Evaluating the effects of desalination antiscalants on phytoplankton and bacterial communities in oligotrophic environments

Antiscalants are used in seawater desalination facilities and are typically discharged with the brine back into the coastal environment. We investigated the impact of two commonly-used antiscalants; polyphosphonates and polycarboxylates-based, on phytoplankton and bacterial communities in the oligotrophic Gulf of Aqaba (Northern Red Sea) using mesocosms simulating different antiscalants concentrations. Our results indicate that the addition of polyphosphonates or poly-carboxylates antiscalants significantly altered phytoplankton and bacterial growth, despite being ‘theoretically’ unavailable for microbial utilization. Specifically, the addition of polyphosphonates resulted in a significant eutrophication leading to increase phytoplankton and heterotrophic bacterial biomass. Contrary, the addition polycarboxylates antiscalants triggered a moderate increase in heterotrophic bacterial biomass and a slight phytoplankton loss, summing to a zero change in bacterioplankton's organic biomass. Altogether, the enhanced autotrophic/heterotrophic biomass attributed to the discharge of polyphosphonates-based antiscalants could impair the pretreatment procedure and increase fouling propensity on the reverse-osmosis membranes. Therefore, it is recommended to minimize/avoid the use of polyphosphonates antiscalants in P-limited oligotrophic seas if the brine is discharged into oligotrophic environments.

Disentangling the lifestyle of bacterial communities in tropical soda lakes

Microbial lifestyles may reveal niche-specific signatures and can contribute to detecting the effects of abiotic fluctuations on biogeochemical cycles. Microorganisms make a tradeoff between optimizing nutrient uptake, improving biomass yield, and overcoming environmental changes according to environmental hostility. Soda lakes are natural environments rich in carbonate and bicarbonate water, resulting in elevated pH and salinities that frequently approach saturation. We hypothesized that during the dry period (elevated pH and salinity), microorganisms try to overcome this harshness by allocating energy to the cellular maintenance process. As these environmental conditions improve during the wet period, microorganisms will begin to invest in nutrient uptake. To test this hypothesis, we evaluated four soda lakes in two different seasons by applying metagenomics combined with flow cytometry (estimate heterotrophic bacterial biomass). The natural occurrence of cyanobacterial blooms in some lakes is the main driver of carbon. These primary producers provide organic carbon that supports heterotrophic bacterial growth and, consequently, a high biomass yield. Under harsh conditions (dry season), cyanobacteria invest in nutrient uptake mechanisms, whereas heterotrophic bacteria allocate energy to survive at the expense of biomass yield. Lakes without cyanobacteria blooms invest in nutrient uptake independent of environmental hostility. This study clarifies the microbial tradeoffs in hostile environments and the impact of this choice on carbon and energy flux in tropical alkaline lakes.

Structure, proximate composition, nutrient dynamics and growth characteristics of Penaeus vannamei in indoor biofloc systems with three different salinities and carbon sources

In the biofloc systems the continuous application of carbon sources led to the assimilation of nitrogenous nutrients and formation of heterotrophic bacterial biomass. The present experiment was conducted in 0.22m3 capacity tanks in the triplicate to find out the structure, proximate composition, nutrient dynamics and growth characteristics of Penaeus vannamei in indoor biofloc systems with three different salinities (35, 20 and 5ppt) and carbon sources (molasses, sugar and rice flour) with an estimated C:N ratio of 15:1. pre salinity acclimatized P. vannamei , specific pathogen free seeds (0.2±0.04g) were stocked @100 numbers per m3 and fed 4 times a day based on the % of body weight (from 22% to 2.5%) and reared for 10 weeks. The results revealed that the carbon application at different salinities significantly influenced the biofloc structure (Scanning Electron Microscopic images of biofloc 4000x) showed round and rod-shaped structures. Proximate composition: crude protein (p<0.001), ether extract (p<0.001) and crude fibre (p<0.05) showed significant difference between treatments. Similarly, elemental carbon composition (p<0.01), nitrogenous waste assimilation of TAN (p<0.01), nitrite (p<0.05), and nitrate (p<0.05) showed significant differences in the treatments vs. the control group. The variables also influenced the microbiota abundance {the metagenomics analysis of biofloc showed Bray-Curtis overall average dissimilarity index of 81%, and it varied from 49% to 100% between treatments & controls} and growth characteristics {average body weight (p<0.001) and survival rate (p<0.01) between treatments and controls} of P. vannamei indoor biofloc systems. Hence, the application of carbon sources at different salinities influenced the biofloc structure, proximate, elemental carbon composition, biofloc microbiota abundance, enhanced the nitrogenous waste assimilation and growth performance of P. vannamei in indoor biofloc systems.

