Protein kinase FaSnRK2.6 phosphorylates transcription factor FabHLH3 to regulate anthocyanin homeostasis during strawberry fruit ripening.

Strawberry (Fragaria×ananassa) is a model plant for studying non-climacteric fruit ripening regulated by abscisic acid (ABA). However, the signaling of ABA in the regulation of fruit coloration is not fully understood. Here, a transcription factor FabHLH3 key to fruit coloration is identified by yeast two hybrid library screening using FaSnRK2.6 as a bait, an ABA core signaling component negative to ripening. Indeed, this interaction is also confirmed by firefly luciferase complementation assay and pull-down assay. RT-qPCR and Western blotting analysis confirm FabHLH3 is expressed ubiquitously in strawberry and stably during fruit development. Manipulating both FabHLH3 and FaSnRK2.6 expression by overexpression and interference demonstrates that FabHLH3 and FaSnRK2.6 promote and inhibit strawberry fruit coloration, respectively, using the marker gene FaUFGT, key to anthocyanin biosynthesis. FaSnRK2.6 can phosphorylate FabHLH3, which promotes FaUFGT expression by the directly binding to its promoter. The phosphorylation inhibits the binding of FabHLH3 to FaUFGT promoter, consequently suppressing FaUFGT expression. Altogether, FaSnRK2.6, a negative kinase in ripening, interacts with and phosphorylates FabHLH3 to suppress FaUFGT expression. With the increase of ABA content in strawberry fruit ripening, the expression of FaSnRK2.6 decreased, which released FabHLH3 transcription activity and enhanced FaUFGT expression, finally promoting the coloration. Thus, our findings fill a gap how FaSnRK2.6 negatively regulates strawberry fruit coloration and ripening by FabHLH3.

NnARF17 and NnARF18 from lotus promote root formation and modulate stress tolerance in transgenic Arabidopsis thaliana

Auxin response factors (ARFs) play a crucial role in regulating gene expression within the auxin signal transduction pathway, particularly during adventitious root (AR) formation. In this investigation, we identified full-length sequences for ARF17 and ARF18, encompassing 1,800 and 2,055 bp, encoding 599 and 684 amino acid residues, respectively. Despite exhibiting low sequence homology, the ARF17- and ARF18-encoded proteins displayed significant structural similarity and shared identical motifs. Phylogenetic analysis revealed close relationships between NnARF17 and VvARF17, as well as NnARF18 and BvARF18. Both ARF17 and ARF18 demonstrated responsiveness to exogenous indole-3-acetic acid (IAA), ethephon, and sucrose, exhibiting organ-specific expression patterns. Beyond their role in promoting root development, these ARFs enhanced stem growth and conferred drought tolerance while mitigating waterlogging stress in transgenic Arabidopsis plants. RNA sequencing data indicated upregulation of 51 and 75 genes in ARF17 and ARF18 transgenic plants, respectively, including five and three genes associated with hormone metabolism and responses. Further analysis of transgenic plants revealed a significant decrease in IAA content, accompanied by a marked increase in abscisic acid content under normal growth conditions. Additionally, lotus seedlings treated with IAA exhibited elevated levels of polyphenol oxidase, IAA oxidase, and peroxidase. The consistent modulation of IAA content in both lotus and transgenic plants highlights the pivotal role of IAA in AR formation in lotus seedlings.

Exogenous abscisic acid and sugar induce a cascade of ripening events associated with anthocyanin accumulation in cultured Pinot Noir grape berries.

