Organic acid preservation of cereal grains improves grain quality, growth performance, and intestinal health of post-weaned pigs

This study investigated the effect of preserving wheat and barley grains with an organic acid liquid surfactant mould inhibitor compared to conventional artificial drying and assessed its effects on the health and performance of pigs post-weaning (PW). A 2 × 2 factorial arrangement was conducted to investigate the interaction between grain preservation method (dried vs. preserved) and zinc oxide (ZnO) inclusion (yes vs. no) on growth performance, diarrhoea scores and incidence, and total tract nutrient digestibility. One hundred and ninety-two pigs (3 pigs/pen; 16 replicates/treatment) were assigned to one of four experimental diets for 35 days: (1) dried grain diet, (2) preserved grain diet, (3) dried grain diet with ZnO and (4) preserved grain diet with ZnO. Diets were formulated to contain similar levels of net energy and standardised ileal digestible lysine. On day 35 PW, 24 pigs (12 replicates/treatment) from the non-ZnO supplemented groups were euthanised and digesta was collected for coefficient of apparent ileal digestibility (CAID), gut microbial population and volatile fatty acid analysis. Stomach tissue was also collected for gene expression analysis, as well as small intestine samples for gut morphology. Notable improvements in grain quality were observed in the preserved grain, including a reduction in grain pH, mould presence, and contamination by mycotoxins, specifically deoxynivalenol in wheat and barley, and ochratoxin A and HT-2 toxin in barley. The preserved grain diet improved average daily gain (ADG; P < 0.01) and body weight (BW; P < 0.01) compared to the dried grain diet. The incorporation of ZnO increased average daily feed intake (ADFI; P < 0.01) and BW (P < 0.05) in the dried grain diet, however, ZnO did not affect ADFI (P > 0.05), and reduced ADG and BW in the preserved grain diet (P < 0.05). Pigs offered the preserved grain diet had reduced faecal scores and diarrhoea incidence compared to those offered the dried grain diet (P < 0.05). Supplementation of ZnO reduced faecal scores and diarrhoea incidence compared to non-supplemented pigs (P < 0.001). On day 35 PW, the preserved grain diet had improved CAID of nitrogen and gross energy (P < 0.05), lower levels of colonic branched-chain volatile fatty acids (P < 0.05), and beneficial shifts in gut microbial populations. Specifically, there was a reduction in ileal Streptococcus (P < 0.001) and an increased abundance of colonic Faecalibacterium (P < 0.05). In conclusion, organic acid preservation improved grain quality, benefiting post-weaned pigs through improved nutrient digestibility and gut health, thus enhancing overall growth performance PW.

Effect of dietary supplementation of two fiber sources differing on fermentability and hydration capacity on performance, nutrient digestibility and cecal fermentation in broilers from 1 to 42 d of age

A total of 378 Cobb-500 male broilers were used to evaluate the effects of 2 fiber sources, differing in hydration capacity and fermentability, on gastrointestinal tract development, apparent ileal digestibility and performance from 1 to 42d of age. There were 9 replicates per each of the 3 dietary treatments, all in mash form: a wheat-soybean control (CON) diet, CON diet diluted with 1.5% of wood lignocellulose (LC diet) as a non-fermentable insoluble fiber with high hydration capacity; and CON diluted with 1.5% of a mixture of fibers (ISFC diet) containing both lignified insoluble fiber and a prebiotic soluble fiber fraction from fructooligosaccharides. Additionally, the fermentability of both fiber sources (LC and ISFC) was determined by in vitro using cecal inoculum from broilers fed the experimental diets. Both LC and ISFC treatments impaired by 4% feed conversion ratio only during the first 7d (P = 0.003) compared with CON group. In the grower period (21-42d), the ISFC group showed the best growth (P = 0.039), and at 42d tended to show the highest body weight (P = 0.095). This agrees well with the highest ileal dry matter (P = 0.033) and organic matter (P = 0.043) digestibility observed in ISFC group and the similar trend observed for ileal protein digestibility (P = 0.099) at 42d. Also, at 42 d, absolute and relative (% body weight) digestive tract weights (P ≤ 0.041) and empty gizzard weights (P ≤ 0.034) were greater for LC and ISFC groups compared to CON. The cecal molar proportion of valeratewas greatest in ISFC group (P = 0.039). In vitro gas production was higher for ISFC than for LC substrate when using either a diet-adapted or non-adapted cecal inoculum (P < 0.05). These results show the interest in combining IF with prebiotic highly fermentable fiber, such as fructooligosaccharides, in broilers to improve nutrient digestibility and finishing performance.

