Periparturient blood T-lymphocyte PD-1 and CTLA-4 expression as potential predictors of new intramammary infections in dairy cows during early lactation (short communication)

BackgroundThe periparturient period in dairy cows is marked by immunosuppression which increases the likelihood of infectious disorders, particularly also mastitis. An in-depth understanding of peripartum leukocyte biology is vital for the implementation of highly successful post-partum disease prevention measures. Immune checkpoint molecules, such as programmed death 1 (PD-1) and cytotoxic T-lymphocyte antigen 4 (CTLA-4), are critical inhibitory receptors expressed on immune cells, particularly T cells, that drive immunosuppressive signaling pathways. However, the potential role of immune checkpoint molecules expression in T-cells on udder health has never been explored. Thus, the association between the occurrence of new postpartum intramammary infections (IMIs) and the expression of programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte-associated antigen-4 (CTLA-4) on blood T-cells during the peripartum period was investigated.ResultsIn this study, the incidence of IMIs by any pathogen in early lactation was not associated with a higher expression of PD-1 and CTLA-4 in the periparturient period. However, the incidence of IMIs by major pathogens throughout the first month of lactation was significantly associated with higher expression of PD-1 at 14 days before calving (P = 0.03) and CTLA-4 at parturition (P = 0.03) by blood T-cells. Also, the expression of CTLA-4 at D0 (P = 0.012) by T-cells was associated with the occurrence of persistent IMIs during the first month of lactation.ConclusionsTo our knowledge, this is the first report to investigate the expression of PD-1 and CTLA-4 by blood T-lymphocytes during the periparturient period in dairy cows and to explore their relationship with the incidence of new IMIs in the postpartum period. Thus, a comprehensive understanding of leukocyte biology during peripartum would appear to be a prerequisite for the identification of resilient dairy cows or targets innovative (immunological) non-antibiotic approaches in the transition period.

Associations of cow- and herd-level factors during the dry period with indicators of udder health in early-lactation cows milked by automated milking systems

This observational study aimed to determine the association of cow-level factors and herd-level housing and management practices during the dry period with indicators of udder health in early-lactation cows in automated milking system (AMS) herds. Data were collected from 166 commercial AMS dairy farms (mean ± standard deviation = 116 ± 111 milking cows; range = 39 to 1,200) across Canada between October 2018 and September 2020. Information on herd demographics, housing, and management practices was obtained on each farm using 2 surveys. On each farm, we selected all cows that had available Dairy Herd Improvement (DHI) somatic cell count (SCC) data for their last milk test before dry-off (>250 d in milk) and their first milk test after calving (5-45 d in milk). Data from 14,007 cows were included after excluding cows with a dry period of <30 d and >120 d. Using the SCC data, we calculated for each cow the somatic cell score (SCS) for the last milk test before dry-off (PreSCS) and the first milk test after calving (PostSCS), which we then averaged per herd at a test-day level. Intramammary infection (IMI) was estimated using cow SCC data. Each cow was classified as not infected (SCC <200,000 cells/mL) or infected (SCC ≥200,000 cells/mL) at her last milk test before dry-off and her first milk test after calving. Based on this classification, cows were further categorized as never infected, always infected, new IMI, or cured IMI. At the cow level, a higher PostSCS was associated with longer dry periods. The odds of having a new IMI were higher for cows of higher parity and that had lower 305-d milk yield before dry-off. Cows with lower parity were more likely to cure an IMI. At the herd level, a higher 305-d milk yield before dry-off was associated with a lower incidence of new IMI and a higher incidence of cured IMI. Separating cows into a different pen as preparation for dry-off tended to be associated with a lower PostSCS and incidence of new IMI. At dry-off, herds that used teat sealants and blanket antibiotic dry cow therapy also had lower PostSCS. During the dry period, housing cows in different groups was associated with a higher PostSCS and a lower incidence of cured IMI, while housing cows in both pack pens and stalls compared with only pack pens was associated with a lower incidence of new IMI. Finally, placing cows onto the AMS to be milked one or more days after calving tended to be associated with a lower PostSCS compared with placing them in the AMS within the first day postpartum. In summary, indicators of udder health in early-lactation cows in AMS herds were associated with several cow-level factors and herd-level housing and management practices before dry-off, at dry-off, during the dry period, and at the beginning of lactation. Thus, if some of the associations identified are causal, AMS producers may be able to improve udder health through modifications of housing and management practices.

