Effects of Pesticides on the Survival of Shredder Nectopsyche sp. (Trichoptera) and Leaf Decomposition Rates in Tropical Andes: A Microcosm Approach.

Andean streams are becoming increasingly impacted by agricultural activities. However, the potential effects of pesticides on their aquatic biodiversity remain unassessed. In order to address this knowledge gap, we conducted an experiment over 37 days in microcosms to assess the effect of two pesticides commonly used in Ecuador (Engeo and Chlorpyrifos) on the aquatic insect Nectopsyche sp. (Trichoptera: Leptoceridae) at 0, 0.10, 5 and 10 μg L-1 concentrations. The highest concentration corresponds to the maximum concentration allowed by the Equatorian legislation. We assessed insect mortality every 24 h, with leaf litter decomposition rates of organic matter determined by deploying Andean alder (Alnus acuminata) dry leaf packs in the microcosms. We found significant mortality of Nectopsyche sp. at high concentrations of Chlorpyrifos, whereas leaf litter was not significantly affected by any of the treatments. We conclude that the environmental legislation of Ecuador might not be fully protecting aquatic biodiversity from pesticide pollution. Further studies are needed, especially when considering that the maximum permitted concentration is very likely exceeded in many areas of the country. We also suggest that the maximum permissible values should be reviewed, considering each pesticide individually.

Estimation of chlorpyrifos distribution in forensic visceral samples and body fluids using LCMS method

The ingestion of the insecticide chlorpyrifos leads to fatal intoxication in suicidal cases, and its distribution can be assessed only after post-mortem. This study attempted to investigate the distribution of chlorpyrifos in forensic visceral tissue samples like stomach, liver, kidney, heart, brain, lung, spleen, muscle and body fluids like blood and urine by liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) with the liquid-liquid extraction method. An analysis time of 8.77 min with 1 μl as an injection volume was chosen. The results obtained are analysed using Labsolutions insight LCMS software, considering Multiple Reaction Monitoring (MRM) and Retention Time (RT). Our findings depict that the stomach tissue has very high concentrations of chlorpyrifos, which suggests the possibility of high distribution and aspiration in the stomach tissue. The concentration of chlorpyrifos in stomach tissue ranged from 816 to 901 μg/g in male cadavers and from 443 to 612 μg/g in female cadavers. Among all the tissues, the stomach tissue showed the highest concentration, while the lowest concentration was found in muscle. Metabolic distribution of chlorpyrifos from the stomach contents to the other surrounding organs may occur and hence the concentrations of CPF were also seen in other organs like the liver, lung, kidney, brain, and muscle. These toxicological results from autopsy findings, together with LC-MS/MS indicate that stomach tissue examination gives an accurate profile of insecticide poisoning in forensic samples with acute poisoning.

Developmental Exposure to Trace Concentrations of Chlorpyrifos Results in Nonmonotonic Changes in Brain Shape and Behavior in Amphibians.

Despite regulations and improved design, pesticides remain ubiquitous in the environment at relatively low, trace concentrations. To understand how prolonged exposure to trace pesticide concentrations impacts vertebrate brain development and behavior, we raised larval amphibians (northern leopard frogs, Lithobates pipiens) in 0, 1, or 10 μg/L of the organophosphorus pesticide chlorpyrifos (CPF) from hatching to metamorphosis. Tadpoles exposed to 1 μg/L CPF, but not 10 μg/L CPF, had changes in relative brain mass, relative telencephalon shape, and behavioral responses to a novel visual cue. Tadpoles exposed to 10 μg/L CPF had altered behavioral responses to predator-associated olfactory cues. After metamorphosis, frogs raised in 1 μg/L CPF, but not 10 μg/L CPF, had changes in the shape of their optic tectum and medulla. Thus, we provide robust evidence that even trace, yet ecologically realistic, concentrations of CPF have neurodevelopmental and behavioral effects that carry over to later life-history stages, further emphasizing the potent effects of trace levels of CPF on vertebrate development. Also, some but not all effects were nonmonotonic, meaning that effects were evident at the lowest but not at the higher concentration of CPF.

