Multiresidue analysis of 200 pesticides in jasmine (Jasminum sambac (L.) Aiton) flowers

A multiresidue analysis method was established for 200 pesticides in jasmine (Jasminum sambac(L.) Aiton) flowers based on QuEChERS procedure and gas chromatography-tandem mass spectrometry (GC–MS/MS). For better recoveries and purification effects, extraction solvents, extractive salts and purification methods were optimized. The limits of quantification (LOQ) of test pesticides are in the range of 0.01 and 0.1 mg kg−1, most of which were below the maximum residue limits (MRLs) prescribed by EU and China. Recoveries at 0.01, 0.1, 1.0 mg kg−1 were predominantly in the range of 70–120 % (n = 3) with corresponding RSDs lower than 20 %, which suggests satisfactory accuracy. The multiresidue analysis method was applied in monitoring pesticide residues in 8 jasmine flower samples. 50 % of those samples present with 1–2 pesticides residues. This study provides a sensitive and high-throughput method for detection of multi-pesticide residues in jasmine flowers.

Optimization of Plant Oxalate Quantification and Generation of Low-Oxalate Maize (Zea mays L.) through O7 Overexpression.

Oxalate, the simplest dicarboxylic acid, is a prevalent antinutrient that chelates with various metals and can lead to the formation of kidney stones in humans. The accurate detection of the oxalate concentration in food and the cultivation of low-oxalate crops are important for enhancing public health. In this study, we established a high-throughput and highly sensitive technique for oxalate detection using ultra-high-performance liquid chromatographic-triple quadrupole tandem mass spectrometry (UPLC-QqQ-MS/MS). Additionally, we overexpressed the gene O7, which encodes oxalyl-CoA synthetase in the maize oxalate degradation pathway, resulting in O7-OE lines. By employing the UPLC-QqQ-MS/MS method to measure oxalate levels in these transgenic lines, we observed that the oxalate content in the kernels of O7-OE lines was reduced by approximately 43%, with a concurrent increase in some micronutrients such as zinc. Importantly, the transgenic maize showed normal seed storage compound accumulation or other agronomic characteristics. In summary, we developed a high-throughput detection method that advances oxalate measurement. Furthermore, by generating new maize germplasm with diminished oxalate, our work offers potential health advantages to consumers.

Simultaneous Detection of Clenbuterol and Higenamine in Urine Samples Using Interference-Free SERS Tags Combined with Magnetic Separation.

Sports doping remains a significant challenge in competitive sports. Given that urine analysis is the standard for detecting doping, developing rapid, sensitive, accurate, and high-throughput methods for stimulant detection in urine is crucial. Surface-enhanced Raman scattering (SERS) tag-based immunoassays have emerged as powerful analytical tools known for their high sensitivity and specificity, holding particular promise for stimulant detection in urine samples. However, both the Raman signals of typical SERS tags and sample matrices are within the Raman fingerprint region (<1800 cm-1), which could lead to spectrum overlap, potentially reducing detection accuracy and sensitivity. By recognizing this, we designed a competitive immunoassay that integrates two types of zero-background SERS tags and magnetic separation. These innovative SERS tags exhibit distinctive Raman peaks within the Raman-silent region (1800-2800 cm-1), effectively mitigating potential spectrum overlap with background sample signals. Moreover, magnetic separation not only enhances operational simplicity but also improves the system's anti-interference capability. Using clenbuterol (CL) and higenamine (HM) as model targets, the SERS-based competitive immunoassay demonstrated sensitive detection of individual CL or HM standards, with limits of detection (LODs) of 0.87 and 0.71 pg/mL, respectively. In multiplex mode, CL and HM can be simultaneously detected with LODs of 1.0 and 0.81 pg/mL, respectively. Furthermore, the recovery rates in urine samples ranged from 83 to 116% (relative standard deviation, RSD ≤ 6.4%) for CL and from 82 to 103% (RSD ≤ 5.1%) for HM, further confirming the reliability of the SERS-based immunoassay for practical applications.

