Role of phytoplankton composition on exudation of dissolved organic carbon in the Bay of Bengal

Phytoplankton in the marine environment exudates part of the primary production as dissolved organic carbon (DOC) into the surrounding waters. The rate of exudation of DOC is depended on the hydrographic condition, nutrient availability, phytoplankton composition, and their size structure. To examine this, samples were collected from the coastal and offshore regions where different hydrographic conditions exists in the Bay of Bengal. The coastal waters were relatively low saline, rich in inorganic nutrients, high phytoplankton biomass (Chl-a) and primary production in the coastal compared to offshore regions. The photic zone integrated Chl-a displayed insignificant difference between coastal and offshore regions whereas higher photic zone integrated primary production was observed in the former than latter region. Dominance of microplankton was observed in the coastal waters associating with high inorganic nitrogen concentrations, in contrast, picoplankton dominated in the offshore region associating with low inorganic nitrogen but high organic nitrogen concentrations. Due to high surface-to-volume ratio of picoplankton, ∼45% of the primary production exudated as DOC in the offshore compared to the coastal region (∼12%) due to dominance of microplankton. The sum of primary production and DOC exudation, called total primary production, was almost equal in the coastal and offshore regions. The mean phytoplankton biomass normalized primary production (pB) in the Bay of Bengal was low (17 ± 8 mgC mgChl-a−1 d−1) compared to Arabian Sea (37 ± 5 mgC mgChl-a−1 d−1). In contrast pB estimated based on total primary production is close (30 ± 16 mgC mgChla−1 d−1) to that of the Arabian Sea (37 ± 5 mgC mgChl-a−1 d−1) suggesting that the Bay of Bengal is equally productive compared to that of Arabian Sea than hitherto hypothesized due to strong stratification and less nutrients input in the former basin. The low sinking carbon flux observed below the photic zone is attributed to the release of primary production as DOC. The released DOC from the phytoplankton may support heterotrophic activity as evidenced by the existence of strong net heterotrophy in the Bay of Bengal. This may lead to the time lag between primary and export productions. None of these processes were incorporated into the numerical models resulting in inaccurate simulations of carbon cycling in the northern Indian Ocean. Modifications in the models by incorporating these processes may improve model simulations for a better understanding the modifications in biogeochemical processes due to climate change in the Bay of Bengal.

Cultivation of Microalgae Spirulina platensis Biomass Using Palm Oil Mill Effluent for Phycocyanin Productivity and Future Biomass Refinery Attributes

Palm oil mill effluent is a type of wastewater which contains a high concentration of organic nitrogen, phosphorus, and different supplement substances. These substances support and enhance the growth of microalgae. Cyanobacteria Spirulina platensis is a blue-green microalga with two phycobiliproteins as its primary constituents. Phycocyanin is a natural blue colourant used in biomedicines, cosmetics, diagnostics, treatments, and environmental protection. Chlorophyll pigment plays a significant role in photosynthesis where the photosynthetic process uses atmospheric carbon to produce starch and lipid which can eventually be converted into desirable products such as biodiesel. Therefore, in this study, Spirulina platensis was cultivated in different concentrations of diluted POME (10-50%, v / v ) at 26 ± 1 °C room temperature, 90 μmol photon m-2 s-1, and aerated for 24 h continuously. The growth of Spirulina platensis was monitored through optical density at 680 nm for 15 days. The highest biomass yield obtained in the control medium and 30% POME medium were 1.27 ± 0.02 and 1.16 ± 0.01 g/L, respectively. The highest phycocyanin yield obtained from the biomass harvested from 30% nutrient media was 175.12 ± 22.32 mg and followed by 163.55 ± 20.15 mg from control media with purified phycocyanin of 87.11 ± 5.20 mg and 85.31 ± 4.33 mg, respectively. The peak properties of phycocyanin such as the amide group at 1655.17 cm-1 (C=O stretching), FT-IR analysis revealed well-formed Spirulina platensis with all characteristic peaks and distinct fingerprints of phycocyanin. The ultrasound method produces the highest lipid yield ( 28.6 ± 0.7 %) which consists of stearic (38.45%), palmitoleic ( 25.72 ± 0.02 ), and palmitic ( 17.71 ± 0.04 ) fatty acid methyl esters. The FAME produced from the extracted lipid has the potential to be used in biodiesel applications. Since POME contains the essential nutrients which can support the growth of Spirulina platensis in the optimum environment for biomass and lipid productivity, it revealed the potential for biodiesel production.

