Favipiravir Protects Enterocytes From Cell Death After Inflammatory Storm.

Over the past years, inflammatory bowel disease (IBD) treatment has become more targeted, anticipating the use of immune-modifying therapies at an earlier stage. During the treatment process prevention and management of viral infections hold significant importance. The protective role of favipiravir on enterocytes which are affected by inflammation is still unknown. We aim to analyze the effects of favipiravir on enterocytes after an inflammatory condition. We conducted a 2,5-diphenyl-2H-tetrazolium bromide (MTT) assay to assess the cytotoxicity of favipiravir on intestinal epithelioid cells (IEC-6). To mimic the inflammation model in cell culture conditions, we exposed IEC-6 cells to tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ). The cells were categorized into four groups: control, inflammation model, application of favipiravir before inflammation (prophylactic), and application of favipiravir after inflammation (treatment). We assessed the presence and distribution of caspase 1, caspase 3, interleukin6 (IL6), interleukin 8 (IL8), mixed lineage kinase domain-like protein (MLKL), receptor-interacting protein kinase 1 (RIPK1), and TNF-α using indirect immunoperoxidase staining. TNF-α and IL8 levels were analyzed with enzyme-linked immunosorbent assay (ELISA) in a culture medium. Caspase 1 was observed to be strong (+++) in the treatment group and weak (+) in the prophylactic group compared to the inflammation group. Caspase 3 was weak (+) in the inflammation group, and it was strong (+++) in the prophylactic and treatment group, the increase in the treatment group was significant. Therefore administering favipiravir before inducing inflammation appears to control the inflammatory caspase pathway in intestinal enterocytes, protecting them from inflammatory responses, while the caspase 3-dependent apoptotic pathway may not be active in enterocytes during inflammation. IL6 and IL8 were negative (-) in control, IL6 was weak (+) in inflammation and favipiravir treated groups; IL8 increased significantly in favipiravir groups compared to control and inflammation groups. Consequently, favipiravir may trigger IL6 release, initiating the inflammatory pathway and potentially enhancing IL8 interactions with other cytokines. TNF-α immunoreactivity was strong (+++) in the inflammation group, while it was moderate (++) in favipiravir-administered groups. MLKL immunoreactivity was strong (+++) in all groups, RIPK1 was weak (+) in control, strong (+++) in the inflammation and treatment group, moderate (++) in the prophylactic group, and the increase in inflammation and treatment group was significant compared to control. Our findings suggest that in the treatment group, necroptosis was triggered by increased MLKL and RIPK1, key players in inflammation and cell death. After immunocytochemical evaluation, our findings suggest that, after the onset of inflammation, favipiravir may play a role in cell death by increasing necroptosis rather than apoptosis.

Comparative phytochemical studies on the roots of Polygala azizsancarii and P. peshmenii and neuroprotective activities of the two xanthones

Six known sucrose mono-, di- and triesters and five xanthone derivatives were isolated from the roots of Polygala peshmenii Eren, Parolly, Raus & Kürschner which is a narrow species endemic to Türkiye. Among the xanthones, 1,7-dihydroxy-2,3-methylenedioxy-5,6-dimethoxy-xanthone is an undescribed compound isolated for the first time from a natural source. The studies on the roots of P. azizsancarii Dönmez have resulted in the isolation of four known compounds including sucrose mono-, di- and triesters. The structures of the sucrose esters and xanthones isolated from P. azizsancarii and P. peshmenii were established by spectroscopic methods, including 1D-NMR (1H NMR, 13C NMR, DEPT-135), 2D-NMR (COSY, NOESY, HSQC, HMBC). Neuroprotective activities of two xanthones, 1,3,6-trihydroxy-2,5,7-trimethoxyxanthone and 3-O-β-D-glucopyranosyloxy-1,6-dihydroxy-2,5,7-trimethoxyxanthone isolated from the roots of P. azizsancarii were evaluated in vitro using in a cellular model of Alzheimer's disease. SKNAS human neuroblastoma cells were used in the study and treated with different consecrations of Aβ₂₅₋₃₅ oligomer for up to 48 h. Cell viability was evaluated using MTT assay. The distribution of β-amyloid, α-synuclein, tau, JAK2, STAT3, caspase 3 and BMP-2 were investigated using indirect immunoperoxidase staining. Our results suggested that both xanthones control tau aggregation with no effect on β-amyloid plaque formation. In addition, for neuronal pathophysiology in AD cell model, decreased distributions of JAK/STAT3 and BMP2 signaling pathways were demonstrated, therefore they play a role in the protective effect on neurons in neurodegenerative disease. A significant decrease in caspase 3 immunoreactivity was detected after the administration of both compounds in AD cells. Therefore, both compounds control neuronal pathophysiology and rescue cell death in AD disease.