Randomized noninferiority field trial evaluating a postmilking teat dip for the prevention of naturally occurring intramammary infections

The aim of this study was to perform a positive-controlled field study under natural exposure conditions to test the efficacy of a newly developed chlorine dioxide-based postmilking teat disinfectant (experimental product, EX) for noninferiority compared with an already established chlorine dioxide-based teat disinfectant (positive control product, PC). After blocking by parity, approximately 200 Holstein cows in early to mid-lactation stages from a dairy farm near Padua, Italy, were randomly assigned to one of 2 groups. Over a 13-wk period between September and December 2021, the teats of cows were dipped with the EX or the PC after each milking. Milk samples were collected from individual quarters of enrolled cows for 13 wk to determine infection status. Teat condition was assessed at wk 1, 5, and 9. Mixed logistic regression was used to analyze the effect of treatment on the incidence of new intramammary infections. For the noninferiority analysis, the upper limit of the 95% confidence interval for the difference in new intramammary infection (NIMI) rate between the 2 treatments (EX - PC) had to be to the left of the critical value d (0.035) to conclude that EX was noninferior to PC in terms of the risk of NIMI. The results showed that the incidence of new infections in the quarters treated with EX (3.1%) was not different from that in the udder quarters treated with PC (2.6%). No overall difference was found between the treatments in terms of teat condition. As the upper limit of the 95% confidence interval of the NIMI rate difference was smaller than the predefined noninferiority limit, we concluded that the EX was noninferior compared with the PC.

The effectiveness of an internal teat sealant in preventing new intra mammary infections in dairy cattle during dry period

The aim of our study was to evaluate the effect of an internal teat sealant infused upon drying off on the incidence of new intra-mammary infections (IMI) during the dry period in dairy cattle. Due to the non-availability of the product on the Indian market, the product was prepared in a laboratory using bismuth subnitrate and liquid paraffin. A total of 64 quarters free from infection on culture were selected for the study at 60 days before the expected date of parturition. The quarters were randomly divided into two groups (group A and group B), with 32 quarters in each group. The group A quarters were infused with teat sealant at the time of drying off, and the quarters in group B were kept as the control and no treatment was provided to them. Milk samples from all the selected quarters were subjected to cultural examination at the time of drying off and 1-3 days post calving. The incidence of new IMI’s between drying off and calving was significantly lower for group A quarters when compared to group B quarters (12.5% vs 34.4%). The incidence of new IMI’s in group A and group B quarters was significantly lower for Streptococcus uberis (3.13% vs 18.75%) and Streptococcus dysgalactiae (3.13% vs 12.5%). The study concluded that infusion of teat sealant at drying off is helpful in lowering the incidence of new IMI’s during the dry period.

Noninferiority study evaluating the efficacy of a teat disinfectant containing copper and zinc for prevention of naturally occurring intramammary infections in an automatic milking system

The aim of this study was to demonstrate the noninferiority of a novel teat disinfectant based on copper and zinc (ZkinCu; Copper Andino, Santiago de Chile, Chile) compared with a previously proven glycolic acid active disinfectant (OceanBlu; DeLaval, Kansas City, MO) as a positive control, with respect to the incidence of new intramammary infections under natural challenge conditions on a commercial robotic dairy farm. This study was conducted in 6 robotic pens of approximately 60 milking cows each. The pens were randomly assigned to 1 of the 2 studied disinfectants. Throughout the 8 wk study, the same pre- and post-milking teat disinfectant was used in each pen. The same milking procedures were used in each robot throughout the study. Pre-milking hygiene consisted of applying the disinfectant (OceanBlu or ZkinCu) with the robotic arm. The same product was applied on the teats after milking. At the beginning of the study, all quarters of all study cows were sampled. In successive samplings (wk 2, 4, 6, and 8), composite milk samples were collected on farm to determine SCC. Once composite SCC results were available (2 d) and based on an SCC of ≥100,000 cells/mL, quarter milk samples underwent bacteriological culture. Clinical mastitis was identified by study personnel. Intramammary infection in biweekly quarter milk samples was determined based on composite SCC levels (≥100,000 cells/mL) and the presence of bacteria. A new IMI was defined as a quarter in which the organism isolated was not present in the previous bacteriological sample, or the previous composite SCC sample was <100,000 cells/mL. Clinical mastitis samples were also considered to be new IMI. The trial was designed as a positive control field trial, in which the objective was to show noninferiority of ZkinCu versus the control (OceanBlu). The overall crude incidences of new IMI for 2 wk at risk were 4.9 and 7.3% for the ZkinCu and OceanBlu groups, respectively. The predominant organisms recovered from quarters with new IMI were Streptococcus uberis, Corynebacterium spp., and coagulase-negative staphylococci in both the ZkinCu and OceanBlu groups. The risk of infection in the OceanBlu group was higher (β = 0.644; 95% confidence interval = 0.05-1.22). The interaction of treatment by week was not significant. The new IMI rate estimates (95% confidence interval) for ZkinCu and OceanBlu were 1.7% (0.8-2.5) and 3.2% (1.7-4.7), respectively. One novel aspect of this study is that it was one of the first commercial noninferiority trials to evaluate a new pre- and post-milking teat disinfectant in a dairy herd with an automatic milking system. The experimental teat disinfectant ZkinCu, evaluated in this field trial with naturally occurring IMI, showed noninferiority relative to the positive control for the prevention of new IMI. This study was conducted in a herd with an automatic milking system, and the results are applicable to herds with similar characteristics. Additional studies are needed to ensure reproducibility under different management conditions.