Nitrogen Sources Affect the Long-Chain Polyunsaturated Fatty Acids Content in Thraustochytrium sp. RT2316-16.

The psychrophilic marine microorganism Thraustochytrium sp. RT2316-16 can produce carotenoids as well as lipids containing the omega-3 polyunsaturated fatty acids (PUFA) eicosapentaenoic acid and docosahexaenoic acid. This work reports on the effects of the composition of the culture medium, including certain amino acids, on growth and lipid synthesis by RT2316-16. Compared with the culture on glutamate, the use of lysine, alanine, or serine, increased the content of the omega-3 PUFA in total lipids. In the media that contained yeast extract, glutamate, and glucose, lipid accumulation occurred when organic ammonium was exhausted earlier than glucose. In contrast, lipid mobilization was promoted if glucose was exhausted while organic ammonium (supplied by yeast extract and glutamate) remained in the medium. The total content of carotenoids in the lipid-free biomass decreased during the first 12 to 24 h of culture, simultaneously with a decrease in the total lipid content of the biomass. The experimental data suggested a possible interrelationship between the metabolism of carotenoids and lipids. A high content of omega-3 PUFA in the total lipids could be obtained by growing the thraustochytrid in a medium with a low glucose concentration (6 g L-1) and a high concentration of organic nitrogen (yeast extract 12 g L-1; glutamate 1.06 g L-1), after glucose was exhausted. These observations may guide the development of a strategy to enhance omega-3 PUFA in the biomass.

Peat soils contain high concentrations of organic nitrogen, but in a form that is not available to plants. South Kalimantan has NH4+ in the range 4.51- 736.86 mg.kg-1 and NO3- content is in the range of 0.37-54.69 mg.kg-1. The low availability of N nutrients in peatlands is due to the slow nitrogen mineralization process and the very high mobility of N in the soil. This study aims to determine the effect of the composition of peat and chicken manure on nitrogen mineralization. This study used a single factor in completely randomized design (CRD) with 5 treatments and 4 replications. carried out in greenhouses and in the Soil Laboratory of the Faculty of Agriculture, Lambung Mangkurat University, Banjarbaru. The treatment is the composition of peat and chicken manure, namely 100% peat material, 80% peat material + 20% chicken manure, 60% peat material + 40% chicken manure, 40% peat material + 60% chicken manure, 20% peat material + 80% chicken manure. The best composition for nitrogen mineralization is based on the high nitrate content, which is found in the composition of 80% peat soil + 20% chicken manure in the first month of incubation, and on the composition of 80% peat soil + 20% chicken manure in the second month. This composition is the best composition from a combination of peat material and chicken manure against nitrogen mineralization in the form of nitrates.

[EFFECTS OF PALM OIL SLUDGE SUBTITUTION FOR SYNTHETIC N FERTILIZER ON GROWTH AND YIELD OF SWEET CORN]. Palm oil sludge (POS) contains high concentrations of organic nitrogen dan its can serve as an alternative for synthetic nitrogen fertilizer. Objective of this study was to compare the growth and yield of sweet corn as amended with different compositions of synthetic N fertilizer and palm oil sludge. Six compositions, each containing 138 kg N/ha were assigned as the treatment and arranged in a randomized complete block design with three replications. These were 100% synthetic fertilizer, 80% synthetic fertilizer + 20% POS, 60% synthetic fertilizer + 40% POS, 40% synthetic fertilizer + 60% POS, 20% synthetic fertilizer + 80% POS, and 100% POS. The results of this study indicated that the plot amended with 60% synthetic fertilizer + 40% POS produced the highest plant stature (176.4 cm), stem diameter (2.40 cm), leaf area (8712.82 cm2 ), plant fresh weight (375.19 g), plant dry weight (136.68 g), and husked ear length (17.93 cm). However, the highest observed husked ear yield was only about 61.6 % the yield potential of the sweet corn variety.

