The Potential of Black Cumin (Nigella sativa) as an Immunostimulant in Aquaculture (Review)

Research on the use of black cumin (Nigella sativa) as an immunostimulant in aquaculture has shown interesting potential. In this review, we explore the potential of black cumin as an immunostimulant in aquatic animals. Based on a recent literature review, we highlight the positive effects of black cumin extract in enhancing immune responses in aquatic organisms. The active compounds in black cumin, such as thymoquinone and α-hederin, are thought to have the ability to stimulate the immune system, increase cytokine production, and increase phagocytic activity. Implications of using black cumin in aquaculture include the potential to improve the health and productivity of aquatic animals. However, challenges such as dose optimization, effectiveness in various aquatic species, and environmental risk management need to be addressed. Suggestions for further research include more in-depth studies of the mechanism of action, clinical trials on various aquatic species, as well as environmental impact assessments. With a careful and coordinated approach, the use of black cumin as an immunostimulant has the potential to increase sustainability and productivity in aquaculture practices.

Dietary administration impact of olive pulp on growth performance, metabolic profile, immune status, and antioxidant potential of gilthead seabream (Sparus aurata L.)

The use of natural immunostimulants in aquaculture is intended to foster overall health and bolster resilience against diseases in farmed fish populations. It constitutes a crucial strategy that can contribute to securing the sustainability of the aquaculture industry, and it is an area that warrants ongoing exploration and development. Therefore, the aim of the present work was to evaluate the effect of dietary administration of olive pulp on growth rate, metabolic profile, serum antioxidant potential, and humoral and cellular innate immune parameters of gilthead seabream (Sparus aurata L.). For this, fish were fed control diet or olive pulp enriched diets (50, 100, and 200 mg kg feed−1) for 4 weeks. Our results demonstrated that the highest inclusion level improved the growth rates and the biological antioxidant potential in the serum of fish. However, after 4 weeks of feeding, most of the assayed metabolic parameters (Ca2+, TP, ALB, K+, and Na+) were increased in the serum of fish fed with a diet containing the lowest level of olive pulp (50 mg kg feed−1). Regarding the innate immune parameters, the IgM levels decreased in the serum of fish fed 50′s diet after 2 and 4 weeks of trial. However, the serum of fish fed with diets containing 100 and 200 showed an increase in hemolytic complement activity after 2 weeks whilst this increase was only sustained in the 200′s group after 4 weeks. After 2 weeks of feeding, the serum of the fish showed an increase in peroxidase activity due to the highest olive inclusion. Concerning cellular innate parameters, peroxidase activity, respiratory burst, and phagocytic ability were increased in head-kidney leucocytes of fish fed 100′s diet at 2 weeks compared to values from control fish. These results suggest that the administration of olive pulp-enriched diets can benefit fish growth, antioxidant, and immune status of gilthead seabream.

The Potential of Fish Protein Hydrolysate Supplementation in Nile Tilapia Diets: Effects on Growth and Health Performance, Disease Resistance, and Farm Economic Analysis.

Fish protein hydrolysate (FPH) has shown immense potential as a dietary protein supplement and immunostimulant in aquaculture, especially in Nile tilapia production. Four isoproteic diets (30% crude protein) were prepared by including FPH at varying percentages (0%, 0.5%, 1%, and 2%). Nile tilapia fed with FPH diets for 90 days, and their growth performance, feed utilization, blood biochemistry, liver and gut morphology, and resistance against Streptococcus iniae were investigated. The findings revealed that diets physical attributes such as pellet durability index and water stability were remarkably (p < 0.05) varied between experimental diet groups. Furthermore, the test diets were more palatable when FPH was included at 1% and 2%. Fish that were fed with a 2% FPH-treated diet had significantly (p < 0.05) greater growth indices than other treatments. Additionally, their feed utilization was significantly (p < 0.05) improved. The experimental diets and intestinal total bacteria count (TBC) exhibited a rising trend with FPH levels, where the 2% FPH-treated diet recorded the highest TBC. Neutrophil (109/L), lymphocyte (109/L), eosinophil (109/L), and red blood cell(1012/L) counts were significantly (p < 0.05) higher in the 2% FPH-treated group, while the white blood cell (109/L), and basophil (109/L) counts were not influenced by the FPH inclusion. Moreover, the FPH-treated groups displayed lower creatinine, bilirubin, and urea levels than the control. The histological examination demonstrated that themid-intestine of 2% FPH-fed Nile tilapia had an unbroken epithelial wall, more villi with frequent distribution of goblet cells, wider tunica muscularis, and stronger stratum compactum bonding than other treatments. Additionally, this group exhibited more nuclei and erythrocytes and less vacuolar cytoplasm in liver than their counterparts. Nile tilapia that were given a diet containing 2% FPH had significantly (p < 0.05) higher resistance (83.33%) to S. iniae during the bacterial challenge test. A significant (p < 0.05) enhancement in farm economic efficiency was observed in the higher inclusion of FPH in diets. In summary, 2% FPH supplementation in Nile tilapia diets improved their growth performance, feed utilization, health status, disease resistance, and farm economic efficiency.

