Effect of rovatirelin, a novel thyrotropin-releasing hormone analog, on the central noradrenergic system

Rovatirelin ([1-[-[(4S,5S)-(5-methyl-2-oxo oxazolidin-4-yl) carbonyl]-3-(thiazol-4-yl)-l-alanyl]-(2R)-2-methylpyrrolidine) is a novel synthetic agent that mimics the actions of thyrotropin-releasing hormone (TRH). The aim of this study was to investigate the electrophysiological and pharmacological effects of rovatirelin on the central noradrenergic system and to compare the results with those of another TRH mimetic agent, taltirelin, which is approved for the treatment of spinocerebellar degeneration (SCD) in Japan. Rovatirelin binds to the human TRH receptor with higher affinity (Ki=702nM) than taltirelin (Ki=3877nM). Rovatirelin increased the spontaneous firing of action potentials in the acutely isolated noradrenergic neurons of rat locus coeruleus (LC). The facilitatory action of rovatirelin on the firing rate in the LC neurons was inhibited by the TRH receptor antagonist, chlordiazepoxide. Reduction of the extracellular pH increased the spontaneous firing of LC neurons and rovatirelin failed to increase the firing frequency further, indicating an involvement of acid-sensitive K+ channels in the rovatirelin action. In in vivo studies, oral administration of rovatirelin increased both c-Fos expression in the LC and extracellular levels of noradrenaline (NA) in the medial prefrontal cortex (mPFC) of rats. Furthermore, rovatirelin increased locomotor activity. The increase in NA level and locomotor activity by rovatirelin was more potent and longer acting than those by taltirelin. These results indicate that rovatirelin exerts a central nervous system (CNS)-mediated action through the central noradrenergic system, which is more potent than taltirelin. Thus, rovatirelin may have an orally effective therapeutic potential in patients with SCD.

Taltirelin is a superagonist at the human thyrotropin-releasing hormone receptor

Taltirelin (TAL) is a thyrotropin-releasing hormone (TRH) analog that is approved for use in humans in Japan. In this study, we characterized TAL binding to and signaling by the human TRH receptor (TRH-R) in a model cell system. We found that TAL exhibited lower binding affinities than TRH and lower signaling potency via the inositol-1,4,5-trisphosphate/calcium pathway than TRH. However, TAL exhibited higher intrinsic efficacy than TRH in stimulating inositol-1,4,5-trisphosphate second messenger generation. This is the first study that elucidates the pharmacology of TAL at TRH-R and shows that TAL is a superagonist at TRH-R. We suggest the superagonism exhibited by TAL may in part explain its higher activity in mediating central nervous system effects in humans compared to TRH.

Effect of thyrotropin-releasing hormone on the locomotor and sensitizing effects of cocaine in rats

The present study was designed to find out whether single and repeated treatment with thyrotropin-releasing hormone (TRH) changed the cocaine-evoked hyperactivation or sensitization, and whether cross-sensitization occurred between TRH and cocaine. Like cocaine (10 mg/kg), TRH (10 mg/kg) increased the basal activation of rats; however, when given in combination with cocaine (10 mg/kg), TRH (5–10 mg/kg) did not change the locomotor effect of cocaine. On day 10, cocaine challenge of rats treated repeatedly with the psychostimulant (days 1–5) significantly enhanced locomotor hyperactivity compared to the effect of acute cocaine injection in saline-treated (days 1–5) animals. When co-administered with cocaine for 5 days during the development of sensitization, TRH (10 mg/kg) enhanced the effect of the challenge dose of cocaine (10 mg/kg) following a 5-day withdrawal. Given acutely with cocaine on day 10 to cocaine-treated animals, TRH (5–10 mg/kg) did not change the expression of cocaine sensitization. Significant enhancement of the locomotor response to TRH (10 mg/kg) challenge was observed in animals treated repeatedly with TRH. The response to TRH (5–10 mg/kg) was stronger in repeated cocaine-treated rats than in saline-injected ones; similarly, the response to cocaine (10 mg/kg) was more potent in TRH-treated animals compared to saline-injected ones (cross-sensitization). In conclusion, our results indicate that exposure to TRH induces sensitization to its locomotor hyperactivity effect. They also show that TRH enhances the development of cocaine sensitization, but affects neither the expression phase of the phenomenon nor the locomotor hyperactivity induced by a single dose of cocaine. Moreover, cross-sensitization between cocaine and TRH has also been demonstrated. These findings also may provide an insight into the relationship between TRH and cocaine in humans exposed to the psychostimulant.

