Mercury Speciation and Stable Isotopes in Emperor Penguins: First Evidence for Biochemical Demethylation of Methylmercury to Mercury-Dithiolate and Mercury-Tetraselenolate Complexes

Apex marine predators, such as toothed whales and large petrels and albatrosses, ingest mercury (Hg) primarily in the form of methylmercury (MeHg) via prey consumption, which they detoxify as tiemannite (HgSe). However, it remains unclear how lower trophic level marine predators, termed mesopredators, with elevated Hg concentrations detoxify MeHg and what chemical species are formed. To address this need, we used high energy-resolution X-ray absorption near edge structure spectroscopy paired with nitrogen (N) and Hg stable isotopes to identify the chemical forms of Hg, Hg sources, and species-specific δ202Hg isotopic values in emperor penguin, a mesopredator feeding primarily on Antarctic silverfish. The penguin liver contains variable proportions of MeHg and two main inorganic Hg complexes (IHg), Hg-dithiolate (Hg(SR)2) and Hg-tetraselenolate (Hg(Sec)4), each characterized by specific isotopic values (δ202MeHg = 0.3 ± 0.2‰, δ202Hg(SR)2 = −1.6 ± 0.2‰, δ202Hg(Sec)4 = −2.0 ± 0.1‰). Using δ15N as a tracer of food source, we show that Hg(SR)2 is likely not obtained through dietary intake, but rather is present as a biochemical demethylation product. Furthermore, on average, female penguins transferred Hg to the egg strictly as MeHg in the egg albumen but as mixtures of MeHg and IHg in the membrane (89% and 11%, respectively) and yolk (32% MeHg and 68% Hg(Sec)4). Despite IHg species in eggs, MeHg is still the main species quantitatively transferred by the mother to the chick because of the disproportionate mass of the MeHg-rich albumen compared to the yolk. This work highlights the transformation of MeHg to Hg(SR)2 during demethylation for the first time in multicellular organisms, but further work is needed to understand the formation of Hg(SR)2 in the presence of relatively abundant Se biomolecules in lower trophic level predator species.

U(V) Stabilization via Aliovalent Incorporation of Ln(III) into Oxo-salt Framework.

Pentavalent uranium compounds are key components of uranium's redox chemistry and play important roles in environmental transport. Despite this, well-characterized U(V) compounds are scarce primarily because of their instability with respect to disproportionation to U(IV) and U(VI). In this work, we provide an alternate route to incorporation of U(V) into a crystalline lattice where different oxidation states of uranium can be stabilized through the incorporation of secondary cations with different sizes and charges. We show that iriginite-based crystalline layers allow for systematically replacing U(VI) with U(V) through aliovalent substitution of 2+ alkaline-earth or 3+ rare-earth cations as dopant ions under high-temperature conditions, specifically Ca(UVIO2)W4O14 and Ln(UVO2)W4O14 (Ln=Nd, Sm, Eu, Gd, Yb). Evidence for the existence of U(V) and U(VI) is supported by single-crystal X-ray diffraction, high energy resolution X-ray absorption near edge structure, X-ray photoelectron spectroscopy, and optical absorption spectroscopy. In contrast with other reported U(V) materials, the U(V) single crystals obtained using this route are relatively large (several centimeters) and easily reproducible, and thus provide a substantial improvement in the facile synthesis and stabilization of U(V).

Detection and Characterization of Hydride Ligands in Copper Complexes by Hard X-Ray Spectroscopy.

Transition metal complexes, particularly copper hydrides, play an important role in various catalytic processes and molecular inorganic chemistry. This study employs synchrotron hard X-ray spectroscopy to gain insights into the geometric and electronic properties of copper hydrides as potential catalysts for CO2 hydrogenation. The potential of high energy resolution X-ray absorption near-edge structure (HERFD-XANES) and valence-to-core X-ray emission (VtC-XES) is demonstrated with measurement on Stryker's reagent (Cu6H6) and [Cu3(μ3-H)(dpmppe)2](PF6)2 (Cu3H), alongside a non-hydride copper compound ICu(dtbppOH) (Cuy-I). The XANES analysis reveals that coordination geometries strongly influence the spectra, providing only indirect details about hydride coordination. The VtC-XES analysis exhibits a distinct signal around 8975 eV, offering a diagnostic tool to identify hydride ligands. Theoretical calculations support and extend these findings by comparing hydride-containing complexes with their hydride-free counterparts.

