Antihypertensive effects of whey protein hydrolysate involve reshaping the gut microbiome in spontaneously hypertension rats

Novel angiotensin-converting enzyme (ACE) inhibitory peptides were identified from whey protein hydrolysates (WPH) in vitro in our previous study and the antihypertensive abilities of WPH in vivo were further investigated in the current study. Results indicated that WPH significantly inhibited the development of high blood pressure and tissue injuries caused by hypertension. WPH inhibited ACE activity (20.81 %, P < 0.01), and reduced renin concentration (P < 0.05), thereby reducing systolic blood pressure (SBP) (12.63 %, P < 0.05) in spontaneously hypertensive rats. The increased Akkermansia, Bacteroides, and Lactobacillus abundance promoted high short chain fatty acid content in feces after WPH intervention. These changes jointly contributed to low blood pressure. The heart weight and cardiomyocyte injuries (hypertrophy and degeneration) were alleviated by WPH. The proteomic results revealed that 19 protein expressions in the heart mainly associated with the wingless/integrated (Wnt) signaling pathway and Apelin signaling pathway were altered after WPH supplementation. Notably, WPH alleviated serum oxidative stress, indicated by the decreased malondialdehyde content (P < 0.01), enhanced total antioxidant capacity (P < 0.01) and superoxide dismutase activity (P < 0.01). The current study suggests that WPH exhibit promising antihypertensive abilities in vivo and could be a potential alternative for antihypertensive dietary supplements.

Enzymatic hydrolysates from sea cucumber body wall prevent low calcium-induced osteoporosis by regulating calcium absorption and gut microbiota

Inadequate calcium intake or intestinal malabsorption is closely related to osteoporosis. The aim of this study was to investigate the effects of sea cucumber enzymatic hydrolysates (SCEH) prepared from body wall on calcium absorption in calcium-deficient rats. Fifty-six female Sprague-Dawley rats on a low calcium diet that induced osteoporosis were fed SCEH and calcium carbonate for 3 months. The results showed that oral administration of SCEH and calcium carbonate both increased the body weight (bw) and bone mineral density (BMD) in calcium-deficient rats to varying degrees, thereby alleviating the adverse effects of osteoporosis. In addition, oral administration of a combination of SCEH (2000 mg/kg bw/day) and calcium (333 mg/kg bw/day) upregulated the expression of calcium transporter genes (TRPV6 and Calbindin-D9k) in the intestines, increased serum 1,25(OH)2D3 levels and BMD, decreased trabecular bone separation, and inhibited parathyroid hormone (PTH) release in calcium-deficient rats compared to calcium carbonate groups. Thus, this combination promoted calcium absorption and improved the efficiency of calcium utilization. These results might be associated with a higher short chain fatty acid (SCFA) level in the gut and an increased abundance of butyrate and acetate producing bacteria, such as Christensenellaceae R7 group, UCG-005 and Clostridium UCG-014. Therefore, a diet containing SCEH and calcium combinations might be recommended to increase bone mass and inhibit osteoporosis.

AhR Activation Ameliorates Intestinal Barrier Damage in Immunostressed Piglets by Regulating Intestinal Flora and Its Metabolism.

The primary factor leading to elevated rates of diarrhea and decreased performance in piglets is immunological stress. The regulation of immune stress through the intestinal flora is a crucial mechanism to consider. In total, 30 weaned piglets were randomly allocated to five groups: the basal diet group (Control), basal diet + lipopolysaccharides group (LPS), basal diet + 250 μg/kg 6-Formylindolo [3,2-b] carbazole + LPS group (FICZ), basal diet + 3mg/kg Cardamonin + LPS group (LCDN), and basal diet + 6mg/kg Cardamonin + LPS group (HCDN/CDN). The results showed that compared with those of the LPS group, the expression of tight junction proteins (occludin; claudin-1) in the FICZ group was significantly increased, and the mRNA levels of IL-1β and TNF-α were significantly reduced (p < 0.05). HCDN treatment had a better effect on LPS-induced intestinal barrier damage in this group than it did in the LCDN group. HCDN treatment leads to a higher villus height (VH), a higher ratio of villi height to crypt depth (V/C), higher tight junction proteins (ZO-1; occludin), and higher short-chain fatty acids (SCFAs). In addition, correlation analyses showed that Succinivibrio was positively correlated with several SCFAs and negatively correlated with prostaglandin-related derivatives in the FICZ group and CDN group (p < 0.05). In summary, Cardamonin alleviates intestinal mucosal barrier damage and inflammatory responses by regulating the intestinal microbiota and its metabolism.