Thyrotropin-Releasing Hormone Receptor (TRHR) Gene Is Associated With Essential Hypertension

In essential hypertension, a polygenic and multifactorial syndrome, several genes interact with the environment to produce high blood pressure. Thyrotropin-releasing hormone (TRH) plays an important role in central cardiovascular regulation. We have described that TRH overexpression induces hypertension in a normal rat, which was reversed by TRH antisense treatment. This treatment also reduces the central TRH hyperactivity in spontaneously hypertensive rats and normalizes blood pressure. Human TRH receptor (TRHR) belongs to the G protein-coupled seven-transmembrane domain receptor superfamily. Mutations of these receptors may result in constitutive activation. As it has been demonstrated that hypertensive patients have a blunted TSH response to TRH injection, suggesting a defect in the TRHR, we postulate that the TRHR gene is involved in human hypertension. We studied 2 independent populations from different geographic regions of our country: a sample of adult subjects from a referral clinic and a population-based sample of high school students. In search of molecular variants of TRHR, we disclosed that a polymorphic TG dinucleotide repeat (STR) at -68 bp and a novel single nucleotide polymorphism, a G-->C conversion at -221 located in the promoter of the TRHR are associated with essential hypertension. As STRs detected in gene promoters are potential Z-DNA-forming sequences and seem to affect gene expression, we studied the potentially different transcriptional activity of these TRHR promoter variants and found that the S/-221C allele has a higher affinity than does the L/G-221 allele to nuclear protein factor(s). Our findings support the hypothesis that the TRHR gene participates in the etiopathogenesis of essential hypertension.

Transcription of the Human Thyrotropin-Releasing Hormone Receptor Gene—Analysis of Basal Promoter Elements and Glucocorticoid Response Elements

The gene for the human thyrotropin-releasing hormone receptor (TRHR) spans 35 kb and contains three exons and two introns (Matreet al.(1999)J. Neurochem.72, 1–11). Despite a reported transcription start site (TSS) mapped to position −885 upstream of the translation initiation codon (Iwasakiet al.(1996)J. Biol. Chem.271, 22183–8), we found cell type specific promoter activity directed by a fragment downstream of this site (−770 to +1). To elucidate the basis for this unexpected activity, we analyzed basal promoter elements in this region of the gene. One divergent TATA box, TTTAAA in position −759, was found by mutational analysis to be critical for promoter activity, providing a likely explanation for the basal activity observed. This proximal region apparently contains several promoter elements, including Pit-1 binding sequences within the first intron of the TRHR gene as previously reported. Here we describe the analysis of two putative glucocorticoid response elements (GREs) that we identified in this region, one (distal) half site overlapping the proposed TSS at −885 and one (proximal) full site within the first intron at position −624. Accordingly, stimulation of rat pituitary GH3and GH4C1cells with dexamethasone strongly enhanced transcription activity of a reporter construct containing the distal GRE half site and the proximal GRE site. Both sites bound the glucocorticoid receptor (GR) in a specific manner. Deletion of the distal GRE half site abolished the dexamethasone induction of CAT transcription, as did mutations in the proximal site. We therefore conclude that both sites are necessary for regulation of the TRHR gene transcription by glucocorticoids.

Human thyrotropin-releasing hormone-associated peptide 3 (hTAP-3) in serum.

Human thyrotropin-releasing hormone (TRH)-associated peptide 3 (hTAP-3), one of the cryptic peptides resulting from the proteolytic processing of preproTRH to produce TRH, was measured in human plasma from normal, hyperthyroid, and hypothyroid subjects. The dilution curve of hTAP-3 immunoreactivity in the serum paralleled the standard curve of the radioimmunoassay. HPLC analysis revealed a single strong immunoreactive peak, which corresponded to the authentic peptide, hTAP-3. The half-life of hTAP-3 in serum was approximately 3.5 min, and the addition of aprotinin and EDTA completely prevented its degradation. In hyperthyroid patients, plasma concentrations of hTAP-3 were significantly higher than those in the control group and hypothyroid patients, but no correlation was found between its level and that of thyroid hormone. These findings indicate the existence of intact hTAP-3 in the human serum and increases in plasma hTAP-3 levels in hyperthyroid patients, suggesting that blood hTAP-3 may be derived from the peripheral organs rather than the hypothalamus.

The thyrotropin-releasing hormone gene 1998: cloning, characterization, and transcriptional regulation in the central nervous system, heart, and testis.

