Effect of a thiazole derivative on the activity of antioxidant enzymes in murine lymphoma cells

Previous in vitro studies have demonstrated a pronounced cytotoxic effect of the thiazole derivative N-(5-Benzyl-13-thiazole-2-yl)-35-dimethyl-1-benzofuran-2-carboxami­de (BF1) on tumor cells. Additionally, it has been determined that scavengers of reactive Oxygen species (ROS) significantly reduce the cytotoxic effect of BF1. In this study, the influence of BF1 on the activity of superoxide dismutase (SOD) and catalase (CAT), both normally and in the presence of ascorbic acid, in mouse Nemeth-Kellner lymphoma (NK/Ly) cells has been studied to evaluate the possible role of antioxidant activity during the action of this substance. The experiments were performed using nonlinear male mice weighing 20–30 g. Intraperitoneal inoculation of 10–15 million cancer cells into the mice induced the ascites form of lymphoma. The thiazole derivative (BF1) was dissolved in dimethyl sulfoxide and added to the test samples at 1, 10, and 50 μM final concentrations. Superoxide dismutase (SOD) and catalase (CAT) activities were determined spectrophotometrically in a homogenate of the lymphoma cells after incubation with the drug for 30 minutes. The baseline level of SOD in the lymphoma of the mice was 0.33±0.02 activity units/min×mg protein. BF1 significantly increased the enzyme activity by 35 % and 29 % at concentrations of 10 (p<0.01) and 50 μM (p<0.05), respectively. The baseline level of CAT activity was 4.61±0.17 nmoles H2O2/min×mg protein, and this significantly decreased by 15 % (p<0.05) and 20 % (p<0.01) following the action of the thiazole derivative at a concentration of 10 and 20 μM, respectively. The increase of SOD activity, coupled with a decrease or absence of changes in CAT activity, may be cytotoxic to cancer cells. Simultaneously, upon the addition of ascorbic acid as a scavenger of ROS to the environment, the activities of SOD and CAT did not change under the action of BF1 at any of the investigated concentrations. Therefore, the effect of the thiazole derivative BF1 has been canceled in the presence of ROS scavengers in the environment. This may indicate the dependence of the cytotoxic effect of BF1 on the presence of ROS in tumor cells.

Endurance exercise attenuates Gαq-RNAi induced hereditary obesity and skeletal muscle dysfunction via improving skeletal muscle Srl/MRCC-I pathway in Drosophila.

G protein alpha q subunit (Gαq) can binds to the G protein-coupled receptor (GPCR) for signaling and is closely related to lipid metabolism. Endurance exercise is an effective means of combating acquired obesity and its complications, but the mechanisms by which endurance exercise modulates hereditary obesity and its complications are unknown. In this study, we achieved knockdown of Gαq in drosophila adipose tissue and skeletal muscle by constructing the Gαq-UAS-RNAi/Ppl-Gal4 and Gαq-UAS-RNAi/Mef2-GAl4 systems. Drosophila were subjected a three-week endurance exercise intervention, and changes in relevant indicators were detected and observed by RT-PCR, ELISA, oil red staining, immunofluorescence staining, and transmission electron microscopy. The results showed that knockdown of Gαq in both adipose tissue and skeletal muscle induced a significant increase in triglycerides accompanied by a decrease in rapid climbing ability, a decrease in Superoxide Dismutase (SOD) activity level, and a decrease in Mitochondrial respiratory chain complexI (MRCC I) content in Drosophila whole body and skeletal muscle, and down-regulated the expression of the G protein alpha q subunit (Gαq), the skeletal muscle myosin heavy chain expression gene (Mhc), mitochondrial biogenesis gene Spargal(the PGC-1alpha homologue in Drosophila). Endurance exercise significantly improved the triglyceride levels in the whole body and skeletal muscle of drosophila with Gαq knockdown in adipose tissue and skeletal muscle, as well as their ability to climb, increased SOD activity level and MRCCI content level, and up-regulated the expression of Gαq, Mhc, and Spargal(Srl). Thus, the present findings suggest that genetic defects in the Gαq gene in adipose and skeletal muscle tissues induce hereditary obesity and skeletal muscle dysfunction, and that endurance exercise attenuates this hereditary obesity and concomitant skeletal muscle dysfunction in drosophila by improving skeletal muscle fiber contractile proteins, mitochondrial function and function, and antioxidant capacity via mediating the Gαq/Mhc, Gαq/Srl/MRCC-I, and Gαq/SOD pathways.

