Ethylene and hydrogen sulfide regulate hexavalent chromium toxicity in two pulse crops: Implication on growth, photosynthetic activity, oxidative stress and ascorbate glutathione cycle

Sustainable agriculture has become prime importance to feed growing population. To achieve this goal application of exogenous hormones and signaling molecules are gaining important. In this context, we have investigated potential of ethylene (25μM ethephon; donor) and H2S (10μM NaHS; donor) in mitigating hexavalent chromium [Cr (VI), 50μM] toxicity in two pulse seedlings: black bean and mung bean. Cr(VI) declined growth and gas exchange parameters (photosynthetic rate, stomatal conductance, sub cellular CO2 concentration, and transpiration level) which was accompanied by intracellular accumulation of Cr in both pulse crops and the damaging effect was greater in mung bean seedlings. The suppression in the growth and related parameters was occurred due to higher buildup of oxidative stress markers; O2•‾, H2O2, lipid peroxidation (as malondialdehyde, MDA equivalents) and membrane injury in leaf and root of both pulse crops. Cr induced disturbance in AsA-GSH cycle (reduction in the activity of glutathione reductase, ascorbate peroxidase, monodehydroascorbate reductase and dehydroascorbate reductase, and the amount of ASA and GSH) could be one of the reasons for greater accumulation of H2O2. Further, exogenous application of ethylene and H2S significantly ameliorated Cr toxicity on growth and photosynthetic activity by significantly lowering the intracellular Cr accumulation and oxidative biomarkers, and also by strengthening the activity of AsA-GSH cycle. The exogenous application of biosynthesis inhibitors of ethylene (AVG) and H2S (PAG) caused greater damaging effect on these parameters due to more accumulation of Cr(VI), thereby suggesting that the endogenous levels of these regulators are critical for Cr(VI) tolerance. Interestingly, ET did not rescue adverse effects of Cr(VI) in absence of endogenous H2S, while H2S could do so even without endogenous ethylene, suggesting that H2S played downstream signaling to ethylene in regulating Cr(VI) toxicity. Hence, being cheap and easily available theses growth regulators may be considered for sustainable agriculture.

Rapidly and simply detecting Cr (VI) in aqueous media via a diketopyrrolopyrrole-based chemosensor with both high selectivity and low LOD

BackgroundThe high toxicity of hexavalent chromium [Cr (VI)] could not only cause harmful effects on humans, including carcinogenicity, respiratory issues, genetic damage, and skin irritation, but also contaminate drinking water sources, aquatic ecosystems, and soil, impairing the reproductive capacity, growth, and survival of organisms. Due to these harmful effects, detecting toxic Cr (VI) is of great significance. However, the rapid, simple, and efficient detection at a low Cr (VI) concentration is extremely challenging, especially in an acidic condition (existing as HCrO4−) due to its low adsorption free energy. ResultsA diketopyrrolopyrrole-based small molecule (DPPT-PhSMe) is designed and characterized to act as a chemosensor, which allows a high selectivity to Cr (VI) at an acidic condition with a low limit of detection to 10−8 M that is two orders of magnitude lower than the cut of limit (1 μM) recommended by World Health Organization (WHO). Mechanism study indicates that the rich sulfur atoms enhance the affinity to HCrO4−. Combining with favorable features of diketopyrrolopyrrole, DPPT-PhSMe not only allows dual-mode detection (colorimetric and spectroscopic) to Cr (VI), but also enables disposable paper-based sensor for naked-eye detection to Cr (VI) from fully aqueous media. The investigation of DPPT-PhSMe chemosensor for the quantification of Cr (VI) in real life samples demonstrates a high reliability and accuracy with an average percentage recovery of 102.1 % ± 4 (n = 3). SignificanceDPPT-PhSMe represents the first diketopyrrolopyrrole-derived chemosensor for efficient detection to toxic Cr (VI), not only providing a targeted solution to the bottleneck of Cr (VI) detection in acidic conditions (existing as HCrO4−) caused by its low adsorption free energy, but also opening a new scenario for simple, selective, and efficient Cr (VI) detection with conjugated dye molecules.

