Early Toxic Effects in a Central American Native Fish (Parachromis dovii) Exposed to Chlorpyrifos and Difenoconazole.

In Costa Rica, agriculture is one of the most important economic activities. Chlorpyrifos and difenoconazole have been identified as agrochemicals widely used in banana and pineapple crops in the Caribbean area of the country and are constantly recorded in aquatic ecosystems. The toxicity of these pesticides in Parachromis dovii was studied. Median lethal concentrations (LC50s) for each substance were obtained from 96-h acute tests. Then, fish were exposed to sublethal concentrations of both substances (10% of LC50), individually and in mixture, to evaluate biomarker responses. Ethoxyresorufin-O-deethylase (EROD), catalase, and glutathione S-transferase activities as well as lipid peroxidation were measured in liver and gill tissues as markers of biotransformation and oxidative stress processes. Cholinesterase activity in brain and muscle tissue was also quantified as a biomarker of toxicity. The LC50s were 55.34 μg/L (95% confidence interval [CI] 51.06-59.98) for chlorpyrifos and 3250 μg/L (95% CI 2770-3810) for difenoconazole. Regarding the biomarkers, a significant inhibition of brain and muscle cholinesterase activity was recorded in fish exposed to 5.50 μg/L of chlorpyrifos. This activity was not affected when fish were exposed to the mixture of chlorpyrifos with difenoconazole. Significant changes in lactate dehydrogenase activity were observed in fish exposed to 325 μg/L of difenoconazole, whereas fish exposed to the mixture showed a significant increase in EROD activity in the liver. These results suggest harmful effects of chlorpyrifos insecticide at environmentally relevant concentrations. There is also evidence for an interaction of the 2 substances that affects the biotransformation metabolism at sublethal levels of exposure. Environ Toxicol Chem 2021;40:1940-1949. © 2021 SETAC.

Occurrence of chemical pollutants in major e-waste sites in West Africa and usefulness of cytotoxicity and induction of ethoxyresorufin-O-deethylase (EROD) in determining the effects of some detected brominated flame retardants and e-waste soil-derived extracts.

We investigated the occurrence of chemical pollutants in major e-waste sites in West Africa and usefulness of cytotoxicity and induction of ethoxyresorufin-O-deethylase (EROD) in determining the effects of some detected brominated flame retardants (BFRs) and e-waste soil-derived extracts. Analysis of the e-waste site samples using AAS and GC-MS techniques revealed the presence of a range of toxic metals as well as persistent and toxic organic pollutants, respectively, in the vicinity of the e-waste sites. As expected, the occurrence (%) of all the detected chemical pollutants in experimental soils significantly (P < 0.05) differs from occurrence (%) in control soil. The calculated LC50 values on RBL-2H3 cells of the detected tetrabromobisphenol A (TBBPA) and hexabromocyclododecane (HBCD) were 3.75 μM and 4.2 μM, respectively. Tribromophenol (TBP), dibromobiphenyl (DBB), and decabromodiphenyl ether (DBDE) were remarkably less toxic on RBL-2H3 cells compared with TBBPA and HBCD as they did not reduce RBL-2H3 cell viability below 50% in the tested concentration range (0-20 μM). The study revealed that TBBPA and HBCD could induce significant RBL-2H3 cell death through caspase-dependent apoptosis. The study further shows that the cytotoxicity of some of these BFRs could increase synergistically when in mixtures and potentially activate inflammation through the stimulation of mast cell degranulation. The e-waste soil-derived extracts induced a concentration-dependent increase in EROD activity in the exposed RTG-W1 cells. Ultimately, nonpolar extracts had higher EROD-inducing potency compared with polar extracts and hence suggesting the presence in higher amounts of AhR agonists in nonpolar e-waste soil-derived extracts than polar extracts. Overall, there is urgent need for actions in order to improve the environmental quality of the e-waste sites.

