Participatory Geomonitoring for Future Mining—Resilience Management in the Cavern Storage Epe (Germany)

Integrated geo- and environmental monitoring in mining represents a high-dimensional challenge (location, altitude/depth, time and sensors). This is challenging for experts but poses great problems for a multitude of participants and stakeholders in building up a complete process understanding. The Epe research cooperation aims to elucidate the ground movement at the Epe cavern storage facility with a public participation process. The research cooperation was founded by the city of Gronau, the citizens’ initiative cavern field Epe, the company EFTAS, Münster, and the Research Center of Post-Mining at the Technische Hochschule Georg Agricola, Bochum. This research cooperation is the first in Germany to involve direct collaboration between science and the public. In the cavern field, which has been in operation since the 1970s, brine is extracted, and at the same time natural gas, crude oil and helium, as well as hydrogen in the future, are stored in the subsurface. The technical focus of this work was the development of a high-resolution spatiotemporal analysis of ground movements. The area is monitored annually by the mining company’s mine surveyor. The complexity of the monitoring issue lies in the fact that the western part is a bog area and a former bog area. Furthermore, the soils in the eastern part are very humus-rich and show strong fluctuations in the groundwater and therefore complex hydraulic conditions. At the same time, there are few fixed scatterers or prominent points in the area that allow high-resolution spatiotemporal monitoring using simple radar interferometry methods. Therefore, the SBAS method (Small Baseline Subset), which is based on an aerial method, was used to analyze the radar interferometric datasets. Using an SBAS analysis, it was possible to evaluate a time series of 760 scenes over the period from 2015 to 2023. The results were integrated with the mine survey maps on the ground movement and other open geodata on the surface, the soil layers and the overburden. The results show complex forms of ground movement. The main influence is that of mining. Nevertheless, the influence of organic soils with drying out due to drought years and uplift in wet years is great. Thus, in dry years, ground subsidence accelerates, and in wet years, ground subsidence not only slows down but in some cases also causes uplift. This complexity of ground movements and the necessary understanding of the processes involved has been communicated to the interested public at several public information events as part of the research cooperation. In this way, an understanding of the mining process was built up, and transparency was created in the subsurface use, also as a part of the energy transition. In technical terms, the research cooperation also provides a workflow for developing the annual mine survey maps into an integrated geo- and environmental monitoring system with the development of a transparent participatory geomonitoring process to provide resilience management to a mining location.

Mortality from leading cancers in districts of England from 2002 to 2019: a population-based, spatiotemporal study

Cancers are the leading cause of death in England. We aimed to estimate trends in mortality from leading cancers from 2002 to 2019 for the 314 districts in England. We did a high-resolution spatiotemporal analysis of vital registration data from the UK Office for National Statistics using data on all deaths from the ten leading cancers in England from 2002 to 2019. We used a Bayesian hierarchical model to obtain robust estimates of age-specific and cause-specific death rates. We used life table methods to calculate the primary outcome, the unconditional probability of dying between birth and age 80 years by sex, cancer cause of death, local district, and year. We reported Spearman rank correlations between the probability of dying from a cancer and district-level poverty in 2019. In 2019, the probability of dying from a cancer before age 80 years ranged from 0·10 (95% credible interval [CrI] 0·10-0·11) to 0·17 (0·16-0·18) for women and from 0·12 (0·12-0·13) to 0·22 (0·21-0·23) for men. Variation in the probability of dying was largest for lung cancer among women, being 3·7 times (95% CrI 3·2-4·4) higher in the district with the highest probability than in the district with the lowest probability; and for stomach cancer for men, being 3·2 times (2·6-4·1) higher in the district with the highest probability than in the one with the lowest probability. The variation in the probability of dying was smallest across districts for lymphoma and multiple myeloma (95% CrI 1·2 times [1·1-1·4] higher in the district with the highest probability than the lowest probability for women and 1·2 times [1·0-1·4] for men), and leukaemia (1·1 times [1·0-1·4] for women and 1·2 times [1·0-1·5] for men). The Spearman rank correlation between probability of dying from a cancer and district poverty was 0·74 (95% CrI 0·72-0·76) for women and 0·79 (0·78-0·81) for men. From 2002 to 2019, the overall probability of dying from a cancer declined in all districts: the reductions ranged from 6·6% (95% CrI 0·3-13·1) to 30·1% (25·6-34·5) for women and from 12·8% (7·1-18·8) to 36·7% (32·2-41·2) for men. However, there were increases in mortality for liver cancer among men, lung cancer and corpus uteri cancer among women, and pancreatic cancer in both sexes in some or all districts with posterior probability greater than 0·80. Cancers with modifiable risk factors and potential for screening for precancerous lesions had heterogeneous trends and the greatest geographical inequality. To reduce these inequalities, factors affecting both incidence and survival need to be addressed at the local level. Pronounced Seasonal and Spatial Variability in Determinants of Phytoplankton Biomass Dynamics along a Near–Offshore Gradient in the Southern North Sea