Fruit quality is dependent on various factors including flavour, texture and colour. These factors are determined by the ripening process, either climacteric or non-climacteric. In grape berry, which is non-climacteric, the process is signalled by a complex set of hormone changes. Abscisic acid (ABA) is one of the key hormones involved in ripening, while sugar availability also plays a significant role in certain ripening aspects such as anthocyanin production. To understand the relative influence of hormone and sugar signalling in situ can prove problematic due to the physiological and environmental (abiotic and biotic) factors at play in vineyards. Here we report on the use of in vitro detached berry culture to investigate the comparative significance of ABA and sugar in the regulation of Pinot noir berry anthocyanin production under controlled conditions. Using a factorial experimental design, pre-véraison berries were cultured on media with various concentrations of sucrose and ABA. After 15 days of in vitro culture, the berries were analysed for changes in metabolites, hormones and gene expression. Results illustrated a stimulatory effect of sucrose and ABA on enhancing berry colour and a corresponding increase in anthocyanins. Increased ABA concentration was able to boost anthocyanin production in berries when sucrose supply was low. The sucrose and ABA effects on berry anthocyanins were primarily manifested through the up-regulation of transcription factors and other genes in the phenylpropanoid pathway, while in other parts of the pathway a down-regulation of key proanthocyanindin transcription factors and genes corresponded to sharp reduction in berry proanthocyanidins, irrespective of sucrose supply. Similarly, increased ABA was correlated with a significant reduction in berry malic acid and associated regulatory genes. These findings suggest a predominance of berry ABA over berry sugar in coordinating the physiological and genetic regulation of anthocyanins and proanthocyanins in Pinot noir grape berries.

Genome-wide analysis of carotenoid cleavage oxygenases and identification of ripening-associated DzNCED5a in durian (Durio zibethinus) fruit

Durian (Durio zibethinus L.), popularly known as the “King of fruits,” holds significant economic importance in Southeast Asia, including Thailand. During its ripening process, the phytohormone abscisic acid (ABA) content has been reported to increase. However, a comprehensive understanding of ABA's specific role in durian fruit ripening remains elusive. Furthermore, little is known about the molecular aspects of the carotenoid cleavage pathway in this iconic fruit. Therefore, we performed genome-wide identification of the carotenoid cleavage oxygenase (CCO) family in durian. This family includes the nine-cis-epoxycarotenoid dioxygenases (NCEDs) responsible for ABA production and the carotenoid cleavage dioxygenases exhibiting diverse substrate specificities. Through phylogenetic analysis, we classified 14 CCOs in durian into 8 distinct subfamilies. Notably, each DzCCO subfamily displayed a conserved motif composition. Cis-acting element prediction showed that cis-elements related to plant hormones and environmental stress responses were distributed in the DzCCO promoter. In addition, transcriptome analysis was performed to examine the expression pattern during the fruit development and ripening stages. Interestingly, DzNCED5a, a ripening-associated gene, exhibited the highest expression level at the ripe stage, outperforming other CCOs. Its expression markedly correlated with increased ABA contents during the ripening stages of both the “Monthong” variety and other durian cultivars. Transiently expressed DzNCED5a in Nicotiana benthamiana leaves confirmed its function in ABA biosynthesis. These findings highlight the involvement of DzNCED5a in ABA production and its potential importance in durian fruit ripening. Overall, this study provides insights into the significance of CCOs in durian fruit ripening.

Ethanol suppresses rice seed germination through inhibiting ROS signaling

Ethanol is frequently used not only as priming but also as a solvent to dissolve hardly water-soluble phytohormones gibberellic acid (GA3) and abscisic acid (ABA) in seed germination. However, the molecular and physiological mechanisms of ethanol's impact on seed germination remain elusive. In this report, we investigated how ethanol affected reactive oxygen species (ROS) during rice seed germination. Ethanol at a concentration of 3.5% (v/v) inhibited 90% seed germination, which was almost reversed by H2O2. H2O2 contents in embryos were reduced by ethanol after 18 h imbibition. Antioxidant enzymes assays revealed that only superoxide dismutase (SOD) activities in seed embryos were lowered by ethanol, in line with the suppressed mRNA expression of SOD genes during imbibition. Additionally, compared to the mock condition, ethanol increased ABA contents but decreased GA (GA1 and GA3) in seed embryos, resulting in disharmonizing GA/ABA balance. Conceivably ethanol induced transcription of OsNCEDs, the key genes for ABA biosynthesis, and OsABA8ox3, a key gene for ABA catabolism. Furthermore, ethanol promoted ABA signaling by upregulating ABA receptor genes and ABA-responsive element (ABRE)-binding protein/ABRE-binding factors during imbibition. Overall, our results demonstrate that lowering of H2O2 levels due to suppressed SOD activities in rice germinating seed embryos is the decisive factor for ethanol-induced inhibition of seed germination, and GA/ABA balance and ABA signaling also play important roles in ethanol's inhibitory impact on seed germination.