Degradation insight of organophosphate pesticide chlorpyrifos through novel intermediate 2,6-dihydroxypyridine by Arthrobacter sp. HM01

Organophosphates (OPs) are hazardous pesticides, but an indispensable part of modern agriculture; collaterally contaminating agricultural soil and surrounding water. They have raised serious food safety and environmental toxicity that adversely affect the terrestrial and aquatic ecosystems and therefore, it become essential to develop a rapid bioremediation technique for restoring the pristine environment. A newly OPs degrading Arthrobacter sp. HM01 was isolated from pesticide-contaminated soil and identified by a ribotyping (16S rRNA) method. Genus Arthrobacter has not been previously reported in chlorpyrifos (CP) degradation, which shows 99% CP (100 mg L−1) degradation within 10 h in mMSM medium and also shows tolerance to a high concentration (1000 mg L−1) of CP. HM01 utilized a broad range of OPs pesticides and other aromatic pollutants including intermediates of CP degradation as sole carbon sources. The maximum CP degradation was obtained at pH 7 and 32 °C. During the degradation, a newly identified intermediate 2,6-dihydroxypyridine was detected through TLC/HPLC/LCMS analysis and a putative pathway was proposed for its degradation. The study also revealed that the organophosphate hydrolase (opdH) gene was responsible for CP degradation, and the opdH-enzyme was located intracellularly. The opdH enzyme was characterized from cell free extract for its optimum pH and temperature requirement, which was 7.0 and 50 °C, respectively. Thus, the results revealed the true potential of HM01 for OPs-bioremediation. Moreover, the strain HM01 showed the fastest rate of CP degradation, among the reported Arthrobacter sp.Graphical

Analysis of Pesticide Residues in Some Market Garden Crops in Santa, North West Cameroon

The increased use of pesticides in crop production, especially in Santa, has left consumers with great fear and uncertainty regarding the fate of residues after consumption in the human body. This study was aimed at performing a multi-residue analysis of crop samples to detect and quantify pesticide residues present, and thus determine their level of contamination. Twelve (12) crop samples (4 samples each of Solanum tuberosum (Irish potato), Solanum lycopersicum (tomato), Capsicum annum (sweet pepper) and Apium graveolens (celery)), were randomly collected from four villages in Santa. The samples were analyzed using the QuEChERS Buffered AOAC 2007.01 method. The results showed the presence of pesticide residues in 58.3% of the samples. Cypermethrin and chlorpyrifos insecticides were detected, though cypermethrin was found in traces. After quantification, the concentration of chlorpyrifos was determined to be far above the CODEX Maximum Residue Limits (MRLs) in 28.6% of the samples. Samples with this high concentration of chlorpyrifos were: tomato from Pinyin and Baligham (2.0 and 1.8 μg/g respectively), whereas the Codex MRLs for tomato is 1.0 μg/g. This study raised concerns on the contamination of some garden crops by pesticide residues and calls for the assessment of the health situation of consumers and the analysis of pesticide residues in drinking water resources in this area.

Bioaccumulation potential of chlorpyrifos in resistant Hyalella azteca: Implications for evolutionary toxicology

Given extensive use of pesticides in agriculture, there is concern for unintended consequences to non-target species. The non-target freshwater amphipod, Hyalella azteca has been found to show resistance to the organophosphate (OP) pesticide, chlorpyrifos, resulting from an amino acid substitution in acetylcholinesterase (AChE), suggesting a selective pressure of unintended pesticide exposure. Since resistant organisms can survive in contaminated habitats, there is potential for them to accumulate higher concentrations of insecticides, increasing the risk for trophic transfer. In the present study, we estimated the uptake and elimination of chlorpyrifos in non-resistant US Lab, and resistant Ulatis Creek (ULC Resistant), H. azteca populations by conducting 24-h uptake and 48-h elimination toxicokinetic experiments with 14C-chlorpyrifos. Our results indicated that non-resistant H. azteca had a larger uptake clearance coefficient (1467 mL g−1 h−1) than resistant animals (557 mL g−1 h−1). The half-life derived from the toxicokinetic models also estimated that steady state conditions were reached at 13.5 and 32.5 h for US Lab and ULC, respectively. Bioaccumulation was compared between non-resistant and resistant H. azteca by exposing animals to six different environmentally relevant concentrations for 28 h. Detection of chlorpyrifos in animal tissues indicated that resistant animals exposed to high concentrations of chlorpyrifos were capable of accumulating the insecticide up to 10-fold higher compared to non-resistant animals. Metabolite analysis from the 28-h concentration experiments showed that between 20 and 50 % parent compound was detected in H. azteca. These results imply that bioaccumulation potential can be more significant in chlorpyrifos resistant H. azteca and may be an essential factor in assessing the full impacts of toxicants on critical food webs, especially in the face of increasing pesticide and chemical runoff.

