Porphyrin Aluminum Metal-Organic Framework in Liquid Water, its Interaction with the Oxidized Organosulfur Compound Diethyl Sulfoxide, and its Sorption from Aqueous Solution.

Oxidized organosulfur compounds and, in particular, sulfoxides are of interest as solvents in the semiconductor and pharmaceutical industry, environmental contaminants, and simulants in deactivation of chemical warfare agents. An experimental study is reported of the interaction of porphyrin aluminum metal-organic framework Al-MOF-TCPPH2 (Compound 2) with diethyl sulfoxide (DESO) in pure form and in aqueous solution. First, the suitability of Compound 2 as sorbent in aqueous solution was assessed; namely, its long-term stability (up to 15 days) in liquid water has been investigated at room temperature and under stirring. Here, a novel facile spectroscopic method has been used, a periodic micro-sampling of sorbent from suspension, followed by vacuum mini-filtration and an ex situ time-dependent attenuated total reflection Fourier transform infrared spectroscopy (ATR FT-IR) analysis. Next, the interaction of Compound 2 with pure liquid DESO under ambient conditions was investigated, which yields the stoichiometric adsorption complex (Al-MOF-TCPPH2)1(DESO)2 denoted Compound 3. In this adsorption complex, molecules of DESO interact with the OH group and carboxylate group of the sorbent. Then, the removal of DESO from Compound 3 was assessed, using facile treatment with warm water in the micro Soxhlet apparatus followed by the ATR FT-IR analysis. Finally, Compound 2 was tested in sorption of DESO from diluted aqueous solution. In the initial step, the sorption proceeds very quickly (in <1 min the concentration of DESO decreases by about 20%) followed by a much slower step. The maximum amount of adsorbed DESO corresponds to half of the amount adsorbed from pure DESO as found by the high-performance liquid chromatography-ultraviolet detection method. This adsorbed amount corresponds to 1 mol DESO adsorbate per mol of sorbent. Porphyrin aluminum metal-organic framework Compound 2 is promising for the removal of DESO from diluted aqueous solution, and it is of interest for the removal of similar oxidized organosulfur compounds.

A Polyacrylate Cotton-based Pipette Tip Micro-solid-phase Extraction Technique Coupled with High-performance Liquid Chromatography for Carvedilol Determination in Aqueous Media

Introduction: In this work, a polyacrylate polymer was synthesized into a pipette tip containing cotton fibers and used to extract carvedilol from water and urine samples. Methods: A high-performance liquid chromatography-ultraviolet detection method was developed, which demonstrated the suitability of the purposed pipette tip micro-solid-phase extraction device. Factors affecting the fabrication procedure and polymer quality were studied and optimized. In the next step, the sample preparation process (including extraction and desorption) was fully optimized, and the optimized method was validated. Results: A coating with suitable mechanical and chemical stability was achieved. Its structure was successfully characterized by Fourier transform infrared spectroscopy and scanning electron microscopy. Within-batch and between-batch fabrication reproducibility were obtained at 3.0 and 9.0 %, respectively. The developed method displayed a limit of detection of 1.1 µg L-1 when 1.5 mL of sample was processed, and it was linear in the concentration range of 3.3-350 µg L-1 with LLOQ of 5 µg L-1 . The polyacrylate cotton-based pipette tip was finally used to extract carvedilol from aqueous media with acceptable recoveries of 92-106%. Conclusion: The proposed method is simple and fast and requires low sample volumes. In addition, this method has been evaluated in terms of greenness with three different tools, and the evaluation results with all three tools have shown that this method is one of the green and environmentally friendly methods.

Development, optimization and validation of an analytical method for the determination of voriconazole in plasma by high-performance liquid chromatography-ultraviolet detection: Application for comprehensive study

Voriconazole is a widely used antifungal agent in clinical settings. However, its use has been associated with neurological side effects in some patients. For this reason, it is crucial to monitor its plasma levels to ensure that they are within the therapeutic range. Thus, in this study, we aimed to develop a simple, fast, and efficient method for the determination of voriconazole in plasma using reversed-phase HPLC-UV. We also aimed to validate the method for its application to routine analysis of immunocompromised patients. Plasma samples from immunocompromised patients were subjected to deproteinization with acetonitrile followed by centrifugation. Chromatographic separation was carried out on a C18 column with UV detection at 254 nm in isocratic mode. The concentrations were calculated by comparing peak areas to those of the internal standard, ketoconazole. The method was validated using the accuracy profile, which uses a calibration curve established for the therapeutic range of 1 to 5.5 µg/mL. The developed method was proved to be rapid by giving a short analysis time for voriconazole at around 5.5 min. Additionally, no interference with the biological matrix was detected. The obtained recoveries were higher than 90%. The accuracy profile showed that the method was accurate and precise for the determination of voriconazole in plasma. The developed method was proved to be simple, efficient, that requires minimal sample preparation. Thus it can be routinely applied for the therapeutic monitoring of voriconazole.

