to low K+ treatment (LK). LK significantly inhibited dry mass, K+ concentration and accumulation, as well as the root elongation (length) and branching (forks and crossings) in sweetpotato seedlings. However, exogenous IAA increased the length, ratio, and density of lateral roots and promoted absorption and accumulation of K+, which effectively alleviated the inhibitory effect of low K+. Exogenous IAA also increased the expression levels of auxin synthesis (IbYUC6 and IbTAR2) and transport (IbPIN1, IbPIN3, and IbPIN8) genes in leaves and roots, which promoted the increase of endogenous IAA content. Furthermore, exogenous IAA was more effective on low-K-tolerant variety (XS32) than low-K-sensitive variety (NZ1) under LK stress, depending on their different IAA synthesis and transport strategies. These results indicated that exogenous IAA enhanced root responsiveness of sweetpotato to low K+ stress by modulating auxin biosynthesis and transport, thereby improving the tolerance of sweetpotato to low K+ stress.

Boron supply restores aluminum-blocked auxin transport by the modulation of PIN2 trafficking in the root apical transition zone.

The supply of boron (B) alleviates the toxic effects of aluminum (Al) on root growth; however, the mechanistic basis of this process remains elusive. This study filled this knowledge gap, demonstrating that boron modifies auxin distribution and transport in Al-exposed Arabidopsis roots. In B-deprived roots, treatment with Al induced an increase in auxin content in the root apical meristem zone (MZ) and transition zone (TZ), whereas in the elongation zone (EZ) the auxin content was decreased beyond the level required for adequate growth. These distribution patterns are explained by the fact that basipetal auxin transport from the TZ to the EZ was disrupted by Al-inhibited PIN-FORMED2 (PIN2) endocytosis. Experiments involving the modulation of protein biosynthesis by cycloheximide (CHX) and transcriptional regulation by cordycepin (COR) demonstrated that the Al-induced increase of PIN2 membrane proteins was dependent upon the inhibition of PIN2 endocytosis, rather than on the transcriptional regulation of the PIN2 gene. Experiments reporting on the profiling of Al3+ and PIN2 proteins revealed that the inhibition of endocytosis of PIN2 proteins was the result of Al-induced limitation of the fluidity of the plasma membrane. The supply of B mediated the turnover of PIN2 endosomes conjugated with indole-3-acetic acid (IAA), and thus restored the Al-induced inhibition of IAA transport through the TZ to the EZ. Overall, the reported results demonstrate that boron supply mediates PIN2 endosome-based auxin transport to alleviate Al toxicity in plant roots.

Integration of root architecture, root nitrogen metabolism, and photosynthesis of ‘Hanfu’ apple trees under the cross-talk between glucose and IAA

Sugars and auxin have important effects on almost all phases of plant life cycle, which are so fundamental to plants and regulate similar processes. However, little is known about the effect of cross-talk between glucose and indole-3-acetic acid (IAA) on growth and development of apple trees. To examine the potential roles of glucose and IAA in root architecture, root nitrogen (N) metabolism and photosynthetic capacity in ‘Hanfu’ (Malus domestica), a total of five treatments was established: single application of glucose, IAA, and auxin polar transport inhibitor (2, 3, 5-triiodobenzoic acid, TIBA), combined application of glucose with TIBA and that of glucose with IAA. The combined application of glucose with IAA improved root topology system and endogenous IAA content by altering the mRNA levels of several genes involved in root growth, auxin transport and biosynthesis. Moreover, the increased N metabolism enzyme activities and levels of genes expression related to N in roots may suggest higher rates of transformation of nitrate (NO3--N) into amino acids application of glucose and IAA. Contrarily, single application of TIBA decreased the expression levels of auxin transport gene, hindered root growth and decreased endogenous IAA content. Glucose combined with TIBA application effectively attenuated TIBA-induced reductions in root topology structure, photosynthesis and N metabolism activity, and mRNA expression levels involved in auxin biosynthesis and transport. Taken together, glucose application probably changes the expression level of auxin synthesis and transport genes, and induce the allocation of endogenous IAA in root, and thus improves root architecture and N metabolism of root in soil with deficit carbon.

