Exploring the impact of titanium dioxide nanoparticles (nTiO2) at varied concentrations in combination with Azospirillum brasilense on wheat growth and physiology

BackgroundNanoparticles (NPs), as a novel source of Nano fertilizers in crop production. Titanium dioxide nanoparticles (nTiO2) also have the potential to improve plant growth, but their effect on the wheat crop is not studied enough. Nothing is known about that how nTiO2 specifically effect on wheat crops especially with soil microbes and how much its dose is effective. Azospirillum brasilense is a promising bio-fertilizer that can be applied in combination with nano-fertilizers to increase crop productivity and can enhance the efficiency of other fertilizers. MethodsThe present study was planned to investigate the role of different doses of titanium dioxide nanoparticles (nTiO2) with Azospirillum brasilense on the growth and physiology of wheat, having three replications. Eleven treatments were planned in the field condition (T0 = Control (No A. brasilense and No nTiO2), T1 = nTiO2 @20 mg/L, T2 = nTiO2 @20 mg/L + A. brasilense, T3 = nTiO2 @30 mg/L, T4 = nTiO2 @30 mg/L + A. brasilense, T5 = nTiO2 @40 mg/L, T6 = nTiO2 @40 mg/L + A. brasilense, T7 = nTiO2 @50 mg/L, T8 = nTiO2 @50 mg/L + A. brasilense, T9 = nTiO2 @60 mg/L, T10 = nTiO2 @60 mg/L + A. brasilense) having randomized complete block design (RCBD). ResultsResults revealed that the individual application of A. brasilense showed significantly higher results in all treatments, but nTiO2 application shows a positive impact on wheat growth, yield, and physiological parameters when used in a lower concentration. nTiO2 @30 mg/L with A. brasilense gives the highest results as compared to all other treatments with the production of higher antioxidant enzymes, nutrient uptake, higher leaf area index, and photosynthesis. Use of nTiO2 @40 mg/L or with the higher dose negatively affects wheat crops, but with the A. brasilense application its negative effect control up to a certain level. ConclusionThis study highlights the potential advantages of combining A. brasilense with nTiO2, for the growth of wheat crops. While A. brasilense alone consistently produced favorable results, lesser doses of nTiO2 also showed promising results but with the combination of nTiO2 and A. brasilense @ 30 mg/L producing the greatest outcomes. The detrimental effects of excessive nanoscale titanium dioxide on plant growth and development, which may result in stress and physiological abnormalities in the plants, are probably the cause of the decrease in crop output at greater nTiO2 concentrations. However, care should be used while utilizing larger nTiO2 concentrations since they could harm wheat crop growth and yield.

Adverse effects of titanium dioxide nanoparticles on beneficial gut bacteria and host health based on untargeted metabolomics analysis

Titanium dioxide (TiO2) is a common additive in foods, medicines, and personal care products. In recent years, nano-scale particles in TiO2 additives have been an increasing concern due to their potential adverse effects on human health, especially gut health. The objective of this study was to determine the impact of titanium dioxide nanoparticles (TiO2 NPs, 30 nm) on beneficial gut bacteria and host response from a metabolomics perspective. In the in vitro study, four bacterial strains, including Lactobacillus reuteri, Lactobacillus gasseri, Bifidobacterium animalis, and Bifidobacterium longum were subjected to the treatment of TiO2 NPs. The growth kinetics, cell viability, cell membrane permeability, and metabolomics response were determined. TiO2 NPs at the concentration of 200 μg/mL showed inhibitory effects on the growth of all four strains. The confocal microscope results indicated that the growth inhibitory effects could be associated with cell membrane damage caused by TiO2 NPs to the bacterial strains. Metabolomics analysis showed that TiO2 NPs caused alterations in multiple metabolic pathways of gut bacteria, such as tryptophan and arginine metabolism, which were demonstrated to play crucial roles in regulating gut and host health. In the in vivo study, mice were fed with TiO2 NPs (0.1 wt% in diet) for 8 weeks. Mouse urine was collected for metabolomics analysis and the tryptophan metabolism pathway was also significantly affected in TiO2 NPs-fed mice. Moreover, four neuroprotective metabolites were significantly reduced in both in vitro bacteria and in vivo urine samples. Overall, this study provides insights into the potential adverse effects of TiO2 NPs on gut bacteria and the metabolic responses of both bacteria and host. Further research is needed to understand the causality between gut bacteria composition and the metabolism pathway, which is critical to monitor the gut-microbiome mediated metabolome changes in toxicological assessment of food components.

