Investigation of Apoptosis-mediated Cytotoxic Effects of Royal Jelly on HL-60 Cells

In recent years, the use of nontoxic natural products that can be effective on cancer cells as new agents has attracted the attention of scientists in order to reduce the negative side effects of existing cancer drugs and their toxicity to normal cells. Some in vivo and in vitro studies have shown that royal jelly (RJ) inhibits cell proliferation and induces apoptosis. In this research, we aimed to investigate the effects of RJ on proliferative and apoptotic processes in the human acute promyelocytic leukemia cell line (HL-60). The HL60 cell line was treated with different concentrations of RJ for 24, 48, and 72 hours. The half maximum inhibitory concentration (IC50) of RJ was determined using 3-(4.5 dimethylthiazol-2-yl)-2.5-diphenyltetrazolium bromide test (MTT) and the proliferation activity of HL-60 cells was evaluated. Flow cytometry analysis was performed to measure apoptosis in HL-60 cells. IC50 values for RJ were calculated as 13.98, 6.45, and 2.06 mg/mL for 24, 48, and 72 hours, respectively. Flow cytometry results also showed that RJ had apoptotic effects at the concentrations found. The results showed that RJ treatment significantly induced apoptosis and reduced the proliferation of HL-60 cells. This study shows that RJ can be a complementary treatment against HL-60 acute myeloid leukemia cells due to its anticancer and antiproliferative effects.

Exosomal membrane proteins analysis using a silicon nanowire field effect transistor biosensor

Exosomes are of great significance in clinical diagnosis, due to their high homology with parental generation, which can reflect the pathophysiological status. However, the quantitative and classification detection of exosomes is still faced with the challenges of low sensitivity and complex operation. In this study, we develop an electrical and label-free method to directly detect exosomes with high sensitivity based on a Silicon nanowire field effect transistor biosensor (Si-NW Bio-FET). First, the impact of Debye length on Si-NW Bio-FET detection was investigated through simulation. The simulation results demonstrated that as the Debye length increased, the electrical response to Si-NW produced by charged particle at a certain distance from the surface of Si-NW was greater. A Si-NW Bio-FET modified with specific antibody CD81 on the nanowire was fabricated then used for detection of cell line-derived exosomes, which achieved a low limit of detection (LOD) of 1078 particles/mL in 0.01 × PBS. Furthermore, the Si-NW Bio-FETs modified with specific antibody CD9, CD81 and CD63 respectively, were employed to distinguish exosomes derived from human promyelocytic leukemia (HL-60) cell line in three different states (control group, lipopolysaccharide (LPS) inflammation group, and LPS + Romidepsin (FK228) drug treatment group), which was consistent with nano-flow cytometry. This study provides a highly sensitive method of directly quantifying exosomes without labeling, indicating its potential as a tool for disease surveillance and medication instruction.

MiRNA-132-5p mediates a negative feedback regulation of IL-8 secretion through S100A8/A9 downregulation in neutrophil-like HL-60 cells.

Neutrophils are an important source of pro-inflammatory and immunomodulatory cytokines. This makes neutrophils efficient drivers of interactions with immune and non-immune cells to maintain homeostasis and modulate the inflammatory process by notably regulating the release of cytokines. Ca2+-dependent regulatory mechanism encompassing cytokine secretion by neutrophils are not still identified. In this context, we propose to define new insights on the role of Ca2+-binding proteins S100A8/A9 and on the regulatory role of miRNA-132-5p, which was identified as a regulator of S100A8/A9 expression, on IL-8 secretion. Differentiated HL-60 cells, a human promyelocytic leukemia cell line that can be induced to differentiate into neutrophil-like cells, were used as a model of human neutrophils and treated with N- formyl-methionyl-leucyl-phenylalanine (fMLF), a bacterial peptide that activates neutrophils. shRNA knockdown was used to define the role of selected targets (S100A8/A9 and miRNA-132-5p) on IL-8 secretion. Different types of cytokines engage different signaling pathways in the secretion process. IL-8 release is tightly regulated by Ca2+ binding proteins S100A8/A9. miRNA-132-5p is up-regulated over time upon fMLF stimulation and decreases S100A8/A9 expression and IL-8 secretion. These findings reveal a novel regulatory loop involving S100A8/A9 and miRNA-132-5p that modulates IL-8 secretion by neutrophils in inflammatory conditions. This loop could be a potential target for therapeutic intervention in inflammatory diseases.