Neuroprotective Effects of Hesperidin and Naringin in SK-N-AS Cell as an In Vitro Model for Alzheimer’s Disease

Objective: Hesperidin and naringin are flavonoids that are found in citrus fruits. Our aim was to create an in vitro model of Alzheimer’s disease (AD) and to evaluate the neuroprotective effects of hesperidin and naringin in SK-N-AS and AD model cells. Methods: Aβ25-35 was used to create an AD model in SK-N-AS cells. The cytotoxicity of hesperidin and naringin was evaluated using MTT. β-amyloid, tau and α-synuclein distributions were analyzed using indirect immunoperoxidase staining to investigate the neuroprotective effects of hesperidin and naringin. Results: The AD model was created by 1 µM of Aβ25-35 for 48 hours after ThT staining. The intensity of β-amyloid was reduced through both hesperidin and naringin treatment in AD model cells. Both flavonoids significantly decreased the intensity of α-synuclein in SK-N-AS and AD model cells. Conclusions: Hesperidin and naringin can be potentially used as neuroprotective agents. Naringin may be more effective than hesperidin in the accumulation of β-amyloid and tau proteins.

Expressions of heat shock protein 90, inducible nitric oxide synthase, and vascular endothelial growth factor in the skin of diabetic rats.

Background and Aim:Chronic skin ulceration is a common and painful condition that affects about 15% of patients with diabetes worldwide. The aim of this study was to evaluate the expressions of heat shock protein 90 (HSP 90), inducible nitric oxide synthase (iNOS), and vascular endothelial growth factor (VEGF) in the skin of diabetic rats.Materials and Methods:A total of 20 rats were divided randomly into two equal groups. Diabetes mellitus (DM) was induced in the rats of Group 2, whereas the rats in Group 1 were kept healthy and served as control. DM was induced by a single intraperitoneal injection of alloxan monohydrate at 120 mg/kg. Rats were considered diabetic if the blood glucose level was above 200 mg/dL. After induction of DM, the rats were monitored daily for 28 days. On day 28, the rats were humanely euthanized, and full-thickness skin punch biopsy was obtained from the dorsal side of the thoracolumbar region. Indirect immunoperoxidase staining was used to evaluate the expressions of HSP 90, iNOS, and VEGF in the skin tissue specimens.Results:The expressions of HSP 90, iNOS, and VEGF in the skin were significantly higher in diabetic rats than in the control rats. On day 28 in diabetic rats, a positive correlation (r=0.65, p=0.01) was detected between mean blood glucose level and the expression levels of HSP 90, iNOS, and VEGF.Conclusion:The results of this study indicated that DM upregulated the expressions of HSP 90, iNOS, and VEGF in the skin tissues of diabetic rats and may impact the healing of skin wounds. However, this study was preliminary and further studies to investigate this relationship are warranted.