Nitrogen forms and concentration influence the impact of titanium dioxide nanoparticles on the biomass and antioxidant enzyme activities of Microcystis aeruginosa

Nanoparticles (NPs) are becoming more widely produced, used, and released into the aquatic environment. In aquatic ecosystems, these NPs affect different populations of photosynthesizing organisms, such as cyanobacteria. This study aimed to evaluate the effects of titanium dioxide (TiO2) NPs (48mgl-1) combined with low (0.04mM) and high (9mM) concentrations of urea and nitrate on Microcystis aeruginosa. Microcystins (MCs) production and release were monitored in the cyanobacterium. The results showed that high urea concentration (9mM) combined with TiO2 NPs inhibited growth, pigment, and malondialdehyde (MDA) content by 82%, 63%, and 47%, respectively. The treatment also increased the reactive oxygen species (ROS) and glutathione S-transferase (GST) activity by 40.7% and 67.7%, respectively. Similarly, low nitrate (0.04mM) combined with TiO2 NPs inhibited growth by 40.3% and GST activity by 36.3% but stimulated pigment production and ROS concentration in M. aeruginosa. These responses suggest that high urea combined with TiO2.NPs and high nitrate combined with TiO2 NPs induced oxidative stress in cyanobacteria. The peroxidase (POD) activity of M. aeruginosa decreased by 17.7% with increasing urea concentrations. Our findings suggest that TiO2 NPs combined with changing nutrient (urea and nitrate) concentrations may adversely affect cyanobacterial development and antioxidant defense enzymes.

Comparative assessment of oxygen uptake rate of activated sludge and Escherichia coli exposed to nanomaterials.

The adverse effects of engineered nanomaterials (ENMs) on bacterial populations found in wastewater treatment plants (WWTPs) or natural systems have been studied for more than a decade, but conflicting evidence on the matter still makes it a subject of considerable concern. In this paper, the short-term exposure impact of titanium dioxide nanoparticles (nTiO2), carboxyl-functionalized multiwall carbon nanotubes (f-MWCNT), and zero-valent iron nanoparticles (nZVI) toward activated sludge and Escherichia coli (E. coli) was investigated through respiration inhibition experiments. Microorganisms were exposed to nanoparticle concentrations of 50, 100 and 200 mg/L (nTiO2, f-MWCNT) and 20, 50 and 100 mg/L (nZVI). The experiments showed that nTiO2 produced no inhibition in activated sludge or E. coli; up to 100 mg/L of nZVI did not inhibit the activated sludge respiration but 50 mg/L inhibited 24 ± 3% the respiration of E. coli and damaged its cell membrane. Activated sludge respiration was inhibited 17 ± 3% with 200 mg/L of f-MWCNT while for E. coli the inhibition was 36 ± 15% and the cell membrane was damaged with a 100 mg/L dose. Transmission electron microscopy (TEM) showed nTiO2-bacteria and nZVI-bacteria surface interaction while bacteria appeared punctured by f-MWCNT. E. coli was more susceptible than activated sludge to the nanomaterials and nZVI was more toxic than f-MWCNT for E. coli. These results demonstrated the absence of acute toxicity effects of the studied nanomaterials at those concentrations expected to occur in activated sludge facilities, and it would only be a concern in case of extremely high inputs, underscoring the resilience of WWTPs biological treatment.

Titanium dioxide nanoparticles induced reactive oxygen species (ROS) related changes of metabolomics signatures in human normal bronchial epithelial (BEAS-2B) cells

