Influence of Prebiotic Immunobeta and the Combination Immunobeta + Zoovit Probiotic on Blood Biochemical Parameters in Ile-de-France Lambs

The aim of the present study was to determine the effect of the addition of prebiotic Immunobeta and probiotic Zoovit on the biochemical parameters of blood in Ile-de-France lambs. The research was carried out at the Agricultural Institute - Stara Zagora. It includes a total of 45 Ile de France lambs, divided into three groups of 15 - one control and two experimental. The groups were formed by the method of analogues, equalized by live weight at the beginning of the experiment, type of birth and sex. The animals of the I experimental group received 8 g of the prebiotic Immunobeta individually once a day, and those of the II experimental group the same amount of prebiotic with the addition of 4 g of the Zoovit probiotic. Blood for the study of 8 animals from each group was taken at the beginning and at the end of the experiment. Evaluation of efficacy and safety of simplified procedure for the percutaneous closure of patent foramen ovale: the Ile De France Foramen Ovale Permeable(IDFFOP) prospective, multicenter cohort study

Abstract The superiority of device closure over medical therapy for the prevention of recurrent stroke in patients with cryptogenic stroke and patent foramen ovale (PFO) was reported in several meta –analyses of randomized controlled trials. The growing number of PFO procedures requires to find alternatives to conventional guidance by transoesophageal echocardiography (TOE) needing general anesthesia. The aim of our study was to assess prospectively the efficacy and safety of a simplified procedure based on trans thoracic echocardiography (TTE) and local anesthesia rather than TOE during PFO closure. Methods Design and study oversight. The IDFFOP study was a French prospective, multicenter cohort study performed in four centers expert in TTE-guided device deployment. All patients had a stroke history, a PFO and a large or moderate right to left shunt defined by the number of microbubbles in the left atrium ( large shunt > 30, moderate : 10 to 30 microbubbles). A(Risk of Paradoxical Embolism) score>5 was required to confirm PFO imputability in stroke history. The primary end point was efficacy of TTE during the transcatheter PFO closure defined as the successful implantation of the device without malposition, without need of TOE, and absence of significant residual shunt at 3-month TTE control. Safety was defined as the absence of any periprocedural complications. Results 280 patients were enrolled from January 2018 to October 2020 ( male sex : 59.5%; Average age 57 years old ). Average ROPE score was 7; Cumulative immediate procedural success with the TTE-guided method was 99%, without any device embolization. However, in 2 patients, TOE was needed to guide device deployment. There was no severe vascular complication, or any ischaemic event during or after the procedure. 6 post-procedure groin hematomas occurred but none needed anyblood transfusion .One patient experienced a transient periprocedural atrial fibrillation. At 3 months there were only 8 (2,8%) residual right to left shunts (3 large, 5 moderate). Conclusion A simplified procedure for percutaneous PFO closure based on TTE and local anesthesia is safe and effective. Randomized controlled trials should be performed to confirm these results.

Efeito do uso da Promixalicina no controle de verminose e parâmetros sanguíneos de ovelhas

Sheep producers are increasingly looking for strategies to tackle the problem of worms in animals, as well as to improve the functioning of the animals' immune system. Therefore, two experiments were carried out, in two different properties (with high and low challenges in relation to verminosis) with the objective of, in both, evaluating the effect of using Ovine Promixalicin on OPG (Eggs per gram of Feces) and blood parameters of pregnant and non-pregnant sheep, on two different properties. The indication for use of the product is to, among others, contribute to the control of verminosis. The animals evaluated on the properties came from a pasture production system, with the use of concentrated and/or roughage supplementation if necessary. The animals used were of the Dorper, Texel, Ile de France and Sulfolk breed. The parameters evaluated were OPG, Famacha©, Body Condition Score and blood parameters (lymphocytes, leukocytes and erythrocytes). In relation to experiment one, with a low challenge in relation to worms, the use of Promyxalicin significantly helped to reduce worms and animal well-being, thus helping the herd to increase productive parameters such as weight gain and fertility. In experiment 2, a system with a high challenge in terms of verminosis, the animals remained uniform and controlled by the use of Promyxalicin, including beneficial results in relation to blood parameters, which did not increase during its use. Therefore, the use of Ovine Promixalicin does not alter the consumption of food by animals, as well as being effective in controlling verminosis, including Haemonchus contortus, Trichostrongylus colobriformis, Oesophagostomum columbianum e Strongyloides papillosus, promoting well-being and maintenance of the immune system, especially in pregnant animals, bringing benefits to the sheep production system.

