Measurements and simulations of microtubule growth imply strong longitudinal interactions and reveal a role for GDP on the elongating end

In the present study, we used interference reflection microscopy (IRM) to observe the in vitro growth rates and fluctuations of bovine brain microtubules at high temporal resolution. We performed measurements in the presence of GTP and in the presence of the hydrolysis-resistant GTP analog GMPCPP. We fit a computational to the resulting measurements to provide insight into the molecular interactions governing microtubule elongation. Microtubules growing with GMPCPP showed a very low apparent critical concentration, reflecting high affinity interactions between αβ-tubulin subunits and the microtubule end.

HPLC method to resolve, identify and quantify guanine nucleotides bound to recombinant ras GTPase

The Ras superfamily of small G proteins play central roles in diverse signaling pathways. Superfamily members act as molecular on-off switches defined by their occupancy with GTP or GDP, respectively. In vitro functional studies require loading with a hydrolysis-resistant GTP analogue to increase the on-state lifetime, as well as knowledge of fractional loading with activating and inactivating nucleotides. The present study describes a method combining elements of previous approaches with new, optimized features to analyze the bound nucleotide composition of a G protein loaded with activating (GMPPNP) or inactivating (GDP) nucleotide. After nucleotide loading, the complex is washed to remove unbound nucleotides then bound nucleotides are heat-extracted and subjected to ion-paired, reverse-phase HPLC-UV to resolve, identify and quantify the individual nucleotide components. These data enable back-calculation to the nucleotide composition and fractional activation of the original, washed G protein population prior to heat extraction. The method is highly reproducible. Application to multiple HRas preparations and mutants confirms its ability to fully extract and analyze bound nucleotides, and to resolve the fractional on- and off-state populations. Furthermore, the findings yield a novel hypothesis for the molecular disease mechanism of Ras mutations at the E63 and Y64 positions.

Affinity of transducin for photoactivated rhodopsin: dependence on nucleotide binding state

The interaction of the rod GTP binding protein, Transducin (G(t)), with bleached Rhodopsin (R(*)) was investigated by measuring radiolabeled guanine nucleotide binding to and release from soluble and/or membrane-bound G(t) by reconstituting G(t) containing bound GDP (G(t-)GDP) or the hydrolysis-resistant GTP analog guanylyl imidodiphosphate (G(t-)p[NH]ppG) with R* under physiological conditions. Release of GDP and p[NH]ppG from G(t) occurred to the same extent and with the same light sensitivity both in the presence and absence of added GTP. Significant amounts of G(t) without bound nucleotide (G(t-)) were generated. When ROS containing bleached rhodopsin (R(*)) were centrifuged in low ionic strength buffer, G(t-) remained associated with the membrane fraction, whereas G(t-)GDP remained in the soluble fraction. These results suggest that G(t-)GDP and G(t-)p[NH]ppG have similar affinities for R(*). The results also suggest that G(t-), rather than G(t-)GDP, is the moiety which exhibits tight, "light-induced" binding to rhodopsin.

Liposomes Comprising Anionic but Not Neutral Phospholipids Cause Dissociation of Rac(1 or 2)·RhoGDI Complexes and Support Amphiphile-independent NADPH Oxidase Activation by Such Complexes