Aquaculture is an important industry that plays a role in meeting the world's animal protein needs. One of the main challenges in aquaculture is maintaining the health of cultivated aquatic organisms. Spirulina, a type of nutrient-rich blue-green microalgae, has garnered attention as a natural immunostimulant capable of enhancing the immune system of aquatic organisms. This article explains the immunostimulant properties of spirulina, including the bioactive compounds it contains, such as polysaccharides, proteins, photosynthetic pigments, and lipopolysaccharides. These compounds can boost the production of immune cells and phagocytic activity, which can help aquatic organisms combat pathogens. Furthermore, the article discusses the potential benefits of using spirulina extract in aquaculture, including increased resistance to diseases, faster growth, and improved nutritional quality of aquatic organisms. However, there are also challenges to address, such as determining the appropriate dosage and optimal application methods. This article contributes to the understanding of how spirulina extract can be utilized to enhance the health and productivity of aquatic organisms in aquaculture. With further research and the development of improved application methods, the potential of spirulina extract as an immunostimulant in aquaculture can be more fully realized, helping to meet the increasing global demand for protein.

Ketapang leaves have been of interest as a potential immunostimulant in aquaculture to boost the non-specific immune response in fish. In this review, various studies exploring the effects of ketapang leaf extract on the non-specific immune response of fish have been examined. The findings suggest that the administration of ketapang leaf extract can lead to an increase in non-specific immune parameters, such as phagocytosis activity, lysozyme activity, and total protein content. The potential use of ketapang leaf extract as an immunostimulant in aquaculture shows promising results in enhancing the non-specific immune response of fish. Strengthening the non-specific immune system can help fish combat various pathogens and diseases, thereby improving their overall health and survival in aquaculture settings. However, further research is needed to fully understand the mechanisms of action and optimal dosage of ketapang leaf extract as an immunostimulant. Additionally, considering its potential positive impact on fish health, the use of ketapang leaf extract in aquaculture warrants comprehensive studies to ensure its safe and effective implementation.

Potential Role of Plant Polysaccharides as Immunostimulants in Aquaculture – A Review

Abstract Aquaculture is one of the primary food-producing sectors in the world that ensures human nourishment. However, aqua farmers are facing serious problems due to disease outbreaks and development of antimicrobial resistance. Until now, chemical or antibiotic based strategies have been applied to control disease related concern in aquaculture. Frequent usage of antibiotics in feed or usage of disinfectant to overcome the disease may end up with negative impacts to the environment and human. Utilization of plant derived polysaccharides has been drastically increased due to their effective roles and could serve as a best replacement for chemical agents and antibiotics. In addition, plant derived compounds and plant extracts were utilized to improve the immunity, intestinal health and growth performance of aquaculturable organisms. In addition, large number of plant-based polysaccharides was utilized as immunostimulants in aquaculture. Hence, this review aims to highlight the multifunctional properties of plant-based polysaccharides in aquaculture. Moreover, advantages and different concentration of plant polysaccharides as a feed additive in aquaculture sector have been discussed herein.