Evaluating young people's listening practices: a public health issue

Listening to music through headphones or earphones has become commonplace with the development of new technologies. If this listening is prolonged over long periods and at high noise levels, it exposes young people to risks to their hearing systems. The WHO (2021) indicates that one person in four is expected to have hearing problems by 2050, and 1.1 billion young people worldwide could be at risk of hearing loss due to dangerous listening habits. Three French organizations (Le RIF, Bruitparif and CidB) have joined forces, with the support of the regional health agency of Ile de France, to coordinate their hearing risk prevention campaign and carry out a study ahead of the campaign to find out about listening practices using headphones or earphones, concert attendance and the precautions taken by teenagers to protect themselves from hearing risks. The results, based on a sample of 2,685 teenagers (secondary school), will provide a snapshot of practices so that prevention messages can be adapted during awareness campaigns.

Performance, ruminal and fecal microbiome of lambs fed diets supplemented with probiotics.

The present study aimed to investigate the impact of adding two doses of a commercial probiotic on productive performance, ruminal and fecal microbiome in growing lambs. Forty-two Texel or Ile de France crossbred lambs aged 86.9 ± 8.0days (body weight: 27.4 ± 3.7kg) were distributed into three groups: basal diet without probiotic supplementation (CG); basal diet + 1g/animal/day of probiotic (GP1) and basal diet + 5g/animal/day of probiotic (GP5). The experimental period was 84days. The weight was evaluated weekly and dry matter intake (DMI) and leftovers were measured daily. At the end of the experiment, lambs were slaughtered. Feces and rumen fluid were collected for microbiome analysis and rumen fragments for histological evaluation. The use of probiotics did not affect weight gain, but GP1 showed a higher silage and DMI intake than CG (p < 0.001). The CG had a greater thickness of keratinized epithelium and stratum corneum (< 0.001) than GP1 and GP5, and greater total papilla width (p = 0.039) than GP1. There was no difference in the general abundance in the rumen and fecal microbiomes. GP5 had a higher proportion of Azoarcus and Dialister taxa in the rumen fluid (p = 0.012 and p = 0.017, respectively) and higher proportion of Treponema and Fibrobacter taxa in the fecal microbiome (p = 0.015 and p = 0.026, respectively), whereas CG had a higher proportion of Anaeroplasma than the other groups (p = 0.032). These results demonstrated the benefits of probiotics for ruminal epithelium protection and microbial diversity. However, there was no effect on performance parameters.

Immunohistochemistry analyses of the abomasal mucosa show differences in cellular-mediated immune responses to Haemonchus contortus infection in resistant and susceptible young lambs

Haemonchus contortus is known for its high pathogenicity in sheep, and the uncontrolled use of anthelmintics resulted in the emergence of multiple drug-resistant populations. Breeding sheep for gastrointestinal nematode resistance is a sustainable alternative to reduce dependence of anthelmintic drugs, and differences in the degree of resistance between breeds have been reported. Here we compare two sheep breeds (Santa Ines and Ile de France), concerning the differences in innate and adaptive immune response involved in the resistance against H. contortus infection. Immunohistochemical analyses of the abomasum were conducted in naïve Santa Ines (n = 14) and Ile de France (n = 12) lambs randomized into four groups: infected Santa Ines (n = 8), non-infected control Santa Ines (n = 6), infected Ile de France (n = 8), and non-infected control Ile de France (n = 4). The infected lambs were initially infected with H. contortus infective larvae at 14 days of age, and multiple infections were conducted every second day until they reached 66 days of age. There was a significant effect (P < 0.001) of the infection with increase in numbers of CD3+ T; CD79α+ B; GATA3+ Th2/ILC2; POU2F3+ tuft cells; FOXP3+ T reg; and IgE + cells in the fundus of the abomasal mucosa in both Santa Ines and Ile de France lambs. Nevertheless, the infected Santa Ines lambs presented the highest averages for CD79α+ B; GATA3+ Th2/ILC; IgE + cells; and POU2F3+ tuft cells and there was a significant association of the breed and infection status with regards to POU2F3+ tuft cells, with the highest mean in the infected Santa Ines group. The infected Santa Ines group had three lambs with high degree of resistance and five lambs that showed a moderate infection. Our results suggest a mechanism of synergistic coordination between different immune-cell types in promoting resistance of suckling lambs under H. contortus infection.