In vivo EROD assays with the zebrafish (Danio rerio) as rapid screening tools for the detection of dioxin-like activity

The present study compares two alternative in vivo approaches for the measurement of ethoxyresorufin-O-deethylase (EROD) activity in zebrafish (Danio rerio) following exposure to acetonic model sediment extracts: (1) the live-imaging EROD assay for the direct detection of EROD induction in individual livers via epifluorescence, and (2) the fish embryo EROD assay in subcellular fractions derived from entire zebrafish embryos after in vivo exposure.For toxicity assessment, each sediment extract was tested with the standard fish embryo test (FET). Upon completion of a functioning liver after 72h, the embryos gave a distinct fluorescent signal in the liver, and a corresponding EROD activity could be detected in the fish embryo EROD assay. The exposure time in the live-imaging EROD assay was reduced to 3h, which resulted in a stronger, less variable and more sensitive EROD response. Overall, the live-imaging and the fish embryo EROD assays showed the same tendencies and gave comparable results, e.g. a concentration-dependent increase in EROD activity at concentrations one order of magnitude below concentrations producing macroscopically visible abnormalities. At higher concentrations, however, a decrease of EROD activity was observed in either test. Both tests ranked the three model sediment extracts in the same order. Results indicate that both test systems complement each other and together provide a rapid and reliable in vivo tool to investigate the presence of dioxin-like substances in environmental samples.

Developmental toxicity, EROD, and CYP1A mRNA expression in zebrafish embryos exposed to dioxin-like PCB126.

Dioxin-like PCB126 is a persistent organic pollutant that causes a range of syndromes including developmental toxicity. Dioxins have a high affinity for aryl hydrocarbon receptor (AhR) and induce cytochrome P4501A (CYP1A). However, the role of CYP1A activity in developmental toxicity is less clear. To better understand dioxin induced developmental toxicity, we exposed zebrafish (Danio rerio) embryos to PCB126 at concentrations of 0, 16, 32, 64, and 128 μg L(-1) from 3-h post-fertilization (hpf) to 168 hpf. The embryonic survival rate decreased at 144 and 168 hpf. The fry at 96 hpf displayed gross developmental malformations, including pericardial and yolk sac edema, spinal curvature, abnormal lower jaw growth, and non-inflated swim bladder. The pericardial and yolk sac edema rate significantly increased and the heart rate declined from 96 hpf compared with the controls. PCB126 did not alter the hatching rate. To elucidate the mechanism of PCB126-induced developmental toxicity, we conducted ethoxyresorufin-O-deethylase (EROD) in vivo assay to determine CYP1A enzyme activity, and real-time PCR to study the induction of CYP1A mRNA gene expression in embryo/larval zebrafish at 24, 72, 96, and 132 hpf. In vivo EROD activity was induced by PCB126 at 16 μg L(-1) concentration as early as 72 hpf but significant increases were observed only in zebrafish exposed to 64 and 128 μg L(-1) doses (p < 0.005) at 72, 96, and 132 hpf. Induction of CYP1A mRNA expression was significantly upregulated in zebrafish exposed to 32 and 64 μg L(-1) at 24, 72, 96, and 132 hpf. Overall, the severe pericardial and yolk sac edema and reduced heart rate suggest that heart defects are a sensitive endpoint, and the general trend of dose-dependent increase in EROD activity and induction of CYP1A mRNA gene expression provide evidence that the developmental toxicity of PCB126 to zebrafish embryos is mediated by activation of AhR.

Vertical distribution of AhR-activating compounds in sediments contaminated by modernized pulp and paper industry

Increased ethoxyresorufin-O-deethylase (EROD) activity is a sensitive biomarker of exposure to the chemicals which activate the aryl hydrocarbon receptor (AhR) and induce the cytochrome P450 system, such as many polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and polychlorinated biphenyls (PCBs). Pulp bleaching was one of the main sources of PCDDs and PCDFs until elemental chlorine free (ECF) and total chlorine free bleaching processes since 1990s have remarkably decreased but not completely eliminate discharges of these chemicals. In addition, historically contaminated sediments may act as a source of these persistent contaminants. In this study, the contamination history and recovery of a watercourse heavily loaded by the chemical wood industry were studied by analyzing PCDDs, PCDFs and PCBs from vertical sediment samples and by measuring hepatic EROD activity from rainbow trout intraperitoneally dosed with the sediment extracts. No PCDDs or PCDFs were found above the chromatographic limit of detection from the study area and only small amounts of PCB congeners 101, 138, 153, and 180 were present. No increased EROD activity was observed in fish indicating the absence of any AhR-activating compounds in the surface sediment, to about 15 cm depth, representing about the last 20 years when kraft pulping and ECF bleaching with activated wastewater treatment have been used. It can be concluded that nowadays organochlorines and other AhR-ligands do not harm the previously heavily polluted watercourse.