Pompon mum-like ionic covalent organic framework nanocomposites for efficient solid-phase extraction of nonsteroidal anti-inflammatory drugs

Molecularly imprinted ionic covalent organic framework nanocomposites (MI-IC-COF@SnO2) were prepared as potential adsorbents for the enhanced adsorption of nonsteroidal anti-inflammatory drugs (NSAIDs) from aqueous solution. The resulting material exhibited a pompon mum-like structure, featuring a large surface area, and well-defined mesopores. The presence of uniform positive ions within the three-dimensional skeleton of MI-IC-COF@SnO2 facilitated a rapid adsorption rate and high adsorption capacity for target analytes. Thermodynamic fitting revealed the adsorption process of NSAIDs to be feasible, endothermic, and spontaneous. Additionally, the adsorbent material exhibited respectable selectivity, as evidenced by imprinting factor values ranging from 2.8 to 6.7. Utilizing MI-IC-COF@SnO2 as the sorbent, a solid-phase extraction method coupled with high-performance liquid chromatography-ultraviolet detection (SPE-HPLC-UV) was developed and optimized. The proposed method demonstrated good linear range with determination coefficients of 0.998-0.999, and low limit of detection (0.18-1.35 µg L−1). Recoveries of NSAIDs in urine and river water samples were 78.1%–106.1%, with relative standard deviations lower than 12.5%. This rapid and sensitive method enables the determination of NSAIDs at trace levels in complex matrices, providing reliable and reproducible results.

Fabrication of magnetic hydrophilic imprinted polymers via two-step immobilization approach for targeted detecting bisphenol A

Molecularly imprinted polymer with water-compatibility for effective separation and enrichment of targeted trace pollutants from complicated matrix has captured extensive attention in terms of their high selectivity and matrix compatibility. This study focuses on modified β-cyclodextrin is used as a hydrophilic functional monomer to develop magnetic molecularly imprinted polymers (MMIPs). MMIPs were prepared using Fe3O4 nanoparticles as carriers and bisphenol A (BPA) as templates using a two-step fixation strategy and surface imprinting technology. The structural characteristic and binding properties of the prepared MMIPs were thoroughly studied. The MMIPs exhibited high crystallinity, high adsorption capacity, fast rebinding rate, remarkable selectivity and distinguish reusability. In addition, through magnetic solid-phase extraction separation technology and high-performance liquid chromatography ultraviolet quantitative detection technology, MMIPs are used for selective enrichment and detection of BPA in complex media such as environmental water and milk. This work provides a new route to construct the hydrophilic molecularly imprinted materials and a new sight on developing more effective sample pretreatment strategies for monitoring targeted pollution in complicated aqueous media.

Natural kapok fiber as a green and efficient adsorbent for in-syringe solid-phase extraction in the determination of antidepressants in biofluids

Effective antidepressant monitoring is crucial for safe treatment, with liquid chromatography being the primary detection method. Due to the complexity of biofluids, meticulous sample preparation is vital to remove interferences and concentrate analytes. In this study, a novel solid-phase extraction method utilizing unprocessed kapok fiber (KF) as an eco-friendly and efficient sorbent, ingeniously incorporated within an in-syringe device for the extraction of antidepressants from biofluids was introduced. The extraction process displayed several advantages, including simplicity and rapidity (less than 3 min with minimal manual intervention), as well as the potential for automation, which represents significant advancements in sample preparation techniques. The method demonstrated excellent analytical performance when coupled with high-performance liquid chromatography-ultraviolet detection, achieving good linearity (R2 ≥ 0.991), excellent relative recovery (92.6–107.5 %), high precision in intraday and interday analyses (RSDs within 0.8–6.0 % and 1.2–7.9 %, respectively), and good sensitivity with limits of quantification between 5–10 ng/mL. Notably, given the use of a renewable and biodegradable material with KF, and a low-toxicity desorption solvent with ethanol, the developed method stands out for its environmental sustainability, aligning with green analytical chemistry principles. Its application to real biological samples validates its efficacy as a rapid and reliable tool for clinical and forensic monitoring of antidepressants. Moreover, the low cost of this method facilitated by the economical nature of KF and the reusability of syringes further enhances its practicability. This study presents a unique approach by employing natural and renewable KF as adsorbents, thereby eliminating the need for complex material synthesis and making a pioneering contribution to the field of analytical chemistry for drug analysis in complex matrices.