Relevance of Nutrient-Limited Phytoplankton Production and Its Bacterial Remineralization for Carbon and Oxygen Fluxes in the Baltic Sea

The Baltic Sea is prone to oxygen deficiency due to the restricted water exchange with the North Sea in coincidence with a high biological oxygen demand. The partitioning of organic carbon between respiration, accumulation and export is co-determined by phytoplankton primary production and its subsequent bacterial remineralization. Here, we investigated net phytoplankton primary production, heterotrophic bacterial biomass production and dark CO2 fixation by on-board incubations with radiolabeled tracers in the Baltic Proper and in the Gulf of Riga after the main spring bloom. Results show that low phytoplankton standing stocks of ≤1.6 µg chlorphyll a L-1 sustained net primary production of 161-724 mg C m-2 d-1 under nitrogen limitation. Estimates of bacterial carbon remineralization suggest that freshly produced organic carbon was supplied to the aphotic zone at all stations. In the southern Baltic Proper, net primary production exceeded the bacterial carbon demand in the upper mixed layer, suggesting that organic matter derived from nutrient-limited primary production was available for export to bacterial communities below the oxycline. On average, 46% of heterotrophic bacterial production was mediated in oxygen minimum zones, revealing the high importance of organic matter recycling under hypoxic and anoxic conditions for the carbon budget. Dark CO2 fixation of up to 4.33 µg C L-1 d-1 in sulfide-free waters equaled 9-54% of the co-inciding heterotrophic bacterial carbon demand and may have provided another organic carbon source for heterotrophic activity. Substantially higher dark CO2 fixation up to 25.46 µg C L-1 d-1 was determined in sulfidic waters. Since our study was conducted five months after the major Baltic inflow event in winter 2014/2015, potential effects of deep water ventilation could be investigated. In the Gotland Basin, heterotrophic bacterial production in renewed oxygen-rich bottom water was similar to that in the uplifted oxygen-deficient former bottom water, while it was significantly reduced in sulfidic waters. Hence, our results suggest that the removal of hydrogen sufide by inflow events has a high potential to increase bacterial carbon remineralization.

Response of Microbial Communities to Changing Climate Conditions During Summer Cyanobacterial Blooms in the Baltic Sea.

Frequencies and biomass of Baltic Sea cyanobacterial blooms are expected to be higher in future climate conditions, but also of longer duration as a result of increased sea surface temperature. Concurrently, climate predictions indicate a reduced salinity in the Baltic Sea. These climate-driven changes are expected to alter not solely the phytoplankton community but also the role of microbial communities for nutrient remineralization. Here, we present the response of summer plankton communities (filamentous cyanobacteria, picocyanobacteria, and heterotrophic bacteria) to the interplay of increasing temperature (from 16 to 18°C and 20°C) and reduced salinity (from salinity 6.9 to 5.9) in the Baltic Proper (NW Gotland Sea) using a microcosm approach. Warmer temperatures led to an earlier peak of cyanobacterial biomass, while yields were reduced. These conditions caused a decrease of nitrogen-fixers (Dolichospermum sp.) biomass, while non nitrogen-fixers (Pseudanabaena sp.) increased. Salinity reduction did not affect cyanobacterial growth nor community composition. Among heterotrophic bacteria, Actinobacteria showed preference for high temperature, while Gammaproteobacteria thrived at in situ temperature. Heterotrophic bacteria community changed drastically at lower salinity and resembled communities at high temperature. Picocyanobacteria and heterotrophic bacterial biomass had a pronounced increase associated with the decay of filamentous cyanobacteria. This suggests that shifts in community composition of heterotrophic bacteria are influenced both directly by abiotic factors (temperature and salinity) and potentially indirectly by cyanobacteria. Our findings suggest that at warmer temperature, lower yield of photosynthetic cyanobacteria combined with lower proportion of nitrogen-fixers in the community could result in lower carbon export to the marine food web with consequences for the decomposer community of heterotrophic bacteria.

