Regulation of Myocardial Lipid Accumulation in the Development of Obesity

Energy metabolism is important in the development of heart disease, which is the leading cause of death worldwide. The importance of energy metabolism in disease is becoming even more clear with the rise in obesity, metabolic diseases, and the recognition that they all fit together as with the recent offcial report of a new syndrome named Cardiovascular-Kidney-Metabolic (CKM) Syndrome. A complete understanding of myocardial lipid storage and subsequent utilization of lipids is essential to understanding the impact of these metabolic diseases in the heart and on the development of cardiac dysfunction and hypertrophy. In this study we investigate how the heart controls fuel storage by taking advantage of the impact of the transcription factor peroxisome proliferator activated receptor α (PPARα) on 1) fatty acid metabolism and lipid accumulation, 2) its key role in sex differences in cardiac hypertrophy which we previously reported, and 3) the regulation of lipid droplets. It is controversial whether a decline in myocardial PPARα mediates cardiac hypertrophy and whether it increases or reduces lipid accumulation. Using our tamoxifen inducible cardiac-specific PPARα knockout mouse (cPPAR−/−) we see less cardiac lipid accumulation in response to a 5 week high fat diet. Our data indicate that this difference in triglyceride levels is present in both males and females. Importantly, this is not driven by a difference in fatty acid supply to these hearts. Serum fatty acid levels are not lower in cPPAR−/− vs Control mice fed a high fat diet. Further, we have not observed a difference in high fat diet induced body weight gain between cPPAR−/− and Control mice. We have begun to examine how these differences in high fat diet-induced lipid accumulation correlate with cardiac hypertrophy by looking at heart weight normalized to tibia length. While we did not expect 5 weeks of a high fat diet to induce cardiac hypertrophy, it is possible that the difference in cardiac lipid storage in the cPPAR−/− hearts could impact this. This measurement did not indicate a difference in cardiac hypertrophy between high fat diet fed cPPAR−/− and Control mice. We are utilizing this mouse as a model to investigate the regulation of lipid droplets and the lipid droplet proteome. So far, our results are indicating that the regulation of lipid droplet proteins is not affected in a uniform manner. For example, our data is suggesting that the expression of different members of the perilipin family are affected in distinct ways. Overall, these data indicate that a reduction in cardiac PPARα protects against obesity induced cardiac lipid accumulation. Going forward, it will be interesting to see how these changes occur in relation to the lipid droplet proteome and whether any changes that occur differentially in males vs female may be involved in sex differences in cardiac hypertrophy. Better understanding these factors will be essential for the development of safe and effective future therapies for CKM syndrome and other cardiovascular diseases. ND-EPSCoR. Regulation of energy metabolism in human pluripotent stem cells.

All living organisms need energy to carry out their essential functions. The importance of energy metabolism is increasingly recognized in human pluripotent stem cells. Energy production is not only essential for cell survival and proliferation, but also critical for pluripotency and cell fate determination. Thus, energy metabolism is an important target in cellular regulation and stem cell applications. In this review, we will discuss key factors that influence energy metabolism and their association with stem cell functions.

Systemic Bioinformatic Analyses of Nuclear-Encoded Mitochondrial Genes in Hypertrophic Cardiomyopathy.

Hypertrophic cardiomyopathy (HCM) is an autosomal dominant disease and mitochondria plays a key role in the progression in HCM. Here, we analyzed the expression pattern of nuclear-encoded mitochondrial genes (NMGenes) in HCM and found that the expression of NMGenes was significantly changed. A total of 316 differentially expressed NMGenes (DE-NMGenes) were identified. Pathway enrichment analyses showed that energy metabolism-related pathways such as “pyruvate metabolism” and “fatty acid degradation” were dysregulated, which highlighted the importance of energy metabolism in HCM. Next, we constructed a protein-protein interaction network based on 316 DE-NMGenes and identified thirteen hubs. Then, a total of 17 TFs (transcription factors) were predicted to potentially regulate the expression of 316 DE-NMGenes according to iRegulon, among which 8 TFs were already found involved in pathological hypertrophy. The remaining TFs (like GATA1, GATA5, and NFYA) were good candidates for further experimental verification. Finally, a mouse model of transverse aortic constriction (TAC) was established to validate the genes and results showed that DDIT4, TKT, CLIC1, DDOST, and SNCA were all upregulated in TAC mice. The present study represents the first effort to evaluate the global expression pattern of NMGenes in HCM and provides innovative insight into the molecular mechanism of HCM.

