Highly Efficient AIE-Active palmatine photosensitizer for photodynamic inactivation of Listeria monocytogenes in apple juice preservation

Photodynamic sterilization is a promising alternative to conventional antibacterial approaches with merits of high efficiency and safety. The exploration of aggregation-induced emission (AIE)-type photosensitizers from natural sources is highly valued by researchers and food industry. Herein, the use of palmatine (PA), an aggregation-induced emission-active natural product from traditional Chinese medicine, as photosensitizers for photodynamic inactivation of a tenacious foodborne pathogen of Listeria monocytogenes is investigated. Antibacterial and antibiofilm activity against L. monocytogenes of PA-mediated photodynamic process were first assessed. The results showed that PA-mediated photodynamic process could inhibit the growth of L. monocytogenes, and the minimum bactericidal concentration was determined as 80 μM. At this PA dosing level, well-established biofilms of L. monocytogenes can be effectively destroyed under light irritation. Afterward, cell- and gene-level investigations were conducted to explore the antibacterial mechanism. It is concluded that the exceptional photodynamic antibacterial activity of PA against L. monocytogenes is attributed to the disruption of the cell wall and membrane, increased cell permeability, leakage of functional proteins, and damage to DNA structure. Subsequently, PA-mediated photodynamic process was applied to reduce bacterial activity in apple juice while preserving its quality. Overall, this work highlights the significant potential of PA-mediated photodynamic strategy for controlling foodborne pathogens in food systems.

Effects of Thermally-Assisted and High-Pressure Processing on Background Microbiota and the Listeria monocytogenes Load of a Minimally Processed Commodity.

The current study investigated the impact of treatments with elevated hydrostatic pressure (500 MPa) for inactivation of Listeria monocytogenes on smoked rainbow trout (Oncorhynchus mykiss) at high and low inoculation levels. The temperature values of the trials were set at 4.4 and 60.0 °C, adjusted with a circulating water bath connected to a stainless steel jacket surrounding the pressure processing chamber. Before pressure processing, the counts (selective counts of PALCAM, mean ± SD) of L. monocytogenes were 6.45 ± 0.1 log CFU/g and were reduced (p < 0.05) to 3.72 ± 0.3, and <1.48 ± 0.8 log CFU/g after 10 min of treatment at 4.4 and 60.0 °C, respectively. Treatments of low inoculation level samples were similarly efficacious and resulted in a reduction (p < 0.05) of the pathogen to 1.62 ± 0.3 and <0.82 ± 0.0 log CFU/g for treatments at 4.4 and 60.0 °C, respectively. At 4.4 °C, linear D-value and non-linear kmax1 were 8.68 and 0.50, and 5.81 and 2.41 for high-inoculation and low-inoculation samples, respectively. Application of hydrostatic pressure at 500 MPa at cold and elevated temperatures was efficacious for up to 5.03 log CFU/g reduction of L. monocytogenes, illustrating the potential for further adaptation of this technology.

Inactivation of Listeria monocytogenes in ready-to-consume liquid infant milk treated with Ohmic Heating

Ohmic heating is a promising alternative method to conventional heating for microbial inactivation. This study aimed to inactivate Listeria monocytogenes and assess some quality parameters in ready-to-consume liquid infant milk treated with different voltage gradients of ohmic heating. Different ohmic heating voltage gradients (5, 10, and 20V cm-1) were applied to the samples inoculated with Listeria monocytogenes (ATCC 13932) for 5 minutes. The application of 20 V cm-1 ohmic heating induced inactivation of Listeria monocytogenes at 4 minutes and resulted in approximately 5.34 log reduction; however, there was no significant reduction for the 5 and 10V cm-1 groups. Moreover, 20V cm-1 ohmic heating application did not cause any changes in pH, L*, and b* values. A significant decrease in a* value and an increase in hydroxymethylfurfural value was noted in this group. In conclusion, the effectiveness of ohmic heating in pathogen inactivation depends on the applied electric field intensity and the application time. As a result, the ohmic heating conditions must be carefully determined for the infant milk to inactivate pathogens and ensure public health protection. The results of this study contain significant and beneficial data that can be disposed of the listeriosis risk associated with the consumption of infant milk in infants.