Anti-microbial activity and immunomodulation of recombinant hepcidin 2 and NK-lysin from flounder (Paralichthys olivaceus)

Infections due to pathogens impact global aquaculture economy, where diseases caused by bacteria should be in particular focus due to antibiotic resistance. Hepcidin and NK-lysin are important innate immune factors having potential to be exploited as alternatives to antibiotics due to their antimicrobial activity and immunomodulatory capacity. In this study, the recombinant proteins of hepcidin 2 and NK-lysin (rPoHep2 and rPoNKL) from flounder (Paralichthys olivaceus) were obtained via a prokaryotic expression system. The results exhibited that rPoHep2 and rPoNKL killed both gram-negative and gram-positive bacteria mainly via attachment and disruption of the membrane. Interestingly, both peptides could bind to bacterial DNA. The antiviral assay showed that both peptides have antiviral activity against hirame nonvirhabdovirus. They exhibited no cytotoxicity to the mammalian and fish cell lines. PoHep2 was found localized in G-CSFR-positive peritoneal cells. Moreover, rPoHep2 significantly enhanced the phagocytosis of flounder leukocytes in vitro. These findings suggested that neutrophils contained rPoHep2 and may respond to the immunoreaction of neutrophils. In summary, both rPoHep2 and rPoNKL possess antimicrobial activities and may be exploited to replace traditional antibiotics. rPoHep2 possess immune regulatory functions, that can be further investigated as an immunostimulant in aquaculture.

Use of Pineapple (Ananas comosus) as an Immunostimulant in Aquaculture (Review)

The use of pineapple fruit extract as an immunostimulant in aquaculture has become the focus of significant research in efforts to improve the immune systems of aquatic organisms, especially farmed fish. Pineapple fruit extract, which contains bromelain, has attracted attention as a potential natural solution to improve the immune response of fish against pathogens and disease. The main aim of this review is to evaluate the extent to which the use of pineapple as an immunostimulant is effective in improving the immune system of fish or other aquatic organisms in an aquaculture context. This article uses literature study methods from several national, accredited national journals and reputable international journals. Literature study is the process of searching, collecting and analyzing various sources of information relevant to a particular topic. Pineapple extract contains bromelain, which has immunomodulatory and anti-inflammatory properties. It can improve the fish's immune response and reduce inflammation, helping to improve fish health. The use of pineapple fruit extract can reduce dependence on antibiotics in the treatment of sick fish, which is in line with global efforts to overcome antibiotic resistance and environmental impacts. With the right efforts, the use of pineapple fruit extract as an immunostimulant in aquaculture has the potential to increase the productivity and sustainability of the aquaculture industry, as well as reduce dependence on antibiotics and synthetic chemicals. Therefore, further research and development of best practices in the use of pineapple fruit extract in aquaculture are necessary steps to realize its potential as a more environmentally friendly and sustainable alternative.

PENGGUNAAN KULIT MANGGIS (GARCINIA MANGOSTANA) SEBAGAI IMUNOSTIMULAN DALAM BIDANG AKUAKULTUR (ARTIKEL REVIEW)

Mangosteen peel contains various bioactive compounds, such as xanthones, flavonoids, and tannins, which have attracted researchers' attention as potential natural immunostimulants to enhance the immune system of fish and other aquatic organisms. In this article, various recent studies examining the effects of mangosteen peel extract on the immune response of fish and aquatic organisms have been reviewed. Some research findings include increased phagocytosis activity, survival rate, and more active total hematocrit in species such as common carp (Cyprinus carpio), Nile tilapia (Oreochromis niloticus), whiteleg shrimp (Litopenaeus vannamei), and African catfish (Clarias gariepinus) treated with mangosteen peel extract. Furthermore, administering mangosteen peel extract can promote growth in aquatic animals. In conclusion, the use of mangosteen peel extract as an immunostimulant in aquaculture holds the potential to enhance the immune response and resilience of fish and aquatic organisms. Improved immune responses can help protect them from pathogens and diseases (such as Aeromonas hydrophila and Vibrio harvey), as well as increase survival and productivity in aquaculture systems. Although these research findings are promising, further studies are needed to better understand the detailed mechanisms of action of mangosteen peel as an immunostimulant, and to evaluate optimal dosages and potential side effects. The use of mangosteen peel as an immunostimulant could serve as an effective natural alternative to strengthen the immune system of fish and aquatic organisms, but more comprehensive research is necessary before widespread and safe implementation in aquaculture practices.

