Detoxification and biodegradability enhancement of antibiotic production wastewater by hybrid process of resin adsorption and Fenton oxidation

Antibiotic production wastewater exhibits high concentration of antibiotics and toxic intermediates, which would pose a serious threat to the ecological environment. The biodegradation methods are prevailing for this wastewater. However, it imposes a considerable toxic burden on biological treatment units in wastewater treatment plants (WWTPs), ultimately resulting in subpar treatment efficacy. For improving the biodegradability of antibiotic production wastewater, hybrid process of resin adsorption and Fenton oxidation was proposed. The adsorption experiments showed that NDA-150 exhibited removal rates of up to 99.1% and 94.8% for two key pollutants 8-hydroxyquinoline (8HQ) and o-aminophenol (OAP), respectively. The constant removal rates demonstrated stable performance and capacity of NDA-150 for practical industrial operation. The remaining high COD due to other organic contaminants in the effluent required the involvement of Fenton oxidation for the further processing. The results of single factor experiments and orthogonal tests showed that the COD removal rate of 56.5% could be realized under the optimal operating conditions. This work proposed a novel process which could alleviate obstacles between the antibiotic manufacture factories and biological treatment units, shed light on the mechanisms and feasibility of it, and provide a promising avenue for the toxicant organic industrial sewage.

Sulfadiazine proliferated antibiotic resistance genes in the phycosphere of Chlorella pyrenoidosa: Insights from bacterial communities and microalgal metabolites

The phycosphere is an essential ecological niche for the proliferation of antibiotic resistance genes (ARGs). However, how ARGs’ potential hosts change and the driving mechanism of metabolites under antibiotic stress in the phycosphere have seldom been researched. We investigated the response of Chlorella pyrenoidosa and the structure and abundance of free-living (FL) and particle-attached (PA) bacteria, ARGs, and metabolites under sulfadiazine by using real-time quantitative PCR, 16 S rRNA high-throughput. The linkage of key bacterial communities, ARGs, and metabolites through correlations was established. Through analysis of physiological indicators, Chlorella pyrenoidosa displayed a pattern of "low-dose promotion and high-dose inhibition" under antibiotic stress. ARGs were enriched in the PA treatment groups by 117 %. At the phylum level, Proteobacteria, Bacteroidetes, and Actinobacteria as potential hosts for ARGs. At the genus level, potential hosts included Sphingopyxis, SM1A02, Aquimonas, Vitellibacter, and Proteiniphilum. Middle and high antibiotic concentrations induced the secretion of metabolites closely related to potential hosts by algae, such as phytosphingosine, Lysophosphatidylcholine, and α-Linolenic acid. Therefore, changes in bacterial communities indirectly influenced the distribution of ARGs through alterations in metabolic products. These findings offer essential details about the mechanisms behind the spread and proliferation of ARGs in the phycosphere.

Occurrence and Removal Efficiencies of Four Antibiotics in Kisii and Kabarnet Waste Water Treatment Plants, Kenya