Staphylococcal intramammary infection dynamics and the relationship with milk quality parameters in dairy goats over the dry period

The objectives of this study were (1) to report the rates of new intramammary infection (IMI) and spontaneous IMI cure over the dry period in 3 dairy goat herds; (2) to evaluate the factors predicting infection dynamics over the dry period; and (3) to define milk quality parameter thresholds that predict infection dynamics over the dry period. Two consecutive udder-half milk samples were collected 10 to 14 d apart before dry-off from 288 goats in 3 herds, and 2 consecutive udder-half samples were collected 7 to 14 d apart in the following lactation, with the first sample being collected ≤10 d in milk, from 200 of the same goats. In 2 of the herds, udder-half milk samples were also collected at the same time points (n = 312 halves; 157 goats) for measurement of milk quality parameters. Standard aerobic culture of milk samples was performed for the detection of mastitis pathogens. To rule out the presence of Mycoplasma spp. IMI, milk samples were also cultured on modified Hayflick medium. Non-Mycoplasma isolates were speciated using MALDI-TOF mass spectrometry. Staphylococcal isolates, when not identified by MALDI-TOF, were speciated using partial gene sequence analysis of rpoB or tuf. When >1 sample from an udder half yielded the same species, available isolates from the first and last positive samples for that species were strain-typed using pulsed-field gel electrophoresis. Incidence of new IMI and cure rate were computed. Generalized linear mixed regression models were built to evaluate the associations between new IMI and pre-dry somatic cell score (SCS), between IMI persistence and half-level SCS, and between IMI persistence and pre-dry IMI species. Thresholds for pre-dry SCS and lactose concentration were computed to predict IMI persistence. Overall, 12.6% (48/380) of halves had a persistent IMI. Cumulative incidence of new IMI over the dry period was 13.2%, and cure rate was 52.0%. Pre-dry SCS was not associated with odds of new IMI or IMI persistence. Pre-dry IMI species was not associated with odds of persistence. Lactose concentration was not associated with odds of persistence. Regardless of culture data, the optimal pre-dry SCS threshold to detect IMI that would persist into the next lactation was 8.7, with sensitivity and specificity of 50 and 73.8%, respectively. Further studies on the effect of control measures on species-specific incidence and cure rates during the dry period are warranted.

Investigation of the relationship between bedding materials, bedding characteristics, and intramammary infection in late lactation dairy cows

Many cows acquire subclinical intramammary infections (IMI) during lactation, necessitating antimicrobial treatment at dry-off (dry cow therapy). If the dairy industry moves toward the adoption of selective dry cow therapy, one essential strategy to reduce mastitis-associated antimicrobial use at dry off will be to reduce the incidence of new IMI during lactation. One important source of mastitis-causing pathogens is bedding. However studies are needed to investigate the association between bedding management, bedding bacteria count (BBC), and IMI risk in late lactation cows.&#x0D; The objectives of this prospective cross-sectional study are to: 1) describe associations between BBC and IMI prevalence for individual pathogens and specific pathogen groups in late lactation cows, 2) identify bedding-related risk factors for high BBC, and 3) identify critical levels of bacteria in bedding that might be used as cut-points to guide practitioners when interpreting BBC reports. This study is ongoing, with winter 2018 results currently being assembled.

Randomized noninferiority study evaluating the efficacy of a postmilking teat disinfectant for the prevention of naturally occurring intramammary infections