Titanium dioxide often enters the respiratory tract in the form of nano-dust in occupational sites. Metabolomics may be a promising method for studying the toxicology of nano titanium dioxide. The intention of this study was to explore the possible impact of titanium dioxide nanoparticles (TiO2 NPs) on the metabolomics signatures of human normal bronchial epithelial (BEAS-2B) cells and to search for investigate the role of reactive oxygen species (ROS). In this study, BEAS-2B cells were treated by TiO2 NPs (0, 25, 50 and 100 μg/mL) for 48 h. The metabolites extracted from BEAS-2B cells were determined by untargeted metabolomics technique, and the differential metabolites and metabolic pathways were discovered by using multivariate analysis. Intracellular ROS was detected by DCFH-DA probe and flow cytometry method. Machine learning was used to explore the relationship between ROS and metabolomics changes. Totally, seventy-six differential metabolites and the steroid biosynthesis pathway of BEAS-2B cells were observed after treatment of TiO2 NPs. Lipid and lipid-like metabolites were the most significant classes among the metabolite products induced by TiO2 NPs. TiO2 NPs resulted in a dose-dependent increase in intracellular ROS levels, and correlated with most of the differential metabolites. In conclusion, TiO2 NPs increased the level of the oxidative stress, which could contribute to the altered signature represented by lipid metabolism in BEAS-2B cells.

Foliar spray and seed priming of titanium dioxide nanoparticles and their impact on the growth of tomato, defense enzymes and some bacterial and fungal diseases

Bacterial and fungal pathogens are major constraints in successful cultivation of tomato resulting into rigorous yield losses. The impact of titanium dioxide nanoparticles (TiO2 NPs) on bacteria, i.e. Pseudomonas syringae pv. tomato, Xanthomonas campestris pv. vesicatoria, Pectobacterium carotovorum subsp. carotovorum and Ralstonia solanacearum and fungi Fusarium oxysporum f. sp. lycopersici and Alternaria solani and on the growth of tomato, chlorophyll, carotenoid contents, proline and defense enzymes viz., superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and phenylalanine ammonia lyase (PAL) activities were determined. TiO2 NPs were applied at 0.10 mL·L−1 and 0.20 mL·L−1concentration as foliar spray and seed priming. Inoculation of tomato with either of these pathogens reduced plant growth, chlorophyll and carotenoid contents, but increased proline and activities of SOD, CAT, APX and PAL. Maximum increase in plant growth, chlorophyll, carotenoid, proline, SOD, CAT, APX and PAL, and reduction in disease indices was observed at foliar spray with 0.20 mL·L−1 TiO2 NPs, followed by seed priming. Use of 0.10 mL·L−1 NPs as seed priming was less effective. Scanning electron microscopy revealed the absorption of TiO2 NPs by trichomes, stomata and seed surface. Foliar spray with 0.20 mL·L−1 TiO2 NPs was useful in increasing plant growth and provided substantial management of bacterial and fungal diseases of tomato.

Impact of titanium dioxide nanoparticles on lead acetate-induced genotoxicity in the major histocompatibility complex region and 16S rRNA sequence in mice

This manuscript aimed to study the effect of co-exposure of both lead acetate and titanium dioxide nanoparticles (TiO2 NPs) on the major histocompatibility complex (Mhc) region and 16S rRNA sequence in vivo. Twenty-four male albino Swiss mice have randomly separated into four groups; Group 1 (control): was received only distilled water. Groups 2-4 received lead acetate (400 mg/kg body weight, orally by gavage for 15 consecutive days) + either distilled water (orally), or ZnCl2 (4 mg/kg body weight, intraperitoneal injection), or titanium deoxidize nanoparticles TiO2 (200 mg/kg body weight, intraperitoneal injection) for additional 15 consecutive days, respectively. According to the results of 16S rRNA sequence that the highest P-distance value (0.002) found between control group and both (lead acetate and lead acetate+TiO2)-treated groups which reflected the genetic effects of both lead acetate and TiO2 NPs. The results of microsatellites revealed that the exposure to lead acetate affected the genetic structure, where the genetic similarity between control group and lead acetate-treated group was 0.83, while zinc chloride decrease lead acetate-induced genotoxicity where the genetic similarity was 0.88 comparing to the control group. Moreover, the co-exposure to lead acetate and TiO2 NPs led to more genotoxicity and DNA damage, that is clear in the genetic similarity between control group and lead acetate+TiO2-treated group which was 0.74.

