Polycyclic Aromatic Hydrocarbons (PAHs) Mediate Transcriptional Activation of the ATP Binding Cassette Transporter ABCB6 Gene via the Aryl Hydrocarbon Receptor (AhR)

Liver is endowed with a mechanism to induce hepatic cytochromes P450 (CYP450s) in response to therapeutic drugs and environmental contaminants, leading to increased detoxification and elimination of the xenobiotics. Each CYP450 is composed of an apoprotein moiety and a heme prosthetic group, which is required for CYP450 activity. Thus, under conditions of CYP450 induction, there is a coordinate increase in heme biosynthesis to compensate for the increased expression of CYP450s. ABCB6, a mitochondrial ATP binding cassette transporter, which regulates coproporphyrinogen transport from the cytoplasm into the mitochondria to complete heme biosynthesis, represents a previously unrecognized rate-limiting step in heme biosynthesis. However, it is not known if exposure to drugs and environmental contaminants induces ABCB6 expression, to assure an adequate and apparently coordinated supply of heme for the generation of functional cytochrome holoprotein. In the present study, we demonstrate that polycyclic aromatic hydrocarbons (PAHs), the widely distributed environmental toxicants shown to induce porphyrin accumulation causing hepatic porphyria, up-regulate ABCB6 expression in both mice and humans. Using siRNA technology and Abcb6 knock-out mice, we demonstrate that PAH-mediated increase in hepatic porphyrins is compromised in the absence of ABCB6. Moreover, in vivo studies in aryl hydrocarbon receptor (AhR) knock-out mice demonstrate that PAH induction of ABCB6 is mediated by AhR. Promoter activation studies combined with electrophoretic mobility shift assay and chromatin immunoprecipitation assay demonstrate direct interactions between the AhR binding sites in the ABCB6 promoter and the AhR receptor, implicating drug activation mechanisms for ABCB6 similar to those found in inducible cytochrome P450s. These studies are the first to describe direct transcriptional activation of both mouse and human ABCB6 by xenobiotics.

Hexachlorobenzene-induced alterations on neutral and acidic sphingomyelinases and serine palmitoyltransferase activities. A time course study in two strains of rats

Hexachlorobenzene (HCB) induces porphyria both in humans and rodents, and hepatocarcinoma in rodents. In a previous work we observed that HCB produces a continuous decrease in hepatic sphingomyelin (SM) content in Wistar rats. A distinguishing characteristic of sphingolipids breakdown products is their participation in anti-proliferative and apoptotic processes and in the suppression of oncogenesis. As a first step to elucidate the role of SM decrease in the hepatotoxicity induced by HCB, the present study evaluates the metabolic causes of the continuous decrease in hepatic SM content observed in Wistar rats with HCB intoxication, and its relation with porphyria development. For this purpose, the time-course (3, 7, 15, 21 and 28 days) of the effects of HCB on hepatic SM levels and on some of the enzymes of SM synthesis (serine palmitoyltransferase, SPT) and catabolism (sphingomyelinases, SMases) was followed, using two strains of rats differing in their susceptibility to acquire porphyria: Chbb THOM (low) and Wistar (high). HCB (1 g kg −1 b.w. per day) was administered by gastric intubation as an aqueous suspension. After 5 days of HCB treatment, animals were allowed a 2-day recovery period without HCB administration. Two phases in the HCB-induced damages to sphingolipid metabolism were observed. The first stage (7 days of treatment), common to both strains of rats, was characterized by a decrease in hepatic SM levels (17–25%) and in SPT activity (50–43%), while strain differences were found for the later stage. In Chbb THOM rats, hepatic SM content was restored to normal values concomitantly with an increase in SPT activity (44%, at day 28), and without any increase in SM catabolism. In addition, the level of the other phospholipids was not altered. In Wistar rats, hepatic SM levels decreased continuously throughout the experiment, accompanied by increases in SPT, acidic sphingomyelinase (A-SMase) and neutral sphingomyelinase (N-SMase) activities (86, 28.5 and 78% increase, respectively). A role for glutathione (GSH) in the interstrain differences or a direct effect of HCB on SM metabolism was not found. The present study: (a) demonstrates that N-SMase, A-SMase, and SPT are some of the enzymes that play a role in the HCB-induced decrease of hepatic SM content; (b) finds that HCB-induced alterations of SM metabolism do not correlate with HCB-induced accumulation of hepatic porphyrins; and (c) proposes a link between HCB-induced alterations in phospholipid pattern and in SM metabolism. The increased SM hydrolysis produced as a consequence of SMases induction could be regarded as a cellular response to liver injury elicited by HCB, perhaps acting through the activation of SM signal transduction pathway delaying the proliferative processes observed after long-term treatment with HCB in some rodent species. However, such protective mechanism appears to be strain-dependent.