Effect of Exogenous Plant Growth Regulators and Rejuvenation Measures on the Endogenous Hormone and Enzyme Activity Responses of Acer mono Maxim in Cuttage Rooting.

The cuttage rooting method for Acer species is difficult to achieve a good efficacy as trees maintain good characteristics at the rejuvenation stage, thus improving the rooting of Acer species. The addition of exogenous hormones and rejuvenation can improve the rooting effect of cuttings; however, the specific regulatory mechanism is still unclear. Here, Acer mono Maxim rejuvenation and non-rejuvenation cuttings were used as test subjects, to investigate the effects of exogenous hormones on the activities of endogenous hormones and antioxidant enzymes in the rooting process of young cuttings. The results showed that exogenous growth-regulating substances significantly improved the rooting rate of A. mono. Exogenous hormones naphthylacetic acid (NAA) + indolebutyric acid (IBA) increased the initial levels of the endogenous hormones, indoleacetic acid (IAA) and abscisic acid (ABA), and the enzyme activities of peroxidase (POD) and polyphenol oxidase (PPO). Rejuvenation treatment prolonged the time of increase in ABA content and indoleacetic acid oxidase (IAAO) activity at the root primordium induction stage, while increasing trans-zeatin riboside (ZR) content and decreasing POD enzyme activity in cuttings. These results demonstrate that A. mono cuttings can achieve the purpose of improving the rooting rate by adding the exogenous hormone (NAA + IBA), which is closely related to the changes of endogenous hormone content and enzyme activity, and these changes of A. mono rejuvenation cuttings are different from non-rejuvenation cuttings.

Allelic variation in transcription factor PtoWRKY68 contributes to drought tolerance in Populus.

Drought stress limits woody species productivity and influences tree distribution. However, dissecting the molecular mechanisms that underpin drought responses in forest trees can be challenging due to trait complexity. Here, using a panel of 300 Chinese white poplar (Populus tomentosa) accessions collected from different geographical climatic regions in China, we performed a genome-wide association study (GWAS) on seven drought-related traits and identified PtoWRKY68 as a candidate gene involved in the response to drought stress. A 12-bp insertion and/or deletion and three nonsynonymous variants in the PtoWRKY68 coding sequence categorized natural populations of P. tomentosa into two haplotype groups, PtoWRKY68hap1 and PtoWRKY68hap2. The allelic variation in these two PtoWRKY68 haplotypes conferred differential transcriptional regulatory activities and binding to the promoters of downstream abscisic acid (ABA) efflux and signaling genes. Overexpression of PtoWRKY68hap1 and PtoWRKY68hap2 in Arabidopsis (Arabidopsis thaliana) ameliorated the drought tolerance of two transgenic lines and increased ABA content by 42.7% and 14.3% compared to wild-type plants, respectively. Notably, PtoWRKY68hap1 (associated with drought tolerance) is ubiquitous in accessions in water-deficient environments, whereas the drought-sensitive allele PtoWRKY68hap2 is widely distributed in well-watered regions, consistent with the trends in local precipitation, suggesting that these alleles correspond to geographical adaptation in Populus. Moreover, quantitative trait loci analysis and an electrophoretic mobility shift assay showed that SHORT VEGETATIVE PHASE (PtoSVP.3) positively regulates the expression of PtoWRKY68 under drought stress. We propose a drought tolerance regulatory module in which PtoWRKY68 modulates ABA signaling and accumulation, providing insight into the genetic basis of drought tolerance in trees. Our findings will facilitate molecular breeding to improve the drought tolerance of forest trees.

