Association Between Prostatitis and Some Biochemical Markers in Patients of the AL-Najaf Province, Iraq

One of the most common clinical conditions in men is prostate disease, especially prostatitis, which causes various symptoms, that negatively affect patient life. Our current study investigated the relationship between prostatitis and IL-6, zinc, and ESR levels for patients who visited AL-Sader Medical City in the province of AL-Najaf, between March and November 2023. our study included 90 samples of people infected with prostatitis and 30 samples of healthy people between the ages of (20-40) years. The level of IL-6 was calculated using the ELISA technique, The level of zinc as an antioxidant asses by using the spectrophotometer and ESR was measured by using the Westergren technique to determine the inflammation case in patients and compared with the healthy group. The results showed an increase in the level of IL-6 in patients with prostatitis compared with a healthy group (908.72±140.19,214.16±85.56) pg/ml respectively. but showed a decrease in the level of Zinc in prostatitis patients compared with the healthy group (38.93±10.92U/ml, 89.36±9.61U/ml) respectively. Also showed an elevated level of ESR, where according to (48.41±13.49 ) mm/hour in patients group compared with a healthy group (14.60±4.14) mm/ hour. The data was analyzed by SPSS program version 23, where the factor effect was considered significant when p<0.05. we conclude from our current study the significant relationship between IL-6, Zinc levels, and prostatitis, as well as the role of the ESR test as an indicator of prostatitis

The FLARE Score and Circulating Neutrophils in Patients with Cancer and COVID-19 Disease.

Inflammation and neutrophils play a central role in both COVID-19 disease and cancer. We aimed to assess the impact of pre-existing tumor-related inflammation on COVID-19 outcomes in patients with cancer and to elucidate the role of circulating neutrophil subpopulations. We conducted a multicenter retrospective analysis of 524 patients with cancer and SARS-CoV-2 infection, assessing the relationship between clinical outcomes and circulating inflammatory biomarkers collected before and during COVID-19 infection. Additionally, a single-center prospective cohort study provided data for an exploratory analysis, assessing the immunophenotype of circulating neutrophils and inflammatory cytokines. The primary endpoints were 30-day mortality and the severity of COVID-19 disease. Prior to COVID-19, 25% of patients with cancer exhibited elevated dNLR, which increased to 55% at the time of COVID-19 diagnosis. We developed the FLARE score, incorporating both tumor- and infection-induced inflammation, which categorized patients into four prognostic groups. The poor prognostic group had a 30-day mortality rate of 68%, significantly higher than the 23% in the favorable group (p < 0.0001). This score proved to be an independent predictor of early mortality. This prospective analysis revealed a shift towards immature forms of neutrophils and higher IL-6 levels in patients with cancer and severe COVID-19 infection. A pre-existing tumor-induced pro-inflammatory state significantly impacts COVID-19 outcomes in patients with cancer. The FLARE score, derived from circulating inflammatory markers, emerges as an easy-to-use, globally accessible, effective tool for clinicians to identify patients with cancer at heightened risk of severe COVID-19 complications and early mortality who might benefit most from immediate and intensive treatment strategies. Furthermore, our findings underscore the significance of immature neutrophils in the progression of COVID-19 in patients with cancer, advocating for further investigation into how these cells contribute to both cancer and COVID-19 disease.

Evaluation of circulating microvesicles and their procoagulant activity in patients with COVID-19

ObjectiveSeveral pathological conditions trigger the formation of microvesicles (MVs), including infectious diseases such as COVID-19. The shedding of MVs increases the levels of inflammatory factors (e.g., interleukin-6; IL-6) and ultimately leads to an inflammatory cascade response, while also increasing the procoagulant response. The current study aimed to evaluate the level of circulating MVs and their procoagulant activity as well as the serum level of IL-6 in patients with COVID-19 and healthy controls. In this case-control study, 65 patients with COVID-19 and 30 healthy individuals were sampled after obtaining written informed consent. MVs counting was measured using conjugated CD61, CD45, CD235a, and Annexin-V antibodies. Additionally, the procoagulant activity of MVs and the IL-6 level were estimated using enzyme-linked immunosorbent assay (ELISA).ResultsThe majority of MVs were platelet-derived MVs (PMVs). Patients with COVID-19 had significantly higher levels of MVs, procoagulant MVs, and IL-6 compared to healthy controls (p < 0.001). MVs were significantly correlated with procoagulant MVs, D-Dimer levels, fibrinogen, and IL-6, but not with platelet, lymphocyte, and neutrophil counts.ConclusionElevated levels of procoagulant MVs and their association with inflammatory and coagulation markers in patients with COVID-19 are suggested as a novel circulatory biomarker to evaluate and predict the procoagulant activity and severity of COVID-19.