Molecular Diversity of Chlorpyrifos Degrading Bacteria Isolated from Apple Orchard Soils of Himachal Pradesh

A total of seventy-two bacterial isolates were obtained employing enrichment culture technique from apple orchard soils contaminated with chlorpyrifos. Pure cultures of bacterial isolates were obtained using streak plate method on mineral salt medium. Bacterial isolates were characterized on the basis of morphology, culture and biochemical properties. Six bacterial isolates exhibited high extracellular organophosphorus hydrolase activity along with high tolerance towards high concentrations of chlorpyrifos. Genomic DNA extraction from bacterial isolates was done with phenol/chloroform method. Molecular Diversity of six chlorpyrifos degrading bacterial isolates was done employing RAPD-PCR technique by using 25 decamer primers, where amplification was showed by only 20 primers. A total of 337 amplified bands and 64 unique bands ranging in size from 100 to 4900bps were produced after RAPD analysis. The similarity coefficient estimated by Jaccard’s coefficient for these bacterial isolates was found to range between 31 to 64 percent.

Investigating a human pesticide intoxication incident: The importance of robust analytical approaches

Abstract A human intoxication incident attributed to pesticide abuse was investigated using cutting-edge analytical methodologies. An LC-ESI-MS/MS method, based on a hybrid solid-phase extraction protocol (hybrid-SPE), was applied for the detection and quantification of several pesticides and metabolites in human biological fluids. Concomitantly, an UHPLC-HRMS method was applied to investigate potential metabolites, assisted by a complementary GC-MS method to elucidate the presence of plausible pesticides co-formulants. The LC-ESI-MS/MS method exhibited acceptable mean recoveries at the lower limit of quantification (LLOQ) and three additional levels, varying from 85 to 106% for all analytes and matrices. In serum, urine, and gastric fluid samples, the suspect compounds, namely chlorpyrifos and myclobutanil, predominated. Gastric fluid samples contained the highest concentrations of chlorpyrifos (39,800 ng/mL) and myclobutanil (18,800 ng/mL), while the neonicotinoid imidacloprid was also quantified, below 30 ng/mL. Notwithstanding, the UHPLC-HRMS analysis unveiled several metabolites of chlorpyrifos and myclobutanil. In parallel, GC-MS analysis, corroborated the presence of several co-formulants in gastric fluid samples, exemplified by m- and o-xylene, and cyclohexanone. Overall, three analytical methods were implemented to elucidate the chemical causality of a human intoxication incident. The presence of suspected active substances, one additional, and several metabolites and co-formulants were documented.

Human dietary exposure to chemicals in sub-Saharan Africa: safety assessment through a total diet study

Human dietary exposure to chemicals can result in a wide range of adverse health effects. Some substances might cause non-communicable diseases, including cancer and coronary heart diseases, and could be nephrotoxic. Food is the main human exposure route for many chemicals. We aimed to assess human dietary exposure to a wide range of food chemicals. We did a total diet study in Benin, Cameroon, Mali, and Nigeria. We assessed 4020 representative samples of foods, prepared as consumed, which covered more than 90% of the diet of 7291 households from eight study centres. By combining representative dietary surveys of countries with findings for concentrations of 872 chemicals in foods, we characterised human dietary exposure. Exposure to lead could result in increases in adult blood pressure up to 2·0 mm Hg, whereas children might lose 8·8-13·3 IQ points (95th percentile in Kano, Nigeria). Morbidity factors caused by coexposure to aflatoxin B1 and hepatitis B virus, and sterigmatocystin and fumonisins, suggest several thousands of additional liver cancer cases per year, and a substantial contribution to the burden of chronic malnutrition in childhood. Exposure to 13 polycyclic aromatic hydrocarbons from consumption of smoked fish and edible oils exceeded levels associated with possible carcinogenicity and genotoxicity health concerns in all study centres. Exposure to aluminium, ochratoxin A, and citrinin indicated a public health concern about nephropathies. From 470 pesticides tested across the four countries, only high concentrations of chlorpyrifos in smoked fish (unauthorised practice identified in Mali) could pose a human health risk. Risks characterised by this total diet study underscore specific priorities in terms of food safety management in sub-Saharan Africa. Preparation of Chlorpyrifos Degradation and Plant Growth Promoting Bacterial Formulation