The objective of this study was to complete a positive-control, natural exposure, noninferiority design field study to test the efficacy of a novel glycolic acid-based postmilking teat disinfectant as compared with a previously proven iodine-based postmilking teat disinfectant (positive control). The primary outcome of interest was the effect of treatment on incidence of new intramammary infections. Secondary outcomes included the effect of treatment on prevalence of infection, somatic cell count, and teat condition. After blocking by parity, approximately 300 early- to mid-lactation cows on a large Wisconsin dairy farm were randomly assigned to 1 of 2 groups. For a 12-wk period between May and August 2014, the 2 groups were dipped after each milking with either the experimental (EX) or positive control (PC) product. Individual quarters were sampled to establish bacteriological infection status at the beginning of the study, and every 2 wk thereafter, by use of a 2-stage process evaluating somatic cell count (SCC), and then culturing milk samples only when SCC exceeded a parity-specific threshold. Teat condition scoring was completed at the beginning of the study and on wk 4, 8, and 12. Mixed logistic regression was used to evaluate the effect of treatment on dichotomous outcome measures including the odds of acquiring a new infection during a given 2-wk sampling interval (incidence), the odds for presence of infection at sampling (prevalence), and odds for a normal teat skin condition score. Mixed linear regression was used to evaluate the effect of treatment on somatic cell count. For the noninferiority analysis, the upper bound of the 95% confidence interval for the difference in new infection rate between the 2 treatments (EX − PC), had to be to the left of the critical value d (0.035) to conclude that EX was noninferior relative to PC with respect to risk for new infections. Results showed that the incidence of new infections was not different for quarters dipped with EX (3.2%) as compared with PC (4.2%). Similarly, the prevalence of infection tended to be lower for quarters dipped with EX (3.92%) as compared with PC (5.03%). No overall difference was found between treatments when evaluating somatic cell count measures and teat condition scores. Because the upper bound of the 95% confidence interval of the new IMI rate difference was smaller than the predefined noninferiority limit, it was concluded that the experimental product was not inferior compared with the positive control. As such, the glycolic acid-based teat disinfectant evaluated in this study can be considered an effective postmilking teat disinfectant, as well as safe, in so far as the product was not irritating to teat skin and did not negatively affect skin condition measures, as compared with the positive control group.

Randomised controlled trials demonstrate efficacy of a novel internal teat sealant to prevent new intramammary infections in dairy cows and heifers

AIMS: The main aim was to investigate the effectiveness of a novel internal teat sealant (ITS) formulation containing bismuth subnitrate and the antiseptic chlorhexidine, in adult cows (Trial A) and nulliparous heifers (Trial B), to reduce the incidence of new intramammary infection (IMI) between treatment and calving and reduce the prevalence of IMI within 6 days following calving.METHODS: In Trial A dairy cows (n=326) without history of clinical mastitis and with a maximum composite milk somatic cell count <200,000 cells/mL during the current lactation were enrolled from three spring-calving mainly pasture-based herds immediately following the final milking. In Trial B all heifers (n=166) were enrolled from two herds approximately 3 weeks before the start of calving. Glands were randomly assigned to controls or treatment with ITS, and mammary secretion samples were taken from all glands prior to treatment, and on two occasions 0–4 and 3–6 days post-calving to determine the incidence of new IMI and the prevalence of IMI immediately post-calving. Additionally, cows in Trial A were monitored for cases of clinical mastitis (CM) in the non-lactating period and animals in both trials were monitored for cases of CM within 30 days of calving.RESULTS: The ITS reduced the estimated incidence of new IMI with both major and any pathogens compared to control glands in cows (6.2 vs. 14.2% and 14.7 vs. 32.8%, respectively) and in heifers (3.0 vs. 13.1% and 13.4 vs. 25.3%, respectively) (p<0.001). The ITS reduced the estimated prevalence of a major or any pathogen compared to untreated controls at both 0–4 and 3–6 days post-calving, in both cows and heifers. Additionally, the ITS reduced the incidence of CM in the non-lactating period in cows (−1.0%, p=0.01) and in the 30 days following calving in heifers (−2.0%, p=0.06), and tended to reduce the estimated incidence of CM in cows post-calving (−1.7%, p=0.10).CONCLUSIONS: The novel ITS reduced the incidence of new IMI, and the prevalence of IMI at calving in both adult cows and heifers, and reduced the incidence of CM in the non-lactating period of cows and in the first 30 days after calving in heifers.CLINICAL RELEVANCE: The results of these trials demonstrate the efficacy of a novel ITS for the control of mastitis in likely uninfected dairy cows over the non-lactating period and in heifers in late pregnancy.

