Gemini Surfactant Mediated Catansomes for Enhanced Singlet Oxygen Generation of Rose Bengal and Their Phototoxicity against Cancer Cells.

Photodynamic therapy (PDT) is an innovative technique for cancer treatment with minimal side effects, based on the use of a photosensitizer, oxygen, and light. Photosensitizers (PSs) have several limitations, that may limit their clinical use, like poor solubilization, self-aggregation, and lack of specific targeting, which can be addressed with the use of nanomaterials. Herein, a unique type of catansomes (CaSs) was prepared using a gemini imidazolium-based surfactant (1,3-bis[(3-octadecyl-1-imidazolio)methyl]benzene dibromide (GBIB) and a double chain surfactant, diaoctyl sodium sulfosuccinate or Aerosol OT (AOT). The formation of CaS GBIB/AOT was optimized in various ethanol/water (E/W) solvent ratios by employing a facile, quick, and most reliable solution-solution mixing method. The CaS was characterized by dynamic light scattering (DLS) and field emission gun scanning electron microscopy (FEG-SEM) techniques. The experimental results reveal that stable CaSs with a spherical shape were obtained at lower concentration (100 μM). Rose Bengal (RB), a PS of the xanthene family, was incorporated into these prepared CaSs, as proven by fluorescence spectroscopy, UV-visible absorption spectroscopy, and confocal laser scanning microscopy. Singlet oxygen (1O2) generation studies revealed the relevant role of the E/W solvent ratio as there was a 4-fold boost in the 1O2 production for GBIB/AOT in E/W = 50:50 and around 3-fold in E/W = 30:70. Also, the GBIB-rich 80:20 fraction was more efficient in increasing the 1O2 generation as compared to the AOT rich fraction (20:80). Further, their phototoxicity was tested in a water-rich solvent ratio (E/W = 30:70) against MCF-7 cells. Upon irradiation with a 532 nm laser (50 mW) for 5 min, RB@GBIB/AOT(20:80) fraction caused 50% decrease in the metabolic activity of MCF-7 cells, and RB@GBIB/AOT(80:20) fraction produced a maximum 85% decrease in cell viability. Furthermore, the enhancement in intracellular 1O2 generation by RB@GBIB/AOT, as compared to pure RB, was confirmed with singlet oxygen sensor green (SOSG). This new type of CaS based on gemini surfactants exhibiting a large amount of 1O2 generation, holds great interest for several applications, such as use in photomedicine in future.

Enhanced vitamin C skin permeation from supramolecular hydrogels, illustrated using in situ ToF-SIMS 3D chemical profiling

Vitamin C (ascorbic acid) is a naturally occurring, powerful anti-oxidant with the potential to deliver numerous benefits to the skin when applied topically. However, topical use of this compound is currently restricted by an instability in traditional formulations and the delivery and eventual fate of precursor compounds has been largely unexplored. Time of flight secondary ion mass spectrometry (ToF-SIMS) is an emerging technique in the field of skin research and offers detailed chemical analysis, with high mass and spatial resolution, as well as profiling capabilities that allow analysis as a function of sample depth. This work demonstrates the successful use of ToF-SIMS to obtain, in situ, accurate 3D permeation profiles of both ascorbic acid and a popular precursor, ascorbyl glucoside, from ex vivo porcine skin. The significant permeation enhancing effect of a supramolecular hydrogel formulation, produced from an amphiphilic gemini imidazolium-based surfactant, was also demonstrated for both compounds. Using ToF-SIMS, it was also possible to detect and track the breakdown of ascorbyl glucoside into ascorbic acid, elucidating the ability of the hydrogel formulation to preserve this important conversion until the targeted epidermal layer has been reached. This work demonstrates the potential of ToF-SIMS to provide 3D permeation profiles collected in situ from ex vivo tissue samples, offering detailed analysis on compound localisation and degradation. This type of analysis has significant advantages in the area of skin permeation, but can also be readily translated to other tissue types.