Quantification and modeling of disinfectant inactivation of Listeria monocytogenes biofilms on stainless steel surfaces

The present study evaluates and models the efficacy of three different disinfectants-sodium hypochlorite (SH), peracetic acid (PAA), and chlorine dioxide (ClO2) solution in inactivating biofilm formation of Listeria monocytogenes on stainless steel (SS) surfaces. The nonlinear survival curves of L. monocytogenes biofilm cells treated with different disinfectants were fitted using the Weibull model and four supervised machine learning models (Artificial Neural Networks (ANN), Random Forest (RF), Support Vector Regression (SVR), and Gradient Boosting Decision Tree (GBDT)). RF model exhibited superior predictive ability compared to other methods (RMSE: 0.12, MAE: 0.10, MAPE: 2.38, and R2: 0.99). Both RF and ANN showed better predictive effectiveness than the Weibull model. This research quantifies and models the efficacy of three disinfectants on L. monocytogenes biofilm on SS surfaces, providing valuable insights into the disinfection process of equipment surfaces in food processing.

Anti-Listeria efficacy of peroxyacetic acid in bacterial buffered solution and on bell peppers

This study aims to 1) evaluate the efficacy of peroxyacetic acid (PAA) to inactivate Listeria monocytogenes in 0.1 % buffered-peptone-water; and 2) evaluate the reduction and mitigation of cross-contamination on peppers. In study-1, aliquots of 1.0-ml PAA solutions (133, 265, and 448 ppm) were added to the first 6 wells of 8-strip-deep well microplates. Then, 0.1 ml of serially diluted nalidixic-acid-resistant (NaL) L. monocytogenes was added and mixed immediately with a multichannel pipette. After exposure for 0, 5, 15, 30, and up to 120-s, 1 ml of 2 × D/E neutralized solution was added to terminate the reaction followed by spread-plating onto tryptic soy agar +200 ppm NaL and incubated at 35 °C for 48 h. In study-2, 5 inoculated red peppers were triple-washed with 15 uninoculated green peppers with 0, 23, 53, and 96 ppm of PAA and analyzed using MPN-method. L. monocytogenes counts were <0.3 log10 CFU/ml after exposure to 133, 265, and 448 ppm of PAA for 120, 90 and 60 s, respectively and fit the Linear and Weibull Models. Applying 23–96 ppm of PAA reduced L. monocytogenes by 2.03–3.32 log10MPN/g and transferred cross-contaminated cell counts by 2.36–3.17 log10MPN/g, with no differences (P > 0.05) compared to the water only treatment. Results suggested that applying allowable concentrations (23–96 ppm) of PAA during triple-wash does not show promising anti-Listeria activity on peppers. Future studies are needed to determine the minimum concentrations of the PAA solution required to effectively reduce and mitigate microbial cross-contamination of Listeria monocytogenes on peppers.

Inactivation of Listeria monocytogenes on a laboratory medium and enoki mushrooms using organic acid and essential oil vapors

The aim of this study was to determine the antimicrobial activity of organic acid (OA) and essential oil (EO) vapors against Listeria monocytogenes on a laboratory medium and enoki mushrooms. Among 14 OA and 8 EO vapors tested, 3 OA vapors (acetic, formic, and propionic acid) and 3 EO vapors (cinnamon bark, citronella, and lemongrass) showed relatively strong antilisterial activity. On tryptic soy agar (TSA), all three OA vapors had a minimum inhibitory concentration (MIC) of 12.5 mg/L, while the minimum lethal concentration (MLC) of formic acid vapor was the lowest, at 50 mg/L, followed by acetic acid vapor (200 mg/L) and propionic acid vapor (400 mg/L). The MIC values of lemongrass, cinnamon bark, and citronella EO vapors were 25, 100, and 100 mg/L, respectively. The MLC of lemongrass EO vapor was 100 mg/L and MLCs of other EO vapors were >400 mg/L. Formic acid and lemongrass EO vapors showed a partial synergism on TSA (fractional inhibitory concentration index of 0.7500). For L. monocytogenes inoculated on enoki mushrooms, formic acid vapor at a concentration of ≥50 mg/L, as well as formic acid and lemongrass vapors in combination (1:2 vol ratio) at ≥ 75 mg/L, inactivated L. monocytogenes. These results indicate that formic acid vapor is effective at controlling the growth of L. monocytogenes on enoki mushrooms.