THE USE OF GARLIC (Allium sativum) AS AN IMMUNOSTIMULANT IN AQUACULTURE

The improvement of fish immunity is a critical factor in successful aquaculture. In recent years, the use of garlic (Allium sativum) as an immunostimulant has attracted researchers and farmers' interest in enhancing fish's health and disease resistance. This abstract discusses an overview of the use of garlic as an immunostimulant in aquaculture. Garlic is widely known for its antimicrobial, antioxidant, and immunostimulant properties. Several studies have evaluated the effects of garlic supplementation on various species of cultured fish. The results have shown that the administration of garlic in fish feed can enhance immune responses, such as increased immune enzyme activity and antibody production. Furthermore, active components in garlic, such as allicin and diallyl sulfide, have been proven effective against fish pathogens such as bacteria, viruses, and parasites. However, garlic as an immunostimulant in aquaculture still requires further research. Factors such as the appropriate dosage, interactions with farming conditions, and a deeper understanding of the underlying mechanisms must be considered. To optimise the use of garlic, further research is required to explore optimal dosages, interactions with environmental factors, and a better understanding of its effects on fish immune responses. In conclusion, garlic as an immunostimulant in aquaculture shows promising potential. The oral administration of garlic extract through the fish feed is capable of preventing infections by Aeromonas sp. and Aeromonas hydrophila in fish species such as Tawes, Catfish, Dumbo catfish, and Nile tilapia, while also enhancing non-specific immunity, such as increased total hematocrit, total haemoglobin, total erythrocytes, and phagocytic activity.

Identification and analysis of differentially expressed microRNAs in gibel carp Carassius auratus gibelio responding to polyinosinic-polycytidylic acid (poly I:C) stimulation

Polyinosinic-polycytidylic acid (poly I:C) is a synthesized analogue of viral double-strand RNA and considered as a potential immunostimulant in aquaculture. MicroRNAs (miRNAs) have been reported to play important roles in the development of the immune system and in regulation of host antiviral responses. In our earlier study, it was found that poly I:C pre-treatment could stimulate the resistance against cyprinid herpesvirus 2 (CyHV-2) infection and enhance the antiviral immune response in gibel carp. To understand the role of miRNAs in regulating the host response to poly I:C treatment, we investigated the expression profiles of miRNAs in the head kidney of poly I:C-treated gibel carp with small RNA sequencing technology. When compared with the untreated group, a total of 24 differentially expressed miRNAs were identified in the poly I:C-stimulated fish, among which, 7 and 17 miRNAs were upregulated and downregulated, respectively. Analysis of target genes of these differentially expressed miRNAs found that most targeted mRNAs were involved in catalytic activity, peptidase activity and endopeptidase activity, and were enriched in the metabolic, protein processing in endoplasmic reticulum and oxidative phosphorylation signaling pathways, suggesting that poly I:C could alter the expression of metabolism-related miRNAs in the kidney of gibel carp. Besides, it was noted that some immune-related miRNAs, including inflammation-related miRNAs (miR-192 and miR-731) and interferon-related miRNAs (miR-194a and miR-122), were downregulated after poly I:C treatment. In summary, it was found that poly I:C could regulate the cellular levels of specific miRNAs involved in metabolism and immune responses in the head kidney of gibel carp, which may increase the capacity of the immune cells to fight against pathogens infection.