Spontaneous Porphyria of the Long–Evans Cinnamon Rat: An Animal Model of Wilson's Disease

To confirm and extend our previous microspectrophotometric observations of 30-week-old male Long–Evans Cinnamon (LEC) rats, an animal model of human Wilson's disease, we analyzed the porphyrin patterns of the organs, urine, and plasma of LEC rats. Abnormal accumulation of porphyrins, especially highly carboxylated porphyrins (uro- and heptaporphyrin), in the kidneys and liver was seen in male and female LEC rats aged 30 weeks and also in 10-week-old rats, before the onset of spontaneous hepatic dysfunction. Accumulation of copper and iron in the kidneys was not observed in the 10-week-old rats. Massive accumulation of porphyrins was observed only in the kidneys of the 30-week-old male LEC rat, indicating that this symptom is related to sex and age. Renal accumulation of porphyrins was reflected in the rate of urinary porphyrin excretion. Hepatic accumulation of porphyrins appeared to be independent of sex and age. These results indicate that neither renal nor hepatic porphyrin accumulation is the result of renal deposition of metals or of spontaneous hepatic dysfunction and that porphyrinuria in the LEC rat is closely related to the renal accumulation of porphyrins. In contrast to these organs, a reduction in the porphyrin levels was observed in the brain of the LEC rat. This was independent of sex and age. The present work stresses the existence of an abnormal heme metabolism in the LEC rat, and thus, the necessity to study the heme metabolism in human Wilson's disease is strongly suggested.

Formation of N-alkylprotoporphyrin IX after interaction of porphyrinogenic xenobiotics with rat liver microsomes

Mechanism-based inactivation of selected cytochrome P450 (CYP) isozymes by xenobiotics can lead to N-alkylation of the heme moiety and the formation of ferrochelatase-inhibitory N-alkylprotoporphyrin IX (N-alkylPP), resulting in hepatic porphyrin accumulation and porphyria. Therefore, it is important to determine which of the CYP isozymes are responsible for N-alkylPP formation. Our objective was to determine if N-alkylPPs could be isolated from rat liver microsomes following interaction in vitro with porphyrinogenic xenobiotics, and if so, whether they are produced in amounts sufficient for further studies using microsomes containing cDNA-expressed human hepatic CYP isozymes. The in vitro formation of N-alkylPP was observed following incubation of 3-[(arylthio)ethyl]sydnone; allylisopropylacetamide; and 3,5-diethoxycarbonyl-1,4-dihydro-2,6-dimethyl-4-ethylpyridine; but not 1-aminobenzotriazole, with rat hepatic microsomes. However the overall N-alkylPP yield per nmol CYP in vitro was much lower than previously observed in vivo. It was concluded that microsomal preparations containing cDNA-expressed CYP isozymes do not contain sufficient CYP for in vitro studies designed to isolate N-alkylPP and human liver microsomes would be a more appropriate source of N-alkylPP because they contain higher levels of total CYP.

Effects of selected antihypertensives and analgesics on hepatic porphyrin accumulation : Implications for clinical porphyria

When patients with acute porphyrias are treated with antihypertensives and analgesics, they could be placed at increased risk of developing porphyric attacks, since little is known about the potential for many of these drugs to induce these attacks. We used primary chick embryo liver cells, which maintain intact heme synthesis and regulation, to study the effects of antihypertensives and analgesics on porphyrin accumulation. Cells were treated with desferrioxamine to block heme synthesis partially, simulating conditions encountered in porphyric patients. Typically, cells were treated for 20 hr with the test drugs (3.16 to 1000 μM), along with desferrioxamine. Porphyrins were measured spectrofluorometrically, as uro-, copro,- and protoporphyrin. The evaluated drugs included six antihypertensives (two calcium channel blockers, an angiotensin receptor antagonist, and three inhibitors of angiotensin converting enzyme) and eight analgesics. Of the calcium channel blockers tested, nifedipine greatly increased porphyrin accumulation, whereas diltiazem caused only a slight increase. Losartan (an angiotensin receptor antagonist), captopril, or lisinopril (two angiotensin converting enzyme inhibitors) produced only small increases in porphyrin accumulation. In contrast, enalapril (another angiotensin converting enzyme inhibitor) substantially increased porphyrin accumulation when given in high concentrations. Among the analgesics tested, fentanyl and tramadol produced the highest porphyrin accumulations. Nalbuphine, hydrocodone, oxycodone, and dezocine were moderately or weakly porphyrogenic, whereas buprenorphine and morphine did not increase porphyrin accumulation. These studies suggest that patients with acute porphyrias may be at greater risk for developing porphyric attacks when treated with nifedipine (compared with diltiazem), enalapril (compared with captopril or lisinopril), and tramadol (compared with the other analgesics).