Effects of Fermentation Temperature, Drying Temperature, Caliber Size, Starter Culture, and Sodium Lactate on Listeria monocytogenes Inactivation During Salami Production

The effect of fermentation and drying temperatures, caliber, and sodium lactate on Listeria monocytogenes inactivation was studied in salami, produced in a pilot scale, inoculated with 107 CFU/g of Listeria innocua ATCC® 33090 as a surrogate microorganism for L. monocytogenes. Fermentation temperature varied between 24 and 30°C, drying temperature between 14 and 20°C, caliber between 5.1 and 13.2 cm, and sodium lactate initial concentrations in salamis were 0 and 2%. L. innocua counts, pH and water activity were determined in salamis over time. Sodium lactate (2%) decreased pH drop and Listeria inactivation during fermentation. Baranyi & Roberts equation was used to fit the experimental data and to estimate, for each test condition, inactivation rate (k), initial (Y0), and final counts of L. innocua (YEND). Total inactivation was calculated as Y0 minus YEND (Y0-YEND). Then, using a Box Benkhen experimental design, a quadratic model for k and a two-factor interaction model (2FI) for Y0 − YEND were obtained as functions of fermentation temperature, drying temperature, and caliber size. The models predicted that maximum k and Y0 −YEND, −2.62 ± 0.14 log10 CFU/g/day and 4.5 ± 0.1 log10 CFU/g, respectively, would be obtained fermenting at 30°C and drying at 20°C regardless of caliber. Drying at 14°C allowed Listeria growth until a water activity (aw) of 0.92 was reached. Therefore, if initial Listeria contamination is high (3 log10 CFU/g), drying at low temperatures will compromise product safety.

Pulsed light treatment of water enriched with short‐chain fructooligosaccharides

AbstractShort‐chain fructooligosaccharides (sc‐FOS) are a soluble dietary fiber with a low calorific value and a sweet taste. The beverage industry is leveraging sc‐FOS in a variety of innovative functional drinks. However, sc‐FOS drinks are vulnerable to microbial spoilage due to the presence of free sugars and high water activity. This study explored the potential of pulsed light (PL) technology to pasteurize water enriched with sc‐FOS at 3% and 6% levels of usage. PL treatments were carried out at 1.7, 1.9, and 2.1 kV for 120, 180, and 240 s, corresponding to fluence ranging between 0.28 and 1.03 J/cm2. The microbial inactivation (of Saccharomyces cerevisiae, Escherichia coli, and Listeria monocytogenes), pH, titratable acidity (TA), total soluble solids (TSS), color profile, and sc‐FOS integrity were investigated. A 5‐log10 inactivation in the microbial populations was obtained at 1.9 kV/ 240 s (equivalent to 0.85 J/cm2 fluence). Of the log‐linear and Weibull models evaluated, Weibull model was found to best‐fit the inactivation kinetics. Insignificant effects of PL were observed on pH, TA, TSS, and color. More than 98% sc‐FOS content was retained in all samples after PL flashing. Therefore, PL technology can be explored as an industrial antimicrobial intervention for functional beverages, including water enriched with sc‐FOS.Practical applicationsShort‐chain fructooligosaccharides (sc‐FOS) are a soluble dietary fiber with a low calorific value and a sweet taste. The study establishes that pulsed light (PL) treatment can be an effective nonthermal pasteurization condition for functional water enriched with short‐chain fructooligosaccharides (sc‐FOS). A PL fluence of 0.85 J/cm2 can be employed for the pasteurization of a 3% sc‐FOS drink (considered a source of dietary fiber) and a drink with 6% sc‐FOS (considered a fiber‐enriched drink). PL fluence up to 1.03 J/cm2 has no concerning effect on the total sugars or sc‐FOS content of the water enriched with FOS after PL treatment, thus making it suitable for an industrial application.