Impact of dietary Laminaria digitata with alginate lyase or carbohydrase mixture on nutrient digestibility and gut health of weaned piglets

Laminaria digitata is a brown seaweed rich in prebiotic polysaccharides, mainly laminarin, but its alginate-rich cell wall could compromise nutrient access. Carbohydrase supplementation, such as individual alginate lyase and carbohydrases mixture (Rovabio® Excel AP), could enhance nutrient digestibility and prebiotic potential. This study aimed to evaluate the effect of these enzymes on nutrient digestibility and gut health of weaned piglets fed with 10% L. digitata. Diets did not affect growth performance (P > 0.05). The majority of the feed fractions had similar digestibility across all diets, but the supplementation of alginate lyase increased hemicellulose digestibility by 3.3% compared to the control group (P = 0.047). Additionally, we observed that algal zinc was more readily available compared to the control group, even without enzymatic supplementation (P < 0.001). However, the increased digestibility of some minerals, such as potassium, raises concerns about potential mineral imbalance. Seaweed groups had a higher abundance of beneficial bacteria in colon contents, such as Prevotella, Oscillospira and Catenisphaera. Furthermore, the addition of alginate lyase led to a lower pH in the colon (P < 0.001) and caecum (P < 0.001) of piglets, which is possibly a result of released fermentable laminarin, and is consistent with the higher proportion of butyric acid found in these intestinal compartments. L. digitata is a putative supplement to enhance piglet gut health due to its prebiotic polysaccharides. Alginate lyase supplementation further improves nutrient digestibility and prebiotic potential. These results suggest the potential use of L. digitata and these enzymatic supplements in commercial piglet-feeding practices.

PSI-14 Effect of dietary xylanase supplementation on growth performance, nutrient digestibility, and digesta viscosity of weaned pigs fed wheat-soybean meal-based diets

Abstract Xylanase supplementation can improve nutrient digestibility in pig diets by degrading cell wall non-starch polysaccharides and reducing digesta viscosity as well as promoting intestinal health of pigs after weaning. This study determined the effects of dietary xylanase supplementation on growth performance, apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of nutrients, and digesta viscosity in different digestive sites of weaned pigs. A total of 312 weaned pigs [body weight (BW) = 5.1 ± 0.9 kg] were weaned into 48 pens and assigned to one of six dietary treatments, each treatment with 8 pens, 6 to 7 pigs per pen, and sex ratios maintained within BW blocks. Experimental diets were formulated in a 3-phase nursery feeding program: Phase 1 (d 0 - d 7), Phase 2 (d 8 - d 21), and Phase 3 (d 22 – d 42). Experimental diets consisted of a wheat/soybean meal-based diet formulated to meet pig requirements (positive control, PC), the PC diet with reduction of 100 kcal of ME (negative control, NC), and the NC diet supplemented with either 900, 1,800, 3,600, or 7,200 units of xylanase. Feed disappearance and body weight were measured at d 7, 14, 21, and 42. Pen fecal score was assessed daily during d 0 to 14 and three times a week during d 15 to 28. On d 21 to 24 of the experiment (12 pigs per day), one pig per pen was selected for sample collection: ileal, cecal, and mid-colon digesta for viscosity and ileal digesta and feces for nutrient digestibility. Orthogonal polynomial contrasts were used to determine the linear and quadratic effects of increasing levels of xylanase in NC diets. Contrast analysis was used to compare PC and NC groups with xylanase-supplemented groups. Fecal score data were analyzed using the PROC FREQ procedure. Supplementing xylanase to NC diets did not improve pig growth performance (P &amp;gt; 0.05) through the nursery period. In wk 2 and wk 3, pigs fed the PC and NC diets had a greater (χ2 &amp;lt; 0.05) incidence of fecal scores 3 &amp; 4 (diarrhea) than the xylanase-supplemented pigs. The AID of DM and ATTD of crude protein, gross energy, and NDF were greater (P &amp;lt; 0.05) in the PC and 900-xylanase supplemented diets than in the other dietary treatments. In addition, the ATTD of NDF and ADF increased linearly (P &amp;lt; 0.1) in response to xylanase supplementation. Xylanase supplementation (900 to 7,200 U) decreased digesta viscosity in colon when compared with PC and NC diets. Overall, xylanase supplementation can improve nutrient digestibility, particularly at total tract level, and reduce viscosity in the hindgut, which could be related with decreasing the occurrence of looseness. However, the effects on growth performance were not expressive when xylanase was supplemented to wheat-soybean meal-based diets with reduction of 100 kcal of ME.