Factors Affecting The Indirect Immunoperoxidase Staining Of Infectious Bursal Disease Virus Antigens In Cells

THE application of the Immunoperoxidase (IP) antibody technique for the definitive diagnosis of Infectious Bursal Disease (IBD) in chickens has earlier been described (Okeke and Lang, 1982). During that study it was noted that the Indirect method of IP was more sensitive in the demonstration of IBD viral antigens than the direct method. Consequently, more formation was sought on the specific technical parameter and the consequences that resulted by deviations from the codified procedures employed in the Indirect technique with the aim of obtaining an even better result. Studies of pH, serum dilutions, serum incubation time, conjugate dilutions, conjugate incubation time and developing time were conducted to find out what Influence such factors could have on the final staining product.

Evaluation of correlation of EIV(H3N8) replication and pathological changes in lungs of BALB/c mice

Equine influenza (EI) is a highly contagious viral respiratory disease caused by the equine influenza virus (EIV). Frequent antigenic changes of EIV warrants thorough pathological evaluation and in-vivo replication of the circulating viruses in an experimental animal model for selection of vaccine strain. In the present study, EIV (H3N8) was inoculated into BALB/c mice and subsequently, gross, histopathological and immunohistochemical examination of the lungs was performed to evaluate the correlation of virus replication with corresponding pathological changes. Microscopically, at 1dpi, lungs from mice infected with EIV revealed degeneration of epithelial tissues, whereas indirect immunoperoxidase staining (IIPT) results showed high EIV antigen positivity in the bronchial and bronchiolar epithelium and mild positivity in the infiltrating cells. Subsequently from 1 dpi to 3 dpi, the virus replicated and infected other parenchymal cells, which led to recruitment of inflammatory cells. Microscopically moderate peribronchial, peribronchiolar and perivascular cuffing of neutrophils and macrophages were observed, whereas IIPT exhibited, scanty and moderate EIV antigen positive interstitial infiltrating macrophages. At 5 and 7 dpi, severe, diffuse interstitial thickening, bronchial, bronchiolar and alveolar epithelial necrosis and denudation, perivascular and peribronchial mononuclear cells cuffing were observed, which could be correlated with IIPT as high EIV antigen positivity of infiltrating macrophages and type II pneumocytes. However, from 11 dpi onwards, microscopically, moderate interstitium thickening and perivascular and peribronchial cuffing were observed whereas, tissues were negative for EIV antigen in IIPT. Mild, focal inflammation could be observed at 14 dpi. Present findings signify the correlation of virus replication with corresponding histopathological changes, which will be useful for assessing the efficacy of anti-influenza drug molecules and EIV vaccines.

Characterization of Progesterone Receptor Expression in Intracranial Meningiomas of Patients Treated in a High-Complexity Hospital in Bogota, Colombia.

BackgroundMeningiomas are the single most common brain tumor. The incidence of these tumors increases with age; different studies have shown that meningiomas usually appear after the age of 50. These tumors are more common in women than in men, and women are twice as likely to suffer from the condition. Surgery is the primary form of treatment, which can be curative with complete resection. If the tumor is unresectable or other treatments such as surgery and radiotherapy have failed, hormonal therapy or chemotherapy may be considered. There is limited information about the clinical, demographic, and histopathological characteristics of these tumors in the population of Bogotá, Colombia.ObjectiveTo evaluate the expression of progesterone receptors in patients over 18 years old who have been diagnosed with meningiomas in a high-complexity hospital in Bogota, Colombia, and to describe the demographic and histopathological characteristics of these patients.MethodsThis is a descriptive and retrospective case series. Patients with meningioma who underwent surgical resection at a high-complexity hospital in Bogota, Colombia, from 2016 to 2019 were retrospectively identified and studied. Demographic variables, such as age and gender, were extracted from the clinical chart. Indirect immunoperoxidase staining was carried out for the progesterone receptor (PR) and Ki67. PR is analyzed as positive and negative, and the Ki67 proliferation index was determined.ResultsThirty-two meningiomas from patients who underwent surgery were available for analysis. Twenty-five (78.1%) were positive for PR, 71.8% were females, and 93% were World Health Organization (WHO) grade I. Meningothelial (28%), fibrous (25%), and transitional (25%) meningiomas were the most frequent subtypes, correspondingly. The Ki67 mean value was 1.14 (0.11-10.71).ConclusionOur case series showed a greater frequency of meningiomas in women, with a high PR expression and a low Ki67 proliferation rate. These data correlate with literature worldwide.