Low Short-Chain-Fatty-Acid-Producing Activity of the Gut Microbiota Is Associated with Hypercholesterolemia and Liver Fibrosis in Patients with Metabolic-Associated (Non-Alcoholic) Fatty Liver Disease

The aim of this study was to investigate the short-chain fatty acid (SCFA) activity of the gut microbiota of patients with metabolic-associated fatty liver disease (MAFLD). The level and spectrum of short-chain fatty acids (SCFAs) were determined via gas–liquid chromatography. Liver fibrosis was assessed using the FIB-4 index and elastography. Among 42 non-cirrhotic MAFLD patients, 24 had high fecal SCFA levels (group H) and 18 had low fecal SCFA levels (group L). Patients in group H had lower serum uric acid, total cholesterol, and LDL cholesterol levels but a higher BMI than those in group L. All patients in group L and only 37.9% of those in group H were found to have hypercholesterolemia. In patients with hypercholesterolemia, the level of SCFAs was lower than that in patients without hypercholesterolemia. Patients in group H had less liver fibrosis than patients in group L. A total of 50.0% of the patients in group H and 92.3% of those in group L had significant liver fibrosis (≥F2). Patients with significant liver fibrosis had lower levels of fecal SCFAs—particularly acetate and butyrate. The fecal SCFA levels were positively correlated with gamma-glutamyl transferase, total bilirubin levels, BMI, and platelet count and were negatively correlated with FIB-4, liver stiffness, serum total, and LDL cholesterol levels.

Potency of fermented jicama extract cultured with Lactobacillus plantarum B1765 for producing short chain fatty acid

Short-chain fatty acid (SCFA) is the end product of inulin metabolism, which has various health benefits, such as affecting lipid metabolism and as an antidiabetic agent. This research studied the effect of the fermentation time of jicama extract with Lactobacillus plantarumB1765 as a starter culture on producing SCFA. Fermentation was conducted for 2, 12, 24, and 36 hours at a temperature of 37°C with 5% (v/v) of the starter culture, then determined the growth of LAB, pH, Total Titratable Acid (TTA), fructose, and Short Chain Fatty Acid content. The total LAB count was measured using the Total Plate Count; fructose was measured using the Nelson-Somogyi method, while SCFA was measured using HPLC with eluents H3PO4 0.1% and acetonitrile 25%. The length of fermentation affected the increase in total Lactic Acid Bacteria (LAB), Total titratable acidity (TTA), fructose, SCFA, and the decrease in pH. The results showed that the optimal growth of Total LAB at 24 hours of fermentation correlated with fructose content, but increasing TTA and SCFA until 36 hours of fermentation and pH decreased to 3,92 at the end of fermentation and decreasing pH due to lactic acid and SCFA producing. SCFA was identified as consisting of acetic acid (1.31±0.006 mg/mL), propionic acid (1.58±0.0055mg/mL), and butyric acid (1.05±0.0038mg/mL), with the highest SCFA was propionic acid. Therefore, fermented jicama extract with Lactobacillus plantarum B1765 has the potential to produce SCFA, which could be used for various health benefits.

Anaerobic Digestion of Hemp and Flax Straw and Shives and Rapeseed Straw by the Ruminal Microbiota

To mitigate greenhouse gas emissions and improve energy supply security, there is an increasing effort toward the use of non-fossil energy sources. Crop residues have a great potential to be exploited as biomass for biogas production. However, due to their lignocellulosic structures they are difficult to degrade and do not reach competitive performance. A feasible option to mine these substrates is present in the forestomach of ruminants. Therefore, the aim of the present study was to use rumen microorganisms to improve anaerobic digestion (AD) of crop residues. For this purpose, hemp straw, mechanically pre-treated hemp fibers and shives, flax straw, flax shives, and aged and fresh rapeseed straw were evaluated using the rumen simulation technique. The AD of the substrates was divided into three batches. In two batches, hay was added as a control substrate. In summary, none of the analyzed substrates had an equivalent performance as the control hay, but pre-treated hemp fibers and shives had better AD parameters compared to all other alternative substrates, with the lowest pH (mean: 6.81), highest short chain fatty acid (20.0 mmol/day) and H2 production (25.6 mM) and highest degradability (25.2%). Flax straw had the second-best performance (6.81, 17.4 mmol/day, 20.6 mM and 22.2%, respectively), followed by fresh rapeseed straw, hemp straw, aged rapeseed straw and flax shives. Therefore, hemp fibers and shives demonstrated to be the most suitable substrates for AD. However, since pre-treatment can represent significant additional costs for biogas production, flax straw also demonstrated to be a good alternative.