Interactions between chromium species and DNA in vitro and their potential role in the toxicity of hexavalent chromium.

Epidemiological and animal studies have supported the carcinogenicity of hexavalent chromium [Cr(VI)]; however, molecular changes responsible for the induction of cancer by Cr(VI) are not entirely understood. Numerous mechanistic studies suggested the role of oxidative stress and genotoxicity in Cr(VI)-mediated carcinogenesis; however, specific types of DNA damage have not yet been conclusively attributed to specific chromium species or other reactive byproducts generated in biological systems exposed to Cr(VI). Due to the remarkably complex chemistry and biological effects of chromium species generated through the intracellular reduction of Cr(VI), their relevance for Cr(VI)-mediated carcinogenesis has not yet been fully elucidated and continues to be a subject of ongoing discussions in the field. In this report, we describe a complex world of chromium species and their reactivity with DNA and other biologically relevant molecules in vitro to inform a more complete understanding of Cr(VI)-mediated toxicity. In addition, we discuss previous results in the context of in vitro models and analytical methods to reconcile some conflicting findings on the biological role of chromium species.

A review of the treatment technologies for hexavalent chromium contaminated water.

The toxicity of hexavalent chromium (Cr(VI)) present in the environment has exceeded the current limits or standards and thus may lead to biotic and abiotic catastrophes. Accordingly, several treatments, including chemical, biological, and physical approaches, are being used to reduce Cr(VI) waste in the surrounding environment. This study compares the Cr(VI) treatment approaches from several areas of science and their competence in Cr(VI) removal. As an effective combination of physical and chemical approaches, the coagulation-flocculation technique removes more than 98% of Cr(VI) in less than 30min. Most membrane filtering approaches can remove up to 90% of Cr(VI). Biological approaches that involve the use of plants, fungi, and bacteria also successfully eliminate Cr(VI) but are difficult to scale up. Each of these approaches has its benefits and drawbacks, and their applicability is determined by the research aims. These approaches are also sustainable and environmentally benign, thus limiting their effects on the ecosystem.

Efficient hexavalent chromium removal by nano-cerium oxide functionalized biochar: Insight into the role of reduction

The strong toxicity of hexavalent chromium (Cr(VI)) have spurred great interests in developing an effective technology to remove it from water. In this study, a cost-effective and recyclable composite nano-cerium oxide functionalized peanut shell biochar (BC-Ce) was synthetized via impregnation-precipitation-pyrolysis process and employed for simultaneous adsorption and reduction of Cr(VI) in water. The BC-Ce possessed better Cr(VI) adsorption potential, and its maximum Cr(VI) adsorption capacity calculated by Langmuir achieved 47.83 mg g−1. Simultaneously, the as-prepared composite displayed obvious reduction behavior for Cr(VI) and partially adsorbed Cr(VI) was reduced to less toxic Cr(III). The effects of composite dosage (0.2–4 g L−1), initial Cr(VI) concentration (20–200 mg L−1) and pH (3−9) on Cr(VI) removal were also investigated. Characterization results indicated that the Cr(VI) reduction induced by BC-Ce was mainly attributed to the comprehensive effect of oxygen-containing functional group (OFGs), carbon-centered persistent free radicals (PFRs), oxygen vacancies and graphitic structure in BC-Ce. The CeO2 supported on the surface of BC facilitated the generation of more OFGs (phenolic-OH and C-O), PFRs and oxygen vacancies, thus provided abundant sites for chemical reduction of Cr(VI). Moreover, the graphitic structure in BC-Ce promoted the transfer of electrons from graphite carbon to Cr(VI). This study provided a new insight into the removal mechanisms of Cr(VI) in water by BC-Ce. Except for the adsorption, the reduction behavior during Cr(VI) removal process by BC-based materials should not be overlooked.

Titanium dioxide nanoparticles potentially regulate the mechanism(s) for photosynthetic attributes, genotoxicity, antioxidants defense machinery, and phytochelatins synthesis in relation to hexavalent chromium toxicity in Helianthus annuus L.