Three-Component Reaction of 3-Formyl-6-Methylchromone, Primary Amines, and Secondary Phosphine Oxides: A Synthetic and Mechanistic Study.

A fast, mild, and efficient catalyst-free approach has been developed for the synthesis of chromonyl-substituted α-aminophosphine oxides by the three-component reaction of 3-formyl-6-methylchromone, primary amines, and secondary phosphine oxides at ambient temperature. Carrying out the reaction with aliphatic amines or aminoalcohols at a higher temperature (80 °C), phosphinoyl-functionalized 3-aminomethylene chromanones were formed instead of the corresponding chromonyl-substituted α-aminophosphine oxides. No reaction occurred when 3-formyl-6-methylchromone and secondary phosphine oxides were reacted with aromatic amines in the absence of any catalyst. Applying a basic catalyst, the formation of the phosphinoyl-functionalized 3-aminomethylene chromanones was observed; however, the reaction was not complete. Detailed experimental and quantum chemical studies were performed to study the transformation. Moreover, the in vitro cytotoxicity of phosphinoyl-functionalized 3-aminomethylene chromanones was also investigated in three different cell lines, such as human lung adenocarcinoma (A549), mouse fibroblast (NIH/3T3), and human promyelocytic leukemia (HL60) cells. Several derivatives showed modest activity against the human promyelocytic leukemia (HL60) cell line.

Serum amyloid A1 recruits neutrophils to the invasive front of T1 colorectal cancers.

The tumor microenvironment plays an essential role in the development and progression of colorectal cancer (CRC). We recently reported that crosstalk between CRC cells and tumor-associated macrophages (TAMs) via serum amyloid A1 (SAA1) promotes invasion by T1 CRCs. In the present study, we aimed to clarify the role of neutrophils in early CRCs. Immunohistochemical analysis of CD66b, chemokine CXC motif ligand 8 (CXCL8 or interleukin-8, IL-8) and matrix metalloproteinase-9 (MMP-9) was performed using primary T1 CRCs (n=49). The HL-60 human promyelocytic leukemia cell line and THP-1 human monocytic leukemia cell line were used to obtain neutrophil-like and macrophage-like cells, respectively. Boyden chamber assays were used to analyze cell migration and invasion, and quantitative RT-PCR was used to analyze gene expression. Immunohistochemical analysis revealed accumulation of neutrophils at the SAA1-positive invasive front of T1 CRCs. Experiments using HL-60 cells suggested that treatment with SAA1 induced neutrophil migration and expression of CXCL8 and MMP-9 in neutrophils and that neutrophils promote CRC cell migration and invasion. Immunohistochemistry confirmed accumulation of CXCL8- or MMP-9-positive neutrophils at the SAA1-positive invasive front of T1 CRCs. Moreover, co-culture experiments using CRC, THP-1 and HL-60 cells suggested that CRC cells activated by macrophages upregulate CXCL8 and MMP-9 in neutrophils. Our results suggest that interplay between macrophages and CRC cells leads to recruitment of neutrophils to the invasive front of T1 CRCs and that SAA1 secreted by CRC cells activate neutrophils to promote invasion.

Ultraviolet Radiation Promoted Hypoxia-Induced Apoptosis in HL-60 Human Promyelocytic Leukemia Cell Line.