Suppression of a core metabolic enzyme dihydrolipoamide dehydrogenase (dld) protects against amyloid beta toxicity in C. elegans model of Alzheimer's disease

A decrease in energy metabolism is associated with Alzheimer's disease (AD), but it is not known whether the observed decrease exacerbates or protects against the disease. The importance of energy metabolism in AD is reinforced by the observation that variants of dihydrolipoamide dehydrogenase (DLD), is genetically linked to late-onset AD. To determine whether DLD is a suitable therapeutic target, we suppressed the dld-1 gene in Caenorhabditis elegans that express human Aβ peptide in either muscles or neurons. Suppression of the dld-1 gene resulted in significant restoration of vitality and function that had been degraded by Aβ pathology. This included protection of neurons and muscles cells. The observed decrease in proteotoxicity was associated with a decrease in the formation of toxic oligomers rather than a decrease in the abundance of the Aβ peptide. The mitochondrial uncoupler, carbonyl cyanide 4-(trifluoromethoxy) phenylhydrazone (FCCP), which like dld-1 gene expression inhibits ATP synthesis, had no significant effect on Aβ toxicity. Proteomics data analysis revealed that beneficial effects after dld-1 suppression could be due to change in energy metabolism and activation of the pathways associated with proteasomal degradation, improved cell signaling and longevity. Thus, some features unique to dld-1 gene suppression are responsible for the therapeutic benefit. By direct genetic intervention, we have shown that acute inhibition of dld-1 gene function may be therapeutically beneficial. This result supports the hypothesis that lowering energy metabolism protects against Aβ pathogenicity and that DLD warrants further investigation as a therapeutic target.

Dyslipidemia in retinal metabolic disorders.

The light‐sensitive photoreceptors in the retina are extremely metabolically demanding and have the highest density of mitochondria of any cell in the body. Both physiological and pathological retinal vascular growth and regression are controlled by photoreceptor energy demands. It is critical to understand the energy demands of photoreceptors and fuel sources supplying them to understand neurovascular diseases. Retinas are very rich in lipids, which are continuously recycled as lipid‐rich photoreceptor outer segments are shed and reformed and dietary intake of lipids modulates retinal lipid composition. Lipids (as well as glucose) are fuel substrates for photoreceptor mitochondria. Dyslipidemia contributes to the development and progression of retinal dysfunction in many eye diseases. Here, we review photoreceptor energy demands with a focus on lipid metabolism in retinal neurovascular disorders.

CitE Enzymes Are Essential for Mycobacterium tuberculosis to Establish Infection in Macrophages and Guinea Pigs.

Bacterial citrate lyase activity has been demonstrated in various eukaryotes, bacteria and archaea, underscoring their importance in energy metabolism of the cell. While the bacterial citrate lyase comprises of three different subunits, M. tuberculosis genome lacks CitD and CitF subunits of citrate lyase complex but encodes for 2 homologs of CitE subunits, Rv2498c and Rv3075c. Using temperature sensitive mycobacteriophages, we were able to generate both single and double citE mutant strains of M. tuberculosis. The survival experiments revealed increased susceptibility of the double mutant strain to oxidative stress in comparison to the parental strain. Also, simultaneous deletion of both citE1 and citE2 in M. tuberculosis genome resulted in impairment of intracellular replication in macrophages. The double mutant strain displayed reduced growth in lungs and spleens of guinea pigs. This is the first study demonstrating that M. tuberculosis critically requires CitE subunits of citrate lyase for pathogenesis. Taken together, these findings position these enzymes as potential targets for development of anti-tubercular small molecules.

Evolving Concepts of Myocardial Energy Metabolism: More Than Just Fats and Carbohydrates.