Relative Potencies of Polychlorinated Dibenzo-p-dioxins, Dibenzofurans, and Biphenyls Derived from Hepatic Porphyrin Accumulation in Mice

Hepatic porphyrin accumulation was studied after subchronic dosing of female B6C3F1 mice by gavage with single congeners of polychlorinated or polybrominated dibenzo-p-dioxins (PCDDs, PBDDs), polychlorinated dibenzofurans (PCDFs), and polychlorinated biphenyls (PCBs). Quantitative hepatic porphyrin profile analyses in selected samples showed uroporphyrin and heptacarboxylporphyrin as the main porphyrins detected. Dose-dependent increases in total hepatic porphyrins were found for all congeners tested. At lower dose levels, relative potencies, based on administered dose as well as target tissue dose, of PCDDs, PCDFs, and coplanar PCBs, using 2,3,7,8-tetrachlorodibenzo-p-dioxin as a reference compound, were in the same range as those previously derived from the induction of hepatic CYP1A1 and CYP1A2 enzyme activities. CYP1A2 has been reported to be involved in the oxidation of uroporphyringen III to uroporphyrin III. All these facts suggest the involvement of an aryl hydrocarbon receptor-mediated mechanism in hepatic porphyrin accumulation, possibly via CYP1A2. However, the relative potencies of the mono-ortho-substituted PCBs were higher for hepatic porphyrin accumulation than for hepatic CYP1A1 and CYP1A2 induction. In addition, hepatic porphyrin accumulation was the highest after exposure to mono-ortho-PCBs. Since mono-ortho-substituted PCBs induce the phenobarbital-inducible CYP2B isoforms of cytochrome P450, an additional induction of δ-aminolevulinic acid synthetase may also contribute to hepatic porphyrin accumulation following subchronic exposure to these particular congeners. Relative potencies derived from hepatic porphyrin accumulation after PCDD, PCDF, or coplanar PCB administration are a useful tool in risk assessment. However, the higher relative potencies of the mono-ortho-substituted PCBs have important implications for risk assessment of these compounds.

Synergistic effect of 2,2',4,4',5,5'-hexachlorobiphenyl and 2,3,7,8-tetrachlorodibenzo-p-dioxin on hepatic porphyrin levels in the rat.

We studied the effect of polychlorinated biphenyls (PCBs) on hepatic porphyrin accumulation in female Sprague-Dawley rats by feeding them diets containing 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), 2,2',4,4',5,5'-hexachlorobiphenyl (PCB 153), 2,3,3',4,4',5-hexachlorobiphenyl (PCB 156), 3,3',4,4',5-pentachlorobiphenyl (PCB 126), or combinations of the single PCB congeners with TCDD for 13 weeks. A dose-dependent increase in hepatic porphyrin accumulation occurred after TCDD, PCB 126, or PCB 156 administration, reaching maximal levels of about twice control values. The lowest dose levels for which a significant increase in hepatic porphyrin accumulation was found were 0.7 microgram TCDD/kg diet, 50 micrograms PCB 126/kg diet, or 6 mg PCB 156/kg diet. These doses are equivalent to 47 ng TCDD/kg/day, 3.2 micrograms PCB 126/kg/day, and 365 micrograms PCB 156/kg/day. Relative potencies for hepatic porphyrin accumulation, using TCDD as a reference, ranged from 0.015 to 0.06 for PCB 126 and from 0.0001 to 0.0003 for PCB 156. CYP1A2 activities significantly correlated with hepatic porphyrin levels, with coefficients of 0.629, 0.483, or 0.808 for TCDD, PCB 126, or PCB 156, respectively. Administration of PCB 153 alone did not result in hepatic porphyrin accumulation. Co-administration of PCB 153 and TCDD revealed a strong synergistic effect on porphyrin accumulation (about 800 times control levels). This synergistic effect was significant in rats fed diets containing any combination of PCB 153 with TCDD. Uroporphyrin III and heptacarboxylic porphyrin were accumulated in porphyrinogenic livers. These results suggest that TCDD induction of CYP1A2 may be involved, leading to oxidation of uroporphyrinogen III to uroporphyrin III, in combination with an increase in delta-aminolevulinic acid synthetase induced by PCB 153. Under porphyrinogenic conditions, an inhibitor of CYP1A2 activity may also be formed. The interactive effects on porphyrin accumulation after co-administration of dioxinlike and non-dioxinlike compounds may have significant implications for the risk assessment of these chemicals.