WHIRLY1 Acts Upstream of ABA-Related Reprogramming of Drought-Induced Gene Expression in Barley and Affects Stress-Related Histone Modifications.

WHIRLY1, a small plant-specific ssDNA-binding protein, dually located in chloroplasts and the nucleus, is discussed to act as a retrograde signal transmitting a stress signal from the chloroplast to the nucleus and triggering there a stress-related gene expression. In this work, we investigated the function of WHIRLY1 in the drought stress response of barley, employing two overexpression lines (oeW1-2 and oeW1-15). The overexpression of WHIRLY1 delayed the drought-stress-related onset of senescence in primary leaves. Two abscisic acid (ABA)-dependent marker genes of drought stress, HvNCED1 and HvS40, whose expression in the wild type was induced during drought treatment, were not induced in overexpression lines. In addition, a drought-related increase in ABA concentration in the leaves was suppressed in WHIRLY1 overexpression lines. To analyze the impact of the gain-of-function of WHIRLY1 on the drought-related reprogramming of nuclear gene expression, RNAseq was performed comparing the wild type and an overexpression line. Cluster analyses revealed a set of genes highly up-regulated in response to drought in the wild type but not in the WHIRLY1 overexpression lines. Among these genes were many stress- and abscisic acid (ABA)-related ones. Another cluster comprised genes up-regulated in the oeW1 lines compared to the wild type. These were related to primary metabolism, chloroplast function and growth. Our results indicate that WHIRLY1 acts as a hub, balancing trade-off between stress-related and developmental pathways. To test whether the gain-of-function of WHIRLY1 affects the epigenetic control of stress-related gene expression, we analyzed drought-related histone modifications in different regions of the promoter and at the transcriptional start sites of HvNCED1 and HvS40. Interestingly, the level of euchromatic marks (H3K4me3 and H3K9ac) was clearly decreased in both genes in a WHIRLY1 overexpression line. Our results indicate that WHIRLY1, which is discussed to act as a retrograde signal, affects the ABA-related reprogramming of nuclear gene expression during drought via differential histone modifications.

Abscisic-Acid-Modulated Stomatal Conductance Governs High-Temperature Stress Tolerance in Rice Accessions

Rising air temperature is a major constraint for crop productivity under the current climate change scenario. Rice crops are known to be sensitive to high-temperature (HT) stress at anthesis and post-anthesis stages. Photosynthesis is an important metabolic process and is affected by HT stress. A pot study was planned to screen a set of seventy-three Indian rice accessions based upon changes in the rate of photosynthesis (Pn) and related gas exchange traits under HT, and to characterize the contrasting rice accessions for component traits of HT stress tolerance. All accessions were raised under ambient temperature (AT) until the booting stage and exposed to HT using controlled chambers at anthesis and post-anthesis. HT exposure led to a large reduction (up to 50%) in Pn, but stomatal conductance (gs) and the rate of transpiration (E) increased significantly across the rice accessions. Based on the photosynthetic response under HT, two contrasting rice accessions (IRGC 135883, tolerant, and IRGC 127222, sensitive) were selected and characterized for HT tolerance, along with an NL-44 check. Among them, Pn decreased marginally but gs and E showed significant increases under HT in the tolerant accession, while sensitive accession showed an up to 50% reduction in Pn and marginal increase in gs and E. No significant changes were recorded for chlorophyll fluorescence (Fv/Fm) in both the genotypes, but tissue temperature depression (TTD) was higher in IRGC 135883 accession under HT. Endogenous abscisic acid (ABA) content increased under HT in the flag leaf of both the accessions, and the highest increase was observed in the sensitive accession. Similarly, spikelet fertility and grain yield showed large reductions in sensitive rice accession under HT. A large increase in ABA concentration in the leaves of the sensitive rice accession might be affecting its gs and cooling capacity under an HT environment. Finally, the study concludes that tolerant rice accessions can be recommended as donors and exploited in future rice breeding programs for developing climate-resilient rice genotypes.