Chromium (VI) is one of the hazardous heavy metal, heavily discharged into the soil and severely hampers the plants yield. The TiO2 NPs was selected due to its potential to alleviate the heavy metals toxicity. This manuscript unravels the mechanisms for Cr(VI) induced toxicity and how foliar application of TiO2 NPS potentially ameliorate the toxicity by regulating the photosynthetic attributes, DNA damage, antioxidants defense machinery, and phytochelatins synthesis in Helianthus annuus L. Plants were exposed to Cr(VI) concentrations [0, 15, 30, and 60 mg Cr(VI) kg−1 of soil], and TiO2 NPS (15 mg L−1, 25 nm size) were foliar sprayed thrice to the plants at three days interval. The maximum accumulation of total chromium was recorded in root (12.53 µg g−1 DW) followed by shoot (5.67 µg g−1 DW) at 60 mg Cr(VI) treatment. The presence and localization of TiO2 NPs inside the plant leaf cells were confirmed by TEM-EDS analysis. The results revealed that Cr(VI) exposure had a dose-dependent inhibitory effects on photosynthetic attributes, structure of guard and epidermal cells, photosynthetic pigments; inducing impacts on H2O2 and MDA productions, DNA damage, AsA-GSH cycle, and most importantly on PC2, and PC3 synthesis which is rarely reported. However, TiO2 NPs exposure minimized Cr(VI) induced toxicity through reduction of total chromium accumulation, H2O2 and MDA productions, thereby reducing DNA damage reported first time under combined treatment of Cr(VI)+ TiO2 NPs as evidenced through comet assay. It also positively regulate the photosynthetic pigments, AsA-GSH cycle, and modulates PC2 and PC3 synthesis which have crucial impacts on ROS quenching and Cr(VI) detoxification, respectively, and in turn, minimizes Cr(VI) toxicity in H. annuus L. Besides, this study strengthens the less acknowledged report that Cr(VI) is an inducer of PCs synthesis and also confirms that TiO2 NPs potentially counteract Cr(VI) toxicity.

Detoxification and Removal of Hexavalent Chromium in Aquatic Systems: Applications of Bioremediation

Chromium is a transition metal with a wide range of applications in leather tanning, textile, electroplating, stainless steel production, inorganic chemical production and wood preservation industries due to yellow colouration, corrosion resistance, higher melting-point and crystalline structure with raging of oxidation states from 0 to +6. Trivalent and hexavalent chromium are the most abundant forms of chromium discharged into the aquatic environment by industries. It has been reported that hexavalent chromium is highly toxic than trivalent chromium due to the higher solubility, mobility and tendency to accumulate in higher trophic levels, which, therefore, become bioavailable and causes carcinogenic, mutagenic and teratogenic effects on most microorganisms and animals, growth inhibition, morphological and physiological changes and yield reductions in plants. Therefore, it is essential to detoxify the above hazardous pollutants up to permissible limits, which local and international authorities have legislated concerning its threat towards biotic components. Hexavalent chromium detoxification is possible to achieve using three methods i.e. physical, chemical and biological methods. These remediation processes can eliminate highly toxic hexavalent chromium or transform it into a less toxic form of trivalent chromium, completely or partially by adsorption and reduction. Biological remediation is considered a cost-effective and ecofriendly method compared to physical and chemical remediation. Further, many biological agents have been identified as agents that can tolerate the hexavalent chromium toxicity up to certain higher levels depending on the internal and external environmental factors, indicating different metal tolerance mechanisms that are assumed to be applied in metal remediation aspects. According to the testimonies of novel bioremediation studies, some hexavalent chromium tolerant organisms such as plants, bacteria, unicellular and multicellular fungi and algae are promising eco-friendly alternatives in detoxification and hexavalent chromium removal perspective. This article reviews the bioremediation approaches available for hexavalent chromium detoxification and removal and highlights the strengths and weaknesses of current bioremediation methods.