Dietary Fiber and Microbiota Metabolite Receptors Enhance Cognition and Alleviate Disease in the 5xFAD Mouse Model of Alzheimer's Disease.

Alzheimer's disease (AD) is a neurodegenerative disorder with poorly understood etiology. AD has several similarities with other "Western lifestyle" inflammatory diseases, where the gut microbiome and immune pathways have been associated. Previously, we and others have noted the involvement of metabolite-sensing GPCRs and their ligands, short-chain fatty acids (SCFAs), in protection of numerous Western diseases in mouse models, such as Type I diabetes and hypertension. Depletion of GPR43, GPR41, or GPR109A accelerates disease, whereas high SCFA yielding diets protect in mouse models. Here, we extended the concept that metabolite-sensing receptors and SCFAs may be a more common protective mechanism against Western diseases by studying their role in AD pathogenesis in the 5xFAD mouse model. Both male and female mice were included. Depletion of GPR41 and GPR43 accelerated cognitive decline and impaired adult hippocampal neurogenesis in 5xFAD and WT mice. Lack of fiber/SCFAs accelerated a memory deficit, whereas diets supplemented with high acetate and butyrate (HAMSAB) delayed cognitive decline in 5xFAD mice. Fiber intake impacted on microglial morphology in WT mice and microglial clustering phenotype in 5xFAD mice. Lack of fiber impaired adult hippocampal neurogenesis in both W and AD mice. Finally, maternal dietary fiber intake significantly affects offspring's cognitive functions in 5xFAD mice and microglial transcriptome in both WT and 5xFAD mice, suggesting that SCFAs may exert their effect during pregnancy and lactation. Together, metabolite-sensing GPCRs and SCFAs are essential for protection against AD, and reveal a new strategy for disease prevention.Significance Statement Alzheimer's disease (AD) is one of the most common neurodegenerative diseases; currently, there is no cure for AD. In our study, short-chain fatty acids and metabolite receptors play an important role in cognitive function and pathology in AD mouse model as well as in WT mice. SCFAs also impact on microglia transcriptome, and immune cell recruitment. Out study indicates the potential of specialized diets (supplemented with high acetate and butyrate) releasing high amounts of SCFAs to protect against disease.

Productive, Qualitative, and In Vitro Fermentation Traits of Amaranthus Grains as Potential Ingredients for Pig Diet

The present work compared the agronomic traits, chemical composition, fatty acid profile, and in vitro fermentation characteristics of twelve accessions of Amaranthus spp., belonging to A. cruentus, A. hybridus, A. hypochondriacus, and A. tricolor, grown in a semiarid Mediterranean area. Among accessions, Benin and Arizona (A. cruentus) and Pennsylvania (A. hypochondriacus) showed the highest seed yield (on average, 322.1 g m−2), while Taiwan (A. tricolor) and India and Iowa (A. hypochondriacus) the highest thousand seed weight (on average, 0.81 g). Among the species, A. hypochondriacus showed the highest crude protein (16 g 100g−1), starch (51.5 g 100g−1), and soluble detergent fiber (2.03 g 100g−1) contents and the most favorable in vitro fermentation characteristics with the highest short-chain fatty acid (SCFA 52.6 mmol g−1) and butyric acid (20.7% SCFA) production together with the lowest crude fiber (4.93 g 100g−1) and insoluble dietary fiber (12.5 g 100g−1) content. Arizona (A. cruentus) showed the highest level of monounsaturated fatty acids (32.67 g 100g−1), Ohio (A. hybridus) had the highest levels of polyunsaturated fatty acids (44.62 g 100g−1) and n6-PUFA (44.21 g 100g−1), and India (A. hypochondriacus) had the highest level of n3-PUFA (0.63 g 100g−1). A. hypochondriacus exhibited not only desirable nutritive characteristics, agronomic traits, and suitability to Mediterranean growing conditions, but also a potential beneficial effect. Nonetheless, it is recommended to run longer-term field trials to confirm these findings and to assess the genotype by environment interaction either with current accessions or others from the wide Amaranth germplasm available.