Modification of cellulose substrate by in situ synthesis of metal-organic framework-5 for thin film microextraction of some non-steroidal anti-inflammatory drugs and their measurement by high-performance liquid chromatography-ultraviolet detector

This work, reports the successful preparation a thin film by a simple and inexpensive process for quantification of a model analytes in the urine sample using HPLC-UV. To this end, cellulose paper was employed as a substrate for the in-situ synthesis of MOF-5, to increase the resistance of the prepared film. The prepared film can be reused 26 times with no reduction in its performance. The thin film prepared by MOF-5 modified cellulose substrate was utilized in thin film microextraction (TFME) method for the extraction and preconcentration of naproxen, aspirin, tolmetin, and celecoxib. Under optimal conditions, the linear dynamic range of the target analytes was 2–500 µg L−1 with correlation coefficients (R2) ranging from 0.9961 to 0.9990. Also, the limits of detection (LODs), the limits of quantification (LOQs) and relative standard deviation (RSD%) of the proposed method for selected analytes ranged between 0.57 and 0.77 µg L−1, 1.7 to 2.3 and 3.5 % to 6.2 %, respectively. Moreover, relative recoveries varied from of 94 % to 108 %, indicating the absence of matrices effect in the proposed method. Eventually, the TFME was successfully used for the extraction of selected analytes from urine samples.

Combining EDTA-intercalated (Zn-Al) layered double hydroxides/graphite and two-phase hollow fiber for liquid phase microextraction of nonsteroidal anti-inflammatory drugs

This study synthesized and characterized layered double hydroxides (LDHs) intercalated with ethylenediaminetetraacetic acid(EDTA) to increase the basal spacing of electrosynthesized Zn-Al-LDHs on pencil graphite (PG). The characteristic verification of the Zn-Al-EDTA-LDH nanostructures was conducted utilizing Fourier-transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), energy dispersive and X-ray analysis (EDS), and X-ray diffraction (XRD). To improve the extraction efficiency, Zn-Al-EDTA-LDHs as adsorbent in solid phase microextraction (SPME) fiber was combined with hollow fiber liquid–liquid phase microextraction (HF-LLPME) to investigate the extraction efficiency of HF-LLME@PG-SPME technique the chosen non-steroidal anti-inflammatory drugs (NSAIDs) from aqueous solution Several variables, comprising extraction time, desorption time, stirring rate, desorption volume, pH of donor phase, and salt effects were screened and optimized by performing placket-Burman design (PBD). After evaluating the extraction and desorption procedures under optimal conditions, NSAIDs analysis was performed employing high-performance liquid chromatography-ultraviolet detection (HPLC-UV). Compared to the most common clean-up and sample preparation methods, the current study focused on developing a greener analytical chemistry process due to reduced solvent consumption, cost-effectiveness, miniaturization and simplified design. The proposed method for NSAIDs exhibited a good linear dynamic range (LDRs) (1.00–200.00 µg/L) with a coefficient of determination of > 0.9956, low limits of detection (LODs) (0.26–0.29 µg/L) and acceptable limits of quantifications (LOQs) (0.86–0.96 µg/L). Relative standard deviations of 5.31 % could be determined. The absolute recoveries of the urine sample analyses could be obtained in a 89.91–97.61 % range and enrichment factors (EFs) (178.92 to 183.88). Investigations of the developed method were also conducted in various urine samples containing the selected drugs. As a result of the obtained recoveries, the method proved to be valuable and suitable to complicated biological samples.