Pathogens inactivation and preservation of Pacific white shrimp by atmospheric cold plasma treatment

Atmospheric cold plasma (ACP) efficacy in inactivating Listeria monocytogenes and Vibrio parahaemolyticus was investigated on fresh and frozen shrimp. ACP inactivation efficacy increased with treatment time (5–20 min) and treatment voltages (60–80 kV) due to increased reactive gas species. ACP yielded stronger pathogen inactivation on frozen versus fresh shrimp. ACP treatment at 80 kV for 30 min maximally reduced L. monocytogenes and V. parahaemolyticus by 0.94 and 1.26 log CFU/g on fresh shrimp and by 1.68 and 2.0 log CFU/g on frozen shrimp. During storage at 4 °C, ACP-treated shrimp exhibited slightly higher thiobarbituric acid reactive substances (TBARS) levels but overall improved quality attributes, including lower total viable count, total volatile basic nitrogen, and pH, as well as higher sensory scores compared to the controls. ACP also extended shrimp shelf-life by 2 days. Therefore, ACP holds significant potential for seafood disinfection and preservation. Industrial relevanceACP effectively reduced levels of Listeria monocytogenes and Vibrio parahaemolyticus on both fresh and frozen shrimp with greater inactivation effects on frozen shrimp. In addition to enhancing microbial safety, ACP also improved shrimp quality and extended shelf-life. ACP could readily be coupled with packaging technology and bio preservatives to enhance its efficacy. This research provides a foundation for the eventual industrial adoption of ACP technology to benefit both consumers through reduced pathogen risks, and seafood processors through improved product safety, quality, and shelf-life.

Ohmic heating application with different electric fields on inactivation of Listeria monocytogenes in protein-enriched cow milk.

The aim of this study was to determine the effects of an ohmic heating (OH) process with different electric field intensities on Listeria monocytogenes inactivation in protein-enriched cow milk. Protein powder was added at rates of 2.5%, 5% and 7.5% in 1.5% fat content milk, and L. monocytogenes (ATCC 13932) strain was then inoculated into the samples. The OH process was carried out in a laboratory-type pilot unit created using stainless steel electrodes, a K-type thermocouple, a datalogger and power supply providing AC current at 0-250 V, 10 A. The inoculated milk samples were heated to 63°C by applying an electric field intensity of 10V/cm and 20V/cm. L. monocytogenes counts, pH, color measurement and hydroxymethylfurfurol levels were then determined. OH applied with an electric field intensity of 10 V/cm caused an average decrease of 5 logs in L. monocytogenes level in the samples containing 2.5% protein and decreased below the detection limit (<1 log) at the 9th minute (p<0.05). Similarly, application of an electric field intensity of 20 V/cm in milk containing 2.5% and 5% protein caused the L.monocytogenes level to decrease below the detection limit (<1 log) at 2 minutes 30 seconds (p<0.05). No change was observed in the L* (brightness) values of the samples but it was determined that there was a slight increase in pH, a* (redness) and b* (yellowness) values compared to the control group. It was observed that the inactivation of L. monocytogenes by OH depends on the duration of the OH process, protein concentration in the milk and the applied voltage gradient.