Anthocyanin-rich blue potato meals protect against polychlorinated biphenyl-mediated disruption of short-chain fatty acid production and gut microbiota profiles in a simulated human digestion model.

Polychlorinated biphenyls (PCBs) are ubiquitous environmental pollutants associated with a wide variety of adverse human health outcomes. PCB 126 and PCB 153 are among the most prevalent congeners associated with human exposure. Emerging studies have suggested that PCB exposure leads to lower gut microbial diversity although their effects on microbial production of health promoting short-chain fatty acids (SCFAs) has been scarcely studied. Blue potatoes are rich in anthocyanins (ACNs), which is a class of polyphenols that promote the growth of beneficial intestinal bacteria such as Bifidobacterium and Lactobacillus and increase the generation of SCFAs. A batch-culture, pH-controlled, stirred system containing human fecal microbial communities was utilized to assess whether human gut microbiota composition and SCFA production are affected by: (a) PCB 126 and PCB 153 exposure; and (b) ACN-rich digests in the presence and absence of the PCB congeners. Anthocyanin-rich blue potato meals (11.03 g) were digested over 12 h with and without PCB 126 (0.5 mM) and PCB 153 (0.5 mM) using an in vitro simulated gut digestion model involving upper gastrointestinal digestion followed by metabolism by human fecal microbiota. Fecal digests were collected for analysis of gut microbial and SCFA profiles. Polychlorinated biphenyl-exposed fecal samples showed a significant (p < 0.05) decrease in species richness and a significantly (p < 0.05) different microbial community structure. PCB treatment was associated with an increased (p < 0.05) relative abundance of Akkermansia, Eggerthella, and Bifidobacterium and a decreased (p < 0.05) relative abundance of Veillonella, Streptococcus, and Holdemanella. ACN digests counteracted the altered abundances of Akkermansia and Bifidobacterium seen with the PCB treatment. PCB exposure was associated with a significant (p < 0.05) decrease in total SCFA and acetate concentrations. ACN digests were associated with significantly (p < 0.05) higher SCFA and acetate concentrations in the presence and absence of PCBs. Human fecal matter exposed to PCB 126 and PCB 153 led to decreased abundance and altered gut microbiota profiles as well as lowered SCFA and acetate levels. Importantly, this study showed that prebiotic ACN-rich potatoes counteract PCB-mediated disruptions in human gut microbiota profiles and SCFA production.

Differences in fine arabinoxylan structures govern microbial selection and competition among human gut microbiota

Dietary fibers are known to modulate microbiome composition, but it is unclear to what extent minor fiber structural differences impact community assembly, microbial division of labor, and organismal metabolic responses. To test the hypothesis that fine linkage variations afford different ecological niches for distinct communities and metabolism, we employed a 7-day in vitro sequential batch fecal fermentation with four fecal inocula and measured responses using an integrated multi-omics approach. Two sorghum arabinoxylans (SAXs) were fermented, with one (RSAX) having slightly more complex branch linkages than the other (WSAX). Although there were minor glycoysl linkage differences, consortia on RSAX retained much higher species diversity (42 members) than on WSAX (18–23 members) with distinct species-level genomes and metabolic outcomes (e.g., higher short chain fatty acid production from RSAX and more lactic acid produced from WSAX). The major SAX-selected members were from genera of Bacteroides and Bifidobacterium and family Lachnospiraceae. Carbohydrate active enzyme (CAZyme) genes in metagenomes revealed broad AX-related hydrolytic potentials among key members; however, CAZyme genes enriched in different consortia displayed various catabolic domain fusions with diverse accessory motifs that differ among the two SAX types. These results suggest that fine polysaccharide structure exerts deterministic selection effect for distinct fermenting consortia.