Utilization of a pH-switchable hydrophilicity solvent for the microextraction of clomipramine from human urine samples

Clomipramine (CLP) is a tricyclic antidepressant drug, and its determination in biological samples is of high importance in clinical and forensic evaluations to assure appropriate drug concentrations. In the present study, benzoic acid was employed as a pH-switchable hydrophilicity solvent (SHS) for the microextraction of CLP from authentic human urine samples prior to its determination by high performance liquid chromatography-ultraviolet detection (HPLC-UV). The microextraction protocol was based on the phase transition of the SHS through pH alteration that resulted in its rapid dispersion and simultaneous phase separation. The obtained solid was collected in a syringe filter, dissolved in methanol, and analyzed. The main parameters that affected the efficiency of the microextraction procedure were studied and optimized to ensure high extraction efficiency for CLP and the analytical method was validated. Under optimum conditions, good linearity was observed between 0.05 and 5.0 μg mL−1. The limit of detection and limit of quantification were found to be 0.015 and 0.05 μg mL−1, respectively. The RSD values for intra-day repeatability and inter-day precision were 2.4–8.9 % and 1.7–9.1 %, respectively. The relative recovery values were within 90.0 and 110.0 % in all cases, demonstrating good method accuracy. The proposed SHS microextraction showed cost-efficiency, handling simplicity, and rapidity resulting in enhanced sample throughput. Moreover, the proposed method exhibited a green character and good applicability based on its evaluation by Green Analytical Procedure Index and Blue Applicability Grade Index.

Three-year study of DNA cytosine methylation dynamics in transplanted Malbec grapevines

DNA cytosine methylation, an epigenetic mechanism involved in gene regulation and genome stability, remains poorly understood in terms of its role under changing environmental conditions. Previous research using methylation-sensitive amplified polymorphism (MSAP) markers in a Vitis vinifera L. cv. Malbec clone showed vineyard-specific DNA methylation polymorphism, but no change in overall methylation levels. To complement these findings, the present study investigates the intra-seasonal epigenetic dynamics between genetically identical plants grown in different vineyards through a transplanting experiment. Cuttings of the same clone, showing differential methylation patterns imposed by the vineyard of origin (Agrelo and Gualtallary), were cultivated in a common vineyard (Lunlunta). Using high-performance liquid chromatography-ultraviolet detection, the quantification of global DNA 5-methylcytosine (5-mC) levels revealed relatively low overall 5-mC percentages in grapevines, with higher levels in Agrelo (5.8%) compared to Gualtallary plants (3.7%). The transplanted plants maintained the 5-mC levels differences between vineyards (9.8% vs 6.2%), which equalized in subsequent seasons (7.5% vs 7%). Additionally, the study examined 5-mC polymorphism using MSAP markers in Lunlunta transplanted plants over three seasons. The observed differences between vineyards in MSAP patterns during the initial growing season gradually diminished, suggesting a reprogramming of the hemimethylated pattern following implantation in the common vineyard. In contrast, the non-methylated pattern exhibited greater stability, indicating a potential memory effect. Overall, this study provides valuable insights into the dynamic nature of DNA methylation in grapevines under changing environmental conditions, with potential implications for crop management and breeding strategies.

Simultaneous determination of selected ionophoric coccidiostats and amino acids in feed premixes using high-performance liquid chromatography-ultraviolet detection method.

The combination of ionophoric coccidiostats and amino acids (AAs) is important in poultry feeding to enhance immunity and improve the growth and feed efficiency of birds suffering from coccidiosis. A simple, rapid, and economical high-performance liquid chromatography-ultraviolet detection (HPLC-UV) method for the simultaneous determination of three ionophoric coccidiostats, namely salinomycin (SAL), maduramicin (MAD), and monensin (MON) in addition to three AAs; L-tryptophan (L-TRP), alpha-ketoleucin (KLEU), and L-valine (L-VAL) in feed premixes was developed and validated. Chromatographic separation was achieved in less than 12min using a phenyl hexyl column with a mobile phase consisting of acetonitrile/methanol/water (25:20:55, v/v/v) adjusted to pH 3 using phosphoric acid. Isocratic elution was performed at a flow rate of 1mL/min with UV detection at 210nm. The method showed good linearity in the ranges 0.50-5.0mg/mL for MON, 0.20-2.0mg/mL for MAD and SAL, 10.0-100.0μg/mL for L-TRP and KLEU, and 50.0-500.0μg/mL for VAL. The developed method was successfully applied to determine the studied analytes in feed premixes with good recoveries and precision. The good validation criteria of the proposed method allow its utilization in quality control laboratories.