79 Evaluating the effects of increasing levels of sodium diformate on nursery pig performance

Abstract Organic acids, such as formic acid, are commonly used as acidifiers in swine diets. Forms of formic acid include salts of sodium, calcium, or potassium. Previous research has shown positive results using potassium diformate in swine diets including improved nutrient digestibility, growth performance, and altered gut microbiota. Additionally, recent data has shown increasing levels of sodium diformate improved growth performance in finishing pigs; however, additional data are needed in nursery pigs. The objective of this study was to investigate the effects of increasing sodium diformate in nursery diets on growth performance. Weanling barrows [n = 360; DNA 200 × 400; initial body weight (BW) = 5.9 ± 0.06 kg] were used in a 38-d study. At weaning, pigs were randomly assigned to pens with 5 pigs per pen. Each pen was randomly allocated to 1 of 6 treatments in a completely randomized design with 12 replicates per treatment. Dietary treatments were formulated to provide none, 0.40, 0.60, 0.80, 1.00, or 1.20% sodium diformate (Formi NDF, ADDCON, Nordic AS, Porsgrunn, Norway) at the expense of corn. Experimental diets were fed in 3 phases: phase 1 from weaning to d 9, phase 2 from d 9 to 24, and phase 3 from d 24 to 38. Feces was collected on d 9 and 24 from 3 pigs per pen to determine fecal dry matter (DM). All data were analyzed using a linear mixed model with treatment as a fixed effect and barn as a random intercept. Contrast statements were used to evaluate the linear and quadratic dose response to increasing sodium diformate inclusion. All statistical models were fit using the GLIMMIX procedure of SAS version 9.4 (Cary, NC). From d 0 to 24 (phase 1 and 2), gain to feed ratio (G:F) increased (linear, P = 0.001) as sodium diformate increased in the diet (Table). However, sodium diformate did not affect (P &amp;gt; 0.10) average daily gain (ADG) or average daily feed intake (ADFI). From d 24 to 38 (phase 3), there was no evidence of differences (P &amp;gt; 0.10) due to increasing sodium diformate for any growth response criteria. For the overall experimental period (d 0 to 38), increasing sodium diformate did not affect (P &amp;gt; 0.10) ADG, ADFI, or G:F. Overall mortality was 0.8% and was not influenced (P &amp;gt; 0.10) by treatment. There was no evidence for differences (P &amp;gt; 0.10) in fecal DM on d 9. However, fecal DM decreased (linear, P &amp;lt; 0.05; quadratic, P = 0.097) as sodium diformate increased in the diet on d 24 with pigs fed 0.40% sodium diformate having the greatest fecal DM. In conclusion, these data suggest increasing levels of sodium diformate has the potential to improve feed efficiency in the early nursey period but did not affect performance in the late nursery.

Influence of Jerusalem artichoke tuber as a prebiotic on the nutrient utilization, weaning stress indicators and immunocompetence of crossbred calves during the pre- and post-weaning phases

This study explored the potential prebiotic properties of pulverized Jerusalem artichoke tuber (JAP) in pre- and post-weaned calves. Accordingly, 24 fifteen-day-old calves were randomly assigned to four groups (n = 6). The groups received JAP supplementation at levels of 0 (CON), 1 (JAP1), 2 (JAP2), and 3 (JAP3) percent of dry matter intake (DMI) during the first 60 days of the 120-day experiment. Two 6-days digestion trials were conducted: one during the 4th week of the experiment (pre-wean), and the second during the tenth week (post-wean). Additionally, serum concentrations of cortisol and epinephrine were measured in both the pre- and post-weaning phases to assess the stress levels. Immunocompetence of the calves during both the stages was evaluated through lymphocyte proliferation response. In the pre-weaning phase, the calves supplemented with JAP had better (P < 0.05) protein intake and digestion compared to the control group. In the post-weaning phase, the JAP supplemented groups exhibited significantly (P < 0.05) improved digestibility coefficients for dry matter, organic matter, crude protein, ether extract, and neutral detergent fibre. Although cortisol levels didn't show significant differences (P > 0.05) between the groups during both pre-weaning and post-weaning stages, the post-weaning stage did reveal a significant reduction (P < 0.05) in epinephrine levels in JAP1 compared to the other groups. Additionally, JAP supplementation enhanced (P < 0.05) the calves' response to lymphocyte proliferation during the post-weaning phase. Considering the improved nutrient digestibility, enhanced systemic immunity, and reduced epinephrine levels observed in the JAP supplemented groups, our findings suggest that JAP may have promising potential as a prebiotic for calves.

Exploring the cause of reduced production responses to feeding corn dried distillers grains in lactating dairy cows

An experiment was conducted to identify the factors that cause reduced production of cows fed a diet with high content of corn distillers grains with solubles (DDGS). We hypothesized that the factors could be high sulfur (S) content in DDGS, which may directly (S toxicity) or indirectly (DCAD) cause reduced production. We also hypothesized that high PUFA in DDGS could be another major factor. In a randomized complete block design, 60 lactating cows (15 primiparous and 45 multiparous; average ± SD at the beginning of the trial: milk yield, 44.0 ± 6.9 kg/d; DIM, 123 ± 50; BW, 672 ± 82 kg) were blocked, and cows in each block were randomly assigned to 1 of the following treatments: SBM (4.7% fatty acids [FA], 0.22% S, and 178 mEq/kg DM of DCAD), a diet containing soybean meal as the main protein source; DG, with SBM replacing mainly soybean byproducts and supplemental fat with distillers grains at 30% dietary DM (4.7% FA, 0.44% S, and 42 mEq/kg DM of DCAD); SBM+S, SBM with sodium bisulfate for additional dietary S (4.8% FA, 0.37% S, and 198 mEq/kg DM of DCAD); SBM+CO, SBM with corn oil (4.7% FA, 0.23%, and 165 mEq/kg DM of DCAD); and DG+DCAD, DG with increased DCAD (4.7% FA, 0.40% S, and 330 mEq/kg DM of DCAD). Due to the limited number of tiestalls, blocks 1 to 6 started the experiment first as phase 1, and the rest of the blocks, as phase 2, started the experiment after phase 1. All cows were fed the SBM diet for 10 d as a covariate period followed by the experimental period for 35 d. Data were analyzed using PROC MIXED of SAS (Version 9.4, SAS Institute Inc.); block and phase were random effects; and treatments, repeated week, and interaction were fixed effects. We found an interaction of week by treatment for DMI. Although milk yield did not change, milk fat concentration tended to decrease (2.78% vs. 3.34%) for DG compared with SBM. Dry matter, OM, NDF, and CP digestibilities were lower when cows were fed the DG diet compared with SBM. Additionally, cows fed DG had lower blood concentrations of HCO3-, base excess, and total (t)CO2 compared with SBM. The SBM+S diet did not affect production, nutrient digestibility, or blood parameters compared with SBM. The SBM+CO diet decreased milk fat concentration and yield compared with SBM. The DG+DCAD diet tended to increase milk fat yield and concentration (1.24 vs. 1.47 kg/d; 2.78% vs. 3.37%) and increased ECM (40.9 vs. 45.1 kg/d) compared with DG but did not improve nutrient digestibility. However, blood HCO3-, base excess, and tCO2 were greater for DG+DCAD compared with DG. In conclusion, the indirect role of S-, altering DCAD, along with the high PUFA content in DDGS, appear to be the factors causing reduced production responses to a high DDGS diet. Increasing DCAD to 300 mEq/kg DM in a high DDGS diet can be a feeding strategy to alleviate reduced production responses.