Effect of Air Temperature and Velocity on Listeria monocytogenes Inactivation During Drying of Apple Slices

A wide range of drying parameters and methods are used by industry to produce dried apples. To ensure end-product safety and regulatory compliance, it is essential to evaluate the effectiveness of such industrial practices on microbial inactivation. Therefore, the objective of this study was to evaluate the effects of drying air temperature and velocity on Listeria monocytogenes inactivation during drying of apple slices. Apples (cv. Gala) were cored, sliced as rings (∼6 mm thick), and surface-inoculated with broth-grown culture of an 8-strain cocktail of L. monocytogenes to achieve an inoculation level of 8.6 ± 0.3 log CFU/g. Apple rings were dried in batches using dry air in a pilot-scale impingement oven at 60 or 80 °C air temperature and 0.7 or 2.1 m/s air velocity, and sampled every 30 min for bacterial enumeration, water activity (aw), and moisture content analysis. L. monocytogenes reduction increased (P < 0.05) with higher air velocity or higher drying air temperature. By the end of drying, in which the standard moisture content for dried apple slices of <24% wet basis was reached, L. monocytogenes was reduced by 1.8 ± 0.3 and 2.8 ± 0.7 log CFU/g at 0.7 and 2.1 m/s air velocity, respectively, after 180 min at 60 °C. When using 80 °C drying temperature, L. monocytogenes reduction was 5.2 ± 0.5 log CFU/g at both air velocities after 150 min. Therefore, process conditions should be considered in the validation of fruit drying processes, instead of solely relying on product endpoint properties, such as moisture content.

Transfer and Inactivation of Listeria monocytogenes during Pilot-Scale Dicing and Flume Washing of Onions

This study assessed the extent of L. monocytogenes transfer from onions to the surface of a commercial dicer, from inoculated onions to uninoculated onions, and the efficacy of various sanitizers during the subsequent flume washing of diced onions. Spanish yellow onions (Allium cepa L.) were dip-inoculated in a 3-strain avirulent L. monocytogenes cocktail (5.9 or 4.2 log CFU/50 g) and air-dried. After dicing one 2.2 kg batch of onions inoculated at ~5.9 log CFU/50 g followed by ten uninoculated batches of 2.2 kg each, L. monocytogenes progressively decreased from 4.6 to 2.6 log CFU/50 g in baches 1 through 10, respectively. After onions inoculated at ~4.0 log CFU/g were diced and flume washed for 2 min in tap water, electrolyzed water containing 55 ppm free chlorine, 80 ppm free chlorine from a commercial sanitizer, or 80 ppm peroxyacetic acid and dewatered on a mechanical shaker table, L. monocytogenes populations decreased 0.4, 0.3, 1.4, and 1.0 log, respectively, with populations of ~1.2 log CFU/mL in water for all three sanitizers. These findings should be useful in future risk assessments and aid in the development of improved industry guidelines to better enhance the safety of diced onions.

Photodynamic inactivation of Listeria monocytogenes using a natural aggregation-induced emission photosensitizer and its application in salmon preservation

Photodynamic sterilization has been regarded as a promising alternative to conventional antibacterial approaches. However, the photodynamic efficiency of conventional photosensitizer is often compromised by aggregation-induced quenching effects. Photosensitizers, especially natural products, with aggregation-induced emission (AIE) properties open a new avenue for photodynamic sterilization. Herein, we investigated the photodynamic antibacterial performance and mechanism of the AIE-type natural product of berberine (BBR) against Listeria monocytogenes (L. monocytogenes). The bactericidal effect was assessed through the determination of minimum bactericidal concentration and implementation of anti-biofilm experiments. The results showed that BBR can significantly suppress the proliferation of L. monocytogenes and can efficiently remove the L. monocytogenes-derived biofilm in a concentration-dependent trend. The mechanism of killing L. monocytogenes has been speculated based on bacterial morphology characterization, viability staining results, enzyme activity assays, DNA electrophoresis analysis, and molecular docking. The excellent anti-bacterial activity of BBR mediated photodynamic process may account for its abilities to the irreversibly disruption of bacterial membrane, inhibition of intracellular enzyme activity and decomposition of intracellular DNA molecules. We also demonstrated that BBR embedded sodium alginate/chitosan packaging film is feasible to maintain the freshness of salmon under light illumination by inhibiting the bacterial proliferation.