Background: Antibiotics are presently considered as emerging contaminants with adverse effects in the environment and the population such as the development of antimicrobial resistant genes (ARG) and antimicrobial resistant bacteria (ARB). This study was guided by objective entailing to determine the seasonal occurrences and removal efficiencies of four antibiotics in Kisii and Kabarnet waste water treatment plants in Kenya. 
 Place and Duration of Study: Waste water samples were picked from Suneka wastewater treatment plant in Kisii county and Kabarnet Level V Hospital in Baringo County Kenya in the months of June and December 2020
 Methodology: Multiple samples were picked in Kisii and Kabarnet waste water treatment plants in the months of June and December correspond to the dry and wet seasons in Kenya. Collected waste water samples were centrifuged and filtered with glass microfiber filter papers and subsequently passed through a Solid phase extractor cartridge. High Performance Liquid Chromatography was used for quantification of antibiotics as per international commission for harmonization and subsequently applied for analysis.
 Results: The results revealed that the dry season had higher antibiotic concentrations at 1.29, 0.09, 2.92 and 1.82 µg/l for sulphamethoxazole, trimethoprim, ampicillin and amoxicillin respectively for the Kisii waste water treatment plant and 0.18, 0.05, 1.34 and 0.09 µg/l respectively for Kabarnet waste water treatment plant. During the wet seasons the measured concentrations were 1.11, 0.14, 2.04 and 1.34 µg/l for sulphamethoxazole, trimethoprim, ampicillin and amoxicillin at the Kisii WWTP, and 0.14, 0.06, 1.01 and 0.09 µg/l for Kabarnet WWTP. The removal efficiencies in the WWTPs, ranged from a high of 94 % to a low of 11.11 % depending on the type of antibiotic in both wet and dry seasons.
 Conclusion: Amoxicillin, ampicillin, trimethoprim and sulphamethaxazole were found in both Kisii WWTPs and Kabarnet WWTPs with nearly all antibiotics having a higher concentration of in the dry season than in the wet season. The highest calculated percentage removal was 94.03 % while the lowest calculated percentage removal was 7.14 %. The results suggest that the two WWTPs are effective for the removal of different types of antibiotics.

A role for the stringent response in ciprofloxacin resistance in Pseudomonas aeruginosa

Pseudomonas aeruginosa is a major cause of nosocomial infections and the leading cause of chronic lung infections in cystic fibrosis and chronic obstructive pulmonary disease patients. Antibiotic treatment remains challenging because P. aeruginosa is resistant to high concentrations of antibiotics and has a remarkable ability to acquire mutations conferring resistance to multiple groups of antimicrobial agents. Here we report that when P. aeruginosa is plated on ciprofloxacin (cipro) plates, the majority of cipro-resistant (ciproR) colonies observed at and after 48 h of incubation carry mutations in genes related to the Stringent Response (SR). Mutations in one of the major SR components, spoT, were present in approximately 40% of the ciproR isolates. Compared to the wild-type strain, most of these isolates had decreased growth rate, longer lag phase and altered intracellular ppGpp content. Also, 75% of all sequenced mutations were insertions and deletions, with short deletions being the most frequently occurring mutation type. We present evidence that most of the observed mutations are induced on the selective plates in a subpopulation of cells that are not instantly killed by cipro. Our results suggests that the SR may be an important contributor to antibiotic resistance acquisition in P. aeruginosa.

Characterization of clumpy adhesion of Escherichia coli to human cells and associated factors influencing antibiotic sensitivity.

The study explores a biofilm-like clumpy adhesion phenotype in Escherichia coli, along with various factors and implications for antibiotic susceptibility. The phenotype permitted the bacteria to survive the onslaught of high antibiotic concentrations. Profiles of the transcriptome and proteome allowed the differentiation between adhered bacteria in clumps and planktonic bacteria in the supernatant. The deletion mutants of genes differentially expressed between adhered and planktonic bacteria, i.e., flgH, ffp, pilV, spnT, and yggT, and respective complementations in trans cemented their roles in multiple capacities. ffp, an uncharacterized gene, is involved in motility and resistance to ampicillin in a clumpy state. The work also affirms for the first time the role of the yggT gene in adhesion and its involvement in susceptibility against another aminoglycoside antibiotic, i.e., gentamicin. Overall, the study contributes to the mechanisms of biofilm-like adhesion phenotype and understanding of the antimicrobial therapy failures and infections of E. coli.

