Crop residue heterogeneity: Decomposition by potential indigenous ligno-cellulolytic microbes and enzymatic profiling

The continuous depletion of fossil resources, energy-crisis and environmental pollution has gained popularity for careful selection of suitable microbial consortium to efficiently decompose crop residue and facilitate nutrient cycling. While crop residue is commonly incorporated into soil, the impact of the heterogeneity of residue on decomposition and biological mechanisms involved in extracellular carbon (C) cycle related enzyme activities remain not fully understood. To address this problem, an incubation study was conducted on chemical heterogeneity of straw and root residue with indigenous ligno-cellulolytic microbial consortium on extracellular enzymes as their activity is crucial for making in-situ residue management decisions under field condition. The activity of extracellular enzymes in different substrates showed differential variation with the type of enzyme and ranged from 16.9 to 77.6 µg mL−1, 135.7 to 410.8 µg mL−1, 66.9 to 177.1 µg mL−1 and 42.1 to 160.9 µg mL−1 for cellulase, xylanase, laccase and lignin peroxidase, respectively. Extracellular enzyme activities were sensitive to heterogeneity of biochemical constituent's present in straw and root residues and enhanced the decomposition processes with indigenous ligno-cellulolytic microbial consortium (Bacillus altitudinis, Streptomyces flavomacrosporus and Aspergillus terreus). Correlation matrix elucidated A. terreus and B. altitudinis as potential indigenous ligno-cellulolytic microbial inoculant influencing soil enzymatic activity (p < 0.001). This research work demonstrates a substantial impact of chemically diverse crop residues on the decomposition of both straw and root. It also highlights the pivotal role played by key indigenous decomposers and interactions between different microorganisms in governing the decomposition of straw and root primarily through release of extracellular enzymes. Consequently, it is novel bio-emerging strategy suggested that incorporation of the crop residues under field conditions should be carried out in conjunction with the potential indigenous ligno-cellulolytic microbial consortium for efficient decomposition in a short period of time under sustainable agriculture system.

Advancing biodegradation of petroleum contaminants by indigenous microbial consortia through assembly strategy innovations

Our demand for petroleum has challenged Earth's ecosystems, causing extensive water and soil pollution. Petroleum contamination treatment technology has relied on a range of biochemical processes to treat hydrocarbon-contaminated wastes, of which microbial degradation is one of the most economical and ecological. Engineered microbialconsortia can solve complex and difficult-to-degrade contaminants. However, the functional stability of consortia is challenged during long-term co-cultivation. Here, we introduced a novel strategy to design a stable consortium for degrading hydrocarbon pollutants, developed from ecological coexistence theory and trait-based method. Specifically, the degradation rate of the consortium constructed using the five strains was 31.54% higher than that of a single strain in a 6% (m/v) petroleum concentration after 16 days of co-culture. Besides, molecular characterization of the composition and transformation of petroleum-based substances further reveals the functional stability of consortium and the degradation mechanism and pattern of hydrocarbons by the consortium. Two independent soil experiments showed that the treatment of oil-contaminated soil by consortia from the indigenous microbiota maximizes the removal of hydrocarbon contaminants. Collectively, this strategy is generally applicable, facilitates the design of engineered microbial consortia, and provides a foundation for solving environmental pollution.

Recirculating packed-bed biofilm photobioreactor combined with membrane ultrafiltration as advanced wastewater treatment

Packed-bed biofilm photobioreactor combined with ultrafiltration membrane was investigated for intensifying the process for secondary wastewater effluent treatment. Cylindrical glass carriers were used as supporting material for the microalgal-bacterial biofilm, which developed from indigenous microbial consortium. Glass carriers allowed adequate growth of the biofilm with limited suspended biomass. Stable operation was achieved after a start-up period of 1000 h, where supernatant biopolymer clusters were minimized and complete nitrification was observed. After that time, biomass productivity was 54 ± 18 mg·L−1·day−1. Green microalgae Tetradesmus obliquus and several strains of heterotrophic nitrification–aerobic denitrification bacteria and fungi were identified. Combined process exhibited COD, nitrogen and phosphorus removal rates of 56 ± 5%, 12 ± 2% and 20 ± 6%, respectively. Membrane fouling was mainly caused by biofilm formation, which was not effectively mitigated by air-scouring aided backwashing.