Koumine induces apoptosis in Cyprinus carpio liver cells by regulating JAK-STAT and p53 signaling pathways

Koumine is an alkaloid with significant anti-anxiety, anticancer cell proliferation, and analgesic activities, and our previous studies have shown that koumine can be used as an immunostimulant in aquaculture, but the molecular mechanism of its effect remains unclear. We fed a basal diet with 0, 0.2, 2, and 20 mg/kg koumine to C. carpio for 10 weeks, and comprehensive studies of the histological and biochemical parameters and transcriptomes of the four groups were performed. Histological results indicated that the number of apoptotic cells in the liver increased with increasing koumine concentration. Compared with those of the control group, the malondialdehyde, superoxide dismutase, catalase, acid phosphatase, alkaline phosphatase, and lactate dehydrogenase levels of the treatment group increased to varying degrees. In total, 100.11 GB of clean data, 4774 DEGs, and 138 differentially expressed genes were obtained from the transcriptome data. Differentially expressed genes were classified into 187 signalling pathways, and the circadian rhythm signalling pathway, the JAK-STAT signalling pathway, the p53 signalling pathway and the PPAR signalling pathway were the top enriched pathways. The qRT-PCR results confirmed that the key genes ifnar1, socs3l, epoa, ghra, cMyc, mcl-1, shisa4, and gtse1 involved in balancing cell proliferation and apoptosis were enriched in these pathways. We discovered that the JAK-STAT and p53 pathways are important targets of koumine. Such information contributes to a better understanding of the potential mechanism by which koumine regulates hepatic immunity as well as to lays the theoretical foundation for its application.

The purpose of this community service was to educate the application of probiotics and immunostimulants in aquaculture. This activity was carried out in Cot Darat village, SamatigaDistrict, West Aceh Regency. This activity is divided into three steps, namely: (1) Introduction and socialization of probiotics and immunostimulant; (2) practice to produce probiotics and the application techniques; and (3) the monitoring of fish health and growth during cultivation. The type of bacteria used in this probiotic is Lactobacillus bacteria and the immunostimulant ingredients are turmeric and curcumin. Lactobacillus, turmeric and curcuma are known can improve fish health in aquaculture. The results of this activity indicate that group members have understood the definition of probiotics and immunostimulants, their benefits and how to make and apply them in aquaculture. In addition, fish health and growth are increase during fish culture compare before the probiotics was applied. The differences that occurred during probiotics application in fish cultivation were seen that the fish became healthier, the growth was better, and fish mortality decreased.

Potential of Oral Nanoparticles Containing Cytokines as Intestinal Mucosal Immunostimulants in Pigs: A Pilot Study.

Simple SummaryAntibiotics are essential compounds to cope with bacterial infections. However, their inadequate and excessive use has triggered the rapid arising of antimicrobial-resistant bacteria. In this scenario, immunostimulants, which are molecules that boost the immune system, open up a new approach to face this problem, enhancing treatment efficacy and preventing infections by immune system response. Cytokines are central effector molecules of the immune system, and their recombinant production and administration in animals could be an interesting immune modulation strategy. The aim of this study was the development of a highly stable nanoparticle of porcine cytokines to achieve the immunostimulation of intestinal mucosa in piglets. The outcomes of the present study prove this approach is able to stimulate swine intestinal cells and macrophages in vitro and tends to modulate inflammatory responses in vivo, although further studies are required to definitively evaluate their potential in animals.Antimicrobial resistance is a global threat that is worryingly rising in the livestock sector. Among the proposed strategies, immunostimulant development appears an interesting approach to increase animal resilience at critical production points. The use of nanoparticles based on cytokine aggregates, called inclusion bodies (IBs), has been demonstrated as a new source of immunostimulants in aquaculture. Aiming to go a step further, the objective of this study was to produce cytokine nanoparticles using a food-grade microorganism and to test their applicability to stimulate intestinal mucosa in swine. Four cytokines (IL-1β, IL-6, IL-8, and TNF-α) involved in inflammatory response were produced recombinantly in Lactococcus lactis in the form of protein nanoparticles (IBs). They were able to stimulate inflammatory responses in a porcine enterocyte cell line (IPEC-J2) and alveolar macrophages, maintaining high stability at low pH and high temperature. In addition, an in vivo assay was conducted involving 20 piglets housed individually as a preliminary exploration of the potential effects of IL-1β nanoparticles in piglet intestinal mucosa after a 7 d oral administration. The treated animals tended to have greater levels of TNF-α in the blood, indicating that the tested dose of nanoparticles tended to generate an inflammatory response in the animals. Whether this response is sufficient to increase animal resilience needs further evaluation.