Mechanisms of triiodothyronine (T3) negative regulation of the human thyrotropin-releasing hormone (TRH) gene were investigated with a chimeric construct of the 5' flanking region fused to a luciferase reporter gene, transfected into human neuroblastoma cells (HTB-11). Maximum negative regulation was achieved with constructs containing bases -242 to +54. Four sequences in this region exhibited homology with half sites of thyroid hormone response elements (TRE) (AGGTCA). The most important site was a sequence with an overlapping TRE/CRE, involving bases -53 to -60 (TGACCTCA). Potential combinatorial interactions of thyroid hormone receptors and CREB at this site were explored. Modest promoter stimulation was achieved with dibutyryl cyclic adenosine monophosphate (cAMP) (10(-3) M) plus IBMX (0.5 mM). Stimulation was greatly enhanced (+820%) by cotransfection of a constitutively activated protein kinase A (pPKA) construct. Cotransfection with pCREB increased stimulation further to 1350% above control. Stimulation of pPKA and pCREB interfered with stimulation by unliganded TRbeta1, and co-transfected pPKA and pCREB blocked T3 negative inhibition by TRbeta1-T3 complexes. When this site was mutated by polymerase chain reaction (PCR) mutagenesis, the mutant construct failed to respond to unliganded TRbeta1, and stimulation by pPKA and/or pCREB was inhibited markedly, from 12.5- to 2.1-fold, p < 0.001. Moreover, TRbeta1-T3 complexes failed to show any inhibition of the mutated promoter. These results suggest that negative regulation is achieved by inhibition of CREB stimulation of the TRH promoter at this overlapping TRE/CRE site. The two cosuppressors, NCoR and SMRT, were able to augment stimulation of the TRH promoter by unliganded TRbeta1 and enhance the magnitude of T3 inhibition. The potential role of the TRH gene and the pathophysiology of thyroid hormone resistance was investigated with three mutant TRbeta1 constructs. Thyroid hormone resistance was found to be expressed at the level of TRH gene regulation, due to lowered inhibition by mutant TRbeta1-T3 complexes and by their dominant negative effects on wild-type TRbeta1-T3 inhibition. TRH gene expression has been identified in the heart. Cardiac TRH mRNA was not regulated by T3, in contrast to HTB-11 cells, but cardiac TRH mRNA density could be augmented by glucocorticoids and by testosterone. TRH receptors were identified using Scatchard blots that showed a kilodalton of 1.4 nM and a bmax of 10 pmol/mg protein. TRH-R mRNA was identified also by reverse transcription polymerase chain reaction (RT-PCR). Enhanced ventricular contractility by TRH was demonstrated in both an open-chested dog preparation and in ex vivo ventricular myocytes, using video edge cinematography. Under controlled conditions, myocyte shortening was 13.3%, and TRH (10(-6) M) caused muscle shortening to increase 140%, (p < 0.005). TRH gene expression was demonstrated exclusively in Leydig cells of the testis. High affinity binding sites were identified in testicular membranes with a kilodalton of 1.6 x 10(-6) M. TRH was able to inhibit LH and HCG-activated testosterone secretion significantly. Thus, one paracrine role of TRH in the testis may be to serve as inhibitory modulator of gonadotropin-stimulated testosterone secretion.

Hydropathy Profiles of Predicted Thyrotropin‐Releasing Hormone Precursors are Highly Conserved Despite Low Similarity of Primary Structures

Two types of cDNAs encoding thyrotropin-releasing hormone (TRH) precursors (TRH-A and TRH-B) were amplified from hypothalamic mRNA of sockeye salmon by reverse transcriptase-polymerase chain reaction (RT-PCR). The amplification was achieved using two primers which correspond to TRH progenitor sequence (Lys/Arg-Arg-Gln-His-Pro-Gly-Lys/Arg-Arg). A full length cDNA encoding TRH-A was obtained by 5'- and 3'-RACE methods. It has a length of 1324 base pairs (bp) that contains sequences of 5' and 3' untranslated regions and an open reading frame of 259 codons. The sockeye salmon TRH-A deduced from the nucleotide sequence tandemly contains 8 copies of TRH progenitor sequences. Another cDNA which encodes a part of TRH-B consists of 242 bp, and the sequence homology between TRH-A and -B cDNAs is 90%. The result of Southern blot analysis of sockeye and masu salmon genomic DNAs supported the evidence that there are at least two TRH genes in the salmonid. A RT-PCR analysis of TRH gene expression in various tissues of sockeye salmon showed that strong expression was observed only in the brain. The primary structure of the sockeye salmon TRH-A shares low similarity to those of human, rat and Xenopus TRH precursors (35, 27 and 44%, respectively). However, their hydropathy profiles were almost the same with each other. The profile of sockeye salmon TRH-A showed the presence of two discrete hydrophobic regions, one in the N-terminal region which corresponds to the signal peptide and the other in the C-terminal region. All of the repetitive TRH progenitor sequences are included in three hydrophilic regions easily recognizable. The present results thus suggest that the three-dimensional structures of TRH precursors are highly conserved, although the primary structures of TRH precursors have diverged through the evolutionary pathway of vertebrates.