Coupling of selenate reduction and pyrrhotite oxidation by indigenous microbial consortium in natural aquifer

Pyrrhotite is ubiquitously found in natural environment and involved in diverse (bio)processes. However, the pyrrhotite-driven bioreduction of toxic selenate [Se(VI)] remains largely unknown. This study demonstrates that Se(VI) is successfully bioreduced under anaerobic condition with the participation of pyrrhotite for the first time. Completely removal of Se(VI) was achieved at initial concentration of 10 mg/L Se(VI) and 0.56 mL/min flow rate in continuous column experiment with indigenous microbial consortium and pyrrhotite. Variation in hydrochemistry and hydrodynamics affected Se(VI) removal performance. Se(VI) was reduced to insoluble Se(0) while elements in pyrrhotite were oxidized to Fe(III) and SO42−. Breakthrough study indicated that biotic activity contributed 81.4 ± 1.07% to Se(VI) transformation. Microbial community analysis suggested that chemoautotrophic genera (e.g., Thiobacillus) could realize pyrrhotite oxidation and Se(VI) reduction independently, while heterotrophic genera (e.g., Bacillus, Pseudomonas) contributed to Se(VI) detoxification by utilizing metabolic intermediates generated through Fe(II) and S(-II) oxidation, which were further verified by pure culture tests. Metagenomic and qPCR analyses indicated genes encoding enzymes for Se(VI) reduction (e.g., serA, napA and srdBAC), S oxidation (e.g., soxB) and Fe oxidation (e.g., mtrA) were upregulated. The elevated electron transporters (e.g., nicotinamide adenine dinucleotide, cytochrome c) promoted electron transfer from pyrrhotite to Se(VI). This study gains insights into Se biogeochemistry under the effect of Fe(II)-bearing minerals and provides a sustainable strategy for Se(VI) bioremediation in natural aquifer.

Molecular characterization of indigenous microbes and its potential as a biological control agent of Fusarium stem rot disease (Fusarium verticillioides) on maize

Fusarium stem rot disease caused by Fusarium verticillioides has become one of the most serious issues confronting Indonesian farmers in recent years. An alternative option for suppressing this disease is to use indigenous microbes as an eco-friendly method to reduce synthetic fungicides. The objective of the research was to identify the molecular characteristics and effectiveness of an indigenous microbial consortium in controlling Fusarium stem rot disease. Identification of indigenous microbes is carried out based on molecular characters using universal primers, namely ITS-1/ITS-4 for fungi and 27F/1492R for bacteria. Nucleotide sequences were analyzed using Bioedit 7.2 version and MEGAX software. In vitro testing was carried out using the dual culture method for indigenous fungi and the disc diffusion method for indigenous bacteria. Meanwhile, in planta testing was conducted by evaluating a consortium of fungi and bacteria to control F. verticillioides in the field using a randomized block design with three replications, followed by a 5% DMRT test. The use of universal primer pairs ITS-1/ITS-4 and 27F/1492R succeeded in amplifying DNA bands of indigenous microbial isolates measuring ±600 bp and ±1465 bp, respectively. S6 and S9 bacterial isolates were identified as Bacillus cereus. JRP 7 and SEDF 6A isolates were identified as Trichoderma asperellum and JRP 10 isolate was identified as Penicillium raperi. All identification resulted in homology of >99%. The in vitro inhibitory reactions of isolates JRP 7, JRP 10, SEDF 6A, S6, and S9 against F. verticillioides were >60%. Disease severity of B6B9C10, B6B9C6, B6B9C7, B9C6C7, B9C7C10, and C6C7C10 treatments significantly proved their ability to control F. verticillioides in the field with a lower percentage of disease severity than positive controls, which are 23.33%, 18.89%, 23.33%., 21.85%, 14.07%, and 15.93%, respectively. The B9C7C10 consortium (S9 + JRP 7 + JRP 10 isolates) containing three species of indigenous microbes, i.e. B. cereus, T. asperellum, and P. raperi is the most effective at controlling F. verticillioides and may be developed for use as biopesticide products.

Potential of indigenous ligno-cellulolytic microbial consortium to accelerate degradation of heterogenous crop residues.