The evaluation of next-generation probiotics on broiler growth performance, gut morphology, gut microbiome, nutrient digestibility, in addition to enzyme production of Bacillus spp. in vitro

Considerable research has been conducted into the efficacy of individual probiotics in broiler production, however information on the most effective combinations of synergistic Bacillus probiotic is lacking. This study investigated the impact of different Bacillus strain combinations in broiler chickens, as well as in vitro enzyme production. In experiment one, a total of 576 Ross 308 broilers at 1 d old were grown for 21 d across 6 treatments of maize-soybean diets (n = 12 pens per treatment) to compare three different strain combinations (formulation 1 [F1]: 3 strains Bacillus amyloliquefaciens; F2: Bacillus coagulans and 2 strains B. amyloliquefaciens; F3: B. coagulans, Bacillus licheniformis and 2 strains B. amyloliquefaciens; F5: Bacillus subtilis, B. licheniformis and 2 strains B. amyloliquefaciens), positive control (PC), and a negative control antibiotic treatment group (NC). In Exp. 2, a total of 360 one-day-old ROSS308 broilers were used to test five treatments (n = 9) including PC, NC, F1 and F5 (selected from Exp. 1), and F4 (Bacillus pumilis and 2 strains B. amyloliquefaciens) in a maize-soybean diet. B. amyloliquefaciens F1 demonstrated a significant improvement in feed conversion ratio (FCR) compared to F2 at d 14 (1.49 vs 2.10; P = 0.038) and the body weight (BW) at d 21 (847.0 g vs 787.4 g) compared to other combinations (P = 0.027). The FCR at d 21 tended to be lower in birds fed F1 (1.46 vs 1.66) compared to the control (P = 0.068). Probiotic treatments had significantly improved nutrient digestibility compared to the PC and NC. Also, probiotic treatments supported the growth of Streptococcus, a common commensal genus and reduced the abundance of genera that correlated with low weight gain such as Akkermansia. Experiment two revealed that F4 improved FCR (P < 0.001) and BW at 28 d (P = 0.014). In vitro testing showed a high production of protease and amylase by Bacillus. Thus, the addition of Bacillus probiotics, particularly containing B. amyloliquefaciens strains and Bacillus pumilus, into the diet of broiler chickens improves production performance, nutrient digestibility, and allows the proliferation of beneficial gut microbiota.

Using probiotics to improve nutrient digestibility and gut-health of weaned pigs: a comparison of maternal and nursery supplementation strategies.

Maternal probiotic supplementation has been found to have a positive impact on the gut health of piglets, not only during the lactation period, but also after weaning. Providing probiotics to nursery pigs is also a common strategy for supplementation. The goal of this study was to evaluate which would be the most effective strategy to improve nutrient digestibility, energy metabolism, and intestinal health in weaned pigs considering the maternal or nursery options. A total of 32 newly weaned pigs were randomly split into a 2 × 2 factorial arrangement considering maternal probiotic supplementation (with or without) in gestation-lactation and probiotic supplementation in the nursery period (with or without). After weaning, experimental diets were provided for 22 days. Total fecal and urine collection was performed from day 15 to 21. Blood samples were collected from all pigs on days 3 and 22 of the experiment to assess serum biochemistry and intestinal permeability. All pigs were euthanized on day 22 for intestinal tissue collection. Pigs born from probiotic-fed sows had greater (p < 0.05) total tract digestibility of dry matter (+1%) and gross energy (+1.3%), and greater (p < 0.05) metabolizable energy coefficient (+1.3%), which resulted in a 46 kcal/kg increase (p < 0.05) in the metabolizable energy content of the diet. Nitrogen intake (p = 0.035), uptake (p = 0.007), and retention (p = 0.012) were all increased in these pigs. Fecal moisture was reduced in pigs born from probiotic-fed sows and pigs fed the probiotic diet only in the nursery (p < 0.05). Pigs born from probiotic-fed sows had reduced intestinal permeability by 16% (p < 0.05), whereas pigs fed the probiotic diet in the nursery only tended to improve this response (p < 0.10). The villus:crypt ratio of pigs born from probiotic-fed sows was greater compared to the control (p < 0.05), while serum levels of alanine aminotransferase were lower (p < 0.05). Pigs born from probiotic-fed sows had increased nutrient digestibility and improved gut health. Therefore, it is concluded that supplementing the sow diets with probiotics rather than just providing diets in the nursery phase is an advantageous strategy.