A rapid, sensitive, and high-throughput pathogen detection method and application of identifying pathogens based on multiplex PCR technology combined with capillary electrophoresis

Respiratory infections are one of the leading causes of death worldwide. Rapid and accurate detection of pathogens is crucial for timely treatment. This study established a detection method based on multiplex PCR combined with capillary electrophoresis, capable of simultaneously detecting 28 common respiratory pathogens. We used specially designed homo-tag primers, significantly reducing the formation of primer dimers and improving the specificity and sensitivity of amplification. After two rounds of PCR amplification, the products were subjected to fragment analysis using the ABI 3500XL Genetic Analyzer, enabling automated result interpretation. The detection limit of this method reaches 2.77 × 102 copies/ml, with some pathogens reaching as low as 2.77 × 10 copies/ml. The detection of 147 clinical specimens showed that the overall consistency of this method with culture, colloidal gold, fluorescent quantitative PCR, and NGS was 75.5%. The multiplex PCR detection method established in this study is rapid, sensitive, and high-throughput, can be used for routine screening of common pathogens in respiratory infections, providing an important basis for clinical diagnosis and treatment.

PhIP-Seq: methods, applications and challenges.

Phage-immunoprecipitation sequencing (PhIP-Seq) technology is an innovative, high-throughput antibody detection method. It enables comprehensive analysis of individual antibody profiles. This technology shows great potential, particularly in exploring disease mechanisms and immune responses. Currently, PhIP-Seq has been successfully applied in various fields, such as the exploration of biomarkers for autoimmune diseases, vaccine development, and allergen detection. A variety of bioinformatics tools have facilitated the development of this process. However, PhIP-Seq technology still faces many challenges and has room for improvement. Here, we review the methods, applications, and challenges of PhIP-Seq and discuss its future directions in immunological research and clinical applications. With continuous progress and optimization, PhIP-Seq is expected to play an even more important role in future biomedical research, providing new ideas and methods for disease prevention, diagnosis, and treatment.

Comparison of serum cytokines and chemokines levels and clinical significance in patients with pemphigus vulgaris-A retrospective study.

In this study, we aimed to examine the relationship between the serum cytokine levels of patients with pemphigus vulgaris (PV) and the Pemphigus Disease Area Index (PDAI), along with the presence of anti-desmoglein (Dsg) 1 antibody, anti-Dsg3 antibody and co-infection among patients with pemphigus vulgaris. This retrospective study included 62 PV patients and 59 healthy individuals who attended the Second Affiliated Hospital of Kunming Medical University from November 2014 to November 2022. The serum concentrations of cytokines and chemokines were assessed using the Luminex 200 System (a high-throughput cytokine detection method). Additionally, anti-Dsg1 and anti-Dsg3 antibodies were determined through enzyme-linked immunosorbent assay, while disease severity was evaluated using the PDAI scoring system. The PV group exhibited elevated levels of Th1 cytokines (such as interleukin (IL)-1RA, IL-1β, IL-2, IL-12p70, GM-CSF, TNF-α, IL-18, IFN-γ), Th2 cytokines (IL-5, IL-10, IL-13) and Th17/Th22-related cytokines (IL-17A, IL-22) compared to the healthy control group (p < 0.05). Conversely, the levels of chemokines (macrophage inflammatory protein-1 alpha (MIP-1α), stromal cell-derived factor-1 alpha (SDF-1α), interferon-inducible protein-10 (IP-10), Regulated on Activation in Normal T-Cell Expressed And Secreted (RANTES), growth-regulated on-gene-alpha (GRO-α), MIP-1β) and Th2 (IL-31) were lower in the PV group compared to the healthy control group (p < 0.05). No significant differences were observed in other cytokines and chemokines (p > 0.05). Additionally, IL-7, IFN-γ, IL-18 and GRO-α showed positive correlations with PDAI, IL-6 correlated positively with anti-Dsg3 antibody levels, and IL-12p70, IL-18, and IFN-γ correlated positively with anti-Dsg1 antibody levels. Furthermore, IL-15 exhibited a positive association with skin infections. PV patients have elevated levels of various cytokines and chemokines, and there are different degrees of elevation in cytokines and chemokines associated with the activation of various T cell subsets. PDAI and the Dsg1 antibody levels are mainly related to the Th1-related cytokines.

Evaluating cell growth and hypoxic regions of 3D spheroids via a machine learning approach

Abstract This study investigated the applicability of the area of spheroids and hypoxic regions for efficient evaluation of drug efficacy using machine learning (ML). We initially developed a high-throughput detection method to obtain the area of spheroids and hypoxic regions that can handle over 10 000 images per hour with an error rate of 2%–3%. The ML models were trained using cell growth of six cell lines (i.e. HepG2, A549, Hep3B, BEAS-2B, HT-29, and HCT116) and hypoxic region variations of two cell lines (i.e. HepG2 and BEAS-2B); our model can predict the area of spheroids and hypoxic region of certain growth date with high precision. To demonstrate the applicability, HepG2 spheroids were treated with sorafenib, and the efficacy of the drug was evaluated through a comparison of differences in areas of cell size and hypoxic regions with the predicted results. Furthermore, our ML approach has been shown to be applicable to provide the model-driven evaluative criterion for toxicity and drug efficacy using spheroids.