Effects of partially hydrolyzed guar gums of different molecular weights on a human intestinal in vitro fermentation model

Partially hydrolyzed guar gums (PHGGs) are prebiotic soluble dietary fibers. High molecular-weight PHGGs have rapid fermentation and high short-chain fatty acid (SCFA) productivity rates, compared to low molecular-weight PHGGs. Therefore, low molecular-weight PHGGs may have less pronounced prebiotic effects than high molecular-weight PHGGs. However, the effects of PHGGs of different molecular weights on the human intestinal microbiota, as well as their fermentation ability and prebiotic effects, have not been investigated. The aim of this study was to evaluate the effects of two PHGGs of different molecular weights, Sunfiber-R (SF-R; 20kDa) and Sunfiber-V (SF-V; 5kDa), on human colonic microbiota and SCFA production. A human intestinal invitro fermentation model was operated by fecal samples with and without the PHGGs. The addition of 0.2% SF-R or SF-V increased the relative abundance of Bacteroides spp., especially that of Bacteroides uniformis. This increase corresponded to a significant (p=0.030) and non-significant (p=0.073) increase in propionate production in response to SF-R and SF-V addition, respectively. Both fibers increased the relative abundance of Faecalibacterium and stimulated an increase in the abundance of unclassified Lachnospiraceae and Bifidobacterium. In conclusion, the low molecular-weight PHGG exerted prebiotic effects on human colonic microbiota to increase SCFA production and bacteria that are beneficial to human health in a manner similar to that of the high molecular-weight forms of PHGG.

Butyrate Differentiates Permissiveness to Clostridioides difficile Infection and Influences Growth of Diverse C. difficile Isolates.

A disrupted "dysbiotic" gut microbiome engenders susceptibility to the diarrheal pathogen Clostridioides difficile by impacting the metabolic milieu of the gut. Diet, in particular the microbiota-accessible carbohydrates (MACs) found in dietary fiber, is one of the most powerful ways to affect the composition and metabolic output of the gut microbiome. As such, diet is a powerful tool for understanding the biology of C. difficile and for developing alternative approaches for coping with this pathogen. One prominent class of metabolites produced by the gut microbiome is short-chain fatty acids (SCFAs), the major metabolic end products of MAC metabolism. SCFAs are known to decrease the fitness of C. difficile in vitro, and high intestinal SCFA concentrations are associated with reduced fitness of C. difficile in animal models of C. difficile infection (CDI). Here, we use controlled dietary conditions (8 diets that differ only by MAC composition) to show that C. difficile fitness is most consistently impacted by butyrate, rather than the other two prominent SCFAs (acetate and propionate), during murine model CDI. We similarly show that butyrate concentrations are lower in fecal samples from humans with CDI than in those from healthy controls. Finally, we demonstrate that butyrate impacts growth in diverse C. difficile isolates. These findings provide a foundation for future work which will dissect how butyrate directly impacts C. difficile fitness and will lead to the development of diverse approaches distinct from antibiotics or fecal transplant, such as dietary interventions, for mitigating CDI in at-risk human populations. IMPORTANCE Clostridioides difficile is a leading cause of infectious diarrhea in humans, and it imposes a tremendous burden on the health care system. Current treatments for C. difficile infection (CDI) include antibiotics and fecal microbiota transplant, which contribute to recurrent CDIs and face major regulatory hurdles, respectively. Therefore, there is an ongoing need to develop new ways to cope with CDI. Notably, a disrupted "dysbiotic" gut microbiota is the primary risk factor for CDI, but we incompletely understand how a healthy microbiota resists CDI. Here, we show that a specific molecule produced by the gut microbiota, butyrate, is negatively associated with C. difficile burdens in humans and in a mouse model of CDI and that butyrate impedes the growth of diverse C. difficile strains in pure culture. These findings help to build a foundation for designing alternative, possibly diet-based, strategies for mitigating CDI in humans.

Insight into the long-term impact of birth weight on intestinal development, microbial settlement, and the metabolism of weaned piglets.