Recovery of Historically Contaminated Watercourse Polluted by the Chemical Wood Industry: EROD Activity in Fish as Biomarker

Despite outstanding process alterations over decades, pulp- and paper-mill-contaminated sediments and continuing exposure by the effluents may still have effects on biota. In this study, ecotoxicological impacts in the boreal watercourse were analyzed by measuring ethoxyresorufin-O-deethylase (EROD) induction from wild fish populations and from experimentally exposed fish. In order to assess the role of sediment-borne chemicals, juvenile rainbow trout (Oncorhynchus mykiss) were exposed in the laboratory to the surface sediments of Lake Vatianjärvi and Southern Lake Saimaa, both watercourses impacted by the chemical wood industry for approximately a century. Hepatic EROD activity was also measured from roach (Rutilus rutilus) and perch (Perca fluviatilis) caught in Lake Vatianjärvi. Increased EROD activity was not observed in wild fish caught in Lake Vatianjärvi nor in rainbow trout exposed to the sediment of Lake Vatianjärvi, but it was observed in rainbow trout exposed to the sediment of Southern Lake Saimaa, probably explained by critically high concentrations of retene. The results of both the laboratory and field study indicate the absence of EROD-inducing compounds of the previously heavily contaminated Lake Vatianjärvi.

Impact of Oxidative Stress Indicated by Thiobarbituric Acid Reactive Substances (TBARS) and Protein Carbonyl Levels (PC) on Ethoxyresorufin-O-deethylase (EROD) Induction in Common Carp (Cyprinus carpio)

This study examined the impact of oxidative stress indicated by thiobarbituric acid reactive substances (TBARS) and protein carbonyl (PC), induced by intensive exercise and cadmium chloride (CdCl2) on ethoxyresorufin-O-deethylase (EROD) activity in juvenile carp (Cyprinus carpio). In the first experiment, fish were divided into three groups: (1) control, (2) carp exposed to intensive exercise, and (3) carp that was not exercised but previously, as well as carp in group 2, received single dose of 3-methylcholantrene (3-MC). The third and sixth day fish were sacrificed and the measurements were conducted. In the second experiment, fish were divided into (1) control, (2) carp in water containing CdCl2, and (3) carp in dechlorinated tap water (2 and 3 received single dose of 3-MC on the seventh day after exposure to CdCl2). The carp were killed 6 days later and livers were excised for biochemical analyses. In the first experiment, on the sixth day after treatment with 3-MC, results show statistically significant increase in EROD activity in non-exercised carp, while that increase in carp exposed to intensive exercise was significantly lower. Three days after exposure to 3-MC, statistically significant increase in TBARS was observed in both exercised and non-exercised carp. Six days after exposure to 3-MC, PC levels were significantly higher in exercised carp. Pretreatment with CdCl2, in the second experiment, caused oxidative stress and reduction of EROD activity. Results show linkage between expression of EROD activity and oxidative stress biomarkers and possible influence of oxidative stress on the cell membrane structures and consequently on EROD activity.

Linking behavioural alterations with biomarkers responses in the European seabass Dicentrarchus labrax L. exposed to the organophosphate pesticide fenitrothion

The acute effects of the organophosphate insecticide fenitrothion on Dicentrarchus labrax juveniles were investigated through a bioassay using biomarkers and swimming behaviour as effect criteria. After 96 h of exposure to sub-lethal concentrations of fenitrothion, the swimming velocity and several biomarkers were individually determined, namely: brain acetylcholinesterase (AChE) activity; muscle cholinesterases (ChE), lactate dehydrogenase and isocitrate dehydrogenase activities; liver ethoxyresorufin-O-deethylase (EROD), glutathione S-transferases, glutathione peroxidase, glutathione reductase, catalase and superoxide dismutase (SOD) activities and lipid peroxidation levels (LPO). A significant decrease of the swimming velocity (LOEC = 2 mg l(-1)), an inhibition of both AChE (LOEC = 0.06 mg l(-1)) and ChE activities (LOEC = 0.03 mg l(-1)), and a positive and significant correlation between the swimming velocity and AChE were found in exposed fish, suggesting an influence of the inhibition of these enzymes in the swimming velocity decrease. An increase of EROD activity (LOEC = 1 mg l(-1)), indicating the involvement of this enzyme in fenitrothion biotransformation, and a negative and significant correlation between EROD activity and swimming velocity were also found, suggesting that the two findings may somehow be related. Furthermore, results show a significant induction of SOD (LOEC = 0.13 mg l(-1)) without LPO increase, suggesting that the enzyme is preventing oxidative stress damage. No significant alterations were found in any of the other parameters tested. Thus, exposure of seabass to fenitrothion in the wild at concentrations similar to those tested here may have adverse consequences at population level as neurotransmission and swimming ability are essential for fish performance and survival.