Activation of the phagocyte NADPH oxidase involves the assembly of a membrane-localized cytochrome b559 with the cytosolic components p47(phox), p67(phox), p40(phox), and the GTPase Rac (1 or 2). In resting phagocytes, Rac is found in the cytosol as a prenylated protein in the GDP-bound form, associated with the Rho GDP dissociation inhibitor (RhoGDI). In the process of NADPH oxidase activation, Rac is dissociated from RhoGDI and translocates to the membrane, in concert with the other cytosolic components. The mechanism responsible for dissociation of Rac from RhoGDI is poorly understood. We generated Rac(1 or 2) x RhoGDI complexes in vitro from recombinant Rac(1 or 2), prenylated enzymatically, and recombinant RhoGDI, and purified these by anion exchange chromatography. Exposing Rac(1 or 2)(GDP) x RhoGDI complexes to liposomes containing four different anionic phospholipids caused the dissociation of Rac(1 or 2)(GDP) from RhoGDI and its binding to the anionic liposomes. Rac2(GDP) x RhoGDI complexes were more resistant to dissociation, reflecting the lesser positive charge of Rac2. Liposomes consisting of neutral phospholipid did not cause dissociation of Rac(1 or 2) x RhoGDI complexes. Rac1 exchanged to the hydrolysis-resistant GTP analogue, GMPPNP, associated with RhoGDI with lower affinity than Rac1(GDP) and Rac1(GMPPNP) x RhoGDI complexes were more readily dissociated by anionic liposomes. Rac1(GMPPNP) x RhoGDI complexes elicited NADPH oxidase activation in native phagocyte membrane liposomes in the presence of p67(phox), without the need for an anionic amphiphile, as activator. Both Rac1(GDP) x RhoGDI and Rac1(GMPPNP) x RhoGDI complexes elicited amphiphile-independent, p67(phox)-dependent NADPH oxidase activation in phagocyte membrane liposomes enriched in anionic phospholipids but not in membrane liposomes enriched in neutral phospholipids.

Differential ligand efficacy at h5-HT 1A receptor-coupled G-protein subtypes: a commentary

Activation of heterotrimeric G-protein involves the exchange by α subunits of GDP for GTP. The binding of the hydrolysis-resistant GTP analogue, guanosine-5′- O-(3-[ 35S]thio)-triphosphate ([ 35S]GTPγS), provides a measure of agonist, antagonist and inverse agonist actions at G-protein-coupled receptors. However, classical [ 35S]GTPγS binding methods do not distinguish among the subtypes of G-proteins activated. This is a limitation in view of (i) promiscuity of receptor coupling to multiple families of G-proteins, and (ii) the differential influence of ligands on distinct signal transduction pathways. A recent development of [ 35S]GTPγS binding methodology employing antibody capture and scintillation proximity assays (SPA) permits the targeting of specific Gα subtypes in both recombinant and native tissues. When applied to human serotonin 1A receptor (h5-HT 1A) expressed in Chinese hamster ovary (CHO) cells, this methodology revealed surprising patterns of Gα i3 subunit activation. For example, low but not high concentrations of high-efficacy h5-HT 1A agonists direct receptor signalling to Gα i3. In contrast, partial agonists favour h5-HT 1A receptor signalling to Gα i3 over a wide concentration range. Further, alterations in buffer sodium concentration reversed these actions of agonists: stimulation of Gα i3 activation was observed at high sodium concentration, but inhibition was observed at low sodium concentration, suggestive of protean efficacy, i.e. a switch from positive to negative efficacy dependent on receptor tone. These results indicate that complex changes in both magnitude and direction of response to receptor ligands can occur for specific G-protein subtypes. Interestingly, the inverse agonist, spiperone, inhibited constitutive h5-HT 1A receptor-mediated Gα i3 activation under all conditions tested, consistent with the hypothesis that it selectively stabilises distinct receptor conformation(s). Further, whereas classical [ 35S]GTPγS binding assays in CHO cell membranes failed to demonstrate negative efficacy for the neuroleptic, haloperidol, this compound exhibited robust inverse agonism for the activation of Gα i3 subunits. These data suggest that haloperidol may selectively inhibit constitutive activation of this G-protein subtype and, thus, exhibit inverse agonist-mediated trafficking of receptor signalling.

GTP gamma S increases Nav1.8 current in small-diameter dorsal root ganglia neurons.