Effects of benzophenone-3 on the liver and thyroid of adult zebrafish

1. Benzophenone-3 (BP-3), commonly known as oxybenzone, is an organic compound that acts as a sunscreen, protecting the skin from UVA and UVB rays. Thus, the objective of this study was to investigate the effects of BP-3 on the liver and thyroid using morphological and biochemical approaches. 2. Adult male zebrafish were randomly assigned to three groups, each with three repetitions (n = 10 per group): water control, solvent control (0.01% ethanol), and 1 μg/L of BP-3, using a static exposure system for 96 hours. After the experiment, histopathological analyses of the liver and thyroid were performed, along with histochemical analyses (glycogen) and biochemical evaluations of the antioxidant enzymes Superoxide Dismutase (SOD) and Catalase (CAT). 3. Exposure to BP-3 resulted in significant histopathological changes in the liver of Danio rerio, increasing the frequency of circulatory disturbances, progressive changes, inflammatory responses, and regressive changes. On the other hand, the thyroid gland did not show any morphological changes during exposure to BP-3, maintaining its typical structure with follicles. There was a significant increase in SOD activity, while CAT showed no changes after 96 hours of exposure. 4. The results obtained demonstrate that exposure to BP-3 causes significant morphophysiological changes in the liver of D. rerio, highlighting not only the negative impacts on the health of these organisms but also the ecotoxicological potential of the substance and its consequences for aquatic biota in contaminated environments.

Kaempferol alleviates myocardial ischemia injury by reducing oxidative stress via the HDAC3-mediated Nrf2 signaling pathway

IntroductionKaempferol (KAE) is a flavonoid found in various plants. Recent studies showed that high dietary intake of KAE was associated with a lower risk of myocardial infarction; however, the cardioprotective mechanism of KAE remains unknown. ObjectivesTo determine the effect of KAE on cardiac injury in isoproterenol (ISO)-induced rats and cobalt chloride (CoCl2)-treated cardiomyocytes, and the underlying mechanisms. MethodsMale rats were pretreated with different doses of KAE for 14 days, and then injected with ISO to induce myocardial ischemia injury. We also established a model of myocardial cell injury using rat H9c2 cardiomyocytes stimulated with CoCl2. ResultsWe found that KAE pretreatment significantly alleviated myocardial injury and improved cardiac function in ISO-injected rats. In addition, KAE reduced oxidative stress in rats with myocardial ischemia by decreasing malondialdehyde concentration and increasing superoxide dismutase activity, and protection of the myocardial mitochondrial structure. KAE also attenuated CoCl2-induced injuryof H9c2 cardiomyocytes via suppression ofoxidative stress. With regard to the mechanism, we found that KAE down-regulated HDAC3 expression and up-regulated Nrf2 expression in ISO-induced rats and CoCl2-stimulated cardiomyocytes. Incubation of cardiomyocytes with HDAC3-selective inhibitor RGFP966 augmented the protective effect of KAE and reduced oxidative stress. By contrast, HDAC3 overexpression by adenovirus attenuated the effect of KAE on oxidative stress compared with KAE treatment group. HDAC3 also regulated Nrf2 expression in the cardiomyocytes with RGFP966 or an adenovirus overexpressing HDAC3; but Nrf2 inhibition reduced the effect of KAE on ROS generation in CoCl2-induced cardiomyocytes. Immunoprecipitation assay showed that HDAC3 interacted with Nrf2 in cardiomyocytes. Further studies found that KAE increased the acetylation level of Nrf2, while HDAC3 overexpression decreased the acetylation of Nrf2 compared with KAE treatment group. ConclusionOur data show that KAE ameliorates cardiac injury by reducing oxidative stress via the HDAC3-mediated Nrf2 signaling pathway in cardiomyocytes.

Probiotic and Rice-Derived Compound Combination Mitigates Colitis Severity

Background: This study investigated the ability of Enterococcus lactis (E. lactis) and Hasawi rice protein lysate (HPL) to suppress colitis induced by dextran sulfate sodium (DSS) in miceColitis is characterized by inflammation of the colon, and exploring potential therapeutic agents could lead to improved management strategies. Methods: Male mice were subjected to DSS treatment to induce colitis, followed by supplementation with E. lactis and/or HPL. The study assessed various parameters, including disease activity index (DAI) scores, gut permeability measured using FITC-dextran, and superoxide dismutase (SOD) activity in excised colon tissues from both treated and untreated control groups. Results: E. lactis supplementation significantly alleviated DSS-induced colitis, as evidenced by improved DAI scores and enhanced gut permeability. Notably, E. lactis combined with HPL (0.1 mg/108) exhibited superior tolerance to a 0.5% pancreatin solution compared to E. lactis alone. Both E. lactis and the combination treatment significantly increased SOD activity (5.6 ± 0.23 SOD U/mg protein for E. lactis and 6.7 ± 0.23 SOD U/mg protein for the combination) relative to the Azoxymethane (AOM)/DSS group, suggesting a reduction in oxidative stress. Additionally, pro-inflammatory markers were significantly reduced in the group receiving both E. lactis and HPL compared to the E. lactis-only group. Levels of proteins associated with cell death, such as PCNA, PTEN, VEGF, COX-2, and STAT-3, were significantly decreased by 14.8% to 80% following E. lactis supplementation, with the combination treatment showing the most pronounced effects. Conclusions: These findings suggest E. lactis supplementation may be beneficial for colitis, with HPL potential to enhance its effectiveness.