Inactivation of Listeria monocytogenes and Salmonella spp. During Cooking of Country Ham and Fate of L. monocytogenes and Staphylococcus aureus During Storage of Country Ham Slices

Thermal inactivation studies were undertaken on Listeria monocytogenes and Salmonella spp. inoculated on the surface of country ham. Hams (average = ca. 3.4 ± 0.5 kg each; average = ca. ≥18% shrinkage) were used as provided by the processor (i.e., “salted hams”), desalted in tap water (i.e., “desalted hams”), or dried for an additional period (i.e., “extra-dried hams”). Hams were surface inoculated (ca. 9.5 log CFU/ham) with a multistrain cocktail of L. monocytogenes or Salmonella spp. and cooked within a bag ina circulating water bath to an internal temperature of 130°F (54.4°C) instantaneous, 145°F (62.8°C) and held for 4 min, 153°F (67.2°C) and held for 34 s, or 160°F (71.1°C) instantaneous. Regardless of ham type, all four time and temperature combinations tested herein delivered a ≥6.7-log reduction of cells of L. monocytogenes or Salmonella spp. Differences in product pH, moisture content, or aw did not have an appreciable impact on the thermal inactivation of L. monocytogenes or Salmonella spp. on country ham. In addition, shelf-life studies were undertaken using slices of “salted” country ham that were surface inoculated (ca. 5.5 log CFU/slice) with a multistrain cocktail of L. monocytogenes or Staphylococcus aureus and then stored at 20°C. Levels of S. aureus increased by ca. ≤1.4 log CFU/slice during storage for 90 days, whereas levels of L. monocytogenes remained relatively unchanged (≤0.2 log CFU/slice increase). Our data validated that cooking parameters elaborated in the U.S. Department of Agriculture’s Food Safety and Inspection Service Cooking Guideline for Meat and Poultry Products (Revised Appendix A) are sufficient to deliver significant reductions (ca. ≥6.8 log CFU/ham) in levels of L.monocytogenes and Salmonella spp. on country ham. In addition, in the event of postprocessing contamination, country ham may support the outgrowth of S. aureus or survival of L. monocytogenes during storage at 20°C for 90 days.

Inactivation of Listeria monocytogenes by Hydrogen Peroxide Addition in Commercial Cheese Brines

Commercial cheese brines are used repeatedly over extended periods, potentially for years, and can be a reservoir for salt-tolerant pathogens, such as Listeria monocytogenes. The objective of this study was to determine the inactivation of L. monocytogenes in cheese brines treated with hydrogen peroxide (H2O2) (0, 50, and 100 ppm) at holding temperatures representing manufacturing conditions. In experiment one, four fresh cheese brines were prepared with 10 or 20% salt and pH 4.6 or 5.4 (2x2 design; duplicate trials). Brines were inoculated with L. monocytogenes, treated with H2O2, and stored at 10 and 15.6°C. For experiment two, seven used commercial brines (representing five cheese types, 15–30% NaCl, pH 4.5–5.5; three seasonal trials) were inoculated with L. monocytogenes or S. aureus, treated with H2O2, and stored at 12.8°C (both L. monocytogenes and S. aureus), 7.2 and 0°C (L. monocytogenes only). Each treatment was assayed on Days 0, 1, and 7 for microbial populations and residual H2O2. Data revealed that pathogen populations decreased ≤1 log in cheese brines with no hydrogen peroxide stored for 7 days, regardless of the storage temperature. In fresh brine treated with 50 or 100 ppm of H2O2, populations of L. monocytogenes were reduced to less than the detectable limit by 7 days at 10 and 15.6°C (>4 log reduction). For unfiltered used brines, H2O2 had no effect on L. monocytogenes populations in Brick J (pH 5.4, 15% NaCl) due to rapid inactivation of H2O2, likely by indigenous yeasts (∼3-log CFU/ml). For the remaining brines, the addition of 100 ppm H2O2 killed >4 log L. monocytogenes when stored at 7.2 or 12.8°C for 1 week, but only 3–4 log reduction when stored at 0°C. The addition of 50 ppm H2O2 had similar lethal effects at 12.8°C but was less effective at 7.2 or 0°C. Inactivation rates of S. aureus were similar to that of L. monocytogenes. This study confirmed that high salt, warmer temperature, and 100-ppm H2O2 accelerated the inactivation of L. monocytogenes in cheese brines. Data also suggest that the presence of catalase-positive indigenous microorganisms may neutralize the effect of H2O2.

