Impact of Cluster Front Line Demonstration (CFLD) on Yield & Economics of Toria in Rain-fed North Eastern Coastal Plain Zone of Odisha

Toria is one of the most important oilseed crop after harvest of Kharif rice in rainfed areas of Balasore district of Odisha. Krishi Vigyan Kendra, Balasore conducted Cluster Front Line Demonstration programme on Toria during Rabi 2019-20 to assess its impact on yield and income gap through scientific cultivation practices. The study was conducted in four villages of the district covering 180nos. farmer to find out the extension gap, technology gap, technology index and economic indexes of the crop before and after the intervention. The study revealed that, average yield of 8.5q/ha was obtained in the CFLD plot which was 44.06 per cent higher than the farmer’s practice. Similarly, the technology gap, extension gap and technology index were found to be 1.5q/ha, 2.6q/ha and 15 per cent, respectively. The average gross income in demonstrated plot was found to be Rs.34840/ ha as compared to Rs.23420/ha in farmers practice. Likewise, average net income of Rs.17040/ha was found in demonstration plot while it was Rs.8420/ha in farmers plot. The Benefit-cost ratio was found to be 1.96 in the demonstration plot which is higher as compared to 1.56 at farmer’s practices. The Study was conducted by Krishi Vigyan Kendra, Balasore to analyze the yield gap of Toria (Brassica campestris var. toria) through Cluster frontline demonstration in North Eastern Coastal Plain Zone of Odisha.

MYB transcription factors, their regulation and interactions with non-coding RNAs during drought stress in Brassica juncea

BackgroundBrassica juncea(L.) Czern is an important oilseed crop affected by various abiotic stresses like drought, heat, and salt. These stresses have detrimental effects on the crop’s overall growth, development and yield. Various Transcription factors (TFs) are involved in regulation of plant stress response by modulating expression of stress-responsive genes. The myeloblastosis (MYB) TFs is one of the largest families of TFs associated with various developmental and biological processes such as plant growth, secondary metabolism, stress response etc. However, MYB TFs and their regulation by non-coding RNAs (ncRNAs) in response to stress have not been studied in B. juncea. Thus, we performed a detailed study on the MYB TF family and their interactions with miRNAs and Long non coding RNAs.ResultsComputational investigation of genome and proteome data presented a comprehensive picture of the MYB genes and their protein architecture, including intron-exon organisation, conserved motif analysis, R2R3 MYB DNA-binding domains analysis, sub-cellular localization, protein-protein interaction and chromosomal locations. Phylogenetically, BjuMYBs were further classified into different subclades on the basis of topology and classification in Arabidopsis. A total of 751 MYBs were identified in B. juncea corresponding to 297 1R-BjuMYBs, 440 R2R3-BjuMYBs, 12 3R-BjuMYBs, and 2 4R-BjuMYBs types. We validated the transcriptional profiles of nine selected BjuMYBs under drought stress through RT-qPCR. Promoter analysis indicated the presence of drought-responsive cis-regulatory components. Additionally, the miRNA-MYB TF interactions was also studied, and most of the microRNAs (miRNAs) that target BjuMYBs were involved in abiotic stress response and developmental processes. Regulatory network analysis and expression patterns of lncRNA-miRNA-MYB indicated that selected long non-coding RNAs (lncRNAs) acted as strong endogenous target mimics (eTMs) of the miRNAs regulated expression of BjuMYBs under drought stress.ConclusionsThe present study has established preliminary groundwork of MYB TFs and their interaction with ncRNAs in B. juncea and it will help in developing drought- tolerant Brassica crops.