Although mitochondrial fatty acid and carbohydrate oxidation are the major source of ATP production in the heart, it is becoming increasingly clear that oxidation of other energy substrates, such as ketones and branched chain amino acids (BCAAs), can also contribute to energy production. Of importance is that alterations in cardiac ketone and branched chain amino acids metabolism may also impact the severity of heart failure through alterations in cellular signaling, despite these fuels providing a lower contribution to overall energy production. Dating back to the first studies investigating glucose uptake in the isolated rabbit heart by Locke and Rosenheim in 1907 to the first studies in humans in the late 1940s by Richard Bing and colleagues assessing cardiac substrate extraction via measurement of arterial coronary sinus differences, cardiac energy metabolism has been a topic of interest for cardiologists and scientists alike. The importance of energy metabolism in the heart has become widely appreciated, as has the dynamic nature of the heart’s ability to metabolize a wide range of energy substrates to meet its energy requirements. Fatty acids are recognized as a key source of energy for the heart, as well as carbohydrates, such as glucose and lactate (Figure).1 In 1961, Shipp et al2 were the first to demonstrate that increasing fatty acid availability to the heart results in a marked inhibition of glucose oxidation, though credit for the reciprocal relationship between fatty acids and glucose metabolism (glucose/fatty acid cycle) is attributed to the work of Randle et al.3 Confirmation of Randle’s glucose/fatty acid cycle in the rodent heart has since been demonstrated in the human heart.1 Figure. The importance of nutritional support in patients in intensive care units (ICU) has been highlighted for many years. This is particularly important in those entering the ICU with lung disease that progresses to respiratory failure who required ventilatory support, where the importance of energy metabolism is demonstrated in major complications such as mortality, time on mechanical ventilation, and time in ICU. The present article shows a narrative review of the evidence on the use of nutritional therapy, and its influence on the outcome of patients admitted to the ICU due to respiratory failure.

Mitochondrial ATPase subunit 6 and cytochrome B gene variations in obese Turkish children.

Objective: Due to the importance of energy metabolism in mitochondria, mitochondrial genome variations are evaluated in energy-related diseases such as obesity. To date, several nuclear genes were found to be related to obesity. Our aim in this study was to investigate the presence of polymorphisms in mitochondrial ATPase subunit 6 (mt-ATP6) and cytochrome b (mt-CytB) genes that may be associated with childhood obesity.Methods: The mt-ATP6 and mt-CytB genes were amplified and entirely sequenced in a series of 100 obese and in an equal number of healthy Turkish children aged between 6-14 years.Results: A total of 118 synonymous and nonsynonymous variations were detected in the obese and control groups. Only two previously reported synonymous substitutions (mt.8614T>C and mt.8994G>A) in the mt-ATP6 gene were found to be significantly higher in the obese group compared to the control group (p<0.05). In the mt-ATP6 gene, one novel nonsynonymous substitution (mt.8726C>T) and one novel synonymous substitution (mt.9108A>T) were found. In the mt-CytB gene, one nonsynonymous substitution (mt.14880T>C) and two synonymous substitutions (mt.14891C>T and mt.15091C>T) were novel substitutions.Conclusion: Two synonymous substitutions (mt.8614T>C and mt.8994G>A) in the mt-ATP6 gene may be associated with childhood obesity. Our study provides the first data about mitochondrial genome variations in a Turkish obese population and also the first in obese children. More cases should be screened in obese groups in order to understand the effects of mitochondrial polymorphisms in the development of obesity.

Noninvasive monitoring of lactate dynamics in human forearm muscle after exhaustive exercise by1H-magnetic resonance spectroscopy at 7 tesla

Despite its importance in energy metabolism, lactate in human skeletal muscle has been difficult to detect by noninvasive (1)H-magnetic resonance spectroscopy mainly due to interference from large water and lipid signals. Long echo-time acquisitions at 7 T effectively attenuates the water and lipid signals in forearm muscle allowing direct observation of both lactate resonances, the methine at 4.09 ppm and the methyl at 1.31 ppm. Using this approach, we were able to monitor lactate dynamics at a temporal resolution of 32 s. While lactate was not detectable at rest, immediately after an acute period of exercise to fatigue the forearm muscle, lactate rose to a level comparable to that of creatine (∼30 mmol/kg wet weight). In a typical (1)H-magnetic resonance spectrum collected using a echo-time of 140 ms, the lactate methine and methyl resonances both appear as doublets with an unusually large splitting of ∼20 Hz due to residual dipolar coupling. During muscle recovery following exercise, the lactate signals decay rapidly with a time constant of t½ = 2.0 ± 0.6 min (n = 12 subjects). This fast and simple lactate detection method may prove valuable for monitoring lactate metabolism in cancer and in sports medicine applications.