Genomic Organization and Promoter Function of the Human Thyrotropin-releasing Hormone Receptor Gene

We isolated and characterized the gene for the human thyrotropin-releasing hormone receptor. The gene spanned more than 30 kilobases and contained three exons and two introns. Intron 1 exists in the 5'-untranslated region, and intron 2 is more than 25 kilobases in length which interrupts the coding region before the beginning of the putative sixth transmembrane domain. Exon 3 encodes the rest of the coding region and the entire 3'-untranslated region. The 3'-flanking region contains four potential polyadenylation signals, and 3'-rapid amplification of cDNA ends studies showed that only a signal at 2076 base pairs downstream of the stop codon was functional in the anterior pituitary. Primer extension and anchor-polymerase chain reaction studies indicated a transcriptional start site at 344 base pairs upstream of the translational start site. The promoter region does not contain either a TATA box or a CAAT box in the appropriate location. Transient transfection study revealed significant activity of the promoter in GH4C1 cells, and the region between -338 and -933 bp from the transcriptional start site worked as a negative regulator. Knowledge of the genomic organization and the promoter region of thyrotropin-releasing hormone (TRH) receptor gene will allow further studies of possible disorders of the TRH receptor, as well as facilitate elucidation of transcriptional control of the human TRH receptor gene.

Reversal of TR-T3 inhibition of the hTRH gene by excess TR ligand-binding domain: evidence for novel accessory protein.

The thyrotropin-releasing hormone (TRH) gene is regulated negatively at the transcriptional level by thyroid hormone (T3). T3 positive regulatory effects on other target genes, such as the growth hormone gene, are mediated through heterodimerization of thyroid hormone receptors (TRs) with RXR or other auxiliary nuclear protein(s). To explore whether an accessory co-suppressor protein(s) may be involved in T3 inhibitory regulation of human TRH gene transcription, transient gene expression studies have been carried out using a hTRH-luciferase (TRH-Luc) chimetric reporter construct, an hTR beta 1 expression construct, and pABgal-hTR beta 1 ligand-binding domain (LBD) fusion constructs, cotransfected into a human neuroblastoma cell line (HTB-11,ATCC). Results herein indicate that T3-dependent inhibitory regulation (48-60% of control) of the hTRH gene promoter by hTR beta 1-T3 complexes could be abrogated completely by cotransfection of a 10 x excess of hTR beta 1-LBD (TR 168-456 aa) in a pABgal94 vector. In striking contrast, cotransfection of a 10 x excess of highly truncated hTR beta 1-LBD (TR 452-456 aa) failed to reverse T3-mediated TRH promoter inhibition. This squelching effect by excessive intact TR-LBD, moreover, could not be reversed by raising T3 concentration 100-fold (from 10(-8) to 10(-6) M), thus excluding a squelching effect of T3 itself by excess LBD. These results suggest that negative regulation of the hTRH gene promoter activity by TR beta 1-T3 complexes involves interactions with an accessory co-suppressor protein, which may bridge DNA-bound TR beta 1-T3 complexes to the transcriptional initiation complex.

Activation of the thyrotropin-releasing hormone (TRH) receptor by a direct precursor of TRH, TRH-Gly