Fatty acid-balanced oil improved nutrient digestibility, altered milk composition in lactating sows and fecal microbial composition in piglets.

This study aimed to investigate the effects of dietary supplementation of a fatty acid-balanced oil, instead of soybean oil, on reproductive performance, nutrient digestibility, blood indexes, milk composition in lactating sows, and fecal microbial composition in piglets. Twenty-four sows (Landrace×Yorkshire, mean parity 4.96) were randomly allotted to two treatments with twelve pens per treatment and one sow per pen based on their backfat thickness and parity. The experiment began on day 107 of gestation and continued until weaning on day 21 of lactation, lasting for 28 days. The control group (CG) was fed a basal diet supplemented with 2% soybean oil and the experimental group (EG) was fed the basal diet supplemented with 2% fatty acid-balanced oil. The fatty acid-balanced oil supplementation increased (p<0.05) the apparent total tract digestibility of dry matter, crude protein, and gross energy in sows. The lower (p<0.05) serum high-density lipoprotein cholesterol and albumin levels of sows were observed in the EG on day 21 of lactation. Dietary supplementation with the fatty acid-balanced oil decreased the fat content, increased the immunoglobulin G level, and changed (p<0.05) some fatty acid content in milk. Moreover, the fatty acid-balanced oil supplementation changed (p<0.05) the fecal microbial composition of piglets, where the average relative abundance of Spirochaetota was decreased (p<0.05) by 0.55% at the phylum level, and the average relative abundance of some potentially pathogenic fecal microorganism was decreased (p<0.05) at the species level. The fatty acid-balanced oil improved nutrient digestibility, changed the serum biochemical indices and milk composition of sows, and ameliorated the fecal microbial composition of piglets.

Isolation and Characterization of Soil Probiotic Bacteria in Cauvery River Basin and their Application in Aquaculture

Research on probiotics for aquatic animals is growing along with the demand for environmentally friendly aquaculture. Probiotics have been defined as live microbial supplements that improve the health of humans and terrestrial animals [1]. The micro biota in the digestive tract of fish and crustaceans is particularly dependent on the external environment due to the flow of water through the digestive tract. The need to increase disease resistance, growth of aquatic organisms and feed efficiency has led to the use of probiotics in aquaculture. In recent years, the use of probiotics in aquaculture has begun to increase [2]. There is documented evidence that probiotics can improve nutrient digestibility, increase stress tolerance and promote reproduction. Unlike previous publications, this study focuses on the role of soil probiotic bacteria isolated from the Cauvery basin in Tiruchirapalli district. Isolated microorganisms - Nitrosococcus and Neisseria were tested for their special role in aquaculture, mainly in tilapia fish (Oreochromis sp.). This study also improves the bioremediation capacity of the isolated soil bacteria, which was demonstrated by testing the water quality parameters of the probiotic used. water environment Fish growth and mortality were also measured. It was clearly observed that the use of an isolated soil probiotic from a cave river basin promoted the longevity of tilapia fish. Thus, the cultivation and use of soil probiotic bacteria in aquaculture is a promising approach in modern science.

Integrating dietary supplementation with poppy (Papaver somniferum L.) seed meal: effects on growth performance, nutrient digestibility, and faecal microbiota in broilers

Abstract. The study investigated the effects of supplementing broiler diets with poppy (Papaver somniferum L.) seed meal (PSM) on growth performance, nutrient digestibility, faecal microbiota, and blood profiles. A total of 250 unsexed 1 d old broilers were allocated to five distinct treatment groups. PSM was incorporated into the diets at varying levels: 0 % (PSM0), 5 % (PSM5), 10 % (PSM10), 15 % (PSM15), and 20 % (PSM20). The findings indicated that growth performance, including weight gain, feed intake, and feed conversion ratio (FCR), was significantly improved (P&lt;0.05) in the PSM20 group compared to the control. However, carcass weight experienced a notable decrease (P&lt;0.05) in the PSM20 group. Regarding nutrient digestibility, PSM supplementation led to reduced crude protein digestibility. Nevertheless, apparent metabolizable energy and ash content were significantly enhanced (P&lt;0.05) in the PSM15 and PSM20 groups. Notably, faecal microbiota also experienced substantial improvement (P&lt;0.05) in the PSM20 group. In conclusion, the study demonstrates that incorporating poppy seed meal at a rate of 20 % in broiler diets enhances growth performance, improves nutrient digestibility, and positively influences faecal microbiota composition.