Minimal residual disease (MRD) is an important reason for the failure of autologous hematopoietic stem cell transplantation (auto-HSCT). Reducing MRD in grafts is particularly important to improve the efficacy of auto-HSCT. Previously, we reported that ultraviolet light-emitting diode (UV LED) suppressed the expression of Bcl-2 to induce apoptosis in HL-60 cells. Leukemia can lead to severe hypoxia of the bone marrow. Therefore, this study aimed to investigate the effect of UV LED on leukemia cells under hypoxia. HL-60 cells were irradiated with a UV LED (30 J/m2) and simulated under hypoxia with cobalt chloride. We found that UV LED irradiation or CoCl2 inhibited proliferation, induced apoptosis, decreased the Bcl-2/Bax ratio, and increased the levels of caspase 3, cleaved-caspase 3, and caspase 9 in HL-60 cells. In particular, the combined application of UV and CoCl2 significantly enhanced the apoptosis of HL-60 cells. In conclusion, UV LED in hypoxia exacerbated the inhibition of proliferation and induction of apoptosis and necrosis in HL-60 cells via the regulation of caspase 3/9 and the Bcl-2/Bax ratio-dependent pathway. The application of UV LEDs in hypoxia conditions may be a promising approach to kill residual drug-resistant leukemia cells in autologous grafts.

Antitumor activity against human promyelocytic leukemia and in silico studies of some benzoxazines

Cancer is one of the deadliest diseases in the world today, and the incidence of cancer is increasing. Leukemia is a type of blood cancer defined as the uncontrolled proliferation of abnormal leukocytes in the blood and bone marrow. The HL-60 (human promyelocytic leukemia) cell line, derived from a single patient with acute promyelocytic leukemia, provides a unique in vitro model system for studying the cellular and molecular events involved in the proliferation and differentiation of leukemic cells. In this study, antitumor activities on the HL-60 of some of the resynthesized benzoxazine derivatives (BXN-01 and BXN-02) were investigated. The results of in vitro studies obtained were compared a standard drug, etoposide. In vitro results showed that BXN-01 and BXN-02 were found to be extremely effective compared to etoposide (IC50 value: 10 µM) with IC50 values of 5 nM and 25 nM, respectively. Furthermore, molecular docking studies were carried out for preliminary prediction of possible interaction modes between compounds and the active site of the target macromolecules, hTopo IIα, HDAC2, and RXRA. Then, in silico ADME/Tox studies were performed to predict drug-likeness and pharmacokinetic properties of BXN-01 and BXN-02. Communicated by Ramaswamy H. Sarma

Abstract 5317: A novel approach to the treatment of ATRA/ATO resistant acute promyelocytic leukemia

Abstract Acute Promyelocytic Leukemia (APL) is characterized by proliferation of immature promyelocytes. Combination therapy using retinoic acid (RA) and arsenic trioxide (ATO) is considered standard treatment in the clinical environment. Resistance to both RA and ATO is associated with an increased risk of mortality. In addition, relapse occurs in approximately 10-20% of patients undergoing treatment for APL. Growth Hormone Releasing Hormone (GHRH) is a hypothalamic peptide that stimulates the release of Growth Hormone from the anterior pituitary. Our previous studies have demonstrated that human AML cell lines (K-562, THP-1, and KG-1a) display the GHRH receptor (GHRH-R). Incubation with GHRH-R antagonist MIA-602, inhibited proliferation and induced apoptosis. The purpose of this study is to examine the effects of GHRH-R antagonist MIA-602, on the NB4 Human Promyelocytic Leukemia cell line and its ATRA and ATO resistant sub-clone. NB4 cells expressed both GHRH-R and SV1 variants via western blot analysis. Wild type NB4 cells, as well as NB4 cells resistant to both ATRA and ATO (NB4-RAA), were cultured in suspension in RPMI and 10% fetal calf serum. NB4-RAA were established in Dr. Jimenez's lab from the wild type NB4 parent cell line. NB4 and NB4-RAA were plated in triplicate on multi-wells at 50,000 cells/mL. Cell lysates were prepared after incubation with 0.05-5 µmol/L of MIA-602. Cell viability was measured at 24 and 48h. Flow cytometry was also performed on the naïve NB4 parent cell line in addition to NB4-RAA to assess expression of CD-56 the cell surface marker. Upregulation of CD-56, also known as Neural cell adhesion molecule 1 (NCAM-1), has been shown to confer drug resistance in AML and is an important prognostic marker in APL. The viability of both cell lines decreased to similar levels at 24 h and 48 h when exposed to concentrations of MIA-602 higher than 0.05 μmol/L (p < 0.05). A significant decrease in cell viability was seen above 0.5 μmol/L after 48h. No viable cells were found in either cell line after 48h when exposed to 5 μmol/L of MIA-602. No difference in cell viability was found between naïve NB4 and resistant NB4-RAA when exposed to the same concentrations of MIA-602. MIA-602 induced significant changes in the expression level of many death-related genes including CASP9 (>1.5-fold; P<0.05). Results of flow cytometry revealed significantly increased expression of CD-56 (>5.8-fold; p,0.05) in NB4-RAA cells as compared to the parent cell line. These results indicate that resistance to ATRA and ATO in APL cells does not confer subsequent resistance to MIA-602. As a result, MIA-602 targets a pathway that is distinct from that of ATRA & ATO. More importantly, upregulation of pro-apoptotic genes by MIA-602 indicates an alternative pathway elicited by the GHRH-R antagonist in the setting of resistant APL cells. Synthesis of 4,4-Disubstituted 3,4-Dihydropyrimidin-2(1H)-ones and -thiones, the Corresponding Products of Biginelli Reaction Using Ketone, and Their Antiproliferative Effect on HL-60 Cells.