Effect of dehydration on the inactivation of Listeria monocytogenes and Salmonella enterica on enoki and wood ear mushrooms.

Foodborne illness outbreaks in the U.S. associated with consumption of both fresh and dried specialty mushrooms have recently occurred. Dried wood ear mushrooms were implicated in a salmonellosis outbreak in 2020, while fresh enoki mushrooms were associated with two listeriosis outbreaks in 2020 and 2023. These specialty mushrooms are commercially available in both their fresh and dried states. Due to the short shelf life of mushrooms, dehydration is a common method used in both industry and by consumers to extend the shelf life and preserve quality. Therefore, the aim of this study was to evaluate the use of dehydration on the inactivation kinetics of both Listeria monocytogenes and Salmonella enterica on enoki and wood ear mushrooms. Fresh mushrooms were inoculated with four strain cocktails of either L. monocytogenes or S. enterica and dried at ambient conditions for 10 min. Following drying of the inoculum, mushrooms were placed into food dehydrators preheated to 70, 80, or 90°C and treated for up to 24 h. At treatment intervals, mushrooms were removed from the dehydrators for pathogen enumeration. Inactivation kinetics for both pathogens were modeled using the Weibull, log-linear with tail, and log-linear with shoulder models. Pathogen reductions of >4 log CFU/g were achieved on both enoki and wood ear mushrooms during dehydration at 90°C after only 2-4 h. At 70 and 80°C, log reductions of >4 log CFU/g were observed on wood ear mushrooms after 4-8 h. On enoki mushrooms, a tailing effect was observed with residual populations (>2 log CFU/g) of L. monocytogenes and S. enterica remaining even after 24 h of treatment at both 70 and 80°C. This study emphasizes the need for an individualized dehydration strategy for each mushroom type to ensure the effectiveness of dehydration as a process to reduce pathogen populations. Results of this study will aid in informing proper time and temperature combinations for dehydration of specialty mushrooms to ensure product safety.

Synergistic effect of electrolyzed water generated by sodium chloride combined with dimethyl dicarbonate for inactivation of Listeria monocytogenes on lettuce.

Electrolyzed water (EW) is recognized as an effective way to control and reduce pathogens in vegetables. However, the disinfection efficacy of EW alone is limited. In this work, the bactericidal activity and biofilm removal capability of EW, generated by adding NaCl to a portable EW generator, were investigated with special reference to Listeria monocytogenes. Furthermore, the impact of EW in combination with dimethyl dicarbonate (DMDC) in reducing the microbial load and improving the overall quality of lettuce during refrigerated storage was evaluated. EW with 0.3% NaCl (SEW) had the highest bactericidal activity against L. monocytogenes. The pathogen treated with SEW exhibited lower superoxide dismutase activity and more leakage of proteins and nucleic acids than in the case of EW. Furthermore, the use of SEW resulted in changes in the cell permeability and morphology of L. monocytogenes. A decrease in adhesion and collapse of the biofilm architecture were also observed, indicating that SEW was more effective for inactivating L. monocytogenes cells compared to EW. For untreated lettuce, the populations of the total plate count and inoculated L. monocytogenes decreased by 2.47 and 2.35 log CFU g-1 , respectively, after the combined SEW/DMDC treatment for 3 min. The use of SEW alone or combined with DMDC did not negatively impact the lettuce color values, titratable acid, ascorbic acid and soluble solids compared to the control group. SEW in combination with DMDC can be used as a novel and potentially effective disinfection strategy for ensuring the safety of vegetable consumption. © 2023 Society of Chemical Industry.