Unveiling Hidden Shake-Up Features in the Uranyl M4-Edge Spectrum.

The M4,5-edge high energy resolution X-ray absorption near-edge structure (HR-XANES) spectra of actinyls offer valuable insights into the electronic structure and bonding properties of heavy-element complexes. To conduct a comprehensive spectral analysis, it is essential to employ computational methods that accurately account for relativistic effects and electron correlation. In this work, we utilize variational relativistic multireference configurational interaction methods to compute and analyze the X-ray M4-edge absorption spectrum of uranyl. By employing these advanced computational techniques, we achieve excellent agreement between the calculated spectral features and experimental observations. Moreover, the calculations unveil significant shake-up features, which arise from the intricate interplay between strongly correlated 3d core-electron and ligand excitations. This research provides important theoretical insights into the spectral characteristics of heavy-element complexes. Furthermore, it establishes the foundation for utilizing M4,5-edge spectroscopy as a means to investigate the chemical activities of such complexes. By leveraging this technique, we can gain a deeper understanding of the bonding behavior and reactivity of heavy-element compounds.

Improvement of Oxygen Reduction Reaction Performance of Fe-N-C Catalyst

Fuel cells are gaining worldwide attention as an acceptable technology for achieving carbon neutrality. However, existing fuel cells use large amounts of precious metals, which limits their widespread adoption. Precious metal-free fuel cells have been studied in our laboratory. The oxidation-reduction reaction (ORR) at the cathode of the fuel cell must start at a higher potential and proceed smoothly in a four-electron reduction reaction without producing hydrogen peroxide1,2. We have synthesized iron-based complex catalysts using the method reported by Plamen Atanassov3 et al. Iron nitrate dihydrate is mixed in aqueous solution with glucose, 2-methylimidazole, and zinc nitrate hexahydrate, followed by hydrothermal synthesis at 200°C for 24 hours. It is then acid-treated with nitric acid to dissolve excess metallic iron, and the process is repeated twice at 950°C to complete the Fe-N-C catalyst. However, SEM and XRD analysis confirmed the presence of large amounts of metallic iron. In response to these results, the synthetic process of the Fe-N-C catalyst was repeatedly improved, and the prepared catalyst was subjected to in-situ XAFS (X-ray absorption fine structure) analysis using synchrotron radiation at SPring-8 (Fig. 1). The peak around 7110 eV is significantly different from that of metallic iron (Fe foil), indicating that iron complex, iron phthalocyanine (II), and it is like the peak of iron phthalocyanine (II), confirming that the prepared Fe-N-C catalyst forms a good complex. The performance of the Fe-N-C catalyst with the confirmed complexation was then investigated, and a cathode that exhibited ORR performance comparable to that of the Pt catalyst was successfully synthesized (Fig. 2). In the future, further improvement of the catalyst preparation process will be discussed to develop a catalyst with much better performance than Pt.Reference 1N. Yamamoto, D.Matsumura, Y. Experimental and Theoretical Investigation of High-Resolution X-ray Absorption Spectroscopy (HR-XAS) at the Cu K-Edge for Cu2ZnSnSe4

Energy sustainability is critical for social activities in the human world. The quaternary compound Cu2ZnSnSe4 (CZTSe), as a promising candidate for thin-film solar cell absorption with medium-level thermoelectric performance, is of interest for the purpose of utilizing solar energy. The defect chemistry and atomic ordering in this particular compound also triggers interests in understanding its crystallographic structure as well as defects. Hereby, high energy resolution X-ray absorption spectroscopy is employed to investigate the electronic and geometric structural complexity in pristine and cobalt-doped Cu2ZnSnSe4. The occupational atomic sites of Cu are found to be mixed with the Zn atoms, forming CuZn anti-defects, which serve as a knob to tune local electronic structures. With proper doping, the band structure can be manipulated to improve the optical and thermoelectric properties of the CZTSe compounds.