Marine phytoplankton biomass dynamics are affected by eutrophication, ocean warming, and ocean acidification. These changing abiotic conditions may impact phytoplankton biomass and its spatiotemporal dynamics. In this study, we used a nutrient–phytoplankton–zooplankton (NPZ) model to quantify the relative importance of the bottom-up and top-down determinants of phytoplankton biomass dynamics in the Belgian part of the North Sea (BPNS). Using four years (2014–2017) of monthly observations of nutrients, solar irradiance, sea surface temperature, chlorophyll-a, and zooplankton biomass at ten locations, we disentangled the monthly, seasonal, and yearly variation in phytoplankton biomass dynamics. To quantify how the relative importance of determinants changed along a near–offshore gradient, the analysis was performed for three spatial regions, i.e., the nearshore region (<10 km to the coastline), the midshore region (10–30 km), and the offshore region (>30 km). We found that, from year 2014 to 2017, the phytoplankton biomass dynamics ranged from 1.4 to 23.1 mg Chla m−3. Phytoplankton biomass dynamics follow a general seasonal cycle, as is the case in other temperate regional seas, with a distinct spring bloom (5.3–23.1 mg Chla m−3) and a modest autumn bloom (2.9–5.4 mg Chla m−3). This classic bimodal bloom pattern was not observed between 2003 and 2010 in the BPNS. The seasonal pattern was most expressed in the nearshore region. The relative contribution of factors determining phytoplankton biomass dynamics varied spatially and temporally. Throughout a calendar year, solar irradiance and zooplankton grazing were the most influential determinants in all regions, i.e., they jointly explained 38–65% of the variation in the offshore region, 45–71% in the midshore region, and 56–77% in the nearshore region. In the near- and midshore regions, nutrients were the greatest limit on phytoplankton production in the month following the spring bloom (44–55%). Nutrients were a determinant throughout the year in the offshore region (27–62%). During winter, sea surface temperature was a determinant in all regions (15–17%). By the high-resolution spatiotemporal analysis of the relative contributions of different determinants, this study contributes to a better mechanistic understanding of the spatiotemporal dynamics of phytoplankton biomass in the southern North Sea. This detailed understanding is anticipated to contribute to the definition of targeted management strategies for the BPNS and to support sustainable development in Belgium’s blue economy.

High-resolution spatio-temporal analysis of snowmelt over Antarctic Peninsula ice shelves from 2015 to 2021 using SAR images

ABSTRACT Ice shelves play an essential role in the dynamics of the Antarctic ice sheet. The surface meltwater is important, as it can irreversibly weaken ice shelves by exerting additional hydrostatic pressure. Therefore, high-resolution snowmelt products are urgently needed to accurately analyze melting patterns of ice shelves and further estimate mass loss. In this study, a new high-resolution (40 m) snowmelt dataset over all of the Antarctic Peninsula ice shelves larger than 100 km was developed based on the modified snowmelt detection framework by using a co-orbit normalization method. The dataset provides detailed snowmelt information on each ice shelf, including the image coverage, melting area and melting ratio every 5 days. The melting patterns of three typical ice shelves (George VI, Wilkins and Larsen C Ice Shelves) and the spatio-temporal melting distribution of the Antarctic Peninsula (AP) were further analyzed. The snowmelt information indicates that both the extent and duration of snowmelt have been increasing in the Antarctic Peninsula from 2015 to 2021, and we found that the snowmelt on the Antarctic Peninsula showed a spatial pattern of significantly intense snowmelt on the western side. We believe this study will provide essential data for ice shelf investigation to support other fields of polar research.

