Hypoglycemic and Hypolipidemic Activities of Ethanolic Extract of Elaeocarpus Tectorius (Lour.) Poir. Leaves in Streptozotocin- Nicotinamide Induced Diabetic Rats

The prevalence of diabetes is growing at an alarming rate on a global scale with approximately 536.6 million people having diabetes in 2021. Medicinal plants are gaining recognition in the treatment of diabetes owing to their safety, ability and low cost. Elaeocarpus tectorius (Lour.) Poir. is one of the least explored plants belonging to the genus Elaeocarpus. With this context, the current study was aimed to explore the antidiabetic and hypolipidemic effect of ethanolic extract of E. tectorius leaves in diabetes-induced rats. Phytochemical screening was performed and the oral acute toxicity of the plant extract was explored. Experimental diabetes was induced using streptozotocin and nicotinamide. The experimental animals were orally treated with ethanolic leaf extract of E. tectorius at doses 200 mg/kg b.w. and 400 mg/kg b.w. for 28 days after which body weight, fasting blood glucose levels, markers of serum lipid profile and histological variations in the pancreas were ascertained. Oral treatment of E. tectorius leaf extract daily for 28 days has significantly brought down (p<0.05) the fasting glucose levels in streptozotocin-nicotinamide induced diabetic rats and normalized the levels of serum lipid profile markers. The histological investigation revealed that the extract has caused significant restoration of pancreatic islet cells, and it was found that the dose of 400 mg/kg b.w. was more efficient and successful in treating hyperglycemia and circumventing related complications than 200 mg/kg b.w. The present study suggests that the leaf extract of E. tectorius possesses good antidiabetic and hypolipidemic activity and can be a useful and cheap alternative to treat diabetes.

EVALUATION OF HYPOLIPIDEMIC ACTIVITY OF ETHANOLIC EXTRACT OF LEAVES OF MORINGA CONCANENSIS NIMMO IN GUINEA PIGS

Objective & Scope:To evaluate the hypolipidemic activity of ethanolic extract of leaves of Moringa concanensisnimmo in guinea pigs Methods:48 Guinea pigs of either gender were randomly divided into 6 groups. Hyperlipidemia was produced by giving cholesterol powder (500 mg/kg) for 60 days. Rosuvastatin 1.5 mg/kg served as active control group. While Moringa was given as 200 mg/kg for 30 days as preventive and curative groups for 0 to 30 days and 31 to 60 days respectively. Distilled water with normal diet for 30 dys was served as Normal control group. The preventive and curative effect of ethanolic extract of Moringa were assessed by serological and histopathological parameter. Results:Cholesterol administration produced increased level of Lipids and liver enzymes along with histopathological alteration of liver and thoracic aorta. Preventive and therapeutic administration of 200 mg/kg ethanolic extract of Moringa resulted into a significant decrease of lipid profile and restoration of histopathological architecture of liver and thorasic aorta as compared to disease control group. Conclusion:Because of antioxidant properties, curative and preventive administration200 mg/kg ethanolic extract of Moringa exerts lipid lowering effect and can be used to treat and prevent dyslipidemia.

Hypolipidemic activity of ethanolic extract of Caulerpa recemosa

Background: Caulerpa recemosa, also known as sea grape, is a stout straggling prostrate shrubby plant with spinous stipules and uniaxial siphonous thallus, mostly divided into a creeping axis (stolon), with rhizoids and erect shoots (fronds), either nude, leaf-like or with grape or feather-like ramuli. Caulerpa racemosa has erect fronds up to 9-13 cm high bearing un-crowded vesiculate ramuli that are radially arranged. Fronds are slightly inflated above the attachment to the stolon which are fixed to the substrate by thin short rhizoids. It is collected from St. Martin’s Island locally called ‘Narikel Jinjira’, located on the southernmost tip of Bangladesh for the present study, ethanol extract of Caulerpa recemosa has been screened for its hypolipidemic activity.
 Methods: Hypolipidemic activity was screened by inducing hyperlipidemia with the help of atherogenic diet in wistar albino rats and serum levels of various biochemical parameters such as total cholesterol, triglycerides, low density lipoprotein and high density lipoprotein cholesterol were determined. Atherogenic index shows the measure of the atherogenic potential of the drugs.
 Results: Ethanol extract showed significant (p < 0.01) hypolipidemic effect by lowering the serum levels of biochemical parameters such as significant reduction in the level of serum cholesterol, triglyceride, low density lipoprotein and increase in high density lipoprotein level which was similar to the standard drug atorvastatin. Ethanol extract exhibited significant atherogenic index andpercentage protection against hyperlipidemia. Preliminary phytochemical analysis revealed the presence of phytoconstituents such as steroids, flavonoids, glycosides, alkaloids, phenoliccompounds.
 Conclusion: The overall experimental results suggest that the biologically active phytoconstituents such as flavonoids, glycosides alkaloids present in the ethanolic extract of Caulerpa racemosa may be responsible for the significant hypolipidemic activity and the results may justify the use of Caulerpa racemosa as a significant hypolipidemic agent.