An efficient synthetic method for novel 4,4-disubstituted 3,4-dihydropyrimidin-2(1H)-ones 5 and -thiones 6 was developed. The cyclocondensation reaction of O-methylisourea hemisulfate salt 11 with 8 gives a tautomeric mixture of dihydropyrimidines 12 and 13 following acidic hydrolysis of the cyclized products to produce 5 in high yields. Thionation reaction of 5 at the 2-position smoothly proceeds to give 2-thioxo derivatives 6. These compounds 5 and 6, corresponding to the products of a Biginelli-type reaction using urea or thiourea, a ketone and a 1,3-dicarbonyl compound, have long been inaccessible and hitherto unavailable for medicinal chemistry. These methods are invaluable for the synthesis of 5 and 6, which have been inaccessible by conventional methods. Therefore, the synthetic methods established in this study will expand the molecular diversity of their related derivatives. These compounds were also assessed for their antiproliferative effect on a human promyelocytic leukemia cell line, HL-60. Treatment of 10 µM 6b and 6d showed high inhibitory activity similarly to 1 µM all-trans retinoic acid (ATRA), indicating that the 2-thioxo group and length of two alkyl substituents at the 4-position are strongly related to activity.

Flavonoides isolados de Sida santaremnensis H. Monteiro (“Guanxuma”) e avaliação das atividades biológicas

Introduction: Sida santaremnensis H. Monteiro (Malvaceae) is a plant popularly known as "vassourinha" or "guanxuma" that has been described as vasorelaxant, antiulcerogenic, antinociceptive and antiedematogenic. Objective: To contribute with the phytochemical and pharmacological profile of Sida santaremnensis through the isolation, purification and determination of chemical constituents of this plant, as well as through the evaluation of the anti-inflammatory and antitumor activity and leishmanicidal effect of the isolated constituent in a greater amount. Methods: The isolation of substances from the plant herein studied was performed with column chromatographic and analytical thin-layer methods and structural determination made by spectroscopic methods, such as Nuclear Magnetic Resonance, Hydrogen and Carbon 13, and comparisons with data from the literature. To assess pharmacological activities, cell viability tests, determination of nitric oxide levels, leishmanicidal activity, among others, were performed. Results: Two flavonoids from S. santaremnensis were obtained, kaempferol (S-1) and kaepferol 3-O-β-D-glycosyl-6’’-α-L-rhamnoside (S-2), the latter in a greater amount. The evaluation of the antitumor activity of the glycosylated flavonoid demonstrated that it does not present hemolytic activity against the human promyelocytic leukemia cell line (HL-60), and that it presented weak leishmanicidal activity. The immunopharmacological evaluation of kaempferol 3-O-β-D-glucosyl-6’’-α-L-rhamnoside revealed that it presents a possible anti-inflammatory action related to the inhibition of the production of nitrite by LPS-stimulated macrophages. Conclusion: These data demonstrate that kaempferol 3-O-β-D-glucosyl-6’’-α-L-rhamnoside has low toxicity, in addition to antileishmania, anti-inflammatory and immunomodulatory properties, which makes it a therapeutic potential for infectious and inflammatory diseases mediated by macrophages.