Exceptionally high organic nitrogen concentrations in a semi-arid South Texas estuary susceptible to brown tide blooms

Studies of estuarine eutrophication have tended to focus on systems with continually flowing rivers, while little is known about estuaries from semi-arid/arid regions. Here we report results from an assessment of water quality conditions in Baffin Bay, Texas, a shallow (<2–3 m) subtropical estuary located in a semi-arid region that has agriculture as the dominant (44%) watershed land use. Chlorophyll a averaged 28–30 μg/l in Baffin Bay from 2003 to 2013 and total Kjeldahl nitrogen (TKN) concentrations were also very high (116–120 μM), with concentrations of both variables exceeding those of most other Texas estuaries by 2–5 fold. More recent field sampling (2013–2015) showed that dissolved organic nitrogen concentrations in Baffin Bay (62 ± 14 μM) were >2 fold higher than in three other Texas estuaries. In contrast, inorganic nitrogen (ammonium, nitrate) and phosphate concentrations were relatively low. Statistically significant long-term annual increases in chlorophyll a and salinity were observed in Baffin Bay, while long-term seasonal increases were observed for water temperature and TKN. Overall, Baffin Bay is displaying multiple symptoms of eutrophication, namely very high organic carbon, organic nitrogen and chlorophyll concentrations, as well as symptoms not quantified here such as fish kills and episodic hypoxia. Much of the increase in chlorophyll in Baffin Bay, at least since ∼1990, have coincided with blooms of the mixotrophic phytoplankton species, Aureoumbra lagunensis, which is thought to be favored under high proportions of organic to inorganic nitrogen. Thus the high and possibly increasing organic nitrogen concentrations, coupled with a long-term annual increase in salinity and a long-term seasonal increase in water temperature are likely to promote additional brown tide blooms in this system in the future.

Effects of phenotypic and RAPD variations on ecophysiological characterization of reeds (Phragmites australis) in Suncheon Bay

Genomic DNA of young reed leaves was subjected to PCR-based random amplified polymorphic DNA (RAPD) technique using random primers. Banding patterns from RAPD showed three major clusters. Group I showing vigorous growth was composed of plants from recently emerging reed patches on the coast shore and was established after the 2000s. Group II formed by reed patches was established at around the 1990s and showed non-vigorous growth along with typical symptoms of reed decline. Eutrophication could be the responsible factor for this reed decline, as the mudflat contained high concentrations of organic nitrogen and phosphorous. Group III reeds showed the most vigorous growth, demonstrating high values for shoot height, biomass, and panicle length along with moderate stem density. The state of Group III can be described as sub-optimal due to their old age on terrestrial habitat, as a minimum level of seawater is important for vigorous growth of reeds such as sufficient shoot height and photosynthetic capability. Reed height and panicle length have been shown to be recently declining in Suncheon Bay. These results indicate that old and rotting parts of reeds inhibit reed growth and that removal of these parts can have positive effects on shoot height and biomass.

Inorganic Nitrogen Derived from Foraging Honey Bees Could Have Adaptive Benefits for the Plants They Visit

In most terrestrial ecosystems, nitrogen (N) is the most limiting nutrient for plant growth. Honey bees may help alleviate this limitation because their feces (frass) have high concentration of organic nitrogen that may decompose in soil and provide inorganic N to plants. However, information on soil N processes associated with bee frass is not available. The objectives of this work were to 1) estimate the amount of bee frass produced by a honey bee colony and 2) evaluate nitrogen mineralization and ammonia volatilization from bee frass when surface applied or incorporated into soil. Two cage studies were conducted to estimate the amount of frass produced by a 5000-bee colony, and three laboratory studies were carried out in which bee frass, surface-applied or incorporated into soil, was incubated at 25oC for 15 to 45 days. The average rate of bee frass production by a 5,000-bee colony was estimated at 2.27 to 2.69 g N month−1. Nitrogen mineralization from bee frass during 30 days released 20% of the organic N when bee frass was surface applied and 34% when frass was incorporated into the soil. Volatilized NH3 corresponded to 1% or less of total N. The potential amount of inorganic N released to the soil by a typical colony of 20,000 bees foraging in an area similar to that of the experimental cages (3.24 m2) was estimated at 0.62 to 0.74 g N m−2 month−1 which may be significant at a community scale in terms of soil microbial activity and plant growth. Thus, the deposition of available N by foraging bees could have adaptive benefits for the plants they visit, a collateral benefit deriving from the primary activity of pollination.