Indigenous microbial diversity has potential for rapid decomposition of residue through enzyme activities that is alternative, effective, and environment friendly strategy to accelerate degradation of lignocellulose in agricultural residues and make composting process economically viable. Keeping this view, the main objective of the present study was isolation and characterization of lignocellulosic degrading microbial diversity from long-term residue management practice experiments and to develop potential microbial consortium for rapid degradation of lignocellulosic biomass. In this study, twenty-five bacteria, nine fungi, and four actinomycetes isolates were obtained from the soil samples of different residue management fields from Ludhiana, Punjab, India. All isolates were qualitatively and quantitatively screened for enzyme activities, i.e., cellulase, xylanase, laccase, and lignin peroxidase. On the basis of quantitative estimation of enzyme activities, 3 fungal (S1F1, S2F4, and S6F9), 2 actinomycetes (S1A1 and S6A4), and 2 bacterial strains (S6B16 and S6B17) were further selected for in vitro bio-compatibility assay. Selected bio-compatible microbial strains were identified as Streptomyces flavomacrosporus (S6A4), Aspergillus terreus (S2F4), and Bacillus altitudinis (S6B16) through 16S rRNA and 18S rRNA sequencing. Furthermore, single and developed microbial consortium (S6B16 + S6A4 + S2F4) were screened for quantitative estimation of cellulase, xylanase, laccase, and lignin peroxidase enzymes with 23 biochemically different cereal, legume, and oil seed crop residues for optimization of enzyme activities at different time intervals. Results revealed that Vigna radiata followed by Cajanus cajan and Arachis hypogaea straw residue powder @ 1% in culture broth are a promising carbon source for B. altitudinis, S. flavomacrosporus, and A. terreus to produce higher ligno-cellulolytic microbial degrading enzymes due to variable range of carbon (C):nitrogen (N) ratio and higher ligno-cellulolytic content in the studied crop residues. Thus, the application of indigenous microbial consortium with efficient lignocellulose hydrolysis enzyme machinery might be an attractive alternative for ex situ crop residue management practices under sustainable manners.

Enhanced microbial degradation of PET and PS microplastics under natural conditions in mangrove environment

In-situ bioremediation of mangrove soil contaminated with polyethylene terephthalate (PET) and polystyrene (PS) microplastics was investigated using indigenous microbial consortium with adequate capacity to degrade the plastics. Eight (8) bacteria were isolated from plastic/microplastic-inundated mangrove soil and screened for the ability to degrade PET and PS microplastics. Optical density at 600 nm and colony forming unit counts were measured to evaluate the growth response of the microbes in the presence of PS and PET microplastics at different times of exposure. Structural and surface changes that occurred post biodegradation on the microplastics were determined through EDS and SEM analysis. The obtained results demonstrated the elongation and disappearance of peaks, suggesting that the microbial consortium could modify both types of microplastics. The overall results of the microplastic degradation showed varied degrees of weight loss after 90 experimental days, with the treated plot recorded 18% weight loss. The augmented soil was increased in the concentrations of Si S, and Fe and decreased in the concentrations of C, O, Na, Mg, Al, Cl, and K after bioremediation.

DEGRADATION OF POLYETHYLENE PLASTIC WASTE BY INDIGENOUS MICROBIAL CONSORTIUM AND FUNGI

Aim: The aims of this research is to determine the ability of an indigenous microbial consortium to degrade Low Density Polyethylene plastics. The plastic was cut mechanically into 1 x 1 cm2 pieces because the smaller the size of the plastic, the larger the surface area. The samples are input in an Erlenmeyer flask containing indigenous microbial consortium and fungi (20% v/v) and Stone Mineral Salt solution media (80% v/v). This research lasted 10 days with a pH of 7 controlled and temperature variations of 25, 30, 35°C. As preliminary research, the sensitivity test seeks to demonstrate that the indigenous microbial consortium and fungi are resistant or insensitive to LDPE. The degradation of LDPE plastic was analyzed using gravimetric methods, Fourier Transform Infrared, and a scanning electron microscope. Methodology and results: According to the results of gravimetric and FTIR analysis, the highest removal value was at a temperature variation of 30°C. The gravimetric analysis revealed that the weight loss in LDPE plastic was 0.0082 gr to 0,0074 gr or 9.76 %, while the FTIR analysis revealed that the intensity removal result was 6,27 %. Conclusion, significance, and impact of study: Scanning Electron Microscope (SEM) analysis revealed morphological changes on the surface of LDPE plastic samples, confirming these findings. Several factors influence the changes that occur in this study's LDPE plastic samples.