The herbicidal activity and mechanism of Talaromyces purpureogenus CY-1 metabolites.

Studies have shown that Talaromyces can produce a large number of secondary metabolites in its metabolic process, many of which have good insecticidal, antibacterial, antitumor, antiviral and other biological activities. In order to explore the herbicidal activity and mechanism of Talaromyces purpureogenus CY-1, we determined the inhibitory effect of the fermentation broth of the CY-1 strain on weeds, identified the major active components, and further investigated the herbicidal mechanism. The results showed that CY-1, with IC50 values of 5.40 g/L and 4.39 g/L, respectively, exhibited good herbicidal activity against Xanthium sibiricum and Amaranthus lividus. Spraying CY-1 strain fermentation broth on redroot pigweed resulted in plant protection efficiencies and fresh weight protection efficiencies of 83.7% and 87%, respectively. The active component identified in the broth was 2-(3-hydroxybenzoyloxy) acrylic acid. Treatment of Amaranthus lividus with 2-(3-hydroxybenzoyloxy) acrylic acid resulted in trends of increasing superoxide dismutase activity, peroxidase activity, respiratory rate, cytochrome oxidase activity, and soluble protein content, followed by a decrease. Peroxidase activity, relative conductance and malondialdehyde content gradually increased, while acetyl lactate synthase and glutamine synthetase initially decreased and gradually returned to normal. The soluble sugar content showed a gradual decrease. The findings of the present study indicate that 2-(3-hydroxybenzoyloxy) acrylic acid can induce oxidative stress in plants, damaging plant cell walls, weakening respiratory activity, inhibiting protein synthesis and sugar metabolism, ultimately leading to weed wilting and death. © 2024 Society of Chemical Industry.

Fortunellin attenuates sepsis-induced acute kidney injury by inhibiting inflammation and ferroptosis via the TLR4/NF-κB pathway.

To investigate the potential protective effect of fortunellin in sepsis-induced acute kidney injury (AKI) and its underlying mechanisms. Lipopolysaccharide (LPS)-treated human kidney proximal tubular epithelial (HK-2) cells were used as a cell model and sepsis-induced AKI was induced by cecal ligation and puncture (CLP) surgery in mice. Cell Counting Kit-8 (CCK8) assays and flow cytometry analysis were performed to examine the viability of HK-2 cells. Enzyme-linked immunosorbent assay (ELISA) was performed to investigate the content of tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and interleukin-1 beta (IL-1β) in vivo and in vitro. The levels of malondialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH), and free iron (Fe2+) were measured as indicators of ferroptosis. The phosphorylation levels of Interleukin-1 Receptor-Associated Kinase 4 (p-IRAK4), p65 (p-65), and inhibitor of kappa B alpha (p-IκBα) were detected by western blot as an indication of nuclear factor kappa-B (NF-κB) pathway activation. Our cell and animal experiments revealed that fortunellin exhibits significant anti-inflammatory and cytoprotective properties. Fortunellin counteracted LPS-induced cellular damage in HK-2 cells, enhancing cell survival and suppressing the secretion of pro-inflammatory cytokines. Additionally, fortunellin demonstrated potent antioxidant effects, reducing MDA and Fe2+ levels while increasing SOD activity and GSH content. The protective effect of fortunellin was further corroborated in the mouse model of sepsis-induced AKI. Notably, fortunellin suppressed activation of the TLR4/NF-κB pathway in the AKI model, as evidenced by decreased levels of p-p65 and p-IκBα proteins. Fortunellin ameliorates inflammation and oxidative stress in sepsis-induced AKI, possibly through the modulation of the TLR4/NF-κB pathway. These findings suggest fortunellin's potential as a therapeutic agent for sepsis-associated AKI.