SCFAs improve disease resistance via modulate gut microbiota, enhance immune response and increase antioxidative capacity in the host

To evaluate the effects of dietary short chain fatty acids (SCFAs) on the intestinal health and innate immunity in crucian carp, a six-week feeding trial was carried out with following treatments: basal diet (BD), basal diet supplementation with 1% sodium acetate (BDSA), basal diet supplementation with 1% sodium propionate (BDSP) and basal diet supplementation with 1% sodium butyrate (BDSB). The results showed dietary BDSA, BDSP and BDSB could protect the host against oxidative stress by improving the activity of certain antioxidative enzymes (T-SOD, GSH-Px and CAT). Additionally, dietary SCFAs could enhance mucosal and humoral immune responses by improving certain innate immune parameters in serum and skin mucus productions (IgM, ACH50 and T-SOD). Furthermore, dietary BDSA and BDSP could up-regulate the expression of immune related genes (TNF-α, TGF-β and IL-8) and tight junction protein genes (occludin and ZO-1). Dietary BDSB could also elevate the expression of IL-8, TGF-β, ZO-1 and Occludin in the midgut. Although dietary differences of SCFAs didn't alter the α-diversity of the intestinal flora, they altered the core microbiota. Finally, the challenge trial showed that dietary basal diet supplementation with SCFAs could protect zebrafish against Aeromonas hydrophila. These results suggest that dietary SCFAs could improve innate immunity, modulate gut microbiota and increase disease resistance in the host, which indicated the potential of SCFAs as immunostimulants in aquaculture.

Gut Immune System and the Implications of Oral-Administered Immunoprophylaxis in Finfish Aquaculture.

The gastrointestinal immune system plays an important role in immune homeostasis regulation. It regulates the symbiotic host-microbiome interactions by training and developing the host’s innate and adaptive immunity. This interaction plays a vital role in host defence mechanisms and at the same time, balancing the endogenous perturbations of the host immune homeostasis. The fish gastrointestinal immune system is armed with intricate diffused gut-associated lymphoid tissues (GALTs) that establish tolerance toward the enormous commensal gut microbiome while preserving immune responses against the intrusion of enteric pathogens. A comprehensive understanding of the intestinal immune system is a prerequisite for developing an oral vaccine and immunostimulants in aquaculture, particularly in cultured fish species. In this review, we outline the remarkable features of gut immunity and the essential components of gut-associated lymphoid tissue. The mechanistic principles underlying the antigen absorption and uptake through the intestinal epithelial, and the subsequent immune activation through a series of molecular events are reviewed. The emphasis is on the significance of gut immunity in oral administration of immunoprophylactics, and the different potential adjuvants that circumvent intestinal immune tolerance. Comprehension of the intestinal immune system is pivotal for developing effective fish vaccines that can be delivered orally, which is less labour-intensive and could improve fish health and facilitate disease management in the aquaculture industry.

Feeding with poly(I:C) induced long-term immune responses against bacterial infection in turbot (Scophthalmus maximus)

Poly(I:C) is a kind of chemosynthetic double-stranded RNA (dsRNA) analogue which could act as TLR3 agonist and induce IFN production. It is widely applied in anti-virus treatment and immunoregulation, as well as vaccine adjuvant in farmed animals. However, whether poly(I:C) could activate innate immune response to defense against bacterial infection remains unclear. In this study, we established a feeding trial model with different dose of poly(I:C) in turbot larvae, then challenged with Edwardsiella piscicida after 3–7 weeks resting period. The results show that feeding turbot with poly(I:C) exhibited a stronger inflammatory response and antioxidant stress ability, and significantly elevated the survival rate within the decreased bacterial loads. Importantly, the bacterial infection-induced white feces in hindgut of turbot were significantly alleviated after poly(I:C) feeding, and this administration induced protection could last for about 7 weeks. Taken together, these findings indicate that feeding turbot with poly(I:C) could enhance a long-term intestinal mucosal immunity in response to bacterial infection, suggesting that poly(I:C) might be a promising immunostimulant in aquaculture.