Time-dependent porphyric response in mice subchronically exposed to arsenic

1. A time-course study was carried out in mice subchronically exposed to As III (as sodium arsenite) or As V (as sodium arsenate), via drinking water, relating the pattern of urinary porphyrin excretion to the renal and hepatic enzyme activities of porphobilinogen deaminase (PBGD), uroporphyrinogen III synthetase (URO III-S), uroporphyrinogen decarboxylase (URO-D) and coproporphyrinogen oxidase (COPRO-O), as well as to the hepatic porphyrin accumulation in the treated animals. 2. A time-dependent, wave-like porphyric response was found in mice exposed to As V, and the increases seen in total urinary porphyrins (at 3 weeks of exposure) corresponded to an increased activity of PBGD and Uro III-S in liver. 3. Significant decreases in renal URO-D and hepatic and renal COPRO-O activities were found in treated mice; these inhibitions were more pronounced in animals exposed to As III. 4. The combination of these enzymic effects may explain the time-dependent porphyric response of mice subchronically exposed to As. Finally, the relative magnitudes of URO-D and COPRO-O inhibitions may determine the pattern of porphyrin concentration observed in urine and tissues. 5. The decrease in renal URO-D activity may help to explain the inversion in the coproporphyrin/uroporphyrin ratio previously reported in humans chronically exposed to As; however, there were differences between the urinary porphyrin profiles found in both species. The possible reasons for the similarities and differences are briefly discussed.

The Role of Oxidative Metabolism in Hexachlorobenzene-Induced Porphyria and Thyroid Hormone Homeostasis: A Comparison with Pentachlorobenzene in a 13-Week Feeding Study

Hexachlorobenzene (HCB) induces a broad spectrum of effects including disturbances in the heme synthesis (porphyria) and in thyroid hormone homeostasis. For most of its effects, biotransformation of the parent compound seems to be a prerequisite. The present study was designed to assess the relevance of the oxidative metabolites in HCB-induced toxicity, with special attention to the role of the reactive tetrachlorobenzoquinone (TCBQ). To this end, toxicity and biotransformation of HCR were compared with those of pentachlorobenzene (PCB), since this chemical is oxidized to the same products as HCB, i.e., pentachlorophenol (PCP) and TCBQ. Female Wistar rats received diets containing different dose levels of HCR or PCB for 13 weeks, with or without cotreatment with triacetyloleandomycin (TAO), a selective inhibitor of cytochrome P450IIIA1/2. Rats treated with HCB (high dose) had significantly elevated levels of urinary porphyrins from the 4th week on and had a significant hepatic accumulation of porphyrins at the end of the study. Both urinary porphyrin excretion and hepatic porphyrin accumulation were greatly inhibited in rats receiving cotreatment with HCB and TAO. However, the inhibition of HCB-induced porphyria by TAO cannot be explained by a diminished formation of the highly reactive TCBQ, since rats treated with a high dose of PCB, which had a several fold higher urinary excretion of PCP and TCHQ compared to a high dose of HCB, did not develop porphyria. Instead, the present study points to the involvement of a putative reactive intermediate in the primary oxidative step in HCB-induced porphyria, since based on paired observations of individual rats, the degree of porphyria was correlated to a high degree with excretion of PCP, whereas correlation of porphyria with early excretion of TCHQ was much weaker. This finding fits well with the fact that the mechanisms of oxidation of HCB to PCP and PCB to PCP are different. Both I-ICB and PCB were oxidized to PCP and tetrachlorohydroquinone (TCHQ), the reduced analog of TCBQ. Cytochrome P450IIIA1/2 appears to be involved in the conversion of HCB and PCB, since cotreatment of TAO resulted in a strongly diminished urinary excretion of PCP and TCHQ. Treatment with HCB as well as PCB results in disturbances of retinoid and thyroid hormone homeostasis. These effects, which have also been reported after exposure to polychlorinated biphenyls, originate from interference of hydroxylated metabolites (notably PCP) with the plasma thyroxine transport protein, transthyretine, and since this metabolite is formed from both HCB and PCB, this results in the same toxicity for both compounds.