Analyses of paralytic shellfish toxins and biomarkers in a southern Brazilian reservoir

The Alagados Reservoir (Brazil) is an important source for the supply of water, recreation and fishery. Since 2002, the occurrence of cyanobacterial blooms (paralytic shellfish toxins – PST producers) have been noted. This study was aimed at the monitoring of PST occurrence in the Reservoir's water and fish. Biomarkers such as ethoxyresorufin-O-deethylase (EROD), glutathione S-transferase (GST), catalase (CAT), and acetylcholinesterase (AchE) activities, lipoperoxidation (LPO), histopathology, and comet assay were analyzed in fish. Water and fish were sampled in spring, summer and autumn. The PST concentrations in water were 5.15, 43.84, and 50.78 ng equiv Saxitoxin/L in the spring, summer and autumn, respectively. The PST muscle concentration was below the limit for shellfish. Gonyautoxins (GTX) were found in water samples and fish muscle, and GTX 5 was the major analogous found in muscle. In the summer samples, the LPO, genetic damage, and the GST and AchE activities increased while in the autumn an increase in EROD activity and genetic damage were observed. In all samplings, histopathological alterations in the fish gills and liver were found. The results showed a seasonal variation in the fishes health, which could be related also to farming activities and to the contaminants bioavailability during the year.

Cytochrome P450 induction in Ictalurus punctatus following exposure to wastewater treatment plant effluent

Induction of the cytochrome P450 enzymes in fish, measured as ethoxyresorufin-O-deethylase (EROD) activity, has been extensively used as a biomarker in assessing exposures and responses of aquatic organisms to contaminants. This study focused on whether exposure to Troy (Alabama) wastewater treatment plant (TWWTP) effluent at the Walnut Creek mixing zone, induced transcription of mRNA for cytochrome P4501A1 enzyme production and increased EROD activity in channel catfish (Ictalurus punctatus). Water from Big Creek (Barbour County, AL), regarded as least impacted by pollutants, was used as a reference source for administration of a positive control chemical (PCB – Aroclor 1254). All water samples were transported from field sites to the Troy University laboratory for conducting the study. Reverse transcriptase-polymerase chain reaction (RT-PCR) indicated the presence of compounds capable of inducing transcription of CYP1A1 mRNA in catfish liver. Increased transcription of mRNA for cytochrome P4501A1 enzyme production, confirmed by a 3-fold induction of EROD activity, was found in catfish exposed to water from the TWWTP mixing zone on Walnut Creek compared to catfish exposed to Walnut Creek Upstream. Catfish exposed to water from Big Creek treated with PCBs were found to have only slightly higher enzyme activities than those exposed to water from Big Creek Control, but a 2.7-fold EROD level was found in catfish exposed to Big Creek Control compared to Walnut Creek Upstream. Determining the induction of cytochrome P450 and subsequent enzymatic activity in catfish and other fish species common to this region could be useful early molecular warnings of possible pollution effects, beyond those methods typically used to characterize water quality.

Induction of in vitro EROD activity and in vivo caffeine metabolism in two species of New Zealand birds

In birds, induction of cytochrome P4501A (CYP1A) is usually assessed as liver microsomal ethoxyresorufin O-deethylase (EROD) activity, but in mammals, it can be determined by a caffeine metabolism blood test. We investigated both of these measures in two species of New Zealand birds. Administration of a model CYP1A inducer, β-naphthoflavone (BNF) (80 mg/kg i.p. twice 2 days apart), to paradise shelducks ( Tadorna variegata; herbivore) and southern black-backed gulls ( Larus dominicanus; omnivore) ( n = 5 or 6) caused marked increases in EROD activity (80- and 20-fold, respectively). In both species, BNF treatment also caused significant increases (>8-fold) in caffeine metabolism determined prior to sacrifice as the serum concentration ratio of the major metabolite, paraxanthine, to caffeine, after caffeine administration (1 mg/kg i.p.). The results suggest in vivo caffeine metabolism is a potentially useful non-destructive biomarker of CYP1A induction in wild birds.