Hexavalent chromium-reducing plant growth-promoting rhizobacteria are utilized to bio-fortify trivalent chromium in fenugreek by promoting plant development and decreasing the toxicity of hexavalent chromium in the soil

BackgroundFenugreek is known to have good anti-diabetes properties. Moreover, several studies accounted that the trivalent form of chromium [Cr(III)] also have anti-diabetic properties. However, its hexavalent form i.e., Cr(VI) is known to be highly toxic and carcinogenic to living beings and retarded plant growth even if it is present in low concentration in soil. Many plant growth-promoting rhizobacteria (PGPR) are reported to have the potential to reduce the Cr(VI) into Cr(III) in soil. In view of the above, the present objective was designed to effectively utilize Cr(VI) reducing PGPRs for the growth and development of fenugreek plant in Cr(VI) amended soil, apart from reducing Cr(VI) in soil and fortification of Cr(III) in the aerial part of plants. MethodsThe experiment was carried out to evaluate the effect of Cr(VI)-reducing PGPRs viz. Bacillus cereus (SUCR44); Microbacterium sp. (SUCR140); Bacillus thuringiensis (SUCR186) and B. subtilis (SUCR188) on growth, uptake and translocation of Cr as well as other physiological parameters in fenugreek grown under artificially Cr(VI) amended soil (100 mg kg−1 of Cr(VI) in soil). ResultsThe aforementioned concentration of Cr(VI) in soil cause severe reduction in root length (41 %), plant height (43 %), dry root (38 %) and herb biomass (48 %), when compared with control negative (CN; uninoculated plant not grown in Cr(VI) contaminated soil). However, the presence of Microbacterium sp.˗SURC140 (MB) mitigates the Cr toxicity resulting in improved root length (92 %), plant height (86 %), dry root (74 %) and herb biomass (99 %) as compared with control positive (CP; uninoculated plants grown in Cr(VI) contaminated soil). The maximum reduction in bioavailability (82 %) of Cr(VI) in soil and its uptake (50 %) by the plant were also observed in MB-treated plants. However, All Cr(VI)-reducing PGPRs failed to decrease the translocation of Cr to the aerial parts. Moreover, the plant treated with MB observed diminution in relative water content (13 %), electrolyte leakage (16%) and lipid peroxidation (38 %) as well as higher chlorophyll (37 %) carotenoids (17 %) contents and antioxidants (18%) potential. ConclusionThis study demonstrates that MB can lower the Cr(VI) toxicity to the plant by reducing the bioavailable Cr(VI), consequently reducing the Cr(VI) toxicity level in soil and helping in improving the growth and yield of fenugreek. Additionally, Cr(III) uptakes and translocation may improve the effectiveness of fenugreek in treating diabetes.

Hexavalent-Chromium-Induced Oxidative Stress and the Protective Role of Antioxidants against Cellular Toxicity.

Hexavalent chromium is a highly soluble environmental contaminant. It is a widespread anthropogenic chromium species that is 100 times more toxic than trivalent chromium. Leather, chrome plating, coal mining and paint industries are the major sources of hexavalent chromium in water. Hexavalent chromium is widely recognised as a carcinogen and mutagen in humans and other animals. It is also responsible for multiorgan damage, such as kidney damage, liver failure, heart failure, skin disease and lung dysfunction. The fate of the toxicity of hexavalent chromium depends on its oxidation state. The reduction of Cr (VI) to Cr (III) is responsible for the generation of reactive oxygen species (ROS) and chromium intermediate species, such as Cr (V) and Cr (IV). Reactive oxygen species (ROS) are responsible for oxidative tissue damage and the disruption of cell organelles, such as mitochondria, DNA, RNA and protein molecules. Cr (VI)-induced oxidative stress can be neutralised by the antioxidant system in human and animal cells. In this review, the authors summarise the Cr (VI) source, toxicity and antioxidant defence mechanism against Cr (VI)-induced reactive oxygen species (ROS).

Exposure to Environmentally Relevant Concentrations of Polystyrene Microplastics Increases Hexavalent Chromium Toxicity in Aquatic Animals.

