Chitosan Xylotrupes gideon encapsulated lemongrass leaf ethanol extract reduce H2O2‐induced oxidative stress in human dermal fibroblast

During phagocytosis, phagocyte cells discharge reactive oxygen species referred to as respiratory bursts, inducing a rise in pro‐oxidants and subjecting the cell to oxidative stress. Such stress is a biological mechanism related to an imbalance in pro‐oxidant/antioxidant homeostasis, which generates toxic reactive oxygen. Encapsulation is a coating process to improve the stability of bioactive compounds from lemongrass extract. Therefore, this study aims to determine the encapsulation activity of lemongrass leaf extract with chitosan X. gideon (LEChXg) to reduce the oxidative stress of fibroblasts. The research used the human dermal fibroblast (HDF) cell line, comprising negative and positive controls and use of LEChXg 100, 200, 300, 400, and 500 µg/mL. HDF cell migration was evaluated by employing the scratch wound healing method and the wound closure was oberseved at 0, 2, 4, 6, and 24 h intervals. The cell proliferation was observed at 24, 48, and 72 h using CCK‐8 at a 450 nm wavelength. The results showed that the observations at 0, 2, and 4 h did not demonstrate any significant difference on the cell migration (p > 0.05) among the groups. However, the wound closure at 4 and 6 h showed a significant difference (p < 0.05) with LEChXg 300 µg/mL. Despite the lack of any significant variation observed up to 24 h, fibroblast subjected to the stressor did not achieve complete closure. The groups treated with LEChXg were more stable in maintaining fibroblast proliferation up to the end of the observation than those with stressors at 24, 48, and 72 h. Fibroblast induced with a stressor was also more stable in maintaining migration and proliferation in groups receiving LEChXg 300 µg/mL.

Combined Effects of Defatted Hydrolyzed Collagen from Salmon Skin and Vitamin C on Proliferation and Migration of Human Fibroblast Cell

Skin injury causes fibroblast dysfunction and lowers collagen production. Safe, functional ingredients such as vitamin C (Vit C) and fish hydrolyzed collagen (HC) have been used to alleviate this problem. Defatted HC from salmon (Oncorhynchus nerka) skin could be a potential functional ingredient with skin nourishment activity. This study aimed to investigate the combined effects of HC and Vit C on the proliferation and migration of human dermal fibroblast (HDF). Molecular weight ranging from 102 Da to 10,175 Da and high imino acid content were found in HC. HC (0–800 µg/mL) or vitamin C (Vit C) (0.01–100 µg/mL) was applied for HDF treatment. Higher cell proliferation was found by adding HC at 50 µg/mL or Vit C at 0.01 µg/mL compared to the control and those treated with both compounds at other levels (p < 0.05). Cells treated with HC (50 µg/mL) combined with Vit C (0.01 µg/mL) (HC+Vit C) showed higher proliferation, migration, and lamellipodia formation of HDF cells than those treated with HC or Vit C alone. Moreover, all the samples tested could stimulate the proliferation and migration of HDF cells via FAK/Akt and ERK/p38 MAPK signaling pathways. Thus, HC combined with Vit C could be a promising functional ingredient for skin nourishment and would healing.

The effect of a geometric-shaped tool with blue led light on the activation of human dermal fibroblasts and cancer cells

Light-emitting diode (LED) irradiation has been used as a therapeutic technique for several diseases. It is known that energy levels of light change according to different surfaces, colors and geometric shapes. However, more information is needed regarding the effect of this change on the human body. This study was aimed to investigate the effect of blue LED light applied through an optical path with special geometry on the human dermal fibroblast (HDF) and cancer cell proliferation, apoptosis, migration, and cell cycle. The number of cancerous cells was found to be lower and the rate of necrosis was determined to be significantly higher in the geometrical light-treated group than in the direct light and non-light groups. The migration of HDF cells in blue LED light in the geometrical tool group increased more than the other groups after 72 h of light irradiation. In cancer cells, the wound closure rate of the non-light group was higher than light-exposed groups after 72 h. Blue LED light induces a G1 phase peak followed by apoptosis in cancer cells 24 h after light irradiation, especially in the geometrical tool group. These results indicate that blue LED irradiation applied through an optical path with a special geometrical tool inhibits cancer cell proliferation, migration, and induces cell necrosis.