Isolation and Identification of Ammonibacteria and Ammoniation Characteristic Analysis

Two strains of T1, Z1 ammonibacterium were isolated from the biofilm of the BAF reactor. According to the morphologic observation and physiological biochemistry test, the strains T1, Z1 were preliminarily identified asProteusandMicrococcus respectively. The research of the strains T1, Z1 for ammoniation characteristics indicated that the suitable temperature was 30°C, the suitable pH value was 7.0 and the best inoculum quantity was 5%. The strain T1 was strictly aerobic bacteria and Z1 was facultative aerobic bacteria. In these operating parameters, two strains of T1, Z1 were good of the degradation of organic nitrogen and the growth rate of ammonia nitrogen could respectively reach up to 86.91% and 69.98%. The strains T1, Z1 had a certainly practical value for treating the waste water with the high concentration of organic nitrogen.

Study of a three-stage fluidized bed process treating acrylic synthetic-fiber manufacturing wastewater containing high-strength nitrogenous compounds

Polyacrylonitrile (PAN) is one of the major synthetic fibers commonly used in the mass production of clothing. The chemical synthesis of PAN is carried out by polymerization of the acrylonitrile (AN) monomers with co-monomers such as vinyl acetate, methyl acrylate and cyclohexyl acrylate. Using water quality analysis of the PAN wastewater, high concentration of organic nitrogen was found and the TKN/COD ratios achieved were 0.15-0.26, indicating the complicated biodegradation characteristics for the PAN wastewater. In order to enhance biodegradation of nitrogenous compounds in PAN wastewater, a combined three-stage process of thermophilic anaerobic/anoxic denitrification/aerobic nitrification fluidized bed reactors was employed. The results indicated that the concentration of effluent in the three-stage process of OD and organic nitrogen was 175 mg/L and 13 mg/L, respectively. Furthermore, molecular biotechnology was applied to study the microbial population in the thermophilic anaerobic fluidized bed reactor. From the results of denaturing gradient gel electrophoresis, the diversity of PAN-degrading bacteria would change in different volumetric loading. Furthermore, the bacteria communities in the thermophilic anaerobic fluidized bed reactor were also studied by fluorescence in situ hybridization and confocal laser scanning microscopy. Alpha and delta-Proteobacteria were dominant in the bacteria population, and some high G+C content bacteria and Clostridium could be characterized in this system.

Increasing the fertilizer value of palm oil mill sludge: bioaugmentation in nitrification

Malaysia is essentially an agricultural country and her major polluting effluents have been from agro-based industries of which palm oil and rubber industries together contribute about 80% of the industrial pollution. Palm oil sludge, commonly referred to, as palm oil mill effluent (POME) is brown slurry composed of 4-5% solids, mainly organic, 0.5-1% residual oil, and about 95% water. The effluent also contains high concentrations of organic nitrogen. The technique for the treatment of POME is basically biological, consisting of pond systems, where the organic nitrogen is converted to ammonia, which is subsequently transformed to nitrate, in a process called nitrification. A 15-month monitoring program of a pond system (combined anaerobic, facultative, and aerobic ponds in series) confirmed studies by other authors and POME operators that nitrification in a pond system demands relatively long hydraulic retention time (HRT), which is not easily achieved, due to high production capacity of most factories. Bioaugmentation of POME with mixed culture of nitrifiers (ammonia and nitrite oxidizers) has been identified as an effective tool not only for enhancing nitrification of POME but also for improving quality of POME as source of liquid nitrogen fertilizer for use in the agricultural sector, especially in oil palm plantations. Nitrate is readily absorbable by most plants, although some plants are able to absorb nitrogen in the form of ammoniun. In this study, up to 60% reduction in HRT (or up to 20% reduction in potential land requirement) was achieved when bioaugmentation of POME was carried out with the aim of achieving full nitrification.

Enhanced biodegradation of organic nitrogenous compounds in resin manufacturing wastewater by anoxic denitrification and oxic nitrification process

In this study, a stream of resin manufacturing wastewater with high contents of organic nitrogenous compounds was treated by a conventional activated sludge process with extended aeration and a modified anoxic denitrification unit followed with two oxic activated sludge units respectively. Performance comparison of these two processes showed that the enhanced biodegradation of organic nitrogen in the modified process was attained with 69.0% of organic nitrogen removal that related to 39.0% removal in the conventional process. The anoxic/oxic process also promoted the removal efficiencies of COD, TKN and TN (total nitrogen) from 91%, 49.6% and 7.4% to 95.3%, 83.8% and 74.4% respectively, in comparison with the conventional extended activated sludge process. These cheerful results also corresponded to the acceptable averaged effluent quality: 82 mg/l of CODs, 48 mg/l of Org.-N, 11 mg/l of NH4+-N, and 42 mg N/l of NO3−-N, that could meet the national effluent standard of ROC-EPA in 1998. The successful investigation also demonstrated that about 50% of the total organic nitrogen removal was achieved in the anoxic unit associated with denitrification, while the residual organic nitrogen was removed in the oxic unit. The pilot-study results suggested that the resin manufacturing wastewater containing high concentration of organic nitrogen should be treated by the anoxic denitrification and oxic nitrification process instead of the conventional activated sludge process with extended aeration.