Effect of Abscisic Acid on Growth, Fatty Acid Profile, and Pigment Composition of the Chlorophyte Chlorella (Chromochloris) zofingiensis and Its Co-Culture Microbiome.

Microalga Chlorella (Chromochloris) zofingiensis has been gaining increasing attention of investigators as a potential competitor to Haematococcus pluvialis for astaxanthin and other xanthophylls production. Phytohormones, including abscisic acid (ABA), at concentrations relevant to that in hydroponic wastewater, have proven themselves as strong inductors of microalgae biomass productivity and biosynthesis of valuable molecules. The main goal of this research was to evaluate the influence of phytohormone ABA on the physiology of C. zofingiensis in a non-aseptic batch experiment. Exogenous ABA stimulated C. zofingiensis cell division, biomass production, as well as chlorophyll, carotenoid, and lipid biosynthesis. The relationship between exogenous ABA concentration and the magnitude of the observed effects was non-linear, with the exception of cell growth and biomass production. Fatty acid accumulation and composition depended on the concentration of ABA tested. Exogenous ABA induced spectacular changes in the major components of the culture microbiome of C. zofingiensis. Thus, the abundance of the representatives of the genus Rhodococcus increased drastically with an increase in ABA concentration, whereas the abundance of the representatives of Reyranella and Bradyrhizobium genera declined. The possibilities of exogenous ABA applications for the enhancing of the biomass, carotenoid, and fatty acid productivity of the C. zofingiensis cultures are discussed.

Genome-Wide Characterization and Expression Profiling of HD-Zip Genes in ABA-Mediated Processes in Fragaria vesca.

Members of homeodomain-leucine zipper (HD-Zip) transcription factors can play their roles by modulating abscisic acid (ABA) signaling in Arabidopsis. So far, our knowledge of the functions of HD-Zips in woodland strawberries (Fragaria vesca), a model plant for studying ABA-mediated fruit ripening, is limited. Here, we identified a total of 31 HD-Zip genes (FveHDZ1-31) in F. vesca, and classified them into four subfamilies (I to IV). Promoter analyses show that the ABA-responsive element, ABRE, is prevalent in the promoters of subfamily I and II FveHDZs. RT-qPCR results demonstrate that 10 of the 14 investigated FveHDZs were consistently >1.5-fold up-regulated or down-regulated in expression in response to exogenous ABA, dehydration, and ABA-induced senescence in leaves. Five of the six consistently up-regulated genes are from subfamily I and II. Thereinto, FveHDZ4, and 20 also exhibited significantly enhanced expression along with increased ABA content during fruit ripening. In yeast one-hybrid assays, FveHDZ4 proteins could bind the promoter of an ABA signaling gene FvePP2C6. Collectively, our results strongly support that the FveHDZs, particularly those from subfamilies I and II, are involved in the ABA-mediated processes in F. vesca, providing a basis for further functional characterization of the HD-Zips in strawberry and other plants.

Dual-functional SERRS and fluorescent aptamer sensor for abscisic acid detection via charged gold nanorods.

Abscisic acid (ABA) is a plant hormone, which plays an important role in plant growth, crop cultivation and modern agricultural engineering management. Accordingly, the detection of ABA content combined with new techniques and methods has become a more and more popular problem in the field of agricultural engineering. In this work, a SERRS and fluorescence dual-function sensor based on the fluorescence quenching and Raman enhancement properties of gold nanorods (AuNRs) was developed, and applied to the detection of plant hormone ABA. The dual-function reporter molecule Rhodamine isothiocyanate (RBITC) and complementary DNA (cDNA) were modified on AuNRs (AuNRs@RBITC@cDNA) as signal probes and aptamer modified magnetic nanoparticles (Fe3O4MNPs@Apt) as capture probes. Through the specific recognition of ABA aptamer and its complementary chains, an dual-function aptamer sensor based on SERRS and fluorescence was constructed. When ABA molecules were present in the detection system, the signal probes were detached from the capture probes due to the preferential binding between aptamer and ABA molecules. SERS signal of the reporter molecules appeared in the supernatant after magnetic separation, and it increased with the increase of ABA concentration. If the etching agent that can etch AuNRs was added to the supernatant, the AuNRs was etching disappeared, then the signal molecules fall off from the AuNRs, and the fluorescence signal intensity would recovered. The intensity of fluorescence signal also increased with the increase of ABA concentration. Thus, the quantitative relationship between ABA concentration and SERRS intensity and fluorescence intensity of signal molecules was established. The linear range of SERRS detection was 100 fM–0.1 nM, the detection limit was 38 fM; The linear range of fluorescence detection was 1 pM–100 nM, the detection limit is 0.33 p.m. The constructed dual-effect sensor was used in the recovery laboratory of real ABA samples, the recovery rate was up to 85–108%.