Infant mortality of low birth body weight (LBBW) piglets can reach 10% and is mainly due to gut and immune system immaturity which can lead to a higher risk in the long term. This study aimed to assess the impact of birth body weight (BBW) on piglet metabolism, gut status, and microbial profile from weaning to 21 d postweaning. At birth, 32 piglets were selected for their BBW and inserted into the normal BBW (NBBW:1.38 ± 0.09 g) or the LBBW (0.92 ± 0.07 g) group. The piglets were weighed weekly from weaning (d0) to d21. At d9 and d21, 8 piglets/group were slaughtered to obtain the distal jejunum for morphology, immunohistochemistry, and gene expression analysis, colon content for microbiota and short-chain fatty acid (SCFA) analysis, and intestinal content for pH measurement. Blood was collected for metabolomic, haptoglobin (Hp), and reactive oxygen metabolite (ROM) analysis. The LBBW group had a lower body weight (BW) throughout the study (P < 0.01), a lower average daily gain from d9-d21 (P = 0.002), and lower feed intake (P = 0.02). The LBBW piglets had lower Hp at d9 (P = 0.03), higher ROMs at d21 (P = 0.06), and a net alteration of the amino acid (AA) metabolism at d9 and d21. A higher expression of NFKB2 was observed in the LBBW piglets at d9 (P = 0.003) and d21 (P < 0.001). MYD88 expression was enhanced in NBBW piglets at d9 (P < 0.001). The LBBW piglets had a lower villus height, absorptive mucosal surface (P = 0.01), and villus height:crypt depth ratio (P = 0.02), and a greater number of T-lymphocytes in both the epithelium and the crypts (P < 0.001) at d21. At d21, the LBBW piglets had higher lactic acid, acetate, butyrate, and valerate, and also higher SCFA in the colon (P < 0.05). The LBBW piglets had a higher Shannon index (P = 0.01) at d9 and a higher abundance of SCFA-fermenting bacteria. In conclusion, the present study confirmed that LBBW could impact the gut mucosal structure, immunity, and inflammatory and oxidative status, leading to an altered AA metabolism, and delaying the recovery from weaning.

PH regulated potassium ferrate oxidation promotes acetic acid yield and phosphorous recovery rate from waste activated sludge

To improve the dose efficiency of K2FeO4 in waste activated sludge (WAS) treatment, pH regulation on K2FeO4 pretreatment and acidogenic fermentation was investigated. Four pretreatments were compared, i.e. pH3 + 50 g/kg-TS, pH10 + 50 g/kg-TS, neutral pH + 50 g/kg-TS and neutral pH + 100 g/kg-TS (without pH adjustment). The higher short chain fatty acids (SCFAs) yield and phosphorous dissolution rate was found under the condition of pH 10.0. In pH10 + 50 g/kg-TS, the maximum concentration of SCFAs was 5591 mg-COD/L, which yield was 22.6 times higher than that of the neutral pH + 50 g/kg-TS (237 mg COD/L). The acidogenic fermentation period could be shortened to 5 days and acetic acid accounted for 70 % of SCFAs. Furthermore, PO43--P in the hydrolysate (346.5 mg/L) accounted for 47.59 % of TP, which is easier to be recovered by chemical precipitation. Therefore, a more economical and feasible utilization mode of potassium ferrate was proposed.

Comparative Analyses of Production Performance, Meat Quality, and Gut Microbial Composition between Two Chinese Goose Breeds.

Simple SummaryPoultry is one of the most frequently consumed meats in the world and plays an important role in the daily life of people. Goose meat is consumed by consumers because it contains a relatively high proportion of polyunsaturated fatty acids. Meat quality traits, production performance, and cecal microbiota diversity in two goose breeds (Zi goose and Xianghai flying goose) were evaluated in this study. Understanding these aspects not only provides a reference for the exploration of the relationship between the cecal microbiota and production performance but also guidelines for the human consumption of healthy poultry meat.Goose meat is consumed by consumers because it contains a relatively high proportion of polyunsaturated fatty acids (PUFAs). This study was conducted to explore the main differences in production performance, breast meat quality traits, and cecal microbiota compositions between the Zi goose (ZG) and Xianghai flying goose (FG). The production performance and breast meat quality trait analyses showed that compared with the ZG, the FG had a higher right breast muscle index, ileum villi height/crypt depth ratio (VH/CD), and cecum fermentation rate (higher short-chain fatty acid (SFCA) concentration); a lower abdominal fat index; a higher proportion of PUFAs; and a lower shear force. Spearman’s correlation coefficients between the cecal microbiota composition and production performance indexes suggested that the genus Faecalibacterium was positively associated with production performance; in contrast, the genus Candidatus Saccharimonas was negatively correlated with production performance; moreover, the Ruminococcus torques group, Parasutterella, and Methanobrevibacter were negatively related to the VH/CD. Taken together, in this particular trial, FG had better production performance, healthier meat quality traits, and better intestinal digestion and absorption capacities than ZG. These results not only provide a useful data reference for the production of healthy geese for human consumption but can also help guide the utilization of goose breed resources.