Microbial plankton communities in the coastal southeastern Black Sea: biomass, composition and trophic interactions

Summary We investigated biomass and composition of the pico-, nano- and microplankton communities in a coastal station of the southeastern Black Sea during 2011. We also examined trophic interactions within these communities from size-fractionated dilution experiments in February, June and December. Autotrophic and heterotrophic biomasses showed similar seasonal trends, with a peak in June, but heterotrophs dominated throughout the year. Autotrophic biomass was mainly comprised by nanoflagellates and diatoms in the first half of the year, and by dinoflagellates and Synechococcus spp. in the second half. Heterotrophic biomass was mostly dominated by heterotrophic bacteria, followed by nanoflagellates and microzooplankton. Dilution experiments suggest that nano- and microzooplankton were significant consumers of autotrophs and heterotrophic bacteria. More than 100% of bacterial production was consumed by grazers in all experiments, while 46%, 21% and 30% of daily primary production were consumed in February, June and December, respectively. In February, autotrophs were the main carbon source, but in December, it was heterotrophic bacteria. An intermediate situation was observed in June, with similar carbon flows from autotrophs and heterotrophic bacteria. Size-fraction dilution experiments suggested that heterotrophic nanoflagellates are an important link between the high heterotrophic bacterial biomass and microzooplankton. In summary, these results indicate that nano- and microzooplankton were responsible for comprising a significant fraction of total microbial plankton biomass, standing stocks, growth and grazing processes. This suggests that in 2011, the microbial food web was an important compartment of the planktonic food web in the coastal southeastern Black Sea.

Effect of periphyton (aquamat) on water quality, nitrogen budget, microbial ecology, and growth parameters of Litopenaeus vannamei in a semi-intensive culture system

The present study was conducted to evaluate the role of periphyton in the improvement of water quality, released effluents, microbial ecology and growth parameters of Litopenaeus vannamei in a semi-intensive aquaculture system. The experiment was performed for 120days in 0.66ha pond with and without addition of substrate i.e. treatment (periphyton based) and control pond (feed based), respectively having a stocking density of 20m−2 (PL 15). The significant (P<0.05) reduction in concentration of total ammonia nitrogen (TAN), nitrite-N, and biological oxygen demand (BOD) were found in substrate treated pond when compared with the control. About 39 genera of phytoplankton belonging to Bacillariophyceae (14 genera), Chlorophyceae (12 genera), Cyanophyceae (10 genera) and Euglenophyceae (3 genera) were found adhered on aquamat. Significant (P<0.05) improvement in the growth parameters of L. vannamei and feed utilization (FCR=1.02) were recorded in the substrate treated pond when compared to control (FCR=1.31). At the end of the experiment, a significant reduction (12%) in the concentration of nitrogen present in the effluents (output water) was estimated in substrate treated pond. Significantly low level of nitrogen content was estimated in the sediment of substrate treated pond when compared with the control. Nitrogenous waste generated for producing one ton of shrimp, got reduced by 25% in the periphyton based aquaculture pond. Moreover, a significantly (P<0.05) low total Vibrios counts (1.14×103CFU mL−1) were enumerated when compared with the control pond (1.53×103CFU mL−1) and ratio of total Vibrios counts (TVC) to total heterotrophic bacteria (THB) in the water of substrate treated pond and control was found to be 10.56 and 28.39, respectively. Higher survival rate (93%) of L. vannamei was recorded in the substrate treated pond in comparison to the control (84.4%). In general, addition of substrate to obtain periphyton significantly reduced nitrogenous waste in the effluent, by converting it into heterotrophic bacterial and algal biomass, improved the culture water quality and growth parameters and survival of the shrimp in comparison to the normal feed based aquaculture system. Chytrid parasitism facilitates trophic transfer between bloom-forming cyanobacteria and zooplankton (Daphnia).