The effects of tramadol on cancer stem cells and metabolic changes in colon carcinoma cells lines

Opioids are widely used in the treatment of cancer related pain. They mainly exert their effects on opioid receptors. The most common opioid in the treatment of pain is morphine. Previous studies show that they may have effects on cancer cell behavior. These may include apoptosis, angiogenesis, invasion, inflammation and immune reactions. Tramadol, also an opioid is widely used in the treatment of cancer pain and is not well studied in cancer behavior. We aimed to investigate the effects of tramadol on cancer stem cells and metabolic changes in colon carcinoma cells.We used Colo320 (ATCC, CCL-220), Colo741 (ECACC, 93052621) and HCT116 (ATCC, CCL-247) colon cancer cell lines. CD133 was considered colon cancer stem cell marker and used to sort CD133+ and CD133− cells by magnetic cell sorting. MTT (mitochondria-targeted therapeutics) technique was used to detect tramadol's cytotoxic effect on cells in the study groups. Cells were treated with 1 mg/kg, 1.5 mg/kg and 2 mg/kg tramadol for 24 h at 37 °C and 5% CO2.Caspase-3, Ki-67, Bcl-2 and VGEF distributions were performed using indirect immunoperoxidase staining for immunohistochemical analysis. The study showed that tramadol has triggering effect on apoptosis in Colo320 colon cancer stem cells.

Gross and Pathomorphological Alterations of Infectious Bursal Disease Virus (IBDV) Infection and Its Localization by Indirect Immunoperoxidase in Broilers

The present research was undertaken to study the gross and pathomorphological alterations due to infectious bursal disease virus infection in broilers. A total of 29 outbreaks were recorded from Srinagar and adjoining districts during the study period. Out of the 29 recorded outbreaks, 22 (75.86%) were clinical outbreaks and the rest 7 (24.13%) were subclinical outbreaks. The average size of the affected flocks ranged between 2500 to 6000 birds. The affected birds were mostly between 17 and 35 days of age. Among the affected flocks morbidity varied from 30% to 60%, however mortality varied from 9 % to 25%. On postmortem examination the carcasses were found to be emaciated, dehydrated with darkened discoloration and presence of petechial and ecchymotic hemorrhages in thigh and breast muscles. Histopathological alterations mostly comprised of depletion of B- lymphocytes with RE cell hyperplasia. Indirect immunoperoxidase staining of the tissues revealed viral antigen in the form of coarsely brown deposits.