High-resolution spatiotemporal analysis of single serotonergic axons in an in vitro system.

Vertebrate brains have a dual structure, composed of (i) axons that can be well-captured with graph-theoretical methods and (ii) axons that form a dense matrix in which neurons with precise connections operate. A core part of this matrix is formed by axons (fibers) that store and release 5-hydroxytryptamine (5-HT, serotonin), an ancient neurotransmitter that supports neuroplasticity and has profound implications for mental health. The self-organization of the serotonergic matrix is not well understood, despite recent advances in experimental and theoretical approaches. In particular, individual serotonergic axons produce highly stochastic trajectories, fundamental to the construction of regional fiber densities, but further advances in predictive computer simulations require more accurate experimental information. This study examined single serotonergic axons in culture systems (co-cultures and monolayers), by using a set of complementary high-resolution methods: confocal microscopy, holotomography (refractive index-based live imaging), and super-resolution (STED) microscopy. It shows that serotonergic axon walks in neural tissue may strongly reflect the stochastic geometry of this tissue and it also provides new insights into the morphology and branching properties of serotonergic axons. The proposed experimental platform can support next-generation analyses of the serotonergic matrix, including seamless integration with supercomputing approaches.

High-resolution spatiotemporal analysis in 6,791 English areas reveals declines in life expectancy

BackgroundHealth inequalities in the UK and other advanced economies are receiving renewed attention by the public and governments in light of the Covid-19 pandemic. Current data on mortality and longevity in England lack precision either in space or in time. We estimated trends from 2002 to 2019 in life expectancy for all 6,791 English Middle-layer Super Output Areas (MSOAs, median population 7,985 in 2019).MethodsWe used data for all deaths in England from 2002 to 2019 with information on age, sex and MSOA of residence and data on population by age, sex and MSOA. We used a Bayesian hierarchical model to obtain stable estimates of age-specific death rates by sharing information across age groups, MSOAs and years. We used life-table methods to calculate life expectancy at birth by sex and MSOA.ResultsIn 2002-2006 and 2006-2010, the vast majority of MSOAs experienced a life expectancy increase for both sexes. In 2010-2014, female life expectancy decreased in 322 (5%) of MSOAs for women. By 2014-2019, the number of MSOAs with declining life expectancy was 1,178 (17%) for women and 635 (9%) for men. The life expectancy increase from 2002 to 2019 was smaller where life expectancy had been lower in 2002, mostly northern urban MSOAs, and larger where life expectancy had been higher in 2002, mostly MSOAs in and around London. As a result of these trends, the gap between the lowest and highest MSOA life expectancy increased from 2002 to 2019, to reach 20.6 (95% credible interval 17.3-24.4) years for women and 27.2 (23.4-31.7) years for men.ConclusionsThis study highlights not only rising inequality, but also declining life expectancy in a large proportion of English communities. The post-Covid rebuild policies in England must include pro-equity economic and educational policies, and expansion of public health and healthcare.Key messages Between 2014 and 2019, 17% and 9% of communities in England underwent a decrease in life expectancy for women and men respectively, highlighting the immediate need for equity-focussed policies.For women, 2% of small areas in England have experienced a long-term decline in life expectancy between 2002 and 2019.