IAA Accumulation Promotes the Root Growth of Tea Plants under Aluminum

A moderate supply of aluminum (Al) can promote the root growth of plant species, such as tea, that have adapted to acid soils. The formation and development of roots are regulated by phytohormones. However, the role that phytohormones play in facilitating the beneficial effects of Al on root growth is unclear. In the present study, tea plants cultured hydroponically with either 0 or 0.4 mmol/L Al were characterized and compared in terms of root growth, hormone concentrations, and the expression of related genes. The hormone content in tea roots was measured by UPLC–MS, and the expression levels of genes involved in phytohormone biosynthesis, transport, and response were analyzed by qRT-PCR. Our results showed that the indole acetic acid (IAA) concentrations in the root base were significantly higher in plants supplied with Al compared to control plants without Al during the experimental period from 6 h to 7 d. By using inhibitors of IAA synthesis and transport, we found that Al induces short-term IAA synthesis in the root tip and enhances IAA transport in other parts of the root over the longer term, thus promoting root growth in tea plants by limiting IAA accumulation. The expression levels of genes YUC8 and AMI1, involved in IAA accumulation, were consistently upregulated in the root tip, whereas expression of PIN1/2/8, involved in IAA transport, was induced in the root base under Al supply at 6 h. In addition, the expression of PIN2 and PIN5 was highly induced in root stem by Al after 7 d and probably contributed to alleviating IAA accumulation in the root tip and thereby promoted root growth in tea plants. Meanwhile, the XET and expansin activities were increased under Al supply at 6 h. Thus, we conclude that the mechanism of Al-induced root growth is mediated by auxin accumulation and transport in cooperation with cell wall loosening in tea plants.

Thidiazuron Promotes Leaf Abscission by Regulating the Crosstalk Complexities between Ethylene, Auxin, and Cytokinin in Cotton.

Thidiazuron (TDZ) is widely used as a defoliant to induce leaf abscission in cotton. However, the underlying molecular mechanism is still unclear. In this study, RNA-seq and enzyme-linked immunosorbent assays (ELISA) were performed to reveal the dynamic transcriptome profiling and the change of endogenous phytohormones upon TDZ treatment in leaf, petiole, and abscission zone (AZ). We found that TDZ induced the gene expression of ethylene biosynthesis and signal, and promoted ethylene accumulation earlier in leaf than that in AZ. While TDZ down-regulated indole-3-acetic acid (IAA) biosynthesis genes mainly in leaf and IAA signal and transport genes. Furthermore, the IAA content reduced more sharply in the leaf than that in AZ to change the auxin gradient for abscission. TDZ suppressed CTK biosynthesis genes and induced CTK metabolic genes to reduce the IPA accumulation for the reduction of ethylene sensitivity. Furthermore, TDZ regulated the gene expression of abscisic acid (ABA) biosynthesis and signal and induced ABA accumulation between 12–48 h, which could up-regulate ABA response factor genes and inhibit IAA transporter genes. Our data suggest that TDZ orchestrates metabolism and signal of ethylene, auxin, and cytokinin, and also the transport of auxin in leaf, petiole, and AZ, to control leaf abscission.

Formation of the Secondary Abscission Zone Induced by the Interaction of Methyl Jasmonate and Auxin in Bryophyllum calycinum: Relevance to Auxin Status and Histology.