Investigation of hydrogen superoxide adsorption during ORR on Pt/C catalyst in acidic solution for PEFC by in-situ high energy resolution XAFS

Oxygen Reduction Reaction (ORR) is one of the key electrochemical reactions, especially in Polymer Electrolyte Fuel Cells (PEFCs). In this work, the reaction pathway and intermediate species such as molecular-type oxygen during ORR were discussed. High energy resolution X-ray absorption fine structure spectra at the Pt L3-edge were obtained to observe the electronic structure of Pt catalyst. Carbon-supported Pt nanoparticles as powder catalysts were used for measurement as more realistic conditions, and the species adsorbed on the Pt nanoparticles were analyzed while controlling under the potentials in acidic electrolyte solution with N2 or O2 gas bubbling. Molecular-type oxygen adsorption species is observed only in the O2-bubbled electrolyte solution. This intermediate adsorption species is assigned to a hydrogen superoxide.

Fossil Bioapatites with Extremely High Concentrations of Rare Earth Elements and Yttrium from Deep-Sea Pelagic Sediments

Detailed geochemical and mineralogical insights into some of the richest rare earth elements and yttrium (REY)-containing bioapatites from ocean-floor sediments have been provided by combining laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS), electron probe microanalysis (EPMA), Rietveld X-ray diffraction analysis, and Ce L3-edge high energy-resolution X-ray absorption near edge structure (HR-XANES) spectroscopy. Bioapatites at 1.94 and 4.70 m below the seafloor (mbsf) of the Clarion-Clipperton Zone (CCZ) of the Pacific Ocean have 26,600 (RSD = 15.7%, n = 20) and 30,300 (RSD = 14.6%, n = 10) mg/kg (mg/kg) total REY, respectively, and bioapatites at 2.28 and 6.95 mbsf of the Peru Basin have 15,500 (RSD = 15.6%, n = 20) and 15,700 (RSD = 17.8%, n = 29) mg/kg total REY, respectively. All bioapatite specimens have a variety of isomorphic substitutions in all atomic positions of the crystallographic structure. The average crystallochemical formula of bioapatites at 6.95 mbsf of the Peru Basin is [(PO4)2.71(SiO4)0.04(CO3,SO4)0.25][Ca4.57Na0.29Y0.04][F0.87Cl0.21]. All other substituents are below 0.04 atoms per formula unit. HR-XANES provides the first direct evidence for trivalent Ce in sediment apatites. The strong negative geochemical anomaly of Ce in fossil bioapatites is well explained by the occurrence of four valent Ce-MnO2 and CeO2 within the sediment and in seafloor ferromanganese nodules.

Paving the way for examination of coupled redox/solid-liquid interface reactions: 1 ppm Np adsorbed on clay studied by Np M5-edge HR-XANES spectroscopy

The recently emerged actinide (An) M4,5-edge high-energy resolution X-ray absorption near-edge structure (HR-XANES) technique has proven to be very powerful for oxidation state studies of actinides. In this work, for the first time, Np M5-edge HR-XANES was applied to study Np sorption on illite. By improving the experimental conditions, notably by operation of the spectrometer under He atmosphere, it was possible to measure Np M5-edge HR-XANES spectra of a sample with ≈ 1 μg Np/g illite (1 ppm). This is 30–2000 times lower than Np loadings on mineral surfaces usually investigated by X-ray absorption spectroscopy. A newly designed cryogenic configuration enabled sample temperatures of 141.2 ± 1.5 K and successfully prevented beam-induced changes of the Np oxidation state. The described approach paves the way for the examination of coupled redox/solid-liquid interface reactions of actinide ions via An M4,5-edge HR-XANES spectroscopy at low metal ion concentrations, which are of specific relevance for contaminated sites and nuclear waste disposal studies.