Chlorpyrifos is an organophosphate pesticide used mainly in agriculture against the pests invading the crops. It becomes essential to degrade such pesticides that cause harm to the soil and the environmental biota. Pseudomonas putida is one of the nitrogen fixers found in the soil. In our study, we isolated and screened the chlorpyrifos degrading bacteria from pesticides contaminated agricultural soil. These bacterial isolates were identified and characterized by the VITEK® 2 Systems. The two isolates, namely J6 and J8, were identified based on their ability to grow at high concentrations of chlorpyrifos and used for further tests. The plant growth-promoting trains like production of Indole Acetic Acid, ammonia, protease, HCN, siderophore were analyzed. The phosphate solubilization ability and antimicrobial activities were also studied. A pot study of formulations prepared using J6 and J8 was conducted to examine the effects of these formulations on germination of seeds of Vigna radiata.

Toxicity bioassay of chlorpyrifos on some local fish species of northern Bangladesh

Four fish species namely, stinging catfish (Heteropneustes fossilis), spotted snakehead (Channa punctatus), climbing perch (Anabas testudineus) and tangra (Batasio tengana) were exposed to various concentrations of chlorpyrifos (Expert 20 EC) to investigate the mortality rate of fish species and the toxicity level of the pesticide. The LC50 values of chlorpyrifos on these fish species were 23.10, 20.32, 16.61 and 13.94 ppm, respectively at 96 hours of exposure. Among these results, it is clear that the lethal concentration is varying species to species because their strengths are different. During the experiments, some water quality parameters such as temperature, pH, dissolved oxygen, total dissolved solids and electrical conductivity were also analyzed. The temperature values remained within the ranges of 17-20°C and the pH was 6.64-7.30. Fish behaved irregular and erratic movements followed by hyper excitability, loss of balance and settling to the bottom of the test chamber. At higher concentration of chlorpyrifos, alterations in physiological and behavioural responses especially erratic swimming, gulping, mucus secretion, increased opercular movement and profuse emission of mucus all above the body were observed during the primary stages of contact after which it became occasional.

Cholinesterase activity in the cup oyster Saccostrea sp. exposed to chlorpyrifos, imidacloprid, cadmium and copper

In the present study, the sensitivity and concentration dependence of three functionally-defined components of cholinesterase activity (total: T-ChE; eserine-sensitive: Es-ChE; and eserine-resistant: Er-ChE) were quantified in the gill, digestive gland and adductor muscle of the tropical cup oyster Saccostrea sp., following acute (96h) aqueous exposure to commercial formulations of the organophosphate (OP) insecticide chlorpyrifos and the neonicotinoid (NN) imidacloprid (concentration range: 0.1–100mg/L), as well as to dissolved cadmium and copper (concentration range: 1–1000μg/L). Oysters (1.5–5.0cm shell length), field-collected from a boating marina in Santa Marta, Colombia (Caribbean Sea) were exposed in the laboratory to each substance at five concentrations. T-ChE, Es-ChE, and Er-ChE activity were quantified in the three tissues in pools of 5 individuals (3 replicates per concentration), before and after inhibition with the total cholinesterase inhibitor eserine (physostigmine, 100µM). Oysters exposed to chlorpyrifos, imidacloprid and Cd showed reduced T-ChE and Es-ChE activity in gills at highest exposure concentrations, with Es-ChE activity being inhibited proportionally more so than T-ChE, whereas Er-ChE activity showed no significant concentration-response. Digestive gland also showed diminished T-ChE, Es-ChE and Er-ChE activity for highest chlorpyrifos and Cd concentrations relative to controls, but an increase of T-ChE and Er-ChE activity at the highest imidacloprid concentration (100mg/L). For Cu, T-ChE, Es-ChE and Er-ChE activities in gills and digestive gland were elevated relative to controls in oysters exposed to Cu concentrations > 100µg/L. In adductor muscle, T-ChE, Es-ChE and Er-ChE activity showed no apparent pattern for any of the four xenobiotics and concentration levels tested. Although this study confirms acute (96h) concentration-dependent reduction of tissue T-ChE and Es-ChE activity in gills and digestive glands of Saccostrea sp. exposed to high concentrations of chlorpyrifos (100mg/L), significant changes in T-ChE, Es-ChE and Er-ChE were also caused by exposure to Cd and Cu at concentrations > 100µg/L and by exposure to imidacloprid (100mg/L), indicating that cholinesterase activity is not a specific biomarker of organophosphate exposure in this species, but, rather, a biomarker of diverse xenobiotic exposure.