The interaction of methyl jasmonate (JA-Me) and indole-3-acetic acid (IAA) to induce the formation of the secondary abscission zone in the middle of internode segments of Bryophyllum calycinum was investigated in relation to auxin status and histology. When IAA at 0.1% (w/w, in lanolin) was applied to the segments, the formation of the secondary abscission zone at a few mm above the treatment in the apical direction was observed. On the contrary, IAA at 0.5% (w/w, in lanolin) did not induce the formation of the secondary abscission zone. JA-Me at 0.5% (w/w, in lanolin) applied to the middle of internode segments kept in the normal (natural) or inverted positions also induced the formation of the secondary abscission zone below and above parts of the treatment. IAA at 0.5% applied to the cut surface of the upper part of the segments completely prevented the formation of the secondary abscission zone induced by JA-Me. Simultaneous application of IAA 0.5% with JA-Me 0.5% in the middle part of the internode segments induced the formation of the secondary abscission zone at 10 mm to 12 mm above the treatment. Histological analyses indicated that the formation of the secondary abscission zone was characterized by the presence of newly synthesized cell plates that resulted from periclinal cell division within one layer of mother cells in stems. The effects of IAA (0.1%) and JA-Me (0.5%) on the formation of the secondary abscission zone were histologically similar. Comprehensive analyses of plant hormones revealed that the balance of the endogenous levels of IAA in both sides adjacent to the abscission zone was significantly disturbed when the secondary abscission formation was induced by the application of IAA. These results strongly suggest that an auxin gradient is important in the formation of the secondary abscission zone in the internode segments of B. calycinum, and IAA gradient results from polar IAA transport from the application site. IAA is important in the regulation of formation of the secondary abscission zone induced by JA-Me. Further possible mechanisms of the formation of the secondary abscission zone in the internode segments of B. calycinum are also discussed in the interaction of JA-Me and IAA.

Transport of Indole 3-acetic Acid through Bulk Liquid Membrane Influence of Carrier in the Diffusion Process

The paper presents a study of the phenomena that take place at membrane system interfaces in the process of indole-3-acetic acid (IAA) transport. The results were obtained in a bulk liquid membrane system using trioctylamine, tributylphosphate, trioctilphosphine oxide as carriers in chloroform. The main equilibriums that take place at the interface feed phase% membrane phase were identified and the diffusion coefficient of the indole-3-acetic acid complex (DLS) and the extraction constant (Kex) were assessed. The influence of the chemical potential gradient on these parameters was considered.

Effects of exogenous IAA in regulating photosynthetic capacity, carbohydrate metabolism and yield of Zizania latifolia

The effects of plant hormones on plant growth of Zizania latifolia are still considered to be the difficult and hot spots. The present study was carried out to investigate the regulation mechanism of exogenous indole-3-acetic acid (IAA) and IAA transport inhibitor (2,3,5-triiodobenzoic acid, TIBA) on yield, photosynthesis capacity, carbon assimilation and interaction of plant-fugal of Ustilago esculenta Henn. These results displayed that exogenous IAA led to significant increases in gall growth rate, swollen gall volume and biomass accumulation, promoting yield per unit of swollen gall in Z. latifolia. Exogenous IAA could improve the net photosynthetic rate (Pn), which contributed to higher stomatal conductance (Gs) and transpiration rate (Tr) in plant. TIBA treatment markedly inhibited the plant growth and photosynthetic rate. Contents of sucrose, glucose, fructose and total soluble sugar by IAA were significantly higher than control, whereas these parameters were declined by TIBA. Exogenous TIBA resulted in accumulation of starch (unfavorable to sugar metabolism) and crude fiber contents (declined swollen gall quality). Additionally, it was firstly reported the stimulation of exogenously applied of IAA and TIBA on plant growth and hyphae growth of U. esculenta. Microscopic analyses revealed that abundant hyphae were well-distributed closely to cell wall after IAA treatment, which could produce endogenous plant hormones, promoting the swollen cell structure enlargement. And hyphal aggregation over-growth by TIBA treatment indicated the vigorously increase of hyphae was surpassed the plant self-growth. These changes were greatly contributed to the increase of endogenous IAA content. The utilization of exogenous IAA and TIBA in present study proved IAA played a dominant role in regulating Z. latifolia growth.