Effect of manganese on the speciation of neptunium(V) on manganese doped magnetites

Speciation and fate of radionuclides in environment are strongly dependent on their interaction with various mineral surfaces. Presence of impurity atoms in minerals modifies these interactions. 237Np, a long-lived component of nuclear high-level waste, is highly mobile in environment as neptunyl (NpO2+) cation. Role of impurities in defining its interaction with mineral surfaces present in natural and engineered systems is yet to be completely understood. Manganese is a ubiquitous impurity present in naturally occurring iron corrosion product, magnetite. Here, a series of manganese (Mn)-doped magnetites (FM) was synthesized and the evolution of solid-sorbed speciation of Np(V) was investigated as a function of Mn doping (3–11 wt%) in near neutral pH condition. Combining Mn K-edge and Np L3-edge X-ray absorption fine structure spectra with Np M5-edge high energy-resolution X-ray absorption near-edge structure spectra, we found nanoparticulate NpIVO2 solid as the single speciation of Np sorbed on both doped and undoped magnetites. Without changing the surface speciation of Np, Mn-O sites did show preference for sorbing Np(V) vis-á-vis Fe-O sites. Mn participates through Fe(II)/Mn(III) redox couple in reducing sorbed Np(V). Its participation, however, becomes significant only when the doped-magnetite contains a critical minimum concentration (> 6 wt%) of Mn. This study highlights the significant role of impurity atom wherein it interacts with sorbing species through redox process.

Computational and Spectroscopic Tools for the Detection of Bond Covalency in Pu(IV) Materials.

Plutonium is used as a major component of new-generation nuclear fuels and of radioisotope batteries for Mars rovers, but it is also an environmental pollutant. Plutonium clearly has high technological and environmental importance, but it has an extremely complex, not well-understood electronic structure. The level of covalency of the Pu 5f valence orbitals and their role in chemical bonding are still an enigma and thus at the frontier of research in actinide science. We performed fully relativistic quantum chemical computations of the electronic structure of the Pu4+ ion and the PuO2 compound. Using four different theoretical tools, it is shown that the 5f orbitals have very little covalent character although the 5f(7/2) a2u orbital with the highest orbital energy has the greatest extent of covalency in PuO2. It is illustrated that the Pu M4,5 edge high-energy resolution X-ray absorption near-edge structure (Pu M4,5 HR-XANES) spectra cannot be interpreted in terms of dipole selection rules applied between individual 3d and 5f orbitals, but the selection rules must be applied between the total wavefunctions for the initial and excited states. This is because the states cannot be represented by single determinants. They are shown to involve major redistributions on the 5f electrons over the different 5f orbitals. These redistributions could be viewed as shake-up-like excitations in the 5f shell from the lowest orbital energy from J = 5f(5/2) into higher orbital energy J = 5f(7/2). We show that the second peak in the Pu M4 edge and the high-energy shoulder of the Pu M5 edge HR-XANES spectra probe the 5f(7/2) a2u orbital; thus, these spectral features are expected to change upon bond variations. We describe theoretical and spectroscopy tools, which can be applied for all actinide elements in materials with cubic structure.

Gold speciation in hydrothermal fluids revealed by in situ high energy resolution X-ray absorption spectroscopy