Asiaticoside improves diabetic nephropathy by reducing inflammation, oxidative stress, and fibrosis: An in vitro and in vivo study

BACKGROUND Diabetic nephropathy (DN) is a severe microvascular complication of diabetes characterized by inflammation, oxidative stress, and renal fibrosis. Asiaticoside (AC) exhibits anti-inflammatory, antioxidant, and anti-fibrotic properties, suggesting potential therapeutic benefits for DN. This study aimed to investigate the protective effects of AC against DN and elucidate the underlying mechanisms involving the nuclear factor erythroid 2-related factor 2 (NRF2)/heme oxygenase-1 (HO-1) antioxidant pathway. AIM To investigate the renoprotective effects of AC against DN and elucidate the role of the NRF2/HO-1 pathway. METHODS The effects of AC on high glucose (HG)-induced proliferation, inflammation, oxidative stress, and fibrosis were evaluated in rat glomerular mesangial cells (HBZY-1) in vitro . A streptozotocin-induced DN rat model was established to assess the in vivo impact of AC on renal injury, inflammation, oxidative stress, and fibrosis. The involvement of the NRF2/HO-1 pathway was examined using pharmacological inhibition studies in the cell model. RESULTS AC inhibited HG-induced HBZY-1 cell proliferation and significantly improved various indicators of DN in rats, including reduced body weight, and elevated blood glucose, serum creatinine, blood urea nitrogen, and 24-h urine protein. Both in vitro and in vivo studies demonstrated that AC decreased inflammation and oxidative stress by reducing interleukin (IL)-6, IL-8, tumor necrosis factor-alpha, reactive oxygen species, and malondialdehyde levels while increasing superoxide dismutase activity. Additionally, AC suppressed the expression of fibrogenic markers such as collagen I, collagen IV, and fibronectin. AC activated NRF2 expression in the nucleus and increased HO-1 and NAD(P)H dehydrogenase (Quinone) 1 protein expression in renal tissues and HG-induced HBZY-1 cells. CONCLUSION AC improves DN by reducing inflammation, oxidative stress, and fibrosis through the activation of the NRF2/HO-1 signaling pathway. These findings not only highlight AC as a promising therapeutic candidate for DN but also underscore the potential of targeting the NRF2/HO-1 pathway in developing novel treatments for other chronic kidney diseases characterized by oxidative stress and inflammation.

Continuous Theta Burst Stimulation Inhibits Oxidative Stress-Induced Inflammation and Autophagy in Hippocampal Neurons by Activating Glutathione Synthesis Pathway, Improving Cognitive Impairment in Sleep-Deprived Mice.

Sleep deprivation (SD) has been reported to have a negative impact on cognitive function. Continuous theta burst stimulation (cTBS) shows certain effects in improving sleep and neurological diseases, and its molecular or cellular role in SD-induced cognition impairment still need further exploration. In this study, C57BL/6 mice were subjected to 48h of SD and cTBS treatment, and cTBS treatment significantly improved SD-triggered impairment of spatial learning and memory abilities in mice. Additionally, cTBS reduced malondialdehyde levels, increased superoxide dismutase activities, and inhibited the production of inflammatory cytokines, alleviating oxidative stress and inflammation levels in hippocampal tissues of SD model mice. cTBS decreased LC3II/LC3I ratio, Beclin1 protein levels, and LC3B puncta intensity, and elevated p62 protein levels to suppress excessive autophagy in hippocampal tissues of SD-stimulated mice. Then, we proved that inhibiting oxidative stress alleviated inflammation, autophagy, and death of hippocampal neuron cells through an in vitro cellular model for oxidative stress, and cTBS treatment promoted the production of glutathione (GSH), the nuclear translocation of nuclear factor erythroid 2-related factor 2 (Nrf2) and the mRNA expression of GSH synthesis-related genes to enhance antioxidant capacity in hippocampal tissues of SD mice. An Nrf2 inhibitor ML385 or a GSH synthesis inhibitor BSO reversed the alleviating effects of cTBS treatment on oxidative stress-associated damage of hippocampal tissues and cognitive impairment in SD model mice. Altogether, our study demonstrated that cTBS mitigates oxidative stress-associated inflammation and autophagy through activating the Nrf2-mediated GSH synthesis pathway, improving cognitive impairment in SD mice.

Stress Responses to Bark Beetle Infestations among Pine (Pinus sylvestris), Fir (Abies alba), and Beech (Fagus sylvatica) Trees