Dynamic Analysis of Fermentation Quality, Microbial Community, and Metabolome in the Whole Plant Soybean Silage

Soybean (Glycine max (L.) Merr.) is an important oilseed crop, known for its rich nutritional content and high-quality protein. To address the shortage of feed protein resources and better utilize soybeans as a raw material, this study investigated the feasibility of using whole-plant soybean (WPS) as silage. As the ensiling period is a critical fermentation parameter, identifying the optimal fermentation duration was a key objective. The research involves fermenting WPS for silage production, conducted over five fermentation durations: 7, 15, 30, 60, and 90 days. The fermentation quality, microbial community, and metabolome of WPS silage were analyzed across these different time points. WPS silage fermented for 30 days exhibited optimal fermentation characteristics, with the highest lactic acid (LA) content observed at 30 days (p < 0.05), while butyric acid (BA) was detected only at 60 and 90 days. At 30 days, Enterococcus genera reached its peak relative abundance and was identified as the dominant genus. Random forest analysis highlighted Pantoea genera as the most influential biomarker. Metabolomic analysis revealed that the metabolic pathways involved in the biosynthesis of essential amino acids valine, leucine, and isoleucine were significantly enhanced during the later stages of fermentation compared to the earlier stages. Under natural fermentation conditions, the optimal fermentation period for WPS silage is approximately 30 days. These findings provide a theoretical basis for the utilization of WPS and the subsequent optimization of fermentation quality.

Pangenome Data Analysis Reveals Characteristics of Resistance Gene Analogs Associated with Sclerotinia sclerotiorum Resistance in Sunflower

The sunflower, an important oilseed crop and food source across the world, is susceptible to several pathogens, which cause severe losses in sunflower production. The utilization of genetic resistance is the most economical, effective measure to prevent infectious diseases. Based on the sunflower pangenome, in this study, we explored the variability of resistance gene analogs (RGAs) within the species. According to a comparative analysis of RGA candidates in the sunflower pangenome using the RGAugury pipeline, a total of 1344 RGAs were identified, comprising 1107 conserved, 199 varied, and 38 rare RGAs. We also identified RGAs associated with resistance against Sclerotinia sclerotiorum (S. sclerotiorum) in sunflower at the quantitative trait locus (QTL). A total of 61 RGAs were found to be located at four quantitative trait loci (QTLs). Through a detailed expression analysis of RGAs in one susceptible and two tolerant sunflower inbred lines (ILs) across various time points post inoculation, we discovered that 348 RGAs exhibited differential expression in response to Sclerotinia head rot (SHR), with 17 of these differentially expressed RGAs being situated within the QTL regions. In addition, 15 RGA candidates had gene introgression. Our data provide a better understanding of RGAs, which facilitate genomics-based improvements in disease resistance in sunflower.

Pollen trapping as Honeybees pollination management and identification of dominant pollinators of Guizotia abyssinica (L.f).

Pollination is one of the most fascinating aspects of insect-plant interactions. Pollen is the male reproductive element of flowering plants, gathered by foraging Honeybees from the male parts of flowering plants called the anther. Guizotia abyssinica (L.f) is an important oilseed crop cultivated in Ethiopia and India, which belongs to the family Asteraceae. Although self-incompatibility is found in Guizotia abyssinica, a higher seed set is experienced in places with an active Honeybees population. Agricultural practices usually focus on inputs such as fertilizer application to improve seed yield of Guizotia abyssinica. However, these practices have little effect on yield if the availability of insect pollination is too low. To fill this gap, an experiment was carried out at Dandi district, West Shao zone, Oromia, Ethiopia to see the effect of Honeybees pollination management as an agronomic input. Pollination management of Honeybees was tested under feeding and non-feeding of colonies management. Pollen was trapped with and without sugar syrup feeding. The results discovered that Honeybees fed sugar syrup collected much more pollen than colonies not fed sugar syrup. The proportion of Guizotia abyssinica pollen collected through sugar syrup feeding of the colony was greater (62.2%), compared to the proportion of Guizotia abyssinica pollen trapped without sugar syrup feeding (37.8%). This indicates that sugar syrup feeding enhances the collection of pollen and probably enhances the pollination efficiency of Honeybees since they visit frequently to fulfill their daily protein requirement of pollen. Therefore, Honeybees pollination services should be included as one of the agronomic inputs with sugar syrup feeding as pollination management that might increase the yield of Guizotia abyssinica since it increases visiting frequency.