We studied the mechanism by which thyrotropin-releasing hormone (TRH)-Gly stimulated prolactin and thyrotropin (TSH) secretion in pituitary, using a pituitary mammotropic cell line, GH3 cells, and a cell line stably expressing a human TRH receptor (TRH-R). In GH3 cells expressing endogenous TRH-R, an addition of TRH-Gly evoked an immediate rise of intracellular calcium concentration, indicating that TRH-Gly reacted directly without converting from TRH-Gly to TRH. In order to determine whether this reaction might occur through TRH-R, we established a cell line stably expressing a human TRH-R, by transfecting a human TRH-R cDNA into Chinese hamster ovary cells (CHO cells). In this cell line, 10 nM TRH elevated intracellular calcium significantly; the K d for McTRH was 1.7 nM. One micromolar and 100 nM TRH-Gly also elevated intracellular concentration of calcium significantly, but not in CHO cells which were not transfected with the TRH-R cDNA. Competition studies further revealed that TRH-Gly displaced McTRH binding (IC 50, 12μM). These data indicate that at high concentration, TRH-Gly interacts directly with TRH-R to activate signal transduction pathway, and that release of prolactin and TSH induced by TRH-Gly in vitro may be due, at least in part, to the direct effect of TRH-Gly on the TRH-R.

TRH is the principal positive regulator of TSH synthesis and secretion in man. T3 is able to control TRH synthesis through feedback inhibition at the transcriptional level, presumably by binding to its receptor which interacts with one or more negative thyroid hormone response elements (TREs) present within the human TRH promoter. In the present study we have identified the specific negative TREs within the TRH promoter and characterized their ability to interact with thyroid hormone receptors (TRs), and the retinoid X receptor (RXR). Our analysis demonstrates that ligand-independent and dependent regulation of the human TRH promoter is restricted to the TR beta 1 isoform. Deletional analysis of the TRH promoter identified two discrete regions that are responsible for mediating ligand-dependent negative regulation of the TRH promoter. Mutagenesis of potential TR binding half-sites within these regions identified three separate half-sites (site 4 from -55 to -60 base pairs (bp); site 5, +14 to +19 bp; and site 6, +37 to +42 bp) which act in combination to allow for negative regulation. Mutation and/or deletion of each of these sites leads to a loss of negative regulation of the TRH promoter by T3. Gel-mobility shift assays of site 4 and its surrounding nucleotides revealed that this region of the promoter is capable of binding TR monomers, homodimers, and TR-RXR heterodimers. Mutagenesis of site 4 leads to a loss of all binding to this region. The region encompassing sites 5 and 6 binds only TR monomer, and the addition of RXR to the binding reaction leads to a loss of specific monomeric binding. To assess the functional importance of site 4 and its surrounding nucleotides we cotransfected RXR isoforms along with TR beta with TRH promoter constructs containing either site 4 or its mutant. In the presence of wild type site 4 sequence, cotransfected RXR enhanced negative regulation of the TRH promoter. Mutation and or deletion of site 4 leads to a loss of this enhancement. These data demonstrate that two structurally different negative TREs cooperate to allow for negative regulation of the human TRH promoter and that negative regulation is TR isoform-specific and modulated by the RXR-signaling pathway through a novel negative TRE.

Molecular cloning and functional expression of a human thyrotropin-releasing hormone (TRH) receptor gene

In this study, we isolated genomic DNA fragments coding for the human thyrotropin-releasing hormone (TRH) receptor. Analysis of the nucleotide sequence revealed that the human TRH receptor gene had an exon-intron structure comprising at least two exons. A polypeptide encoded by the gene consisted of 398 amino acid residues with putative seven transmembrane domains. It showed high homology as a whole amino acid sequence with the rat and mouse TRH receptors except for considerable variation in the C-terminal region. Chromosomal mapping study indicated that the human TRH receptor gene was assigned to chromosome 8. Chinese hamster ovary (CHO) cells transfected with a DNA fragment containing the coding regions of the human TRH receptor bound with [ 3H]TRH. This binding was inhibited by adding unlabeled TRH in a dose-dependent fashion. Scatchard analysis indicated that the transfected CHO cells expressed a single class of high affinity binding sites at a dissociation constant ( K d) of approximately 1 nM. These results demonstrated that the isolated gene encoded a specific TRH receptor with high affinity.

Assignment of the gene encoding the human thyrotropin-releasing hormone receptor to 8q23 by fluorescence in situ hybridization.

A cDNA for human thyrotropin-releasing hormone (TRH) receptor has been isolated from a human pituitary cDNA library. By using this cDNA as a biotinylated probe, the gene encoding the TRH receptor has been localized to chromosome 8q23 by in situ hybridization.