Phytochemical Profile, Antioxidant and Anti Proliferative Studies in Different Extracts of Artocarpus kemando Miq. Bark

In this study, the stem bark of Artocarpus kemando was used to find alternative antioxidants from natural sources with fewer side effects. A. kemando wasextracted successively using hexane, chloroform and methanol solvents, and evaluated for antioxidant, cytotoxic, and antiproliferative activities. The extracts were investigated for determination of their total phenolic content (TPC) and total flavonoid content (TFC). Then, the antioxidant activities were evaluated using chemical based assays such as ferric reducing antioxidant power (FRAP), total antioxidant capacity, radical scavenging of 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) and 2,2’-azino-bis (3-ethylbenzothia zoline-6-sulphonic) (ABTS), β-carotene-linoleic acid (BC) assay, oxygen radical absorbance capacity (ORAC), and cell based assay. The cytotoxic study was done using four different cell lines namely human estrogen receptor positive (ER+) breast cancer cell line (MCF7), human ovarian cancer cell line (CAOV-3), human promyelocytic leukemia cell line (HL60), and normal immortalised human ovarian surface epithelial cell line (TI074), and were evaluated using microculture tetrazolium salt (MTT) before morphological change study was done on CAOV-3 cell. In this study, methanol extract displayed the most promising antioxidant activity compared to other extracts when tested with DPPH, FRAP, ABTS, TAOC, BC,ORAC, and cytoprotective assays. The remarkable activity showed by the methanol extract might be due to its high content of phenolic and flavonoid compounds at 855.5 ± 0.01 GAE μg/mL and 145.45 ± 0.06 QAE μg/mL, respectively. Nevertheless, the chloroform extract displayed better scavenging activity compared to other extracts with IC50 value of 618 ± 0.04 μg/mL in DPPH assay. Each extract was analysed using Gas Chromatography Mass Spectrophotometry and the chemical constituents obtained were then analysed. In the cytoprotective activity, the methanol extract showed a comparable cytoprotection with ascorbic acid against the free radicals at the lowest effective concentration (EC50) value of 21.48 μg/mL. However, in the cytotoxicity study, only chloroform extract displayed significant toxicity against the cancer cells with IC50 value of 27.9 ± 0.03, 24.1 ± 0.02 and 9.0 ± 0.04 μg/mL after treatment at 24, 48, and 72 h, respectively. The chloroform extract of A. kemando was found capable of inducing apoptosis as shown with cell membrane blebbing, chromatin condensation and formation of apoptotic bodies. The results obtained from the study showed that A. kemando bark could be a potential antioxidant and antitumor agents particularly on human ovarian cancer cells.

Amplification of Cord Blood-Derived Cytotoxic T Lymphocytes using HL-60 cell Derived Exosomes