Prevalence and incidence of intramammary infections in lactating dairy goats

AIM: To determine the prevalence of intramammary infection (IMI) of lactating dairy goats between 0 and 4 days postpartum, the prevalence and incidence rate of new IMI at Weeks 2, 14 and 27 of lactation, and the relationship between herd-level prevalence of IMI and bulk tank somatic cell count (BTSCC).METHODS: Milk samples were collected from 8% of a herd (total 624 does) from 18 dairy goat herds in the Waikato region of New Zealand, for bacteriology and somatic cell count (SCC) determination, from both glands within 4 days of kidding (Week 0) and again at Weeks 2, 14 and 27. Prevalence of IMI was determined at each time point and incidence rate calculated for Weeks 0–2, 2–14, and 14–27. Greenwood and Reed-Frost models were compared for estimation of the transmission parameter for all pathogens, and for Staphylococcus aureus, coagulase negative staphylococci (CNS) and Corynebacterium spp. separately.RESULTS: Bacteria were isolated from 1,122/4,814 (23.3%) glands, with CNS (13.4%) and Corynebacterium spp. (7.3%) being the most common isolates. Prevalence of any IMI increased with stage of lactation, varied among herds, and increased with age (all p<0.05). Incidence rate was 80, 24 and 7 new IMI/10,000 gland days for Weeks 0–2, 2–14 and 14–27, respectively. Incidence rate for any IMI increased with age and with the presence of an IMI in the contralateral gland, and varied among herds (p<0.001). The transmission of each pathogen was better modelled assuming contagiousness (Reed-Frost models), than not (Greenwood models).At gland level, IMI increased SCC at all stages of lactation (p<0.001). The gland prevalence of IMI within herds was positively associated with ln BTSCC at Week 2 (p=0.02), but not Weeks 14 or 27 (p>0.05).CONCLUSIONS: Prevalence of IMI increased with stage of lactation, but the highest incidence rate of new IMI occurred in early lactation. Models accounting for the contagious nature of infection fitted better than those not accounting for contagiousness. BTSCC was only associated with prevalence of IMI in early lactation.CLINICAL RELEVANCE: Reduction of BTSCC in dairy goats may be best achieved by minimising the prevalence and incidence of new IMI in early lactation. Further studies are required to define management factors associated with the between herd, and stage of lactation, effects on prevalence and incidence rate in order to reduce BTSCC throughout lactation.

The effect of internal teat sealant products (Teatseal and Orbeseal) on intramammary infection, clinical mastitis, and somatic cell counts in lactating dairy cows: A meta-analysis

The objective of this study was to assess the efficacy of internal teat sealant products containing bismuth subnitrate (Teatseal and Orbeseal; Pfizer Animal Health, West Ryde, Australia) when used alone, or in the presence of antibiotic dry cow therapy (ADCT), before or at drying off on the incidence of new intramammary infections (IMI), clinical mastitis, and milk somatic cell count (SCC) during lactation. The literature search identified 18 English-language publications on the use of Teatseal in dairy cattle. A total of 12 studies with 17 subtrials or comparisons including 13 positive control subtrials (internal teat sealant and ADCT vs. ADCT) and 4 negative control subtrials (internal teat sealant vs. untreated) examining IMI were included in the analysis. Internal teat sealants, alone or in the presence of ADCT, reduced the risk of acquiring new IMI after calving by 25% [risk ratio (RR)=0.75; 95% confidence interval (CI): 0.67 to 0.83]. Internal teat sealants reduced the risk of IMI by 73% compared with untreated cows (RR=0.27; 95% CI: 0.13 to 0.55). The results of both meta-analyses of IMI, with positive and negative controls, were heterogeneous [I2 (a statistic that describes the proportion of total variation in study effect estimates that is due to heterogeneity)=65.4 and 92.1%]. No farm or cow factors studied significantly contributed to the heterogeneity of the results. A total of 16 studies (21 subtrials), including 14 positive control subtrials and 7 negative control subtrials, examining clinical mastitis were included in the analysis. Internal teat sealants alone and in the presence of ADCT reduced the risk of clinical mastitis after calving in lactating cows by 29% (RR=0.71; 95% CI: 0.62 to 0.82), and 48% (RR=0.52; 95% CI: 0.37 to 0.75), respectively. The results of the meta-analysis on clinical mastitis with positive controls were homogeneous (I2=33.6%), whereas the results of studies with negative controls were heterogeneous (I2=60.4%). No farm or cow factors studied that had sufficient data to evaluate significantly contributed to the heterogeneity of the results. The estimated linear score (LS) of milk SCC after calving in published studies (n=3) and for studies that provided raw data (n=2), was significantly lower for cattle treated with internal teat sealants and ADCT in 3 studies than for cattle treated with internal teat sealants only. The estimated LS of pooled raw data of 3 studies from 32 herds showed that the LS of cows treated with internal teat sealant and ADCT was not significantly different than those treated with ADCT only. This study found that the application of internal teat sealants in the presence of ADCT or the use of internal teat sealants alone at dry off significantly reduced the incidence of IMI and clinical mastitis in lactating dairy cows compared with respective control groups. Further studies are needed to investigate the effect of internal teat sealants on postpartum milk SCC in lactating dairy cows.