UIO-66-NH2 encapsulated polyoxometalate loaded on graphene oxide accommodated on mixed cellulose esters (MCE) paper for thin film microextraction of pesticides besides their quantification via high-performance liquid chromatography-ultraviolet detection

A POM@UIO-66-NH2/GO composite was designed and prepared via a facile in-situ synthesis method for microextraction purposes. The proper sizes (window size and pore size) of UIO-66-NH2 can effectively seal POM as a monomolecular active component and prevent POM leaching during the extraction. After that, a membrane containing POM@UIO-66-NH2/GO nanocomposite was synthesized and utilized in thin film microextraction (TFME) of various pesticides such as imidacloprid, tebuconazole, and propiconazole as the model analytes in different real samples such as cucumber, tomato, potato, head lettuce, and strawberry. It is worth mentioning that the introduction of GO can improve the electron transfer of the sorbent. The extracted model analytes were desorbed by a suitable organic solvent. The extracted analytes were quantified via high-performance liquid chromatography-ultraviolet detection (HPLC-UV). Under the optimized condition, the method's linear dynamic range (LDR) was obtained in the range of 1.0–200.0 μg L−1 for testing analytes. The obtained correlation coefficients (r2) were higher than 0.9918. The limits of detection (LODs) were also calculated to be between 0.31 and 0.34 μg L−1. The relative standard deviations (%RSDs) were obtained in the range of 2.06 to 4.11. The enrichment factor (EF) values for the studied analytes were also obtained in the range of 89–102. The obtained recoveries indicated that the presented method is useful for analyzing pesticides in various real samples.

In situ growth of ZIF-67 to construct core-shell mosaic structural composites for efficient extraction of benzoylurea insecticides.

A novel mosaic structure Silica@C/Co@ZIF-67 composite was synthesized by successfully embedding Co nanoparticles on the surface of silica spheres with the help of thermoplastic polyethyleneimine by carbon-reduction. The ZIF-67 half-shell layer structure was synthesized by the in-situ growth of ZIF-67 on the surface of silica spheres through the coordination of 2-methylimidazole with Co metal nodes. The composite was used as a magnetic solid-phase extraction adsorbent combined with high performance liquid chromatography-ultraviolet detector (HPLC-UV) for the extraction and determination of benzoylurea insecticides (BUs) in vegetables and tea. Based on the presence of π-π, hydrophobic and hydrogen bonding interactions between Silica@C/Co@ZIF-67 and BUs, the BUs were rapidly captured by the composites resulting in high adsorption performance. Under the optimal extraction parameters, the linear ranges were 0.3-200µg L-1 for diflubenzuron, 0.6-200µg L-1 for chlorbenzuron, and 1.0-200µg L-1 for triflumuron, teflubenzuron, and flufenoxuron, with correlation coefficients (R2) greater than 0.9991. The limits of detection (LODs) of the method were 0.1-0.3μg L-1, and the relative standard deviations (RSDs) were 1.2-3.0% for intra-day and 2.6-4.6% for inter-day. In the spiked recovery experiments of vegetables and tea, the recoveries of the five kinds of BUs ranged from 75.8 to 112.9%. In addition, after 10 repetitions using Silica@C/Co@ZIF-67, the recoveries of the five kinds of BUs were still as high as 78.4 to 83.9%.

Simultaneous determination of four N-lauryl amino acid surfactants in facial cleansers by high performance liquid chromatography-ultraviolet detection

