Effect of wastewater containing mixture of nanoparticles on organic matter removal, power generation, and biofilm integrity in sediment microbial fuel cell

ABSTRACT Industrial wastewaters from the cosmetic industry contain high organic strength and mixture of nanoparticles (NPs). Sediment microbial fuel cell (MFC) is an emerging nature-based technology that can treat complex wastewaters. The aim of this study is to understand the effect of a binary mixture of zinc oxide (ZnO) and copper oxide (CuO) NPs (concentration: 1 + 1 and 10 + 10 mg/L) on the organic matter removal, power generation, and biofilm health of sediment MFCs after a long-term operation of 120 days. The high chemical oxygen demand (COD) removal (>95%) observed for all reactors signified the minimal impact of 10 mg/L NP mixture on treatment. The dissolved organic carbon (DOC) removal from the sediment was reduced by 8% due to NPs. NPs also led to 42.2% higher anode extracellular polymeric substances (EPS) and 46.65% lesser cathode EPS generation. The maximum power density of 0.29 mW/m2 was obtained for the 10 mg/L NP reactor, with the average being 23% higher than the no-NP control reactor. This was the first study to explore the effect of the mixture of NPs on the performance of an MFC. The results indicated that sediment MFCs can sustain high mixture concentrations of NPs. Furthermore, variation of parameters can aid in establishing the feasibility of this technology for treating wastewater with NPs.

Effects of mixtures of polychlorinated biphenyls (PCBs) and three organochlorine pesticides on cognitive function differ between older Mohawks at Akwesasne and older adults in NHANES

BackgroundAkwesasne Mohawks has been exposed to high concentrations of polychlorinated biphenyls (PCBs) and background levels of organochlorine pesticides, hexachlorobenzene (HCB), dichlorodiphenyl dichloroethylene (DDE), and mirex. We have previously reported relative contributions to the mixture of low- and high-chlorinated PCBs, HCB, and DDE on cognitive decrements in Mohawks of various ages. ObjectiveThis study examines differences in the mixture effects of PCB congener groups, HCB, DDE, and mirex on cognitive function in older Mohawks and less PCB-exposed older adults from the National Health and Nutrition Examination Survey (NHANES) 1999–2002 cycles. MethodsWe used Bayesian kernel machine regression (BKMR) to evaluate the mixture effects of different PCB congener groups, HCB, DDE, and mirex on cognitive function in both populations. Models were adjusted for age, sex, education levels, and race/ethnicity focusing on individuals 60 years and older. ResultsOlder Mohawks had 3-fold higher mean total PCB concentrations and 1.8-fold higher mirex, but slightly lower mean DDE and HCB levels than NHANES older adults. Higher mixture concentrations were significantly associated with greater cognitive decline. In older Mohawks, low- and high-chlorinated PCBs, HCB, and DDE contributed to the cognitive score decline. In contrast, score decline in older NHANES adults were primarily from high-chlorinated PCBs and DDE with a threshold dose of approximately 2.08–2.27 ng/g and 2.02–2.40 ng/g, respectively. ConclusionMixtures of PCBs and organochlorine pesticides increase the risk of cognitive decline in both older Mohawks and NHANES older adults. However, contributions to these mixture effects show significant differences. In older Mohawks, high- and low-chlorinated PCBs, DDE, and HCB are the primary contributors, while high-chlorinated PCBs and DDE are important contributors in NHANES older adults. Due to chronic heavy exposures to PCBs, older Mohawks had a significantly increased risk of cognitive decline compared to general older adults from NHANES.

Biodegradation Kinetics of Fragrances, Plasticizers, UV Filters, and PAHs in a Mixture─Changing Test Concentrations over 5 Orders of Magnitude.

Biodegradation of organic chemicals emitted to the environment is carried out by mixed microbial communities growing on multiple natural and xenobiotic substrates at low concentrations. This study aims to (1) perform simulation type biodegradation tests at a wide range of mixture concentrations, (2) determine the concentration effect on the biodegradation kinetics of individual chemicals, and (3) link the mixture concentration and degradation to microbial community dynamics. Two hundred ninety-four parallel test systems were prepared using wastewater treatment plant effluent as inoculum and passive dosing to add a mixture of 19 chemicals at 6 initial concentration levels (ng/L to mg/L). After 1-30 days of incubation at 12 °C, abiotic and biotic test systems were analyzed using arrow solid phase microextraction and GC-MS/MS. Biodegradation kinetics at the highest test concentrations were delayed for several test substances but enhanced for the reference chemical naphthalene. Test concentration thus shifted the order in which chemicals were degraded. 16S rRNA gene amplicon sequencing indicated that the highest test concentration (17 mg C/L added) supported the growth of the genera Acidovorax, Novosphingobium, and Hydrogenophaga, whereas no such effect was observed at lower concentrations. The chemical and microbial results confirm that too high mixture concentrations should be avoided when aiming at determining environmentally relevant biodegradation data.