Effects of two typical quinolone antibiotics in the marine environment on Skeletonema costatum

This study investigated the effects of levofloxacin (LEV) and norfloxacin (NOR) on Skeletonema costatum, focusing on cell growth, chlorophyll a (Chla) content, maximal quantum yield of PSII (Fv/Fm), protein content, enzyme activities of superoxide dismutase (SOD), glutathione reductase (GR), and glutathione peroxidase (GSH-PX), and the membrane lipid peroxidation product malondialdehyde (MDA) content were conducted to analyze the responses of S. costatum under LEV and NOR exposure. Cell growth, Chla content, Fv/Fm, protein content, enzyme activities, and MDA content were assessed to elucidate physiological changes. Both LEV and NOR inhibited S. costatum growth, except for 10 mg/L NOR, which promoted growth. Algal cells exhibited higher sensitivity to LEV, with 96h-IC50 values of 14.770 mg/L for LEV and 44.250 mg/L for NOR. Low NOR concentration (10 mg/L) increased Chla content, while high antibiotic concentrations (>20 mg/L for LEV, >100 mg/L for NOR) decreased Chla content and Fv/Fm, indicating an impact on photosynthesis. Elevated LEV and NOR levels reduced protein and MDA content but increased GR, SOD, and GSH activities, indicating induced oxidative stress. The study provides a comprehensive analysis of LEV and NOR effects on marine microalgae growth and underlying physiological mechanisms, shedding light on potential ecological risks posed by antibiotics in marine ecosystems.

Effects of high antibiotic concentrations applied to continuous local antibiotic perfusion on human bone tissue-derived cells.

Continuous local antibiotic perfusion (CLAP) has recently attracted attention as a new drug delivery system for orthopaedic infections. CLAP is a direct continuous infusion of high-concentration gentamicin (1,200 μg/ml) into the bone marrow. As it is a new system, its influence on the bone marrow is unknown. This study aimed to examine the effects of high-concentration antibiotics on human bone tissue-derived cells. Cells were isolated from the bone tissue grafts collected from six patients using the Reamer-Irrigator-Aspirator system, and exposed to different gentamicin concentrations. Live cells rate, apoptosis rate, alkaline phosphatase (ALP) activity, expression of osteoblast-related genes, mineralization potential, and restoration of cell viability and ALP activity were examined by in vitro studies. The live cells rate (the ratio of total number of cells in the well plate to the absorbance-measured number of live cells) was significantly decreased at ≥ 500 μg/ml of gentamicin on day 14; apoptosis rate was significantly increased at ≥ 750 μg/ml, and ALP activity was significantly decreased at ≥ 750 μg/ml. Real-time reverse transcription-polymerase chain reaction results showed no significant decrease in the ALP and activating transcription factor 4 transcript levels at ≥ 1,000 μg/ml on day 7. Mineralization potential was significantly decreased at all concentrations. Restoration of cell viability was significantly decreased at 750 and 1,000 μg/ml on day 21 and at 500 μg/ml on day 28, and ALP activity was significantly decreased at 500 μg/ml on day 28. Our findings suggest that the exposure concentration and duration of antibiotic administration during CLAP could affect cell functions. However, further in vivo studies are needed to determine the optimal dose in a clinical setting.

Development of novel hybrid 3D-printed degradable artificial joints incorporating electrospun pharmaceutical- and growth factor-loaded nanofibers for small joint reconstruction

Small joint reconstruction remains challenging and can lead to prosthesis-related complications, mainly due to the suboptimal performance of the silicone materials used and adverse host reactions. In this study, we developed hybrid artificial joints using three-dimensional printing (3D printing) for polycaprolactone (PCL) and incorporated electrospun nanofibers loaded with drugs and biomolecules for small joint reconstruction. We evaluated the mechanical properties of the degradable joints and the drug discharge patterns of the nanofibers. Empirical data revealed that the 3D-printed PCL joints exhibited good mechanical and fatigue properties. The drug-eluting nanofibers sustainedly released teicoplanin, ceftazidime, and ketorolac in vitro for over 30, 19, and 30 days, respectively. Furthermore, the nanofibers released high levels of bone morphogenetic protein-2 and connective tissue growth factors for over 30 days. An in vivo animal test demonstrated that nanofiber-loaded joints released high concentrations of antibiotics and analgesics in a rabbit model for 28 days. The animals in the drug-loaded degradable joint group showed greater activity counts than those in the surgery-only group. The experimental data suggest that degradable joints with sustained release of drugs and biomolecules may be utilized in small joint arthroplasty.