Effect of disinfecting teats post-milking or pre- and post-milking on intramammary infection and somatic cell count

AIMS: To determine the effects of (a) post-milking teat disinfection compared with no disinfection and (b) pre- and post-milking teat disinfection compared with post-milking disinfection alone, on the incidence of new intramammary infection (IMI), somatic cell count (SCC) and teat skin abnormalities in dairy cows. METHODS: In Experiment 1, dairy cows in five dairy herds were randomly allocated to a post-milking teat disinfection group (n=230), that was sprayed with an iodine-based disinfectant (TeatguardPlus) for a complete lactation, or to a non-disinfected group (n=239). In Experiment 2, cows were randomly allocated to post-milking teat disinfection (n=239) or both pre- and post-milking teat disinfection (n=235), using a chloramine-T-based disinfectant (Teatsweet) for both treatments, from calving to 118–127 days in milk. The incidence of new IMI was determined by aseptic sampling of all quarters at calving, during lactation, and at trial end or at drying-off, with clinical mastitis cases sampled on detection. SCC and teat skin abnormalities were measured at 2-monthly intervals during lactation. In both experiments, disinfectant was applied by spray application. RESULTS: Cows that received post-milking teat disinfection had a lower incidence of new IMI caused by Staphylococcus aureus, Streptococcus uberis, Corynebacterium spp and coagulase negative staphylococci, had lower bulk milk SCC during lactation, and had fewer teat skin abnormalities compared with the non-disinfected cows (p < 0.05). Pre-milking teat disinfection, in addition to post-milking teat disinfection, did not reduce the incidence of new IMI for any pathogens and did not reduce SCC (p> 0.05). CONCLUSIONS: Post-milking teat disinfection applied as a spray is a key component in mastitis control in New Zealand. There was no benefit from the addition of pre-milking disinfection. CLINICAL RELEVANCE: This study confirms previous findings of the effectiveness of post-milking teat disinfection in reducing the incidence of IMI caused by the common mastitis-causing pathogens in New Zealand, and presents the first results of a controlled study examining pre-milking teat spraying undertaken in New Zealand commercial dairy herds.

The comparative efficacy of two long-acting dry-cow cephalonium products in curing and preventing intramammary infections

AIMS: To test equivalence between two different cephalonium dry-cow therapy (DCT) products with regard to the incidence of bacteriological cure of existing infections of quarters, the incidence of new intramammary infections (IMI) from 2–5 days postpartum and the incidence of clinical mastitis from drying-off to 21 days postpartum. METHODS: All cows (n=1,570) on four commercial dairy farms in Southland were eligible for inclusion. Power analysis using a variance inflation factor indicated a required minimum of 2,101 quarters per treatment group. Cows were blocked by herd, parity, previous history of mastitis and most recent somatic cell count (SCC), and then randomly allocated to either of two cephalonium treatments at the cow level, viz the treatment group (new formulation cephalonium) or the positive control group (existing reference formulation cephalonium). All quarters within a cow were assigned the same treatment. Samples collected from all quarters with a SCC ≥500,000 cells/mL at drying-off, and those with a positive culture postpartum were cultured, as well as samples from all quarters collected between 2–5 days postpartum. All cases of mastitis were recorded and sampled for culture. The risk of quarter-level incidence of bacteriological cure, IMI and clinical mastitis was modelled using a GLM and generalised estimating equation (GEE), including the effects of treatment group, age,farm, udder-health status at drying-off and length of dry period. RESULTS: For 829 infected quarters, the estimated incidence of bacteriological cure for all pathogens was 78.0 (95% CI=64.3–91.6)% for the treatment group and 75.7 (95% CI=61.6–89.8)% for the positive control group (p=0.71). Incidence of cure varied with the farm (p=0.001), type of pathogen pre-treatment (p=0.009) and log10SCC at drying-off (p<0.001). For 4,530 quarters, the estimated incidence of new IMI, with any pathogen, for treatment and positive control groups was 16.1 (95% CI=13.1–19.7)% and 16.0 (95% CI=13.0–19.5)% respectively (p=0.91). Incidence of IMI varied with farm (p<0.001), pathogen and SCC status before treatment (p<0.001), and length of the dry period (in days) (p<0.001). For 5,136 quarters, the estimated incidence of clinical mastitis was 1.0 (95% CI=0.5–1.7)% in the treatment group and 1.1 (95% CI=0.6–2.0)% in the positive control group (p=0.64). CONCLUSIONS: Use of the two different cephalonium products at drying-off in four dairy herds in Southland resulted in equivalence with respect to incidence of bacteriological cure of existing infections, incidence of new IMI postpartum, and incidence of clinical mastitis in early lactation.