We used life table methods to calculate life expectancy at birth and probabilities of death in different ages by sex and MSOA.FindingsIn 2002–06 and 2006–10, all but a few (0–1%) MSOAs had a life expectancy increase for female and male sexes. In 2010–14, female life expectancy decreased in 351 (5·2%) of 6791 MSOAs. By 2014–19, the number of MSOAs with declining life expectancy was 1270 (18·7%) for women and 784 (11·5%) for men. The life expectancy increase from 2002 to 2019 was smaller in MSOAs where life expectancy had been lower in 2002 (mostly northern urban MSOAs), and larger in MSOAs where life expectancy had been higher in 2002 (mostly MSOAs in and around London). As a result of these trends, the gap between the first and 99th percentiles of MSOA life expectancy for women increased from 10·7 years (95% credible interval 10·4–10·9) in 2002 to reach 14·2 years (13·9–14·5) in 2019, and for men increased from 11·5 years (11·3–11·7) in 2002 to 13·6 years (13·4–13·9) in 2019.InterpretationIn the decade before the COVID-19 pandemic, life expectancy declined in increasing numbers of communities in England. High-resolution spatiotemporal transcriptome analyses during cellularization of rice endosperm unveil the earliest gene regulation critical for aleurone and starchy endosperm cell fate specification

The major tissues of the cereal endosperm are the starchy endosperm (SE) in the inner and the aleurone layer (AL) at the outer periphery. The fates of the cells that comprise these tissues are determined according to positional information; however, our understanding of the underlying molecular mechanisms remains limited. Here, we conducted a high-resolution spatiotemporal analysis of the rice endosperm transcriptome during early cellularization. In rice, endosperm cellularization proceeds in a concentric pattern from a primary alveolus cell layer, such that developmental progression can be defined by the number of cell layers. Using laser-capture microdissection to obtain precise tissue sections, transcriptomic changes were followed through five histologically defined stages of cellularization from the syncytial to 3-cell layer (3L) stage. In addition, transcriptomes were compared between the inner and the outermost peripheral cell layers. Large differences in the transcriptomes between stages and between the inner and the peripheral cells were found. SE attributes were expressed at the alveolus-cell-layer stage but were preferentially activated in the inner cell layers that resulted from periclinal division of the alveolus cell layer. Similarly, AL attributes started to be expressed only after the 2L stage and were localized to the outermost peripheral cell layer. These results indicate that the first periclinal division of the alveolus cell layer is asymmetric at the transcriptome level, and that the cell-fate-specifying positional cues and their perception system are already operating before the first periclinal division. Several genes related to epidermal identity (i.e., type IV homeodomain-leucine zipper genes and wax biosynthetic genes) were also found to be expressed at the syncytial stage, but their expression was localized to the outermost peripheral cell layer from the 2L stage onward. We believe that our findings significantly enhance our knowledge of the mechanisms underlying cell fate specification in rice endosperm.

Integrated meteorological drought monitoring framework using multi-sensor and multi-temporal earth observation datasets and machine learning algorithms: A case study of central India

Devising strategies for drought planning and risk mitigation in predominantly small holder agriculture regions like South Asia requires improved understanding of spatio-temporal characteristics of droughts at finer spatial scales. In this study, an integrated framework is developed to generate high-resolution gridded precipitation products from publicly available coarse scale data, and use the downscaled products for analysis of meteorological droughts. Data from a region in Central India is used to develop and test the framework. The Standard Precipitation Index (SPI) is used to characterize meteorological drought. The proposed framework incorporates: (i) a Random Forest machine learning algorithm to downscale CHIRPS (5 km) gridded long term rainfall data to a higher resolution gridded product (1 km) using high resolution multi-sensor and multi-temporal earth observation data of land surface characteristics (vegetation, temperature, and topography) as cofactors; (ii) Computation of 3-month SPI (SPI-3) for the downscaled gridded rainfall; (iii) Run theory applied to downscaled SPI grid to determine drought characteristics - duration, severity, intensity, frequency, and onset and cessation of droughts, and (iv) Principal Components Analysis (PCA) to combine effects of multiple drought characteristics into a single composite Effective Meteorological Drought Index (EMDI), to identify sub-regional drought patterns. The downscaled gridded precipitation product (1 km) was validated using: (i) original CHIRPS (5 km) data, and (ii) measured rainfall data from 65 rain gauge stations in the study area, with respect to conservation of both statistical properties and spatial structure. Comparisons were also made with drought characteristics obtained from Standardized Evapotranspiration Index (SPEI) derived from MODIS evapotranspiration data. The downscaled product captures spatio-temporal variability of droughts at finer village and sub-village scales, compared to original CHIRPS (5 km) data for which each pixel encompassed multiple villages. Spatial distribution of composite index EMDI delineates the study area into two characteristic drought risk regions. In one region, droughts are of longer duration, but are less frequent, with lower severity and intensity. In the second region droughts are of shorter duration but of higher severity, intensity and frequency. The integrated framework developed in this study for high resolution spatio-temporal analysis of droughts, starting from coarse scale precipitation data, is practical and sufficiently general to adopt in other regions to support local drought risk planning and targeting mitigation decisions and actions.