Tetrodotoxin-resistant (TTX-R) sodium current in small-size dorsal root ganglia (DRG) neurons is upregulated by prostaglandin E(2) and serotonin through a protein kinase A (PKA)/protein kinase (PKC) pathway, suggesting G protein modulation of one or more TTX-R channels in these neurons. Recently, GTP(gammaS), a hydrolysis-resistant analogue of GTP, was shown to increase the persistent current produced by the TTX-R Na(v)1.9. In this study, we investigated the modulation of another TTX-R channel, Na(v)1.8, by GTP(gammaS) in small-diameter DRG neurons from rats using whole-cell voltage clamp recordings. Because it has been suggested that fluoride, often used in intracellular recording solutions, may bind to trace amounts of aluminum and activate G proteins, we recorded Na(v)1.8 currents with and without intracellular fluoride, and with the addition of deferoxamine, an aluminum chelator, to prevent fluoride-aluminum binding. Our results show that GTP(gammaS) (100 micro M) caused a significant increase in Na(v)1.8 current (67%) with a chloride-based intracellular solution. Although the inclusion of fluoride instead of chloride in the pipette solution increased the Na(v)1.8 current by 177%, GTP(gammaS) further increased Na(v)1.8 current by 67% under these conditions. While the effect of GTP(gammaS) was prevented by pretreatment with H7 (100 micro M), a non-selective PKA/PKC inhibitor, the fluoride-induced increase in Na(v)1.8 current was not sensitive to H7 (100 micro M), or to inclusion of deferoxamine (1 mM) in the intracellular solution. We conclude that G protein activation by GTP(gammaS) increases Na(v)1.8 current through a PKA/PKC mechanism and that addition of fluoride to the pipette solution further enhances the current, but is not a confounding variable in the study of Na(v)1.8 channel modulation by G proteins independent of a PKA/PKC pathway or binding to aluminum.

Extracellular conversion of guanine-based purines to guanosine specifically enhances astrocyte glutamate uptake

Guanosine (GUO) has been shown to stimulate glutamate uptake in primary astrocyte cultures. The purpose of this study was to determine the effect and specificity of guanine- or adenine-based purines on glutamate and GABA uptake in cultured astrocytes. Stimulatory effect on glutamate uptake was observed with GUO, GMP or GTP. Simultaneous exposure with these guanine-based purines did not show an additive effect. We also investigated a possible interconversion of guanine-based purines during incubation time. Action by GTP was excluded since the hydrolysis resistant GTP analog, GMP–PNP did not stimulate glutamate uptake. Addition of an ecto-5′-nucleotidase inhibitor abolished GMP-stimulatory effect on glutamate uptake, without affecting GUO action. Taken together, these results suggest that GUO is the guanine-based purines responsible for glutamate uptake activation. In addition, the stimulatory effect on glutamate uptake was not observed with adenine-based purines. Moreover, GABA uptake was not activated by GUO. These results point to specificity in the interaction between GUO and the astrocyte glutamate uptake system.

Ran-dependent Signal-mediated Nuclear Import Does Not Require GTP Hydrolysis by Ran

Nuclear import of classical nuclear localization sequence-containing proteins involves the assembly of an import complex at the cytoplasmic face of the nuclear pore complex (NPC) followed by movement of this complex through the NPC and release of the import substrate into the nuclear interior. This process has historically been thought to require nucleotide hydrolysis as a source of energy. We found, using hydrolysis-resistant GTP analogs and a mutant Ran unable to hydrolyze GTP, that transport of classical nuclear localization sequence containing substrate through the NPC and release of the substrate into the nucleus did not require hydrolysis of GTP by Ran. In fact, for movement of this type of import substrate into the nuclear interior we did not observe a requirement for hydrolysis of any nucleotide triphosphate. We did, however, find that a pool of free GTP (or its structural equivalent) must be added, probably because the GDP Ran that is added must be converted to GTP Ran during the import process. We found that a requirement for GTP hydrolysis can be restored to an import mixture consisting of recombinant import factors by the addition of RCC1, the Ran guanine nucleotide exchange factor.