The effect of selenium supplementation before calving on early-lactation udder health in pastured dairy heifers

Selenium (Se) deficiency has been associated with lowered resistance to mastitis in dairy cattle. However, little published data exists on the effect of Se supplementation before calving on udder health of pastured dairy heifers. Further, the relative efficacy of injectable barium selenate and oral organic Se for improving udder health in cows has not previously been tested. The objectives of this study were to determine the effects of precalving Se supplementation and type of supplementation on the blood activity of glutathione peroxidase and measures of udder health immediately after calving and during the first month of lactation in pastured dairy heifers. One hundred forty pregnant Chilean Holstein-Friesian heifers were fed a basal diet containing, on average, 0.15mg of Se/kg of dry matter. One month before predicted calving, heifers were allocated to 1 of 3 groups. Group 1 (n=49) received no supplementary Se, group 2 (n=46) received a single subcutaneous injection of Se (1mg/kg of live weight, as barium selenate), and group 3 (n=45) was fed Se yeast (3mg/heifer/d until calving). Heifers supplemented with barium selenate had a higher glutathione peroxidase activity from 14 d in milk onwards. Selenium supplementation, irrespective of source, tended to reduce the prevalence of intramammary infection (IMI) and decrease the prevalence of quarters with high somatic cell count (SCC) at calving. Overall, Se supplementation did not result in a reduction of the incidence of new IMI or clinical mastitis or in decreased SCC during the balance of the first month of lactation. However, in pasture-based heifers injected with barium selenate before calving, and fed diets with 1.3 and 2.5mg of Se/d precalving and during lactation, respectively, no cases of clinical mastitis were observed in the first month of lactation.

Risk of New Intramammary Infections during the Dry Period in Holstein Dairy Cattle Enrolled in the CBMRN National Cohort of Dairy Farms

Intramammary infections acquired during the dry period are difficult to diagnosis and tend to lead to decreased production in the subsequent lactation. In addition, the use of dry cow therapies (DCT) have virtually eliminated major pathogens, such as Streptococcus agalactiae. However, use of dry cow therapy or teat sealants may have questionable efficacy against other pathogens. Selective dry cow therapy, based on farm- or cow-specific pathogen profiles, tend to decrease usage of antimicrobial products. However, to generate farm-specific pathogen profiles, knowledge of incidence of intramammary pathogens is needed.&#x0D; The objectives of this observational study were to determine the incidence of new intramammary infections (NIMI) in dairy cattle during the dry period and risk factors associated with NIMI incidence risk.

A review of prevention and control of heifer mastitis via non-antibiotic strategies

Clinical and subclinical mastitis is a significant problem in primiparous dairy cattle (heifers) with a higher prevalence and incidence in heifers than cows, especially early in lactation. Differences in management (e.g. nutrition, pasturing, no use of dry cow therapy) as well as differences in physiological status (e.g. continuing growth in heifers) are likely contribute to the observed differences between heifers and cows. These differences may result in the requirement for different approaches for mastitis management in heifers than for cows. Mastitis is a multifactorial disease, hence control requires an understanding of the risk factors before effective interventions can be defined. Control strategies are aimed at reducing the incidence of new intramammary infections and eliminating existing infections. Potential strategies can include improved environmental and animal hygiene, application of internal and external teat sealants, prepartum application of teat antiseptics, prepartum milking and control of horn fly in environments where it acts as vector. Other less well-proven strategies to control heifer mastitis include management of heifers as a physically separate group from older cows and not feeding mastitic milk to calves. It is concluded that several well-proven strategies are available to manage heifer mastitis. However, further research is likely to improve understanding of heifer mastitis and lead to novel managerial approaches to mastitis control in this age group.

Quarter-Level Analysis of Subclinical and Clinical Mastitis in Primiparous Heifers Following the Use of a Teat Sealant or an Injectable Antibiotic, or Both, Precalving

This study investigated the effect of infusion of a bismuth subnitrate teat canal sealant or an injectable antibiotic, or both, in heifers on the cure of existing intramammary infection (IMI), incidence of new IMI, prevalence of postcalving IMI, and incidence of clinical mastitis in the first 2 wk postcalving at the quarter level. Heifers (n = 1,067) in 30 seasonally calving, pasture-fed dairy herds were randomly assigned at the heifer level to 1 of 4 treatments (no treatment; 3 intramuscular injections of 5g of tylosin antibiotic at 24-h intervals; infusion of a teat sealant into all 4 quarters; 3 intramuscular injections of 5g of tylosin antibiotic and infusion of teat sealant into all 4 quarters). Mammary gland secretion samples were collected from each quarter of every heifer before treatment. Heifers within a herd were enrolled on one calendar day, 27 d (on average) before the planned start of the seasonal calving period. Duplicate milk samples were collected from each gland within 5 d after calving for bacterial culture and from glands the herdowners diagnosed as having clinical mastitis. The relative risk of effect of treatment on the incidence of cure, incidence of new IMI, prevalence of postcalving IMI, and incidence of clinical mastitis were calculated at the gland level using multivariate logistic regression analyses. Neither infusion of a teat sealant nor treatment with the injectable antibiotic increased the cure of precalving IMI. Infusion of the teat sealant reduced the risk of new IMI with any pathogen by 74%, reduced the prevalence of postcalving IMI by 65%, reduced the risk of new infection with Streptococcus uberis by 70% in quarters with an IMI precalving, and reduced the incidence of clinical mastitis from which a pathogen was isolated by 70% in quarters with an IMI precalving. Parenteral antibiotic treatment had no effect on any of these outcomes. In conclusion, use of an internal teat-canal sealant in heifers reduced the postcalving IMI prevalence and the incidence of pathogen-associated clinical mastitis postcalving by decreasing the incidence of new infections over this high-risk peripartum period, and may be a useful tool for reducing the risk of mastitis in heifers.