Reflection of hydrocarbon pollution on hepatic EROD activity in the black goby ( Gobius niger)

Hepatic ethoxyresorufin- O-deethylase (EROD) activity was measured in black gobies ( Gobius niger) collected from seven sampling sites along the Slovenian coast during spawning and non-spawning periods. Sediments from the same sites were tested for their toxicity and for total polycyclic aromatic hydrocarbon (PAH) content. EROD activity was significantly higher during the non-spawning period in winter than during the spawning period in spring and generally correlated well with increased total PAH content in sediments and with sediment toxicity. An 11-fold increase of EROD activity was induced 48 h after injecting 10 μg g −1 weight of benzo(a)pyrene into black gobies. On the basis of these results and of its ecological characteristics, the black goby is proposed as a suitable candidate for future biomonitoring programmes.

Effects of medetomidine on hepatic EROD activity in three species of fish

Medetomidine, an antifouling candidate, was investigated for its effects on cytochrome P4501A (CYP1A) activity in fish. Rainbow trout ( Oncorhynchus mykiss), turbot ( Psetta maxima), and Atlantic cod ( Gadus morhua) were exposed to medetomidine either via i.p. injection (<5 μmol (1 mg)/kg) or via water (<50 nM). Enzyme activity was measured as ethoxyresorufin- O-deethylase (EROD) activity in liver microsomes. There was a small (2–7-fold) increase in EROD activity in rainbow trout. In turbot, EROD activity increased (4-fold) after injection, while a non-significant (50%) decrease was observed after water exposure. No effects on EROD activities were observed in Atlantic cod. In vitro inhibition studies of EROD activities in liver microsomes from all three species showed that medetomidine was a very potent CYP1A inhibitor. Thus, median inhibition values (IC 50) were 35±10 nM for rainbow trout, 47±17 nM for turbot, and 111±70 nM for Atlantic cod. These observed effects suggest that medetomidine interferes with CYP1A-dependent metabolism of xenobiotics in these fish species.

Hepatic and immune biological effect assays in C57BL/6 mice to measure polycyclic aromatic hydrocarbon bioavailability under laboratory exposures with increasing environmental relevance

Monitoring biological responses that are mediated via the aryl-hydrocarbon receptor (AhR) in animals exposed to environmental contaminants can indicate both the presence of chemicals that act through this biochemical pathway and whether these chemicals are bioavailable. The use of an ex-situ method that incorporated biological responsiveness monitoring in mice for determining the presence of 'biologically active' hydrocarbons in contaminated soils was investigated. The use of C57BL/6 as a test organism was validated by determining hepatic and immune responsiveness to two polyaromatic hydrocarbons (PAHs): 3,4 benz[a]pyrene (B[a]P) and 1,2 benz (a)anthracene (BA) administered via intraperitoneal (i.p.) injection. The responsiveness of mice exposed to soils spiked with hydrocarbons or ex situ exposures to soil removed from two contaminated sites was also investigated. Mice that were exposed to B[a]P via i.p. injections showed a 14-fold increase in liver microsomal ethoxyresorufin O-deethylase (EROD) activity compared to the control group. In contrast EROD activity following BA exposure at the same level was not significantly enhanced. Mouse immune response was significantly inhibited in a dose-dependent manner by i.p. injections of B[a]P. No significant inhibition occurred with the same doses of BA. Following i.p. exposure, the retention of B[a]P in mouse carcasses was greater than BA. Mice exposed to clean soils spiked with environmentally relevant concentrations of B[a]P and BA failed to show any significantly different hepatic or immune responses. Carcass residue data indicated a limited uptake of PAH from the soil. In contrast, EROD activity in mice exposed (ex situ) to hydrocarbon-contaminated soils removed from a fuel-loading depot and decommissioned gas works was significantly enhanced (4- and 2-fold respectively). However, this increase in EROD activity did not appear to correlate with either soil or carcass PAH concentrations. These results support the assumption that B[a]P has a higher affinity for the aryl hydrocarbon receptor (AhR) compared to BA. Soil parameters such as organic carbon content, structure and particle size distribution can modulate the bioavailability of contaminants to biological receptors. These factors are implicated in the lack of responsiveness demonstrated in the spiked soil experiments. However the responsiveness of EROD activity in mice exposed (ex situ) to soil contaminated with complex mixtures of hydrocarbon compounds confirms the potential usefulness of this model to determine the presence of 'biologically active' compounds in aged soils removed from contaminated sites.