Arabidopsis NPF5.1 regulates ABA homeostasis and seed germination by mediating ABA uptake into the seed coat

ABSTRACT Abscisic acid (ABA) is a plant hormone that induces seed dormancy during seed development and inhibits seed germination after imbibition. Although ABA is synthesized in the seed coat (testa), endosperm, and embryo, the physiological roles of the hormone derived from each tissue are not fully understood. We found that the gene encoding an Arabidopsis ABA importer, NPF5.1, was expressed in the seed coat during seed development. Dry seeds of loss-of-function npf5.1 mutants contained significantly higher levels of dihydrophaseic acid (DPA), an inactive ABA metabolite, than the wild type. The npf5.1 mutant also had a slight increase in ABA content. An increase in DPA was prominent in the fraction containing the seed coat and endosperm. Seed germination of the npf5.1 mutant was similar to the wild type in the presence of ABA or the gibberellin biosynthesis inhibitor paclobutrazol. However, a mutation in NPF5.1 suppressed the paclobutrazol-resistant germination of npf4.6, a mutant impaired in an ABA importer expressed in the embryo. These results suggest that ABA uptake into the seed coat mediated by NPF5.1 is important for ABA homeostasis during seed development and for regulating seed germination.

H2O2 participates in ABA regulation of grafting-induced chilling tolerance in cucumber.

Rootstock provides more abscisic acid (ABA) content to scions to increase the chilling tolerance of seedlings. H2O2 is involved in ABA regulation of grafting-induced chilling tolerance of cucumber. Here we examined the role of ABA in the response of grafted cucumber to chilling stress. The data showed chilling induced an increase in leaf and root ABA content and there was a positive correlation between ABA content and the chilling tolerance of the varieties. The increase of ABA content and NCED mRNA abundance in the leaf of both Cs/Cs (self-root) and Cs/Cm (grafted with pumpkin as rootstock) showed a delay under aerial stress compared with those under whole plant and root-zone stress. Intriguingly, an increase in ABA in xylem was found under whole-plant and root-zone chilling stress but was not detected under aerial stress, implying the increases in ABA content in leaves were mainly from root ABA transportation. Compared to Cs/Cs, a higher ABA content and NCED mRNA abundance were observed in Cs/Cm, which showed that Cm could output more ABA than Cs. The removal of endogenous ABA decreased the difference in chilling tolerance induced by Cm, as evidenced by the observed similar oxidative stress levels and photosynthetic capacity between Cs/Cs and Cs/Cm after chilling stress. Moreover, we found that the H2O2 signal in grafted cucumber could respond to chilling stress earlier than the H2O2 signal in self-rooted cucumber. The inhibition of endogenous H2O2 decreased the chilling tolerance of grafted cucumber induced by ABA by reducing photosynthesis and the mRNA abundance of CBF1 and COR. Thus, our results indicate that H2O2, as the downstream signal, participated in the rootstock-induced chilling tolerance of grafted seedlings induced by ABA.