54 IMMUNOLOCALIZATION OF STEROID SULFATASE AND ESTROGEN-SPECIFIC SULFOTRANSFERASE IN BOVINE FOLLICLES

Steroid hormones are regulators in the fine-tuned mechanism of follicular development in cattle. Their concentration and property can be modulated via different processes. Sulfoconjugation via sulfotransferases (SULT) changes them from being hydrophobic to hydrophilic molecules, thereby preventing them from diffusing freely across the lipid bilayer and necessitating a transport system like the sodium-dependent organic anion transporter (SOAT; SLC10A6). In addition, sulfated steroids can no longer bind to their nuclear receptors, rendering them biologically inactive. Steroid sulfatase (STS) removes the sulfate moiety from conjugated steroids, transforming them to the free active forms. Data about the concentration of sulfated steroids in follicular fluid and the expression of the related enzymes are limited in horses and cattle. Recently, transcripts of the corresponding enzymes have been detected in cumulus cells of immature bovine cumulus-oocyte complex. Furthermore, it has been shown that small amounts of sulphated steroids are present in bovine follicular fluid. The objective of the study was to investigate the steroid metabolizing enzymes at the protein level via immunohistochemistry (IHC). Bovine ovaries collected at a slaughterhouse were categorized according to their oestrus cycle stage. Three pairs of ovaries of each cycle stage (proestrus, oestrus, postestrus, interestrus) were collected. Morphological criteria (ovaries: presence/absence of follicles and/or CL, size and number of follicles; closure of the cervix, amount of mucus) were employed to categorize them. Samples for IHC were fixed in formalin. After washing and dehydration, the samples were finally embedded in paraffin and mounted onto blocks. Indirect immunoperoxidase staining methods were applied using the streptavidin-biotin technique for signal enhancement following standard procedures. Tissue sections of 3 µm were mounted onto glass slides, and a polyclonal rabbit anti-human STS and a polyclonal rabbit anti-bovine SULT1E1 primary antibody were used to detect STS or SULT1E1, respectively. Negative controls were included using nonspecific rabbit IgG. The evaluation of the staining was descriptive in relation to a positive control (bovine placenta). Immunostaining for STS was detected in granulosa cells of antral and secondary follicles and in the endothelium of blood vessels, irrespective of the stage of the oestrus cycle. Moreover, staining for SULT1E1 was restricted to granulosa cells in antral follicles, again irrespective of the stage of the oestrus cycle. However, staining intensity for STS and SULT1E1 was only weak. These data indicate for the first time the presence of the steroid metabolising enzymes STS and SULT1E1 in bovine ovaries at the protein level. However, due to the low expression level, further studies are needed to clarify the function. Inhibition ofPasteurella multocidaAdhesion to Rabbit Respiratory Epithelium Using Lectins

This study aimed to evaluate the ability of a panel of lectins to inhibit the ability of Pasteurella multocida to adhere to and affect the rabbit respiratory epithelium. Nasal septa from rabbit fetuses were cultured with various lectins before the addition of P. multocida. The percentage of bacteria adhering to the epithelium was evaluated semiquantitatively by indirect immunoperoxidase (IIP) staining. The goblet cells (GCs) were counted in semithin sections stained with toluidine blue and served as the main morphological criterion to evaluate the inhibitory effect of the lectins. The lectins PNA, WGA, RCA120, and DBA significantly inhibited the adhesion of P. multocida to the ciliated epithelium (P < 0.05) and prevented the pathogen-induced increase in the number of GCs (P < 0.05) compared with those of positive control tissues. In addition, VVA, SJA, UEA I, DSL, SBA, and ECL significantly inhibited the increase in GCs compared with that of the control tissues. The results suggest that less aggressive therapeutic strategies, such as treatment with lectins, may represent alternative approaches to control bacterial respiratory infections.

Detection of HHV-6-specific mRNA and antigens in PBMCs of individuals with chromosomally integrated HHV-6 (ciHHV-6)

After inheritance of chromosomally integrated HHV-6 (ciHHV-6), viral DNA is found in every nucleated cell. The prevalence of ciHHV-6 is estimated to be 0.2-5% of humans. There are conflicting data on the potential for replication, possibly leading to clinical implications. We analysed peripheral blood mononuclear cells (PBMCs) from individuals with ciHHV-6 proven by fluorescence in situ hybridization (FISH) for HHV-6-specific mRNA (U94, U42, U22) and antigens by means of reverse transcription PCR and an indirect immunoperoxidase staining. U94 transcripts indicative of latent infection were detected in six (54.5%) out of 11 individuals at least once. Transcripts indicative of lytic infection (i.e. U42 and U22) were detected in four (36.4%) out of 11 individuals at least once. HHV-6 antigen was detected in seven (70%) out of 10 individuals at least once. The presence of viral mRNA and proteins supports virus gene expression from ciHHV-6, which may lead to virus replication. Considering the properties of active HHV-6 infection together with obvious replicative activity in individuals with ciHHV-6, pathophysiological effects leading to clinical consequences of chromosomally integrated viral DNA might be considered.