Bioaccumulation of BDE47 in testes by TiO2 nanoparticles aggravates the reproductive impairment of male zebrafish by disrupting intercellular junctions

This study attempts to explore the potential impact of titanium dioxide nanoparticles (n-TiO2) on bioconcentration and reproductive impairments of male zebrafish in the presence of 2,2′,4,4′-tetrabromodiphenyl ether (BDE47), the congener of PBDEs predominant in environment and most abundant in biosamples. n-TiO2 nanoparticles strongly adsorbed BDE47 to form BDE47/TiO2 complex, which was taken up into the testes of zebrafish, and increased the tissue burdens of both BDE47 and n-TiO2. Correspondingly, no observed toxic dose of n-TiO2 (100 μg/L) was found to aggravate the abnormal histological morphology of the testes and the decrease in egg production, gonadosomatic index, sexual hormone levels and related gene expression in zebrafish in the presence of BDE47 at 5 or 50 μg/L. In addition, n-TiO2 exacerbated the destruction resulting from the ultrastructural disassembly of intercellular connectivity of germ cells in zebrafish and the decrease in transepithelial electrical resistance in TM4 cells induced by BDE47. Furthermore, n-TiO2 enhanced BDE47 to initially activate p-JNK MAPK signaling pathway and subsequently triggered the downregulation of junction proteins (i.e., ZO-1, Connexin-43 and N-cadherin), leading to impaired cell-cell junctions in vivo and in vitro. Our results demonstrated that n-TiO2 should act as a carrier to facilitate the accumulation of BDE47 in zebrafish testes and result in a synergistic effect on BDE47-induced adverse reproductive outcomes via disruption of intercellular connectivity of zebrafish testes. This study is beneficial in providing a scientific basis for improving the health risk assessment of environmental pollutants, particularly those that coexist with nanoparticle contamination in realistic environments.

Effect of Titanium Dioxide Nanoparticles on the Expression of Efflux Pump and Quorum-Sensing Genes in MDR Pseudomonas aeruginosa Isolates.

Most of the infections caused by multi-drug resistant (MDR) P. aeruginosa strains are extremely difficult to be treated with conventional antibiotics. Biofilm formation and efflux pumps are recognized as the major antibiotic resistance mechanisms in MDR P. aeruginosa. Biofilm formation by P. aeruginosa depends mainly on the cell-to-cell communication quorum-sensing (QS) systems. Titanium dioxide nanoparticles (TDN) have been used as antimicrobial agents against several microorganisms but have not been reported as an anti-QS agent. This study aims to evaluate the impact of titanium dioxide nanoparticles (TDN) on QS and efflux pump genes expression in MDR P. aeruginosa isolates. The antimicrobial susceptibility of 25 P. aeruginosa isolates were performed by Kirby–Bauer disc diffusion. Titanium dioxide nanoparticles (TDN) were prepared by the sol gel method and characterized by different techniques (DLS, HR-TEM, XRD, and FTIR). The expression of efflux pumps in the MDR isolates was detected by the determination of MICs of different antibiotics in the presence and absence of carbonyl cyanide m-chlorophenylhydrazone (CCCP). Biofilm formation and the antibiofilm activity of TDN were determined using the tissue culture plate method. The effects of TDN on the expression of QS genes and efflux pump genes were tested using real-time polymerase chain reaction (RT-PCR). The average size of the TDNs was 64.77 nm. It was found that TDN showed a significant reduction in biofilm formation (96%) and represented superior antibacterial activity against P. aeruginosa strains in comparison to titanium dioxide powder. In addition, the use of TDN alone or in combination with antibiotics resulted in significant downregulation of the efflux pump genes (MexY, MexB, MexA) and QS-regulated genes (lasR, lasI, rhll, rhlR, pqsA, pqsR) in comparison to the untreated isolate. TDN can increase the therapeutic efficacy of traditional antibiotics by affecting efflux pump expression and quorum-sensing genes controlling biofilm production.

Impact of titanium dioxide nanoparticles on intestinal community in 2,4,6-trinitrobenzenesulfonic acid (TNBS)-induced acute colitis mice and the intervention effect of vitamin E.

