The gelatin-based liquid marbles for cell cryopreservation.

As an alternative and a straightforward cryopreservation biotechnological tool, liquid marble provides a promising cryopreservation approach. Currently, effective cell preservation mainly based on the addition of dimethyl sulfoxide (DMSO) and fetal bovine serum (FBS). As state-of-the-art cryoprotectant (CPA), DMSO, has intrinsic toxicity, which is the bottleneck of its widespread application. The complex compositions of FBS have the potential risks of pathogenic microorganism contamination. However, efficient cell cryopreservation using liquid marbles, a platform independent of DMSO and FBS, has not been well investigated yet. Herein, we explore the cryoprotection role of liquid marbles based on gelatin solution. Gelatin has a superior biocompatibility, which DMSO is incomparable. During a freeze-thaw cycle, gelatin produces negligible osmotic pressure, and has high ice recrystallization inhibition (IRI) activity to induce the formation of smaller and smooth ice crystals. Moreover, the specific structure of liquid marble also provides favorable supports for cell survival. The cryopreservation efficiency of mouse fibroblasts cells L929 via the gelatin-based liquid marble was as high as 90%, and the recovered cells could maintain their normal functionalities. This work opens a new window of opportunity for non-toxic and efficient cryopreservation of liquid marbles without the need of DMSO and FBS addition.

Design of an Ice Recrystallization-Inhibiting Polyampholyte-Containing Graft Polymer for Inhibition of Protein Aggregation.

Freezing-induced damage to proteins, through osmotic stress and ice recrystallization, during protein processing and long-term storage is a serious concern and may lead to loss of protein activity owing to denaturation. In this study, graft copolymers composed of a cryoprotective polymer (capable of preventing osmotic stress) and poly(vinyl alcohol) (PVA; known for its high ice recrystallization inhibition (IRI) property) were developed. The polymers had high IRI activity, albeit slightly lower than that of PVA alone, but substantially higher than that of succinylated ε-poly-l-lysine (PLLSA) alone. The graft polymers showed an efficiency higher than that of PVA or PLLSA alone in protecting proteins from multiple freeze-thaw cycles, as well as during prolonged freezing, indicating a synergy between PVA and PLLSA. The PLLSA-based graft polymer is a promising material for use in protein biopharmaceutics for the long-term storage of proteins under freezing conditions.

Spreading fully at the ice-water interface is required for high ice recrystallization inhibition activity

Although materials for ice recrystallization inhibition (IRI) are essential for the cryopreservation of cells and tissues, there exists no guiding mechanism for the design of such materials. Therefore, the construction of materials for IRI relies on the try-and-error strategy. Herein, through changing the tacticity of hydroxyl groups on poly(vinyl alcohol) (PVA) backbones with the affinities of PVAs to ice unchanged, we experimentally find IRI activity decreases significantly for isotactic PVA in comparison to that of atactic PVA. Molecular dynamics simulation shows atactic PVA spreads fully at the ice-water interface due to its much stronger interaction with water. This indicates atactic PVA can cover more ice surface and possess a higher IRI activity when the same amount of PVAs are used, which is consistent with the results that PVA can cover the same amount of ice surface more efficiently through experimentally measuring the adsorption of PVAs on the ice surface. A guiding mechanism of high active IRI materials can be obtained: only having affinity to ice is not enough to obtain high IRI activity ( i.e ., only small amount of materials is required to reduce the size of ice crystals to ca. (35± 10) μm), IRI agents must also have high affinity to water, i.e. , low interfacial energies, to both ice and water. The former is to guarantee the adsorption of the IRI agent on the ice surface, and the latter is required for the IRI agent to spread sufficiently at the ice-water interface. Therefore, each IRI molecule can effectively block the diffusion of water onto the ice surface, and consequently inhibits the growth of ice. A spreading coefficient of IRI agents is therefore introduced to quantitatively assess the capability of IRI agents to spread at the ice-water interface.

Complex Coacervate Core Micelles Containing Poly(vinyl alcohol) Inhibit Ice Recrystallization.

Complex coacervate core micelles (C3Ms) form upon complexation of oppositely charged copolymers. These co-assembled structures are widely investigated as promising building blocks for encapsulation, nanoparticle synthesis, multimodal imaging, and coating technology. Here, the impact on ice growth is investigated of C3Ms containing poly(vinyl alcohol), PVA, which is well known for its high ice recrystallization inhibition (IRI) activity. The PVA-based C3Ms are prepared upon co-assembly of poly(4-vinyl-N-methyl-pyridinium iodide) and poly(vinyl alcohol)-block-poly(acrylic acid). Their formation conditions, size, and performance as ice recrystallization inhibitors are studied. It is found that the C3Ms exhibit IRI activity at PVA monomer concentrations as low as 1 × 10-3 m. The IRI efficacy of PVA-C3Ms is similar to that of linear PVA and PVA graft polymers, underlining the influence of vinyl alcohol monomer concentration rather than polymer architecture.