Induction of cytochrome P4501A in African brown house snake (Lamprophis fuliginosus) primary hepatocytes

Although there have been numerous studies involving fish, birds, and mammals, little is known about the response of the cytochrome P4501A system of snakes to halogenated aromatic hydrocarbons (HAHs). The present study describes the induction of ethoxyresorufin-O-deethylase (EROD) in primary hepatocytes of the African brown house snake (Lamprophis fuliginosus). Hepatocytes were exposed in multiwell plates to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and four different non-ortho-substituted coplanar polychlorinated biphenyls (PCBs 77, 81, 126, and 169). Exposure to TCDD and PCB 126 resulted in a dose-dependent increase in EROD activity, with maximum inducible EROD activities of 177 +/- 56 (mean +/- SEM) and 101.1 +/- 55 pmol/min/mg protein for TCDD and PCB 126, respectively. None of the other PCBs caused a measurable induction of EROD, which suggests reduced inducibility of snake hepatocytes compared to some vertebrate taxa. Median effective concentrations (EC50s) were 0.16 +/- 0.03 nM for TCDD and 8.25 +/- 4.14 nM for PCB 126. The relative potency (REP20-80) range for PCB 126 was 0.044 to 0.046. Compared to results from in vitro systems using other vertebrate species, both the maximum inducibility and the REPs estimated for L. fuliginosus were within the same range as those reported for mammals and the more sensitive bird species but were greater than the values reported for most fish species. In conclusion, induction of EROD activity in primary hepatocytes appears to be a useful approach for evaluating the dioxin-like potencies of aryl hydrocarbon-receptor agonists in snakes. The test system offers a method for rapid screening of reptilian responsiveness to these compounds using smaller numbers of organisms than with in vivo studies, an important consideration for many declining reptile species.

Effect of dispersant on the composition of the water‐accommodated fraction of crude oil and its toxicity to larval marine fish

Newly hatched mummichog (Fundulus heteroclitus) were exposed in a 96-h static renewal assay to water-accommodated fractions of dispersed crude oil (DWAF) or crude oil (WAF) to evaluate if the dispersant-induced changes in aqueous concentrations of polycyclic aromatic hydrocarbons (PAH) affected larval survival, body length, or ethoxyresorufin-O-deethylase (EROD) activity. Weathered Mesa light crude oil (0.05-1 g/L) and filtered seawater with or without the addition of Corexit 9500 were used to prepare DWAF and WAE At 0.2 g/L, the addition of dispersant caused a two- and fivefold increase in the concentrations of total PAH (sigmaPAH) and high-molecular-weight PAH (HMWPAH) with three or more benzene rings. Highest mortality rates (89%) were observed in larvae exposed to DWAF (0.5 g/L; sigmaPAH, 479 ng/ml). A reduction in body length was correlated with increased levels of sigmaPAH (r2 = 0.65, p = 0.02) and not with HMWPAH. The EROD activity increased linearly with HMWPAH (r2 = 0.99, p = 0.001) and not with sigmaPAH. Thus, chemical dispersion increased both the sigmaPAH concentrations and the proportion of HMWPAH in WAF. Dispersed HMWPAH were bioavailable, as indicated by a significantly increased EROD activity in exposed mummichog larvae, and this may represent a significant hazard for larval fish.

Cytochrome P450 1A1 and 1B1 in Human Blood Lymphocytes Are Not Suitable as Biomarkers of Exposure to Dioxin-like Compounds: Polymorphisms and Interindividual Variation in Expression and Inducibility