Synergistic experimental study by a solar pilot evaporation system for the volume reduction and antibiotics photocatalytic degradation of liquid digestate

The challenges posed by the large volume, low nutrient content, and high antibiotic concentrations in the liquid digestate from anaerobic digestion are becoming increasingly prominent. In this study, a solar pilot system was used for the evaporation and volume reduction of the liquid digestate and the photocatalytic degradation of antibiotics in the liquid digestate. First, given the difficulty in recovering the powdered catalyst, dolomite-supported g-C3N4 (DCN) was synthesized using a simple one-pot method, and its photocatalytic properties were investigated. The results indicated that the photocatalytic performance of DCN was excellent when the urea-to-dolomite doping ratio was 1:8, and the degradation efficiency of tetracyclines (TCs) in the real liquid digestate reached 90.65 % when the amount of DCN/1:8 was 0.625 g/mL. The TCs degradation efficiency increased as the initial pH of the liquid digestate decreased, reaching 95.33 % at an initial pH of 4, which could be attributed to the acidic conditions that are favorable for the generation of hydroxyl radicals and excited oxygen. Furthermore, the DCN was filled directly into a solar pilot system, which was employed for the evaporation reduction treatment of the liquid digestate as well as the photocatalytic degradation of antibiotics under optimal process conditions. The pilot experiment showed that the average TCs degradation efficiency rose from 57.54 % to 95.27 % with the addition of DCN/1:8 to the solar evaporation system. After a 4-day replicated experiment, the average TCs degradation efficiency reached 90.64 %. Importantly, DCN/1:8 had less of an effect on the evaporation efficiency of the liquid digestate and the recovery of key nutrients, such as NH4+-N. This study aims to demonstrate the engineering, theoretical, and technological feasibility of sunlight photocatalytic degradation of antibiotics in complex water quality systems and to provide a reference for the reduction and harmless treatment of liquid digestate.

Chemical-Vapor-Deposition-Synthesized Two-Dimensional Non-Stoichiometric Copper Selenide (β-Cu2-xSe) for Ultra-Fast Tetracycline Hydrochloride Degradation under Solar Light.

The high concentration of antibiotics in aquatic environments is a serious environmental issue. In response, researchers have explored photocatalytic degradation as a potential solution. Through chemical vapor deposition (CVD), we synthesized copper selenide (β-Cu2-xSe) and found it an effective catalyst for degrading tetracycline hydrochloride (TC-HCl). The catalyst demonstrated an impressive degradation efficiency of approximately 98% and a reaction rate constant of 3.14 × 10-2 min-1. Its layered structure, which exposes reactive sites, contributes to excellent stability, interfacial charge transfer efficiency, and visible light absorption capacity. Our investigations confirmed that the principal active species produced by the catalyst comprises O2- radicals, which we verified through trapping experiments and electron paramagnetic resonance (EPR). We also verified the TC-HCl degradation mechanism using high-performance liquid chromatography-mass spectrometry (LC-MS). Our results provide valuable insights into developing the β-Cu2-xSe catalyst using CVD and its potential applications in environmental remediation.