Parasites are rarely included in food web studies, although they can strongly alter trophic interactions. In aquatic ecosystems, poorly grazed cyanobacteria often dominate phytoplankton communities, leading to the decoupling of primary and secondary production. Here, we addressed the interface between predator-prey and host-parasite interactions by conducting a life-table experiment, in which four Daphnia galeata genotypes were maintained on quantitatively comparable diets consisting of healthy cyanobacteria or cyanobacteria infected by a fungal (chytrid) parasite. In four out of five fitness parameters, at least one Daphnia genotype performed better on parasitised cyanobacteria than in the absence of infection. Further treatments consisting of purified chytrid zoospores and heterotrophic bacteria suspensions established the causes of improved fitness. First, Daphnia feed on chytrid zoospores which trophically upgrade cyanobacterial carbon. Second, an increase in heterotrophic bacterial biomass, promoted by cyanobacterial decay, provides an additional food source for Daphnia. In addition, chytrid infection induces fragmentation of cyanobacterial filaments, which could render cyanobacteria more edible. Our results demonstrate that chytrid parasitism can sustain zooplankton under cyanobacterial bloom conditions, and exemplify the potential of parasites to alter interactions between trophic levels.

Linking Heterotrophic Microbial Activities with Particle Characteristics in Waters of the Mississippi River Delta in the Aftermath of Hurricane Isaac

Riverine runoff often triggers microbial responses in coastal marine environments, including phytoplankton blooms and enhanced bacterial biomass production that drive the transformation of dissolved and particulate organic matter on its way from land to the deep ocean. We measured concentrations and characteristics of particulate organic matter (POM), concentrations of dissolved organic carbon (DOC), and bacterial community abundance and activities in the water column at three sites near the Mississippi River Delta two weeks after Hurricane Isaac made landfall in late August 2012. River plumes had salinities of >30 PSU and high levels of DOC (210-380 µM), resulting from the storm surge that pushed large quantities of marine waters upstream. Relatively high concentrations of phytoplankton POM and low levels of microbial exopolymeric particles (TEP and CSP) suggested that storm-induced riverine discharge triggered the development of phytoplankton blooms that were in their initial stages at the time of sampling. Surface water POM had C/N ratios of 5-7 and strong protein-like fluorescence signals in the base-extracted POM (BEPOM) fraction at the two sites closer to the river mouth (Stns. TE and MSP). Freshly produced POM triggered a twofold increase in heterotrophic bacterial biomass production (3H-leucine incorporation) and a fourfold increase in bacterial peptide hydrolysis (activities of leucine-aminopeptidase). In contrast, elevated DOC concentrations coincided with only moderate bacterial community activity, suggesting that heterotrophic bacterial metabolism near the Mississippi River Delta in the aftermath of Hurricane Isaac was more closely linked with autochthonous primary production.

Biomass, production, and control of heterotrophic bacterioplankton during a late phytoplankton bloom in the Amundsen Sea Polynya, Antarctica

We investigated the heterotrophic bacterial biomass and production in February 2012, in four habitats (a polynya, sea-ice zone, ice shelf, and the open sea) in the Amundsen Sea to determine the spatial distribution, controlling factors, and ecological role of the bacteria during a late phytoplankton bloom by Phaeocystis antarctica. Bacterial abundance (BA) and production (BP) were highest at the center of the polynya, and both were significantly correlated with phytoplankton biomass. BP accounted for average 17% of the organic carbon produced by phytoplankton primary production (PP), which is higher than the average BP:PP ratio reported in most open ocean. The abundance of heterotrophic nanoflagellates (HNF) was correlated with the BA, and the average bacteria:HNF ratio (260) was lower than the values reported in most marine environments (400–1000), including the Ross Sea Polynya (800). Evidence for a tight coupling of bacteria and phytoplankton activities on the one hand and intense HNF grazing on bacteria on the other could be found in the high BP:PP and low bacteria:HNF ratios, respectively. Interestingly, these data were accompanied by low particulate carbon export fluxes measured during the late Phaeocystis bloom. Together, these results indicated that the microbial loop plays a significant role in the biogeochemical carbon cycle and food web processes in the Amundsen Sea Polynya.

