Effect of 3D printing and traditional molding on phenolic compounds and antioxidant activity in steamed bread

The production process of fermented black wheat steamed bread is closely related to the overall quality and nutritional content. In this study, we investigated the accuracy, product texture profile and antioxidant activity of fermented black wheat steamed bread samples produced by piston and spiral three-dimensional (3D) printers. The steaming process generally increased the total phenolic content and flavonoid content of the samples. The spiral 3D printer obtained samples with higher accuracy, total phenolic content up to 1960.43 Mg GAE/kg, and higher cellular antioxidant activity (CAA) content. The samples printed by the piston 3D printer showed higher total flavonoid content (575.75 Mg QE/kg), 2, 2′-azobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) values and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) values. This study shows that antioxidant-rich health foods can be prepared using 3D printed black wheat flour. The choice of 3D printing method affects the overall quality and nutritional content of the final product.

Supercritical antisolvent precipitation as a green technology to fractionate an Origanum majorana extract: Relationship between fractions cellular antioxidant activity and phytochemical composition.

The aim of this work was to determine the optimal conditions to fractionate a marjoram extract using supercritical antisolvent precipitation (SAS) technology in order to improve its antioxidant activity. This activity was evaluated using a cellular antioxidant assay (CAA) and compared to a chemical method (ABTS). After extract fractionation, most of the phenolics were recovered in the precipitate, except for the less polar ones that were found in the separators, along with essential oil components, non-volatile terpenes and fatty acids. Precipitate fractions obtained at 40 °C and 20–25 MPa showed the best TEAC values (1.7 mmol Trolox/g extract). Nevertheless, no precipitate fraction exerted better CAA than the unfractionated extract. This activity was related to the presence of arbutin and rosmarinic acid in their composition. In contrast, separators showed a low TEAC value (0.3 mmol Trolox/g), but a higher cellular antioxidant activity, especially fractions obtained at 15–20 MPa and 40–50 °C. This enhanced antioxidant activity was mainly attributed to the presence of ursolic acid, sterubin, menthol, sabinene hydrate and terpineol, compounds with a less polar character that therefore could cross more easily the cell membrane and exert their antioxidant activity inside the cells. Thus, separator fractions obtained at 20 MPa and 40 °C increased a 30% the CAA value respect to UAE extract and could be used for designing nutraceuticals or functional foods with cellular antioxidant activity. Precipitate fractions, at these extraction conditions, also showed an increase in TEAC value (40%) and could be employed as natural antioxidants in food formulations.

Murici (Byrsonima crassifolia (L.) Kunth and verbascifolia (L.)) and Tapereba (Spondias mombin) Improve Hepatic and Inflammatory Biomarkers in High-Fat-Diet Rats.

The present study investigated the effects of murici and tapereba on improving hepatic and inflammatory biomarkers in high-fat-diet rats. Female Wistar rats were divided into five groups (n = 10/group): control (CON), high-fat diet (HF), murici drink + high-fat diet (Mu-HF), tapereba drink + high-fat diet (Tap-HF), and murici and tapereba blend drink + high-fat diet (MT-HF). Drinks were offered daily for 60 days, following which body and liver weights, hepatosomatic indexes, serum parameters, inflammatory profile, and antioxidant activity (DPPH and ORAC) were analyzed. The cell death of hepatic cells was evaluated using flow cytometry. It was observed that weight gain was similar among the groups, while glycemia was lower in the MT-HF group. A high-fat diet increased the concentration of cholesterol total, ALT, IL-1β (in plasma and liver), and TNF-α (in the liver), and this was reduced by treatment with the fruit-based beverages. The other evaluated parameters showed no statistically significant difference. Compared to the CON and HF groups, the groups that received the drinks had higher cellular antioxidant activity and reduced oxidative stress, lipid oxidation, and development of pro-inflammatory cytokines, such as IL-1β. A high-fat diet induced higher cell death in hepatic tissue, which was prevented by the murici, tapereba, and the fruit-blend drinks. The consumption of murici, tapereba, and fruit-blend-based beverages showed beneficial effects on liver metabolism; therefore, they may serve as a nutritional approach for preventing and treating non-alcoholic liver disease.