Spatial and temporal distribution of pesticides and PCBs in the atmosphere using XAD-resin based passive samplers: A case study in the Quequén Grande River watershed, Argentina

XAD-resin based passive air samplers were deployed at 10 sites in the Quequén Grande River watershed in Argentina during three periods to evaluate the spatial and temporal variations of pesticides and PCBs in the atmosphere. Endosulfan and chlorpyrifos were the most prevalent pesticide because of their continued usage in Argentina, while DDTs, HCHs, chlordanes, dieldrin and heptachlors registered lower levels, reflecting their use in the past. Atmospheric endosulfan levels were 1–2 orders of magnitude higher during the application period (application: 800–12,000 pg/m3, pre- and post-application: <2–350 pg/m3), suggesting that its use in the area continued even after a ban came into effect. The remaining organochlorine pesticides also reached higher concentrations during this period, which is more likely attributable to temperature controlled air-surface exchange than current applications. The highest concentrations of chlorpyrifos were recorded during the application period, in particular at agricultural sites, where its use is wide-spread on soybean fields. The fungicide chlorothalonil was found predominantly at urban sites and in proximity to Quequén harbor, suggesting that its use might be domestic and as a biocide in antifouling paints. A different temporal pattern was observed for the herbicide trifluralin, suggesting its use in the early stages of the wheat-growing season during winter. Limited spatial variations in PCBs levels indicate a diffuse contamination source in the study area, while their relatively high correlation with temperature suggests re-volatilization from local sources. Relative enrichment of lighter PCBs congeners could be attributed to re-evaporation from secondary sources as well as atmospheric transport from urban sites.

Bacteria contribute to pesticide degradation in cryoconite holes in an Alpine glacier

Organic contaminants deposited on glacier snow and ice are subject to partitioning and degradation processes that determine their environmental fate and, consequently, their accumulation in ice bodies. Among these processes, organic compound degradation by supraglacial bacteria has been investigated to a lesser extent than photo- and chemical degradation. We investigated biodegradation of the organophosphorus insecticide chlorpyrifos (CPF), a xenobiotic tracer that accumulates on glaciers after atmospheric medium- and long-range transport, by installing in situ microcosms on an Alpine glacier to simulate cryoconite hole systems. We found that biodegradation contributed to the removal of CPF from the glacier surface more than photo- and chemical degradation. The high concentration of CPF (2–3 μg g−1 w.w.) detected in cryoconite holes and the estimated half-life of this compound (35–69 days in glacier environment) indicated that biodegradation can significantly reduce CPF concentrations on glaciers and its runoff to downstream ecosystems. The metabolic versatility of cryoconite bacteria suggests that these habitats might contribute to the degradation of a wide class of pollutants. We therefore propose that cryoconite acts as a “biofilter” by accumulating both pollutants and biodegradative microbial communities. The contribution of cryoconite to the removal of organic pollutants should be included in models predicting the environmental fate of these compounds in cold areas.

Evaluation of chlorpyrifos effects, alone and combined with lipopolysaccharide stress, on DNA integrity and immune responses of the three-spined stickleback, Gasterosteus aculeatus

Organism immune defences might be weakened by pollutants, largely detected in aquatic ecosystems, leading to the facilitation for opportunistic pathogens to infect organisms. In this context, destabilization of fish non-specific immune parameters and erythrocyte DNA integrity was tested, on a model fish species, the three-spined stickleback (Gasterosteus aculeatus), after exposure to chlorpyrifos (CPF). Alone, pesticide exposure induced a genotoxic potential (chlorpyrifos at 1.75 and 0.88µg/L) in addition to a decrease in phagocytosis capacity and a stimulation of respiratory burst. Then, to mimic pathogenic infection, fish exposure to chlorpyrifos was combined with lipopolysaccharides (LPS) stress. In this second experiment, an increase of DNA damage was observed in fish exposed to a lower concentration of chlorpyrifos and LPS. Moreover, at the higher concentration of chlorpyrifos, an early destabilization of innate immunity was observed as suggested by the absence of an increase of lysosomal presence in fish injected with LPS. This study highlighted the usefulness of stress on stress responses to better understand the impact of contaminants on the organism's health.