The Relationship between the Density of Winter Canola Stand and Weed Vegetation

Canola (Brassica napus L.) is an important oilseed crop that provides essential vegetable oil but faces significant competition from weeds that are influenced by various agronomic practices and environmental conditions. This study examines the complex interactions between canola stand density and weed intensity over three growing seasons, identifying a total of 27 weed species. It is important to establish a connection between the density of winter canola stands, the intensity of weeding and the response of individual weed species in real conditions. The case study was executed on plots located in the Přerov district (Olomouc region, Czech Republic). The assessment was carried out during two periods—autumn in October and spring in April. Canola plants (plant density) were counted in each evaluated area, weed species were identified, and the number of plants for each weed species was determined. Half of the plots were covered with foil before herbicide application to prevent these areas from being treated with herbicides. We used redundancy analysis (RDA) to evaluate the relationships between canola density and weed dynamics, both with and without herbicide treatment. The results show the ability of canola to compete with weeds; however, that is factored by the density of the canola stand. In dense stands (over 60 plants/m²), canola is able to suppress Galium aparine L., Geranium pusillum L., Lamium purpureum L., Papaver rhoeas L. and Chamomilla suaveolens (Pursh) Rydb. Nevertheless, there are weed species that grow well even in dense canola stands (Echinochloa crus-galli (L.) P. Beauv., Phragmites australis (Cav.) Steud., Tripleurospermum inodorum (L.) Sch. Bip. and Triticum aestivum L.). These findings highlight the potential for using canola stand density as a strategic component of integrated weed management to reduce herbicide reliance and address the growing challenge of herbicide-resistant weed populations. This research contributes significantly to our understanding of the dynamics of weed competition in canola systems and informs sustainable agricultural practices for improved crop yield and environmental stewardship.

Early monitoring of drought stress in safflower (Carthamus tinctorius L.) using hyperspectral imaging: a comparison of machine learning tools and feature selection approaches

Early detection of drought stress is essential for preventing permanent plant damage and minimizing yield loss. This study utilized hyperspectral imaging at the leaf level to visualize drought stress in safflower plants (Carthamus tinctorius L.). Three safflower genotypes, Palenus, A82, and IL-111, were cultivated under three irrigation levels. Stress conditions were simulated by depleting 50%, 70%, and 90% of soil water content, representing unstressed (US), mild stress (MS), and severe stress (SS) conditions, respectively. Hyperspectral images of leaf samples were captured before any visible signs of water scarcity emerged. Classification analysis was performed using the full mean spectral data with partial least squares discriminant analysis, soft independent modeling of class analogy (SIMCA), support vector machines, and artificial neural network (ANN) classifiers. Feature selection methods were applied to extract the most informative wavebands, and ANN was used to build predictive models. Spatial analysis involved pixel-wise classification using both unsupervised (k-means clustering) and supervised (best classifiers) approaches. ANN outperformed other classifiers using the entire spectral data, effectively distinguishing US, MS, and SS classes in the Palenus, A82, and IL-111 genotypes, achieving F1-scores of 92.22%, 96.01%, and 96.47%, respectively. Among the feature selection methods, SIMCA-based features excelled in monitoring stress conditions in the Palenus and A82 genotypes. In supervised spatial analysis, ANN models clearly depicted the progression of stress in leaves across different genotypes. This study demonstrates the potential of hyperspectral imaging to differentiate various levels of drought stress in safflower, an important oilseed crop.

A gain-of-function mutation in BnaIAA13 disrupts vascular tissue and lateral root development in Brassica napus.

Rapeseed (Brassica napus) is an important oilseed crop worldwide. Plant vascular tissues are responsible for long-distance transport of water and nutrients and for providing mechanical support. The lateral roots absorb water and nutrients. The genetic basis of vascular tissue and lateral root development in rapeseed remains unknown. This study characterized an ethyl methanesulfonate-mutagenized rapeseed mutant, T16, which showed dwarf stature, reduced lateral roots, and leaf wilting. SEM observations showed that the internode cells were shortened. Observations of tissue sections revealed defects in vascular bundle development in the stems and petioles. Genetic analysis revealed that the phenotypes of T16 were controlled by a single semi-dominant nuclear gene. Map-based cloning and genetic complementarity identified BnaA03.IAA13 as the functional gene; a G-to-A mutation in the second exon changed glycine at position 79 to glutamic acid, disrupting the conserved degron motif VGWPP. Transcriptome analysis in roots and stems showed that auxin and cytokinin signaling pathways were disordered in T16. Evolutionary analysis showed that AUXIN/INDOLE-3-ACETIC ACID is conserved during plant evolution. The heterozygote of T16 showed significantly reduced plant height while maintaining other agronomic traits. Our findings provide novel insights into the regulatory mechanisms of vascular tissue and lateral root development, and offer a new germplasm resource for rapeseed breeding.