Transcriptomics of cytokinin and auxin metabolism and signaling genes during seed maturation in dormant and non-dormant wheat genotypes

To gain insights into the roles of cytokinin (CK) and auxin in regulating dormancy during seed maturation in wheat, we examined changes in the levels of CK and indole-3-acetic acid (IAA) and expression patterns of their metabolism and signaling genes in embryonic and endospermic tissues of dormant and non-dormant genotypes. Seed maturation was associated with a decrease in the levels of isopentenyladenine in both tissues mainly via repression of the CK biosynthetic TaLOG genes. Differential embryonic trans-zeatin content and expression patterns of the CK related genes including TacZOG, TaGLU and TaARR12 between maturing seeds of the two genotypes implicate CK in the control of seed dormancy induction and maintenance. Seed maturation induced a decrease of IAA level in both tissues irrespective of genotype, and this appeared to be mediated by repression of specific IAA biosynthesis, transport and IAA-conjugate hydrolysis genes. The differential embryonic IAA content and expression pattern of the IAA biosynthetic gene TaAO during the early stage of seed maturation between the two genotypes imply the role of IAA in dormancy induction. It appears from our data that the expression of specific auxin signaling genes including TaRUB, TaAXR and TaARF mediate the role of auxin signaling in dormancy induction and maintenance during seed maturation in wheat.

Nitric Oxide Affects Rice Root Growth by Regulating Auxin Transport Under Nitrate Supply.

Nitrogen (N) is a major essential nutrient for plant growth, and rice is an important food crop globally. Although ammonium (NH4+) is the main N source for rice, nitrate (NO3-) is also absorbed and utilized. Rice responds to NO3- supply by changing root morphology. However, the mechanisms of rice root growth and formation under NO3- supply are unclear. Nitric oxide (NO) and auxin are important regulators of root growth and development under NO3- supply. How the interactions between NO and auxin in regulating root growth in response to NO3- are unknown. In this study, the levels of indole-3-acetic acid (IAA) and NO in roots, and the responses of lateral roots (LRs) and seminal roots (SRs) to NH4+ and NO3-, were investigated using wild-type (WT) rice, as well as osnia2 and ospin1b mutants. NO3- supply promoted LR formation and SR elongation. The effects of NO donor and NO inhibitor/scavenger supply on NO levels and the root morphology of WT and nia2 mutants under NH4+ or NO3- suggest that NO3--induced NO is generated by the nitrate reductase (NR) pathway rather than the NO synthase (NOS)-like pathway. IAA levels, [3H] IAA transport, and PIN gene expression in roots were enhanced under NO3- relative to NH4+ supply. These results suggest that NO3- regulates auxin transport in roots. Application of SNP under NH4+ supply, or of cPTIO under NO3- supply, resulted in auxin levels in roots similar to those under NO3- and NH4+ supply, respectively. Compared to WT, the roots of the ospin1b mutant had lower auxin levels, fewer LRs, and shorter SRs. Thus, NO affects root growth by regulating auxin transport in response to NO3-. Overall, our findings suggest that NO3- influences LR formation and SR elongation by regulating auxin transport via a mechanism involving NO.

Cadmium and arsenic affect root development in Oryza sativa L. negatively interacting with auxin