Histopathological effects of chlorpyrifos on the gills, hepatopancreas and gonads of the freshwater crab Zilchiopsis collastinensis. Persistent effects after exposure

Sublethal effects of the pesticide chlorpyrifos were evaluated in the crab Zilchiopsis collastinensis (Decapoda, Trichodactylidae). Crabs were exposed to high concentrations of chlorpyrifos at the beginning of the experiment and controlled dilution, under natural light and temperature conditions. A control and three concentrations (22.4, 41.25 and 61.4µg chlorpyrifos L−1) were evaluated in triplicate. Nine crabs per concentration and day were used. The gills, hepatopancreas and ovaries were sampled before pesticide exposure (day 0) and 8, 15 and 22 days later, when concentrations were diluted and below the detection limits. The histopathological effects and their variations in time were observed and quantified. In gills, hyperplasias were observed in several cases, mainly in crabs exposed to chlorpyrifos. The number of collapsed lamellae and the number of affected lamellae quickly increased in exposed crabs, as effects were observed on day 8 and remained until day 22. In hepatopancreas there was an increase in the number of F and B -cells and affected tubules, especially after 22 days of exposure (p<0.05). In ovaries, there were no effects on gonadosomatic indexes or oocyte volume, but there was a significant increase in the atretic oocyte proportion related to pesticide exposure (p<0.05). The histopathological effects on the gills, hepatopancreas and ovaries were observed after exposure and persist even after dilution, and might be related to earlier exposures. Thus, these histopathological effects might be used as pesticide biomarkers even after the pesticide is not detected by chemical methods.

Biochemical and histological biomarkers in the midgut of Apis mellifera from polluted environment at Beheira Governorate, Egypt.

The aim of this study was to analyze the impact of organophosphorus (OP) pollutants on oxidative stress and ultrastructural biomarkers in the midgut of the honeybee Apis mellifera collected from three locations that differ in their extent of spraying load with OP insecticides: a weakly anthropised rural site, Bolin which is considered as a reference site; moderately spraying site, El Kaza; and a strongly anthropised urban site, Tiba with a long history of pesticide use. Results showed that high concentrations of chlorpyrifos, malathion, diazinon, chlorpyrifos-methyl, and pirimiphos-methyl were detected in midgut at locations with extensive pesticide spraying. Reduced glutathione content, superoxide dismutase, catalase, and glutathione peroxidase displayed lowest activities in the heavily sprayed location (Tiba). Lipid peroxidation level in the midgut of honeybees in the sprayed locations was found to be significantly higher compared to the reference values. Meanwhile, various ultrastructural abnormalities were observed in the epithelial cells of midgut of honeybees collected from El Kaza and Tiba, included confluent and disorganized microvilli and destruction of their brush border, the cytoplasm with large vacuoles and alteration of cytoplasmic organelles including the presence of swollen mitochondria with lysis of matrices, disruption of limiting membranes, and disintegration of cristae. The nuclei with indented nuclear envelope and disorganized chromatin were observed. These investigated biomarkers indicated that the surveyed honeybees are being under stressful environmental conditions. So, we suggest using those biomarkers in the assessment of environmental quality using honeybees in future monitoring of ecotoxicological studies.

The Effect of Chlorpyrifos, an Organophosphorus Pesticide, on Glucose Uptake in Whole Blood