Metagonimus yokogawai: a 100-kDa somatic antigen commonly reacting with other trematodes.

This study was undertaken to characterize the properties of a 100 kDa somatic antigen from Metagonimus yokogawai. Monoclonal antibodies (mAbs) were produced against this 100 kDa antigen, and their immunoreactivity was assessed by western blot analysis with patients' sera. The mAbs against the 100 kDa antigen commonly reacted with various kinds of trematode antigens, including intestinal (Gymnophalloides seoi), lung (Paragonimus westermani), and liver flukes (Clonorchis sinensis and Fasciola hepatica). However, this mAb showed no cross-reactions with other helminth parasites, including nematodes and cestodes. To determine the topographic distribution of the 100 kDa antigen in worm sections, indirect immunoperoxidase staining was performed. A strong positive reaction was observed in the tegumental and subtegumental layers of adult M. yokogawai and C. sinensis. The results showed that the 100 kDa somatic protein of M. yokogawai is a common antigen which recognizes a target epitope present over the tegumental layer of different trematode species.

Isolation and identification of infectious bursal disease virus from broiler and layer chickens during the outbreak year 2007 in Bangladesh

The objective of the present study was to isolate and identify infectious bursal disease viruses (IBDVs) from broiler and layer chickens of outbreaks of infectious bursal disease (IBD) in three districts of Bangladesh. A total of 70 bursal samples were collected from dead broiler (n=40) and layer (n=30) chickens showing specific lesions of IBD from seven commercial poultry farms of three different districts (Mymensingh, Chittagong and Tangail) of Bangladesh during the year 2007. Five representative bursal samples from each farm were used for the isolation of IBDVs using 9-day-old embryonated eggs of seronegative flock of layer birds and for identification the samples were subjected to agar gel immunodiffusion test (AGIDT), immunohistochemistry (IHC) and reverse transcription-polymerase chain reaction (RT-PCR). Out of 35 bursal samples, IBDVs were successfully isolated from 28 (80%) samples. By AGIDT, 32 (91.4%) samples were found positive for IBDV antigen. Results of AGIDT clearly indicated that IBDVs detected in 29 bursal samples of six affected farms were identical to each other but not to IBDVs present in the remaining three samples of another farm. Indirect immunoperoxidase staining of the bursal sections revealed the presence of IBDV antigen in 32 (91.4%) samples and the IBDV antigen was detected mainly in the cortex of the lymphoid follicles of the bursal tissues. In histopathology, cell depletion, atrophy and necrosis were observed in many bursal follicles with severe edema of interfollicular septa. Of the 35 bursal samples, 34 (97.1%) samples generated 254 bp product by RT-PCR. In conclusion, the results of virus isolation and identification by AGIDT, IHC and the analysis of viral genome by RT-PCR confirmed the outbreaks of acute IBD in commercial poultry of Bangladesh. Moreover, histopathological findings and results of AGIDT gave a clear indication that the isolates from six outbreaks were different from classical strain and it seems to be of very virulent strain. On the other hand, the isolates from the other outbreak were similar to the classical strain.

Analysis of the expression of platelet antigens CD9 and CD41/61 in transgenic rabbits with the integrated human blood clotting factor VIII gene construct

The objective of this research was to study the expression of cell membrane molecules CD9 and CD41/61 of transgenic rabbit with integrated human factor VIII (rhFVIII) gene construct. The expressions of these molecules have been monitored during two lactations of transgenic rabbits and simultaneously compared with the expression of the same molecules of non-transgenic rabbits. The immunochemical analysis by indirect immunofluorescence, ELISA and indirect immunoperoxidase staining of blood cells and udder tissues show that the insertion of the WAP-hFVIII gene construct into the rabbit genome, do not influence the expression of cell membrane antigens CD9 and CD41/61 on the blood platelets, polymorphonuclear blood cells, milk somatic cells and mammary gland tissues.