RNF34 Is a Cold-Regulated E3 Ubiquitin Ligase for PGC-1α and Modulates Brown Fat Cell Metabolism

The transcriptional coactivator PGC-1α is a master regulator of energy metabolism and adaptive thermogenesis in the brown fat cell. PGC-1α is a short-lived protein, and the molecular components that control PGC-1α turnover and their functional importance in energy metabolism are largely unknown. Here we performed a luciferase-based overexpression screen and identified a Ring-finger-containing protein, RNF34, as a specific E3 ubiquitin ligase for PGC-1α. RNF34 is a nuclear protein that interacts with and ubiquitinates PGC-1α to promote its turnover. Interestingly, RNF34 binds to the C-terminal half of PGC-1α and targets it for degradation independently of the previously identified N-terminal phosphodegron motif. In brown fat cells, knockdown of RNF34 increases the endogenous PGC-1α protein level, uncoupling protein 1 (UCP1) expression, and oxygen consumption, while the opposite effects are observed in brown fat cells ectopically expressing wild-type RNF34 but not in cells expressing the ligase activity-defective mutant. Moreover, cold exposure and β3-adrenergic receptor signaling, conditions that induce PGC-1α expression, suppress RNF34 expression in the brown fat cell, indicating a physiological relevance of this E3 ligase in thermogenesis. Our results reveal that RNF34 is a bona fide E3 ubiquitin ligase for PGC-1α and negatively regulates brown fat cell metabolism.

AMPK γ2 subunit gene PRKAG2 polymorphism associated with cognitive impairment as well as diabetes in old age

Metabolic and cognitive disorders are closely related. However, the molecular mechanism underlying this association is still elusive. Given the importance of energy metabolism in neuronal cells, AMP-activated protein kinase (AMPK), a master switch of energy metabolism, could be an independent factor affecting cognitive as well as metabolic functions. Therefore, we examined the relationship between the AMPK γ2 gene, the PRKAG2 -26C/T polymorphism and cognitive impairment or diabetes in 1609 subjects aged from 60 to 80. We performed multivariate logistic regression analyses with adjustment for age, gender, education, smoking, alcohol, depression, waist circumference, APOE e4, and stroke history. We found a significant association between the -26C/T polymorphism (CC vs. CT/TT) and cognitive impairment (OR, 1.6; 95% CI, 1.1-2.3). Moreover, this polymorphism (CC/CT vs. TT) was also related to the presence of diabetes (OR, 1.8; 95% CI, 1.2-2.8). Importantly, the relationship with cognitive impairment was still significant in non-diabetic individuals (OR, 1.6; 95% CI, 1.1-2.4). Further analyses with a subpopulation (n=611) revealed that CC homozygotes relative to T-allele carriers had significantly better performances in verbal memory and attentional tasks. These findings collectively support a hypothesis that AMPK has a role not only in metabolic functioning but also in cognitive functioning in humans. Extended longitudinal study with a larger number of samples is warranted.

Metabolic adaptation of Escherichia coli to long-term exposure to salt stress

The aim of this work was to understand the relevance of central carbon metabolism in salt stress adaptation of Escherichia coli. The cells were grown anaerobically in batch and chemostat reactors at different NaCl concentrations using glycerol as a carbon source. Enzyme activities of the main metabolic pathways, external metabolites, ATP level, NADH/NAD + ratio, l-carnitine production and the expression level of the main genes related to stress response were used to characterize the metabolic state under the osmotic stress. The results provided the first experimental evidence of the important role played by central metabolism adaptation and cell survival after long-term exposure to salt stress. Increased glycolytic fluxes and higher production of fermentation products indicated the importance of energy metabolism. Carbon fluxes under stress conditions were controlled by the decrease in the isocitrate dehydrogenase/isocitrate lyase ratio and the phosphoenolpyruvate carboxykinase/phosphoenolpyruvate carboxylase ratio, and the increase in the phosphotransferase/acetyl-CoA synthetase ratio. Altogether, the results demonstrate that, under salt stress, E. coli enhances energy production by substrate-level phosphorylation (Pta–Ack pathway) and the anaplerotic function of the TCA cycle, in order to provide precursors for biosynthesis. The results are discussed in relation with the general stress response and metabolic adaptation of E. coli.