Insect infestation triggers multiple defense responses in plants, both locally at the infection site and systemically throughout the plant, including the production of feeding deterrents, toxins, defensive proteins, enzymes, and secondary metabolites. Our study aimed to compare the endogenous levels of antioxidative enzymes, photosynthetic pigments, phytohormones, total phenols, and flavonoids in bark-beetle-infested and uninfested trees. We evaluated the surviving trees in bark-beetle-infested stands, assessing both the condition and defense of uninfested and infested beech (Fagus sylvatica), pine (Pinus sylvestris), and fir (Abies alba) trees. Sampling was performed at six affected sites in the Czech Republic, targeting trees that were resilient to significant health deterioration caused by abiotic and biotic factors. The results showed different levels of most of the measured compounds in the three species. Among all the tested species, photosynthetic pigment levels showed the strongest association with infestation status, which was generally lower in the infested plants. For chlorophyll a, extremely significant reductions were observed from 123 ± 20.6 to 101 ± 17.9 μg/g dry weight (DW) in pine, from 231 ± 33.1 to 199 ± 22.2 μg/g DW in beech, and from 60 ± 5.66 to 51.3 ± 6.27 μg/g DW in fir. In contrast, enzymatic activities indicated only isolated instances of significant association, whereas antioxidative properties (total phenolic content, flavonoids, and 2,2-diphenyl-1-picrylhydrazyl radical scavenging capacity) were not significantly associated with infestation status. There was a statistically significant increase in glutathione reductase activity in infested fir and pine trees. However, this difference was not statistically significant in beech. In contrast, a significant increase in superoxide dismutase activity was detected in infected beech trees. Phytohormones have emerged as the most diverse group of analyzed compounds. Cytokinins were the most distinct, with many of them being significantly increased in infested pines, whereas both beech and fir showed only one significant association. Additionally, derivatives of jasmonic acid also showed a distinct pattern of change associated with bark beetle infestation, with the levels of three out of the four analyzed jasmonates being significantly decreased in infested pines, whereas no effects were observed in beeches and firs. Notably, many phytohormones were significantly elevated in the infested pine, whereas both beech and fir exhibited only one significant association. Overall, the data showed that pines responded differently to bark beetles than to beeches or firs. The greatest changes in phytohormones were observed in pine, whereas the most significant changes in photosynthetic pigments were observed in beech and fir trees.

The pro-oxidant-antioxidant ratio in the cells of plant seedlings from various ecological and geographical growth locations

The balance of pro-antioxidants within the cells of common fir (Abies alba) and mugwort (Artemisia vulgaris) seedlings cultivated across eight distinct climatogeographical regions of Russia and Kharkov (Ukraine) was investigated. The intensity of lipid peroxidation processes was recorded as a pro-oxidant response, and the antioxidant response was measured by assessing the total content of low-molecular-weight antioxidants and the activity of superoxide dismutase. An assessment of the pro-antioxidant balance within the cellular structures of seedlings, originating from seeds of plants grown in diverse climatogeographical environments, was performed. This evaluation included the analysis of seedling survival rates, the degree of lipid peroxidation, and the activity of the antioxidant system, utilizing the coefficient method for measurement. The results suggest that the regions from which plant seeds were collected can be classified into three distinct zones according to the values of the extremity coefficient. A significant positive correlation was identified between the activity of superoxide dismutase in the cells of the examined seedlings and the levels of solar insolation in the regions where the seeds were collected. Adaptation changes in wild plants to temperature, humidity, and light conditions lead to the activation of antioxidant systems. The seedlings of plants whose seeds were formed in environments characterized by high insolation and low precipitation (such as Yakutsk, Ulan-Ude, and Srednekolymsk) exhibited a low total antioxidant content, accompanied by a compensatory increase in superoxide dismutase activity. Our research indicates that the survival rates of seedlings of common fir and mugwort are contingent upon the pro-antioxidant balance that has developed in response to the climatic conditions experienced by the plants. This relationship is characterized by species specificity, nonlinear dynamics, and is predominantly affected by the volume of precipitation received during the active growing season of the maternal plants.

Assessing the Oxidative Stress Reducing Potential of Spilanthes filicaulis (Schumach & Thonn) Ethyl-Acetate Sub-fractions on Plasmodium berghei Infected Female Mice.

Infections by Plasmodium parasite actuate oxidative stress. As malaria parasite actions overwhelm host antioxidant defense by producing excessive reactive species during haemoglobin degradation. This study aimed to evaluate the oxidative status by considering the antioxidant level of ethyl-acetate sub-fractions of Spilanthes filicaulis (ESSF) extract on Plasmodium berghei NK-65 parasitized mice. Sixty female mice weighing 20.0 ± 3.0g were inoculated intraperitoneally with 0.2mL of parasitized erythrocytes randomly selected into five groups of 12 mice, Groups I and II were orally administered with normal saline (10mL/kg) and chloroquine (10mg/kg) while, Groups III, IV and V were administered 250,500 and 750mg/kg per day respectively of ESSF. Mice in each group were sacrificed on days 4 and 8 post treatment, thereafter blood and liver samples were collected and prepared using standard methods to obtain erythrocytes and liver homogenates. Malondialdehyde (MDA), a measure of lipid peroxidation, superoxide dismutase (SOD) and catalase (CAT) level was assessed in the erythrocyte and liver. Administration of ESSF produced a significant (p < 0.05) decrease in the MDA concentration of the parasitized treated group when compared to parasitized untreated group on day 4. Concomitantly, a significant (p < 0.05) increase in SOD and CAT activity in the treated groups with a corresponding decrease in the untreated group on day 4. However, effects of this action were more pronounced on day 8 in both tissues. These findings suggest that ESSF might contribute to the protection of malaria infected mice against oxidative disruptions by improving antioxidant status.