Identification of horse ingredients based on recombinase polymerase amplification, high-throughput DNA isolation and magnetic separation

Adulteration of meat products remains a significant issue worldwide. In this study, we developed a high-throughput, rapid, sensitive, and selective method for qualitative and quantitative detection of horse meat adulteration in meat products. This method, which is not only innovative but also highly practical, involves coupling a 96-well high-throughput DNA isolation process with recombinant enzyme polymerase amplification (RPA), magnetic separation (MS), and high-throughput fluorescence detection (FD). We designed a set of primers targeting the mitochondrial ATP6-8 gene of horses. The upper and lower primers were labeled with 6-FAM fluorescent moiety and biotin, respectively. Following RPA amplification, the amplicons were separated and purified using streptavidin-modified magnetic beads (MB@SA). Subsequently, the amplicons were qualified and quantified using a high-throughput fluorescence detection system. We optimized the conditions for primer addition, incubation time, and the ratio of amplicon to MB@SA. Our results showed that a fluorescence threshold of 834 indicated positive detection. The method exhibited high specificity, with no positive amplification observed in DNA from the other 11 types of meat. It demonstrated high sensitivity and was capable of detecting as little as 0.1% (wt%) of horse meat in adulterated samples. Combined with our developed 96-well high-throughput DNA extraction system, the RPA-MS-FD assay process could analyze 96 samples in 31.5 min, including 11.5 min for rapid DNA extraction, 15 min for RPA amplification, and 5 min for fluorescence detection. Finally, we successfully applied this assay to analyze 21 commercial meat products, proving its practicality and effectiveness.

Novel selection of recycled PET surrogates based on non-targeted screening of non-intentionally added substances and chemometrics

Waste PET is utilized to produce recycled PET (rPET) through bottle-to-bottle recycling in order to reduce environmental pollution. However, non-intentionally added substances (NIAS) may be generated during rPET production, potentially posing a threat to food safety. In this study, a non-target screening of 13 batches with a total of 39 rPET samples was performed using GC-QTOF-MS and 240 compounds were tentatively identified. It was worth noting that the detection of phthalate esters in rPET from China, such as bis(2-ethylhexyl) phthalate, exhibits a higher diversity (17 different types) and frequency (7.8 %–100 %) in comparison with related studies. Subsequently, based on the principles for selection of surrogates used in challenge test, seven molecular descriptors were selected as variables for compound characterization, followed by principal component analysis (PCA) for representation. Based on this analysis and referencing the EFSA and FDA surrogate suggestion lists, eight surrogates were proposed for China, namely chlorobenzene, toluene, benzophenone, bis(2-ethylhexyl) terephthalate, methyl salicylate, methyl stearate, 1-phenyldecane and phenylcyclohexane. High-throughput detection methods were developed for each surrogate with the regression correlation coefficients over 0.999, spiked recoveries ranging from 99.3-107.5 %, LODs of 0.003–2.192 ng/mL, LOQs of 0.009–7.306 ng/mL, RSD of 0.71–4.23 %, respectively. This research provides foundational data and methodological support for the evaluation of rPET challenge testing in China, offering technical support for subsequent safety assessments of rPET.

RNA-binding motif protein 28 enhances angiogenesis by improving STAT3 translation in hepatocellular carcinoma

Hepatocellular carcinoma (HCC) is a prevalent malignant tumor characterized by extensive angiogenesis. However, the underlying mechanisms of HCC pathogenesis remain unclear. Previous studies have shown that RNA-binding proteins (RBPs) are implicated in HCC pathogenesis. In this study, we observed that increased RBM28 expression in HCC tissues was positively correlated with tumor microvascular density and negatively correlated with patient prognosis. Overexpression of RBM28 in HCC cells promoted tubule formation in human umbilical vein endothelial cells, whereas inhibition of RBM28 had the opposite effect, furthermore, the role of RBM28 in the progression of HCC was assessed using transgenic mouse models and chemically induced HCC models. We used various molecular assays and high-throughput detection methods to evaluate the role of RBM28 in promoting angiogenesis in HCC. Increased RBM28 expression in HCC directly binds to STAT3 mRNA, recruiting EIF4E to increase STAT3 expression and enhancing the secretion and expression of vascular endothelial growth factor A; consequently, promoting neovascularization in HCC. The potential of RBM28 as a viable diagnostic and therapeutic target for HCC was assessed using multi-cohort clinical samples and animal models. In summary, our results provide insights into the pathogenesis, clinical diagnosis, and treatment of HCC.