Spatio-Temporal Variability in North Atlantic Oscillation Monthly Rainfall Signatures in Great Britain

The North Atlantic Oscillation (NAO) is the primary atmospheric-oceanic circulation/teleconnection influencing regional climate in Great Britain. As our ability to predict the NAO several months in advance increases, it is important that we improve our spatio-temporal understanding of the rainfall signatures that the circulation produces. We undertake a high resolution spatio-temporal analysis quantifying variability in rainfall response to the NAO across Great Britain. We analyse and map monthly NAO-rainfall response variability, revealing the spatial influence of the NAO on rainfall distributions, and particularly the probability of wet and dry conditions/extremes. During the winter months, we identify spatial differences in the rainfall response to the NAO between the NW and SE areas of Britain. The NW area shows a strong and more consistent NAO-rainfall response, with greater probability of more extreme wet/dry conditions. However, greater NAO-rainfall variability during winter was found in the SE. The summer months are marked by a more spatially consistent rainfall response; however, we find that there is variability in both wet/dry magnitude and directionality. We note the implications of these spatially and temporally variable NAO-rainfall responses for regional hydrometeorological predictions and highlight the potential explanatory role of other atmospheric-oceanic circulations.

Regulation of mRNA translation by a photoriboswitch.

Optogenetic tools have revolutionized the study of receptor-mediated processes, but such tools are lacking for RNA-controlled systems. In particular, light-activated regulatory RNAs are needed for spatiotemporal control of gene expression. To fill this gap, we used in vitro selection to isolate a novel riboswitch that selectively binds the trans isoform of a stiff-stilbene (amino-tSS)-a rapidly and reversibly photoisomerizing small molecule. Structural probing revealed that the RNA binds amino-tSS about 100-times stronger than the cis photoisoform (amino-cSS). In vitro and in vivo functional analysis showed that the riboswitch, termed Werewolf-1 (Were-1), inhibits translation of a downstream open reading frame when bound to amino-tSS. Photoisomerization of the ligand with a sub-millisecond pulse of light induced the protein expression. In contrast, amino-cSS supported protein expression, which was inhibited upon photoisomerization to amino-tSS. Reversible photoregulation of gene expression using a genetically encoded RNA will likely facilitate high-resolution spatiotemporal analysis of complex RNA processes.

Relation between Release of Catecholamines and FFN511 Studied with Electrochemical Detector Arrays