IntroductionDendritic cell (DCs) based cytotoxic T lymphocytes (CTLs) are commonly used in immunotherapy due to their specificity. The selection of appropriate cell origin and tumor antigen is the key point. The objective was to culture DCs and CTLs simultaneously from cord blood, and deliver antigen information using tumor derived exosomes.Material and methodsExosomes were collected from the human promyelocytic leukemia cell line HL-60 using ultracentrifugation. Prepared DCs from adherent cord blood mononuclear cells (MNCs) using SCF, GM-CSF, and IN-4. TNF-α and microRNA removed tumor-exosome were used to induce DCs maturation. DCs matured in the presence of HL-60 cell membrane protein extract or no antigen were set as control. CTLs was cultured from non-adherent MNCs by adding IFN-γ, IL-15, SCF, FLT-3L, anti-CD3, anti-CD28 and IL-2. The CTLs were analyzed by flow cytometry, cytotoxicity experiments and ELISA.ResultsDCs can be obtained from cord blood and express costimulatory molecules. After 15 days, the total number of the cells expanded 26.3 times, and more than 82% of the cells expressed CD3+CD8+ in the most amplified HL-60-Ex-DCs-CTL group. These CD3+CD8+ T cells generated by HL-60-Ex-DCs displayed specific cytotoxicity towards HL-60 and low lethality towards unrelated BALL-1 cells. ELISA results showed that the expressions of TNF-α and IFN-γ in HL-60-Ex-DCs or HL-60mPr-DCs activated CTLs were upregulated compared with the control group.ConclusionsCord blood CTLs generated by HL-60 derived exosome activated DCs displayed specific cytotoxicity towards HL-60 promyelocytic leukemia cells. Therefore cord blood and tumor derived exosomes provided a good source for adoptive immunotherapy.

Synergistic effects of LY294002 and ABT199 on the cell cycle in K562, HL60 and KG1a cells.

The aim of the present study was to investigate the synergistic effect of LY294002 (a PI3K inhibitor) and ABT199 (a BCL2 inhibitor) on the cell cycle in acute myeloid leukemia (AML). The optimal concentration and duration of combined LY294002 and ABT199 were determined in human erythroleukemia (K562), promyelocytic leukemia (HL60) and myeloid leukemia (KG1a) cell lines. The mRNA and protein expression levels of cell cycle‑related molecules, including S‑phase kinase‑associated protein 2 (Skp2), p27, BCL2, Bax, cleaved caspase 3 (caspase‑3) and caspase 9 (caspase‑9) were detected via reverse transcription‑quantitative PCR and western blot analysis, respectively. At the molecular level, LY294002 and ABT199 combination treatment significantly downregulated Skp2, Bcl2, procaspase‑3 and procaspase‑9 expression levels, but markedly upregulated p27, Bax, cleaved caspase‑3 and caspase‑9 expression levels in K562, HL‑60 and KG1a cells. The results of the present study demonstrated that LY294002 and ABT199 combination treatment may serve as a novel therapeutic strategy for AML.

Novel chiral naphthalimide-cycloalkanediamine conjugates: Design, synthesis and antitumor activity

A novel series of enantiopure naphthalimide-cycloalkanediamine conjugates were designed, synthetized and evaluated for in vitro cytotoxicity against human colon adenocarcinoma (LoVo), human lung adenocarcinoma (A549), human cervical carcinoma (Hela) and human promyelocytic leukemia cell lines (HL-60). The cytotoxicity of the compounds was highly dependent on size and relative stereochemistry of the cycloalkyl ring as well as length of the spacer. By contrast, any kind of enantioselection was observed for each pair of enantiomers. Flow cytometric analysis indicated that compounds 22 and 23 could effectively induce G2/M arrest in the four previous cell lines despite a mild apoptotic effect.

Biotransformation of artemisinic acid to bioactive derivatives by endophytic Penicillium oxalicum B4 from Artemisia annua L.