Purification, characterization, and cellular antioxidant activity of 4,4‐dimethylsterols and 4‐desmethylsterols

AbstractA mixture of 4,4‐dimethylsterols (TA) and 4‐desmethylsterols (PS) was produced as valuable byproducts during the large‐scale production of ferulic acid by γ‐oryzanol. However, the utilization of both byproducts is impeded by this mixed form. This study has developed a novel method for separating TA and PS through a one‐step separation procedure using flash chromatography. The optimal conditions were as follows: mobile phase of hexane‐ethyl acetate (9:1, vol/vol), the flow rate of 40 ml/min, loading volume of 20 ml, run time of 20 min. Under optimized conditions, TA with a yield of 41.8% ± 2.8% with a purity of 93.4% ± 0.4% was obtained, and 36.8% ± 1.7% and 98.2% ± 0.8% for that of PS. Their chemical structures were further confirmed by ultraviolet spectroscopy (UV), Fourier‐transform infrared spectroscopy (FT‐IR), and gas chromatography–mass spectrometry (GC–MS). The time‐fluorescence value curve indicated that TA and PS had inhibitory effects on peroxyl radical‐induced 2′,7′‐dichlorofluorescin (DCFH) oxidation. The results showed that TA had higher cellular antioxidant activity (CAA) than PS. TA was more suitable as a potential natural antioxidant. The present study provides a convenient method for high‐purity TA and PS production, which will improve the value‐added byproducts from the large‐scale production of ferulic acid.

Interfacial engineering strategy to improve the stabilizing effect of curcumin-loaded nanostructured lipid carriers

In this work, the interfacial structure of nanostructured lipid carriers (NLCs) stabilized by whey protein isolate (single layer NLCs, S-NLCs) was modified with polysaccharide through two approaches: 1) electrostatic deposition of a polysaccharide layer onto the surface of S-NLCs affording double layer NLCs (D-NLCs); 2) heating the protein-polysaccharide complexes to promote biopolymer assembly, which were utilized as emulsifiers for the composite layer NLCs (C-NLCs). S-NLCs presented a spherical shape, hydrodynamic diameter of about 300 nm and curcumin (Cur) encapsulation efficiency below 30%. However, these values changed following interfacial decoration of NLCs. The interfacial viscoelastic studies suggested that the type 2 strategy enabled a more compact and elastic stabilizing layer than the type 1 method, as the interfacial biopolymers of the former were held together by multiple interactions. Therefore, C-NLCs showed higher capacity to inhibit polymorphic transition or degradation of Cur than other NLCs. Both approaches could slow down the lipolysis rate as well as decrease the lipolysis degree of NLCs during simulated digestion, while increase the bioavailability of the entrapped Cur. Besides, the highest cellular antioxidant activity was observed for Cur in C-NLCs, followed by D-NLCs and S-NLCs, in accordance to the stabilizing effects of these NLCs for the loaded agent.

Enhanced Lignanamide Absorption and Antioxidative Effect of Extruded Hempseed (Cannabis sativa L.) Hull in Caco-2 Intestinal Cell Culture.