Titanium dioxide nanoparticles (TiO2-NPs) are widely applied as additives in foods due to their excellent whitening and brightening capability. Although the toxicity and antibacterial activity of TiO2-NPs have been extensively studied, their impact on the gut microbiota in vivo still remains unclear, especially in animals with gastrointestinal disorders. In the present study, healthy mice and TNBS-induced colitis mice were administered with TiO2-NPs (38.3 ± 9.3 nm) orally at a dose of 100 mg per kg bw daily for 10 days to study the impact of TiO2-NPs on the gut microbiota and colitis development. Moreover, the mechanism of TiO2-NPs on the gut microbiota was also discussed when the colitis mice were additionally administered with vitamin E to remove ROS. Changes in the microbiota community structure and gut-associated function prediction were analyzed through bioinformatics. The result showed that the oral administration of TiO2-NPs mitigated colitis symptoms by reducing the DAI and CMDI scores and TNF-α level. Furthermore, 16S rDNA sequencing analysis showed that the structure and function prediction of gut microbiota could be modified in healthy mice and colitis mice after exposure to TiO2-NPs, but the opposite physiological effect occurred since the dominant flora varied in these two groups. Moreover, vitamin E intervention did not change the effects of TiO2-NPs on the microbiota community structure and gut-associated function, which indicates that the mechanism of the biological effects of TiO2-NPs on the gut microbiota may not be associated with their ability to induce the generation of ROS. In summary, our work firstly found that TiO2-NPs could regulate the gut microbiota of colitis mice and participate in the mitigation of TNBS-induced acute colitis, and the capability of TiO2-NPs to induce the generation of ROS inducement did not contribute to this process.

Impact of Titanium Dioxide Nanoparticles on Cd Phytotoxicity and Bioaccumulation in Rice (Oryza sativa L.).

The impact of engineered nanoparticles (ENPs) on the migration and toxicity of coexisting pollutants is still unclear, especially in soil media. This study aims to evaluate the impact of titanium dioxide nanoparticles (TiO2 NPs) on the phytotoxicity of cadmium (Cd) to Oryza sativa L., and the migration of cadmium (Cd) in the soil-rice system. Three different Cd stress groups (C1 group: 1.0 mg kg−1, C2 group: 2.5 mg kg−1 and C3 group: 5.0 mg kg−1) were set in the pot experiment, and the target concentration of TiO2 NPs in each group were 0 mg kg−1 (T0), 50 mg kg−1 (T1), 100 mg kg−1 (T2) and 500 mg kg−1 (T3). Plant height and biomass decreased with the increasing of Cd content in paddy soil. TiO2 NPs could lower the phytotoxicity of Cd in terms of the changes in the morphological and biochemical characteristics, especially in the tillering and booting stage. In the tillering stage, TiO2 NPs addition caused a significant increase in plant height, biomass and the total chlorophyll content in the leaves of Oryza saliva L. In the booting stage, TiO2 NPs addition caused a 15% to 32% and 24% to 48% reduction of malondialdehyde (MDA) content for the C2 and C3 group, respectively, compared to that of the respective control treatment (T0). TiO2-NPs addition reduced the activity of peroxidase (POD) in the leaves in the booting and heading stage, and the activity of catalase (CAT) in the tillering stage. In the C1 and C2 group, the grain Cd content in the 100 and 500 mg kg−1 TiO2 NPs treatments reached 0.47–0.84 mg kg−1, obviously higher than that of the treatment without TiO2 NPs (0.27–0.32 mg kg−1), suggesting that TiO2-NPs could promote Cd migration in the soil-rice system.

Effect of titanium dioxide nanoparticles on the accumulation and distribution of arsenate in Daphnia magna in the presence of an algal food.

The impact of titanium dioxide nanoparticles (nano-TiO2) on the bioavailability of metals in aquatic filter-feeding organisms has rarely been investigated, especially in the presence of algae as a food source. In this study, we quantified the accumulation and subcellular distribution of arsenate (AsV) in Daphnia magna in the presence of nano-TiO2 and a green alga (Scenedesmus obliquus) food source. Results showed that S. obliquus significantly increased the accumulation of total arsenic (As) and titanium (Ti) in D. magna. The presence of this food source increased As in metal-sensitive fractions (MSF) and as biologically detoxified metals (BDM), while it decreased Ti levels in MSF but increased levels as BDM. The difference in the subcellular distribution of As and Ti demonstrates the dissociation of As from nano-TiO2 during digestion at subcellular partitioning irrespective of food availability. In turn, the presence of algae was shown to increase metal-based toxicity in D. magna due to the transfer of As from BMD to MSF. Furthermore, S. obliquus significantly increased the concentration of As and Ti in soluble fractions, indicating that As and nano-TiO2 ingested by D. magna could be transferred more readily to their predators in the presence of S. obliquus. Our study shows the potential of algae to increase the toxicity and biomagnification of AsV. Furthermore, it highlights food as an important factor in the toxicity assessment of nanomaterials and co-existing pollutants.