An on-chip wound healing assay fabricated by xurography for evaluation of dermal fibroblast cell migration and wound closure

Dermal fibroblast cell migration is a key process in a physiological wound healing. Therefore, the analysis of cell migration is crucial for wound healing research. In this study, lab-on-a-chip technology was used to investigate the effects of basic fibroblast growth factor (bFGF), mitomycin C (MMC), MEK1/2 inhibitor (U0126) and fetal calf serum (FCS) on human dermal fibroblast cell migration. The microdevice was fabricated consisting of microchannels, pneumatic lines and pneumatically-activated actuators by xurographic rapid prototyping. In contrast to current approaches in in vitro wound healing such as scratch assays and silicone inserts in wellplate format, which show high variability and poor reproducibility, the current system aims to automate the wounding procedure at high precision and reproducibility using lab-on-a-chip. Traumatic wounding was simulated on-chip on fibroblast cell monolayers by applying air pressure on the flexible circular membrane actuator. Wound closure was monitored using light microscopy and cell migration was evaluated using image analysis. The pneumatically controlled system generates highly reproducible wound sizes compared to the conventional wound healing assay. As proof-of-principle study wound healing was investigated in the presence of several stimulatory and inhibitory substances and culture including bFGF, MMC, U0126 MEK1/2 inhibitor as well as serum starvation to demonstrate the broad applicability of the proposed miniaturized culture microsystem.

Phytochemical properties of a rare mangrove Aegialitis rotundifolia Roxb. leaf extract and its influence on human dermal fibroblast cell migration using wound scratch model

Background: Aegialitis rotundifolia Roxb. is a small mangrove plant traditionally used for curing pain arising from sundry injuries. At present, there are no scientific evidences of its wound healing properties and few reports have claimed that reduction of pain could accelerate the wound healing process. Aims and Objectives: The present work deals with the qualitative and quantitative study of the phytoconstituents present in A. rotundifolia Roxb., ethanolic leaves extract and also evaluate its wound healing properties in vitro. Materials and Methods: The qualitative phytochemical analysis of the extract was performed using preliminary phytochemical tests whereas the quantitative determination of phytochemicals was evaluated spectroscopically using ultraviolet spectroscopy. The in vitro wound healing activity was determined on human dermal fibroblast (HDF) cells using the wound scratch assay. Results: The quantitative estimation of phytochemicals revealed that total phenolics was the most abundantly found phytochemical followed by a total flavonoid, total flavonol, total tannin, total alkaloid, total proanthocyanidin, and total saponin. In the in vitro wound healing assay, the sample was first examined for possible cytotoxicity on HDF cells where the results showed that it was slightly toxic at higher concentration (inhibit cell growth by 50%: 157.41 ± 3.4 μg/mL); thus, a concentration of 40 and 80 μg/mL was taken to carry out the scratch assay. The wound scratch assay revealed significant (P < 0.01) wound healing activity in a dosedependent manner at 40 and 80 μg/mL concentrations. Conclusion: Thus, the extract proved effective in wound healing which may have been attributed by the presence of several bioactive phytochemicals.

Functional and cell proliferative properties of an exopolysaccharide produced by Nitratireductor sp. PRIM-31

Exopolysaccharides (EPS) produced by bacteria are important source of natural biomolecules with therapeutic applications. In this study EPS produced by a strain PRIM-31 isolated from seawater from south west coast of India identified by 16S rRNA gene sequencing as Nitratireductor sp. was investigated for its biotechnological applications. The isolate produced 650mgL−1 EPS under optimum growth conditions. The purified EPS contained 58.3% carbohydrates with 7.08% uronic acid containing sugars, functional groups such as sulfate (2.68%) and trace amounts of proteins (0.65%). Molecular weight of the EPS was 90kDa and monosaccharide composition was arabinose, glucose, xylose, glucuronic acid and galactose (6.6: 3.5: 2.1: 1.3: 1). The EPS displayed antioxidant activity in terms of total antioxidant capacity, ferric reducing power, scavenging of DPPH (IC50 390μgmL−1) and superoxide radicals (IC50 340μgmL−1). Cell proliferative activity of the EPS was evidenced by significant improvement in human dermal fibroblast (HDF) proliferation compared to control. Further, significant improvement (41%) in HDF cell migration was observed in in vitro scratch assay with EPS treatment. The EPS also showed antiproliferative activity against glioblastoma cells with an IC50 of 234.04μgmL−1. These functional properties of the EPS prospect its biotechnological applications.