Does habitat modification impact morphology, performance, and inflammatory responses in an amphibian with limited dispersal capacity (Lisssotriton helveticus)?

The environment of an organism exerts selective pressures that affect mobility, feeding, reproduction as well as predator-prey and conspecific interactions. Land use changes induced by human activities modify these selective pressures and may result in the adaptation of organisms. Amphibians are ectotherms that typically show a biphasic life cycle with an aquatic and terrestrial phase, which makes them particularly sensitive to environmental change. We studied the impact of habitat modifications on palmate newt populations in the Ile de France region across four types of habitats: urban, mixed, agricultural, and natural with at least two replicates for each habitat type. We measured the morphology of newts using callipers, quantified maximal running and swimming speed and acceleration using high-speed video recordings, and quantified the swelling of the hind limb linked to an inflammatory reaction. Our results show that in urban habitats, newts are larger and heavier and have a better body condition. Females, moreover, have a larger head in natural habitats, possibly due to diet specialisation of females during the breeding season. In mixed and agricultural habitats, newts have longer limbs and show a tendency to run faster, possibly associated with the selective pressures on movement in mixed habitats. Differences in inflammatory responses were observed between sexes but not habitat types. Overall, our results show differences in morphology and trends for differences in performance in newts living in different habitats suggesting that animals are adapting to human-induced changes in their environment.

Exploration of effects of galvanic vestibular stimulation on circadian rhythms and its associations with sleep and spatial memory in patients with breast cancer: The ICANSLEEP-2 protocol.

Patients with breast cancer (BC) exhibit circadian rhythm disruptions, mainly of rest-activity rhythm (RAR), of which sleep is an essential component, and cortisol rhythm. Sleep complaints such as insomnia and cognitive impairments are prevalent in BC. In general population, sleep is known to contribute greatly to cognition. Thus, improving RAR (and particularly sleep) could help limiting cognitive impairments in BC patients. It has recently been suggested that, in addition to its essential role in spatial memory, the vestibular system contributes to RAR synchronization. Its stimulation could therefore limit both sleep disturbances and spatial memory deficits in BC. The main aim of the ICANSLEEP-2 study is to assess the effects of galvanic vestibular stimulation (GVS) on circadian rhythms. The secondary aim is to assess whether GVS improves sleep and spatial memory in BC patients. Two groups with insomnia complaints (Insomnia Severity Index > 7) will be included: a patients' group with BC (n = 50) and a healthy control group without history of cancer (n = 25). There will be two assessment sessions, before and after 2 weeks of GVS. Patients will be randomly assigned to either a GVS group or a sham group (noneffective stimulation). Controls will receive GVS. GVS effects will be quantified and compared between groups. Assessments will include actigraphy, salivary cortisol, polysomnography, a cognitive test battery (including a computer-based task for spatial memory) and validated questionnaires (for psychological functioning and sleep complaints). Current methods for improving sleep in BC have had controversial outcomes regarding sleep structure. We expect GVS to offer a new mean of directly targeting RAR disruptions in BC patients, with beneficial effects on sleep structure. Given the crucial impact of sleep on cognitive functioning, notably spatial memory, improving sleep of BC patients should enhance their cognitive functioning. This study received ethical approval from the Ile de France IV institutional review board on 19 April 2022 (no. ID-RCB: 2022-A00437-36). HPV-driven oropharyngeal cancer burden in Paris and its region (ILE DE FRANCE) from 1981 TO 2021

BackgroundFrance has the sixth highest incidence of oropharyngeal cancer (OPC) in Europe, but the epidemiological impact of high-risk HPV (HR-HPV) remains poorly documented. The objective of our study was to assess the proportion of OPCs caused by HR-HPV in Paris, and its suburbs, over the four past decades. This area accounts for almost one-fifth of the total population of France. MethodsOPCs diagnosed in 1981, 1986, 1991, 1996, 2001, 2006, 2011, 2016 and 2020/2021 in two of the main referral cancer centers for HNCs in Paris and its suburbs were retrieved from the tumor biobanks. HPV status was determined by p16-staining and HPV-DNA detection. Samples were considered HPV-driven if both assays were positive. Results were compared to the French cancer registry data. ResultsSamples from 697 OPC patients were assessed (including 82 % of all samples diagnosed in 2001, 2006, 2011, 2016, 2021). The proportion of HPV-driven cases rose from 2.7 % to 53 % between 1981 and 2021. HPV16 was the dominant genotype during the study period. Of patients with HPV-driven OPC, 81 % were male and 42 % were smokers versus 80 % and 92 % in their HPV-negative counterparts. The age of OPC patients increased significantly, during the study period, independent of their HPV status ConclusionThe proportion of HPV-driven OPCs has significantly increased in Paris and its suburbs, during the last four decades. OPCs has become the 2nd predominant type of head and neck cancer, in France. This may be linked to the rise in HPV-driven cases and the decrease of tobacco and alcohol consumption in men.