Cadmium (Cd) and arsenic (As), non essential, but toxic, elements for animals and plants are frequently present in paddy fields. Oryza sativa L., a staple food for at least the half of world population, easily absorbs As and Cd by the root, and in this organ the pollutants evoke consistent damages, reducing/modifying the root system. Auxins are key hormones in regulating all developmental processes, including root organogenesis. Moreover, plants respond to environmental stresses, such as those caused by Cd and As, by changing levels and distribution of endogenous phytohormones. Even though the effects of Cd and As on the roots have been investigated in some species, it remains necessary to deepen the knowledge about the cross-talk between these toxic elements and auxin during root formation and development, in particular in agronomically important plants, such as rice. Hence, the research goal was to investigate the interactions between Cd and As, alone or combined, and auxin during the development of rice roots. To reach the aim, morphological, histological and histochemical analyses were carried out on seedlings, exposed or not to Cd and/or As, belonging to the wild type and transgenic lines useful for monitoring indole-3-acetic acid (IAA) localization, i.e., OsDR5:GUS, and IAA cellular influx and efflux, i.e., OsAUX1:GUS and OsPIN5b:GUS. Moreover, the transcript levels of the YUCCA2 and ASA2, IAA biosynthetic genes were also monitored in Cd and/or As exposed wild type seedlings. The results highlight that As and Cd affect cyto-histology and morphology of the roots. In particular, they alter the lateral root primordia organization and development with negative consequences on root system architecture. This is due to a disturbance of IAA biosynthesis and transport, as indicated by the altered expression of both ASA2 and YUCCA2 biosynthetic genes, and AUX1 and PIN5b transporter genes.

Mycorrhiza stimulates root-hair growth and IAA synthesis and transport in trifoliate orange under drought stress

Root-hair growth and development regulated by soil microbes is associated with auxin. In this background, we hypothesized that mycorrhizal fungal inoculation induces greater root-hair growth through stimulated auxin synthesis and transport under water stress conditions. Trifoliate orange (Poncirus trifoliata) was inoculated with an arbuscular mycorrhizal (AM) fungus (Funneliformis mosseae) under well-watered (WW) and drought stress (DS) for 9 weeks. Compared with non-AM seedlings, AM seedlings displayed significantly higher density, length, and diameter of root hairs and root indoleacetic acid (IAA) level, whereas lower total root IAA efflux, regardless of soil moisture status. Root PtYUC3 and PtYUC8 involved in IAA biosynthesis were up-regulated by mycorrhization under WW and DS, whereas AM-modulated expression in PtTAA1, PtTAR2, PtYUC4, and PtYUC6 depended on status of soil moisture. Mycorrhizal inoculation down-regulated the transcript level of root auxin efflux carriers like PtPIN1 and PtPIN3, whereas significantly up-regulated the expression of root auxin-species influx carriers like PtABCB19 and PtLAX2 under DS. These results indicated that AMF-stimulated greater root-hair growth of trifoliate orange under DS that is independent on AMF species is related with mycorrhiza-modulated auxin synthesis and transport, which benefits the host plant to enhance drought tolerance.

The Interaction between Auxin and Nitric Oxide Regulates Root Growth in Response to Iron Deficiency in Rice

Fe deficiency (-Fe) is a common abiotic stress that affects the root development of plants. Auxin and nitric oxide (NO) are key regulator of root growth under -Fe. However, the interactions between auxin and NO regulate root growth in response to Fe deficiency are complex and unclear. In this study, the indole-3-acetic acid (IAA) and NO levels in roots, and the responses of root growth in rice to different levels of Fe supply were investigated using wild type (WT), ospin1b and osnia2 mutants. -Fe promoted LR formation but inhibited seminal root elongation. IAA levels, [3H] IAA transport, and expression levels of PIN1a-c genes in roots were reduced under -Fe, suggesting that polar auxin transport from shoots to roots was decreased. Application of IAA to -Fe seedlings restored seminal root length, but not LR density, to levels similar to those under normal Fe (+Fe), and the seminal root length was shorter in two ospin1b mutants relative to WT under +Fe, but not under -Fe, confirming that auxin transport participates in -Fe-inhibited seminal root elongation. Moreover, -Fe-induced LR density and -Fe-inhibited seminal root elongation paralleled NO production in roots. Interestingly, similar NO accumulation and responses of LR density and root elongation were observed in osnia2 mutants compared to WT, and the higher expression of NOA gene under -Fe, suggesting that -Fe-induced NO was generated via the NO synthase-like pathway rather than the nitrate reductase pathway. However, IAA could restore the functions of NO in inhibiting seminal root elongation, but did not replace the role of NO-induced LR formation under -Fe. Overall, our findings suggested that NO functions downstream of auxin in regulating LR formation; NO-inhibited seminal root elongation by decreasing meristem activity in root tips under -Fe, with the involvement of auxin.