Sensor system for analysis of biofilm sensitivity to ampicillin

The resistance of biofilms to antibiotics is a key factor that makes bacterial infections unsusceptible to antimicrobial therapy. The results of classical tests of cell sensitivity to antibiotics cannot be used to predict therapeutic success in infections associated with biofilm formation. We describe a simple and rapid method for the real-time evaluation of bacterial biofilm sensitivity to antibiotics, with Pseudomonas putida and ampicillin as examples. The method uses an electric biosensor to detect the difference between changes in the biofilm electric polarizability, thereby evaluating antibiotic sensitivity. The electric signals showed that P. putida biofilms were susceptible to ampicillin and that at high antibiotic concentrations, the biofilms differed markedly in their susceptibility (dose-dependent effect). The sensor also detected differences between biofilms before and after ampicillin treatment. The electric-signal changes enabled us to describe the physical picture of the processes occurring in bacterial biofilms in the presence of ampicillin. The approach used in this study is promising for evaluating the activity of various compounds against biofilms, because it permits a conclusion about the antibiotic sensitivity of biofilm bacteria to be made in real time and in a short period (analysis time, not longer than 20 min). Source apportionment and predictable driving factors contribute to antibiotics profiles in Changshou Lake of the Three Gorges Reservoir area, China

Lakes, crucial antibiotic reservoirs, lack thorough exploration of quantitative relationships between antibiotics and influencing factors. Here, we conducted a comprehensive year-long investigation in Changshou Lake within the Three Gorges Reservoir area, China. The concentrations of 21 antibiotics spanned 35.6–200 ng/L, 50.3–348 ng/L and 0.57–57.9 ng/g in surface water, overlying water and sediment, respectively. Compared with abundant water period, surface water and overlying water displayed significantly high antibiotic concentrations in flat and low water periods, while sediment remained unchanged. Moreover, tetracyclines, fluoroquinolones and erythromycin posed notable risks to algae. Six primary sources were identified using positive matrix factorization model, with aquaculture contributing 21.2%, 22.7% and 25.4% in surface water, overlying water and sediment, respectively. The crucial predictors were screened through machine learning, redundancy analysis and Mantel test. Our findings emphasized the pivotal roles of water quality parameters, including water temperature (WT), pH, dissolved oxygen, electrical conductivity, inorganic anions (NO3⁻, Cl⁻ and F⁻) and metal cations (Ca, Mg, Fe, K and Cr), with WT influencing greatest. Total nitrogen (TN), cation exchange capacity, K, Al and Cd significantly impacted sediment antibiotics, with TN having the most pronounced effect. This study can promise valuable insights for environmental planning and policies addressing antibiotic pollution.

Unveiling a novel mechanism for competitive advantage of ciprofloxacin-resistant bacteria in the environment through bacterial membrane vesicles

Ciprofloxacin (CIP) is a prevalent environmental contaminant that poses a high risk of antibiotic resistance. High concentrations of antibiotics can lead to the development of resistant bacteria with high fitness costs, which often face a competitive disadvantage. However, it is unclear whether low-cost resistant bacteria formed by exposure to sub-MIC CIP in the environment can evolve competitive mechanisms against sensitive Escherichia coli (SEN) other than stronger resistance to CIP. Our study exposed E. coli to sub-MIC CIP levels, resulting in the development of CIP-resistant E. coli (CIPr). In antibiotic-free co-culture assays, CIPr outcompeted SEN. This indicates that CIPr is very likely to continue to develop and spread in antibiotic-free environments such as drinking water and affect human health. Further mechanism investigation revealed that bacterial membrane vesicles (BMVs) in CIPr, functioning as substance delivery couriers, mediated a cleavage effect on SEN. Proteomic analysis identified Entericidin B (EcnB) within CIPr-BMVs as a key factor in this competitive interaction. RT-qPCR analysis showed that the transcription of its negative regulator ompR/envZ was down-regulated. Moreover, EcnB plays a crucial role in the development of CIP resistance, and some resistance-related proteins and pathways have also been discovered. Metabolomics analysis highlighted the ability of CIPr-BMVs to acidify SEN, increasing the lytic efficiency of EcnB through cationization. Overall, our study reveals the importance of BMVs in mediating bacterial resistance and competition, suggesting that regulating BMVs production may be a new strategy for controlling the spread of drug-resistant bacteria.

Defying the odds: Determinants of the antimicrobial response of Salmonella Typhi and their interplay.