Abstract Gold mobilization, transfer, and concentration in the Earth’s crust are controlled by hydrothermal sulfur- and chloride-bearing fluids. Yet the exact chemical identity, structure, and stability of Au-bearing species and, in particular, the respective contributions of the sulfide (HS−) and trisulfur ion (S3⋅−) ligands to Au transport lack direct in situ evidence. Here we employed high energy resolution fluorescence detection X-ray absorption spectroscopy (HERFD-XAS) on aqueous sulfate/sulfide/S3⋅−-bearing solutions at typical hydrothermal temperatures and pressures (T = 350 °C, P = 600 bar) to reveal differences in dissolved Au spectral signatures indicative of contrasting fluid-phase Au speciation as a function of acidity and redox conditions. Combined with in situ Au solubility measurements and quantum-chemical and thermodynamic modeling, our spectroscopic data provide direct evidence for the Au(HS)S3− and Au(HS)2− complexes predominant at acidic-to-neutral and alkaline conditions, respectively. Our findings thus directly confirm a recent speciation scheme for Au in aqueous S-bearing fluids established using less direct methods, and highlight an important role of the trisulfur ion in gold mobilization and concentration in hydrothermal-magmatic deposits associated with subduction zones. More generally, our results show that HERFD-XAS enables the identification of structural and coordination features in metal complexes virtually unresolvable using classical XAS techniques. By avoiding limitations of less direct techniques, our integrated high-resolution spectroscopic approach opens perspectives for studies of the speciation and solubility of gold and other metals in high T-P fluids, and potentially silicate melts, inaccessible to direct observation in nature.

Structure-Spectroscopy Correlations for Intermediate Q of Soluble Methane Monooxygenase: Insights from QM/MM Calculations.

The determination of the diiron core intermediate structures involved in the catalytic cycle of soluble methane monooxygenase (sMMO), the enzyme that selectively catalyzes the conversion of methane to methanol, has been a subject of intense interest within the bioinorganic scientific community. Particularly, the specific geometry and electronic structure of the intermediate that precedes methane binding, known as intermediate Q (or MMOHQ), has been debated for over 30 years. Some reported studies support a bis-μ-oxo-bridged Fe(IV)2O2 closed-core conformation Fe(IV)2O2 core, whereas others favor an open-core geometry, with a longer Fe–Fe distance. The lack of consensus calls for a thorough re-examination and reinterpretation of the spectroscopic data available on the MMOHQ intermediate. Herein, we report extensive simulations based on a hybrid quantum mechanics/molecular mechanics approach (QM/MM) approach that takes into account the complete enzyme to explore possible conformations for intermediates MMOHox and MMOHQ of the sMMOH catalytic cycle. High-level quantum chemical approaches are used to correlate specific structural motifs with geometric parameters for comparison with crystallographic and EXAFS data, as well as with spectroscopic data from Mössbauer spectroscopy, Fe K-edge high-energy resolution X-ray absorption spectroscopy (HERFD XAS), and resonance Raman 16O–18O difference spectroscopy. The results provide strong support for an open-core-type configuration in MMOHQ, with the most likely topology involving mono-oxo-bridged Fe ions and alternate terminal Fe-oxo and Fe-hydroxo groups that interact via intramolecular hydrogen bonding. The implications of an open-core intermediate Q on the reaction mechanism of sMMO are discussed.

Isotope Fractionation from In Vivo Methylmercury Detoxification in Waterbirds

Here, we used high energy-resolution X-ray absorption near edge structure spectroscopy and isotope ratios of total mercury (δ202THg) and methylmercury (δ202MeHg) to elucidate the chemical speciation of Hg and the resultant MDF as a result of internal MeHg demethylation in waterbirds. In three waterbirds (Clark's grebe, Forster's tern, and south polar skua), between 17 and 86% of MeHg was demethylated to inorganic mercury (iHg) species primarily in the liver and kidneys as Hg-tetraselenolate [Hg(Sec)4] and minor Hg-dithiolate [Hg(SR)2] complexes. Tissular differences between δ202THg and δ202MeHg correlated linearly with %iHg [Hg(Sec)4 + Hg(SR)2] and were interpreted to reflect a kinetic isotope effect during in vivo MeHg demethylation. The product-reactant isotopic enrichment factor (ϵp/r) for the demethylation of MeHg → Hg(Sec)4 was -2.2 ± 0.1‰. δ202MeHg values were unvarying within each bird, regardless of Hg(Sec)4 abundance, indicating fast internal cycling or replenishment of MeHg relative to demethylation. Our findings document a universal selenium-dependent demethylation reaction in birds, provide new insights on the internal transformations and cycling of MeHg and Hg(Sec)4, and allow for mathematical correction of δ202THg values as a result of the MeHg → Hg(Sec)4 reaction.