Phosphorylation of the gamma subunit of the retinal photoreceptor cGMP phosphodiesterase by the cAMP-dependent protein kinase and its effect on the gamma subunit interaction with other proteins.

Cyclic GMP phosphodiesterase, a key enzyme in phototransduction, is composed of P alpha beta and two P gamma subunits. Interaction of P gamma with P alpha beta or with the alpha subunit (T alpha) of transducin is crucial for the regulation of cGMP phosphodiesterase in retinal photoreceptors. Here we have investigated phosphorylation of P gamma by cAMP-dependent protein kinase and its functional effect on the P gamma interaction with P alpha beta or T alpha in vitro. P gamma, but not P gamma complexed with T alpha (both GTP and GDP forms), is phosphorylated. Measurement of 32P radioactivity in phosphorylated P gamma, analysis of phosphorylated P gamma by laser mass spectrometry, identification of phosphoamino acid, and phosphorylation of mutant forms of P gamma indicate that only threonine 35 in P gamma is phosphorylated. Phosphorylation of P gamma mutants also reveals that the C and N terminals of P gamma which are required for the regulation of P alpha beta functions are not involved in the P gamma phosphorylation but that arginine 33, which is ADP-ribosylated by an endogenous ADP-ribosyltransferase, is required for the phosphorylation. Phosphorylated P gamma has a higher inhibitory activity for trypsin-activated cGMP phosphodiesterase than nonphosphorylated P gamma, indicating that the P gamma-P alpha beta interaction is affected by P gamma phosphorylation. Nonphosphorylated P gamma inhibits both the GTPase activity of T alpha and the binding of a hydrolysis-resistant GTP analogue to T alpha, while P gamma phosphorylation reduces these inhibitory activities. These observations suggest that a P gamma domain containing threonine 35 is involved in the P gamma-T alpha interaction, and P gamma phosphorylation regulates the P gamma-T alpha interaction. Our observation suggests that P gamma phosphorylation by cAMP-dependent protein kinase may function for the regulation of phototransduction in vertebrate rod photoreceptors.

Molecular diversity and double regulatory mechanism of activation of phospholipase C in rat brain

Whereas evidence for a G protein-dependent stimulation of phospholipase C (PLC) is abundant, reports on the inhibition of PLC through a G protein-mediated pathway have only recently begun to appear. In the present study, cerebral cortex membranes were chosen since they have a readily measurable Gpp[NH]p and Ca 2+-stimulated PLC activity. Nanomolar concentrations of Gpp[NH]p, a hydrolysis-resistant GTP analogue, inhibited basal inositol 1,4,5-trisphosphate (IP 3) production, with a maximum inhibition of 25% at 10nM. Increasing the concentrations of Gpp[NH]p to over 10nM resulted in a reversal of the inhibitory effect and onset of stimulation of IP 3 production. GDPβS as a G protein inhibitor and U-73122 as a putative PLC-β inhibitor had little effect on basal IP 3 production at 100μM and 1μM, respectively. However, GDPβS and U-73122 completely antagonized both the inhibition and the stimulation of IP 3 production produced by lower and higher concentrations, respectively, of Gpp[NH]p. Rat cortical membranes expressed a greater amount of PLC- β 1. These data suggest that PLC- β 1 isozymes may be regulated by both inhibitory and stimulatory G protein-mediated mechanisms.

Persistent activation of transducin by bleached rhodopsin in salamander rods.

The hydrolysis-resistant GTP analogue GTP-gamma-S was introduced into rods isolated from the retina of the salamander Ambystoma tigrinum to study the origin of the persistent excitation induced by intense bleaching illumination. Dialysis of a dark-adapted rod with a whole-cell patch pipette containing 2 mM GTP-gamma-S resulted in a gradual decrease in circulating current. If the rod was first bleached and its sensitivity allowed to stabilize for at least 30 min, then dialysis with GTP-gamma-S produced a much faster current decay. The circulating current could be restored by superfusion with the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine, suggesting that the decay in current originated from persistent excitation of the phosphodiesterase by transducin bound to GTP-gamma-S. We conclude that the persistent excitation which follows bleaching is likely to involve the GTP-binding protein transducin, which mediates the normal photoresponse. This observation suggests that a form of rhodopsin which persists long after bleaching can activate transducin much as does photoisomerized rhodopsin, although with considerably lower gain.