The prevalence of hexavalent chromium [Cr(VI)] and microplastics (MPs) is ubiquitous and is considered a threat to aquatic biota. MPs can act as a vector for waterborne metals; however, the combined effects of Cr(VI) and MPs on aquatic organisms are largely unknown. In this study, aquatic model animals, such as rotifers (Brachionus calyciflorus and B. plicatilis), water fleas (Daphnia magna), amphipods (Hyalella azteca), polychaetes (Perinereis aibuhitensis), and zebrafish (Danio rerio) were exposed to environmental concentrations (1, 10, and 100 particles L−1) of 1 μm polystyrene MPs alone, Cr(VI) alone, or Cr(VI) combined with MPs. Following exposure, the potential effects were measured by analyzing basic life endpoints (e.g., survival rate and growth). A significant response to MPs alone was not observed in all animals. However, MPs combined with Cr(VI) concentration-dependently increased Cr(VI) toxicity in two rotifer species. The survival rate of water fleas was significantly reduced upon exposure to Cr(VI) + MPs (100 particles L−1) compared with exposure to Cr(VI) alone, and significantly decreased the number of offspring. Although there was no significant effect on the body length of the amphipod, concentration-dependent decreases in their survival rates were observed. In contrast, no significant change was found in the survival rate of polychaetes; however, their burrowing ability was inhibited by Cr(VI) + MPs (100 particles L−1). Further, larval mortality was increased in response to Cr(VI) + MPs (100 particles L−1) in zebrafish. Taken together, the findings suggest that MPs can exacerbate Cr(VI) toxicity, even at environmental levels.

Hydrogen sulfide manages hexavalent chromium toxicity in wheat and rice seedlings: The role of sulfur assimilation and ascorbate-glutathione cycle

The role of hydrogen sulfide (H2S) is well known in the regulation of abiotic stress such as toxic heavy metal. However, mechanism(s) lying behind this amelioration are still poorly known. Consequently, the present study was focused on the regulation/mitigation of hexavalent chromium (Cr(VI) toxicity by the application of H2S in wheat and rice seedlings. Cr(VI) induced accumulation of reactive oxygen species and caused protein oxidation which negatively affect the plant growth in both the cereal crops. We noticed that Cr(VI) toxicity reduced length of wheat and rice seedlings by 21% and 19%, respectively. These reductions in length of both the cereal crops were positively related with the down-regulation in the ascorbate-glutathione cycle, and were recovered by the application NaHS (a donor of H2S). Though exposure of Cr(VI) slightly stimulated sulfur assimilation but addition of H2S further caused enhancement in sulfur assimilation, suggesting its role in the H2S-mediated Cr(VI) stress tolerance in studied cereal crops. Overall, the results revealed that H2S renders Cr(VI) stress tolerance in wheat and rice seedlings by stimulating sulfur assimilation and ascorbate-glutathione which collectively reduce protein oxidation and thus, improved growth was observed.

Hydrogen sulphide ameliorates hexavalent chromium toxicity in two cereal crops: Role of antioxidant enzymes and proline metabolism.

Chromium pollution in soils is a major threat as it reduces crop yields. Hence, researchers seek methods/strategies which could curtail such losses. We report the role of H2 S in alleviating hexavalent chromium [Cr(VI)] stress in two cereals crops, i.e. wheat and rice seedlings, by estimating various physiological attributes. Cr(VI) reduced shoot and root length in both cereals through increased accumulation of Cr(VI) in root tips and increased in oxidative stress markers, i.e. superoxide radicals (SOR), H2 O2 and lipid peroxidation (as MDA equivalent). Supplementation with H2 S alleviated Cr(VI) toxicity in both cereal crops. Application of H2 S increased tolerance to Cr(VI) stress by protecting photosynthesis and enhancing activity of antioxidant enzymes, particularly glutathione-S-transferase and content of proline. Rice was more resistant to Cr(VI) than wheat seedlings.

Reductive Transformation of Hexavalent Chromium in Ice Decreases Chromium Toxicity in Aquatic Animals.