A triple-channel sensor array utilizing fluorescent carbon dots for simultaneous discrimination and detection of multiple fluoroquinolones

The presence of fluoroquinolones (FQs) residues in food and the environment has prompted concerns regarding food safety and public health. Consequently, it is of great significance to analyze the types and levels of FQs present. However, the majority of studies have concentrated on the specific detection of individual FQs, with a notable absence of high-throughput and rapid analysis methods for the simultaneous detection of multiple FQs that may coexist in food and the environment. Hereon, a triple-channel sensor array was successfully constructed utilizing fluorescent carbon dots (TA-CDs), with the assistance of Cu2+ and Fe3+, for the qualitative discrimination and quantitative detection of eight types of FQs. The sensor array can distinguish between different concentrations of FQs and various mixtures of FQs, as well as 100 % accuracy in the discrimination of unknown samples. Impressively, the sensor platform can quantitatively detect FQs in animal-derived foods, such as honey, milk, eggs, and pork, as well as in water samples. This research has the potential to be extended to other analytes with similar chemical structures or properties.

Sensitive and High-Throughput Time-Resolved Luminescence Detection of Tetracycline in Milk for Eliminating Background Fluorescence on a Miniaturized Apparatus.

Fluorescence detection has always suffered from high background fluorescence from real samples such as milk. Therefore, cumbersome pretreatments of samples were necessary to remove the fluorescent substances but led to long processing times and low efficiency. Time-resolved luminescence detection is a powerful technique for eliminating short-lived background fluorescence without additional pretreatments. However, the related instruments are usually equipped with high-speed excitation sources and detectors, which are always bulky and expensive. Herein, we developed a low-cost and miniaturized imaging system for high-throughput time-gated luminescence detection. An UV LED array was used to excite multiple samples, the luminescence of which could be detected by a smartphone simultaneously. An analog circuit was designed to synchronize the LED to the mechanical chopper to eliminate the background signals resulting from scattering and short-lived autofluorescence. Compared to other synchronous circuits based on FPGAs and microcontrollers, this analog circuit required no programming and memory. For the first time, high-throughput time-resolved luminescence detection of tetracycline in milk without any separation or enrichment was achieved by utilizing a smartphone as a camera, and the scattered signals and the background fluorescence were eliminated efficiently. The limit of detection reached as low as 53 nM (∼0.024 ppm), lower than the residue limit set by the European Union. This high-throughput time-gated luminescence detection method can be used for quantitative analysis of many real samples with high background fluorescence.

MassARRAY: a high-throughput solution for rapid detection of foodborne pathogens in real-world settings.

Bacterial foodborne pathogens pose a substantial global public health concern, prompting government agencies and public health organizations to establish food safety guidelines and regulations aimed at mitigating the risk of foodborne illness. The advent of DNA-based amplification coupled with mass spectrometry, known as MassARRAY analysis, has proven to be a highly precise, sensitive, high-throughput, and cost-effective method for bacterial detection. This study aimed to develop, validate, and evaluate a MassARRAY-based assay for the detection and identification of significant enteropathogenic bacteria. The MassARRAY-based assay was developed for the detection of 10 crucial bacterial foodborne pathogens, including Campylobacter coli, Campylobacter jejuni, Clostridium perfringens, Escherichia coli, Enterococcus faecalis, Enterococcus faecium, Listeria monocytogenes, Salmonella spp., Shigella spp., and Staphylococcus aureus. The assay was optimized using the reference gDNA (n = 19), followed by validation using gDNA (n = 85) of reference and laboratory isolates. Additionally, the evaluation of the assay's reaction using a mixture of gDNA from all nine targeted species was performed. The limit of detection of the developed MassARRAY-based assay was determined using bacterial cells. Moreover, the validation method for field samples was evaluated by comparing it with standard microbiological testing methods routinely analyzed. The developed MassARRAY-based assay demonstrated 100% concordance with known bacterial pure cultures. The assay's reaction using a mixture of gDNA from all nine targeted species revealed the MassARRAY's capability to detect all targeted species in a single assay with the lowest concentration of 1 ng/μL of gDNA. The limits of detection of the assay range from 357 ± 101 to 282,000 ± 79,196 cells. Moreover, the validation of the assay in field samples revealed a 100% correlation between the data obtained from the standard microbiological method and the MassARRAY-based assay. These findings suggested that the developed MassARRAY-based assay exhibited the excellence in high-throughput detection of foodborne bacterial pathogens with high accuracy, reliability, and potential applicability within real-world field samples.