Tubulin stimulates adenylyl cyclase activity in rat striatal membranes via transfer of guanine nucleotide to Gs protein

Previous studies of rat cerebral cortex and rat C6 glioma cells have demonstrated that dimeric tubulin is capable of activating the G proteins Gs and Gil via transfer of guanine nucleotide from tubulin to Gs α and Gil α. To provide further information regarding cytoskeletal modulation of adenylyl cyclase, the present study examined effects of tubulin on the activation of the enzyme in rat striatal membranes. Tubulin, prepared from rat brain by polymerization with the hydrolysis-resistant GTP analog 5′-guanylylimidodiphosphate (GppNHp) caused significant activation of adenylyl cyclase by ∼ 130%. Furthermore, tubulin-GppNHp activated SKF 38393-sensitive adenylyl cyclase and potentiated forskolin-stimulated activity of the enzyme. When tubulin, polymerized with the hydrolysis-resistant photoaffinity GTP analog [ 32p]p 3 (4-azidoanilido)-p 1-5′-GTP ([ 32P]AAGTP), was incubated with striatal membranes, AAGTP was transferred from tubulin to Gs α as well as Gi α with the extents of nucleotide transfers being 7.6 ± 0.8% and 17.8 ± 1.4% of AAGTP originally bound to tubulin, respectively. These results indicate that, in rat striatum, the tubulin dimer participates in the stimulatory regulation of adenylyl cyclase by transferring guanine nucleotide to Gs α, supporting the hypothesis that tubulin contributes to the regulation of neuronal signal transduction.

Participation of Tubulin in the Stimulatory Regulation of Adenylyl Cyclase in Rat Cerebral Cortex Membranes

This study examined effects of tubulin on the activation of adenylyl cyclase in rat cerebral cortex membranes. Tubulin, prepared from rat brain by polymerization with the hydrolysis-resistant GTP analogue 5'-guanylylimidodiphosphate (GppNHp) caused significant activation of the enzyme by approximately 156% under conditions in which stimulation rather than inhibition of the enzyme was favored. Tubulin-GppNHp activated isoproterenol-sensitive adenylyl cyclase, potentiated forskolin-stimulated activity of the enzyme, and reduced agonist binding affinity for beta-adrenergic receptors. When tubulin, polymerized with the hydrolysis-resistant photoaffinity GTP analogue [32P]P3(4-azidoanilido)-P1-5'-GTP ([32P]AAGTP), was incubated with cerebral cortex membranes, AAGTP was transferred from tubulin to Gs alpha as well as Gi alpha. These results suggest that, in rat cerebral cortex membranes, the tubulin dimer participates in the stimulatory regulation of adenylyl cyclase by transferring guanine nucleotide to Gs alpha, as well as affecting the Gi-mediated inhibitory pathway.