Cytochrome P450 1A1 (CYP1A1) and 1B1 (CYP1B1) are phase I enzymes, the expression of which can be affected by many environmental compounds, including dioxins and dioxin-like compounds. Because CYP1A1 and CYP1B1 expression can easily be determined in peripheral blood lymphocytes, it is often suggested as biomarker of exposure to these compounds. In this study we investigated the interindividual differences in constitutive and induced CYP1A1-catalyzed ethoxyresorufin-O-deethylase (EROD) activity and CYP1A1 and CYP1B1 gene expression in human blood lymphocytes in a group of ten non-smoking females. Freshly isolated lymphocytes were cultured in medium containing the mitogen PHA and were exposed to the most potent dioxin, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) or the less potent dioxin-like polychlorinated biphenyl 126 (PCB126). In addition, we determined the occurrence of the CYP1A1 MspI and CYP1B1 Leu432Val polymorphisms. All individuals showed a concentration-dependent increase of EROD activity by TCDD, which was significantly correlated with an increase in CYP1A1, but not CYP1B1 expression. The maximum induced EROD activity by TCDD was very different among the individuals, but the EC50 values were about the same. PCB126 also caused a concentration-dependent increase of EROD activity, but was a factor 100-1000 less potent than TCDD among the individuals. The allele frequencies for CYP1A1 MspI and CYP1B1 Leu432Val reflected a normal Caucasian population and in this study the polymorphisms had no apparent effect on the expression and activity of these enzymes. Our study shows a large interindividual variability in constitutive and induced EROD activity, and CYP1A1 and CYP1B1 expression in human lymphocytes. In addition, dioxin concentrations at which effects were observed in our in vitro study are about 10-fold higher than the human blood levels found in vivo, indicating that EROD activity and CYP1A1 and CYP1B1 expression in human lymphocytes might not be applicable as biomarkers of exposure to dioxin and dioxin-like compounds.

Evidence for a new human CYP1A1 regulation pathway involving PPAR-α and 2 PPRE sites

Cytochrome P450 1A1 catalyzes the degradation of endobiotics (estradiol, fatty acids, and so on) and the bioactivation of numerous environmental procarcinogens, such as arylamines and polycyclic aromatic hydrocarbons, that are found in food. Several peroxisome proliferators and arachidonic acid derivatives enhance cytochrome P450 1A1 activity, but the mechanisms involved remain unknown. The aim of this work was to study the role of peroxisome proliferator-activated receptors in cytochrome P450 1A1 gene induction. The role of peroxisome proliferator-activated receptor transcription factors in cytochrome P450 1A1 induction was assessed by means of enzymatic activities, quantitative real-time polymerase chain reaction, gene reporter assays, mutagenesis, and electrophoretic mobility shift assay. We show that peroxisome proliferator-activated receptor-alpha agonists (WY-14643, bezafibrate, clofibrate, and phthalate) induce human cytochrome P450 1A1 gene expression, whereas 2,4-thiazolidinedione, a specific peroxisome proliferator-activated receptor-gamma agonist, represses it. The induction of cytochrome P450 1A1 transcripts by WY-14643 was associated with a marked increase of ethoxyresorufin O -deethylase activity (10-fold at 200 mumol/L). Transfection of peroxisome proliferator-activated receptor-alpha complementary DNA enhanced cytochrome P450 1A1 messenger RNA induction by WY-14643, although WY-14643 failed to activate xenobiotic responsive element sequences. Two peroxisome proliferator response element sites were located at positions -931/-919 and -531/-519 of the cytochrome P450 1A1 promoter. Their inactivation by directed mutagenesis suppressed the inductive effect of WY-14643 on cytochrome P450 1A1 promoter activation. Electrophoretic mobility shift assay and chromatin immunoprecipitation assay experiments showed that the 2 cytochrome P450 1A1 peroxisome proliferator response element sites bind the peroxisome proliferator-activated receptor-alpha/retinoid X receptor-alpha heterodimer. We describe here a new cytochrome P450 1A1 induction pathway involving peroxisome proliferator-activated receptor-alpha and 2 peroxisome proliferator response element sites, indicating that peroxisome proliferator-activated receptor-alpha ligands, which are common environmental compounds, may be involved in carcinogenesis.

Organochlorine contaminants in flounder ( Platichthys flesus) and mullet ( Mugil cephalus) from Douro estuary, and their use as sentinel species for environmental monitoring