Significant developments have recently been achieved in the field of N-lauryl amino acid (NLAA) surfactants derived from renewable resources. Compared with conventional surfactants, NLAAs exhibit remarkable surfactant properties, exceptional biodegradability, good biocompatibility, and high safety profiles. These attributes have led to the widespread use of NLAAs in personal-care products. The detection methods employed for NLAAs include two-phase titration (TT), spectrophotometric analysis (SA), and high performance liquid chromatography (HPLC). However, because both TT and SA measure the total concentration of anionic active matter, identifying and quantifying individual compounds in a sample containing multiple anionic surfactants is impossible. The presence of cationic surfactants in the sample also introduces interferences, which lead to significant errors. Compared with TT and SA, HPLC offers direct and rapid testing procedures. However, compounds with no or weak UV-visible light absorption exhibit low sensitivity when detected by UV, necessitating the use of detectors such as differential refractive index detectors (RIDs), evaporative light scattering detectors (ELSDs), or charged aerosol detectors (CADs). Most HPLC users consider UV light as the fundamental configuration of the instrument, and other detectors are less commonly employed. Therefore, establishing a new HPLC method suitable for the UV detection of NLAAs is of practical significance. In this study, a novel HPLC-UV method was developed for the simultaneous detection of N-lauryl glutamine (LG), N-lauryl glycine (LC), N-lauryl alanine (LA), and N-lauryl sarcosine (LS) by optimizing the mobile-phase composition and selecting an appropriate chromatographic column and detection wavelength. The samples were mixed with acetonitrile-0.10% H3PO4 aqueous solution (60∶40, v/v) and sonicated for 10 min, then stayed at room temperature for 5 min. Subsequently, the mixture was filtered through a 0.22 μm filter membrane and separated on an Agilent Eclipse Plus C18 column (150 mm×4.6 mm, 5 μm). The mobile phase used for separation consisted of acetonitrile-0.10% H3PO4 aqueous solution at a flow rate of 1.0 mL/min. The detection wavelength was set at 205 nm, and the injection volume was 10 μL. The results demonstrated that the four NLAAs exhibited good linearity in the range of 2.0-800.0 mg/L, with correlation coefficients (r)≥0.9995. The limits of detection (LODs) ranged from 0.17 to 0.49 mg/L, and limits of quantification (LOQs) ranged from 0.57 to 1.63 mg/L. The relative standard deviations (RSDs) for precision, repeatability, and stability over 24 h were all below 2.0%. Using this method, the NLAA contents of five facial-cleanser products were determined. The results demonstrated that all five samples contained one or more NLAAs, and the total NLAA contents ranged from 64.58 to 97.01 mg/g. The five spiked-sample recoveries of the NLAAs at four different spiked levels (0.60, 4.50, 15.00, 24.00 mg/g) ranged from 94.3% to 107.4%, indicating satisfactory accuracy. However, the actual NLAA composition and label for one facial-cleanser product were not consistent with our test results. This finding demonstrates the necessity of strengthening market monitoring through testing. The proposed method has the advantages of simple pretreatment, rapid testing, good precision, high accuracy, and appropriate stability. Thus, it is suitable for the determination of NLAA contents in facial cleansers and provides an effective technical reference for the raw-material purity assessment, synthetic yield detection, and product quality control of this type of surfactant.

Development of a simple high-performance liquid chromatography-ultraviolet detection method for olaparib in patients with ovarian cancer.

Olaparib is a small-molecule inhibitor of poly(ADP)-ribose polymerase (PARP) used as maintenance therapy for recurrent ovarian cancer and newly diagnosed advanced ovarian cancer after initial chemotherapy. An exposure-toxicity correlation has been reported between the probability of anemia, a common adverse event associated with olaparib, and the steady-state minimum plasma concentration (Cmin) as well as the predicted maximum plasma concentration (Cmax). On the other hand, olaparib exhibits high interpatient variability with regard to the area under the concentration-time curve, Cmax, and Cmin. Therefore, we developed a simple and sensitive assay based on high-performance liquid chromatography with ultraviolet light (HPLC-UV) for the therapeutic drug monitoring of olaparib. The analysis was performed on an octadecylsilyl column with a mobile phase consisting of 0.5% KH2PO4 (pH 4.5) and acetonitrile (71:29, v/v), at a flow rate of 0.8 mL/min. Olaparib and an internal standard (imatinib) were well separated from the co-extracted material, with retention times of 13.6 and 11.5 min, respectively. The calibration curve for olaparib showed linearity over the concentration range of 0.10-10.0 μg/mL (r2 = 0.9998). The intra- and inter- day validation coefficients ranged from 1.79 to 4.13% and 1.37 to 3.55%, respectively. Measurement accuracy ranged from - 6.07 to 3.26%, with a recovery rate of more than 91.06%. The developed method was then applied to evaluate the plasma olaparib concentrations in patients with ovarian cancer. Our findings demonstrate that HPLC-UV is an economical, simple, and sensitive method for clinical application and holds promise for the effective drug monitoring of olaparib during ovarian cancer treatment.