A mixture of Persistent Organic Pollutants (POPs) and Azoxymethane (AOM) show potential synergistic effects on intestinal tumorigenesis in the A/J Min/+ mouse model

A multitude of cancer types, including breast, testicular, liver and colorectal cancer, have associations with exposure to Persistent Organic Pollutants (POPs). The present study aimed to investigate whether a mixture of POPs could affect intestinal tumorigenesis in the A/J Min/+ mouse, a model for human colorectal cancer (CRC). Pollutants were selected for their presence in Scandinavian food products and the mixture was designed based on defined human estimated daily intake levels. Mice were exposed through the diet, at control, low and high mixture concentrations, for 10 weeks. In a separate experiment, mice also received one subcutaneous injection of Azoxymethane (AOM) to explore whether this carcinogenic compound influenced the effect of the POPs. Intestinal tumorigenesis was examined by surface microscopy and histopathology. Moderate and dose-dependent increases in tumorigenesis were observed after dietary POP exposure. The AOM treatment alone stimulated the growth of colonic lesions, but did not increase the formation of new lesions. Combined AOM treatment and POP exposure demonstrated a synergistic effect on lesion formation in the colon, and to a lesser extent in the small intestine. This synergy was also evident by an increased number of malignant colonic tumors (carcinomas). In conclusion, the study shows that a mixture of POPs interacted synergistically with a known carcinogen (AOM), causing increased intestinal tumorigenesis in the A/J Min/+ mouse model.

Responses of earthworms to repeated exposure to three biocides applied singly and as a mixture in an agricultural field

The study aimed at investigating effects of three differently acting biocides; the insecticide esfenvalerate, the fungicide picoxystrobin and the bactericide triclosan, applied individually and as a mixture, on an earthworm community in the field. A concentration–response design was chosen and results were analyzed using univariate and multivariate approaches. Effects on juvenile proportions were less pronounced and more variable than effects on abundance, but effects in general were species- and chemical-specific, and temporal variations distinct. Esfenvalerate and picoxystrobin appeared to elicit stronger effects than triclosan at laboratory-based ECx values, which is in accordance with our previous laboratory study on Eisenia fetida. The mixture affected abundance and juvenile proportions, but the latter only at high mixture concentrations. Esfenvalerate and picoxystrobin appeared to be the main drivers for the mixture's toxicity. Species-specific toxicity patterns question the reliability of mixture toxicity predictions derived on E. fetida for field earthworms. Biocide concentrations equaling EC50s (reproduction) for E. fetida provoked effects on the field earthworms mainly exceeding 50%, indicating effect intensification from the laboratory to field as well as the influence of indirect effects produced by species interactions. The differing results of the present field study and the previous laboratory study imply that lower- and higher-tier studies may not be mutually exclusive, but to be used in complementary.

Transgenic (cyp19a1b-GFP) zebrafish embryos as a tool for assessing combined effects of oestrogenic chemicals

Endocrine disrupting chemicals and especially oestrogen receptor (ER) agonists have been extensively studied over the years due to their potential effects on sexual development and reproduction in vertebrates, notably fish. As ER agonists can exist as complex mixtures in the aquatic environment, evaluating the impact of combined exposure on oestrogenic effects has become increasingly important. Use of predictive models such as concentration addition (CA) and independent action (IA) has allowed assessment of combined estrogenic effects of complex multi-compound mixtures of ER agonists in various fish in vitro and in vivo experimental models. The present work makes use of a transgenic zebrafish strain, tg(cyp19a1b-GFP), which expresses the green fluorescent protein (GFP) under the control of the cyp19a1b (brain aromatase or aromatase B) gene to determine the oestrogenic potency of ER agonists alone or in mixtures. In these studies, tg(cyp19a1b-GFP) zebrafish embryos were exposed for four days (from one to five days post fertilization) to five different oestrogenic chemicals; 17α-ethinylestradiol (EE2), 17β-estradiol (E2), estrone (E1), bisphenol A (BPA) and 4-tert-octylphenol (OP), and three mixtures of up to four of these compounds. The mixture of BPA, OP and E2 was also tested with primary cultures of rainbow trout hepatocytes by analysing the ER-mediated induction of the oestrogenic biomarker vitellogenin in order to compare the performance of the two methods for assessing oestrogenic effects of complex mixtures. The three tested mixtures were predominantly acting in an additive manner on the expression of GFP. Additivity was indicated by the overlap of the 95% confidence interval of the concentration response curves for the observed data with the CA and IA prediction models, and model deviation ratios within a factor of two for a majority of the mixture concentrations. However, minor deviations determined as more than additive effects for the mixture of EE2, E1 and E2 and less than additive effects for the mixture of BPA, OP, EE2 and E1 were observed at the higher mixture concentrations tested. The successful prediction of additivity by CA and IA in tg(cyp19a1b-GFP) zebrafish embryos and deviations at high mixture concentrations seemed to correspond well to results obtained in the rainbow trout hepatocyte assay. The present results clearly show the usefulness of combining predictive modelling and use of in vitro bioassays for rapid screening of oestrogenic effects of complex mixtures and environmental samples.