Activation of the g protein in porcine oocytes

Fertilization in most animal species involves a transient increase in the intracellular free Ca” concentration of the oocyte in the form of a CaPt wave starting from the site of sperm attachment. It is thought that the origin of this Ca” is inositol triphosphate (IP,) induced Ca” release from the endoplasmic reticulum. IP, is derived from the hydrolyses of phosphatidylinositol4,5biphosphate (PIP,) which is catalyzed by the enzyme phospholipase C under regulation of a G protein. Thus the G protein mediated PIP, hydrolysis seems to play an important role in oocyte activation. G proteins or guanine nucleotide binding proteins are a class of membrane proteins that couple membrane receptors to membrane effector enzymes. They are present in a variety of cells, and in oocytes their activation by GTP-7-S (a hydrolysisresistant analog of GTP) causes resumption of meiosis in many species. The present study was undertaken to determine the possibility of porcine oocyte activation through a G protein. Oocytes from slaughterhouse-obtained porcine ovaries were matured in Waymouth medium supplemented with 2.5 mM pyruvate, 10 IU/ml PMSG, 10 IU/ml hCG, 10% (v/v) heat-treated fetal calf serum (Sigma Chemical Co., St. Louis, MO) and 10% (v/v) porcine follicular fluid for 20 h, then in the same medium without hormonal supplement for an additional 16 h. Mature oocytes were stripped of their cumulus cells by vortexing in 0.3 M mannitol solution containing 0.3 mg/ml hyaluronidase. Then 40 pl of 1 mM GTP-7-S (Sigma Chemical Co., St. Louis, MO) was injected near the center of the oocytes using a microinjector (Narishige Co. Ltd, Tokyo, Japan). The carrier medium in which GTP-7-S was dissolved consisted of 100 PM EGTA and 10 mM Hepes buffered at pH 7.0. Control oocytes were injected with either 40 pl of 1 mM GDP-p-S, a metabolically stable analog of GDP (which inactivates G proteins) or carrier medium. Injected oocytes were incubated in Hepes-buffered Tyrodes medium (HbT) for six hours then they were evaluated for some of the early and late events of oocyte activation. Transmission electron microscopy revealed that in contrast to GDP-p-S, GTP-rS caused the release of the vast majority of the cortical granules. It also triggered changes in the protein profiles (i.e., dephosphorylation of a 25 kDa protein) as determined by one-dimensional polyacrylamide gel electrophoresis (1D SDS PAGE). Following GTP-7-S injection 71.4% (190/266) of the mature oocytes formed pronuclei, which was significantly higher (P<O.OOl) than the rates of pronuclear formation in oocytes injected with GDP-p-S (23.7%; 67/282) or carrier medium (25.9%; 52/201). Furthermore, following 7 d of incubation in ligated porcine oviducts 13.1 ok ( 10/76) of the transferred injected oocytes developed to the compact morula/blastocyst stage. We conclude that a signal transducing pathway involving a G protein does exist in the porcine oocyte, and by stimulating the G protein with GTPq-S porcine oocytes can readily be activated.

Synthetic peptides as probes for G protein function. Carboxyl-terminal G alpha s peptides mimic Gs and evoke high affinity agonist binding to beta-adrenergic receptors.

The molecular interfaces between Gs and the beta-adrenergic receptor were investigated using synthetic peptides corresponding to various regions of its alpha subunit, alpha s. These experiments were carried out on saponin-permeable C6 glioma cells in which the beta-adrenergic receptor appears tightly coupled to Gs. Synthetic site-specific peptides from alpha s (corresponding to amino acids 15-29, 354-372, and 384-394) and alpha i (8-22, 315-324, and 345-455) were tested for their ability to interfere with coupling between the beta-adrenergic receptor and Gs. The two carboxyl-terminal peptides from alpha s blocked beta-adrenergic stimulation of adenylyl cyclase in permeable cells. However, only alpha s-354-372 had this effect in C6 membranes. It is suggested that the partial uncoupling of Gs, which occurs subsequent to cell disruption, may be related to a change in the interaction of the alpha s carboxyl terminus with the beta-adrenoreceptor. Two carboxyl-terminal peptides, 354-372 and 384-394, could also mimic the effect of Gs to increase agonist affinity for the beta-adrenergic receptor. In combination, alpha s-354-372 and alpha s-384-394 increased the ability of isoproterenol to compete with 125I-pindolol binding in a partially additive manner. Synthetic peptides from alpha i and amino-terminal peptides from alpha s had no effect on beta-agonist binding, suggesting a high specificity of peptide effects. Two findings suggest that these peptides bind directly to the beta-adrenergic receptor and stabilize its high agonist affinity conformation. First, GTP and hydrolysis-resistant GTP analogs did not alter the high affinity binding in the presence of high concentrations of the peptides. Second, in S49 lymphoma cyc- cells, which lack Gs, these peptides evoked the high affinity agonist binding state of the beta-receptor. Neither peptide had an effect on antagonist binding affinity, as measured by propranolol displacement of 125I-pindolol. These data suggest that at least two regions on the alpha subunit of Gs participate in high affinity Gs binding to the beta-adrenergic receptor. The fact that these small peptides could mimic the holo-Gs effect on the receptor is rather surprising, and the specificity of the effect suggests that the primary and secondary structure of small regions of alpha s contain much of the information for specific interaction with beta-adrenergic receptors.