As a biosynthetic precursor of the antimalarial drug artemisinin, artemisinic acid (AA) is abundant in Artemisia annua L. with a content of 8-10-fold higher than artemisinin, but less effective. In this study, the biotransformation of AA was carried out with an endophytic fungus Penicillium oxalicum B4 to extend its utility. After 10-day-culture of the endophyte with AA at 2 mg/mL, eight biotransformation metabolites were isolated from the culture broth, including five undescribed metabolites, namely 3α,14-dihydroxyartemisinic acid, 14-hydroxy-3-oxo-artemisinic acid, 15-hydroxy-3-oxo-artemisinic acid, 12, 15-artemisindioic acid and 1,2,3,6-tetradehydro-12, 15-artemisindioic acid. The fungal enzymes possess the selective capacity to hydroxylate, carbonylate and ketonize the allyl group of AA. The major biotransformation metabolite was the hydroxylated product 3-α-hydroxyartemisinic acid (33.3%) in the cultures of early stage (day 1–6), whereas most of the other biotransformation products were synthesized in the later stage (day 8–10). Compared with AA, some metabolites (3α,14-dihydroxyartemisinic acid, 15-hydroxy-3-oxo-artemisinic acid and 1,2,3,6-tetradehydro-12, 15-artemisindioic acid) possessed stronger cytotoxic activity to the human colon carcinoma cell line (LS174T) and promyelocytic leukemia cell line (HL-60). The metabolites 12, 15-artemisindioic acid and 3-α-hydroxyartemisinic acid exhibited significant inhibitory activity to the lipopolysaccharide-induced nitrite production of RAW 264.7 cells at 10.00 μM and 2.50 μM, respectively. The results demonstrated the potential of fungal endophytes on biotransforming AA to its bioactive derivatives.

Development of Efficient Synthetic Method for Tautomeric Dihydropyrimidines and Analysis of Their Functionality

I here present the results of our studies on the synthesis and functional analysis of tautomeric dihydropyrimidines (DPs) and related compounds in two sections. In the first section, we describe our experimental and theoretical studies on the thermodynamics and properties of 2-substituted 1,4- and 1,6-dihydropyrimidine-5-carboxylates by 1H NMR measurements and density functional theory (DFT) calculations, respectively. The concentration ratios of tautomers a/b of DPs 1, 2, and 3 were determined under various conditions to determine the effects of temperature, solvent, and concentration on thermodynamics data. The obtained free energy differences (ΔG), enthalpy differences (ΔH), and entropy differences (ΔS) are discussed in terms of the molecular structures, dipole moments (DM), and electrostatic potential maps obtained by DFT calculations to clarify the nature of DPs 4-8. In the second section, an efficient synthetic method developed for 6-unsubstituted 3,4-dihydropyrimidin-2(1H)-thiones 9 and 2-ones 10 is described. The novelties of the synthesis protocol are as follows: 1) the use of Lewis acid-mediated reaction, 2) good to high yields, and 3) its broad scope of applicability to aldehydes and ureas. Hitherto unavailable 6-unsubstituted 2-amino DP 11 and 2-aryl DP 12 were obtained from 9 by a substitution reaction with the amine and the Liebeskind-Srogl reaction, respectively. The compounds 9, 10, and related 6-methyl derivatives 19-21 were assessed for their antiproliferative effect on the human promyelocytic leukemia cell line HL-60. 4,4-Dipropyl derivative 20 showed relatively strong activity with an IC50 value of 341 nM.

Synthesis of novel 6-unsubstituted 2-aminodihydropyrimidines by Sc(OTf)3-mediated amination and their antiproliferative effect on HL-60 cells

An efficient method of synthesis of novel 6-unsubstituted 2-aminodihydropyrimidines 9 has been developed. The substitution reaction of 2-methylthiodihydropyrimidines 7 and primary amines in the presence of a catalytic amount of Sc(OTf)3 with heating proceeds smoothly to give 9 in good to high yields. This amination protocol can be applied to various aryl- and alkylamines. Other 2-methylthiodihydropyrimidines 10 having different substituent patterns at 4- and 6-positions also reacted with aniline under the same conditions to give corresponding 2-anilinodihydropyrimidines 11 in high yields. These compounds 9 were assessed for their antiproliferative effect on the human promyelocytic leukemia cell line HL-60. The 2-benzyl derivative 9i showed a relatively high activity with an IC50 of <100 nM. The high activity of 9i was comparable to that of all-trans retinoic acid (223.1 nM). The results of the trypan blue exclusion test of 9i indicated that necrosis-like cell death occurred.