Identification of ascorbic acid and phytochemical profiles of Scotch Bonnet pepper (Capsicum chinense) using High-Performance Liquid Chromatography-Ultraviolet Detector (HPLC-UV)

Researchers have discovered that red and green scotch peppers are extremely rich in phytochemicals and that these secondary metabolites are connected to several biological processes in the human body. This study aimed to determine the numerous phytochemicals present in scotch pepper seeds, flesh, and red and green varieties. The samples were pulverized after being air-dried. The phytochemicals and ascorbic acid contents were examined using a High-performance Liquid Chromatography-Ultraviolet detector (HPLC-UV). Twelve phytochemicals and ascorbic acid were identified in the flesh and seed of red scotch pepper, with lycopene having the highest content (83.10 mg/kg), the ascorbic acid content of 8.80 mg/kg; moreover, the seed of red scotch pepper revealed twelve phytochemicals and lycopene being the highest content (75.60 mg/kg) and ascorbic acid content of 6.80 mg/kg, furthermore, fourteen phytochemicals were identified in the flesh of red scotch bonnet pepper with lycopene the highest quantity (86.00 mg/kg) and ascorbic content of 10.60 mg/kg. Twelve phytochemicals were identified in the flesh and seed of green scotch bonnet pepper, lycopene (68.70 mg/kg) being the highest content, ascorbic acid content (6.50 mg/kg), twelve phytochemicals were revealed in the flesh of green pepper and lycopene had highest quantity (76.30 mg/kg), in the seed of green pepper, the lycopene content was 68.70 mg/kg. However, it has been shown that capsaicin, dihydrocapsaicin, and lutein concentrations varied, with red scotch pepper flesh having the highest concentration of capsaicin (7.50 mg/kg). The lycopene and ascorbic acid contents of flesh were the highest (86.00 mg/kg) and (10.60 mg/kg). The study's findings showed that red pepper flesh was better than other components that were looked at.

Determination of alectinib and its active metabolite in plasma by pipette-tip solid-phase extraction using porous polydopamine graphene oxide adsorbent coupled with high-performance liquid chromatography-ultraviolet detection

Alectinib is known as an effective targeted drug, which has excellent therapeutic effect on non-small cell lung cancer and can significantly prolong the survival of patients. Therapeutic drug monitoring is necessary due to the photo-instability of alectinib and the individual differences in patients. In this work, a porous polydopamine graphene oxide composite (PDAG) was prepared by a simple surface modification method. A PDAG-based pipette-tip solid-phase extraction (PT-SPE) coupled with HPLC-UV detection was proposed for the separation and detection of alectinib and its active metabolite M4 in plasma. The method was methodologically validated and showed good linearity in the range of 50–5000 ng mL−1 (R2 > 0.9995). The limit of detection (LOD) was 4.8 ng mL−1 and 3.9 ng mL−1 for alectinib and M4, respectively, and the limit of quantitation (LOQ) was 16.1 ng mL−1 and 13.1 ng mL−1, respectively. The intra-day and inter-day precision expressed by coefficient of variation was less than 4.8 %. The recovery of this method ranged from 84.9 % to 103.5 % with a standard deviation of less than 4.3 %. In conclusion, the established method is accurate, stable and inexpensive, and can be used to monitor the levels of alectinib and M4 in plasma, which provide technical and data support for exploring optimal individualized remedial dosing regimens.

Preparation and extraction property study of corn cob-like magnetic mosaic carbon materials derived from MOF-on-MOF composites

BackgroundLarge accumulations of pesticide residues in the environment eventually enter the human body with food. Based on magnetic solid-phase extraction (MSPE) technology, it is possible to achieve efficient extraction of trace pesticide residues in foodstuffs, and the construction of MSPE adsorbents with excellent magnetic properties and many active sites is still one of the main research topics. Based on this, we developed a new strategy for the preparation of "MOF-on-MOF" composites, which were carbonized and used as adsorbents for the extraction of pesticide residues in cereals. ResultsA novel corn cob core-shell composite Fe2O3@C@ZIF-8 was created by embedding ZIF-8 onto the surface of Fe2O3@C derived from MIL-88A(Fe), and used for the extraction of benzoylurea insecticides(BUs) from cereals. The adsorption behaviour between Fe2O3@C@ZIF-8 and BUs was investigated by static and kinetic experiments and the adsorption mechanism was elaborated. For the trace analysis of BUs, a magnetic solid-phase extraction combined with high performance liquid chromatography-ultraviolet detector (HPLC-UV) approach was also developed. Under the optimized experimental conditions, the limits of detection and quantification were 0.015–0.03 μg L−1 and 0.05–0.1 μg L−1, and the relative standard deviations for the intra-day and inter-day ranges were 1.82%–2.13 % and 3.85%–4.59 %, respectively. The spiked recoveries of the four cereals ranged from 82.72% to 104.45 %. After 10 cycles of use of Fe2O3@C@ZIF-8, the recoveries of BUs ranged from 77.90% to 96.74 %. SignificanceThe analytical method based on Fe2O3@C@ZIF-8 as adsorbent has a low limit of detection, a wide linear range and is highly applicable to the analysis of real samples. In addition, the strategy developed in this study for the preparation of thermoplastic polymer-assisted "MOF-on-MOF" composites has promising applications.