High-density genetic map construction and QTL mapping to identify genes for blight defense- and yield-related traits in sesame (Sesamum indicum L.).

Sesame (Sesamum indicum L.) is an important oilseed crop widely cultivated in subtropical and tropical areas. Low genetic yield potential and susceptibility to disease contribute to low productivity in sesame. However, the genetic basis of sesame yield- and disease-related traits remains unclear. Here, we represent the construction of a high-density bin map of sesame using whole genome sequencing of an F2 population derived from 'Yizhi' and 'Mingdeng Zhima'. A total of 2766 Bins were categorized into 13 linkage groups. Thirteen significant QTLs were identified, including ten QTLs related to yield, two QTLs related to Sesame Fusarium wilt (SFW) disease, and one QTL related to seed color. Among these QTLs, we found that SFW-QTL1.1 and SFW-QTL1.2 were major QTLs related to Fusarium wilt disease, explaining more than 20% of the phenotypic variation with LOD > 6. SCC-QTL1.1 was related to seed coat color, explaining 52% of the phenotypic variation with LOD equal to 25.3. This suggests that seed color traits were controlled by a major QTL. Candidate genes related to Fusarium wilt disease and seed color in the QTLs were annotated. We discovered a significant enrichment of genes associated with resistance to late blight. These genes could be spectral disease resistance genes and may have a role in the regulation of Fusarium wilt disease resistance. Our study will benefit the implementation of marker-assisted selection (MAS) for the genetic improvement of disease resistance and yield-related traits in sesame.

Development of high oleic Spanish bunch groundnut (Arachis hypogaea L.) through marker-assisted backcrossing

ABSTRACT Groundnut (Arachis hypogaea L.) is an important oilseed crop. A specific type of groundnut, known as high-oleate groundnut, is particularly sought-after for its antioxidant properties and benefits to human health due to its favorable fatty acid profile. To breed high-oleic acid varieties of groundnut, two elite varieties of Tamil Nadu, India, TMVGn 13 and GJG 33, were hybridized with a high-oleic groundnut donor released in India, Girnar 4. The resulting progenies were then backcrossed with their recurrent parents for two generations, using marker-assisted backcross selection. Six progenies in the TMVGn 13 × Girnar 4 cross and nine progenies in the GJG 33 × Girnar 4 cross were identified as homozygous mutants for ahFAD2A and ahFAD2B genes, respectively. Three lines with high oleic acid content (>70%), good oil content (48–50%), and pod yield per plant were selected from these progenies. Following multi-location and adaptive research yield trials, these lines may be released as high-oleic acid varieties.

Inheritance Analysis for Quantitative Traits in F2 Populations of Mustard Genotypes Grown Under Heat Stress Environments

Genetic variability is the basic requirement for improving crops which further leads to progress in crop breeding. This study was carried out to determine the inheritance pattern of various agronomically important traits in F2 populations of mustard genotypes. The objective of this study was to determine the genetic variability for agro-morphological traits under control conditions (CC) and heat stress environments (HS) and provide the future strategies to improve this important oilseed crop. The experiment was conducted at the Botanical Garden, Sindh Agriculture University, Tandojam. During 2022-23, a total of 27 genotypes including nine parental mustard genotypes and eighteen F2 progenies were grown under control and heat stress conditions. The experiment was laid out in a randomized complete block design with three replications. The analysis of variance for the genotypes, treatments, and F2 progenies revealed significant differences (P≤0.05) in most of the studied characters, highlighting the importance of genetic variations in the evaluated breeding materials of mustard for stress breeding. When estimating heritability for different characters, including seed yield plant-1, it was found that F2 crosses showed varying levels of heritability for different characteristics, ranging from low to moderate and high. This implies that careful selection may be conducted for traits that have low heritability, especially for seed yield.; however, those characters which showed high heritability would be selected at early generation stage to improve the characters.