Prognosis of vascular endothelial growth factor gene polymorphism in Egyptian patients with acute myeloid leukemia

BackgroundAngiogenesis is an important process in hematological malignancies especially leukemia. Its role in acute leukemia has been discussed since the cloning of the vascular endothelial growth factor (VEGF) gene from acute myelogenous leukemia (AML). HL-60 is a human promyelocytic leukemia cell line. VEGF, a polymorphic gene, plays a key role in angiogenesis and tumor growth. Its different genotypes are associated with many human diseases. Increased angiogenesis, mediated by VEGF, was associated with poor prognosis in AML patients. AML that arises from neoplastic transformation of hematopoietic stem and progenitor cells remains one of the greater challenges in treating this AML. In adults, leukemia is included in the top 15 of the most common forms of cancer. Single-nucleotide polymorphism of the VEGF gene, 699947 is found in peripheral blood mononuclear cells of Iranian, Australian, Caucasian, and African patients with AML. The main target of this study is to investigate the relationship between VEGF gene polymorphism (rs699947) with AML.Patients and methodsThis a controlled study which included 100 and 76 individuals in each group, respectively. Group 1: a healthy control group and group 2: AML diagnosed patients. Blood samples were genotyped using the tetra-primer amplification-refractory mutation system-PCR and the results were confirmed by PCR-restriction fragment length polymorphism method.ResultsOur results suggest that there is a correlation between rs699947 single-nucleotide polymorphism and AML development and prognosis. However, patients with −2578 AA/CC genotypes were associated with significant better complete remission in comparison to patients with −2578 CA genotype (P = 0.0001). We found that certain polymorphisms are associated significantly with remissions while others with worse response to induction chemotherapy.

Curcumin Induces Human Glioma Cell Apoptosis by Promoting Reactive Oxygen Species Production

Malignant glioma by surgery is difficult to completely remove, the recurrence rate is high, the survival period is short and the prognosis is poor, which poses a great threat to human health. This article mainly studies the related process that curcumin induces human glioma cell apoptosis by promoting reactive oxygen species production. In the experiment, glioma cells were processed and Uppsala 87 malignant glioma cells of human were cultured in vitro. 3-(4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide assay was used to detect the effect of curcumin on the proliferation of Uppsala 87 malignant glioma cells. The cells in each reagent group and control group treated with curcumin were treated with 3-(4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide colorimetric treatment at 24 h. Use flow cytometry to detect cell apoptosis. Cellular immunofluorescence was used to detect the effect of oxidase inhibitors on nuclear factor activation. Experimental data showed that the apoptotic rate of 0, 2.5, 5 mg/l, diallyl disulphide treatment groups were 12.3 %, 19.75 % and 26.44 %, respectively, indicating that diallyl disulphide caused an increase in the apoptotic rate of human promyelocytic leukemia cell line cells and the difference was statistically significant (p<0.05). The results show that curcumin can inhibit the proliferation of Uppsala 87 malignant glioma cells cultured in vitro, induce the differential expression of apoptosis-related genes and thus have a significant effect of promoting cell apoptosiss.

Daphnane-type diterpenoids from Euphorbia fischeriana Steud and their cytotoxic activities

Two new daphnane-type diterpenoids fischerianin A (1) and fischerianin B (2), as well as two known ones langduin A (3) and langduin A6 (4), were isolated from the extracts of Euphorbia fischeriana Steud dry roots. Their structures including the absolute stereochemistry were determined by various spectroscopic methods and comparing their experimental and calculated CD spectra. 1 and 2 harbor a ketal group and a 9,13-oxide bridge in their C ring which is rare in daphnane-type diterpenoids. In cytotoxic assays, moderately inhibitory activities of 1–4 against human cancer cell lines (human malignant melanoma cell line, A375; human liver carcinoma cell line, HepG2; human promyelocytic leukemia cell line, HL-60; human Caucasian chronic myelogenous leukemia cell line, K562; human cervix epithelioid carcinoma cell line, HeLa) were observed, with IC50 values ranging from 5.31 to 21.46 μM.