Auxin is Involved in Lateral Root Formation Induced by Drought Stress in Tobacco Seedlings

Drought stress is a common abiotic stress that affects plant root development. The goal of this study was to identify the role of auxin on aspects of root growth under water-deficient conditions in Nicotiana tabacum L. In this study, time-course development of lateral root (LR) growth in response to water deficiency (2.5% polyethylene glycol 6000, PEG) was analyzed in tobacco seedlings. LR formation in tobacco plants in response to drought stress depended on the experimental duration. First- and second-order LR formation was induced, respectively, by 43 and 28% on day 5 and by 57 and 40% on day 7 relative to control treatment. Thereafter, PEG-induced LR formation was not observed, and a decrease of 16% was recorded in first-order LR formation on day 11. A root pruning study suggested that PEG-induced LR formation plays a pivotal role in maintaining tobacco plant growth during drought stress. The change in the indole-3-acetic acid (IAA) concentration in leaves and roots was consistent with that of LR formation. This was corroborated by elevated DR5::GUS expression levels on day 5 and by decreased expression levels on day 9. Application of N-1-naphthylphthalamic acid (NPA) to PEG-treated seedlings reduced LR formation to levels similar to those of the control treatment; DR5::GUS expression was also similar between these treatments after NPA application. Results were similar following α-naphthylacetic acid application to control-treated seedlings. Measurement of [3H]IAA transport indicated that polar auxin transport from shoots to roots was increased by 43% under PEG application. Quantitative reverse transcription PCR revealed higher expression of YUC and PIN genes in PEG-treated plants, indicating that they play key roles in auxin synthesis and transport regulation in drought stress. Overall, these suggest that drought stress regulates LR formation in tobacco seedlings via modification of auxin synthesis and polar transport depending on the experimental stage.

Activation of gibberellin 20-oxidase 2 undermines auxin-dependent root and root hair growth in NaCl-stressed Arabidopsis seedlings

Although salt stress mainly disturbs plant root growth by affecting the biosynthesis and signaling of phytohormones, such as gibberellin (GA) and auxin, the exact mechanisms of the crosstalk between these two hormones remain to be clarified. Indole-3-acetic acid (IAA) is a biologically active auxin molecule. In this study, we investigated the role of Arabidopsis GA20-oxidase 2 (GA20ox2), a final rate-limiting enzyme of active GA biosynthesis, in IAA-directed root growth under NaCl stress. Under the NaCl treatment, seedlings of a loss-of-function ga20ox2-1 mutant exhibited primary root and root hair elongation, altered GA4 accumulation, and decreased root Na+ contents compared with the wild-type, transgenic GA20ox2-complementing, and GA20ox2-overexpression plant lines. Concurrently, ga20ox2-1 alleviated the tissue-specific inhibition of NaCl on IAA generation by YUCCAs, IAA transport by PIN1 and PIN2, and IAA accumulation in roots, thereby explaining how NaCl increased GA20ox2 expression in shoots but disrupted primary root and root hair growth in wild-type seedlings. In addition, a loss-of-function pin2 mutant impeded GA20ox2 expression, indicating that GA20ox2 function requires PIN2 activity. Thus, the activation of GA20ox2 retards IAA-directed primary root and root hair growth in response to NaCl stress.