Supramolecular gels: using an amide-functionalized imidazolium-based surfactant

Studies on supramolecular gel aggregates via supramolecular interactions are very fascinating and impressive. The interactions of gelator with solvent and inorganic salt can functionalize, activate, or control the properties of supramolecular gels. A lot of work on this area has been done, but all these contributions are only at an early stage. In this work, we tuned the gel formation conditions and properties by using low-molecular-weight gelator (LMWGs), amide-functionalized imidazolium-based surfactant, viz., N-cetyl-N′-acetamido imidazolium bromide ([N-C16, N′-CONH2-Im]Br), in different solvents and with inorganic salt additives. Under chemical (Cu2+/H2O2) and physical (temperature) stimuli, gel-sol phase transition occurred in the gel formed from 2 wt% [N-C16, N′-CONH2-Im]Br in the solvent formamide (FM). Accompanied with increased mechanical strength of the gels, morphology alternation from initial straight stripe-shaped structures to curly belt-like textures was observed in the addition of CuBr2. From Small-angle X-ray scattering (SAXS) measurements, bilayer units with interdigitated hydrocarbon tails of the gelator were testified to appear in the gels. Comparative experiments demonstrate that the π-π interaction, H-bonding, and hydrophobic interaction between the gelator molecules are the main driving forces for the gelation. The low-cost gelator, facile preparation, and coordination-driven assembly process make the robust gel a good candidate in various applications, such as material templates and drug deliveries.

Micelle-vesicle-micelle transition in aqueous solution of anionic surfactant and cationic imidazolium surfactants: Alteration of the location of different fluorophores

The presence of different surfactants can alter the physicochemical behaviors of aqueous organized assemblies. In this article, we have investigated the location of hydrophobic molecule (Coumarin 153, C153) and hydrophilic molecule (Rhodamine 6G perchlorate, R6G) during micelle-vesicle-micelle transition in aqueous medium in presence of anionic surfactant, sodium dodecylbenzenesulfonate (SDBS) and cationic imidazolium-based surfactant, 1-alkyl-3-methylimidazolium chloride (CnmimCl; n=12, 16).Initially, the physicochemical properties of anionic micellar solution of SDBS has been investigated in presence of imidazolium-based surfactant, CnmimCl (n=12, 16) in aqueous medium by visual observation, turbidity measurement, zeta potential (ζ), dynamics light scattering (DLS), and transmission electron microscopy (TEM). Zeta potential (ζ) measurement clearly indicates that the incorporation efficiency of C16mimCl in SDBS micelle is better than the other one due to the involvement of strong hydrophobic as well as electrostatic interaction between the two associated molecules. Turbidity and DLS measurements clearly suggest the formation of vesicles over a wide range of concentration.Finally, the rotational motion of C153 and R6G has also been monitored at different mole fractions of CnmimCl in SDBS-CnmimCl (n=12, 16) solution mixtures. The hydrophobic C153 molecules preferentially located in the bilayer region of vesicle, whereas hydrophilic R6G can be solubilized at surface of the bilayer, inner water pool or outer surface of vesicles. It is observed that rotational motion of R6G is altered significantly in SDBS-CnmimCl solution mixtures in presence of different mole fractions of CnmimCl. Additionally, the translational diffusion motion of R6G is monitored using fluorescence correlation spectroscopy (FCS) techniques to get a complete scenario about the location and translational diffusion of R6G.

Tailoring self-assembly behavior of a biological surfactant by imidazolium-based surfactants with different lengths of hydrophobic alkyl tails

Efficient and selective removal of dyes using imidazolium-based supramolecular gels.

A supramolecular gel was constructed by using an imidazolium-based surfactant, N-cetyl-N'-carboxymethyl imidazolium bromide ([N-C16, N'-CO2H-Im]Br), in the DMSO/H2O binary solvent mixtures and investigated as an adsorbent for removing dyes from aqueous solution. The self-assembled gel displays a morphology of microplatelets stacked in bilayer units with interdigitated hydrocarbon tails, and the structure remains unchanged below the sol-gel transition temperature. The gel also exhibits a strong birefringence property and excellent mechanical strength. In particular, the gels show superior performance in removal of anionic dye molecules, for example, removing 80% of eosin Y within 10 min, The constructed gels also present excellent salinity tolerance, even when the concentration of NaCl is 1000 times higher than that of the dye, and can maintain their high efficiency after 25 cycles, indicative of their promise in water treatment.

Self-assembly of imidazolium-based surfactants in magnetic room-temperature ionic liquids: binary mixtures.