Salmonella Typhi, the invasive serovar of S. enterica subspecies enterica, causes typhoid fever in healthy human hosts. The emergence of antibiotic-resistant strains has consistently challenged the successful treatment of typhoid fever with conventional antibiotics. Antimicrobial resistance (AMR) in Salmonella is acquired either by mutations in the genomic DNA or by acquiring extrachromosomal DNA via horizontal gene transfer. In addition, Salmonella can form a subpopulation of antibiotic persistent (AP) cells that can survive at high concentrations of antibiotics. These have reduced the effectiveness of the first and second lines of antibiotics used to treat Salmonella infection. The recurrent and chronic carriage of S. Typhi in human hosts further complicates the treatment process, as a remarkable shift in the immune response from pro-inflammatory Th1 to anti-inflammatory Th2 is observed. Recent studies have also highlighted the overlap between AP, persistent infection (PI) and AMR. These incidents have revealed several areas of research. In this review, we have put forward a timeline for the evolution of antibiotic resistance in Salmonella and discussed the different mechanisms of the same availed by the pathogen at the genotypic and phenotypic levels. Further, we have presented a detailed discussion on Salmonella antibiotic persistence (AP), PI, the host and bacterial virulence factors that can influence PI, and how both AP and PI can lead to AMR.

Dissemination of antibiotic resistance genes through soil-plant-earthworm continuum in the food production environment

Treated municipal wastewater (TMW) can provide a reliable source of irrigation water for crops, which is especially important in arid areas where water resources are limited or prone to drought. Nonetheless, TMW may contain residual antibiotics, potentially exposing the crops to these substances. The goal of this study was to investigate the dissemination of antibiotics resistance genes (ARGs) in the soil–plant–earthworm continuum after irrigation of spinach and radish plants with TMW containing trimethoprim, sulfamethoxazole, and sulfapyridine in a greenhouse experiment, followed by feeding of earthworms with harvested plant materials. Our results showed that antibiotic resistance genes (ARGs) were enriched in the soil–plant–earthworm microbiomes irrigated with TMW and TMW spiked with higher concentrations of antibiotics. The number of ARGs and antibiotic-resistant bacteria (ARB) enrichment varied with plant type, with spinach harboring a significantly higher amount of ARGs and ARB compared to radish. Our data showed that bulk and rhizosphere soils of spinach and radish plants irrigated with MilliQ water, TMW, TMW10, or TMW100 had significant differences in bacterial community (p < 0.001), ARG (p < 0.001), and virulence factor gene (VFG) (p < 0.001) diversities. The abundance of ARGs significantly decreased from bulk soil to rhizosphere to phyllosphere and endosphere. Using metagenome assembled genomes (MAGs), we recovered many bacterial MAGs and a near complete genome (>90 %) of bacterial MAG of genus Leclercia adecarboxylata B from the fecal microbiome of earthworm that was fed harvested radish tubers and spinach leaves grown on TMW10 irrigated waters, and this bacterium has been shown to be an emerging pathogen causing infection in immunocompromised patients that may lead to health complications and death. Therefore, crops irrigated with TMW containing residual antibiotics and ARGs may lead to increased incidences of enrichment of ARB in the soil–plant–earthworm continuum.

Effect of Vancomycin, Gentamicin and Clindamycin on Cartilage Cells In Vitro.