Effect of combined treatment with FK506, FTY720, and ex vivo graft irradiation in rat small bowel transplantation: Expression of mucosal addressin cell adhesion molecule-1

We studied the effect of the combined treatment with FK506, FTY720, and ex vivo graft irradiation. Five groups of SBT animals were studied on days 3, 5, and 7 after operation (untreated, FK506, FTY720, FK506 + FTY720, FK506 + FTY720 + irradiation). Indirect immunoperoxidase staining was performed against CD4 and MAdCAM-1. The numbers of CD4 positive cells in allografts were also analyzed by flow cytometry. The graft survival was prolonged in all of the FK506- and FTY720-treated groups. SBT allografts treated by FK506 and FTY720 demonstrated less infiltration of CD4 positive cells, but the irradiation group did not show any effects on its expression. In FK506- and FTY720-treated groups, MAdCAM-1 expression on the HEVs in PPs was up-regulated, and its expression on the ECVs in the LP was down-regulated compared with other allograft groups. Irradiation did not show any effects on MAdCAM-1 expression on both HEVs in PPs and ECVs in LP. FK506 and FTY720 prevented the infiltration of CD4 positive cells, the down-regulation of MAdCAM-1 expression on HEVs in PPs, and the up-regulation of MAdCAM-1 expression on ECVs in LP during the early phase of SBT.

秋田県において15 年ぶりに確認された古典型つつが虫病の1 例

We report the confirmation of classical tsutsugamushi disease in August 2008. A 17-year-old woman seen for fever and eschar on the back reported having been bitten by an insect nine days earlier while fishing on the Omonogawa river. The suspected culprit was Leptotrombidium akamushi. During convalescence serum IgM and IgG antibody titers rose significantly against the Kato serotype antigen in indirect immunoperoxidase staining. Epidemiology, clinical symptoms and the antibodies detected suggested classical tsutsugamushi disease infection. Such disease transmitted by L. akamushi have not been reported since 1993 in Akita Prefecture. The public should thus be informed about Orientia tsutsugamushi prevention, in case such disease re-care in this area in the future.

Biochemical and genetic investigation of round-headed spermatozoa in infertile men including two brothers and their father.

Acrosin and the outer acrosomal membrane (OAM) were studied in the spermatozoa of 9 infertile patients who differed in the number of round-headed spermatozoa between 14 and 71% in their ejaculates. These sperm components were also investigated in two infertile brothers who exhibited exclusively round-headed spermatozoa in their ejaculates, and in their fertile father. It turned out that round-headed spermatozoa lack both acrosin and the OAM as studied by indirect immunofluorescent and immunoperoxidase staining technique, gelatinolysis tests and by acrosin activity measurements. The normally shaped spermatozoa of 6 of the 9 infertile patients were found to be positive for acrosin and the OAM as expected, but in the remaining three patients even these spermatozoa were abnormal; in one patient they were unstainable for acrosin and in two patients they were unstainable both for acrosin and the OAM. These results have been confirmed by studies with the gelatinolysis test. The father of the two brothers with exclusively acrosomeless spermatozoa had more than 94% of normally shaped spermatozoa. However, only 10% of these spermatozoa were acrosin positive and only 30% were positive for the OAM. On the basis of these results we postulate that the mode of inheritance of the round-headed spermatozoa syndrome is polygenic rather than monogenic as suggested by previous authors.