Exocytosis is an essential vesicle fusion process that mediates transmitter release. For high resolution spatiotemporal analysis of single vesicle fusion events, a combinatorial approach of amperometry and fluorescence imaging was employed. Amperometric spikes from physically stimulated bovine chromaffin cells loaded with the Fluorescent False Neurotransmitter FFN511were detected by microfabricated four-electrode electrochemical detector (ECD) arrays. To determine the location of a fusion event leading to amperometric spikes with different quantal size in the different ECD array electrodes, random walk simulations were performed using the actual ECD array geometry. The location of a measured release event was fitted to match the simulated data. To account for drift of the ECD array, the pixel shift of each frame was fitted relative to the image that was used to generate the simulation map. The transparent glass surface in the center of the ECDs allowed the concurrent fluorescence imaging of localized loss of FFN511from secretory vesicles in TIRF microscopy mode. The frequency of amperometric spikes was much higher than that of FFN511 release events. Only the amperometric spikes that were correlated in time and location with a loss of fluorescence event were considered for analysis. The intensity time course of a 3x3 pixel area (480 x 480 nm2) around the fusion site of the fluorescence movie was determined and different fusion events were aligned at the onset of their respective amperometric spikes. Time super-resolution analysis using the recently developed Event Correlated Microscopy (ECOM) method revealed that FFN511 release followed the amperometric spike onset with a mean delay of ∼115 ms, presumably reflecting slow diffusion of FFN while in amperometric detection catecholamine molecules are rapidly consumed during the oxidation process. Sustainable energy supply is an essential part for economic and social development in every society. Islands as geographical isolated regions have to confront a number of challenges to secure a reliable and clean energy system. Currently, electricity demand on the Greek island of Crete is supplied by oil-fired engines imposed to new emissions restrictions applied from 2020. Thus, a capacity upgrade is necessary and new solutions driven by renewable energy, energy storage and interconnections. This study investigates three scenarios: Business as Usual (BAU); Natural Gas (NG); and the Interconnection of Crete with the National Grid System (Int.) to assess the potential techno-economic and environmental impact of the required transition under the Energy Trilemma Index. A capacity expansion and operation optimisation process has been applied through a high resolution spatio-temporal analysis performed with PLEXOS Integrated Energy Model. It was concluded that no BAU Scenario could facilitate a future plan for the electricity system of Crete or any European island imposed to such constrains. The optimum scenario incorporates interconnectors and energy storage systems that manage to deliver 52% reduction in the total system costs (2020–2040), 79% in electricity generation costs and 48% reduction in GHG emissions by 2040, compared to the BAU.

High resolution spatiotemporal analysis of erosion risk per land cover category in Korçe region, Albania

Some recent land use changes in Albania, such as deforestation, cropland abandonment, and urban sprawl, have caused serious increase of erosion risk. The main objective of this study was to map erosion risk in Korce region and assess the degree at which every land use is concerned. The G2 erosion model was applied, which can provide erosion maps and statistical figures at month-time intervals using input from free European and global geodatabases. The mapping results in Korce region were derived at a 30-m cell size, which is an innovation for G2. Autumn-winter months were found to be the most erosive, with average erosion rates reaching the maximum in November and December, i.e. 2.62 and 2.36 t/ha, respectively, while the annual rate was estimated at 10.25 t/ha/yr. Natural grasslands, shurblands, mixed forests, and vineyards showed to exhibit the highest mean erosion rates, while shrublands, broad-leaved forests and natural grasslands were found to be the most extended land covers risky for non-sustainable erosion rates (i.e. >10 t/ha/yr). A detailed examination of the detected hot spots is now necessary by the competent authorities, in order to apply appropriate, site-specific conservation measures. Notably, use of SPOT VGT data did not prevent the maps from having extended gaps due to cloudiness. Sentinel-2 time series, freely available by the European Space Agency (ESA), have the potential to improve spatiotemporal coverage of V-factor, thus further empowering the G2 model, in the near future.

Climatic cooling potential and building cooling demand savings: High resolution spatiotemporal analysis of direct ventilation and evaporative cooling for the Iberian Peninsula