Impact of Pterostilbene on Lens Epithelial Cells from Cataract Model Rats

Lens epithelial cell (lEC) membrane is one of the pathological process of cataract. Sandalwood stilbene is a kind of non-flavonoid polyphenol extracted from blueberries, grapes and other berries. It is considered as a natural antioxidant through various related mechanisms. This study was to investigate the effect and mechanism of Rosewood astragalus on lEC in oxidative cataract rats, and to provide evidence for clinical cataract treatment. Grouping method: normal control group, model group (H2O2 induced cataract) and Zitan astragalus group. Lens turbidity was observed, LECs was isolated and cultured. MTT assay was used to detect LECs proliferation and Caspase-3 activity was used to detect LECs apoptosis. The expression of NF-κB pathway was analyzed. The activities of superoxide dismutase (SOD), reactive oxygen species (ROS) were detected. The results showed that H2O2 significantly increased lens turbidity, enhanced Caspase-3 activity, inhibited LECs proliferation, increased NF-κB pathway expression, increased ROS activity, and decreased SOD activity (P &lt;0.05). Dansandalus stilbene had the effects of inhibiting NF-κB pathway expression, promoting LECs proliferation, decreasing Caspase-3 activity, decreasing ROS production and increasing SOD activity (P &lt; 0.05). In this study, it is suggested that Rosewood stilbene delaying the oxidation of traumatic cataract by anti-oxidation and apoptosis of LECs, and regulating cell proliferation, inhibiting the expression of NF-κB.

Microplastics and copper impacts on feeding, oxidative stress, antioxidant enzyme activity, and dimethylated sulfur compounds production in Manila clam Ruditapes philippinarum

Microplastics (MPs) are widespread ocean pollutants and many studies have explored their effects. However, research on MPs combined impact with copper (Cu) on dimethylated sulfur compound production is limited. Dimethyl sulfide (DMS) is an important biogenic sulfur compound related to global temperatures. This study examined the ecotoxicological effects of polyamide 6 MPs and Cu on dimethylsulfoniopropionate (DMSP), DMS, and dimethyl sulfoxide (DMSO) production in Manila clams (Ruditapes philippinarum). Our findings showed that MPs and Cu increased oxidative stress, indicated by higher superoxide anion radical production and malondialdehyde levels while decreasing glutathione contents and increasing superoxide dismutase activities. Additionally, MPs and Cu exposure reduced DMS and dissolved DMSO (DMSOd) concentrations due to decreased grazing. These results contribute to a better understanding of the ecotoxicological effects of MPs/Cu on bivalves and their roles in the organic sulfur cycle, suggesting a need for further research on long-term impacts on them.

SPRY4 regulates ERK1/2 phosphorylation to affect oxidative stress and steroidogenesis in polycystic ovary syndrome

Polycystic ovary syndrome (PCOS) is a prevalent endocrine disorder in women of childbearing age. The role of Sprouty RTK Signaling Antagonist 4 (SPRY4) in ovarian function in PCOS was investigated herein, focusing on its regulation of ERK1/2 phosphorylation. PCOS models were established in mice using dehydroepiandrosterone (DHEA). The expression levels of SPRY4 in ovarian tissues were analyzed through RT-qPCR and immunohistochemistry. SPRY4 knockdown was achieved via lentivirus, and its effects on endocrine function, ovarian morphology, oxidative stress, and ERK1/2 phosphorylation were evaluated. Afterwards, granulosa cells were isolated and treated with DHEA and ERK2 agonist tert-Butylhydroquinone. The impacts of ERK2 activation on the regulation of SPRY4 knockdown were assessed using ELISA, fluorescent probes, western blotting, and biochemical assays. SPRY4 knockdown normalized the estrous cycle, reduced serum levels of testosterone, anti-Müllerian hormone, and luteinizing hormone/follicle-stimulating hormone ratio, and improved ovarian morphology. Additionally, SPRY4 knockdown alleviated oxidative stress by decreasing reactive oxygen species and malondialdehyde levels while increasing superoxide dismutase activity. It also restored steroidogenic enzyme expression, which were disrupted by DHEA induction. In vitro, SPRY4 knockdown enhanced granulosa cell viability and reduced ERK1/2 phosphorylation, with tert-Butylhydroquinone reversing these effects and restoring oxidative stress and steroidogenesis disruptions. Together, SPRY4 modulates ERK1/2 phosphorylation to influence oxidative stress and steroidogenesis in PCOS. Targeting SPRY4 may provide novel therapeutic avenues for improving ovarian function and managing PCOS.