Single and joint effects of Zn and Cd on Porcellio scaber (Crustacea, Isopoda) exposed to artificially contaminated food

This study aimed at determining effects of Zn, Cd and their equitoxic mixtures on metal assimilation and food consumption of the terrestrial isopod Porcellio scaber, in relation to metal availability in the food. Cd was four times less water-extractable than Zn. Cd or Zn extractability was affected neither by metal concentration nor by the presence of the other metal. In single metal exposures, assimilation efficiency (AE) was up to five times higher for Cd than for Zn. In a mixture, AE of Cd significantly increased at low mixture concentrations and decreased at high mixture concentrations. AE of Zn significantly increased at intermediate mixture concentrations. Effects of the Zn and Cd mixture on food consumption were additive (28-day EC50,total=1.10TU; EC50,water-extractable=1.18TU) when based on total and water-extractable concentrations but antagonistic when related to internal metal concentrations in the isopods (EC50,internal=1.40TU).

Drinking water contaminants (arsenic, cadmium, lead, benzene, and trichloroethylene). 1. Interaction of contaminants with nutritional status on general performance and immune function in broiler chickens

The objective of this study was to examine possible interactions between drinking water contaminants and suboptimal nutritional status for performance and immune function in male broiler chickens. Experimental drinking water contained a mixture of arsenic, benzene, cadmium, lead, and trichloroethylene (TCE) at low concentrations (0.80, 1.3, 5.0, 6.7, and 0.65 ppm) and high concentrations (8.6, 13, 50, 67, and 6.5 ppm). These chemicals were selected because they are among the most common contaminants found in ground water near hazardous waste sites. The experimental diets included feed containing 50% added vitamins and minerals (V&M) and feed without added V&M. Increasing levels of drinking water contaminants and decreasing levels of V&M in diet resulted in significantly (P < or = 0.05) decreased water and feed intake, decreased weight gain, and suppression of natural, humoral, and cell-mediated immune response. In a paired-water study, feed consumption, body weight, and immune function were decreased in chickens provided low and high concentrations of the chemical mixture in drinking water compared with chickens given control drinking water equal to the volumes consumed by the chickens given the low and high concentration of mixture, respectively. A deficiency of dietary V&M caused increased sensitivity to adverse effects of drinking water contaminants.

Effect of Pollination Frequency and Pollen Concentration on Yield and Fruit Characteristics of Mechanically Pollinated Date Palm Trees (Phoenix dactyliferavar. Khalas)

Abstract An experiment was carried in Al-Ain region, the largest date production district of the United Arab Emirates (UAE), during the production years 1993 and 1994. The experiment tested the contribution of two pollen concentrations (1:10 and 1:5 on a volume basis) and three frequencies of mechanical application (two, three and four times) to yield and fruit characteristics of dates of variety Khalas (one of the most prestigious varieties in UAE) in comparison with the current manual application method (control). The experiment followed a balanced block design with eight replicates. Significant variations existed in yield and fruit characteristics among the various treatments. Although pollen concentration did not have a significant effect on these variations, the effect of the frequency of pollen application and its respective interaction with pollen concentration was significant; these results were consistent in both years. Highest average bunch mass was 5·66 kg resulting from pollinating the bunch twice with a 1:10 concentration but this was not significantly different from the average mass of bunches that were hand pollinated (control 5·46 kg). In general increased pollen application resulting from either higher mixture concentrations or more frequent applications resulted in more fruit drop and consequently a significant drop in yield.

Assessment of the Combined Effects of Substances: The Relationship between Concentration Addition and Independent Action

The assessment of the combined effects of substances is usually based on one of two different concepts: concentration addition or independent action. Both concepts are founded on different pharmacological assumptions about sites and modes of actions of substances, but in toxicology and ecotoxicology such knowledge is rare for most chemicals. In order to validate experimental results and to allow for precautious assessments, the quantitative relationships between concentration addition and independent action are therefore of interest. In this paper, we derive for the Weibull, the logistic, and the normal distribution functions the concentrations where the response probability due to concentration addition exceeds that due to independent action and vice versa. This is done (a) by analytically comparing both models for low and high mixture concentrations and (b) by numerically calculating the response probabilities when concentration addition and independent action agree. It is shown that the relationships between the models for joint action depend on the distribution functions, the corresponding slope parameters, and on the mixture concentrations administered.