Porcine in vitro digestion and solubilization of non-starch polysaccharides in corn and wheat supplemented with xylanase and feruloyl esterase

A study evaluated porcine in vitro digestion and non-starch polysaccharide (NSP) solubilization of corn and wheat without or with xylanase alone or xylanase plus feruloyl esterase (FE). The enzymes supplied 4000 U of xylanase and 35 U of FE per kilogram of sample. Samples were digested with pepsin at pH of 2.0 and then with pancreatin at pH of 6.8.. The digested samples were filtered to obtain unhydrolyzed residue, which was then washed using alcohol and acetone. The cereal grains (CG) before in vitro digestion and the washed unhydrolyzed residues were analyzed for dry matter and NSP. Xylanase improved (P &lt; 0.05) IVDDM for wheat (85 vs. 89%), but not for corn (78 vs. 79%). Addition of FE to the xylanase-supplemented CG increased (P &lt; 0.05) IVDDM for both CG, and reduced (P &lt; 0.05) the arabinoxylans in the unhydrolyzed residue for corn, but not for wheat. In conclusion, xylanase product used in the current study is more effective in improving digestibility of nutrients in wheat than in corn. Starter protein content and supplemental soybean oil or hydrogenated palm fatty acids in Holstein dairy calves: growth performance, protozoa population, and nitrogen utilization efficiency

The study hypothesized that a 24% protein starter diet supplemented with inert fat (palm fatty acids; PLFs) instead of triglyceride-rich soybean oil (SBO) would promote the growth of dairy calves. The effects of different protein contents (20 vs 24% CP) and fat sources (SBO vs PLF at 2.5% DM) on growth, digestibility, blood metabolites, rumen protozoa and urinary nitrogen in dairy calves were investigated. A total of 52 female dairy calves (mean age 3 days and BW 40.7 kg) were allocated to the following treatments in a completely randomized design: (1) 20% CP starter diet supplemented with SBO (20CP-SBO), (2) 20% CP starter diet supplemented with PLF (20CP-PLF), (3) 24% CP starter diet supplemented with SBO (24CP-SBO), and (4) 24% CP starter diet supplemented with PLF (24CP-PLF). Calves were weaned on day 53, and the study ended on day 73. The 24CP diets enhanced starter feed intake, average daily gain (ADG), withers height, hip width, organic matter (OM) digestibility, and preweaning blood glucose compared to the 20CP diets. In contrast, SBO supplementation reduced feed intake, ADG, withers height, OM and CP digestibility, and pre- and postweaning blood glucose and beta-hydroxybutyrate levels, while increasing preweaning aspartate aminotransferase, compared to PLF. SBO supplementation led to lower urinary excretion of purine derivatives and microbial protein synthesis postweaning, and a reduction in protozoa population both pre- and postweaning. Calves fed PLF had higher starter intake than those fed SBO, regardless of the diet being 20CP or 24CP. Calves fed the 24CP-PLF diet had the highest ADG, while calves fed the 20CP-SBO diet had the lowest ADG. Calves fed the 20CP-SBO diet had lower feed efficiency than calves fed the other diets during the preweaning period and throughout the experimental period. Calves fed the 24CP-PLF diet had higher hip heights at weaning and on day 73 than calves fed the other diets. Calves fed the 20CP-SBO diet had lower neutral detergent fiber digestibility than calves fed the other diets. Calves fed the 24CP-SBO diet had higher postweaning blood urea nitrogen concentration than calves fed the other diets. Feeding the 20CP-SBO diet to dairy calves decreased urinary allantoin excretion and rumen microbial protein synthesis but increased urinary nitrogen excretion during the preweaning period. Our results suggest that PLF is more suitable than SBO in calf diets, especially when calves fed 24% CP, possibly due to improved nutrient digestibility.

Exogenous Enzymes as Zootechnical Additives in Monogastric Animal Feed: A Review

Feed enzymes have been extensively used in livestock diets to enhance nutrient digestion and promote their growth performance. Indeed, recent research has indicated that feed enzymes, notably phytase, protease, and xylanase, function as catalysts, facilitating the breakdown of phytic acid, proteins, and β-1,4-xylan bonds and offering prospective advantages linked to the intestinal well-being and microbiota of young pigs and chickens. Various feed enzymes are currently being added to the diets of swine and broiler chickens. The potential enzymes used in the feed industry include cellulase, β-mannanase, β-glucanases, xylanases, phytases, proteases, lipases, and galactosidases. Though significant research has been conducted on phytase, protease, and xylanase, consistent findings, particularly in terms of improving nutrient digestibility and promoting growth performance of monogastric animals, are still limited. Also, the outcome of recent studies raises the question whether phytase and xylanase could play functional roles beyond increasing nutrient digestibility and intestinal health, such as positively modulating the intestinal microbiota and reducing environmental problems. Therefore, in this review we aimed to address the functional roles of exogenous enzyme activities in monogastric animal diets. Also, we sought to explore the advantages of these enzymes in enhancing the nutritional value of both alternative and conventional feedstuffs.