The role of available phosphorous in vanadate decontamination by soil indigenous microbial consortia

Indigenous microbial consortia are closely associated with soil inherent components including nutrients and minerals. Although indigenous microbial consortia present great prospects for bioremediation of vanadate [V(V)] contaminated soil, influences of some key components, such as available phosphorus (AP), on V(V) biodetoxification are poorly understood. In this study, surface soils sampled from five representative vanadium smelter sites were employed as inocula without pretreatment. V(V) removal efficiency ranged from 81.7 ± 1.4% to 99.5 ± 0.2% in batch experiment, and the maximum V(V) removal rates were positively correlated with AP contents. Long-term V(V) removal was achieved under fluctuant hydrodynamic and hydrochemical conditions in column experiment. Geobacter and Bacillus, which were found in both original soils and bioreactors, catalytically reduced V(V) to insoluble tetravalent vanadium. Phosphate-solubilizing bacterium affiliated to Gemmatimonadaceae were also identified abundantly. Microbial functional characterization indicated the enrichment of phosphate ABC transporter, which could accelerate V(V) transfer into intercellular space for efficient reduction due to the structural similarity of V(V) and phosphate. This study reveals the critical role of AP in microbial V(V) decontamination and provides promising strategy for in situ bioremediation of V(V) polluted soil.

A comprehensive feasibility study of effectiveness and environmental impact of PAH bioremediation using an indigenous microbial degrader consortium and a novel strain Stenotrophomonas maltophilia CPHE1 isolated from an industrial polluted soil

Polycyclic Aromatic Hydrocarbons (PAHs) are major toxic and recalcitrant pollutants in the environment. This study assessed the capacity of an isolated soil microbial consortium (OMC) to biodegrade PAHs. OMC was able to reach 100% biodegradation of naphthalene, acenaphthylene, acenaphthene, fluorene and phenanthrene in solution, and up to 76% and 50% of anthracene and fluoranthene, respectively, from a mix of 16 PAHs. To measure phenanthrene (PHE) mineralization, OMC and eight strains isolated from OMC were used and identified by PCR amplification of the gene 16S ribosomal RNA. A novel Stenotrophomonas maltophilia CPHE1, not previously described as a PAH degrader, was able to mineralize almost 40% PHE and biodegrade 90.5% in solution, in comparison to OMC that reached 100% PHE degradation, but only 18.8% mineralization. Based on metabolites identified during PHE degradation and on the detection of two genes (PAH RHDα and nahAc) in OMC consortium, two possible via were described for its degradation, through salicylic and phthalic acid. PAH RHDα, which codified the first step on PHE biodegradation pathway, was also found in the DNA of S. maltophilia CPHE1. An ecotoxicology study showed that PHE bioremediation after inoculating S. maltophilia CPHE1 for 30 days decreased by half the solution toxicity.

Biodegradation of anthraquinone dyes: Interactive assessment upon biodecolorization, biosorption and biotoxicity using dual-chamber microbial fuel cells (MFCs)

In this work, indigenous microbial consortium was employed to degrade four (4) anthraquinone dyes. Apart from biodecolorization, the interactive effects of biosorption and biotoxicity via microbial fuel cells (MFCs) were investigated thereby suggesting feasible strategies to assess toxicities of anthraquinone dyes. Serial microbial acclimatization upon indigenous mixed consortia was first implemented to obtain optimal consortia for decolorization. The well-acclimated consortium was capable to degrade 50 mg L−1 Acid blue 62 (AB62) at 47.07 ± 1.411 mg L−1 h−1. The ranking of decolorization performance at 50 mg L−1 was AB62 > Acid Blue 25 (AB25) > Acid Blue 40 (AB40) > Reactive Blue 19 (RB19). The appreciably decreased rates of decolorization at higher concentrations (300 mg L−1) was mainly biosorption due to dye inhibition. However, as power densities in MFCs at < 300 mg L−1 were still not significantly attenuated, indicating that dyes at such levels were still under threshold of toxicity for bioenergy extraction. After decolorization, considerable increases in power generation were exhibited due to accumulated degraded intermediates as electron shuttles. Microbial community analysis suggested that an increase in species diversity and abundance of both dye-decolorizing and electroactive bacteria was revealed after the consortium underwent successful serial acclimation.