Evaluation of in vivo Synergistic Hypoglycemic & Hypolipidemic Activity of Ethanolic Extract of Calotropis gigantean Leaves in Combination to Metformin in Alloxan Induced Rats

Aim: The present study was designed to investigate the antidiabetic & hypolipidemic activity of Calotropis gigantean (Family: Apocynaceae) in alloxan-induced diabetic rat model.
 Study Design: In vivo study was carried out by ethanolic leaf extract was administered in 250 mg/kg body weight concentration and then subjected to different rats models to authenticate the antidiabetic and hyperlipidimic properties of the plant.
 Place and Duration of Study: Department of Pharmacy, Southeast University, Banani, Dhaka-1213,Bangladesh within a period of July 2018 to December, 2018.
 Methodology: Diabetes was induced in rats by an intraperitoneal injection (i.p) of alloxan (100 mg/kg B.W). Ethanolic leaf extract of C. gigantean (250 mg/kg B.W) was administrated orally as a single dose per day to the diabetic rats for 7 days. The negative control group received 0.5 ml of sterile normal saline water orally & positive control group received metformin orally. Synergistic effect of plant was evaluated by combination with 100 mg/kg B.W & 50 mg/kg B.W oral administration of metformin. After 7 days study period, fasting blood glucose, total cholesterol, triglyceride, high-density lipoprotein cholesterol, liver weight & body weight were measured only for diabetic group to observe the effects of diabetes induction.
 Results: Individual plant extract (250 mg/Kg B.W) & Metformin (100 mg/kg B.W) reduced FBG significantly by 52% (P<0.001) & 55.3% (P<0.001) correspondingly. Metformin (100 mg/kg B.W) potentiated reduction (68%) (P<0.001) when combined to plant extract (250 mg/Kg B.W). Significant dose dependent manner was followed when metformin (50 mg/kg B.W) was combined to plant extract (250 mg/Kg B.W). Our results clearly suggests that C. gigantean exhibit hypoglycemic & hypolipidemic activity with an alteration in body-liver weight. The present study also suggested to develop a combination therapy of extract along with metfromin in different doses to minimize the intake of synthetic drug. Significant reduction of TG, TC were noted by extract (250 mg/kg B.W) with 32.42% (P<0.001) & 41.32% (P<0.001) respectively where standard shown the diminution 43.43% (P<0.05) & 47.21% (P<0.001) respectively as compare to Untreated diabetic rats. 50.21% (P<0.01) & 42.38% (P<0.001) reduction of TG & TC were estimated by C.gigantea extracts (250 mg/kg B.W) when combined with Metformin (100 mg/kg B.W). 34.53% (P<0.05) & 41.54% (P<0.001) reduction of TG & TC by C.gigantea extracts (250 mg/kg B.W) were confirmed when combined to Metformin (50 mg/kg B.W). Combination therapy also has shown synergistic effect in elevation of plasma HDL-cholesterol.
 Conclusion: The results of the study concluded that C. gigantean have potential antidiabetic and antioxidant properties.

The hypolipidemic activity of ethanolic extract of seed kernel of Caesalpinia bonducella fleming on serum lipids and atherogenesis in albino rats fed with high fat diet

Background: An increase in the incidence and prevalence of hyperlipidemia in India has led to an increased demand for drugs with a lipid lowering potential. Thus, the present study was undertaken to evaluate the hypolipidemic activity of ethanolic extract of seed kernel of Caesalpinia bonducella Fleming (EESKCB) in albino rats fed with high fat diet.Methods: The extract was daily administered to healthy adult albino rats of either sex, fed with high fat diet for a period of 60 days. At the end of 60 days, i.e. on the 61st day, serum cholesterol, serum triglyceride, serum low density lipoprotein cholesterol, serum high density lipoprotein cholesterol, atherogenic index, the body weights of the rats and the serum levels of malondialdehyde (MDA) and catalase were estimated. Phytochemical analysis of the plant extract was also carried out. For statistical analysis, the methods employed were One-way ANOVA, Dunnett’s multiple comparison test and Student’s ‘t’ test (Paired). Values of p < 0.05 were considered significant.Results: The test drug brought about significant (p<0.05) decrease in the raised levels of all the lipid parameters except the High density lipoprotein cholesterol. There was also a significant (p<0.05) increase in the serum levels of catalase and a significant (p<0.05) reduction in the serum levels of MDA in the test drug treated group. The test drug also significantly (p<0.05) inhibited the rise in body weights of the rats.Conclusions: From this study, it can be said that the seed kernel of Caesalpinia bonducella Fleming has significant hypolipidemic activity.

