Negative terpinen-4-ol modulate potentially malignant and malignant lingual lesions induced by 4-nitroquinoline-1-oxide in rat model.

Our aim was to verify the modulative TP-4-ol capacity in 4-nitroquinoline-1-oxide induced oral rat cancer. The stereoisomers of TP-4-ol were used against the human tongue squamous cell line and the negative stereoisomer showed lower IC50. Thirty-one Holtzman rats (120-130g) were cancer-induced by 4-nitroquinoline-1-oxide (4-NQO/8weeks/25ppm) and 32 Holtzman rats (120-130g) were used to healthy and TP-4-ol toxicity experiments. Six groups were used, healthy, 0.1nL/g of TP-4-ol, 8nL/g of TP-4-ol, 4-NQO, 4-NQO + 0.1nL/g of TP-4-ol, and 4-NQO + 8nL/g of TP-4-ol. We performed the toxicity analysis by biochemical and histopathological analysis. The biochemistry analysis includes alkaline phosphatase (ALP), alanine aminotransferase (ALT), aspartate transaminase (AST), urea, and creatinine and the histopathology analysis includes the liver, kidney, lung, and spleen. Specifically, for malign modulation, we performed a macroscopic and microscopic analysis. The group exposed to 0.1nL/g of TP-4-ol demonstrated a reduced risk of malignancy in dysplasia considering the criteria of architecture and cytology. Similarly, a drop of percentual rats with SCC diagnosis was observed in 4-NQO + 0.1nL/g (41.6%) when compared to 4-NQO (87.5%). Moreover, the 4-NQO group presented a median of 2.62 SCC/rat and the 4-NQO + 0.1nL/g demonstrated a median of 0.75 SCC/rat. For toxicity analysis, 4-NQO + 0.1nL/g showed focal necrosis in the kidney and 4-NQO showed lung hemorrhagic areas. The concentration of 0.1nL/g was more effective in reducing the tongue induction of potentially malignant and malignant lesions by 4-NQO. A kidney toxicity was observed in healthy animals exposed to 0.1nL/g of TP-4-ol. The negative isoform of terpinen-4-ol negatively modulates the development of potentially malignant and malignant lesions in rats (Rattus nonverdicts albinos, Holtzman) exposed to 4-NQO. (-)-Terpinen-4-ol reduced the mice percentual with squamous cell carcinoma, 87.5 to 41.6%, and decreased the cancer/rat ratio of 2.62 in 4-NQO to 0.75 in 4-NQO + 0.1nL/g. This represents 52.4% by group and 71.3% in the cancer/rat ratio.

Model of Human Tongue Squamous Cell Lines Stably Transfected with Human Papillomavirus (HPV)16 E6 and E7 Genes and Biological Characteristic Analysis.

Background Human tongue squamous cell carcinoma (TSCC) is the most common oral cancer with the highest human papillomavirus (HPV) infection rate in oral cancer. The purpose of this study was to research the correlation between HPV and TSCC. Method Plasmid pEGFP/HPV16 E6E7 and plasmid pEGFP/no HPV16 E6E7 were constructed. TSCC cell lines SCC9 and SCC15 were infected by liposome transfection and would be highly selected by antibiotic. Fluorescence imaging, PCR, and Western blot were used to detect the expression of HPV16 E6E7 in cells. The biological characteristics were detected by CCK-8, wound healing assay, qRT-PCR, and Western blot. Result TSCC cell lines transfected with HPV16 E6E7 gene were successfully established and identified. And the proliferation and migration ability of the TSCC cell lines infected with HPV16 E6E7 gene were significantly stronger than that of the blank group. Conclusion TSCC cell lines infected with HPV16 E6E7 with significantly higher ability of proliferation and migration were more malignant than those not infected with HPV16 E6E7.

Crosstalk between Fisetin-induced Apoptosis and Autophagy in Human Oral Squamous Cell Carcinoma.

Fisetin (3,3-,4-,7-tetrahydroxyflavone), a naturally occurring flavonoid, has antioxidant, anti-inflammatory, and anticancer effects. Oral squamous cell carcinoma (OSCC) has a 5-year survival rate lower than that of most other carcinomas, and can create functional and aesthetic problems for the patient. New therapies for OSCC are necessary, and treatment using plant-derived natural substances has recently become a trend. It has been suggested that autophagy may play an important role in cancer therapy. Several studies demonstrated that autophagy inhibition enhances apoptotic cell death. Therefore, autophagy inhibition might be a promising therapeutic method against OSCC. Our results showed that fisetin induced apoptotic cell death in human tongue squamous cell line Ca9-22 could be enhanced by inhibition of autophagy. Thus, autophagy process in fisetin treated OSCC might presumed to play a role of pro-survival. The combination of fisetin and an effective autophagy inhibitor could be a potentially adjuvant and useful treatment for oral cancer.