Hormonal responses to short-term and long-term water deficit in native Scots pine and Norway spruce trees

Water stress has a continuing major negative impact on global forest performance, which necessitates to understand how plants coordinate multiple physiological responses to adverse conditions. Therefore, we studied the effects of short-term and long-term water deficit on the biosynthesis and signalling of phytohormones, especially abscisic acid (ABA), in current-year needles of Scots pine and Norway spruce trees growing in a natural forest in the Bryansk region of Russia. In spruce, clear physiological effects of long-term water shortage were evident, whereas pine trees were affected more by short-term water deficit. However, both species presented similar pronounced increases in ABA content under long-term water deficit. Long-term ABA accumulation was not associated with changes in stomatal density or osmotic adjustment in either species. The dynamics of jasmonates and salicylic acid were similar; the contents of both decreased throughout the entire short-term water deficit period, and the contents were correlated with the decrease in phenolic compounds in the pine trees. The same decrease was observed for auxin, whereas cytokinins decreased only in more tolerant pine. Possible physiological reasons for the dynamics of plant hormone pools in trees under water deficit are discussed. • Pine and spruce were differentially affected by short-term and long-term water deficit. • Sustained increases in ABA were induced in response to long-term water deficit in pine and spruce. • Jasmonate and SA contents decreased uniformly under short-term water deficit. • Needle morphophysiological traits were not correlated with phytohormone dynamics.

Changes in the abscisic acid, phenylpropanoids and ascorbic acid metabolism during strawberry fruit growth and ripening

The functional compounds and sensory attributes of strawberries are acquired during ripening; however, the molecular mechanisms regulating this process have not yet been fully elucidated. Therefore, in this study, we aimed to investigate the accumulation of abscisic acid (ABA), phenylpropanoids, and l-ascorbic acid (AsA) as well as their relative gene expression levels during this process. The accumulation of ABA during the fruit growth and ripening stages depends on the upregulation of FaNCED1 and FaNCED2 gene expression, and the downregulation of FaCYP707 expression. Moreover, in the initial stages, the ABA glycosylation pathway is activated as a provision for the ABA levels required for future events. As the ABA levels increase, FaPAL, FaC4H1, and FaC4H4 genes expression are upregulated, aiding in the synthesis of flavonoid precursors, which, in addition to the higher FaUFGT expression, directly reflects an improvement in anthocyanin. AsA content increases during fruit development and ripening and is dependent on the synthesis pathway, due to the upregulation of FaGLDH expression, and on the oxidation and recycling pathways via the regulation of expression levels of FaAPX and FaMR. These results suggest that the activation of FaNCED1 and the inhibition of FaCYP707A1 may contribute to the increase in ABA content, thereby stimulating anthocyanin and AsA synthesis and accumulation.

Genome-wide association study identifies variants of GhSAD1 conferring cold tolerance in cotton.

Cold stress is a major environmental factor affecting plant growth and development. Although some plants have developed resistance to cold stress, the molecular mechanisms underlying this process are poorly understood. Using genome-wide association mapping with 200 cotton accessions collected from different regions, we identified variations in the short chain alcohol dehydrogenase gene, GhSAD1, that responds to cold stress. Virus-induced gene silencing and overexpression in Arabidopsis revealed that GhSAD1 fulfils important roles in cold stress responses. Ectopic expression of a haploid genotype of GhSAD1 (GhSAD1HapB) in Arabidopsis increased cold tolerance. Silencing of GhSAD1HapB resulted in a decrease in abscisic acid (ABA) content. Conversely, overexpression of GhSAD1HapB increased ABA content. GhSAD1HapB regulates cold stress responses in cotton through modulation of C-repeat binding factor activity, which regulates ABA signalling. GhSAD1HapB induces the expression of COLD-REGULATED (COR) genes and increases the amount of metabolites associated with cold stress tolerance. Overexpression of GhSAD1HapB partially complements the phenotype of the Arabidopsis ABA2 mutant, aba2-1. Collectively, these findings increase our understanding of the mechanisms underlying GhSAD1-mediated cold stress responses in cotton.

The yellowhorn AGL transcription factor gene XsAGL22 contributes to ABA biosynthesis and drought tolerance in poplar.