Ligand T3 Dependent and Independent Effects of Thyroid Hormone Receptors upon Human TRH Gene Transcription in Neuroblastoma Cells

Thyrotropin releasing hormone (TRH) gene is regulated negatively at the transcriptional level by thyroid hormone (T3) in rat anterior hypothalamus. The actions of T3 upon other target genes are known to be mediated through the thyroid hormone receptors (TR), TRα and TRβ. To explore whether the inhibitory regulation of human (h) TRH gene transcription by T3 is TR isoform specific and whether TRH gene transcription can be modulated as well by unliganded TR isoforms, transient gene expression studies have been carried out using hTRH-luciferase (TRH-Luc) chimeric constructs and TR expression constructs, co-transfected into a human neuroblastoma cell line (HTB-11). Data herein demonstrate T3-dependent inhibitory regulation of the hTRH gene promoter by TR-T3 complexes. Moreover, significant inhibition (39%-60%) could be achieved by T3 bound to either hTRα1, hTRβ1, or rTRβ1, β2 and was comparable quantitatively, indicating an absence of TR isoform specificity for T3 inhibition. Conversely, basal promoter activity of the hTRH gene could be activated significantly by unliganded hTRα1, β1, rTRβ1, and β2 (150% to 334%), but not by hTRα2. Thus, TRs appear to exert opposite effects on hTRH gene transcription, depending on the presence or absence of ligand T3. These dual effects of TR suggest that the addition of the T3 ligand effects conformational changes that can abrogate the initiation of transcription.

Scopolamine effects on the pressor response to thyrotropin-releasing hormone in humans

Thyrotropin-releasing hormone (TRH) produces a marked pressor effect, which may be mediated by central cholinergic neurons, which in turn enhance sympathetic nervous system activity. In this study, 22 subjects (10 patients with Alzheimer's disease and 12 elderly controls) were administered IV scopolamine or placebo prior to administration of IV high-dose TRH (0.5 mg/kg). Systolic blood pressure was less on the day scopolamine was administered prior to TRH administration, as compared with placebo (F[1,20]=6.12, p<0.02). Results indicate that the pressor effect of TRH is attenuated by scopolamine, indicating a role of the cholinergic system in this response in humans.

Assignment of human thyrotropin-releasing hormone (TRH) receptor gene to chromosome 8.

The human gene encoding thyrotropin-releasing hormone receptor was assigned to chromosome 8, using human-Chinese hamster ovary somatic cell hybrids, analyzed by Southern hybridizations. Hybridization was carried out with a 32P-labeled fragment of the human TRH-R genomic DNA. Hybridization of this probe to a human specific 10.5-kb DNA fragment of EcoRI-digested WBC DNA was used to localize the human TRH-R gene. No hybridization, by contrast, was seen with this probe and hamster DNA after EcoRI treatment. Results from 18 somatic cell hybrids corroborated unequivocally that the human TRH-R gene can be assigned to human chromosome 8.

We cloned a human thyrotropin-releasing hormone receptor (TRH-R) gene and its pituitary cDNA, and examined whether TRH-R mRNA may be expressed and function in the GH-secreting pituitary adenomas of patients with acromegaly. The human TRH-R consists of 398 amino acids and is a member of the G protein-coupled receptor family. The gene has a single intron in the coding sequence. An addition of TRH caused an obvious increase in intracellular calcium concentration in chinese hamster ovary cells expressing this receptor. Although highly homologous to the rat and mouse TRH-Rs, the human TRH-R has several differences in the carboxyl-terminal structure. The sequence and functioning analyses showed the same expression of TRH-R mRNA in the pituitary adenomas as that in the normal pituitaries, indicating that 1) some adenomas of patients with acromegaly express an intact and functional TRH-R, and that 2) the paradoxical response of GH secretion to TRH found in the patients might be a direct effect of TRH on TRH-R expressed in the adenoma.

Molecular Cloning of a Functional Human Thyrotropin-Releasing Hormone Receptor

A cDNA encoding the human thyrotropin-releasing hormone receptor (hTRH-R) was isolated from a human brain cDNA library. Screening of 1.2 million clones resulted in 2 candidates. The largest clone contained TRH-R homologous sequences starting in the third transmembrane domain and included a long 3′ untranslated sequence. The smaller clone contained a potential start of the open reading frame, but was interrupted by an intron in the sixth transmembrane domain. The two clones had 497 bp of overlapping identical sequences and it was possible to assemble a complete cDNA thus restoring the assumed coding sequence. Electrophysiological studies of frog oocytes injected with in vitro transcribed mRNA showed TRH-specific inward currents, demonstrating that the reconstituted cDNA encoded a functional receptor. The predicted amino acid sequence of the hTRH-R protein showed high homology with the rat and mouse TRH-Rs with the exception of their C-terminal region. The human TRH-R gene seems to contain two introns.