Effect of Indigenous Microbial Consortium on Bioleaching of Arsenic from Contaminated Soil by Shewanella putrefaciens

The effects of indigenous microbial consortium on removal of As from As-contaminated soil using an Fe(III)-reducing bacterium Shewanella putrefaciens were investigated under circumneutral pH condition. Sequential extraction of As revealed that more than 30% of As was associated with Fe(III)-(oxy)hydroxides in the soil. Bioleaching experiments were conducted anaerobically with a supply of lactate as a carbon source. The highest As removal efficiency (57.5%) was obtained when S. putrefaciens and indigenous bacterial consortium coexisted in the soil. S. putrefaciens and indigenous bacteria solely removed 30.1% and 16.4% of As from the soil, respectively. The combination of S. putrefaciens and indigenous bacteria led to a higher amount of labile As after microbial dissolution of Fe(III)-(oxy)hydroxides. After microbial treatment, soil quality represented by pH and organic content appeared to be preserved. The results indicated that the ecological and physiological understanding of the indigenous microbiome might be important for the efficient application of bioleaching technology to remove As from contaminated soils.

Enrichment and key features of a robust and consistent indigenous marine-cognate microbial consortium growing on oily bilge wastewaters.

Oily bilge wastewater (OBW) is a hazardous hydrocarbon-waste generated by ships worldwide. In this research, we enriched, characterized and study the hydrocarbon biodegradation potential of a microbial consortium from the bilges of maritime ships. The consortium cZ presented a biodegradation efficiency of 66.65% for total petroleum hydrocarbons, 72.33% for aromatics and 97.76% removal of n-alkanes. This consortium showed the ability to grow in OBWs of diverse origin and concentration. A 67-fold increase in biomass was achieved using a Sequential Batch Reactor with OBW as the only carbon and energy source. The bacterial community composition of the enriched OBW bacterial consortium at the final stable stage was characterized by 16S amplicon Illumina sequencing showing that 25 out of 915 of the emerged predominant bacterial types detected summed up for 84% of total composition. Out of the 140 taxa detected, 13 alone accumulated 94.9% of the reads and were classified as Marinobacter, Alcanivorax, Parvibaculum, Flavobacteriaceae, Gammaproteobacteria PYR10d3, Novispirillum and Xanthomonadaceae among the most predominant, followed by Thalassospira, Shewanella, Rhodospirillaceae, Gammaprotobacteria, Rhodobacteriaceae and Achromobacter. The microbial community from OBW bioreactor enrichments is intrinsically diverse with clear selection of predominant types and remarkably exhibiting consistent and efficient biodegradation achieved without any nutrient or surfactant addition. Due to there is very little information available in the OBW biodegradation field, this work contributes to the body of knowledge surrounding the treatment improvement of this toxic waste and its potential application in wastewater management.

Application of enhanced bioreduction for hexavalent chromium-polluted groundwater cleanup: Microcosm and microbial diversity studies

Hexavalent chromium (Cr6+) is a commonly found heavy metal at polluted groundwater sites. In this study, the effectiveness of Cr6+ bioreduction by the chromium-reducing bacteria was evaluated to remediate Cr6+-contaminated groundwater. Microcosms were constructed using indigenous microbial consortia from a Cr6+-contaminated aquifer as the inocula, and slow-releasing emulsified polycolloid-substrate (ES), cane molasses (CM), and nutrient broth (NB) as the primary substrates. The genes responsible for the bioreduction of Cr6+ and variations in bacterial diversity were evaluated using metagenomics assay. Complete Cr6+ reduction via the biological mechanism was observed within 80 days using CM as the carbon source under anaerobic processes with the increased trivalent chromium (Cr3+) concentrations. Cr6+ removal efficiencies were 83% and 59% in microcosms using ES and NB as the substrates, respectively. Increased bacterial communities associated with Cr6+ bioreduction was observed in microcosms treated with CM and ES. Decreased bacterial communities were observed in NB microcosms. Compared to ES, CM was more applicable by indigenous Cr6+ reduction bacteria and resulted in effective Cr6+ bioreduction, which was possibly due to the growth of Cr6+-reduction related bacteria including Sporolactobacillus, Clostridium, and Ensifer. While NB was applied for specific bacterial selection, it might not be appropriate for electron donor application. These results revealed that substrate addition had significant impact on microbial diversities, which affected Cr6+ bioreduction processes. Results are useful for designing a green and sustainable bioreduction system for Cr6+-polluted groundwater remediation.