In order to monitor the presence of organic pollutants in Douro estuary (NW Portugal), two sentinel fish species, grey mullet ( Mugil cephalus) and flounder ( Platichthys flesus), were periodically sampled from May 2001 to April 2002. At all sampling periods, several specimens of both species were allowed to depurate in clean sea water. Levels of PCBs and DDTs were quantified in liver and muscle of both species. The accumulation of PCBs was higher in muscle of mullet, with a maximum of 345 ng/g dw, than in flounder, with a maximum 52 ng/g dw. In the liver, flounder showed the highest levels (811 ng/g dw). Of the 18 congeners analysed, CBs 180 (hepta), 153 and 138 (hexachlorobiphenyls) were predominant in the tissue of both species. The maximum concentration of tDDT was measured in flounder liver (301 ng/g dw). In contrast, a 10-fold higher tDDT was recorded in mullet muscle (63–69 ng/g dw and 8–16 ng/g dw for mullet and flounder, respectively). The hepatic ethoxyresorufin O-deethylase (EROD) activity was periodically determined. Consistently, a 10-fold higher enzymatic activity was present in mullet in comparison with flounder (1536 pmol/min /mg protein in mullet and 156 pmol/min /mg protein in flounder). In mullet, no correlation could be found between EROD activity and gonado-somatic index (GSI) or hepato-somatic index (HSI). On the contrary, during the reproductive season, female flounder showed a negative correlation between EROD activity and GSI. Despite being in clean sea water for 1 month period, no significant decrease in the tissue content of PCBs and tDDT was found. However, mullet’s EROD activity followed a clear pattern, with a decrease enzymatic activity after being in captivity. Female flounder displayed a similar trend during the resting season. Yet, during the reproductive season, an increased EROD activity was recorded after being in captivity for 1 month, which may be associated with a modulation effect of steroids on CYP 1A1. Histological analyses of gonad revealed that 21% of male mullet displayed testis-ova, while no male flounder was found to show gonadal abnormalities. Overall, the study reports the accumulation of significant levels of PCBs and DDTs in the tissues of the studied species. It also provides important evidences supporting the use of grey mullet as a sentinel species for monitoring the presence of organic contaminants and xeno-estrogenic pollution in southern European estuaries.

Some polybrominated diphenyl ether (PBDE) flame retardants with wide environmental distribution inhibit TCDD-induced EROD activity in primary cultured carp ( Cyprinus carpio) hepatocytes

Ethoxyresorufin- O-deethylase (EROD) activity, a catalytic function of the cytochrome P450 1A (CYP1A) microsomal oxygenase subfamily, is a popular biomarker for exposure to xenobiotics, polyhalogenated aromatic hydrocarbons (PHAHs) in particular. It has found wide use in aquatic pollution assessment both in vivo and in vitro. In such studies, subjects are often exposed to complex mixtures where various constituents can interfere with EROD-activity, possibly resulting in inadequate estimation of toxic hazard or biological response. The present study investigates the effects of polybrominated diphenyl ethers (PBDEs), a relatively new and increasingly detected group of environmental contaminants, on the validity of EROD activity as exposure marker in carp ( Cyprinus carpio) hepatocytes. Freshly isolated hepatocytes of a genetically uniform strain of male carp were co-exposed to 2,3,7,8-tetrachlorodibenzo- p-dioxin (TCDD) at concentrations of 0, 1, 3, 10, 30, and 100 pM, and one of the highly purified PBDE/PCB congeners (at concentrations of 0, 0.25, and 2.5 μM) or cleaned-up and untreated DE-71 samples (0, 0.1, and 1 μM). PBDEs were selected from the 209 possible congeners based on their relative abundance in environmental samples: BDE-47, BDE-99, BDE-100, and BDE-153. A tentative metabolite of BDE-47, 6OH-BDE-47, was also included. In addition, a commercial pentabrominated dipenylether mixture (DE-71) was tested for interference with EROD activity both with and without clean-up by carbon fractionating which removed possible planar contaminants. Polychlorinated biphenyl (PCB)-153, a reported inhibitor of EROD activity in flounder, was included for comparison. Cells were cultured for a total period of 8 days; exposure started at day 3 after cell isolation. After 5 days of exposure, cell pellets were frozen before EROD activity was determined. Upon exposure to TCDD, the cells responded with increased EROD activity as expected. Significant reduction of TCDD-induced EROD activity was found in the presence of BDE-47, BDE-99, and BDE-153, but not with BDE-100 and 6-hydroxylated BDE-47. Of these PBDE congeners, the most abundant congener in environmental samples, BDE-47, exhibited the strongest inhibition (down to 6% of the TCDD control value). The cleaned-up fraction of commercial penta-BDE (DE-71) mixture proved an even more potent inhibitor, resulting in reduction of EROD activity to 4% of the control values observed at 1.0 μM. BDE-47 and BDE-153 did not reduce TCDD-induced EROD activity when added shortly prior to measurement, suggesting possible interaction with TCDD at the level of CYP1A biosynthesis. PCB-153 did not show significant effects on EROD activity in carp in this study. The present results indicate that environmentally relevant PBDEs can interfere with determination of EROD activity in vitro, at levels reported earlier for PCBs. The observation that detected PBDE levels are rising, stresses the need for caution when interpreting EROD data on environmental samples.