The treatment of grafts with vancomycin for ligament reconstruction in knee surgery is the current standard. However, high antibiotic concentrations have chondrotoxic effects. To test the chondrotoxicity of clindamycin, gentamicin and vancomycin in comparable concentrations. In vitro and in vivo effective concentrations hugely vary from drug to drug. To allow for comparisons between these three commonly used antibiotics, the concentration ranges frequently used in orthopedic surgical settings were tested. Controlled laboratory study. Human cartilage from 10 specimens was used to isolate chondrocytes. The chondrocytes were treated with clindamycin (1 mg/mL and 0.5 mg/mL), gentamicin (10 mg/mL and 5 mg/mL) or vancomycin (10 mg/mL and 5 mg/mL), at concentrations used for preoperative infection prophylaxis in ligament surgery. Observations were taken over a period of 7 days. A control of untreated chondrocytes was included. To test the chondrotoxicity, a lactate dehydrogenase (LDH) test and a water-soluble tetrazolium salt (WST-1) assay were performed on days 1, 3 and 7. In addition, microscopic examinations were performed after fluorescence staining of the cells at the same time intervals. All samples showed a reasonable vitality of the cartilage cells after 72 h. However, clindamycin and gentamicin both showed higher chondrotoxicity in all investigations compared to vancomycin. After a period of 7 days, only chondrocytes treated with vancomycin showed reasonable vitality. The preoperative treatment of ligament grafts with vancomycin is the most reasonable method for infection prophylaxis, in accordance with the current study results regarding chondrotoxicity; however, clindamycin and gentamicin cover a wider anti-bacterial spectrum. The prophylactic antibiotic treatment of ligament grafts at concentrations of 5 mg/mL or 10 mg/mL vancomycin is justifiable and reasonable. In specific cases, even the use of gentamicin and clindamycin is appropriate.

Enhancing biofilm penetration and antibiofilm efficacy with protein nanocarriers against pathogenic biofilms

Nanocarriers play an important role in enhancing the efficacy of antibiotics against biofilms by improving their penetration and prolonging retention in pathogenic biofilms. Herein, the multifunctional nanocarriers including nanospheres (NS) and nanotubes (NT) with a high biocompatibility and biodegradability were prepared through self-assembly of partially hydrolyzed α-lactalbumin. The effects of these two different shaped nanocarriers on the delivery of antibiotics for biofilm treatment were examined by conducting in vitro antibiofilm experiment and in vivo infected wound model. The strong affinity of NS and NT for the bacterial surface allows antibiotics to be concentrated in the bacteria. Notably, the high permeability of NT into biofilms facilitates deeper penetration and the easier diffusion of loaded antibiotics within the biofilm. Furthermore, the acidic biofilm environment triggers the release of antibiotics from the NT, resulting in the accumulation of high local antibiotic concentrations. Therefore, NT could efficiently clean and inhibit the biofilm formation while also destroying the mature biofilms. In a S. aureus infected wound animal model, treatment with antibiotic-loaded NT demonstrated accelerated healing of S. aureus infected wounds when compared to free antibiotic treatment. These findings indicate that NT nanocarrier strategy is promising for treating bacterial biofilm infections, offering the potential for lower antibiotics dosages and preventing the overuse of antibiotics.

Cultivation of black soldier fly larvae in substrates containing antibiotics

The use of antibiotics in agriculture to treat and prevent infectious diseases and to stimulate growth results in antibiotics accumulating in animal tissues and wastes. Further utilization of such animal waste causes an increase in antibiotic-resistant bacteria. One of the promising ways of manure processing leading to reduction of antibiotic concentrations and obtaining useful biomass from organic waste may be bioconversion with the help of insects, particularly black soldier flies (Hermetia illucens) ( Diptera: Stratiomyldae). It is important to know how the presence of antibiotics in the feeding substrate affects the insects and their characteristics for further utilization. The effect of adding the antibiotics ceftriaxone, coliflox, levofloxacin and their mixtures to feed substrates on the growth of black soldier fly larvae was studied. When ceftriaxone, coliflox and levofloxacin were given at a concentration of 1–100 mg/kg feed, an increase in mean larval weight was observed compared to the control group after 5 days of culturing. At antibiotic concentrations of 500–1000 mg/kg, this effect was compensated, presumably by the negative effect of the antibiotic on the microbiota of the insect digestive system. At antibiotic concentrations of 2000 mg/kg feed, growth of Hermetia illucens larvae was retarded. No effect of antibiotics on fatty acid composition, moisture and ash content of insects was found. The results indicate that the use of black soldier fly for the treatment of animal waste is feasible, but the likely negative effect on insect growth at high antibiotic concentrations should be considered.