Short-Chain Fatty Acids Modulate Permeability, Motility and Gene Expression in the Porcine Fetal Jejunum Ex Vivo.

Postnatally, short-chain fatty acids (SCFA) are important energetic and signaling agents, being involved in host nutrition, gut imprinting and immune and barrier function. Whether SCFA exert similar effects during the late fetal phase has been insufficiently elucidated. This study aimed to evaluate whether the fetal jejunum senses SCFA and whether SCFA modify the muscle tension and epithelial permeability and related signaling in jejunal tissue from the porcine fetus in late gestation. Exposure of fetal jejunal tissue to a mix of SCFA (70 µmol/mL) in an organ bath for 20 min lowered the muscle tension. Moreover, SCFA decreased the transepithelial conductance while increasing the short-circuit current in the Ussing chamber, indicating reduced permeability and increased SCFA absorption. Gene expression in the tissues harvested from the Ussing chamber after 30 min indicated downregulation of the expression of receptors (i.e., FFAR2 and TLR2), MCT1 and tight-junction and adherens proteins, which may be a negative feedback response to the applied high SCFA concentration compared with the micromolar concentration detected in fetal gastric fluid. Taken together, our data demonstrate that the fetal jejunum senses SCFA, which trigger electrophysiological, muscle contraction and related gene transcription responses. Hence, SCFA may play a role in prenatal gut nutrition and imprinting.

Non-Alcoholic Components in Huangjiu as Potential Factors Regulating the Intestinal Barrier and Gut Microbiota in Mouse Model of Alcoholic Liver Injury.

Different alcoholic beverages and drinking patterns might exert divergent impacts on alcoholic liver disease (ALD) progression. Whether the abundant non-alcoholic components (NAC) in fermented wine could alleviate ethanol (EtOH)-induced adverse influences on the liver remains unknown. Hence, the chronic ALD mouse model was established to compare the effects of Huangjiu (a typical fermented wine) and EtOH feeding on the liver, intestinal barrier, gut microbiota, and intestinal short-chain fatty acids (SCFAs) content. Although Huangjiu intake led to slight hepatic steatosis, it mitigated oxidative stress, inflammation, and intestinal damage relative to EtOH intake. In comparison with EtOH feeding, Huangjiu significantly improved the intestinal barrier integrity and reduced hepatic lipopolysaccharide levels by up-regulating the expression of intestinal tight junction proteins (ZO-1 and occludin) and antimicrobial activity peptides (Reg3β and Reg3γ). The administration of Huangjiu NAC partially restored alcohol-induced gut microbiota dysbiosis via recovering the abundance of Lactobacillus, Faecalibaculum, and Akkermansia. Moreover, mice receiving Huangjiu showed higher SCFAs levels (such as acetic acid and butyric acid) than those receiving EtOH. Huangjiu consumption resulted in lower hepatotoxicity than pure EtOH, at the same alcohol dose. The NAC in Huangjiu might attenuate the progression of ALD by regulating intestinal barrier function and microbiota-meditated gut ecology.

The Analysis of Phagocytic Myeloid Cells in Low and High Fiber Fed Mice after Three Weeks of Hypoxia