Development and validation of an ELISA kit for the detection of Staphylococcus aureus enterotoxin A, B, C1, C2, C3, D, E from food samples

Staphylococcus aureus (S. aureus) is a significant food-borne pathogen that poses a threat to human health. The classical serotypes staphylococcal enterotoxin (SE) A, B, C1, C2, C3, D, E produced by S. aureus are the primary cause of its food-borne pathogenic toxicity. Therefore, establishing a highly sensitive, rapid, robust, high-throughput detection method for the total amount of classical serotypes of SEs is crucial for food safety and public health. This study prepared 10 rabbit polyclonal antibodies and 40 mouse monoclonal antibodies targeting SEA, SEB, SEC1, SEC2, SEC3. Upon identifying the optimal antibody pairs through square array method and optimization, a sandwich ELISA kit for the detection of SEs (SEA, SEB, SEC1, SEC2, SEC3, SED, SEE) was developed, with respective limits of detection at 0.169–0.688 ng/mL. In addition, this sandwich ELISA kit could detect SEs in raw cow milk, pasteurized milk, fermented milk, milk powder, whey powder, rice, raw pork, and canned beef samples with coefficients of variation below 7%. Meanwhile, this sandwich ELISA kit was stable at 4 °C for one-year storage. The study fills a research gap in the detection of the total amount of classical serotypes of SEs.

A Dual and Rapid RPA-CRISPR/Cas12a Method for Simultaneous Detection of Cattle and Soybean-Derived Adulteration in Goat Milk Powder.

The adulteration of goat milk powder occurs frequently; cattle-derived and soybean-derived ingredients are common adulterants in goat milk powder. However, simultaneously and rapidly detecting cattle-derived and soybean-derived components is still a challenge. An efficient, high-throughput screening method for adulteration detection is needed. In this study, a rapid method was developed to detect the adulteration of common cattle-derived and soybean-derived components simultaneously in goat milk powder by combining the CRISPR/Cas12a system with recombinant polymerase amplification (RPA). A dual DNA extraction method was employed. Primers and crRNA for dual detection were designed and screened, and a series of condition optimizations were carried out in this experiment. The optimized assay rapidly detected cattle-derived and soybean-derived components in 40 min. The detection limits of both cattle-derived and soybean-derived components were 1% (w/w) for the mixed adulteration models. The established method was applied to a blind survey of 55 commercially available goat milk powder products. The results revealed that 36.36% of the samples contained cattle-derived or soybean-derived ingredients, which revealed the noticeable adulteration situation in the goat milk powder market. This study realized a fast flow of dual extraction, dual amplification, and dual detection of cattle-derived and soybean-derived components in goat milk powder for the first time. The method developed can be used for high-throughput and high-efficiency on-site primary screening of goat milk powder adulterants, and provides a technical reference for combating food adulteration.

High-Throughput Multitarget Molecular Detection in an Automatic Light-Addressable Photoelectrochemical Sensing Platform.

Successively emerged high-throughput multitarget molecular detection methods bring significant development tides in chemical, biological, and environmental fields. However, several persistent challenges of intricate sample preparation, expensive instruments, and tedious and skilled operations still need to be further addressed. Here, we propose an automatic light-addressable photoelectrochemical (ALA-PEC) sensing platform for sensitive and selective detection of multitarget molecules. With Au nanoparticle-decorated TiO2 nanotube photonic crystals (Au-TiO2 NTPCs) as a photoelectrode and 8 kinds of antibiotics as target molecules, the ALA-PEC sensing system implements automatic detection of multimolecules in a short time with high sensitivity and good selectivity. Random samples with different amounts of antibiotics have been well-distinguished in the ALA-PEC system, and both the chemical components and concentrations have been well-illustrated in a pattern recognition model. It is worth noting that 8 samples are not the limit of the ALA-PEC sensing platform, which can be easily expanded to more complex detection arrays based on practical needs. The emerging ALA-PEC sensing platform provides a new solution for rapid screening and detection of multitarget and high-throughput substances and potentially brings the automatic, portable, sensitive, high-throughput, and cost-effective detection technique to an entire new realm.