Preparation of metal-organic framework @molecularly imprinted polymers for extracting N-nitrosamines in salted vegetables

In this paper, the novel metal-organic framework @molecularly imprinted polymers were prepared and applied in extracting N-nitrosamines from salted vegetables. The imprinted polymers were coated on the surface of MIL-101 using multi-dummy template molecules (5-nonanol, benzhydrol and N-formylpyrrolidine). The characterization and adsorbing experiments showed that the hybrid imprinted polymers presented spherical particles with typically core-shell structure, and exhibited high adsorption capacity (maximum capacity: 46.85 mg/g) and fast equilibrium rate (only 5 min) for N-nitrosamines. Various parameters (sample loading solvent, pH, washing solvent, elution solvent and elution volume) affecting solid-phase extraction were optimized. Under the optimum conditions, the solid-phase extraction process based on the hybrid polymers combined with high performance liquid chromatography-ultraviolet detection method was established and applied to analyze N-nitrosamines in different salted vegetables. The results showed that the developed method produced the linear relationship between the peak areas versus the N-nitrosamines concentrations of 0.2–10 µg/g with limit of detections from 20.6 to 76.1 ng/g. The spiked recovery of N-nitrosamines in the salted vegetable samples was in the range of 66–100.5 % with relative standard deviation from 0.1 to 3.4 %. Those results demonstrated that the established method was sensitive and efficient for directly enriching and analyzing trace N-nitrosamines in salted vegetables.

Development of a universal sol–gel sorbent for fabric phase sorptive extraction and its application in tandem with high performance liquid chromatography-ultraviolet detection for the analysis of phthalates in environmental and drinking water samples

Most sorbent-based sample preparation methods are designed to interact with a class of analytes i.e., a polar sorbent for polar analytes, a non-polar sorbent for non-polar analytes, an ion-exchange sorbent for ionized compounds. A big challenge surfaces when making a choice of an appropriate sorbent when there is a mixture of compounds of varying polarities and ionization states in the complex sample matrices. Hence, the aim of this study was to develop a universal sol–gel sorbent coating for fabric phase sorptive extraction that would effectively and simultaneously extract polar, nonpolar, organic acids and bases via different intermolecular/interionic interactions. The universal sol–gel sorbent coated fabric phase sorptive extraction membrane was created by exploiting sol–gel surface coating technology to implant different functional moieties, ion exchanging ligands and an organic polymer that results in a 3-D organic–inorganic hybrid structure on the fabric substrate with a high level of selectivity/affinity for the target compounds. The preconcentrated extracts containing the isolated phthalates after the fabric phase sorptive extraction procedure were injected into the high-performance liquid chromatography-ultraviolet detection (HPLC-UV) system for the quantification. The FPSE-HPLC-UV method demonstrated a linear range between 50 and 1000 ng/mL for dimethyl phthalate, diethyl phthalate, dipropyl phthalate, diallyl phthalate, benzyl butyl phthalate, 1–10000 ng/mL for di-n-heptyl phthalate, 1–5000 ng/mL for di-n-heptyl phthalate, 10–5000 ng/mL for di-n-ethyl hexyl phthalate and 100–7500 ng/mL di-n-octyl phthalate. The intra-day and inter-day reproducibility (in terms of percentage relative standard deviation, %RSD) did not exceed 13.6. This method proves to be a selective, accurate, reproducible, cheap, and fast analytical approach for the quantitative and qualitative determination of the phthalates which is useful in crafting regulations for phthalates content in beverages and juices as well as in environmental monitoring.