Relationship between serum metabolic indexes and immune function in patients with insomnia and their mechanism

We conducted a study to examine how certain serum metabolic markers (reactive oxygen species (ROS), homocysteine (Hcy), and reduced glutathione (GSH)) impact the levels of IL-4 in patients with insomnia. The study involved 60 insomnia patients, including 20 with primary insomnia and 20 with somatopathy insomnia, aged 23-84 years with a mean age of 61.20 ± 12.59 years and a mean disease duration of 6.97 ± 8.45 years. There were 20 normal controls, 11 males and 9 females, aged 26-63 years, with an average age of 49.95 ± 10.52 years. We measured ROS levels using immunofluorescence, Hcy levels using enzyme-linked immunosorbent assay, and GSH levels using ELISA. The IL-4 level in serum was also detected by ELISA to assess the patient's immune function. Our analysis revealed that changes in ROS, Hcy, and GSH levels were associated with changes in IL-4 levels in serum. Therefore, early detection of serum metabolic changes in insomnia patients and proactive intervention can help reduce susceptibility to various infections and tumors.

Analysis of the expression of cytokines and chemokines, platelet-leukocyte aggregates, sCD40L and sCD62P in cutaneous melanoma.

Cutaneous melanoma (CM) is a malignancy with a variable incidence worldwide and a poor advanced-stage prognosis. Melanoma growth is closely associated with the immune system. A cross-sectional study was performed on CM patients admitted at the Hospital de Cancer de Pernambuco (HCP) between 2015 and 2018. Fifty-one CM patients were included, and 30 healthy individuals. The study aimed to evaluate the association of platelet activation mechanisms and inflammatory response in patients with cutaneous melanoma. Elevated serum IL10 and low serum TNF levels in CM patients compared to controls (p < 0.05). High IL6 levels in patients with negative lymph nodes LN (-) compared to positive lymph nodes group (LN +, p = 0.0005). Low RANTES levels in patients compared to controls (p < 0.05). Elevated levels of platelet-lymphocyte (PLA), platelet-monocytes (PMA), and platelet-neutrophils (PNA) aggregates were observed in patients compared to controls (p < 0.05). CM patients with stage II had lower PMA levels than stages I and III (p < 0.05). High PMA levels were observed in patients with LN (+) compared to the LN (-) group (p < 0.0001). Patients with SSM had high levels of sCD40L and sCD62P compared to controls (p < 0.05)). High sCD40L levels in stage II compared to the stage III group, and sCD62P in stages I and II compared to the stage III group (p < 0.05). High sCD62P levels in patients with LN (-) compared to the group LN (+) (p < 0.05). It was observed the immunosuppressive profile in CM may favor tumor progression. High levels of platelet-leukocyte aggregates, sCD40L, and sCD62P may be associated with the worst prognosis.