Spotted Fever GroupRickettsiasp. Closely Related toR. japonica,Thailand

To the Editor: In response to a recent report that suggested human infection with Rickettsia japonica in northeastern Thailand (1), we phylogenetically reexamined spotted fever group rickettsiae (SFGR) from Thailand. The organism had been isolated from a male Haemaphysalis hystricis tick found on Mt. Doi Suthep, Chiang Mai, northern Thailand, in December 2001. The strain was designated TCM1 and was not distinguishable from R. japonica by indirect immunoperoxidase stain using monoclonal antibody (2). After propagating strain TCM1 in L-929 cell culture, we extracted DNA by using a Wizard Genomic DNA Purification Kit (Promega, Madison, WI, USA). We subjected the DNA to sequencing that targeted a 491-bp fragment of rickettsial outer membrane protein A (ompA), a 394-bp fragment of the rickettsial genus–specific 17-kDa antigen gene, and a 1,250-bp fragment of citrate synthase gene (gltA). Direct sequencing of amplicons was performed as previously described (3). Phylogenetic analyses based on ompA indicated that strain TCM1 was closely related to and clustered within the same clade as R. japonica strain YH (98.4% identity) (Figure, panel A). Also, a 17-kDa antigen gene obtained from strain TCM1 showed 99.5% identity to the corresponding gene of R. japonica (Figure, panel B). Our phylogenetic analysis with ompA and 17-kDa antigen gene showed that strain TCM1 was closely related to R. japonica but distinguished from Rickettsia sp. PMK94 (which was closely related to R. heilongjiangensis from northeastern China) (3); another SFGR agent, R. honei from Ixodes granulatus ticks in Thailand (4), was apparently different from strain TCM1 (Figure). Phylogenetic analyses based on gltA (99.4%–99.6% identity) showed that strain TCM1 is also closely related to R. japonica and Rickettsia sp. strain PMK94 (data not shown). Thus, we describe the R. japonica group in Thailand. R. japonica is the specific pathogen of Japanese spotted fever, which has been found mainly in southwestern Japan (5). The present strain, closely related to R. japonica, is likely to have been isolated from H. hystricis in Thailand because R. japonica frequently has been isolated, or detected by PCR, from the same tick species in Japan (6). Such tick species–specificity of SFGR should be considered when speculating on any geopathologic relationships of rickettsioses among different SFGR-endemic areas. Previous reports on spotted fever–positive results of human serosurveys (7,8) and on a clinical case (9) in northern Thailand may provide epidemiologic background. In Asia, multiple species of rickettsiea (e.g., R. japonica, R. heilongjiangensis, R. honei) are the causative agents of spotted fever rickettsioses, so the agent closely related to R. japonica could cause spotted fever in Thailand. Additionally, R. japonica has been found in Korea (10), and our current study indicates that R. japonica and its genetic variants are widely distributed in Far Eastern countries, including Japan (Grant-in-Aid for International Cooperative Research, unpub. data). Therefore, the epidemiology and genetic variation of SFGR throughout Asia should be examined by molecular studies.

Expression of Intercellular Adhesion Molecule-1 in the Livers of Rats Treated with Diethylnitrosamine

It has been reported that levels of soluble intercellular adhesion molecule-1 (ICAM-1) in the blood are elevated in hepatocellular carcinoma patients. In the present study, serial observations of the localization of ICAM-1 in the liver were made by light and electron microscopy in rats with carcinogen-induced cancer. Male Fisher rats were given diethylnitrosamine (DEN) orally in their drinking water. Rats were sacrificed at 6, 8, 12, or 14 weeks after the start of DEN administration and the liver tissue was collected. ICAM-1 expression in liver was assessed using indirect immunoperoxidase staining with anti-rat ICAM-1 antibody. Although ICAM-1 expression by endothelial cells in livers of DEN-treated rats was lower than in the control group at 8 weeks, it was higher in the membrane and cytoplasm of hepatocytes. The expression of ICAM-1 in mesenchymal cells was decreased, paralleling development of cellular atypia, whereas in hepatocyte membranes and cytoplasm it was increased in these atypia. ICAM-1 was localized to the cytoplasm of cancer cells, but to the membrane of hepatocytes in the treated livers at 14 weeks. Furthermore, the levels of ICAM-1 in mesenchymal cells tended to be lower in the cancerous area than in the atypical hyperplastic nodule, and were reduced as the density of cell atypia increased, in comparison to cells in areas without cancerous nodules. We concluded that ICAM-1 may be influenced the development of cancer induced in the rat liver by a chemical carcinogen.