Algae alleviate carbon limitation of heterotrophic bacteria in a boreal peatland

Summary In most high‐latitude wetlands, carbon accumulation as peat represents a balance between plant net primary productivity and heterotrophic decomposition. We hypothesized that this assessment of ecosystem metabolism is incomplete as it does not include information on energy inputs from microalgae, which form complex biofilms with heterotrophic micro‐organisms on the peat surface. To investigate the potential for algae (and associated exudates) to support heterotrophic metabolism under ambient and elevated nutrient levels, we conducted a factorial enrichment of nutrients (nitrogen and phosphorus) and carbon (glucose) in mesocosms with and without the presence of algae (using light‐transparent and dark treatments, respectively) in an Alaskan fen. We measured respiration rates and changes in bacterial biomass to characterize the response of heterotrophic bacteria to our experimental treatments. During incubation assays, algae released up to 38% of their net productivity as exudates and there was a positive relationship between algal production and concentrations of dissolved organic carbon inside mesocosms. Elevated algal productivity in the presence of nutrient enrichment stimulated heterotrophic bacterial respiration and biomass. These responses did not occur with nutrient enrichment in the dark (without algae). The response of bacteria to algae was similar in magnitude to bacterial responses to glucose enrichment. Synthesis. We conclude that bacteria in this boreal fen were primarily limited by labile carbon, and this constraint was alleviated in the presence of elevated algal production. Consequently, algae may facilitate hotspots of microbial activity in northern peatlands, especially in conditions of greater nutrient availability associated with more variable hydrology expected for this region with ongoing climate change.

Linking summer conditions to CO2 undersaturation and CO2 influx in a subtropical coastal lake

In this study, we tested the hypothesis that pCO2 and air–water CO2 fluxes in the surface waters of a subtropical lake vary on two time scales (diel and seasonally) and that CO2 concentrations would decrease during the day and in summer. We estimated the variability of pCO2 and the air–water CO2 flux from pH-alkalinity in four 48-h periods that were representative of each subtropical season. There was high variability in pCO2 and the air–water gas flux over 48 h, but there was no clear pattern between day and night. CO2 concentrations showed a strong positive correlation with heterotrophic bacterial biomass and a negative correlation with dissolved organic carbon concentrations and water temperature. The lake was predominantly diel and seasonally CO2 supersaturated; the highest CO2 efflux was observed in the spring and a CO2 influx was observed in summer. Our hypothesis was confirmed; pCO2 was lowest in summer and during the daytime in spring and summer due to physical and biological conditions that favoured photosynthetic activities. These findings suggest that temporal shifts in the microbial community and meteorological variables, which are indirectly related to temperature, may be important drivers of CO2 concentrations in Peri Lake. In conclusion, pCO2 and the air–water CO2 flux vary temporally (diel and seasonally) in the littoral zone of this subtropical coastal lake, with shifts between CO2 influx and efflux throughout the sampling periods.

Microbial metabolism of transparent exopolymer particles during the summer months along a eutrophic estuary system.

This study explores the role of transparent exopolymer particles (TEP) as an additional carbon source for heterotrophic microbial activity in the eutrophic Qishon estuary. From the coastal station and upstream the estuary; TEP concentrations, β-glucosidase activity, bacterial production and abundance have gradually increased. TEP were often found as bio-aggregates, scaffolding algae, detritus matter and bacteria that likely formed “hotspots” for enhance microbial activity. To further demonstrate the link between TEP and heterotrophic bacterial activity, confined incubations with ambient and polysaccharide-enriched estuary water were carried out. Following polysaccharide addition, elevated (~50%) β-glucosidase activity rates were observed, leading to TEP hydrolysis. This newly formed bioavailable carbon resulted in significantly higher growth rates, with up to a 5-fold increase in heterotrophic bacterial biomass, comprising mostly high nucleic acid content bacteria. Taking together the findings from this research, we conclude that even in highly eutrophic environments heterotrophic bacteria may still be carbon limited. Further, TEP as a polysaccharide matrix can act as a metabolic surrogate, adding fresh bioavailable carbon through tight associations with bacteria in eutrophic ecosystems such as the Qishon estuary.