Structural Elucidation of Glycosaminoglycans in the Tissue of Flounder and Isolation of Chondroitin Sulfate C.

Glycosaminoglycans (GAGs) are valuable bioactive polysaccharides with promising biomedical and pharmaceutical applications. In this study, we analyzed GAGs using HPLC-MS/MS from the bone (B), muscle (M), skin (S), and viscera (V) of Scophthalmus maximus (SM), Paralichthysi (P), Limanda ferruginea (LF), Cleisthenes herzensteini (G), Platichthys bicoloratus (PB), Pleuronichthys cornutus (PC), and Cleisthenes herzensteini (CH). Unsaturated disaccharide products were obtained by enzymatic hydrolysis of the GAGs and subjected to compositional analysis of chondroitin sulfate (CS), heparin sulfate (HS), and hyaluronic acid (HA), including the sulfation degree of CS and HS, as well as the content of each GAG. The contents of GAGs in the tissues and the sulfation degree differed significantly among the fish. The bone of S. maximus contained more than 12 μg of CS per mg of dry tissue. Although the fish typically contained high levels of CSA (CS-4S), some fish bone tissue exhibited elevated levels of CSC (CS-6S). The HS content was found to range from 10-150 ug/g, primarily distributed in viscera, with a predominant non-sulfated structure (HS-0S). The structure of HA is well-defined without sulfation modification. These analytical results are independent of biological classification. We provide a high-throughput rapid detection method for tissue samples using HPLC-MS/MS to rapidly screen ideal sources of GAG. On this basis, four kinds of CS were prepared and purified from flounder bone, and their molecular weight was determined to be 23-28 kDa by HPGPC-MALLS, and the disaccharide component unit was dominated by CS-6S, which is a potential substitute for CSC derived from shark cartilage.

A high-throughput differential scanning fluorimetry method for rapid detection of thermal stability and iron saturation in lactoferrin

Thermal stability and iron saturation of lactoferrin (LF) are of great significance not only for the evaluation of the biological activities of LF but also for the optimization of the isolation and drying process parameters. Differential scanning calorimetry (DSC) is a well-established and efficient method for thermal stability and iron saturation detection in LF. However, multiple DSC measurements are typically performed sequentially, thus time-consuming and low throughput. Herein, we introduced the differential scanning fluorimetry (DSF) approach to overcome such limitations. The DSF can monitor LF thermal unfolding with a commonly available real-time PCR instrument and a fluorescent dye (SYPRO orange or Glomelt), and the measured melting temperature of LF is consistent with that determined by DSC. On the basis of that, a new quantification method was established for determination of iron saturation levels using the linear correlation of the degree of ion saturation of LF with DSF measurements. Such DSF method is simple, inexpensive, rapid (<15 min), and high throughput (>96 samples per experiment), and provides a valuable alternative tool for thermal stability detection of LF and other whey proteins.

Identification of Garlic Viruses Associated with Seed Bulbs and Consumption Bulbs from Several Locations in Indonesia

Virus infection is one of the major constraints in garlic production since the viruses are readily accumulated on vegetative propagation material (bulbs). This research aimed to detect garlic common latent virus (GCLV), shallot latent virus (SLV), onion yellow dwarf virus (OYDV), and leek yellow stripe virus (LYSV) infecting local garlic as seed bulb and imported garlic as consumption bulb. Seed bulb samples were obtained from seed breeders in several garlic growing centers in Indonesia. In contrast, consumption bulb samples were obtained from plant quarantine warehouses and three local markets in Bogor. Some bulb samples were used for morphological observations, and some were germinated in the laboratory until the leaves emerged. Leaves were collected for virus detection by RT-PCR using specific primers for GCLV, SLV, OYDV, and LYSV. Seed and consumption bulbs have differences in their morphological characteristics, especially in the type of neck hardness and the size of the bulb diameter. OYDV and LYSV infections were successfully detected in seed and consumption bulbs, while SLV was only found in consumption bulbs. Nucleotide sequence analysis showed that SLV from consumption bulbs formed one group, GCLV from seed bulbs formed one group, while OYDV and LYSV from seed and consumption bulbs were in different groups, indicating that the viruses came from different strains. Further research through high-throughput detection methods and providing virus-free planting material are needed to anticipate the spread of new strains of garlic viruses in Indonesia.