Curcumin Prevents Renal Damage of l-NAME Induced Hypertension in by Reducing MMP-2 and MMP-9.

In the present study, we investigated whether curcumin administration would interfere with the main renal features of l-NAME-induced hypertension model. For this purpose, we conducted both in vitro and in vivo experiments to evaluate renal indicators of inflammation, oxidative stress, and metalloproteinases (MMPs) expression/activity. Hypertension was induced by l-NAME (70 mg/kg/day), and Wistar rats from both control and hypertensive groups were treated with curcumin (50 or 100 mg/kg/day; gavage) or vehicle for 14 days. Blood and kidneys were collected to determine serum creatinine levels, histological alterations, oxidative stress, MMPs expression and activity, and ED1 expression. l-NAME increased blood pressure, but both doses of curcumin treatment reduced these values. l-NAME treatment increased creatinine levels, glomeruli area, Bowman's space, kidney MMP-2 activity, as well as MMP-9 and ED1 expression, and reduced the number of glomeruli. Curcumin treatment prevented the increase in creatinine levels, MMP-2 activity, and reduced MMP-2, MMP-9, ED1, and superoxide levels, as well as increased superoxide dismutase activity and partially prevented glomeruli alterations. Moreover, curcumin directly inhibited MMP-2 activity in vitro. Thus, our main findings demonstrate that curcumin reduced l-NAME-induced hypertension and renal glomerular alterations, inhibited MMP-2 and MMP-9 expression/activity, and reduced oxidative stress and inflammatory processes, which may indirectly impact hypertension-induced renal outcomes.

Lovastatin Combination Therapy Increases the Survival and Proliferation of Rat Bone Marrow-Derived Mesenchymal Stem Cells Against the Inflammatory Activity of Lipopolysaccharide.

Oxidative stress hurts the survival of transplanted mesenchymal stem cells (MSCs). Lipopolysaccharide (LPS) preconditioning inhibits apoptotic death in MSCs. Also, Lovastatin's protective effect was reported on MSCs. Here, we investigated the potential of LPS and Lovastatin combination therapy on the survival and proliferation of MSCs. MSCs harvested from adult rats (240-260 g) femur and tibia bone marrow. Third passage MSCs were divided into 6 groups control group, LPS, LPS + Lovastatin (10 and 15 µM), and Lovastatin (10 and 15 µM). Cell survival and proliferation were assessed using an MTT assay 24 h after LPS, Lovastatin, or LPS + Lovastatin treatment. Also, Malondialdehyde (MDA) as a lipid peroxidation marker and antioxidant enzymes such as Glutathione peroxidase (GPX) and Superoxide dismutase (SOD) activity levels evaluated. Finally, the expression level of tumor protein P53 (P53) and octamer-binding transcription factor 4 (OCT4) genes were measured by qRT-PCR test. Lovastatin 10 μM potentiated proliferation and survival of MSCs. It can increase the activity of GPX and SOD. 10 µM Lovastatin could not affect MDA amounts but decreased the expression levels of P53 and Oct4 significantly. Nevertheless, treatment with LPS reduced the survival and proliferation of MSCs, along with a significant reduction in GPX activity. LPS + Lovastatin could increase SOD activity, however, GPX enzyme activity and MSCs proliferation did not change so, and it was not effective. We propose Lovastatin at the dose of 10 µM as a suitable combination agent to increase the survival and proliferation of MSCs in oxidative stress conditions.

Effect of different concentrations of inulin on ram sperm quality during cryopreservation

BACKGROUND: In reproductive biotechnology, sperm cryopreservation has a vital role to play. Cryopreservation of sperm produces reactive oxygen species (ROS), which disrupt sperm function and structural competence. Numerous protective chemicals, including fructans, have been used during sperm cryopreservation. OBJECTIVES: To evaluate the effect of different concentrations of the fructosan inulin on ram sperm quality parameters, superoxide dismutase (SOD) activity, and malondialdehyde (MDA) production after freezing and thawing. MATERIALS AND METHODS: The pooled samples from four healthy rams were divided into seven equal aliquots and diluted in a Tris-base extender supplemented with 1, 2, 4, 8, 16, and 28 mM of inulin or without inulin supplementation (control). By using liquid nitrogen vapor, the semen was frozen and stored at 196°C. RESULTS: The total motility, viability, and DNA integrity were significantly improved after freezet-hawing with 28 mM inulin, compared to other treatment groups (P&lt;0.05). A Tris-based extender containing 16 and 28 mM of inulin displayed the highest levels of ram sperm membrane integrity when compared with the control (p&lt;0.05). The abnormality of ram sperm was increased during freeze-thawing at control and 1 mM of inulin, compared to 16 and 28 mM of inulin (P&lt;0.05). Additionally, 28 mM of inulin decreased MDA and increased SOD activity in ram sperm in comparison with the other treatments (P&lt;0.05). CONCLUSION: As a result, 28 mM of inulin could be beneficial for the cryopreservation industry and reduce the harmful effects of freeze-thawing on ram sperm.