The phase behaviour of binary mixtures of ionic surfactants (1-alkyl-3-imidazolium chloride, C(n)mimCl with n=14, 16 and 18) and imidazolium-based ionic liquids (1-alkyl-3-methylimidazolium tetrachloroferrate, C(n)mimFeCl4, with n=2 and 4) over a broad temperature range and the complete range of compositions is described. By using many complementary methods including differential scanning calorimetry (DSC), polarised microscopy, small-angle neutron and X-ray scattering (SANS/SAXS), and surface tension, the ability of this model system to support self-assembly is described quantitatively and this behaviour is compared with common water systems. The existence of micelles swollen by the solvent can be deduced from SANS experiments and represent a possible model for aggregates, which has barely been considered for ionic-liquid systems until now, and can be ascribed to the rather low solvophobicity of the surfactants. Our investigation shows that, in general, C(n)mimCl is a rather weak amphiphile in these ionic liquids. The amphiphilic strength increases systematically with the length of the alkyl chain, as seen from the phase behaviour, the critical micelle concentration, and also the level of definition of the aggregates formed.

Small-Angle Neutron-Scattering Studies of Mixed Micellar Structures Made of Dimeric Surfactants Having Imidazolium and Ammonium Headgroups

Planar imidazolium cation based gemini surfactants [16-Im-n-Im-16], 2Br(-) (where n = 2, 3, 4, 5, 6, 8, 10, and 12), exhibit different morphologies and internal packing arrangements by adopting different supramolecular assemblies in aqueous media depending on their number of spacer methylene units (CH(2))(n). Detailed measurements of the small-angle neutron-scattering (SANS) cross sections from different imidazolium-based surfactant micelles in aqueous media (D(2)O) are reported. The SANS data, containing the information of aggregation behavior of such surfactants in the molecular level, have been analyzed on the basis of the Hayter and Penfold model for the macro ion solution to compute the interparticle structure factor S(Q) taking into account the screened Coulomb interactions between the dimeric surfactant micelles. The characteristic changes in the SANS spectra of the dimeric surfactant with n = 4 due to variation of temperature have also been investigated. These data are then compared with the SANS characterization data of the corresponding gemini micelles containing tetrahedral ammonium ion based polar headgroups. The critical micellar concentration of each surfactant micelle (cmc) has been determined using pyrene as an extrinsic fluorescence probe. The variation of cmc as a function of spacer chain length has been explained in terms of conformational variation and progressive looping of the spacer into the micellar interior upon increasing the n values. Small-angle neutron-scattering (SANS) cross sections from different mixed micelles composed of surfactants with ammonium headgroups, 16-A(0), [16-Am-n-Am-16], 2Br(-) (where n = 4), 16-I(0), and [16-Im-n-Im-16], 2Br(-) (where n = 4), in aqueous media (D(2)O) have also been analyzed. The aggregate composition matches with that predicted from the ideal mixing model.

Imidazolium-Based Zwitterionic Surfactant: A New Amphiphilic Pd Nanoparticle Stabilizing Agent

Palladium nanoparticles (NPs) with an average size of 3.4 nm were prepared in water using imidazolium-based surfactant 3-(1-dodecyl-3-imidazolio)propanesulfonate (ImS3-12) as a stabilizer. The Pd NPs are highly dispersible in water and chloroform and were characterized by transmission electron microscopy, energy-dispersive X-ray spectroscopy, powder X-ray diffraction, and dynamic light scattering. The results indicate that in water the NP surface is covered with a double layer of ImS3-12 molecules. The NPs were effective in the aqueous biphasic hydrogenation of cyclohexene, with easy recycling and no loss of catalytic activity after four successive runs.

Surface active ionic liquids: Study of the micellar properties of 1-(1-alkyl)-3-methylimidazolium chlorides and comparison with structurally related surfactants

The impetus for the increasing interest in studying surface active ionic liquids (SAILs; ionic liquids with long-chain “tails”) is the enormous potential for their applications, e.g., in nanotechnology and biomedicine. The progress in these fields rests on understanding the relationship between surfactant structure and solution properties, hence applications. This need has prompted us to extend our previous study on 1-(1-hexadecyl)-3-methylimidazolium chloride to 1-(1-alkyl)-3-methylimidazolium chlorides, with alkyl chains containing 10, 12, and 14 carbons. In addition to investigating relevant micellar properties, we have compared the solution properties of the imidazolium-based surfactants with: 1-(1-alkyl)pyridinium chlorides, and benzyl (2-acylaminoethyl)dimethylammonium chlorides. The former series carries a heterocyclic ring head-group, but does not possess a hydrogen that is as acidic as H2 of the imidazolium ring. The latter series carries an aromatic ring, a quaternary nitrogen and (a hydrogen-bond forming) amide group. The properties of the imidazolium and pyridinium surfactants were determined in the temperature range from 15 to 75°C. The techniques employed were conductivity, isothermal titration calorimetry, and static light scattering. The results showed the important effects of the interactions in the interfacial region on the micellar properties over the temperature range studied.