Pulmonary hypertension (PH) is a devastating disease that is characterized by increased blood pressure (BP) in the pulmonary vasculature. Immune cells participate in adverse cardiovascular remodelling of pulmonary vessels and could represent targets for PH treatment. It has been shown that patients with PH have increased numbers of interstitial macrophages and dendritic cells in the lungs that originate from circulating immune cells. These myeloid immune cells migrate to the vessel wall and promote vascular hypertrophy. Attenuation of immune cell invasion into the vessel wall could potentially slow the progression of PH. One of the possible ways to reduce inflammation in the cardiovascular system is dietary modification with increased content of soluble fiber. Soluble fiber increases the level of short chain fatty acids (SCFAs) in the plasma which demonstrate anti‐inflammatory properties in systemic hypertension and cardiac overload. In different models of cardiovascular diseases high fiber (HFi) diet and SCFAs improve cardiovascular functions reducing the level of immune cells at the site of injury. The effects of HFi diet on the pulmonary circulation are unknown. This study establishes the effects of low (LFi) and HFi diet on macrophage and dendritic cell composition in the lungs in the model of hypoxia‐induced PH.C57bl/6j mice were fed with LFi and HFi diet five weeks before and three weeks during hypoxic exposure (10% O2). After that hematocrit, cecum, spleen and body weight were measured. Flow cytometry was used to identify the proportion of alveolar and interstitial macrophages and dendritic cells in the lungs.Hypoxic animals had increased hematocrit level compared to normoxic controls. Hematocrit did not differ between LFi fed animals and HFi fed animals in hypoxic conditions but in normoxia hematocrit was higher in LFi diet group. Cecum weight was significantly higher in HFi fed mice than in LFi fed mice whereas total body weight and spleen weight was not affected by diet composition. The percentage of alveolar macrophages (CD11b+CD11c+CD64+) did not differ between groups. Hypoxia increased the proportion of dendritic cells (CD11b+MHCll+CD24+) and reduced the proportion of interstitial macrophages (CD11b+MHCll+CD64+) among CD11b+MHC ll+ cells. Dietary fiber did not affect the proportion of dendritic cells and interstitial macrophages in normoxic and hypoxic animals.This data demonstrates that three weeks of hypoxia changes the proportion of CD11b+ dendritic cells and interstitial macrophages in the mouse lungs however soluble dietary fiber did not affect it.

Short-chain fatty acids promote jejunal barrier function and caecal muscle contractibility in laying hens ex vivo

ABSTRACT 1. Short-chain fatty acids (SCFA) exert beneficial actions in the gut; nevertheless, information about the effect of SCFA on physiological responses in the small intestine of chickens is rare. 2. The aim of this study was to assess the effect of 1) different molar acetate:butyrate ratios (Ac:But; Experiment 1; 78.5% acetate and 7.3% butyrate versus 71.4% acetate and 14.0% butyrate) and 2) SCFA concentrations (Experiment 2; final concentration in chambers: 70.5 versus 141 µmol SCFA/ml buffer) on the jejunal and caecal contractibility and jejunal barrier function in laying hens. The change in muscle contractibility due to the SCFA was measured in mid-jejunal and caecal segments (n = 4 each per hen) from four laying hens using the organ bath system after precontraction with acetylcholine for 15 min. Changes in short-circuit current (ISC) and transepithelial tissue conductivity (GT) as indicators for net ion flux and barrier function, respectively, were measured in mid-jejunal tissue (n = 3/hen and treatment), mounted into Ussing chambers. 3. In Experiment 1, the addition of SCFA, irrespective of the Ac:But ratio, decreased jejunal muscle tension (P < 0.05), jejunal GT as well as caused a less negative ISC (P < 0.05). In Experiment 2, the increasing SCFA concentrations increased the caecal muscle contraction and jejunal ISC by 75.6% while decreasing the GT by up to 19.6% (P < 0.05). 4. In conclusion, results demonstrate that increasing butyrate proportions and SCFA concentrations stimulate caecal muscle contraction, thereby increasing caecal mixing and emptying in vivo. Jejunal ISC and GT support a strong SCFA sensing capacity in the jejunum, as both, more butyrate and higher SCFA, increased mucosal ion uptake and barrier function.

Valorization of hydrothermal carbonization products by anaerobic digestion: Inhibitor identification, biomethanization potential and process intensification

Integrating hydrothermal carbonization (HTC) and anaerobic digestion for biorefinery-oriented full utilization of wet organic wastes is a promising emerging technology. The objectives of this study were to identify the potential inhibitory substances, evaluate the biomethane potential of mixed and aqueous products and explore process intensifying strategies. The results indicated that the high HTC temperature of 240 °C resulted in a significantly low methane yield of 60 ± 5 mL/g COD and a high Short chain fatty acid (SCFAs) accumulation of 4174 ± 76 mg/L. GC-MS analysis showed that the contents of inhibitory pyrazines, pyridines and ketones in aqueous fraction at 240 °C substantially increased from 13.14%, 0.4%, 0.55% at 180 °C to 23.34%, 2.89%, 5.13%, respectively. When the aqueous products obtained from 240 °C-HTC was supplemented or pretreated by carbonaceous material, the methane yields were greatly improved and increased to 1.3-fold and 1.8-fold, respectively. These finding could provide some valuable technical information for HTC based biorefinery of organic waste.