The changes and the potential clinical applications of cytokines in Taiwan’s major venomous snakebites patients

BackgroundTaiwan habu (Protobothrops mucrosquamatus), green bamboo viper (Viridovipera stejnegeri), and Taiwan cobra (Naja atra) are the most venomous snakebites in Taiwan. Patients commonly present with limb swelling but misdiagnosis rates are high, and currently available diagnostic tools are limited. This study explores the immune responses in snakebite patients to aid in differential diagnosis. MethodsThis prospective observational study investigated the changes in cytokines in snakebite patients and their potential for diagnosis. ResultsElevated pro-inflammatory cytokines IL-6 and TNF-α were observed in all snakebite patients compared to the healthy control group. While no significant disparities were observed in humoral immune response cytokines, there were significant differences in IFN-γ levels, with significantly higher IL-10 levels in patients bitten by cobras. Patients with TNF-α levels exceeding 3.02 pg/mL were more likely to have been bitten by a cobra. ConclusionThis study sheds light on the immune responses triggered by various venomous snakebites, emphasizing the potential of cytokine patterns for snakebite-type differentiation. Larger studies are needed to validate these findings for clinical use, ultimately improving snakebite diagnosis and treatment.

Efficacy of non-surgical therapy on IL-6 levels in patients with chronic periodontitis: A systematic review

Chronic periodontitis-affected tissues have an increased production of pro-inflammatory cytokines relative to anti-inflammatory counterparts, including IL-6. Therefore, this systematic review aimed to synthesize and appraise the available literature to provide a comprehensive analysis of the efficacy of non-surgical therapy, specifically focusing on the therapeutic effect of IL-6 in patients with chronic periodontitis. This review focused on seven studies including 308 patients with moderate to severe chronic periodontitis across five different countries. The RoB assessment showed a moderate to low risk of bias studies. Moreover, the studies found a statistically significant decrease in IL-6 levels in patients after receiving non-surgical therapy. There were statistically significant alterations in periodontal indices and IL-6 levels over time, which showed the efficacy of non-surgical treatment based on various literature. Hence, decreasing IL-6 could serve as a therapeutic indicator of inflammatory response.

Altered serum concentrations of IL-8, IL-32 and IL-10 in patients with lung impairment 6 months after COVID-19

Post-COVID symptoms are reported in 10–35 % of patients not requiring hospitalization, and in up to 80 % of hospitalized patients and patients with severe disease. The pathogenesis of post-COVID syndrome remains largely unknown. Some evidence suggests that prolonged inflammation has a key role in the pathogenesis of most post-COVID manifestations. We evaluated a panel of inflammatory and immune-mediated cytokines in individuals with altered HRCT features and in patients without any long-term COVID symptoms. Blood samples of 89 adult patients previously hospitalized with COVID-19 were collected and stratified as patients with and without HRCT evidence of fibrotic lung alterations. Serum analyte concentrations of IL-4, IL-2, CXCL10 (IP-10), IL-1β, TNF-α, CCL2 (MCP-1), IL-17A, IL-6, IL-10, IFN-γ, IL-12p70 and TGF-β1 (free active form) were quantified by bead-based multiplex assay. Clinical and functional data were recorded in a database.With the use of machine learning approach, IL-32, IL-8, and IL-10 proved to be associated with the development of HRCT evidence of lung sequelae at follow-up. Direct comparison of cytokine levels in the two groups showed increased levels of IL-32 and decreased levels of IL-8 in patients with lung impairment. After further stratification of patients by severity (severe versus mild/moderate) during hospitalization, IL-10 emerged as the only cytokine showing decreased levels in severe patients. These findings contribute to a better understanding of the immune response and potential prognostic markers in patients with lung sequelae after COVID-19.

Determination of IL-6 Gene Promoter Polymorphism in Patients with Hepatitis C and Its Impact on RNA Secondary Structure.

Background and Objectives: A polymorphism in the promoter region of the IL-6 gene would influence the level of IL-6 expression in patients with HCV, resulting in a pro-inflammatory response. Few studies have shown the association between -174G>C (rs1800795) and -1363G>T (rs2069827) polymorphisms and HCV infection, and their results have been contradictory. There are no data published in our population to study such an IL-6 stimulus against HCV infection and its impact on RNA secondary structure. Therefore, we isolated human subjects from the province of Punjab, Pakistan. The objective was to screen for IL-6 gene promoter polymorphisms -174G/C and -1363G/T and those correlated with serum concentrations of IL-6 in patients with HCV and compared with a control. Materials and Methods: In conventional PCR, measurement of serum IL-6 by CLIA and statistical analysis were performed to observe the genotype association studies. By integrating bioinformatics and computational tools, our study aimed to provide a comprehensive understanding of how variations in the promoter region of IL-6 may have functional implications on gene expression. Results: The -174G>C and -1363G>T genotypes in the promoter region of patients with HCV were in strong allelic association (Δ = 0.97, p < 0.001). Interestingly, the bioinformatics analysis was well aligned with our experimental data. Conclusions: Based on the data, it can be inferred that IL-6 gene promoter polymorphisms are important in the dysregulation of IL-6 levels in patients with HCV.