Despite the latest pursuit to discover novel phenolic compounds in hempseed and its biological properties, it remains a mystery whether they can be absorbed in the human body. Extrusion treatment and extraction of the free phenolic fraction significantly (p < 0.05) improved human Caco-2 intestinal cell absorption for hempseed hull lignanamides including cannabisin B (Papp value of 1.35 × 10-5 ± 1.0 × 10-6) as compared to the bound fraction of raw hull (Papp value of 2.82 × 10-6 ± 5.2 × 10-7). Co-supplementation of the flavonoid naringenin (20 μM) further improved these absorption rates. Higher cellular antioxidant activity was observed in the free extraction fractions. Treatment with the free phenolic fraction of extruded hempseed hull (100 mg/mL) alleviated tert-butyl hydroperoxide's (25 μM) negative effects on cell viability, intracellular malondialdehyde levels, apoptosis induction, glutathione S-transferase, and glutathione levels. RNA-sequencing with the limma method unveiled a total of 2795 differentially expressed genes in 21 day-old Caco-2 intestinal cells, suggesting the changes in cell metabolism after exposure to extruded hempseed hull extract. This study could promote the utilization of extrusion technology to improve the absorption and antioxidant capacities of bioactive phenolics in plant food processing byproducts.

Effect of enzymatic digestion, chemical and boiled water extraction techniques on apparent antioxidant bioactivities of apple peel

Apple peel, is rich in bioactive phytochemicals with a wide of range of biological activities and health benefits. Phenolics, antioxidant and anti-proliferative properties of apple peel were evaluated using a simulated in vitro digestion and compared with a chemical extraction method to explore how the nature products by digestion to obtain a magnitude of antioxidant benefits. Digested samples were subjected to anti-proliferative evaluation against human liver cancer cells. Furthermore, antioxidant of digested apple peel was evaluated via the cellular antioxidant activity (CAA) method. Results revealed that the phenolic compounds obitained from enzymatic digestion were different from the chemical extracts. The contents of some component of phenolic acids and flavonoids (such as Q-3-G, Phloridzin, coumaric acid) in the digestion method were higher than other methods, which was contributed to higher in vitro antioxidant activity than other methods. Apple peel treated with cellulose-digestion also showed the highest CAA (No PBS wash) value. This finding indicated that enzyme-digestion of phytochemical rich materials could provide additional health benefits from natural products and would be a healthy strategy for food processing.

Ethanol Extracts of Coleus tuberosusFlesh and Peel as a Potential Source of Natural Antioxidant and Breast Cancer Prevention Agent

This study examines the effects of ethanol extracts of Coleus tuberosus flesh and peel on T47D cancer cells. Antioxidant potential was evaluated through cellular antioxidant activity experiment, and anti-proliferation activity was evaluated using MTT assay (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide). The effects on cell cycle were evaluated with a flow cytometry, while induction of apoptosis was evaluated based on morphological changes by staining with acridine orange and ethidium bromide. The results indicate that the peel extract had higher cellular antioxidant activity than the flesh extract. The IC50 of celullar antioxidant activity of the flesh and peel extracts were 287.13±10.35 g/ml and 217.86±12.96 g/ml, respectively. The peel extract also had higher anti-proliferative activity. The IC50 of anti-proliferative activity were 887.05±5.03 µg/ml (flesh extract) and 548.18±4.52 µg/ml (peel extract). The peel and flesh extracts can cause cell cycle arrest in the S phase and G2-M phase. The extracts induce apoptosis within T47D cancer cells, showing an orange color. Therefore, these extracts could be used as potential sources of natural antioxidants and breast cancer prevention agents.

Heat treatment improves the immunomodulatory and cellular antioxidant behavior of a natural flavanone: Eriodictyol

Plants and natural molecules are generally consumed not in raw state but after different processing conditions (heating, mechanical agitation or cooking). The understanding of the chemistry and biological outcome of thermal treatment is still scarce. In the current study, Eriodictyol, a natural flavanone, has undergone heat treatment, generating hence three different products ((3-(3,4-dihydroxyphenyl)-3-hydroxypropanoic acid, (3-(3,4-dihydroxyphenyl) propanal) and an unidentified component).The consequences of aforementioned treatment on the immunomodulatory behavior of resulted molecules were evaluated. The amount of nitric oxide production and the lysosomal enzyme activity were determined in vitro on mouse peritoneal macrophages. The kinetic of cellular antioxidant activity in splenocytes and macrophages was measured. The present investigation demonstrates that heat-processed eriodictyol significantly enhanced the proliferation of lymphocytes B and T compared to native eriodictyol. Indeed, this compound showed an important improvement on cytotoxic T lymphocyte (CTL) and natural killer (NK) activities. In addition, the production of nitric oxide (NO) and suppression of phagocytic activity of activated macrophages have been increasingly important after thermal processing. Furthermore, it was also revealed that heat-treated Erio in comparison with the native (non heat-treated) molecule has a highest cellular anti-oxidant activity in splenocytes and macrophages cells. These findings highlight the importance of heat-process as feasible and effective strategy to improve the immunomodulatory and the antioxidant efficiency of an known flavanone Eriodictyol.