Spatial and temporal variability of small-sized copepod assemblages in a shallow semi-enclosed embayment (Kalloni Gulf, NE Mediterranean Sea)

The small-sized copepod community structure (&lt;1 mm) and its response to environmental variability was examined during an annual cycle in Kalloni Gulf, a semi-enclosed coastal system in the NE Aegean Sea. Α 90 µm net was used in order to adequately sample the smaller copepod fractions. In the copepod ecology the spatial patterns dominated over the seasonal. Total copepod abundance increased towards the inner gulf area while diversity indices followed the opposite trend. The inner gulf copepod assemblage area was affected by the increasing magnitude of eutrophication, characterized by high abundance values, low species richness and dominance of a single species (e.g.Oithona nana). A clear seasonal succession of species was observed, ordered by temperature, salinity, chlorophyll a, heterotrophic bacterial biomass, organic and inorganic nutrients. Freshwater inputs and the subsequent nutrient inflow was a dominant phenomenon enhancing copepod production and negatively affecting the biodiversity of the assemblage. The great contribution of copepod larval stages in the total community and their year-round presence indicate the continuous production of copepods during the annual cycle, highlight the importance of the smaller size fraction and encourage the use of finer mesh-size nets when assessing the structure and dynamics of copepod communities.

Phylogenetic constraints on elemental stoichiometry and resource allocation in heterotrophic marine bacteria

The objective of this study was to evaluate the contribution of evolutionary history to variation in the biomass stoichiometry and underlying biochemical allocation patterns of heterotrophic marine bacteria. We hypothesized that phylogeny significantly constrains biochemical allocation strategy and elemental composition among taxa of heterotrophic marine bacteria. Using a 'common-garden' experimental design, we detected significant interspecific variation in stoichiometry, macromolecule allocation and growth rate among 13 strains of marine Proteobacteria. However, this variation was not well explained by 16S rRNA phylogenetic relationships or differences in growth rate. Heterotrophic bacteria likely experience C-limitation when consuming resources in Redfield proportions, which consequently decouples growth rate from allocation to rRNA and biomass P content. Accordingly, overall bacterial C : nutrient ratios (C : P = 77, C : N = 4.9) were lower than Redfield proportions, whereas the average N : P ratio of 17 was consistent with the Redfield ratio. Our results suggest that strain-level diversity is an important driver of variation in the C : N : P ratios of heterotrophic bacterial biomass and that the potential importance of non-nucleic acid pools of P warrants further investigation. Continued work clarifying the range and controls on the stoichiometry of heterotrophic marine bacteria will help improve understanding and predictions of global ocean C, N and P dynamics.

Effect of different biofloc levels on microbial activity, water quality and performance of Litopenaeus vannamei in a tank system operated with no water exchange