Background: The sub-chronic organophosphorous pesticide exposure increases insulin resistance and thereby the risk of developing Diabetes mellitus. Its acute exposure also leads to hyperglycemia. However, the mechanism is not clearly understood. Hence the present study explores the effect of low and high concentrations of chlorpyrifos, an organophosphorous pesticide on glucose uptake by whole blood and its possible mechanism. Methods: Level of hemolysis was determined at 2 and 4 hours of 1% DMSO (Dimethyl sulfoxide) and chlorpyrifos (0.3 and 10 mg/ml) exposure. The whole blood was treated in vitro with vehicle i.e., 1% DMSO or high (10 mg/mL) or low (0.3 mg/mL) concentrations of chlorpyrifos dissolved in DMSO. Then plasma glucose was assayed by glucose oxidase method at 2 and 4 hours of DMSO/chlorpyrifos exposures. Results: There was no effect of DMSO or 0.3 mg/ml chlorpyrifos treatment on hemolysis even after 4 hours. 10 mg/ml chlorpyrifos causes 2% and 14% hemolysis at 2 and 4 hours respectively. Plasma glucose level decreased in a time dependent manner after collection of blood (15.9% at 2 hours and 34.2% at 4 hour) in presence of DMSO. Chlorpyrifos did not influence glucose uptake by whole blood in low concentration but suppressed it in high concentration. Conclusion: Chlorpyrifos does not impair glucose uptake at low concentration in whole blood probably because major glucose uptake in blood is insulin independent. The impaired glucose uptake at high concentration is mostly due to toxic metabolic impairment and partly due to its hemolytic effect.

Ecotoxicity of two organophosphate pesticides chlorpyrifos and dichlorvos on non-targeting cyanobacteria Microcystis wesenbergii.

Organophosphate pesticides (OPs), as a replacement for the organochlorine pesticides, are generally considered non-toxic to plants and algae. Chlorpyrifos and dichlorvos are two OPs used for pest control all over the world. In this study, the dose-response of cyanobacteria Microcystis wesenbergii on OPs exposure and the stimulating effect of OPs with and without phosphorus source were investigated. The results showed that high concentrations of chlorpyrifos and dichlorvos caused significant decrease of chlorophyll a content. The median inhibitory concentrations (EC50) of chlorpyrifos and dichlorvos at 96 h were 15.40 and 261.16 μmol L(-1), respectively. Growth of M. wesenbergii under low concentration of OPs (ranged from 1/10,000 to 1/20 EC50), was increased by 35.85 % (chlorpyrifos) and 41.83 % (dichlorvos) at 120 h, respectively. Correspondingly, the highest enhancement on the maximum quantum yield (F v/F m) was 4.20 % (24 h) and 9.70 % (48 h), respectively. Chlorophyll fluorescence kinetics, known as O-J-I-P transients, showed significant enhancements in the O-J, J-I, and I-P transients under low concentrations of dichlorvos at 144 h, while enhancements of chlorophyll fluorescence kinetics induced by low concentrations of chlorpyrifos were only observed in the J-I transient at 144 h. Significant decreases of chlorophyll content, F v/F m and O-J-I-P transients with OPs as sole phosphorus source were found when they were compared with inorganic phosphate treatments. The results demonstrated an evidently hormetic dose-response of M. wesenbergii to both chlorpyrifos and dichlorvos, where high dose (far beyond environmental concentrations) exposure caused growth inhibition and low dose exposure induced enhancement on physiological processes. The stimulating effect of two OPs on growth of M. wesenbergii was negligible under phosphate limitation.

Isolation and characterization of a highly efficient chlorpyrifos degrading strain of Cupriavidus taiwanensis from sludge.

In this study, a highly effective chlorpyrifos (CP)-degrading bacterium (termed strain X1) was isolated from the sludge of drain outlet of a chlorpyrifos manufacturer. Strain X1 was identified as Cupriavidus taiwanensis based upon the analysis of the 16S rDNA gene and biochemical characteristics, which is capable of transforming CP into 3,5,6-trichloro-2-pyridinol (TCP), and the resulting TCP was further metabolized when performed in an aqueous medium. Degradation experiments were carried out under different conditions at the range of pH (5.0∼9.0) and temperature (22∼42 °C), and the optimized pH and temperature were 7.0 and 32 °C respectively. Biotransformation of high concentration of CP was also determined; 400 mg l(À1) of CP was completely transformed within 36 h; approximately 95% of CP was removed within 48 h when concentration of CP was up to 500 mg l(À1) . A genomic library was successfully constructed to clone the gene encoding the CP hydrolase, and a positive transformant with clear hydrolytic zones was obtained and analyzed. The insert gene sequence exhibited close relationship with 99% similar to opdB gene encoding parathion hydrolase, whereas, transformant failed in degrading the accumulated TCP. These results highlight the potential of this bacterium to be used in the cleanup of CP.