Regulation of NPA and ACC on H2O2-Induced Pea Primary Horizontal Bending Root

Existing reports have described a notable fact that 1-naphthylphthalamic acid (NPA) coupled with 1-aminocylopropane-1-carboxylic acid (ACC), a precursor of ethylene, can regulate gravitropism of pea primary root by altering the volume of indole-3-acetic acid (IAA) and gibberellin A3 (GA3) in the root. However, the mechanism behind this is not quite clear. In this study, various concentration setups of NPA and ACC were used during cultivation of pea seeds (exposed to H2O2) to study the effect of NPA and ethylene on contents of IAA and GA3 in curving pea primary roots. Results showed that generally the horizontal bending of pea primary roots induced by H2O2 could be alleviated by NPA and ACC. Alpha-amylase activity in pea primary roots declined gradually with the rise of NPA and ACC concentrations, consequently boosting the content of starch while reducing the content of soluble sugar. ACC instigated the ethylene triple response. In addition, IAA content in primary roots declined gradually with the rise of NPA concentration, but GA3 content did not show any consistent fluctuation. The inhibition of NPA on IAA polar transport in pea was only effective in primary roots, with no effect on the IAA transport from bud to other parts.

Ethylene and auxin interaction in the control of adventitious rooting in Arabidopsis thaliana.

Adventitious roots (ARs) are post-embryonic roots essential for plant survival and propagation. Indole-3-acetic acid (IAA) is the auxin that controls AR formation; however, its precursor indole-3-butyric acid (IBA) is known to enhance it. Ethylene affects many auxin-dependent processes by affecting IAA synthesis, transport and/or signaling, but its role in AR formation has not been elucidated. This research investigated the role of ethylene in AR formation in dark-grown Arabidopsis thaliana seedlings, and its interaction with IAA/IBA. A number of mutants/transgenic lines were exposed to various treatments, and mRNA in situ hybridizations were carried out and hormones were quantified In the wild-type, the ethylene precursor 1-aminocyclopropane-1-carboxylic acid (ACC) at 0.1 μM enhanced AR formation when combined with IBA (10 μM), but reduced it when applied alone; this effect did not occur in the ein3eil1 ethylene-insensitive mutant. ACC inhibited the expression of the IAA-biosynthetic genes WEI2, WEI7, and YUC6, but enhanced IBA-to-IAA conversion, as shown by the response of the ech2ibr10 mutant and an increase in the endogenous levels of IAA. The ethylene effect was independent of auxin-signaling by TIR1-AFB2 and IBA-efflux by ABCG carriers, but it was dependent on IAA-influx by AUX1/LAX3.Taken together, the results demonstrate that a crosstalk involving ethylene signaling, IAA-influx, and IBA-to-IAA conversion exists between ethylene and IAA in the control of AR formation.

Regulation of polar auxin transport in grapevine fruitlets (Vitis vinifera L.) and the proposed role of auxin homeostasis during fruit abscission.

BackgroundIndole-3-acetic acid (IAA), the most abundant auxin, is a growth promoter hormone involved in several developmental processes. Auxin homeostasis is very important to its function and this is achieved through the regulation of IAA biosynthesis, conjugation, degradation and transport. In grapevine, IAA plays an essential role during initial stages of berry development, since it delays fruitlet abscission by reducing the ethylene sensitivity in the abscission zone. For this reason, Continuous polar IAA transport to the pedicel is required. This kind of transport is controlled by IAA, which regulates its own movement by modifying the expression and localization of PIN-FORMED (PIN) auxin efflux facilitators that localize asymmetrically within the cell. On the other hand, the hormone gibberellin (GA) also activates the polar auxin transport by increasing PIN stability. In Vitis vinifera, fruitlet abscission occurs during the first two to three weeks after flowering. During this time, IAA and GA are present, however the role of these hormones in the control of polar auxin transport is unknown.ResultsIn this work, the use of radiolabeled IAA showed that auxin is basipetally transported during grapevine fruitlet abscission. This observation was further supported by immunolocalization of putative VvPIN proteins that display a basipetal distribution in pericarp cells. Polar auxin transport and transcripts of four putative VvPIN genes decreased in conjunction with increased abscission, and the inhibition of polar auxin transport resulted in fruit drop. GA3 and IAA treatments reduced polar auxin transport, but only GA3 treatment decreased VvPIN transcript abundance. When GA biosynthesis was blocked, IAA was capable to increase polar auxin transport, suggesting that its effect depends on GA content. Finally, we observed significant changes in the content of several IAA-related compounds during the abscission period.ConclusionsThese results provide evidence that auxin homeostasis plays a central role during grapevine initial fruit development and that GA and IAA controls auxin homeostasis by reducing polar auxin transport.Electronic supplementary materialThe online version of this article (doi:10.1186/s12870-016-0914-1) contains supplementary material, which is available to authorized users.