The Controversial Effect of Antibiotics on Methicillin-Sensitive S. aureus: A Comparative In Vitro Study.

Methicillin-sensitive Staphylococcus (S.) aureus (MSSA) bacteremia remains a global challenge, despite the availability of antibiotics. Primary treatments include β-lactam agents such as cefazolin and flucloxacillin. Ongoing discussions have focused on the potential synergistic effects of combining these agents with rifampicin or fosfomycin to combat infections associated with biofilm formation. Managing staphylococcal infections is challenging due to antibacterial resistance, biofilms, and S. aureus's ability to invade and replicate within host cells. Intracellular invasion shields the bacteria from antibacterial agents and the immune system, often leading to incomplete bacterial clearance and chronic infections. Additionally, S. aureus can assume a dormant phenotype, known as the small colony variant (SCV), further complicating eradication and promoting persistence. This study investigated the impact of antibiotic combinations on the persistence of S. aureus 6850 and its stable small colony variant (SCV strain JB1) focusing on intracellular survival and biofilm formation. The results from the wild-type strain 6850 demonstrate that β-lactams combined with RIF effectively eliminated biofilms and intracellular bacteria but tend to select for SCVs in planktonic culture and host cells. Higher antibiotic concentrations were associated with an increase in the zeta potential of S. aureus, suggesting reduced membrane permeability to antimicrobials. When using the stable SCV mutant strain JB1, antibiotic combinations with rifampicin successfully cleared planktonic bacteria and biofilms but failed to eradicate intracellular bacteria. Given these findings, it is reasonable to report that β-lactams combined with rifampicin represent the optimal treatment for MSSA bacteremia. However, caution is warranted when employing this treatment over an extended period, as it may elevate the risk of selecting for small colony variants (SCVs) and, consequently, promoting bacterial persistence.

Impact analysis of hydraulic loading rate and antibiotics on hybrid constructed wetland systems: Insight into the response to decontamination performance and environmental-associated microbiota

Hybrid constructed wetlands (HCWs) are a promising solution for water ecology and environmental treatment, not only for conventional types of water pollution but also for antibiotics. Among the critical parameters for wetlands, the hydraulic loading rate (HLR) is especially important given the challenges of antibiotics treatment and frequent extreme rainfall. To investigate the removal performance of different HLRs on nutrients and antibiotics, as well as the response of antibiotics to nutrient removal, and the impact of HLRs on microbial communities, new HCWs with vertical flow constructed wetlands (VFCWs) and floating constructed wetlands (FCWs) in series were built. The results of the study showed that: (1) HCWs are highly effective in removing chemical oxygen demand (COD), NH4+−N, NO2−−N, and total phosphorus (TP) at low HLR (L_HLR), with removal efficiencies as high as 97.8%, 99.6%, 100%, and 80.5%. However, high HLR (H_HLR) reduced their removal efficiencies; (2) The average removal efficiency of fluoroquinolones (FQs) under different HLRs was consistently high, at 99.9%, while the average removal efficiency of macrolides (MLs) was 96.3% (L_HLR) and 88.4% (H_HLR). The removal efficiency of sulfonamides (SAs) was susceptible to HLRs, and the removal of antibiotics occurred mainly in the rhizosphere zone of wetland; (3) High concentrations of antibiotics in HCWs were found to inhibit and poison plant growth and to reduce the removal efficiency of TP by 12%. However, they had a minor effect on the removal efficiency of carbon and nitrogen nutrients; (4) H_HLR altered the diversity and abundance of microbial communities in different compartments of the wetland and also reduced the relative abundance of Bacillus, Hydrogenophaga, Nakamurella, Denitratisoma and Acidovorax genera, which are involved in denitrification and phosphorus removal processes. This alteration in microbial communities was one of the main reasons for the reduced performance of nitrogen and phosphorus removal.