Genome-wide analysis of the NYN domain gene family in Brassica napus and its function role in plant growth and development

The NYN domain gene family consists of genes that encode ribonucleases that are characterized by a newly identified NYN domain. Members of the family were widely distributed in all life kingdoms and play a crucial role in various RNA regulation processes, although the wide genome overview of the NYN domain gene family is not yet available in any species. Rapeseed (Brassica napus L.), a polyploid model species, is an important oilseed crop. Here, the phylogenetic analysis of these BnaNYNs revealed five distinct groups strongly supported by gene structure, conserved domains, and conserved motifs. The survey of the expansion of the gene family showed that the birth of BnaNYNs is explained by various duplication events. Furthermore, tissue-specific expression analysis, protein–protein interaction prediction, and cis-element prediction suggested a role for BnaNYNs in plant growth and development. Interestingly, the data showed that three tandem duplicated BnaNYNs (TDBs) exhibited distinct expression patterns from those other BnaNYNs and had a high similarity in protein sequence level. Furthermore, the analysis of one of these TDBs, BnaNYN57, showed that overexpression of BnaNYN57 in Arabidopsis thaliana and B. napus accelerated plant growth and significantly increased silique length, while RNA interference resulted in the opposite growth pattern. It suggesting a key role for the TDBs in processes related to plant growth and development.

EFFECT OF PACLOBUTRAZOL AND ABSCISIC ACID APPLICATION ON MORPHO-PHYSIOLOGICAL GROWTH AND PRODUCTIVITY OF CANOLA (Brassica napus L.)

Canola (Brassica napus L.) is an important non-conventional oilseed crop that can be grown in diverse soil and climatic conditions. However, seed shattering after maturity is a major problem in canola production as it can cause up to 50% yield loss if harvesting is delayed due to adverse conditions. Different agronomic and physiological interventions can be opted to enhance yield stability and shattering resistance in canola. A field study was conducted to analyze the effect of paclobutrazol and abscisic acid on growth and yield attributes of canola. Two canola cultivars Hybrid (45S42) and Inbred Sandal Canola were sown and paclobutrazol and abscisic acid were applied with the dose of 50 mg L-1 and 0.5 mM respectively, alone or in combination. The experiment was laid out under Randomized Complete Block Design (RCBD) with factorial arrangement having three replications. The results showed that exogenous application of paclobutrazol and abscisic acid alone or in combination posed significant effects on both canola cultivars. The plant height decreased significantly, however, leaf area index, seed weight, number of seeds per silique and number of branches per plant were increased compared with control. Moreover, the combination of paclobutrazol and abscisic acid recorded a higher increase in leaf area index, dry matter accumulation plant yield and number of seeds per pod. The effects of the combined application of paclobutrazol and abscisic acid recorded higher effects compared with their individual application. Between cultivars, the Hybrid cultivar (45S42) recorded better yield as compared to the Inbred Sandal Canola. These results thereby suggest that Hybrid (45S42) cultivar under combined application of paclobutrazol and abscisic acid can be a suitable option for gaining high yield and productivity.

Comprehensive characterization of protease inhibiting gene family, cis-regulatory elements, and protein interaction network in linseed and their expression upon bud fly infestation

Linseed, also known as flax is an important oilseed crop with many potential uses in paint, textile, food and pharmaceutical industries. Susceptibility to bud fly (Dasyneura lini Barnes) infestation is a serious biotic concern leading to severe yield penalty in linseed. Protease inhibitors (PIs) are potential candidates that activate during the insect-pest attack and modulate the resistance. In the present study, we explored the PI candidates in the linseed genome and a total of 100 LuPI genes were identified and grouped into five distinct subgroups. The analysis of cis-acting elements revealed that almost all LuPI promoters contain several regulatory elementary related to growth and development, hormonal regulation and stress responses. Across the subfamilies of PIs, the specific domains are consistently found conserved in all protein sequences. The tissue-specific in-silico expression pattern via RNA-seq revealed that all the genes were regulated during different stress. The expression through qRT-PCR of 15 genes revealed the significant up-regulation of LuPI-24, LuPI-40, LuPI-49, LuPI-53, and LuPI-63 upon bud fly infestation in resistant genotype EC0099001 and resistant check variety Neela. This study establishes a foundation resource for comprehending the structural, functional, and evolutionary dimensions of protease inhibitors in linseed.