Assessing pretreatment and seawater reverse osmosis performance using an ATP-based bacterial growth potential method

Various bacterial growth potential (BGP) methods have been developed recently to monitor biofouling in seawater reverse osmosis (SWRO) systems such as assimilable organic carbon and bacterial regrowth potential. However, the relationship between these methods and biofouling in SWRO desalination plants has not yet been demonstrated. In this research, an attempt is made to investigate if a correlation exists between BGP of SWRO feed water and the chemical cleaning frequency in SWRO plants using an ATP-based BGP method employing an indigenous microbial consortium. Using ATP-based BGP method at 5 different seawater locations showed low variations of bacterial yield.The BGP method was applied to assess the pretreatment performance of three full-scale SWRO plants with different pretreatment processes. Dual media filtration (DMF) showed the highest BGP removal (>50%) in two SWRO plants. Removal of BGP and hydrophilic organic carbon in dissolved air floatation combined with ultrafiltration was similar to the removal achieved with DMF in combination with inline coagulation. For the three SWRO plants investigated, a higher BGP in SWRO feed water corresponded to a higher chemical cleaning frequency. However, more data is required to confirm if a real correlation exists between BGP and biofouling in SWRO plants.

Diesel uptake by an indigenous microbial consortium isolated from sediments of the Southern Gulf of Mexico: Emulsion characterisation

In this study a microbial consortium, dominated by members of the genera Marinobacter and Alcanivorax (Gammaproteobacteria) isolated from marine sediments of Southern Gulf of Mexico, was assessed to grow in a bubble column bioreactor using 13 g L−1 of diesel (aliphatic and aromatic hydrocarbons mix including nonane and hexadecane) as the sole carbon source. The consortium was able to produce 3.3 g L−1 of biomass, measured as suspended solids. Microbial growth was detectable, even substrate depletion, after 8 days of cultivation. The emulsifier activity and its influence on the droplet size were also evaluated: it was observed that droplet diameter decreases as emulsifier activity increases. The bubble column bioreactor system proposed in this research could be used as a biotechnological process for the remediation of a contaminated body in important petrochemical regions, for example, Veracruz, México, where some points of sea and fresh-water bodies were analysed to find nonane and hexadecane in all sample water. It is important due to a lack of information, regarding hydrocarbon pollution in this port area, is filled.

Bioremediation of contaminated urban river sediment with methanol stimulation: Metabolic processes accompanied with microbial community changes

The intense pollution of urban river sediments with rapid urbanization has attracted considerable attention. Complex contaminated sediments urgently need to be remediated to conserve the ecological functions of impacted rivers. This study investigated the effect of using methanol as a co-substrate on the stimulation of the indigenous microbial consortium to enhance the bioremediation of petroleum hydrocarbons (PHs) and polycyclic aromatic hydrocarbons (PAHs) in an urban river sediment. After 65 days of treatment, the PAHs degradation efficiencies in the sediment adding methanol were 4.87%–40.3% higher than the control. The removal rate constant of C31 was 0.0749 d−1 with 100 mM of supplied methanol, while the corresponding rate was 0.0399 d−1 in the control. Four-ring PAHs were effectively removed at a degradation efficiency of 65%–69.8%, increased by 43.3% compared with the control. Sulfate reduction and methanogenesis activity were detected, and methane-producing archaea (such as Methanomethylovorans, with a relative abundance of 25.87%–58.53%) and the sulfate-reducing bacteria (SRB, such as Desulfobulbus and Desulfobacca) were enriched. In addition, the chemolithoautotrophic sulfur-oxidizing bacteria (SOB, such as Sulfuricurvum, with a relative abundance of 34%–39.2%) were predominant after the depletion of total organic carbon (TOC), and markedly positively correlated with the PHs and PAHs degradation efficiencies (P < 0.01). The SRB and SOB populations participated in the sulfur cycle, which was associated with PHs and PAHs degradation. Other potential functional bacteria (such as Dechloromonas) were also obviously enriched and significantly positively correlated with the TOC concentration after methanol injection (P < 0.001). This study provides a new insight into the succession of the indigenous microbial community with methanol as a co-substrate for the enhanced bioremediation of complexly contaminated urban river sediments.