Chemical Forms of Mercury in Blue Marlin Billfish: Implications for Human Exposure

Although fish is an important source of nutrients, including some of the healthiest proteins, long-chain fatty acids, and essential selenium, species at the top of the food chain frequently contain large amounts of toxic mercury (Hg). The provisional tolerable weekly intake (PTWI) of Hg from fish consumption is calculated from the total concentration of Hg and assuming that all Hg is speciated as organic methylmercury (MeHg). Using high energy-resolution X-ray absorption near-edge structure (HR-XANES) spectroscopy, we show that blue marlin (Makaira sp.), a common top predator consumed by humans, contains high concentrations of inorganic Hg(II) complexed as 57 ± 10% Hg-tetraselenolate [Hg(Sec)4] and 43 ± 10% tiemannite (HgSe). The stable Hg–Se chemical bond likely attenuates the bioavailability of Hg and counteracts some of its health hazards to consumers. Thus, monitoring the concentration of MeHg, rather than total Hg, in top predators such as marlin would provide a more robust measure of potential Hg exposure and may be sufficient for food safety controls. The bonding of Hg atoms to four selenocysteine (Sec) residues in the Hg(Sec)4 complex severely depletes the stock of bioavailable Se, and quantification shows that blue marlin is not a chief source of dietary Se essential to selenoenzyme synthesis and activity.

High Energy Resolution Fluorescence Detection of the Pt L3,2-Edge Whitelines of Pt-Based Bimetallic Systems: Implications for the Pt 5d5/2,3/2 Density of States

This study utilizes the state-of-the-art high energy resolution fluorescence detection (HERFD) and X-ray absorption fine-structure spectroscopy (XAFS) to revisit the Pt L3- and L2-edge whitelines (WLs) for Pt metal and to explore the Pt L3,2-edge WL–5d5/2,3/2 density of states (DOS) relationship for multiple Pt-Ni and Pt-Cu bimetallic fcc systems. A generalized linear correlation of the WL area with the unoccupied 5d states has been established. The unoccupied Pt 5d5/2 and 5d3/2 state counts are found to be correlated linearly despite the diversity of the systems under investigation. This finding allows for a straightforward and effective way to obtain the number of unoccupied Pt 5d states by only measuring the Pt L3-edge in these or similar systems. This observation also introduces a new spin on the Pt L3,2-edge WL intensity anomaly in that the Pt 5d3/2 states are not as full as previously thought. Guidance for the XAFS experiment design and charge redistribution implications using this correlation are discussed.

In Vivo Formation of HgSe Nanoparticles and Hg-Tetraselenolate Complex from Methylmercury in Seabirds-Implications for the Hg-Se Antagonism.

In vivo and in vitro evidence for detoxification of methylmercury (MeHg) as insoluble mercury selenide (HgSe) underlies the central paradigm that mercury exposure is not or little hazardous when tissue Se is in molar excess (Se:Hg > 1). However, this hypothesis overlooks the binding of Hg to selenoproteins, which lowers the amount of bioavailable Se that acts as a detoxification reservoir for MeHg, thereby underestimating the toxicity of mercury. This question was addressed by determining the chemical forms of Hg in various tissues of giant petrels Macronectes spp. using a combination of high energy-resolution X-ray absorption near edge structure and extended X-ray absorption fine structure spectroscopy, and transmission electron microscopy coupled to elemental mapping. Three main Hg species were identified, a MeHg-cysteinate complex, a four-coordinate selenocysteinate complex (Hg(Sec)4), and a HgSe precipitate, together with a minor dicysteinate complex Hg(Cys)2. The amount of HgSe decreases in the order liver > kidneys > brain = muscle, and the amount of Hg(Sec)4 in the order muscle > kidneys > brain > liver. On the basis of biochemical considerations and structural modeling, we hypothesize that Hg(Sec)4 is bound to the carboxy-terminus domain of selenoprotein P (SelP) which contains 12 Sec residues. Structural flexibility allows SelP to form multinuclear Hgx(Se,Sec)y complexes, which can be biomineralized to HgSe by protein self-assembly. Because Hg(Sec)4 has a Se:Hg molar ratio of 4:1, this species severely depletes the stock of bioavailable Se for selenoprotein synthesis and activity to one μg Se/g dry wet in the muscle of several birds. This concentration is still relatively high because selenium is naturally abundant in seawater, therefore it probably does not fall below the metabolic need for essential selenium. However, this study shows that this may not be the case for terrestrial animals, and that muscle may be the first tissue potentially injured by Hg toxicity.