MEM&SO protocol: understanding the determinants of social learning in neurodegenerative diseases

BackgroundPeople with neurodegenerative diseases may have difficulty learning new information, owing to their cognitive impairments. Teaching them techniques for learning in social contexts could alleviate this difficulty. The present study will examine the performances of patients with Alzheimer’s disease and patients with the semantic variant of primary progressive aphasia on a memory test administered in three social contexts. The protocol will make it possible to identify determinants of social interactions, social abilities, cognition, and personality that can explain the potentially beneficial effect of social context on learning in these patients.MethodsThirty dyads (patient with primary memory impairment who meets criteria for Alzheimer’s disease paired with caregiver), 16 dyads (patient meeting criteria for semantic variant of primary progressive aphasia paired with caregiver), and 46 dyads (healthy controls with no cognitive complaints) will be recruited. A nonverbal memory test (social memory task) will be administered to each dyad in three different social contexts (presence-only, observation, collaboration). Patients and healthy controls will also undergo a neuropsychological assessment to measure social (interactions and abilities), cognitive and personality aspects. Patients will be compared with controls on differential social scores calculated between the presence-only and collaboration contexts, and between the presence-only and observation contexts. A multiple comparative case study will be conducted to identify social, cognitive and personality variables that potentially explain the differential scores in the collaboration and observation contexts.DiscussionFor the first time, memory will be assessed in patients with Alzheimer’s disease and patients with the semantic variant of primary progressive aphasia in three different contexts (presence-only, observation, collaboration). The multiple comparative case study will make it possible to identify the determinants of memory performance in the social context, in order to create the most beneficial learning context for individual patients, according to their profile.Trial registrationThis study was approved by the Ile de France XI institutional review board (2022-A00198-35), and registered on ClinicalTrials.gov (no. Age at death during the Covid-19 lockdown in French metropolitan regions: a non parametric quantile regression approach

BackgroundLockdowns have been implemented to limit the number of hospitalisations and deaths during the first wave of 2019 coronavirus disease. These measures may have affected differently death characteristics, such age and sex. France was one of the hardest hit countries in Europe with a decreasing east–west gradient in excess mortality. This study aimed at describing the evolution of age at death quantiles during the lockdown in spring 2020 (17 March—11 May 2020) in the French metropolitan regions focusing on 3 representatives of the epidemic variations in the country: Bretagne, Ile-de-France (IDF) and Bourgogne-Franche-Comté (BFC).MethodsData were extracted from the French public mortality database from 1 January 2011 to 31 August 2020. The age distribution of mortality observed during the lockdown period (based on each decile, plus quantiles 1, 5, 95 and 99) was compared with the expected one using Bayesian non-parametric quantile regression.ResultsDuring the lockdown, 5457, 5917 and 22 346 deaths were reported in Bretagne, BFC and IDF, respectively. An excess mortality from + 3% in Bretagne to + 102% in IDF was observed during lockdown compared to the 3 previous years. Lockdown led to an important increase in the first quantiles of age at death, irrespective of the region, while the increase was more gradual for older age groups. It corresponded to fewer young people, mainly males, dying during the lockdown, with an increase in the age at death in the first quantile of about 7 years across regions. In females, a less significant shift in the first quantiles and a greater heterogeneity between regions were shown. A greater shift was observed in eastern region and IDF, which may also represent excess mortality among the elderly.ConclusionsThis study focused on the innovative outcome of the age distribution at death. It shows the first quantiles of age at death increased differentially according to sex during the lockdown period, overall shift seems to depend on prior epidemic intensity before lockdown and complements studies on excess mortality during lockdowns.