Thyroid dysfunction in women with metabolic syndrome and type 2 diabetes

The most common endocrinopathies are pathology of the thyroid gland and diabetes. Thyroid hormones affect glucose metabolism in diabetes, just as diabetes affects thyroid functionObjective. The aim of this study was to explore the relationship between components of the metabolic syndrome, thyroid function, and IL-6 levels in patients with type 2 diabetes.Material and methods. We examined 65 women with type 2 diabetes (average age – (45±0.88) years), and 30 practically healthy women of the control group (average age – (44.14 ± 0.74) years). All patients underwent clinical, anthropometric and biochemical examination. The level of thyroid-stimulating hormone, free thyroxine, free triiodothyronine, antibodies to thyroperoxidase, glucose, glycated hemoglobin, HOMA index, total cholesterol, triglycerides, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol were determined. The research was carried out on a Cobas 6000 biochemical analyzer using Roche reagents. In all patients, the level of IL-6 was determined by the method of solid-phase enzyme immunoassay.Results. Among all examined, 19 (29.2%) women were found to have subclinical hypothyroidism on the background of an autoimmune process, their average TSH level was within 4.85±0.21 mU/l, ATPO –572.89±12.29 IU /ml, 9 (13.8%) had hypothyroidism on the background of an autoimmune process, their average TSH level was within 10.10±0.19 mU/l, ATPO –823.76±11.91 IU/ml, and 12 (18.4%) people were carriers of antithyroid antibodies, their average TSH level was within 1.76±0.26 mU/l, ATPO –368.81±14.26 IU/ml. It was found that the average level of IL-6 in patients with type 2 diabetes and AIT was 21.76±0.19 pg/ml, in patients with type 2 diabetes, AIT and SCH – 24.85±0.21 pg/ml, in patients with type 2 diabetes, AIT and HT – 27.43±0.24 pg/ml, patients with type 2 diabetes with normal thyroid function – 18.45±0.19 pg/ml, control group patients - 5,74±1.12 pg/ml. Conclusions. In patients with autoimmune thyroiditis and type 2 diabetes, a relationship of medium strength was found between IL-6 and OT (r=0.32; p&lt;0.05), IL-6 and BMI (r=0.34; p&lt; 0.05), IL-6 and HbA1c (r=0.31; p&lt;0.05), IL-6 and HOMA-IR (r=0.35; p&lt;0.05), IL-6 and TC ( r=0.31; p&lt;0.05) weak correlation between IL-6 and LDL (r=0.21; p&lt;0.05), IL-6 and TG (r=0.18; p &lt;0.05), strong correlation relationship between IL-6 and ATPO (r=0.78; p&lt;0.05), IL 6 and TSH (r=0.62; p&lt;0.05).

The effect of abdominal obesity on the level of inflammatory markers in young people with hypertension