Evaluation of pulsed xenon UV irradiation on inactivation of Listeria monocytogenes on stainless-steel surfaces

UV-C disinfection is a safe and effective method for the inactivation of bacteria, viruses, spores, and other microorganisms. Pulsed xenon UV lamps (PX-UV) can provide high intensity UV-C irradiation in a short time. To improve our understanding of which factors affect the disinfection efficacy of PX-UV lamps, a rigorous investigation of the effects of exposure time, frequency, UV fluence, and angle of irradiance on log reduction of Listeria monocytogenes on stainless steel surfaces was characterized. Total UV fluence received proved to be the main determinant of log reduction regardless of pulse frequency, angle, or exposure time. However, 30 s of exposure to UV light at 25 Hz resulted in a 5.59-log reduction which corresponds to a fluence of 151.2 mJ/cm2. Kinetic constants of UV inactivation was determined to allow end users to calculate the required fluence needed to achieve the desired log-reduction. This work confirms that UV-C fluence should be the primary consideration when deploying PX-UV lamps as a no-touch disinfection technology and that operational parameters should be selected to achieve the desired fluence.

Determination of the effect of milk fat on the inactivation of Listeria monocytogenes by ohmic heating

Research in recent years has focused on innovative technologies that provide pathogen inactivation without damaging the structural properties of foods. Ohmic heating (OH) is an innovative technology, that provides an effective microbial inactivation with massive and rapid heating. This study aims to determine the effects of milk fat on the inactivation of Listeria monocytogenes by OH with a low voltage gradient. L. monocytogenes (ATCC 13932) inoculated 3.1%, 1.5%, and 0.1% fat-milk samples were heated up to 62°C by OH and conventional heating (CH) process. OH treatment lead to the inactivation of L. monocytogenes in both 1.5% and 0.1% groups and led to approximately 5.30 log decrease, however, there was a 3.10 log decrease in the 3.1% group at 6 min. CH lead to a few reduction as 0.21, 0.29 and 0.39 log in 3.1%, 1.5% and 0.1% fat-milk respectively. In OH, the sublethal injury ratio was higher than CH in all milk groups. However, OH did not statistically change color and pH values at the 6th min of the process, had a significant effect on hydroxymethylfurfural value only in 3.1% fat-milk. In conclusion, the increased fat content may have important inhibitory effects on pathogen inactivation in OH. Thus, the OH conditions should be chosen carefully to sufficient inactivation of pathogens in milk with high-fat content.

Insights into inactivation and response mechanisms of sublethal Listeria monocytogenes treated by cold plasma with joint transcriptomics and metabolomics

The aim of the current study is to elucidate the inactivation and molecular response pattern of sublethal Listeria monocytogenes to cold plasma mediated two-pronged oxidative microenvironments from a high-throughput multi-omics perspective. First joint transcriptomics and metabolomics analyses revealed that significantly expressed genes and metabolites were mainly involved in enhanced transmembrane transport and Fe2+/Cu+ efflux, amino acids limitation, cytoplasmic pH homeostasis, reconfiguration of central carbon metabolism flux and energy conservation strategy, which triggered the surge of intracellular endogenous oxidative stress and finally mediated bacterial ferroptosis and pathogenicity attenuation. Typical antioxidant systems such as the TrxR-Trx system and common antioxidant genes (e.g. sodA, katA, ahpC, trxA, spxA) were inhibited, and the more prominent antioxidant pathways include methionine metabolism, pentose phosphate pathway and glutathione metabolism as well as the DNA repair systems. Therefore, our work confirmed from the transcriptional and metabolic as well as physiological levels that cold plasma-mediated intracellular oxidative stress induced big perturbations in pathways as a driving force for the inactivation and pathogenicity attenuation of L. monocytogenes.