Impact of titanium dioxide nanoparticles on purification and contamination of nematic liquid crystals.

We have investigated the impact of titanium dioxide nanoparticles on the ionic contamination of liquid crystals. Nematic liquid crystals with high and low initial ionic contamination have been examined. It has been shown that titanium dioxide nanoparticles reduced the ion density of liquid crystals with high initial ionic contamination from 134.5 × 1012 cm−3 to 63.2 × 1012 cm−3. In the case of liquid crystals with low initial ionic contamination, the nanoparticles led to an insignificant increase of ion density from 19.8 × 1012 cm−3 to 25.7 × 1012 cm−3.

Metabolomic and proteomic investigations of impacts of titanium dioxide nanoparticles on Escherichia coli.

In a previous study, it was demonstrated that the toxic impact of titanium dioxide nanoparticles on Escherichia coli starts at 10 ppm and is closely related to the presence of little aggregates. It was also assumed that only a part of the bacterial population is able to adapt to this stress and attempts to survive. Proteomic analyses, supported by results from metabolomics, reveal that exposure of E. coli to nano-TiO2 induces two main effects on bacterial metabolism: firstly, the up-regulation of proteins and the increase of metabolites related to energy and growth metabolism; secondly, the down-regulation of other proteins resulting in an increase of metabolites, particularly amino acids. Some proteins, e.g. chaperonin 1 or isocitrate dehydrogenase, and some metabolites, e.g. phenylalanine or valine, might be used as biomarkers of nanoparticles stress. Astonishingly, the ATP content gradually rises in relation with the nano-TiO2 concentration in the medium, indicating a dramatic release of ATP by the damaged cells. These apparently contradictory results accredit the thesis of a heterogeneity of the bacterial population. This heterogeneity is also confirmed by SEM images which show that while some bacteria are fully covered by nano-TiO2, the major part of the bacterial population remains free from nanoparticles, resulting in a difference of proteome and metabolome. The use of combined–omics has allowed to better understand the heterogeneous bacterial response to nano-TiO2 stress due to heterogeneous contacts between the protagonists under environmental conditions.

Toxicity of titanium dioxide nanoparticles: Effect of dose and time on biochemical disturbance, oxidative stress and genotoxicity in mice.

The toxic impact of titanium dioxide nanoparticles (TiO2NPs) on human health is of prime importance owing to their wide uses in many commercial industries. In the present study, the effect of different doses and exposure time durations of TiO2NPs (21nm) inducing oxidative stress, biochemical disturbance, histological alteration and cytogenetic aberration in mice liver and bone marrow was investigated. Different doses of (TiO2NPs) (50, 250 and 500mg/kg body weight) were each daily intrapertioneally injected to mice for 7, 14 and 45days. Aspartate and alanine aminotransferases (AST &ALT), gamma glutamyl transpeptidase (GGT), total protein, total antioxidant capacity (TAC), malondialdehyde (MDA), glutathione (GSH), catalase (CAT) and nitric oxide (NO) levels were measured. The work was extended to evaluate the liver histopathological pattern and the chromosomal aberration in mice spinal cord bone marrow. The results revealed severe TiO2NPs toxicity in a dose and time dependent manner with positive correlation (r=0.98) for most investigated biochemical parameters. The same observation was noticed for the histological analysis. In case of cytogenetic study, chromosomal aberrations were demonstrated after injection of TiO2NPs with 500mg/kg b. wt. for 45days. In conclusion, the selected biochemical parameters and the liver architectures were influenced with dose and time of TiO2NPs toxicity, while the genetic disturbance started at the high dose of exposure and for long duration. Further studies are needed to fulfil the effect of TiO2NPs on pharmaceutical and nutritional applications.

Impact of titanium dioxide nanoparticles on the bacterial communities of biological activated carbon filter intended for drinking water treatment.