Background: Diabetes mellitus (DM) is the most common non-communicable disease of the modern world. The study of plants having antihyperglycemic and hypolipidemic activities may give a new approach in the treatment of DM. The study was intended to evaluate the antihyperglycemic and hypolipidemic activity of ethanolic extract of Clerodendrum infortunatum Linn. (EECL) in alloxan-induced diabetic albino rats. Methods: Diabetes was induced in albino rats by administration of alloxan monohydrate (160 mg/kg, intraperitoneal). Rats were divided into six groups of six animals each. First group served as non-diabetic control, second group as diabetic control, third group as standard and was treated with 0.1 mg/kg/day of glibenclamide. Group 4, 5, and 6 received 200 and 400 mg/kg body weight of EECL. Blood samples were analyzed for blood glucose on day 1, 3, 7, 14, 21, and 28 and lipid profile on day 28. Results: The EECL showed a significant reduction (p<0.001) in blood glucose level and serum lipid profile levels with 400 mg/kg body weight in alloxan induced diabetic rats as compared with control. Conclusion: It is concluded that EECL is effective in controlling blood glucose levels and in improving lipid profile in diabetic rats.

HYPOLIPIDEMIC ACTIVITY OF ETHANOLIC EXTRACT OF DAUCUS CAROTA SEEDS IN NORMAL RATS

Objective: Hypolipidemic activity of ethanolic extract of Daucus carota seeds in Normal rats. Method: Rats were divided in to 4 groups. First control group, treated with vehicle only for 7 days. Second group was standard, treated with lovastatin (7.2mg/kg) for 7 days. Third and fourth group were test, treated with Daucus carota seeds extract (200mg/kg and 400mg/kg) for 7 days. 8th day blood sample was collected by retro orbital route and lipid profiles were estimated in serum. Result and Conclusions: Standard and test (400mg/kg DCSE) groups were showed significant (P <0.001) reduction in the total cholesterol, triglyceride HDL and VLDL as compared control group. Test group (200mg/kg) was showed significant (P <0.05) reduction as compared control group. The antioxidant potential of Daucus carota seeds has contributed to the reduction of oxidative stress and Lipid levels in experimental rats.

Hypolipidemic activities of Ficus racemosa Linn. bark in alloxan induced diabetic rats.

Ficus racemosa (Moraceae family) is used in traditional system of medicine for the treatment of several disorders including diabetes mellitus. The aim of the study was to investigate the antihyperglycemic and hypolipidemic activities of ethanolic extract of Ficus racemosa bark (FrEBet) in alloxan-induced diabetic rats. A total number of 30 animals were divided into 5 groups of six each. Diabetes mellitus was induced by single intraperitoneal injection of freshly prepared solution of alloxan monohydrate (150 mg/kg bw) dissolved in physiological saline in overnight fasted wistar rats. Dose dependent studies for FrEBet (100-500 mg/kg bw) was carried out to find out the effective pharmacological dose (antidiabetic and hypolipidemic) to alloxan-induced diabetic rats. Blood glucose, plasma insulin, total cholesterol, phospholipids, triglycerides, free fatty acids, HDL cholesterol and LDL cholesterol levels in plasma, erythrocyte membranes, liver and kidney were determined by specific colorimetric methods. An increase in blood glucose was accompanied by an increase in total cholesterol, phospholipids, triglycerides, FFA and decrease in HDL cholesterol in diabetic rats. Oral administration of FrEBet (300 mg/kg bw) to diabetic rats restored the status of blood glucose, lipids and lipoproteins to near normal range. Our investigation thus shows that FrEBet has potent antidiabetic and hypolipidemic effects in alloxan-induced diabetic rats and these effects were much comparable to that of the standard reference drug, glibenclamide.

Hypoglycemic and hypolipidemic activity of ethanolic extract of Salvadora oleoides in normal and alloxan-induced diabetic rats.

Objective:To find out the hypoglycemic and hypolipidemic activity of an ethanolic extract of the aerial part of Salvadora oleoides Decne in euglycemic and alloxan-induced diabetic albino rats.Materials and Methods:Diabetes was induced in albino rats by administration of alloxan monohydrate (120 mg/kg, i.p.). Normal as well as diabetic albino rats were divided into groups (n = 6) receiving different treatments: vehicle (control), ethanolic extract (1 g and 2 g/kg b.w), and standard antidiabetic drug tolbutamide (0.5 g/kg b.w.). Blood samples were collected by cardiac puncture and were analyzed for blood glucose and lipid profile on days 0, 7, 14, and 21.Results:The ethanolic extract of S oleoides produced significant reduction (P < 0.001) in blood glucose and also had beneficial effects (P < 0.001) on the lipid profile in euglycemic as well as alloxan-induced diabetic rats at the end of the treatment period (21st day). However, the reduction in the blood glucose and improvement in lipid profile was less than that achieved with the standard drug tolbutamide.Conclusion:We concluded that an ethanolic extract of S oleoides is effective in controlling blood glucose levels and improves lipid profile in euglycemic as well as diabetic rats.