“The least significant change on bone mineral density scan increased in patients with higher degrees of obesity”

Background: The least significant change (LSC) threshold of 0.03 g/cm² is used to interpret bone mineral density (BMD) scans in the general population. Our working hypothesis was that the current LSC threshold would not be applicable in obese populations. Aims: The aim of this study was to calculate the LSC in an obese population. Methods: We performed an interventional study among 120 obesity patients, in whom two measurements of BMD were performed at 3 sites. Pairs of measures were used to calculate the LSC, using the Bland and Altman method. Results: We calculated that the LSC was 0.046 g/cm² at the lumbar spine, 0.069 g/cm² at the femoral neck, and 0.06 g/cm² at the total hip. We also calculated the LSC for each class of obesity and observed an increase in LSC with increasing body mass index (BMI). We calculated a LSC of 0.05 g/cm² in patients with class 2 or class 3 obesity, whereas the LSC in patients with class 1 obesity is similar to the threshold used in the general population. Discussion: In obese population, like BMD, LSC is higher than the threshold value of the general population, and increases with increasing BMI.Conclusion: LSC of 0.05 g/cm² could be used in clinical practice in patients with class 2 or 3 obesity. These findings should help to improve the interpretation of BMD scans in these patients and optimize their management. OLYMPUS-POPGEN: A synthetic population generation model to represent urban populations for assessing exposure to air quality.

With the new individual- and activity-based approaches to simulating exposure to air pollutants, exposure models must now provide synthetic populations that realistically reflect the demographic profiles of individuals in an urban territory. Demographic profiles condition the behavior of individuals in urban space (activities, mobility) and determine the resulting risks of exposure and environmental inequalities. In this context, there is a strong need to determine the relevance of the population modeling methods to reproduce the combinations of socio-demographic parameters in a population from the existing databases. The difficulty of accessing complete, high-resolution databases indeed proves to be very limiting for the ambitions of the different approaches. This work proposes to evaluate the potential of a statistical approach for the numerical modeling of synthetic populations, at the scale of dwellings and including the representation of coherent socio-demographic profiles. The approach is based on and validated against the existing open databases. The ambition is to be able to build upon such synthetic populations to produce a comprehensive assessment of the risk of environmental exposure that can be cross-referenced with lifestyles, indicators of social, professional or demographic category, and even health vulnerability data. The approach implemented here is based on the use of conditional probabilities to model the socio-demographic properties of individuals, via the deployment of a Monte Carlo Markov Chain (MCMC) simulation. Households are assigned to housing according to income and house price classes. The resulting population generation model was tested in the Paris region (Ile de France) for the year 2010, and applied to a population of almost 12 million individuals. The approach is based on the use of census and survey databases. Validation, carried out by comparison with regional census data, shows that the model accurately reproduces the demographic attributes of individuals (age, gender, professional category, income) as well as their combination, at both regional and sub-municipal levels. Notably, population distribution at the scale of the model buildings remains consistent with observed data patterns. The outcomes of this work demonstrate the ability of our approach to create, from public data, a coherent synthetic population with broad socio-demographic profiles. They give confidence for the use of this approach in an activity-based air quality exposure study, and thus for exploring the interrelations between social determinants and environmental risks. The non-specific nature of this work allows us to consider its extension to broader demographic profiles, including health indicators, and to different study regions.

Wool production and quality traits of pure- and crossbred Merino-type sheep

Very little information is available on the quality of wool produced by terminal crosses out of wool producing dam lines. This study was therefore undertaken to elucidate the wool quality traits of four crossbred lines relative to Merinos and Dohne Merinos, which served as dam lines. Each dam line was mated to rams of their own breed as well as to Dormer or Ile de France rams to create four crossbred lines, namely, Dohne x Dormer, Dohne x Ile de France, Merino x Dormer, and Merino x Ile de France, in addition to the purebred Dohne Merino and Merino lines. Four rams and four ewes from each of these lines were reared up to one year of age under optimal growth conditions and shorn at the end of that time. Fleeces were weighed and samples collected for analysis. Neither sex nor genotype influenced clean fleece weight or clean yield percentage, but fibre diameter differed between genotypes. Purebred Merino had the finest wool (18.26 μm) and the Merino x Dormer cross the coarsest (26.01 μm). However, all lines still produced wool that could be used in manufacturing clothing, while fleeces showed good uniformity. The two purebred lines had the highest comfort factor (> 98%). The crossbred lines produced a similar quantity of wool as the purebreds, although of poorer quality. All genotypes except Merino x Dormer appear to produce wool that could be processed into garments, although the crossbred wool would only be suitable for outerwear.