Feasibility and Growth Dynamics of Mustard Farming in Uttar Pradesh and Bundelkhand Region of U.P. India

Bundelkhand is a region of central India lying in Uttar Pradesh and Madhya Pradesh. It is one of the least socio-economically developed regions of the country mainly due to regular incidence of drought and erratic rainfall. Mustard is one of the most important oilseed crop of rabi season for Bundelkhand region on the basis of area under the crop. With the limited resources, there has been a noticeable change in the area, production and productivity of mustard. Apart from edible oil, it also has industrial use in producing items like varnishes, soaps, paints, hydrogenated oil, perfumery, lubricants, etc. The study is an attempt to examine the pattern of growth, instability, cost and return of production of mustard crops at state (Uttar Pradesh) and regional (Bundelkhand region of Uttar Pradesh) level. Study is based on secondary data obtained from various published sources. Compound Annual Growth Rate, coefficient of variation, Commission for Agricultural Costs & Prices (CACP) cost concept and returns were used as analytical tools to assess the financial suitability and performance of the crop in both the Uttar Pradesh and Bundelkhand region. It was observed that the growth in area (2.94%) and production (4.36%) of mustard crop in Bundelkhand region was increasing at a higher rate as that of Uttar Pradesh. Higher net return was earned by the farmer growing mustard in Uttar Pradesh than the farmer of Bundelkhand region.

GC-MS analysis and antifungal potential of flower extract of Acacia nilotica subsp. Indica against Macrophomina phaseolina

Macrophomina phaseolina is a wide host ranged soil-borne fungal plant pathogen. It infects more than 500 host plant species belonging to 100 families. Many important oil-seed and leguminous crops are known to be attacked by this devastating plant pathogen. In the present study, antifungal potential of flowers of a leguminous tree Acacia nilotica subsp. indica, was assessed against this pathogen through bioassays guided fractionation. Initially, methanolic extracts of 1 %–5 % of leaf, flower, root-bark and stem-bark of the plant species under consideration were evaluated for their antifungal potential against the target pathogen. Among these, the best antifungal activity was shown by flower extract. The reduction in growth of the test fungal strain was 27–49 %, 4–40 % and 2–27 % due to flower, root-bark and leaf extracts, respectivey, over control. Flower extract was partitioned using n-hexane, chloroform, ethyl acetate and n-butanol as the solvents. Bioassays guided study of these fractions of methanolic extract of flower revealed that high antifungal potential was shown by n-hexane and chloroform fractions against M. phaseolina causing 26–53 % and 28–50 % decline in fungal biomass, respectively, as compared to that of control. GC-MS analysis of chloroform fraction revealed the presence of 27 compounds in this fraction. Among these cyclopentanol,-1-methyl (10.93 %) was the predominant compound followed by methyl, 4,4-dimethyl butanoate (7.04 %), 1-pentanol (6.80 %), 2-propanol, 1-cyclopropyl (6.11 %), 1H,imidazole-4-5-dihydro-2-methyl (5.93 %), trichloroethane (5.91 %), carbonic acid-ethyl hexyl ester (4.59 %), 1,4-butandiol,2,3-bis(methylene)- (4.54 %) and [S]-3,4-dimethyl pentanol (4.48 %).

Exploration of GmDof11-lncRNA13082 Module Regulating Oil Synthesis in Plants.

Soybean (Glycine max [Linn.] Merr.) is an important oilseed crop. Although transcription factors (TFs) can coordinate the expression of mRNA and lncRNA, their coordination in the soybean oil synthesis pathway remains unclear. This study examined the interaction between the TF GmDof11 and lncRNA13082 and found that overexpression of GmDof11 led to an increase in the number of Arabidopsis seeds, thousand seed weight, crude protein, hydrolysis amino acid, and soluble sugar. Additionally, it reduced the triglyceride and starch contents and affected the proportion of fatty acids, increasing the contents of palmitic acid, stearic acid, and linolenic acid. The yeast two-hybrid experiments revealed that GmDof11 interacts with GmBCCP1, GmLEC1b, and GmFAB2 proteins. In the RT-qPCR analysis of transgenic soybean roots, it was found that GmDof11 can activate the production of lncRNA13082 and work in conjunction with lncRNA13082 to oversee oil synthesis and nutrient storage. Our research provides robust theoretical evidence for a comprehensive resolution of TF-lncRNA regulation in the soybean oil synthesis network.