In this study, the toxicity of hexavalent chromium [Cr(VI)] reduced by citric acid in ice was measured using representative aquatic model invertebrates (i.e., rotifer, water flea, amphipod, and polychaete) and a vertebrate (zebrafish) by analyzing short- and/or long-term endpoints that are frequently applied to each animal. Cr(VI) reduction in the presence of citric acid was markedly enhanced in the ice phase compared to that in an aqueous solution through the freeze concentration effect. The highly concentrated Cr(VI) and citric acid in ice grain boundaries were also confirmed using in situ cryogenic confocal Raman spectroscopy. Overall, exposure to Cr(VI) resulted in higher acute and/or chronic effects on aquatic animals, such as drastic mortality, growth inhibition, and decrease in offspring number, whereas the animals were increasingly tolerant to Cr(VI) that was reduced in the ice phase. Sublethal concentrations of Cr(VI) significantly decreased the antioxidant capacity in the aquatic animals. However, when the same concentrations of Cr(VI) were reduced in ice, these treatments showed no modulation or increase in the antioxidant defense system. Taken together, our results suggest that Cr(VI) reduction into Cr(III) was successfully achieved in ice and that this methodology can decrease the actual toxicity of Cr(VI) in aquatic animals.

OLEANOLIC ACID, A PROSPECTIVE PROTECTIVE AGENT AGAINST BRAIN ENERGY METABOLISM AND OXIDATIVE DYSFUNCTIONS FOLLOWING HEXAVALENT CHROMIUM EXPOSURE IN MICE

Objective: Effect of oleanolic acid against hexavalent chromium-induced altered brain energy metabolism associated with oxidative stress was evaluated in the present study. Methods: Swiss albino mice were divided into three groups, Control (n=6), chromium-treated (n=6), and oleanolic acid (OA) supplemented (n=6). The chromium treated group was orally administered with K2Cr2O7 for 30 days at a dose of 10 mg/kg b.w/day. OA supplementation was given at a dose of 5 mg/kg bw/day for the past 14 days of chromium treatment. Control group received the vehicle only. After the treatment, whole brain was removed for examining the parameters such as pyruvic acid, free amino nitrogen, tissue protein, TCA cycle enzyme activities, NADH dehydrogenase function, and oxidative stress markers. Results: Significant decrease in cerebral pyruvic acid content associated with suppressed malate dehydrogenase and succinate dehydrogenase activities were observed. The NADH dehydrogenase activity was inhibited owing to enhanced accumulation of chromium in cerebral tissue. Depletion of proteins and increased free amino acid nitrogen were accompanied with inhibited cathepsin, pronase and trypsin activities, and increased transaminase function. In addition, GSH content was decreased along with increased lipid peroxidation, oxidized GSSG content, TG/GSSG ratio, carbonylated protein content, and tissue free hydroxyl radical formation. Superoxide dismutase, catalase, glutathione reductase, and glutathione peroxidase were also inhibited by hexavalent chromium. Oleanolic acid supplementation was found to have significant protective effect against brain metabolic and oxidative dysfunctions. Conclusion: The present study elucidated therapeutic efficacy of oleanolic acid against hexavalent chromium toxicity in brain tissue of mice.

Smart PMMA‑cerium oxide anticorrosive coatings: Effect of ceria content on structure and electrochemical properties

Organic-inorganic hybrids are considered an effective and environmentally compliant alternative to chromate-based anticorrosive coatings, currently banned due to the high toxicity of hexavalent chromium. In this work, hybrid nanocomposites based on poly(methyl methacrylate) (PMMA), covalently bonded to cerium oxide nanoparticles through the 2-hydroxyethyl methacrylate (HEMA) coupling agent, were tailored by carefully tuning the inorganic colloidal precursor to provide films with active corrosion protection for metallic substrates. Lithium hydroxide was exploited as oxidizing agent of cerium nitrate to form ceria nanoparticles. Precursor solutions and solid nanocomposites with different LiOH to Ce(NO3)3.6H2O molar ratios were analyzed using spectroscopic and electron microscopy techniques and theoretical simulations. Electrochemical impedance spectroscopy (EIS) was used to study the anticorrosive performance of the coatings on carbon steel and 7075 aluminum alloy. The results showed that increasing amounts of LiOH lead to the formation of higher ceria content and larger primary nanoparticles, ranging from 1.7 to 2.8 nm. The homogenous ~20 μm-thick coatings present excellent anticorrosive performance on carbon steel and AA7075 substrates. Coatings on carbon steel with low LiOH loading (1Li:1Ce) showed long-term durability and high impedance modulus of up to 29 GΩ cm2 after 1 day in saline solution. On the AA7075 alloy, the presence of larger cerium oxide particles at higher LiOH content (3Li:1Ce) acted as effective reservoirs of cerium ions, providing high barrier coatings (395 GΩ cm2) with active corrosion protection.