Oviposition by the Cinnabar Moth, Tyria Jacobaeae, in Relation to Nitrogen, Sugars and Alkaloids of Ragwort, Senecio Jacobaea

Recent studies have demonstrated that relationships between nitrogen levels and oviposition, food selection, larval survival and/or damage to host plants are not simple. We studied the effect of different levels of nitrogen on oviposition in the cinnabar moth in relation to the concentrations of alkaloids, that are positively correlated with nitrogen, and of soluble carbohydrates that are negatively correlated with nitrogen. We could not demonstrate that total alkaloid concentration was important as a proximate factor in the selection of food plants. Cinnabar moth selected plants with a high concentration of organic nitrogen and sugars. Plants poor in both, as well as plants rich in only one of these substances received fewer egg batches.

Geologic nitrogen and the occurrence of high nitrate soils in the western San Joaquin Valley, California

The content of total, nitrate, fixed, exchangeable, and organic nitrogen in Cretaceous and Tertiary formations in two representative drainage basins of the east central Diablo Range was determined. The results of these analyses showed that high concentrations of organic nitrogen (up to 1,200 ?g/g) occur predominantly in the Cretaceous formations while high concentrations of nitrate (up to 4,800 ?g/g) occur predominantly in the younger Tertiary sediments. The concentrations of fixed and exchangeable ammonium were variable and generally low. Available geomorphic data for the western side of the San Joaquin Valley were analyzed to seek a possible cause for the observed large accumulations of nitrates in west side soils. It was concluded that the occurrence of mudflows and intermediate flows in the alluvial fans bordering the Diablo Range on its east side, in conjunction with a semi-arid climate, is the principal factor responsible for the retention of soluble nitrogen. A method for predicting high nitrate soils in the western San Joaquin Valley was developed by assigning a score to each basin and associated fan based on the occurrence of high nitrate formations, the percentage of fine-grained sediments, and the predominance of mudflow deposits.

Atmospheric nitrogen fixation by hydrocarbon‐oxidizing bacteria

AbstractMicroorganisms have been found which concomitantly convert hydrocarbons, selected naphthenic acids, and atmospheric nitrogen into cellular substance. Bacteria are included in the genera Pseudomonas, Mycobacterium, and Azotobacter. Carbon sources utilized include the hydrocarbons methane, n‐butane, n‐tetradecane, toluene, and a naphthenic acid, cyclohexane‐carboxylate. Uptake of isotopic nitrogen was employed as a criterion of nitrogen fixation. The results indicate a rather wide prevalence in nature of hydrocarbon‐oxidizing bacteria capable of fixing atmospheric nitrogen. Their occurrence helps explain the high concentration of organic nitrogen commonly found in soils exposed to gas leakage from pipelines or natural‐gas seeps, and suggests further consideration of the possibility of applying selected petroleum residua to soils in order to increase the agricultural potential by nitrogen‐fixing processes.

Bacteriol. 88:468-472. 1964.-Methane-oxidizing bacteria capable of fixing atmospheric nitrogen were isolated from garden soil, pond mud, oil field soil, and soil exposed to natural gas, indicating a rather wide prevalence in nature. This may explain the high concentration of organic nitrogen commonly found in soils exposed to gas leakage from pipelines or natural-gas seeps. Added molybdenum was a requirement for growth in a nitrogen-free mineral salts medium. All nitrogen-fixing, methane-oxidizing bacteria isolated were gram-negative, nonsporeforming, usually motile rods. Colonies were light yellow, yellow, or white. The most common isolate, which formed light-yellow colonies, is referred to as Pseudomonas methanitrificans sp. n., and is distinguished from Pseudomonas (Methanomonas) methanica by nitrogen-fixing ability and a preponderance of poly-beta-hydroxybutyrate in the cellular lipid fraction.