Comparative analysis of cyto-genotoxicity of zinc using the comet assay and chromosomal abnormality test.

In this study, the toxicity of the trace element zinc (Zn) in Allium cepa L. test material was examined. Toxicity was investigated in terms of physiological, cytogenetic, biochemical, and anatomical aspects. Germination percentage, root length, weight gain, mitotic index (MI), micronucleus (MN) frequency, chromosomal abnormalities (CAs), malondialdehyde (MDA), proline and chlorophyll levels, superoxide dismutase (SOD) and catalase (CAT) enzyme activities, and meristematic cell damage were used as indicators of toxicity. Additionally, the comet test was used to measure the degree of DNA damage. Four groups of A. cepa bulbs-one for control and three for applications-were created. While the bulbs in the treatment groups were germinated with Zn at concentrations of 35, 70, and 140mg/L, the bulbs in the control group were germinated with tap water. Germination was carried out at room temperature for 72h and 144h. When the allotted time was over, the root tips and leaf samples were collected and prepared for spectrophotometric measurements and macroscopic-microscopic examinations. Consequently, Zn treatment led to significant reductions in physiological indicators such as weight gain, root length, and germination percentage. Zn exposure caused genotoxicity by decreasing the MI ratios and increasing the frequency of MN and CAs (p < 0.05). Zn promoted various types of CAs in root tip cells. The most observed of CAs was the sticky chromosome. Depending on the dose, Zn was found to cause an increase in tail lengths in comet analyses, which led to DNA damage. Exposure to Zn led to a significant decrease in chlorophyll levels and an increase in MDA and proline levels. It also promoted significant increases in SOD and CAT enzyme activities up to 70mg/L dose and statistically significant decreases at 140mg/L dose. Additionally, Zn exposure caused different types of anatomical damage. The most severe ones are epidermis and cortex cell damage. Besides, it was found that the Zn dose directly relates to all of the increases and decreases in physiological, cytogenetic, biochemical, and anatomical parameters that were seen as a result of Zn exposure. As a result, it has been determined that the Zn element, which is absolutely necessary in trace amounts for the continuation of the metabolic activities of the organisms, can cause toxicity if it reaches excessive levels.

The protective effect of hydroalcoholic Citrus aurantifolia peel extract against doxorubicin-induced nephrotoxicity

Background and purpose: Doxorubicin chemotherapy is a widely used treatment for various cancers, including breast, ovarian, and uterine cancers, among others. However, long-term use can cause nephrotoxicity side effects. Some citrus flavonoids have demonstrated nephroprotective activity; therefore, this study aimed to test the nephroprotective effectiveness of Citrus aurantifolia peel extract in protecting and reducing kidney damage caused by doxorubicin. Experimental approach: Citrus aurantifolia peel was dried, ground, and extracted by ultrasonication (70% ethanol), then the extract was dried. Twenty-five female Sprague-Dawley rats were divided into 5 groups including the normal group (control), positive control (doxorubicin) group receiving doxorubicin at the repeated intraperitoneal (i.p.) dose of 4 mg/kg/day on days 2, 6, 10, and 14, and treatment groups receiving Citrus aurantifolia peel extract (CPE) with the doses of 100, 200, and 400 mg/kg/day orally for 14 days, and doxorubicin (4 mg/kg/day, i.p.) on days 2, 6, 10 and 14. On day 15, the rats were euthanized for the measurements of MDA, superoxide dismutase (SOD), catalase, kidney function (measuring blood urea nitrogen (BUN), creatinine, albumin serum levels), and renal histopathology. Findings/Results: The CPE yield was 16.13%. CPE could significantly reduce the levels of MDA, and increase SOD and catalase activities compared with the doxorubicin-induced nephrotoxic model. CPE could increase renal function by reducing BUN and creatinine levels, increasing albumin, and improving the histopathology of the kidney. Conclusion and implications: CPE has a potential effect as nephroprotective against doxorubicin-induced toxicity in renal through antioxidant capacities and increased renal function.