Antioxidant Revival of Cucumis melo l. Modulates the Oxidant-antioxidant Status on Chromium VI Toxicity Induced Male Albino Rats

Objective: To assess the natural bio-remediation in Hexavalent chromium (Cr VI) toxicity induced rats. Methodology: Male albino rats were given Hexavalent chromium for 42 days to demonstrate that this carcinogenic compund increased LPO while reducing SOD, GSH, GPx, and CAT levels. Rats were randomly divided into five experimental groups (n=6). Group I was a control group as they were only given with clean water to drink. Potassium dichromate 10 g/kg was given to Group II for 42 days. Group III administered with 500 mg/kg of Cucumis melo L, Group IV was administered with 500 mg/kg of Ascorbic acid and Group V was administered with 1:1 ratio of Cucumis melo L and ascorbic acid, respectively. All results were provided as mean SD for a total of 6 rats/ per each group. The significance of differences between male albino rats was determined using the Student’s t-test. At p < 0.05 and p < 0.001, differences were judged significant. Findings: Cucumis melo L. fruit aqueous extracts significantly protected rats from chromiuminduced oxidative damage. The LD50 was determined to be 500 mg/kg using the DPPH assay. As a consequence, we believe that giving fruit extracts to people can help them recover from oxidative stress-related cell damage.Novelty: The administration of chromium 6 resulted in a substantial reduction in body weight and an increase in the organ to body weight ratio. Chromium administration significantly enhanced LPO while decreasing serum levels of SOD, GSH, GPx, and CAT. Cucumis melo aqueous fruit extract was shown to be a protective agent against chromium-induced oxidative stress at a dosage of 500 mg/kg. Synergistic action of silicon nanoparticles and indole acetic acid in alleviation of chromium (CrVI) toxicity in Oryza sativa seedlings

The present study investigates ameliorative effect of silicon nanoparticles (SiNPs) and indole acetic acid (IAA) alone and in combination against hexavalent chromium (CrVI) toxicity in rice seedlings. The results of the study revealed protective effects of SiNPs and IAA against CrVI toxicity. The 100 μM of CrVI imposed toxic effects in rice seedlings at morphological, physiological and biochemical levels which coincided with increased level of intracellular CrVI and declined level of endogenous nitric oxide (NO). The CrVI enhanced levels of superoxide radicals (SOR) (59.51% and 50.1% in shoot and root, respectively) and H2O2 (19.5% and 23.69% in shoot and root, respectively). However, when SiNPs and IAA were applied to plants under CrVI stress, they enhanced tolerance and defence mechanisms as manifested in terms of increased biomass, endogenous NO, photosynthetic pigments, and antioxidants level. It was also noticed that CrVI arrested cell cycle at G2/M phase whereas growth was restored as compared to control when SiNPs and IAA were supplemented. Thus, the hypothesis that combined application of SiNPs and IAA will be effective in alleviating CrVI toxicity is validated from the results of this study. Moreover, in SiNPs and IAA-mediated mitigation of CrVI toxicity, endogenous NO has a positive role. The importance of the study will be that the combination of SiNPs and IAA can be utilized against heavy metal stress and even when supplied alone, they will enhance the crop productivity parameters with and without stress conditions.