Antioxidant and antiproliferative activities of proanthocyanidins from Chinese bayberry (Myrica rubra Sieb. et Zucc.) leaves

Chinese bayberry leaves proanthocyanidin (BLPs) contain epigallocatechin-3-O- gallate (EGCG) as its terminal unit and most of its extension units, which is unusual in the plant kingdom. The present study compared the antioxidant and anti-proliferative activities of BLPs prepared from different purification stages. Low-molecular-weight BLPs were prepared by ultrasonic treatment. Results revealed that the antioxidant activities of BLPs were strongly related to the content of total phenolics and proanthocyanidins; however, the cellular assay indicated that the mean degree of polymerization (mDP) played a vital role in the cellular antioxidant activity (CAA) of BLPs. The ultrasonic depolymerized BLPs with lower mDP (3.6 ± 0.3) exhibited a much higher CAA (EC50 = 0.042 ± 0.005 mg/mL) than other groups, which suggests that bioavailability is crucial to the cellular antioxidant potential of BLPs. Additionally, BLPs strongly inhibited HepG2 proliferation in a dose-dependent manner. Our results indicated that BLPs could be a valuable resource of bioactive compounds to promote consumer health.

Antioxidant Activity and α-Glucosidase Inhibitory Activities of the Polycondensate of Catechin with Glyoxylic Acid.

In order to investigate polymeric flavonoids, the polycondensate of catechin with glyoxylic acid (PCG) was prepared and its chemically antioxidant, cellular antioxidant (CAA) and α-glucosidase inhibitory activities were evaluated. The DPPH and ABTS radical scavenging activities and antiproliferative effect of PCG were lower than those of catechin, while PCG had higher CAA activity than catechin. In addition, PCG had very high α-glucosidase inhibitory activities (IC50 value, 2.59 μg/mL) in comparison to catechin (IC50 value, 239.27 μg/mL). Inhibition kinetics suggested that both PCG and catechin demonstrated a mixture of noncompetitive and anticompetitive inhibition. The enhanced CAA and α-glucosidase inhibitor activities of PCG could be due to catechin polymerization enhancing the binding capacity to the cellular membrane and enzymes.

Evaluation of Cellular Antioxidant and Antiproliferative Activities of Five Main Phyllanthus Emblica L. Cultivars in China.

The cell-based antioxidant activity assay as more biological relevant assay was considered to be more accurate to predict antioxidant activity in vivo than chemical activity assays. In the present study, the five main Phyllanthus emblica L. cultivars in China were subjected for cellular antioxidant activity based on HepG2 cells as well as antiproliferative activity. Total phenolics, total flavonoids and oxygen radical absorbance capacity were also measured. The results showed that Qingyougan, Binggan and Boligan (832±100, 774±52 and 704±28 μmol of quercetin equivalents/100 g) had higher cellular antioxidant activity than Tianyougan and Yougan (553±50 and 457±24 μmol of quercetin equivalents/100 g) in phosphate buffered saline wash protocol whereas, Boligan (3735±217 μmol of quercetin equivalents/100 g) had the highest cellular antioxidant activity and Tianyougan (2025±171 μmol of quercetin equivalents/100 g) had the lowest cellular antioxidant activity in no phosphate buffered saline wash protocol. The highest and lowest antiproliferative activities were observed in Binggan and Tianyougan (median effective dose: 6.95±0.11 and 14.03±0.10 mg/ml), respectively. The significant correlation was only observed between total flavonoids and cellular antioxidant activity from no phosphate buffered saline wash protocol (R(2) =0.908, P<0.05), and total flavonoids and antiproliferative activity (R(2) =0.887, P<0.05), suggesting the major contribution of flavonoids to the bioactivities of emblica. Overall, the data obtained revealed that different Phyllanthus emblica L. cultivars had strong cellular antioxidant and antiproliferative activities, thus should be recommended to increase consumption for health.