Reduction of Organic Load and Biodegradation of Palm Oil Mill Effluent by Aerobic Indigenous Mixed Microbial Consortium Isolated from Palm Oil Mill Effluent (POME)

This study was designed to determine the potential of indigenous mixed microbial consortium isolated from palm oil mill effluent (POME) in reducing organic load and biodegradation of POME. Isolation and identification of indigenous microorganisms was subjected to standard microbiological methods and sequencing of the 16S rRNA and 18S rRNA genes. Sequencing of the 16S rRNA and 18S rRNA genes of the microbial strains suggests that they were identified as Micrococcus luteus101PB, Stenotrophomonas maltophilia102PB, Bacillus cereus103PB, Providencia vermicola104PB, Klebsiella pneumoniae105PB, Bacillus subtilis106PB, Aspergillus fumigatus107PF, Aspergillus nomius108PF, Aspergillus niger109PF, and Meyerozyma guilliermondii110PF. Results revealed that total percent reduction efficiency by the aerobic mixed microbial consortium for all bacteria–fungi combination (ABFC) gave a biochemical oxygen demand (BOD) reduction efficiency of about 90.23%, chemical oxygen demand (COD) 91.06%, and total suspended solids (TSS) 92.23% and bacteria–fungi stepwise (BFSW) recorded BOD reduction efficiency of 85.28%, COD 84.45%, and TSS 86.18% in 1000 mL of POME. The HPLC chromatogram results revealed increase in glucose level due to breakdown of cellulose which represents the cellulosic materials in POME by mixed microbial consortium signifying biodegradation of cellulose as a clean-up process for the tested POME sample. Therefore, the indigenous microbial strains are promising organisms for industrial applications. These microbes have direct applications in industrial process such as bioremediation and biodegradation of wastewaters.

Bioaugmentation of chlorothalonil-contaminated soil with hydrolytically or reductively dehalogenating strain and its effect on soil microbial community

Although bioaugmentation of pollutant-contaminated sites is a great concern, there are few reports on the relationships among indigenous microbial consortia, exogenous inocula, and pollutants in a bioaugmentation process. In this study, bioaugmentation with Pseudochrobactrum sp. BSQ1 and Massilia sp. BLM18, which can hydrolytically and reductively dehalogenate chlorothalonil (TPN), respectively, was studied for its ability to remove TPN from soil; the alteration of the soil microbial community during the bioaugmentation process was investigated. The results showed that TPN (50 mg/kg) was completely removed in both bioaugmentation treatments within 35 days with half-lives of 6.8 and 9.8 days for strains BSQ1 and BLM18 respectively. In high concentration of TPN-treated soils (100 mg/kg), the bioaugmentation with strains BSQ1 and BLM18 respectively reduced 76.7% and 62.0% of TPN within 35 days. The TPN treatment significantly decreased bacterial richness and diversity and improved the growth of bacteria related to the elimination of chlorinated organic pollutants. However, little influence on soil microbial community was observed for each inoculation treatment (without TPN treatment), showing that TPN treatment is the main force for the shift in indigenous consortia. This study provides insights into the effects of halogenated fungicide application and bioaugmentation on indigenous soil microbiomes.

Potential of dissimilatory nitrate reduction pathways in polycyclic aromatic hydrocarbon degradation

This study investigates the potential of an indigenous estuarine microbial consortium to degrade two polycyclic aromatic hydrocarbons (PAHs), naphthalene and fluoranthene, under nitrate-reducing conditions. Two physicochemically diverse sediment samples from the Lima Estuary (Portugal) were spiked individually with 25 mg L−1 of each PAH in laboratory designed microcosms. Sediments without PAHs and autoclaved sediments spiked with PAHs were run in parallel. Destructive sampling at the beginning and after 3, 6, 12, 30 and 63 weeks incubation was performed. Naphthalene and fluoranthene levels decreased over time with distinct degradation dynamics varying with sediment type. Next-generation sequencing (NGS) of 16 S rRNA gene amplicons revealed that the sediment type and incubation time were the main drivers influencing the microbial community structure rather than the impact of PAH amendments. Predicted microbial functional analyses revealed clear shifts and interrelationships between genes involved in anaerobic and aerobic degradation of PAHs and in the dissimilatory nitrate-reducing pathways (denitrification and dissimilatory nitrate reduction to ammonium - DNRA). These findings reinforced by clear biogeochemical denitrification signals (NO3− consumption, and NH4+ increased during the incubation period), suggest that naphthalene and fluoranthene degradation may be coupled with denitrification and DNRA metabolism. The results of this study contribute to the understanding of the dissimilatory nitrate-reducing pathways and help uncover their involvement in degradation of PAHs, which will be crucial for directing remediation strategies of PAH-contaminated anoxic sediments.