In the present study a new methodology allowing the assessment of building's cooling demand savings by the use of ventilated passive cooling systems is presented in a twofold innovative way. Firstly, using a redefined concept of the climatic cooling potential (CCP), which allows for the direct estimation of savings in building's cooling demand by the use of different passive cooling systems on a large spatiotemporal scale. Secondly, this assessment relies on high resolution climate dataset built using a regional climate model covering the Iberian Peninsula (IP) with a 9 km horizontal spacing and the period between 1989 and 2008. Here, the CCP concept is applied for direct ventilation and evaporative cooling, in such a way that it allows for a comparison with the building monthly cooling demand, providing a direct assessment on the cooling demand savings for any building, for three air flow rates. The results show that CCP is asymmetrically distributed both spatially and temporally within the IP. During the cooling season CCP values are above 1 kWh per m3 of building and 3 kWh per m3 of building, for direct ventilation and evaporative cooling, respectively. Evaporative cooling provides a less heterogeneous annual cycle of CCP than direct ventilation, with a relative difference in the south and central part of the Iberian Peninsula superior to 100% during summer. Nonetheless, despite the consistently higher values offered by evaporative cooling, in the coastal regions the relative difference between the two systems drops to less than 10% due to the higher moisture in the air. For the case of a typical office room in the region of Lisbon, in the month of August, the cooling demand savings provided by the use of direct ventilation and evaporative cooling can represent more than 27% and 40% of the cooling demand, respectively.

Monitoring the spatiotemporal dynamics of proteins at replication forks and in assembled chromatin using isolation of proteins on nascent DNA

Understanding the processes of DNA replication, chromatin assembly and maturation, and the replication stress response requires the ability to monitor protein dynamics at active and damaged replication forks. Detecting protein accumulation at replication forks or damaged sites has primarily relied on immunofluorescence imaging, which is limited in resolution and antibody sensitivity. Here we describe a procedure to isolate proteins on nascent DNA (iPOND) that permits a high-resolution spatiotemporal analysis of proteins at replication forks or on chromatin following DNA replication in cultured cells. iPOND relies on labeling of nascent DNA with the nucleoside analog 5-ethynyl-2'-deoxyuridine (EdU). Biotin conjugation to EdU-labeled DNA using click chemistry facilitates a single-step streptavidin purification of proteins bound to the nascent DNA. iPOND permits an interrogation of any cellular process linked to DNA synthesis using a 3- to 4-d protocol.

Increased expression of golli myelin basic proteins enhances calcium influx into oligodendroglial cells.

The myelin basic protein (MBP) gene encodes two families of proteins: the classic MBP constituents of myelin and the golli-MBPs, the function of which is less well understood. Previous work suggests that golli proteins may play a role in Ca2+ homeostasis in oligodendrocytes (OLs) and in T-cells. Overexpression of golli in OL cell lines induces elaboration of sheets and processes. Live imaging of these cells revealed a rapid retraction of the processes and sheets after depolarization with high K+. This phenomenon was associated with a significant increase in [Ca2+]int without changes in cell viability. The results indicated that golli produced its effect through Ca2+ influx, rather than Ca2+ release from intracellular stores. Furthermore, a specific [Ca2+]int chelator (BAPTA) or Cd2+, a specific blocker of voltage-operated Ca2+ channels, abolished the ability of golli to promote process extension in a dose-dependent manner. Analysis of the golli protein identified a myristoylation site at the C terminus of the golli domain, which was essential for the action of golli on Ca2+ influx, suggesting that binding of golli to the plasma membrane is important for modulating Ca2+ homeostasis. High-resolution spatiotemporal analysis along N19 processes revealed higher-amplitude local Ca2+ influx in regions with elevated levels of golli. These findings suggest a key role for golli proteins in regulating voltage-gated Ca2+ channels in OLs during process remodeling. Our observations are consistent with the hypothesis that golli proteins, as a part of a protein complex, modulate Ca2+ influx at the plasma membrane and along OL processes.

Dynamic imaging of T cell-dendritic cell interactions in lymph nodes.

T cell immune responses begin within organized lymphoid tissues. The pace, topology, and outcomes of the cellular interactions that underlie these responses have, so far, been inferred from static imaging of sectioned tissue or from studies of cultured cells. Here we report dynamic visualization of antigen-specific T cells interacting with dendritic cells within intact explanted lymph nodes. We observed immunological synapse formation and prolonged interactions between these two cell types, followed by the activation, dissociation, and rapid migration of T cells away from the antigenic stimulus. This high-resolution spatiotemporal analysis provides insight into the nature of cell interactions critical to early immune responses within lymphoid structures.