In zero-exchange superintensive culture systems, flocculated particles (bioflocs) accumulate in the water column. Consequently, some control over the concentration of these particles must be performed. The objective of this study is to evaluate the effects of three concentrations of bioflocs on microbial activity, selected water quality indicators and performance of Litopenaeus vannamei in a tank system operated with no water exchange. A 44-day study was conducted with juvenile (6.8g) shrimp stocked in twelve 850L tanks at a stocking density of 459shrimpm−3. Biofloc levels were expressed as three presets of total suspended solids (TSS) concentrations, as follows: 200mgL−1 (T200), 400–600mgL−1 (T400–600), and 800–1000mgL−1 (T800–1000). TSS levels were controlled by attaching a 40L settling tank to each culture tank. Reduction of TSS to concentrations close to 200mgL−1 decreased the time of bacterial cell residence and significantly reduced the nitrification rates in the water (P<0.05). The tanks in the T200 treatment had a greater variability of ammonia and nitrite (P<0.05), which led to the need to increase the C:N ratio of the organic substrate to control ammonia through its assimilation into heterotrophic bacterial biomass. But the higher production of heterotrophic bacteria in T200 (P<0.05) increased the dissolved oxygen demand. Nitrification rates were higher (P<0.05) in tanks with TSS concentrations above 400mgL−1, and ammonia and nitrite were significantly lower than in the T200 tanks. We suggest that ammonia and nitrite in the T400–600 and T800–1000 tanks were controlled primarily by nitrifying bacteria, which provided higher stability of these parameters and of dissolved oxygen. Regarding shrimp performance, the reduction of TSS to levels close to 200mgL−1 was associated with better nutritional quality of bioflocs. Nevertheless, differences in biofloc levels and nutritional quality were not sufficient to affect the weight gain by shrimp. The rate of shrimp survival and the final shrimp biomass were lower (P<0.05) when the TSS concentrations were higher than 800mgL−1. Analysis of the shrimps’ gills showed a higher degree of occlusion in the T800–1000 treatment (P<0.05), which suggests that the shrimp have an intolerance to environments with a solids concentration above 800mgL−1. Our results show that intermediate levels of bioflocs (TSS between 400 and 600mgL−1) appear to be more suitable to superintensive culture of L. vannamei since they create factors propitious for maintaining the system’s productivity and stability

Bacteria contribute to Artemia nutrition in algae-limited conditions: A laboratory study

We investigated the effect of the stimulation of bacterial growth on Artemia performance in combination with a standard and with a low algal feeding regime. In both regimes, organic carbon (supplied as sucrose or soluble potato starch) and 15N labeled inorganic nitrogen (supplied as NaNO3) were used to stimulate bacterial growth in the Artemia cultures at C/N ratio 10 and 50. After a culture period of 15days, significantly improved biomass production was obtained in all treatments with the low algae feeding regime, supplemented by carbohydrate addition. In addition, results of 15N accumulation and fatty acid analysis in Artemia indicated that Artemia utilized more bacteria in algae-limited conditions. Our study shows that bacteria can be used as a nutrient source for Artemia compensating for suboptimal algae supply. In Artemia pond cultures, carbohydrate addition may hence potentially be used to stimulate the conversion of nitrogen waste into heterotrophic bacterial biomass. This can be converted into protein-rich Artemia biomass, especially when algae are in sub-optimal supply. These findings open perspectives for alternative Artemia pond production protocols, in addition to the present management procedures that exclusively focus on phytoplankton blooms as nutrient source to sustain dense Artemia populations.

Year-round patterns in bacterial production and biomass in Lake Simcoe, Ontario, Canada: are heterotrophic bacteria a significant contributor to low hypolimnetic oxygen?

Bacteria serve an important function in aquatic environments and are associated with nutrient regeneration, carbon (C) metabolism, and secondary production. This is the first study to measure heterotrophic bacterial production, abundance, biomass, and biovolume in Lake Simcoe, Ontario. Excessive phosphorus loading resulting in low hypolimnetic oxygen concentrations has impacted the cold-water fishery in Lake Simcoe. We tested the hypothesis that bacteria contributed to dissolved oxygen declines in Lake Simcoe and examined the environmental factors impacting bacterial activity. Spatial and temporal variations in the microbial community were measured from June 2010 to July 2011. A dual-isotope method (3H-TdR and 14C-leu) was used, resulting in mean (± standard deviation) annual epilimnetic bacterial production estimates of 0.130 ± 0.173 and 0.268 ± 0.304 μg C L−1 h−1, respectively. The mean annual bacterial abundance was 1.49 ± 1.53 cells × 109 L−1, with a mean biomass of 1.21 ± 1.34 μg C L−1 and a mean biovolume of 0.0043 ± 0.0030 μm3 cell−1. These estimates had distinct seasonal patterns, with consistently lower bacterial activity in the winter relative to the spring, summer, and fall. Differences between epilimnetic and hypolimnetic bacterial activity were inconsistent for the 4 bacterial parameters measured. Lake temperature, chlorophyll a, and dissolved organic C concentrations were the most significant factors influencing the annual epilimnetic patterns in the examined bacterial parameters. Annual bacterial production was low in Lake Simcoe and does not seem to be a major contributor to the low hypolimnetic oxygen concentrations in the lake.