Phenolic Compounds and Antioxidant Capacities of 10 Common Edible Flowers from China

The free and bound phenolic compounds in 10 common Chinese edible flowers were investigated using reversed phase high-performance liquid chromatography. Their antioxidant capacities were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical-scavenging activity, oxygen radical absorption capacity (ORAC), ferric reducing antioxidant power (FRAP), and cellular antioxidant activity (CAA). Free factions were more prominent in phenolic content and antioxidant capacity than bound fractions. Paeonia suffruticosa and Flos lonicerae showed the highest total phenolic content (TPC) 235.5 mg chlorogenic acid equivalents/g of dry weight and total flavonoid content 89.38 mg rutin equivalents/g of dry weight. The major phenolic compounds identified were gallic acid, chlorogenic acid, and rutin. P. suffruticosa had the highest antioxidant capacity in the DPPH, ABTS, and ORAC assays, which were 1028, 2065, 990 μmol Trolox equivalents/g of dry weight, respectively, whereas Rosa chinensis had the highest FRAP value (2645 μmol Fe(2+) equivalents /g of dry weight). The P. suffruticosa soluble phenolics had the highest CAA, with the median effective dose (EC50 ) 26.7 and 153 μmol quercetin equivalents/100 g of dry weight in the phosphate buffered saline (PBS) and no PBS wash protocol, respectively. TPC was strongly correlated with antioxidant capacity (R = 0.8443 to 0.9978, P < 0.01), which indicated that phenolics were the major contributors to the antioxidant activity of the selected edible flowers.

Comparative Evaluation of the Antioxidant Potential of Infant Cereals Produced from Purple Wheat and Red Rice Grains and LC-MS Analysis of Their Anthocyanins

Cellular oxidative damage by endogenous and exogenous sources of free radicals and reactive oxygen species is a particular threat in infants. Antioxidant protection is normally achieved through a balance between pro-oxidants and endogenous and/or dietary antioxidants. Comprehensive research is required on optimization to achieve good antioxidant protection through infant foods, in particular, the commercially available infant cereals. This study therefore investigated the properties of whole purple wheat, unpolished red rice, and partially polished red rice before and after processing to produce infant cereals. Total phenolic content (TPC), total anthocyanin content (TAC), oxygen radical absorbance capacity (ORAC), individual anthocyanin components, and cellular antioxidant activity were measured. Home-made and laboratory-made pigmented infant cereals differed in that the latter required longer exposure to higher temperature and enzymatic hydrolysis. Home-made and laboratory-made unpolished red rice infant cereals showed higher total phenolic contents and peroxyl radical scavenging activity than home-made and laboratory-made purple wheat infant cereals; however, the latter had higher TAC. Pigmented infant cereals generally had higher TPC, TAC, and ORAC than the commercial ones (p < 0.05). Anthocyanins were identified in whole purple wheat, but they were not detected in unpolished red rice. C-Glycosyl apigenin was found in both whole purple wheat and unpolished red rice. Processing significantly decreased anthocyanin and C-glycosyl apigenin contents (p < 0.05). Purple wheat infant cereals had higher cellular antioxidant activity than unpolished red rice ones (p < 0.05). Whole purple wheat infant cereals showed higher antioxidant activity than the commercial infant cereal, suggesting a possibility of improving infant antioxidant status by incorporating this grain in their diet.