Synthesis and evaluation of new imidazolium‐based aromatic ether functionalized cationic mono and gemini surfactants

Synthesis of a novel series of imidazolium-based surfactants, [(ROArCH2MIm)Br] with aromatic ether functionalized hydrophobic tail of varying chain lengths and also their gemini counterparts were conducted in the present work. Synthesis involves initial conversion of 4-hydroxy benzoic acid to its methyl ester followed by its O-alkylation with long chain fatty alcohols, which was subsequently reduced and brominated to get 4-alkyloxy benzyl bromide. For the synthesis of monocationic surfactant, 4-alkyloxy benzyl bromide is quaternized with N-methylimidazole. Gemini surfactants are synthesized by initial coupling of imidazole to 4-alkyloxy benzyl bromide followed by its quaternization with dibromoalkane. The surface properties of all the synthesized surfactants were determined using surface tensiometry. Within the same homologous series, expected decrease in critical micelle concentration (cmc) with the increase in hydrophobicity was observed for shorter chain homologs. However, the deviation in cmc value from regularity was observed when the number of carbon atoms in the hydrophobic chain exceeded a certain number. The cmc values of the geminis were found to be remarkably low compared to their monomeric counterparts.

Improved activity of enzymes in mixed cationic reverse micelles with imidazolium-based surfactants

This work reports the significant enhancement in performance of interfacially active enzymes, Chromobacterium viscosum (CV) lipase and horseradish peroxidase (HRP) in mixed reverse micelles of cetyltrimethylammonium bromide (CTAB) and imidazolium-based amphiphiles having varying tail lengths. Lipase activity in these mixed systems was always higher than that in the individual cationic reverse micelles of CTAB or any imidazolium surfactant, highest being observed in the mixed system of CTAB (50 mM) and 6 (1-tetradecyl-3-methyl imidazolium bromide, 40 mM)/water/isooctane/ n-hexanol (0.24 M), second-order rate constant, k 2 = 1301 ± 5 cm 3 g −1 s −1, ∼200% higher compared to that in CTAB and ∼65% more than the most popular AOT-microemulsion. Activity increased with concentration of imidazolium surfactant and also with its alkyl tail length. To have a more profound view on the structure–activity relationship, CTAB was replaced by cetyltriethylammonium bromide (CTEAB) and cetyltripropylammonium bromide (CTPAB) with subsequent increase in the headgroup size. The generalized influence of these mixed cationic systems on surface-active enzyme was also verified using HRP, where the activity improved ∼100%. This enhancement in enzyme activity is presumably due to the activating effect of the imidazolium cation in the enzymatic reactions by improving the nucleophilicity of interfacial water in vicinity of enzyme through hydrogen bonding.

Use of a Polyhedral Oligomeric Silsesquioxane (POSS)-Imidazolium Cation as an Organic Modifier for Montmorillonite

Recent studies on organically modified clays (OMCs) have reported enhanced thermal stabilities when using imidazolium-based surfactants over the typical ammonium-based surfactants. Other studies have shown that polyhedral oligomeric silsesquioxanes (POSS) also improve the thermal properties of composites containing these macromers. In an attempt to utilize the beneficial properties of both imidazolium surfactants and POSS macromers, a dual nanocomposite approach to prepare OMCs was used. In this study, the preparation of a new POSS-imidazolium surfactant and its use as an organic modifier for montmorillonite are reported. The purity, solubility, and thermal characteristics of the POSS-imidazolium chloride were evaluated. In addition, several OMCs were prepared by exchanging the Na+ with POSS imidazolium cations equivalent to 100%, 95%, 40%, 20%, and 5% of the cation exchange capacity of the clay. The subsequent OMCs were characterized using thermal analysis techniques (DSC, SDT, and TGA) as well as 29Si NMR to determine the POSS content in the clay interlayer both before and after thermal oxidation degradation. Results indicate the following: (1) the solvent choice changes the efficiency of the ion-exchange reaction of the clay; (2) self-assembled crystalline POSS domains are present in the clay interlayer; (3) the d-spacing of the exchanged clay is large (3.6 nm), accommodating a bilayer structure of the POSS-imidazolium; and (4) the prepared POSS-imidazolium exchanged clays exhibit higher thermal stabilities than any previously prepared imidazolium or ammonium exchanged montmorillonite.