Higher Ammonium Transamination Capacity Can Alleviate Glutamate Inhibition on Winter Wheat (Triticum aestivum L.) Root Growth under High Ammonium Stress.

Most of the studies about NH4+ stress mechanism simply address the effects of free NH4+, failing to recognize the changed nitrogen assimilation products. The objective of this study was to elucidate the effects of glutamate on root growth under high ammonium (NH4+) conditions in winter wheat (Triticum aestivum L.). Hydroponic experiments were conducted using two wheat cultivars, AK58 (NH4+-sensitive) and Xumai25 (NH4+-tolerant) with either 5 mM NH4+ nitrogen (AN) as stress treatment or 5 mM nitrate (NO3-) nitrogen as control. To evaluate the effects of NH4+-assimilation products on plant growth, 1 μM L-methionine sulfoximine (MSO) (an inhibitor of glutamine synthetase (GS)) and 1 mM glutamates (a primary N assimilation product) were added to the solutions, respectively. The AN significantly reduced plant biomass, total root length, surface area and root volume in both cultivars, but less effect was observed in Xumai25. The inhibition effects were alleviated by the application of MSO but strengthened by the application of glutamate. The AN increased the activities of GS, glutamate dehydrogenase (GDH) in both cultivars, resulting in higher glutamate contents. However, its contents were decreased by the application of MSO. Compared to AK58, Xumai25 showed lower glutamate contents due to its higher activities of glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT). With the indole-3-acetic acid (IAA) contents decreasing in roots, the ratio of shoot to root in IAA was increased, and further increased by the application of glutamate, and reduced by the application of MSO, but the ratio was lower in Xumai25. Meanwhile, the total soluble sugar contents and its root to shoot ratio also showed similar trends. These results indicate that the NH4+-tolerant cultivar has a greater transamination ability to prevent glutamate over-accumulation to maintain higher IAA transport ability, and consequently promoted soluble sugar transport to roots, further maintaining root growth.

Potassium deficiency inhibits lateral root development in tobacco seedlings by changing auxin distribution

Potassium deficiency (LK) is a common abiotic stress that affects plant root development. We aimed to identify the role of auxin in LK effects on root growth. We investigated the effects of LK on the formation and elongation of first- and second-order lateral roots (LRs), auxin concentration, DR5::GUS expression, [3H]IAA transport and the expressions of six PIN genes in roots of tobacco plants. We also examined the effects of exogenous auxin (NAA) and a transport inhibitor (NPA) on LR growth and DR5::GUS expression. Potassium deficiency reduced root growth, mostly by impairing the formation and elongation of first-order LRs. Indole acetic acid (IAA) concentration and DR5::GUS expression levels decreased in leaves and roots subjected to LK, indicating that LK affected auxin distribution. [3H]IAA transport and the expression levels of PIN genes were reduced by LK, showing that auxin polar transport was inhibited by LK. Application of NAA to LK-treated plants increased first-order LR formation and elongation to levels similar to the control, an outcome that was consistent with the similarity in DR5::GUS expression levels between treatments. NPA acted in a converse manner. Potassium deficiency inhibited LR formation and elongation in tobacco plants by shifting auxin distribution.