Oceanic heterotrophic bacterial nutrition by semilabile DOM as revealed by data assimilative modeling

Previous studies have focused on the role of labile dissolved organic matter (DOM) (defined as turnover time of ~1 d) in supporting heterotrophic bacterial production, but have mostly neglected semilabile DOM (defined as turnover time of ~100 to 1000 d) as a potential substrate for heterotrophic bacterial growth. To test the hypothesis that semilabile DOM supports substantial amounts of heterotrophic bacterial production in the open ocean, we constructed a 1-dimensional epipelagic ecosystem model and applied it to 3 open ocean sites: the Arabian Sea, Equatorial Pacific and Station ALOHA in the North Pacific Subtropical Gyre. The model tracks carbon, nitrogen and phosphorus with flexible stoichiometry. This study used a large number of observations, including measurements of heterotrophic bacterial production rates and standing stocks, and DOM concentra- tion data, to rigorously test and constrain model output. Data assimilation was successfully applied to optimize the model parameters and resulted in simultaneous representation of observed nitrate, phosphate, phytoplankton and zooplankton biomass, primary production, heterotrophic bacterial biomass and production, DOM, and suspended and sinking particulate organic matter. Across the 3 ocean ecosystems examined, the data assimilation suggests semilabile DOM may support 17 to 40% of heterotrophic bacterial carbon demand. In an experiment where bacteria only utilize labile DOM, and with more of the DOM production assigned to labile DOM, the model poorly represented the observations. These results suggest that semilabile DOM may play an important role in sustaining heterotrophic bacterial growth in diverse regions of the open ocean.

Influence of allochthonous organic matter on bacterioplankton biomass and activity in a eutrophic, sub-tropical estuary

Heterotrophic bacterial and phytoplankton biomass, production, specific growth rates, and growth efficiencies were studied in the Northern region of the Cananéia–Iguape estuarine system, which has recently experienced an intense eutrophication due to anthropogenic causes. Two surveys were carried out during spring and neap tide periods of the dry season of 2005 and the rainy season of 2006. This region receives large freshwater inputs with organic seston and phosphate concentrations that reach as high as 1.0 mg l −1 and 20.0 μM, respectively. Strong decreasing gradients of seston and dissolved inorganic nutrients were observed from the river/estuary boundary to the estuary/coastal interface. Gradients were also observed in phytoplankton and bacterial production rates. The production rates of phytoplankton were 5.6-fold higher (mean 8.5 μg C l −1 h −1) during the dry season. Primary production rates (PP) positively correlated with salinity and euphotic depth, indicating that phytoplankton productivity was light-limited. On the other hand, bacterial biomass (BB) and production rates (BP) were 1.9- and 3.7-fold higher, respectively, during the rainy season, with mean values of up to 40.4 μg C l −1 and 7.9 μg C l −1 h −1, respectively. Despite such a high BP, bacterial abundance remained <2 × 10 6 cells ml −1, indicating that bacterial production and removal were coupled. Mean specific growth rates ranged between 0.9 and 5.5 d −1. BP was inversely correlated with salinity and positively correlated with temperature, organic matter, exopolymer particles, and particulate-attached bacteria; this last accounted for as much as 89.6% of the total abundance. During the rainy season, BP was generally much higher than PP, and values of BP/PP > 20 were registered during high freshwater input, suggesting that under these conditions, bacterial activity was predominantly supported by allochthonous inputs of organic carbon. In addition, BB probably represented the main pathway for the synthesis of high-quality (low C:N) biomass that may have been available to the heterotrophic components of the plankton food web, particularly nanoheterotrophs.