Unoccupied β-adrenoceptor-induced adenylyl cyclase stimulation in turkey erythrocyte membranes

The influence of β-adrenoceptor ligands and a β-adrenoceptor-derived peptide was studied on stimulation of adenylyl cyclase in membrane preparations of turkey erythrocytes. In the absence of receptor ligands, the hydrolysis-resistant GTP analogs, guanosine 5′-[γ-thio]triphosphate and guanosine 5′-[β,γ-imino]triphosphate, caused a slow, time-dependent increase in adenylyl cyclase activity, which was accelerated and potentiated by the additional presence of the agonist, isoproterenol. In contrast, the β-adrenoceptor antagonists, propranolol and pindolol, almost completely prevented stimulation of adenylyl cyclase by the GTP analogs alone, i.e. in the absence of an agonist, in a concentration-dependent and stereo-selective manner. This antagonist action was mimicked by a peptide corresponding to the second cytoplasmic loop of the turkey erythrocyte β-adrenoceptor. On the other hand, GTP analog-preactivated cyclase activity and enzyme stimulation by fluoride and forskolin were not or only slightly β-adrenoceptors can cause significant G s protein and subsequent adenylyl cyclase activation, and that this unoccupied receptor action can be blocked by β-adrenoceptor antagonists, thus exhibiting by themselves a negative intrinsic activity.

Evidence for a novel adenylyl cyclase in human epididymal sperm.

Since cAMP is considered to play a major role in the acquisition of maturation and fertilizing capacity of mammalian sperm, we investigated the expression of cAMP-synthesizing adenylyl cyclase (AC) in sperm retrieved directly from the human epididymis. Particulate fractions were prepared from purified epididymal sperm samples and AC was monitored by the direct conversion of ATP into cAMP. We report that in great contrast to human ejaculated sperm and other mammalian sperm cells, the human epididymal sperm do not express a Mn(2+)-sensitive AC. However, a functional AC was readily detectable in these sperm cells in the presence of saturating concentrations of Ca2+ (50mM) and bicarbonate (HCO3-, 50mM), a combination that causes maximal activation in human ejaculated sperm. Using these conditions, human epididymal sperm AC showed similar capacity to generate cAMP compared to human ejaculated sperm AC. When assays were performed in the presence of Mg2+ and a saturating concentration of GMP-P(NH)P (50 microM), the hydrolysis-resistant GTP analog, and forskolin (100 microM), no activity was detected indicating that the epididymal sperm AC differs from that in somatic cells. These data demonstrate that human epididymal sperm contain an AC that is unique and different from the enzyme system described in somatic cells and other mammalian sperm cells, including human ejaculated sperm.

Presynaptic glutamate receptors depress inhibitory postsynaptic transmission in lobster neuromuscular synapse.