Synthesis of Degradable Poly(vinyl alcohol) by Radical Ring-Opening Copolymerization and Ice Recrystallization Inhibition Activity.

Poly(vinyl alcohol) (PVA) is the most active synthetic mimic of antifreeze proteins and has extremely high ice recrystallization inhibition (IRI) activity. Addition of PVA to cellular cryopreservation solutions increases the number of recovered viable cells due to its potent IRI, but it is intrinsically nondegradable in vivo. Here we report the synthesis, characterization, and IRI activity of PVA containing degradable ester linkages. Vinyl chloroacetate (VClAc) was copolymerized with 2-methylene-1,3-dioxepane (MDO) which undergoes radical ring-opening polymerization to install main-chain ester units. The use of the chloroacetate monomer enabled selective deacetylation with retention of esters within the polymer backbone. Quantitative IRI assays revealed that the MDO content had to be finely tuned to retain IRI activity, with higher loadings (24 mol %) resulting in complete loss of IRI activity. These degradable materials will help translate PVA, which is nontoxic and biocompatible, into a range of biomedical applications.

Ice and anti-nucleating activities of an ice-binding protein from the annual grass, Brachypodium distachyon.

Plants exposed to sub-zero temperatures face unique challenges that threaten their survival. The growth of ice crystals in the extracellular space can cause cellular dehydration, plasma membrane rupture and eventual cell death. Additionally, some pathogenic bacteria cause tissue damage by initiating ice crystal growth at high sub-zero temperatures through the use of ice-nucleating proteins (INPs), presumably to access nutrients from lysed cells. An annual species of brome grass, Brachypodium distachyon (Bd), produces an ice-binding protein (IBP) that shapes ice with a modest depression of the freezing point (~0.1 °C at 1 mg/mL), but high ice-recrystallization inhibition (IRI) activity, allowing ice crystals to remain small at near melting temperatures. This IBP, known as BdIRI, is unlike other characterized IBPs with a single ice-binding face, as mutational analysis indicates that BdIRI adsorbs to ice on two faces. BdIRI also dramatically attenuates the nucleation of ice by bacterial INPs (up to -2.26 °C). This 'anti-nucleating' activity is significantly higher than previously documented for any IBP.

Antifreeze proteins enable plants to survive in freezing conditions.

Overwintering plants secrete antifreeze proteins (AFPs) to provide freezing tolerance. These proteins bind to and inhibit the growth of ice crystals that are formed in the apoplast during subzero temperatures. Antifreeze activity has been detected in more than 60 plants and AFPs have been purified from 15 of these, including gymnosperms, dicots and monocots. Biochemical characterization of plant antifreeze activity, as determined by the high ice recrystallization inhibition (IRI) activities and low thermal hysteresis (TH) of AFPs, showed that their main function is inhibition of ice crystal growth rather than the lowering of freezing temperatures. However, recent studies showed that antifreeze activity with higher TH also exists in plants. Calcium and hormones like ethylene and jasmonic acid have been shown to regulate plant antifreeze activity. Recent studies have shown that plant AFPs bind to both prism planes and basal planes of ice crystals by means of two flat ice binding sites. Plant AFPs have been postulated to evolve from the OsLRR-PSR gene nearly 36 million years ago. In this review, we present the current scenario of plant AFP research in order to understand the possible potential of plant AFPs in generation of freezing-tolerant crops.

A Theoretical Model of a Plant Antifreeze Protein from Lolium perenne

Antifreeze proteins (AFPs), found in certain organisms enduring freezing environments, have the ability to inhibit damaging ice crystal growth. Recently, the repetitive primary sequence of the AFP of perennial ryegrass, Lolium perenne, was reported. This macromolecular antifreeze has high ice recrystallization inhibition activity but relatively low thermal hysteresis activity. We present here a theoretical three-dimensional model of this 118-residue plant protein based on a β-roll domain with eight loops of 14–15 amino acids. The fold is supported by a conserved valine hydrophobic core and internal asparagine ladders at either end of the roll. Our model, which is the first proposed for a plant AFP, displays two putative, opposite-facing, ice-binding sites with surface complementarity to the prism face of ice. The juxtaposition of the two imperfect ice-binding surfaces suggests an explanation for the protein’s inferior thermal hysteresis but superior ice recrystallization inhibition activity and activity when compared with fish and insect AFPs.