PSVIII-28 Effect of Replacement Dietary of Animal Protein with Insect Meal (Hermetia illucens) on Growth Performance, Nutrient Digestibility, Blood Profile, Gas Emission, and Fecal Microbial in Weaned Pigs

Abstract This study was conducted to investigate the effects of defatted and hydrolyzed forms of black soldier fly (Hermetia illucens; HI) larvae as fishmeal substitutes in the diet of weaned pigs on growth performance, nutrient digestibility, blood profile, gas emission, and fecal microbial. The 48 4-week-old weaned pigs [(Landrace × Yorkshire) × Duroc] with an initial body weight (BW) of 9.39 ± 1.02 kg were randomly assigned to a completely randomized three dietary treatments. Each treatment had 4 replicate pens, and 3 pigs were assigned to each pen. The three treatments were as follows: 1) a basal diet (CON), 2) a basal diet without a fish meal and substitute with defatted HI powder (T1), and 3) a basal diet without a fish meal and substitute with hydrolyzed HI powder (T2). The experiments progressed in 6 weeks. There were no significant differences (P &amp;gt; 0.05) among the treatment groups in BW, average daily gain, average daily feed intake, and feed efficiency during the entire experimental period (Table 1). At 2 wk, the CON and T1 groups increased (P &amp;lt; 0.05) crude protein (CP) digestibility compared with the T2 group (Table 2). The CON group had greater (P &amp;lt; 0.05) CP digestibility than the T2 group at 6 wk. At 6 wk, the T1 group showed a tendency for greater gross energy (P = 0.095) than the T2 group. The T1 group had greater (P &amp;lt; 0.05) lysine digestibility than the CON group at 2 wk (Table 3). Also, The T1 and T2 groups showed a tendency for greater (P = 0.070) glutamic acid digestibility than the CON group. At 6 wk, the T2 group increased (P &amp;lt; 0.05) lysine, arginine, and methionine digestibility compared with the CON group (Table 4). There were no significant differences (P &amp;gt; 0.05) among the treatment groups in blood profile and gas emission (Table 5 and 6). The T2 group had significantly greater (P &amp;lt; 0.05) lactobacillus counts than the T1 group at 4 wk (Table 7). In conclusion, it was found that the defatted and hydrolyzed forms of HI as a substitute for a fish meal did not negatively affect growth performance, blood profiles, and gas emissions. Also, the defatted and hydrolyzed forms of HI could improve nutrient digestibility and lactobacillus counts, which are beneficial bacteria in the intestine. Therefore, it is considered that the diets of weaned pig containing defatted or hydrolyzed HI has sufficient potential to replace fishmeal, which is an existing animal protein source in the diet.

Moringa oleifera leaf meal supplementation improves nutrient digestibility, milk yield, and reproductive performances in dairy cows during early lactation.

The study aimed to evaluate the effects of Moringa oleifera leaf meal (MOLM) supplementation on nutrient utilization, milk yield, and reproductive performance of early lactating Sahiwal cows. Control cows (GC) received a basal diet, while the treatment cows (GM) were supplemented with concentrate comprising 12% MOLM. Ovarian activity and uterine involution were monitored by trans-rectal ultrasonography on the 21st, 28th, 35th, and 42nd days postpartum. The result indicated that MOLM-supplemented cows required fewer days (P ≤ 0.05) to complete uterine involution. As lactation progresses, there was a significant reduction (P ≤ 0.05) in the diameter of the cervix and uterine horns in GM than GC. There was a significant increase in the number of follicles on the 21st day and average milk yield in GM than GC. The incidence of endometritis and cystic ovarian disease was less in MOLM supplemented group. The use of MOLM in the diet reduced the total cost per cow per successful service. It is concluded that MOLM can be safely included at 12% in the diet of early lactating cows to modulate the reproductive performances of dairy cows. Dairy farmers can use moringa leaf meal to feed their dairy cows, which is cheaper and improves production and reproduction performance.

PSIII-30 Effects of a Bacillus-Based Direct-Fed Microbial on in Vitro Gas Production and Fiber Digestibility of Feedstuffs