Prevalence and incidence of subclinical mastitis in goats and dairy ewes in Vermont, USA

The prevalence of bacterial isolation and the somatic cell count (SCC) of the milk from goats ( n=110 from six herds) and sheep ( n=153 from three herds) was determined at parturition and approximately 40 days later. Incidence of new intramammary infection (IMI) and the spontaneous cure rate were determined over this time period. In the goats, 27.3 and 25.5% were infected at parturition and 40 days later, respectively, while 15.0 and 9.1% of sheep were infected at parturition and 40 days later, respectively. Incidence of new infection was 0.039 and 0.034 cases/half/30 days for goats and sheep, respectively, and did not differ between the species ( P>0.2). Spontaneous cure occurred in 93.8% of sheep halves infected at parturition but only 50.0% of goat halves ( P<0.05). Coagulase negative staphylococci (CNS) were the most common isolates from both sheep and goats. Milk from bacteriologically positive halves had a significantly higher somatic cell count than halves from which no bacteria were isolated in both sheep and goats. It was concluded that bacterial infection of the mammary gland was associated with an elevated somatic cell count. Additionally, differences were detected between the species in incidence of new infection and spontaneous cure rate which resulted in a decline in prevalence in sheep, but not goats, with time postpartum.

Effect of localised antibiotic infusions applied to the teat-canal and teat sinus at drying-off on mastitis in the dry-period and at calving.

An experiment using three New Zealand herds and a total of 632 cows, examined the effect of localised prophylactic treatments with antibiotic at drying-off on the incidence of new intramammary infection during the dry period and at calving. Antibiotic was infused either into the teat canal (0.22 g of dry-cow formulation) or the teat sinus (3.1 g of lactating-cow formulation) of uninfected quarters to eliminate any bacteria present in these locations at the last milking of lactation. These treatments were compared with a negative control (nil treatment) and a positive antibiotic control (infusion of 3.6 g of dry-cow formulation). All antibiotic formulations used the same active ingredient, sodium cloxacillin. No significant reduction in new dry period clinical mastitis was observed for the two localised treatments whereas the positive control treatment achieved 100% reduction in new clinical mastitis compared with untreated control quarters. A 41% reduction (P < 0.05) in new Streptococcus uberis infections at calving was associated with the teat canal antibiotic treatment, compared with an 82% reduction (P < 0.001) for the positive antibiotic control. Both localised treatments showed a reduced incidence of new intramammary infection (P < 0.001) when pooled across periods and pathogens. Teats receiving either the teat canal antibiotic treatment or a full infusion of long acting dry-cow antibiotic had a lower incidence of open teat canals (P < 0.05) at 3 weeks after drying-off.

The prophylactic effect of a teat sealer on bovine mastitis during the dry period and the following lactation

Aim. To determine the prophylactic efficacy of a teat sealer, administered at drying off, in reducing new intramammary infections in the dry period and the following lactation. Methods. A total of 528 cows with late lactation somatic cell counts < 200 000 cells/ml was identified in three commercial herds. Of these, bacteriological examination showed 482 cows were uninfected in all four quarters and 46 were infected in only one quarter. At drying off, uninfected quarters were randomly allocated to the following treatments: no infusion (negative controls), infusion with a bismuth subnitrate based teat sealer, infusion with teat sealer + antibiotic, or infusion with a cephalonium-based dry cow antibiotic (positive control). New infections were identified during the dry period by periodic udder palpations and at calving by bacteriological culture. Results. All three infused treatments reduced the incidence of new intramammary infections due to Streptococcus uberis, both during the dry period and at calving, by about 90% (p < 0.01 ). The majority of the infections were due to Streptococcus uberis. For all treatments, a 50% lower incidence of clinical mastitis over the first 5 months of the ensuing lactation was reported by farmers. X-ray imaging of 19 teats showed that the teat sealer material was retained, at least in part, in the lower teat sinus over about 100 days of the dry period. Conclusions. Closure of the teat canal from day one of the dry period as achieved by the teat sealer was as effective in reducing new dry period infections as the infusion of a long-acting dry cow antibiotic formulation. The lower incidence of new infections in the ensuing lactation among the infused quarters implies that fewer subclinical infections persisted from the dry period. Use of teat sealers at drying off appears to offer the same prophylactic efficacy as the dry cow antibiotic approach.