Screening of Castor Germplasm and Efficacy Studies of Bioagents and Fungicides Against Botryotinia ricini (Godfrey) Whetzel

Castor is an important non-edible oilseed crop having exceptional oil characteristics for the chemical industry. The crop hosts several diseases, among them gray mold is the most destructive one hindering its productivity. This article presents a study assessing the resistance of different germplasm lines screened against Botrytis ricini under artificial conditions. It also examines the effect of various fungicides on B. ricini isolates, which were obtained from infected Ricinus communis (castor oil plant) using standard tissue isolation techniques. The fungus was isolated from infected Castor raceme by standard tissue isolation technique and its pathogenicity was established by proving Koch’s postulates. Out of 47 germplasm lines screened, TSIL-19 showed resistance response to gray mold under both detached spike and capsule method. Under in vitro assessment, Trichoderma viride (86.11 %) and Pseudomonas fluorescens (71.48 %) bio agents exhibited maximum mycelial growth inhibition of the pathogen. Among the seven combination fungicides evaluated, Fenamidone 10% + Mancozeb 50% WG recorded highest mycelial growth inhibition (100%) of B. ricini.

Genome-wide identification of the GDSL-type esterase/lipase protein (GELP) gene family in Ricinus communis and its transcriptional regulation during germination and seedling establishment under different abiotic stresses

GDSL-type esterase/lipase protein (GELP) genes are crucial in the specialized lipid metabolism, in the responses to abiotic stresses, and in the regulation of plant homeostasis. R. communis is an important oilseed crop species that can sustain growth and productivity when exposed to harsh environmental conditions. Herein, we raised the question of whether the GELP gene family could be involved in the acquisition of R. communis tolerance to abiotic stresses during seed germination and seedling establishment. Thus, we used bioinformatics and transcriptomics to characterize the R. communis GELP gene family. R. communis genome possesses 96 GELP genes that were characterized by extensive bioinformatics, including phylogenetic analysis, subcellular localization, exon-intron distribution, the analysis of regulatory cis-elements, tandem duplication, and physicochemical properties. Transcriptomics indicated that numerous RcGELP genes are readily responsive to high-temperature and salt stresses and might be potential candidates for genome editing techniques to develop abiotic stress-tolerant crops.

Genome-wide identification of SIMILAR to RCD ONE (SRO) gene family in rapeseed (Brassica napus L.) reveals their role in drought stress response

ABSTRACT Rapeseed (Brassica napus L.) is an important oilseed crop widely cultivated worldwide, and drought is the main environmental factor limiting its yield enhancement and the expansion of planted areas. SIMILAR TO RCD ONE (SRO) is a plant-specific small gene family that plays a crucial role in plant growth, development, and responses to abiotic stresses such as drought. However, the functional role of SROs in rapeseed remains poorly understood. In this study, 19 BnaSROs were identified from the rapeseed genome, with 9, 10, 10, 18, and 20 members identified from the genomes of Brassica rapa, Brassica nigra, Brassica oleracea, Brassica juncea, and Brassica carinata, respectively. We then analyzed their sequence characteristics, phylogenetic relationships, gene structures, and conserved domains, and explored the collinearity relationships of the SRO members in Brassica napus and Brassica juncea. Next, we focused on the analysis of tissue expression and stress-responsive expression patterns of rapeseed SRO members and examined their expression profiles under ABA, MeJA and water-deficit drought treatments using qPCR. Transcriptome data analysis and qPCR detection indicated that BnaSROs exhibit multiple stress-responsive expression patterns. BnaSRO1 and BnaSRO11, which are likely to function through interactions with NAC transcription factors, were screened as major drought-regulated members. Our results provide a solid foundation for functional analysis of the role of the SRO gene family in abiotic stress responses, especially drought stress responses, in rapeseed.