Regulation of abscisic acid (ABA) biosynthesis helps plants adapt to drought stress, but the underlying molecular mechanisms are largely unclear. Here, a drought-induced transcription factor XsAGL22 was isolated from yellowhorn (Xanthoceras sorbifolium Bunge). Yeast one-hybrid and electrophoretic mobility shift assays indicated that XsAGL22 can physically bind to the promoters of the ABA biosynthesis-related genes XsNCED6 and XsBG1, and a dual-luciferase assay showed that XsAGL22 activates the promoters of the later two genes. Transient overexpression of XsAGL22 in yellowhorn leaves also increased the expression of XsNCED6 and XsBG1 and increased cellular ABA levels. Finally, heterologous overexpression of XsAGL22 in poplar increased ABA content, reduced stomatal aperture and increased drought resistance. Our results suggest that XsAGL22 is a powerful regulator of ABA biosynthesis and plays a critical role in drought resistance in plants.

Concentrations-dependent effect of exogenous abscisic acid on photosynthesis, growth and phenolic content of Dracocephalum moldavica L. under drought stress

At the same time, the plant increases the production of phenolic compounds and essential oil as a response to stress conditions.In the semi-arid regions, drought stress is the most important environmental limitations for crop production. Abscisic acid (ABA) plays a crucial role in the reactions of plants towards environmental stress such as drought. Field experiments for two consecutive years in 2016 and 2017 were conducted to evaluate the effect of three watering regimes (well-watered, moderate and severe drought) and five exogenous ABA concentrations (0, 5, 10, 20 and 40 μM) on growth, photosynthesis, total phenolic and essential oil content of Dracocephalum moldavica L. Without ABA application, the highest photosynthetic rate (6.1 μmol CO2 m−2 s−1) was obtained under well-watered condition and, moderate and severe drought stress decreased photosynthesis rate by 26.39% and 34.43%, respectively. Some growth parameters such as stem height, leaf area, leaf dry weight and biological yield were also reduced by drought stress. ABA application showed a decreasing trend in photosynthesis rate and mentioned plant growth parameters under all moisture regimes. The highest seed yield (1243.56 kg ha−1) was obtained under well-watered condition without ABA application. Increasing ABA concentration decreased seed yield in all moisture regimes. The highest total phenolic content (8.9 mg g−1 FW) and essential oil yield (20.58 kg ha−1) were obtained from 20 and 5 μM ABA concentration, respectively, under moderate drought stress.

Transcriptome and proteome analyses of the molecular mechanisms underlying changes in oil storage under drought stress in Brassica napus L.

AbstractRapeseed (Brassica napus L.) is the second most important oilseed crop in edible vegetable oil and bioenergy; however, drought stress generally causes a decrease in rapeseed yield and oil content, especially during the reproductive stage. In our study, we measured the oil and protein contents and gibberellic acid (GA) and abscisic acid (ABA) levels in seeds that were acquired on the 30th, 40th, and 50th days after flowering under control and drought treatments. RNA and protein libraries were constructed from the stressed seeds to perform transcriptome and proteome analyses, respectively. Our results demonstrated that the oil content decreased due to four primary mechanisms: downregulation of fatty acid biosynthesis‐associated genes and proteins; upregulation of fatty acid degradation‐associated genes and proteins; enhancement of protein storage due to changes in the abundances of relevant genes and proteins; and upregulation of Gly‐Asp‐Ser‐Leu (GDSL) gene expression, potentially as the result of upregulating the GA biosynthesis gene GA20ox3 and downregulating the GA inactivating gene GA2ox3 and thus an increase in GA content. During seed maturation, oil storage change may also relate to increasing ABA content as the upregulation of two members of NCED6 (9‐cis‐epoxycarotenoid dioxygenase) gene family involved in ABA biosynthesis, and the upregulation of genes involved in ABA signal transduction. These results will help to establish a foundation for breeding excellent varieties of rapeseed with high oil content for areas with frequent droughts to promote the supply of edible vegetable oil and biofuel.