Effects of dietary supplementation of L-carnitine on performance, carcass and meat characteristics of quails

The present study was conducted to determine the effects of feeding diets containing two levels of metabolizable energy (12.13 or 11.72 MJ ME/kg) and two different fat sources (sunflower- and fish-oil) with or without supplemental L-carnitine (0 or 50 mg/kg diet) on growth performance and carcass and meat characteristics of Japanese quails. Two hundred and forty day-old male quail chicks were randomly assigned to eight treatment groups, each subdivided into three replicates of 10 chicks. The chicks were raised from hatch until 5 wks of age. Feeding the diet containing 12.13 MJ ME/kg increases body weight and body weight gain significantly, and improved the feed conversion ratio above that of the lower energy diet. Feed intakes of the birds were unaffected by treatments. The cold carcass yield of quails fed the diet containing sunflower oil was significantly higher than those receiving the diets containing fish oil. After 35 days of feeding the diet containing the standard energy level (12.13 MJ ME/kg) the thigh yield of the birds was significantly higher than that of the chicks on the lower energy diet. Dietary treatments did not affect pH values of edible meat in the quails. Feeding diets containing sunflower oil and L-carnitine significantly decreased malonaldehyde (MA) amounts in the edible meat. The crude protein content of the edible meat fraction was significantly higher when the dietary energy level was decreased from 12.13 to 11.72 MJ ME/kg diet. Decreasing dietary energy levels significantly decreased the “L” (lightness) and “b” values (less yellow) of the meat, while dietary L-carnitine supplementation resulted in a significant increase in “L” value. The total edible meat of the quails on the diets containing fish oil had a higher “a” value (more red) than the quails receiving sunflower oil in their diets. It was concluded that additional studies are required to clarify the role of dietary L-carnitine in the oxidation of long-chain fatty acids, its antioxidant properties and its importance in energy metabolism in Japanese quails. Molecular and cellular adaptations to carbohydrate and fat intake.

Adaptation to carbohydrate and fat intake involves changes in a number of biochemical parameters at the cellular level. A change in the concentration of fat or carbohydrate in the blood acts directly to influence metabolic pathways by altering the flux of intermediates into cells. This in turn alters the concentration of hormones and other signaling molecules and changes the rate of expression of genes coding for key regulatory proteins or enzymes in metabolic pathways. These effects occur at different rates and in a tissue-specific manner in response to diet. A key metabolic adaptation involves changes in the level of expression of genes coding for proteins of critical importance in energy metabolism; this is largely due to an altered rate of transcription of selected genes under the control of hormones and/or carbohydrate and lipid. The mediators of this effect are transcription factors, that is, nuclear proteins which integrate the effects of hormones and substrates with the transcription process by binding to response elements in the promoters of regulated genes and interacting with the transcription machinery at the TATA box, thereby altering the activity of RNA polymerase II. In this review we will outline the hierarchy of cellular adaptations to diet and will emphasize the latest concepts of gene regulation in response to metabolites and hormones. In particular, we will review the role of the various transcription factors involved in the regulated expression of the gene for the cytosolic form of phosphoenolpyruvate carboxykinase (GTP) (EC 4.1.132), a key gluconeogenic enzyme.

Behavior of certain parameters of lipid and energy metabolism. IV. Regulation of lipid and energy metabolism in livers and race and mini pigs

To characterize the lipid and the energy metabolism in the livers of genetically different types of pigs (land race pig and mini pig), the authors determined the activities of enzymes typical of and limiting these metabolic pathways. Furthermore, they measured the concentrations of typical metabolites and ascertained parameters that are of importance in energy metabolism. The concentrations of acetyl CoA and free fatty acids in the livers of mini pigs were significantly greater than those in the livers of land race pigs, whereas the cholesterol, glycerol, triglyceride and acetoacetate concentrations were reduced. The activities of glucose-6-phosphate dehydrogenase (E.C.1.1.1.49.), citrate synthase (E.C.4.1.3.7.) and ATP citrate lyase (E.C.4.1.3.8.) were lower in the livers of mini pigs than in the livers of land race pigs, whereas the activity of fatty acid synthase was higher. The concentrations of cyclic 3',5'-adenosine monophosphate (AMP) and adenosine diphosphate (ADP) were lower in the livers of mini pigs than in those of land race pigs. In land race pigs, the metabolic process seems, therefore, to be determined in favour of the degradation of free fatty acids and of the generation of energy. In mini pigs, lipogenesis in the liver appears to be the decisive metabolic pathway. The possibility of a higher coordinating control mechanism of the lipid and the energy metabolism is discussed.