Demethylation of Methylmercury in Bird, Fish, and Earthworm.

Toxicity of methylmercury (MeHg) to wildlife and humans results from its binding to cysteine residues of proteins, forming MeHg-cysteinate (MeHgCys) complexes that hinder biological functions. MeHgCys complexes can be detoxified in vivo, yet how this occurs is unknown. We report that MeHgCys complexes are transformed into selenocysteinate [Hg(Sec)4] complexes in multiple animals from two phyla (a waterbird, freshwater fish, and earthworms) sampled in different geographical areas and contaminated by different Hg sources. In addition, high energy-resolution X-ray absorption spectroscopy (HR-XANES) and chromatography-inductively coupled plasma mass spectrometry of the waterbird liver support the binding of Hg(Sec)4 to selenoprotein P and biomineralization of Hg(Sec)4 to chemically inert nanoparticulate mercury selenide (HgSe). The results provide a foundation for understanding mercury detoxification in higher organisms and suggest that the identified MeHgCys to Hg(Sec)4 demethylation pathway is common in nature.

Mercury in the tissues of five cephalopods species: First data on the nervous system

Mercury (Hg), one of the elements most toxic to biota, accumulates within organisms throughout their lifespan and biomagnifies along trophic chain. Due to their key role in marine systems, cephalopods constitute a major vector of Hg in predators. Further, they grow rapidly and display complex behaviours, which can be altered by neurotoxic Hg. This study investigated Hg concentrations within 81 cephalopod specimens sampled in the Bay of Biscay, which belonged to five species: Eledone cirrhosa, Sepia officinalis, Loligo vulgaris, Todaropsis eblanae and Illex coindetii. Hg concentrations were measured in the digestive gland, the mantle muscle and the optic lobes of the brain. The digestive gland and the mantle were tissues with the most concentrated Hg among all species considered (up to 1.50 μg.g−1 dw), except E. cirrhosa. This benthic cephalopod had 1.3-fold higher Hg concentrations in the brain (up to 1.89 μg.g−1 dw) than in the mantle, while other species had 2-fold lower concentrations of Hg in the brain than in the mantle. Brain-Hg concentrations can be predicted from muscle-Hg concentrations for a given species, which facilitates the assessment of Hg toxicokinetics in cephalopods. In the most contaminated E. cirrhosa individual, the chemical form of Hg in its digestive gland, mantle muscle and optic lobes, was determined using High energy-Resolution X-ray Absorption Near Edge Structure (HR XANES) spectroscopy. In the digestive gland, 33 ± 11% of total Hg was inorganic Hg speciated as a dicysteinate complex (Hg(Cys)2), which suggested that the demethylation of dietary MeHg occurs in this organ. All Hg found in the mantle muscle and the optic lobes is methylated and bound to one cysteinyl group (MeHgCys complex), which implies that dietary MeHg is distributed to these tissues via the bloodstream. These results raised the questions regarding interspecific differences observed regarding Hg brain concentrations and the possible effect of Hg on cephalopod functional brain plasticity and behaviour.