The toxicity of hexavalent chromium to soil microbial processes concerning soil properties and aging time

Chromium (Cr) pollution has attracted much attention due to its biological toxicity. However, little is known regarding Cr toxicity to soil microorganisms. The present study assesses the toxicity of Cr(VI) on two microbial processes, potential nitrification rate (PNR) and substrate-induced respiration (SIR), in a wide range of agricultural soils and detected the abundance of soil bacteria, fungi, ammonia-oxidizing bacteria and archaea. The toxicity thresholds of 10% and 50% effective concentrations (EC10 and EC50) for PNR varied by 32.18- and 38.66-fold among different soils, while for SIR they varied by 391.21- and 16.31-fold, respectively. Regression model analysis indicated that for PNR, CEC as a single factor explained 27% of the variation in EC10, with soil clay being the key factor explaining 47.3% of the variation in EC50. For SIR, organic matter and pH were found to be the most vital predictors for EC10 and EC50, explaining 34% and 61.1% of variation, respectively. In addition, extended aging time was found to significantly attenuate the toxicity of Cr on PNR. SIR was mainly driven by total bacteria rather than fungi, while PNR was driven by both AOA and AOB. These results were helpful in deriving soil Cr toxicity threshold based on microbial processes, and provided a theoretical foundation for ecological risk assessments and establishing a soil environmental quality criteria for Cr.

Acute Toxicity of Hexavalent Chromium on Behavioural Parameters and Histopathology Damages in Freshwater Common Carp, Cyprinus carpio

Water sources of almost all regions in India are carrying the unbearable burden of dangerous pollutants. The release of waste products and anthropogenic wastes run-off has contributed to aquatic pollution. The environment has become a storehouse for chemical pollutant which infiltrate into the aquatic environment, including estuarine, thus immobilizing the aquatic biota, among the heavy metals. Chromium is a heavy metal which has both beneficial and harmful effect on organisms. It is highly toxic and carcinogenic. Many industries are disposing of chemical pollutants to the water. These disposals contain many chemicals, including Chromium. The purpose of this review was to check the accumulation and concentration of heavy metals in different organs of freshwater fishes that come in contact with the water contaminated with heavy metals. The subjected fish were exposed to Chromium (Cr) at the sub-lethal level at a concentration of 40mg/L in 96 hours. During the observation period, the fishes shown some behavioral changes like erratic swimming, slow motility, suffocation, and the scales become thin and decolorized. In the gill region, the gill filaments become swollen, and gill rackers become thick, and curling of lamellae was noticed, and it is caused due to the Chromium build-upon gills. At the region of the caudal fin, permanent bending of the tail was observed. The Chromium also affects other vital organs like the spleen and gut region. LC50 was found in 96 hours. The result indicates that Chromium is highly toxic and has deleterious effects on aquatic life. Humans are also affected by the intake of fishes for primary people of those areas where the leading food is fish.

Bioremediation of hexavalent chromium from wastewater using bacteria-a green technology.

Hexavalent chromium has high toxic effect on the ecological system. The aim of the present study is to isolate and characterize the bacteria that can reduce the toxicity of hexavalent chromium from liquid effluent. The bacterial isolate was identified as Bacillus sp. ltds1 after 16S rRNA gene sequencing, and annotation has been submitted in National Center for Biotechnology Information (NCBI) GenBank. The bacterial strain was found able to grow in Luria Broth medium at 100mg/L Cr6+ concentration. A maximum Cr6+ bioremediation (95.24 ± 2.08 %) could be achieved using the said isolate at 40mg/L, pH 7, and inoculum concentration 4 % at 24h. The residual chromium was found in the form of less toxic trivalent chromium (Cr3+), which confirms that the bacterial isolate can transform toxic Cr6+ to non-toxic Cr3+. Fourier Transform Infra-Red (FTIR) study was performed to analyze the functional groups and overall nature of chemical bonds involved in the remediation process, whereas, Energy-Dispersive Spectroscopy (EDS) studies of native and treated cells showed the changes in elemental composition in response to metal stress. Artificial Neural Network (ANN) based prediction model is developed based on experimental points. The developed model was found to predict the bioremediation of Cr6+ at various operating conditions. Particle Swarm Optimization (PSO) is used to optimize the variables like the initial concentration of metal, pH, temperature, and inoculum concentration for the said bacterial strain. The results showed that the isolate could be applied as a potential bioremediation agent for Cr6+ removal.