Abstract Direct-fed microbials (DFM) support the health and performance of the herd through different modes of action, including improvements on nutrient digestibility. In fact, Bacillus spp. has been identified as an important enzyme-producing bacteria that could benefit overall nutrient utilization by the beef and dairy cattle herd. Based on this rationale, we hypothesized that incubation of a Bacillus-based DFM would improve nutrient digestibility of feedstuffs often fed to ruminants. Therefore, our objective was to evaluate the effects of a Bacillus-based DFM on in vitro nutrient digestibility of different single feedstuffs. Six individual feedstuffs were sampled from China and South Africa and used herein. Feedstuffs were incubated or not (CON) with a Bacillus-based DFM (DFM; BOVACILLUS; B. licheniformis and B. subtilis; 3.2 × 109 CFU/gram; Chr. Hansen A/S., Hørsholm, Denmark) over a 48-h in vitro period. The dose of DFM was adjusted to the volume of the flasks being used and to represent the commercial dose of 3 grams/head per day (9.6 × 109 CFU/head). In vitro gas production, dry matter (DM), and neutral detergent fiber (NDF) digestibility were evaluated at 24 and 48 h. Data were analyzed using PROC MIXED of SAS (version 9.4). Mean total in vitro gas production increased by 5.1 and 6.5% when DFM was incubated at 24 and 48 h, respectively (P &amp;lt; 0.01). At 24 h, NDF digestibility was 12.6% greater for DFM-treated feedstuffs vs. CON (P = 0.02; 27.7 vs. 24.6% for DFM and CON, respectively), but no effects were observed on DM digestibility (P = 0.26). Conversely, 48-h DM and NDF digestibility were greater for DFM vs. CON (P ≤ 0.05), with improvements that were in the order of 3.7 and 15.1%, respectively. In conclusion, we observed greater in vitro fiber degradability and gas production of feedstuffs collected from China and South Africa over a 48-h period when the DFM BOVACILLUS was added at a dose representative of 9.6 × 109 CFU per head. Greater improvements in fiber degradability are likely due to the enzymes produced by the DFM BOVACILLUS.

The feasibility of partial replacement of berseem hay by spent mushroom (Pleurotus osteratus) substrate in rabbit diets on growth performance, digestibility, caecum fermentation, and economic efficiency

ABSTRACTNowadays, agricultural by-product disposal is a major concern. The mushroom by-products could be used as an alternative feed source in rabbit diets. Therefore, partial replacement of berseem hay (BH) with spent mushroom substrate (SMS) was conducted in four experimental groups as follows: 0, 20, 40, and 60% of SMS. Forty weaned New Zealand White (NZW) rabbits at 6th weeks of age with an initial body weight of 520.25 ± 70.01 g were fed the experimental diet for 8 weeks. The results showed that ash content and cell wall constituents in SMS were higher than in BH, while the other nutrient compounds in SMS were lower than in BH. Dietary SMS at a level of 60% increased the final body weight (p = 0.05) and feed conversion ratio (p ≤ 0.05). However, average daily gain and total feed intake were not affected by treatments. The replacement of SMS at levels of 40 and 60% significantly improved nutrient digestibility and total digestible nutrient value. SMS 60% decreased (p = 0.040) faecal N and improved (p = 0.006) retained nitrogen. The SMS replacement increased caecum length (p = 0.001), and full and empty caecum weight (p = 0.001 and 0.021, respectively) compared to the control. The SMS inclusion caused a decrease (p = 0.021 and 0.007) in the pH and NH3-N concentrations, respectively. Total VFA, acetic acid, butyric acid, and propionic acid proportions increased with the dietary inclusion of SMS in a level-dependent manner. Using SMS as a replacement for BH in growing rabbit diets reduced the total feed cost, and consequently improved net revenue, economic efficiency, and relative economic efficiency.

317 Awardee Talk: Functional Roles of Phytase and Xylanase for Nursery Pigs and Broiler Chickens

Abstract The objective of this research was to investigate the functional roles of phytase and xylanase in nursery pigs and broiler chickens. These enzymes have been widely studied and have shown consistent results improving nutrient digestibility and growth performance. However, recent studies have raised the hypothesis that phytase and xylanase could also have potential effects on intestinal and bone health, and intestinal microbiota. The first study studied at the effects of a bacterial 6-phytase on nutrient digestibility and retention of nursery and growing pigs. The results showed that supplementing a bacterial 6-phytase in pig diets can be a potential alternative for reducing high levels of inorganic phosphorus sources while still increasing the digestibility and utilization of phosphorus by the animals. The second study investigated the efficacy and optimal level of a bacterial 6-phytase supplemented beyond traditional dose levels on bone and intestinal health, nutrient digestibility, and growth performance of nursery pigs. The results revealed that the supplementation of the bacterial 6-phytase enhanced bone health, nutrient digestibility, and growth performance in nursery pigs. The third study aimed to determine the effects of phytase supplementation on the intestinal microbiota and morphology, bone health, nutrient digestibility, and growth performance of broiler chickens. The results showed that phytase supplementation had potential benefits on the microbiota by reducing potential harmful bacteria and increasing beneficial bacteria. Additionally, phytase had positive effects on bone health, intestinal morphology, and nutrient digestibility of broiler chickens. The fourth study examined the functional roles of xylanase on the intestinal health and growth performance of nursery pigs. The results showed that xylanase supplementation had beneficial effects on intestinal health by reducing the digesta viscosity, the relative abundance of potentially harmful bacteria, and the oxidative stress in the jejunal mucosa. All of these factors collectively reflected in improvements on the intestinal morphology, nutrient digestibility, and growth performance of nursery pigs. In conclusion, the results of these studies reaffirmed the benefits of phytase and xylanase related to nutrient digestibility and growth performance in nursery pigs and broiler chickens, as well as showed potential benefits on intestinal health and microbiota of the animals. Supplementing these enzymes could also contribute to a reduction in feed costs and environmental concerns associated with a poor hydrolysis and breakdown of antinutritional factors in animal diets.