Objective: to investigate pro- and anti-inflammatory blood markers in young people with arterial hypertension (AH) against the background of abdominal obesity (AO).&#x0D; Materials and methods: the study included 530 people, including 267 people with hypertension, of which 169 were with AO. In the control group (without AH) there were 263 people comparable in gender and age, with AO - 106 people. The content of TNF-α, IL-6, IL-8, MCP-1 (monocytic chemoatractant protein), PAI-1 (plasminogen activator inhibitor-1), IL-10, IL-17a, IL-17e, IL-17f were determined in all blood by multiplex analysis. Statistical processing was carried out in the SPSS 13.0 program.&#x0D; Results: The level of IL-17a was 1.64 times higher in patients with hypertension (p0.05) compared to the control. The level of IL-6 in patients with hypertension was higher (p0.0001) by 52.91% compared to the group without hypertension. There was no difference between the control and the group with hypertension for the remaining biomarkers. The influence of AO on the level of the studied markers in the control group was not revealed. In the group with AH, a significantly higher level of PAI-1 (p0.05) was in the subgroup with AO. For subgroups with AO, the difference between patients without AH and with AH was manifested in a decrease in IL-17e and an increase in IL-6 in patients with AH (p0.05). No statistically significant difference was found for the remaining indicators. The relative chance of early hypertension was associated with the presence of AO and an increase in the level of IL-6.&#x0D; Conclusion: From the markers of inflammation studied by us, an increased level of IL-6 and IL-17a can serve as potential biomarkers indicating a high probability of developing early hypertension in people under 45 years of age.

Association Between Interleukin-6 Gene Polymorphism and Severity of Coronary Artery Disease in Patients with Diabetes.

Previous studies have identified diabetes as a risk factor for coronary heart disease. This study determined the correlation between the IL-6 gene -572 G/C polymorphism and the incidence and severity of coronary heart disease in patients with diabetes. One hundred four patients with diabetes who were admitted to our hospital from January 2019 to December 2020 were retrospectively enrolled in the current study. These patients were divided into a diabetes only group (group A, 27 patients) and a diabetes complicated by coronary heart disease group (group B, 77 patients). Seventy patients in the latter group were further divided into low, medium, and high Syntax score groups based on coronary angiography results. A correlation analysis between IL-6, blood lipids, and the IL-6 -572 G/C gene levels was performed. The serum IL-6 level in patients with the IL-6-572G/C-GG genotype was higher than patients with the GC and CC genotypes. In patients with diabetes, the presence of the IL-6-572G/C-GG and GC genotypes was associated with a significantly increased risk of developing coronary heart disease. Patients with the IL-6-572G/C-GG genotype and diabetes were shown to have more severe coronary artery lesions compared to patients with the CC genotype. Moreover, the G allele of the IL-6-572G/C gene was linked to a higher risk of coronary heart disease and more severe coronary artery lesions in patients with diabetes compared to the C allele. The IL-6-572G/C gene polymorphism is associated with the incidence and severity of coronary heart disease in patients with diabetes.

IL-8 Triggers Neutrophil Extracellular Trap Formation Through an Nicotinamide Adenine Dinucleotide Phosphate Oxidase- and Mitogen-Activated Protein Kinase Pathway-Dependent Mechanism in Uveitis.

To explore the mechanism underlying IL-8-induced neutrophil extracellular trap (NET) formation in patients with ocular-active Behçet's disease (BD) and the effect of inhibiting NET formation on the severity of inflammation in experimental autoimmune uveitis (EAU) mice. The serum extracellular DNA and neutrophil elastase (NE) and IL-8 levels in patients with ocular-active BD, the expression of myeloperoxidase, NE, and histone H3Cit in IL-8-induced neutrophils isolated from healthy controls, and the effects of NETs on HMC3 cells were detected. Female C57BL/6J mice were immunized with IRBP651-670 to induce EAU and EAU mice received intravitreal injection of the CXCR2 (IL-8 receptor) antagonist SB225002 or PBS. The serum levels of extracellular DNA, NE, and keratinocyte-derived chemokine (the mouse ortholog of human IL-8) and expression of myeloperoxidase, NE, and histone H3Cit in mouse retinas were detected. Disease severity was evaluated by clinical and histopathological scores. Serum keratinocyte-derived chemokine expression levels in EAU mice and IL-8 expression levels in patients with ocular-active BD increased. IL-8 notably increased NET formation in a dose-dependent manner through an nicotinamide adenine dinucleotide phosphate oxidase and mitogen-activated protein kinase pathway dependent mechanism. IL-8-induced NET formation damaged HMC3 cells in vitro. Pretreatment with SB225002 notably ameliorated the production of NETs in EAU mice. Our data confirm that NET formation is induced by IL-8. IL-8-induced NET formation was found to be related to mitogen-activated protein kinase and nicotinamide adenine dinucleotide phosphate pathways. Pretreatment with the CXCR2 antagonist SB225002 alleviated neutrophil infiltration and suppressed NET formation in EAU mice.