Two New Eu(III)-Based Coordination Polymer Nanoparticles Constructed from the V-Shaped Carboxylic Linkers: Synthesis, Crystal Structures and Anti-oral Cancer Studies

Nowadays, great attention has been focused on the development of nanomedicine for their various advantageous properties. In this work, two new mixed ligand coordination polymers [Eu2(PDDA)2(ox)(phen)2](DMA)2 (1, H2ox = oxalic acid, phen = 1,10-phenanthroline, H2PDDA is in short of 4,4′-(pyridine-2,6-diyl)dibenzoic acid) and [Eu2(bpda)3(phen)2(H2O)2](H2O)3 (2, H2bpda is in short of 1,1′-biphenyl-3,3′-dicarboxylic acid) have been prepared by an one pot reaction of Eu(NO3)3·6H2O with two newly designed bicarboxylic acid ligands, which demonstrate interesting structural diversity depending on the assistant ligands used. Complex 1 is a three-dimensional pillar-layered network showing a two-fold interpenetrated alpha-Po primitive cubic type 6-connected net, while complex 2 reveals a 2D layered network with a sql/Shubnikov tetragonal topology. Furthermore, both complexes have been successfully downsized to nanoscale morphology to study their anticancer activities against the human tongue squamous cell lines CAL27 via the MTT assay, which reveals that the anticancer activities of the both complexes are greatly enhanced by miniaturizing to nanoparticles.

Influence of COX‐2 inhibitor on expression of COX‐2, VEGF, Ang‐1 and Ang‐2 gene in human tongue squamous cell line Tca‐8113

Through observing the change of COX‐2, vascular endothelial growth factor, angipoietin‐1 and angiopoietin‐2 gene in human tongue squamous cell carcinoma Tca‐8113 cell line by ihibiting the activity of cox‐2 to investigate the affect and mechanism of the selective COX‐2 inhibitor nimesulide in the biological behavior of human tongue squamous cell carcinoma.Human tongue squamous cell carcinoma Tca‐8113 cell were cultured in vitro and treated with NIM for 48h to measure the expression of COX‐2, VEGF, Ang‐1 and Ang‐2 mRNA in Tca‐8113 cells by the semiquantitative reverse transcription polymerase chain reaction analysis.Human tongue squamous cell carcinoma Tca‐8113 cell were treated with NIM for 48h,the expression of COX‐2 ,VEGF, Ang‐1, Ang‐2 mRNA level between the each treated group and the control group was statistical significance(P<0.01), The expression of COX‐2, VEGF mRNA reduced in a concentration depengent manner r= ‐0.963,0.934 . Human tongue squamous cell carcinoma Tca‐8113 cell were treated with NIM,the expression of COX‐2, VEGF, Ang‐1 ,Ang‐2 mRNA level in the each treated group compared with the control group was reduced, and the reduced in a concentration depengent manner.

Celecoxib enhances the lethal effects of bleomycin in human tongue squamous carcinoma cell line Tca8113

To investigate the role of COX-2 inhibitor celecoxib in enhancing the lethal effects of bleomycin in Tca8113 cell line. Tca8113 cells were treated with different concentrations of celecoxib and bleomycin for 24, 48, 72 h. Methyl thiazolyl tetrazolium assay was used to calculate cell growth inhibition rate and Jin Zheng Jun's method was used to evaluate the interaction of celecoxib and bleomycin on Tca8113 cells. Flow cytometry was used to evaluate the effects of combined use of celecoxib and bleomycin on cell cycle progress and apoptosis. Low dose of celecoxib (10 micromol/L, < IC(50)) combined with bleomycin showed synergism or additive lethal effect on Tca8113 cell line. Celecoxib could notably enhance the inhibitory effect of bleomycin on Tca8113 cells by blocking cell cycle progress and thus resulting in the increasing G(0)/G(1) cells [(60.93 +/- 0.32)%] distribution and inducing apoptosis [(1.87 +/- 0.11)%]. Low doses of celecoxib could significantly enhance the lethal effect of bleomycin on Tca8113 cells by inhibiting cell growth and proliferation through blocking cell cycle progress and inducing apoptosis. The ways of these interactions on inhibiting Tca8113 cell growth were synergistic or/and additive.