Titanium dioxide nanoparticles (TiO2 NPs) are inevitably present in the aquatic environment owing to their increasing production and use. However, knowledge of the potential effects of TiO2 NPs on the treatment of drinking water is scarce. Herein, the effects of two types of anatase TiO2 NPs (TP1, 25nm; TP2, 100nm) on the bacterial community in a biological activated carbon (BAC) filter were investigated via quantitative polymerase chain reaction (Q-PCR) analysis, ATP quantification, and 454 pyrosequencing analysis. Both TP1 and TP2 significantly inhibited the bacterial ATP level (p < 0.01) and induced a decrease in the abundance of bacterial 16S rDNA gene copies at doses of 0.1 and 100mgL(-1). Simultaneously, the diversity and evenness of the bacterial communities were considerably reduced. The relative abundances of bacteria annotated to OTUs from Nitrospira class and Betaproteobacteria class decreased upon TiO2 NP treatment, whereas those of Bacilli class and Gammaproteobacteria class increased. TiO2 NP size showed a greater effect on the bacterial composition than did the dose based on Bray-Curtis distances. These findings identified negative effects of TiO2 NPs on the bacterial community in the BAC filter. Given the fact that BAC filters are used widely in drinking water treatment plants, these results suggested a potential threat by TiO2 NP to drinking water treatment system.

Effect of titanium dioxide nanoparticles on zebrafish embryos and developing retina.

To investigate the impact of titanium dioxide nanoparticles (TiO2 NPs) on embryonic development and retinal neurogenesis. The agglomeration and sedimentation of TiO2 NPs solutions at different dilutions were observed, and the ultraviolet-visible spectra of their supernatants were measured. Zebrafish embryos were experimentally exposed to TiO2 NPs until 72h postfertilization (hpf). The retinal neurogenesis and distribution of the microglia were analyzed by immunohistochemistry and whole mount in situ hybridization. The 1 mg/L was determined to be an appropriate exposure dose. Embryos exposed to TiO2 NPs had a normal phenotype. The neurogenesis was initiated on time, and ganglion cells, cones and rods were well differentiated at 72 hpf. The expression of fms mRNA and the 4C4 antibody, which were specific to microglia in the central nervous system (CNS), closely resembled their endogenous profile. These data demonstrate that short-term exposure to TiO2 NPs at a low dose does not lead to delayed embryonic development or retinal neurotoxicity.

Titanium dioxide nanoparticles as carrier facilitate bioaccumulation of phenanthrene in marine bivalve, ark shell (Scapharca subcrenata)

To evaluate the impact of titanium dioxide nanoparticles (nTiO2) on the uptake of hydrophobic organic chemicals by marine bivalves, we conducted a comparative bioaccumulation study by exposing clam, Scapharca subcrenata, to phenanthrene (Phe) in the presence and absence of nTiO2. The large surface area of nTiO2 resulted in adsorption of co-existing Phe in aqueous solution to form nTiO2-Phe complexes. Accumulation of nTiO2 was not observed in clams at exposed concentration (500 μg/L) in this study. However, enhanced uptake of Phe by clams was observed in the presence of nTiO2, with ku and BAFs values being 2 and 1.7 times higher than that of Phe alone, respectively. The enhanced uptake can be explained by ingestion of nTiO2-Phe complexes into the gut and subsequent desorption of Phe there. Therefore, nTiO2 as a carrier facilitated the uptake of Phe by marine bivalves.

The impact of titanium dioxide nanoparticles on biological nitrogen removal from wastewater and bacterial community shifts in activated sludge

The potential impact of titanium dioxide nanoparticles (TiO2 NPs) on nitrogen removal from wastewater in activated sludge was investigated using a sequencing batch reactor. The addition of 2-50mgL(-1) of TiO2 NPs did not adversely affect nitrogen removal. However, when the activated sludge was exposed to 100-200mgL(-1) of TiO2 NPs, the effluent total nitrogen removal efficiencies were 36.5% and 20.3%, respectively, which are markedly lower than the values observed in the control test (80%). Further studies showed that the decrease in biological nitrogen removal induced by higher concentrations of TiO2 NPs was due to an inhibitory effect on the de-nitrification process. Denaturing gradient gel electrophoresis profiles showed that 200mgL(-1) of TiO2 NPs significantly reduced microbial diversity in the activated sludge. The effect of light on the antibacterial activity of TiO2 NPs was also investigated, and the results showed that the levels of TiO2-dependent inhibition of biological nitrogen removal were similar under both dark and light conditions. Additional studies revealed that different TiO2 concentrations had a significant effect on dehydrogenase activity, and this effect was most likely the result of decreased microbial activity.