Особенности экспрессии CD163+ и HLA-DR+ на моноцитах крови при раке молочной железы

Circulating monocytes are important actors of the tumor growth pathogenesis. It has been shown that some features of the monocyte populations expressing endocytosis receptors or the major histocompatibility complex components are observed in blood of patients with breast cancer. The study was aimed to assess the relationship of tumor parameters and blood cytokine profile with the composition of circulating monocyte populations in patients with localized and locally advanced breast cancer. The study has shown that the circulating monocytes’ phenotypic characteristics are correlated to the clinical and morphological features of the neoplastic process. The content of populations with the CD14+CD16++CD163+ and CD14++CD16+CD163+ phenotypes positively correlates with the disease stage, while larger primary tumor size is associated with lower CD14+CD16++ monocyte levels. Elevated serum levels of IL8 and МСР-1 are observed in breast cancer patients. High IL6 levels in patients with breast cancer are associated with the reduced percentage of CD14++CD16-HLA-DR+, CD14+CD16++HLA-DR+ and CD14++CD16-СD163+ monocytes. Thus, CD163+ and HLA-DR+ monocytes are associated with the clinical and morphological parameters and blood levels of cytokines, which suggests that these populations are involved in breast cancer progression and indicates that further research is advisable for translation of the findings into clinical practice.

EFFECT OF MELATONIN ON THE LEVEL OF CYTOKINES IN PREECLAMPSIA

Introduction. The literature reports a decrease in melatonin levels in preeclampsia, when the degree of decrease correlates with the severity of the process, as well as a decrease in the expression of melatonin receptors in the placental tissue in case of fetal growth restriction.Regarding changes in the cytokine profile in PE: there is evidence that the levels of proinflammatory cytokines, namely TNF-α and IL-6, increase in preeclampsia, while the concentrations of anti-inflammatory cytokines, namely IL-4 and IL-10, decrease. Melatonin, on the contrary, reduces the secretion of proinflammatory cytokines, in particular TNF-α, and increases the production of anti-inflammatory cytokines, namely IL-10. Therefore, normalization of melatonin levels may be a promising direction in the treatment of PE in pregnant women.Objective of the study. Assess the pathogenetic mechanisms of complications arising from a decrease in melatonin, and, in particular, find out more about the changes in melatonin, IL-6 and IL-10, determine the correlation between them.Material and research methodsThe study conducted at the Department of Pregnancy Pathology of the Chernivtsi Regional Perinatal Center, involved 32 women whose pregnancy was complicated by preeclampsia. The control group consisted of 33 women with uncomplicated pregnancies observed in the antenatal clinic of the Chernivtsi Regional Perinatal Center.The patient's informed consent form and the patient's examination card were approved by the Biomedical Ethics Committee of Bukovinian State Medical University (BSMU) of the Ministry of Health of Ukraine (Chernivtsi). The work was guided by the general provisions of the Declaration of Helsinki "Recommendations for Physicians for Biomedical Research Involving Human Subjects" (1964), the World Medical Association on Ethical Principles for Scientific Medical Research Involving Human Subjects (1964-2000), considering the requirements of Directive 2001/20/EC of the European Parliament and of the Council, ICH GCP, the Council of Europe Convention on Human Rights and Biomedicine (04.04.1997), Order of the Ministry of Health of Ukraine No. 690 of 23.09.2009.Statistical data were calculated using MedCalc software developed by MedCalc Software (Ostend, Belgium). The results were evaluated using the Mann-Whitney U-test for small groups. The P value of &lt;0.05 was considered valid. The level of melatonin in venous blood taken from women with diagnosed PE was significantly lower (p=0.029), as well as levels of placental growth factor PlGF (p&lt;0.0001), compared to healthy women. In our study we also found that the concentrations of both proinflammatory IL-6 and anti-inflammatory IL-10 were elevated in women with PE compared to women with uncomplicated pregnancies. We performed a prognostic assessment of the impact of the biochemical parameters we studied on the outcome of pregnancies in the examined patients. Melatonin, if present in the blood of a pregnant woman in a concentration higher than the threshold values we established (6.71 pg/ml), reduces the chances of clinical manifestations of severe preeclampsia with a high degree of reliability (p=0.00173). We assume that placental dysfunction in particular leads to impaired synthesis of the antioxidant melatonin, which causes disorders in the tissues of the mother, placenta and fetus and, as a result, provokes clinical manifestations of preeclampsia. The condition of the placenta also worsens the subclinical inflammatory process, as evidenced by an increase in IL-6 levels in patients with diagnosed preeclampsia; an increase in IL-10 in the study group, in our opinion, is a kind of compensatory reaction that prevents preterm labour. In women with pregnancies complicated by preeclampsia, melatonin is a prognostic criterion for delivery before 38 weeks of gestation, and this delivery is mainly associated with the progression of preeclampsia to its severe stage. In the group of patients with uncomplicated pregnancy, proinflammatory interleukin-6 is predictive of spontaneous onset of labour before 38 weeks of gestation. Placental growth factor PlGF was a predictor of the birth of children with a body weight of less than 3000 g in both study groups.Conclusions. Concentrations of melatonin and of placental growth factor in venous blood in women whose pregnancy was complicated by preeclampsia in the third trimester of pregnancy are significantly lower compared to uncomplicated pregnancies. On the other hand, the levels of proinflammatory IL-6 and anti-inflammatory IL-10 were significantly increased in the group of patients with diagnosed preeclampsia compared with normal pregnancy.