1. We examined the functional role of GTP-coupled glutamate receptor (GluB-R) in the presynaptic membrane of lobster neuromuscular synapse. 2. Injection of guanosine 5'-O-(3-thiotriphosphate) (GTP gamma S), a hydrolysis-resistant analogue of GTP, into the excitatory axon mimicked the presynaptic glutamate response and effectively suppressed excitatory postsynaptic potentials or excitatory postsynaptic currents (EPSCs). 3. Statistical analysis revealed that the coefficient of variation (standard deviation divided by the mean of EPSC amplitude) was increased after injection of GTP gamma S into the excitatory axon, indicating a presynaptic inhibition of transmitter release. 4. The effect of glutamate on inhibitory postsynaptic potentials (IPSPs) or inhibitory postsynaptic currents (IPSCs) was studied when the postsynaptic glutamate receptors were blocked by the Joro spider toxin (JSTX). Glutamate depressed IPSPs or IPSCs in the JSTX-treated preparation. Furthermore, repetitive stimulation of the excitatory nerve produced effective inhibition of IPSCs. 5. Quisqualate and kainate suppressed IPSCs in a similar way to glutamate. In contrast, N-methyl-D-aspartate, ibotenic acid, trans-D,L-1-amino-1,3-cyclopentanedicarboxyloc acid, and 2-amino-4-phosphonobutanate had no effect on GluB-R. 6. Our results indicate that GluB-R, which exists in both excitatory and inhibitory nerve terminals, regulates transmitter release by a presynaptic inhibitory mechanism.

Regulation of Ca2+ current in frog ventricular cardiomyocytes by guanosine 5'-triphosphate analogues and isoproterenol.

Calcium currents (ICa) were measured in frog ventricular myocytes using the whole-cell patch clamp technique and a perfused pipette. To gain insight into the role of G proteins in the regulation of ICa in intact cells, the effect of internal perfusion with hydrolysis-resistant GTP analogues, guanylyl 5'-imidodiphosphate (GppNHp) or guanosine 5'-thiotriphosphate (GTP gamma S), on ICa stimulated by isoproterenol (Iso) or forskolin (Forsk) was examined. Significant differences were observed between the effects of the two GTP analogues. Internal perfusion of GppNHp resulted in a near-complete (approximately 80%) and irreversible inhibition of Iso-stimulated ICa. In contrast, internal perfusion with GTP gamma S resulted in only a partial (approximately 40%) inhibition of Iso- or Forsk-stimulated ICa. The fraction of the current not inhibited by GTP gamma S remained persistently elevated after the washout of Iso but declined to basal levels upon washout of Forsk. Excess internal GTP or GppNHp did not reduce the persistent ICa. Internal adenosine 5'-thiotriphosphate (ATP gamma S) mimicked the GTP gamma S-induced, persistent ICa. GppNHp sometimes induced a persistent ICa, but only if GppNHp was present at high concentration before Iso exposure. Inhibitors of protein kinase A inhibited both the GTP gamma S- and ATP gamma S-induced, persistent ICa. We conclude that: (a) GTP gamma S is less effective than GppNHp in inhibiting adenylyl cyclase (AC) via the inhibitory G protein, Gi; and (b) the persistent ICa results from a long-lived Gs-GTP gamma S complex that can activate AC in the absence of Iso. These results suggest that different hydrolysis-resistant nucleotide analogues may behave differently in activating G proteins and imply that the efficacy of G protein-effector molecule interactions can depend on the GTP analogue with which the G protein is activated.

GTPγS stimulates exocytosis in patch-clamped rat melanotrophs

We investigated the molecular mechanisms regulating exocytosis in patch-clamped melanotrophs by measuring the membrane capacitance. Ca 2+-dependent exocytosis could be induced by membrane depolarization or by including solutions containing 2 μM free Ca 2+ in the patch pipette. Caz+-dependent exocytosis was inhibited by GDPβS, suggesting involvement of a GTP-binding protein. The hydrolysis-resistant GTP analogs, GTPγS and GppNHp, were able to stimulate exocytosis at low free Ca 2+ concentrations. The stimulatory response to GTPγS was abolished by both GDPβS and GTP. The latter suggests that a sustained activation of a GTP-binding protein is necessary for exocytosis. This behavior is similar to the stimulation of exocytosis by guanine nucleotides in mast cells and other nonexcitable cells and suggests a common regulatory mechanism.