Expression and Prognostic Value of Cytokines in Patients with Newly Diagnosed Diffuse Large B-Cell Lymphoma

To investigate the expression and prognostic value of cytokines in patients with newly diagnosed diffuse large B-cell lymphoma (DLBCL). Clinical data of 62 patients diagnosed with DLBCL in the First People's Hospital of Yunnan Province from June 2017 to November 2018 were collected. The differences in expression levels of 14 serum cytokines [interleukin (IL)-1β, IL-2, IL-4, IL-5, IL-6, IL-8, IL-10, IL-12p70, IL-17A, IL-17F, IL-22, interferon (IFN)-γ, tumor necrosis factor (TNF)-α, TNF-β] in patients with different survival outcomes, and the impact of the cytokines on 3-year progression-free survival (PFS) and 3-year overall survival (OS) of patients with DLBCL were analyzed retrospectively. Among the 14 cytokines, only the expression of IL-10 was significantly different in patients with different survival outcomes (P =0.007). According to the receiver operating characteristic (ROC) curve, the optimal cut-off value for IL-10 was 11.74 pg/ml. Serum IL-10 was positively correlated with infection markers procalcitonin (PCT) (r =0.321, P =0.029), C-reactive protein (CRP) (r =0.320, P =0.013) and tumor burden index lactate dehydrogenase (LDH) (r =0.439, P <0.001) in newly diagnosed DLBCL patients. The level of IL-10 in patients with pulmonary infection was significantly higher than that in patients without pulmonary infection (P =0.012). However, there was no statistically significant difference on the 3-year survival outcomes between patients with or without pulmonary infection. There was no significant difference in IL-10 level in patients with different Ann Arbor stages (P >0.05). Patients with high IL-10 level (IL-10>11.74 pg/ml) had significantly higher LDH level than those with low IL-10 level (IL-10≤11.74 pg/ml) (P <0.001). The 3-year PFS rate and 3-year OS rate of DLBCL patients with high IL-10 level were significantly lower than those of low IL-10 level group [(44.4±11.7)% vs (81.8±5.8)%, P <0.001; (61.6±11.5)% vs (93.2±3.8)%, P =0.001]. Serum IL-10 level in newly diagnosed DLBCL patients can reflect the inflammatory state of the body, which may be related to tumor load. Newly diagnosed DLBCL patients with serum IL-10>11.74 pg/ml have higher early mortality and worse prognosis.