Hepatic protoporphyria is associated with a decrease in ligand binding for the mitochondrial benzodiazepine receptors in the liver

Protoporphyrin IX (PP) and N-methylprotoporphyrin IX ( N-MePP) added in vitro to liver membranes reduced dose-dependently the affinity of [ 3H]PK 11195 for the mitocnondrial benzodiazepine receptors (MBRs), the latter being about 20 times more potent ( K i 4.5 and 0.25 muM). Preincubation of these two porphyrins with liver homogenates for 120 min at 4° resulted in significant inhibition of [ 3H]-PK 11195 binding even after repeated washings of the membranes due to the residual presence in the membranes of about 35 and 5% of PP and N-MePP, respectively. Thus, the hypothesis that an in vivo increase in the hepatic porphyrin content modifies the binding of the isoquinoline PK 11195 to the MBRs was investigated in an experimental model of protoporphyria. PP and N-MePP were allowed to accumulate in vivo through treatment with 3,5-diethoxycarbonyl-1,4-dihydrocollidine (DDC) (100 mg/kg i.p., once), and rats were killed 5 h after treatment when hepatic porphyrin accumulation was marked (10-fold increase), PP predominating. In the liver, treatment reduced the affinity ( K d ) of [ 3H]PK 11195 for MBRs (from 3.56 to 15.37 nM, P < 0.01) and the maximum number of binding sites ( B max) (55% decrease, P < 0.05); the affinity ( K i) of RO 5-4864 for [ 3H]PK 11195 binding sites was also reduced (from 23.9 to 72.99 nM, P < 0.05). No significant differences were found in the brain cortex. Liver and brain diazepam binding inhibitor levels and plasma corticosterone levels were unchanged. The reduction in [ 3H]PK 11195 binding to MBRs in the liver of DDC-treated rats thus appears to be attributable to a specific effect of the DDC-induced formation of the two protoporphyrins; this conclusion suggests that in hepatic protoporphyria processes modulated by MBRs may be altered.

Cyclophosphamide and its metabolite acrolein. Some studies on their porphyrinogenic action in 17 day old chick embryo

1. Some studies of cyclophosphamide (CP) and its metabolite acrolein in chick embryo liver were carried out in order to investigate the mechanism of the porphyrinogenic action of CP. 2. in vitro and in vivo studies revealed that CP induced but did not activate (5-aminolaevulinic acid (ALA) synthase. 3. Pretreatments with phenobarbital (PB) or SKF-525A did not modify ALA-synthase induction produced by CP. 4. Acrolein administration neither induced ALA-synthase activity nor increased cytochrome P-450 content or led to hepatic porphyrin accumulation. 5. Time course induction of cytochrome P-450 content after administration of CP or PB was similar. 6. The results obtained would indicate that CP is a strong inducer of both ALA-synthase activity and microsomal cytochrome P-450 content in liver of 17 day-old chick embryos and that its porphyrinogenic activity is not mediated by its metabolite acrolein.

Protective Effect of S-Adenosyl-L-Methionine in Hepatic Uroporphyria: Evaluation in an Experimental Model

The potential use of S-adenosyl-L-methionine (SAMe) as therapy for human porphyria cutanea tarda was investigated in an experimental model of hepatic porphyria--that is, chronic treatment of female rats with 0.2% hexachlorobenzene (HCB) in the diet. Administration of SAMe (25 mg/kg subcutaneously twice daily) during the last 15 days of HCB administration halved porphyrin accumulation in the liver but did not alter HCB-induced massive inhibition of uroporphyrinogen decarboxylase. Equally unaffected were inhibition of glutathione peroxidase and stimulation of lipid peroxide formation induced by HCB. Hypothetically, the beneficial effect of SAMe on hepatic porphyrin accumulation might be linked to modifications of the cellular availability of adenosine triphosphate.

2,3,7,8-Tetrachlorodibenzo- p-dioxin-induced porphyria in genetically inbred mice: Partial antagonism and mechanistic studies

2,3,7,8-Tetrachlorodibenzo- p-dioxin (TCDD) (233 nmol/kg) causes a significant increase of hepatic uroporphyrin, heptacarboxyporphyrin, and total porphyrins in female C57BL/6 mice, ovariectomized C57BL/6 mice, male C57BL/10 mice, and male C57BL/6 mice 3 weeks after treatment. In contrast, 6-methyl-1,3,8-trichlorodibenzofuran (MCDF) was inactive at a dose of 750 μmol/kg. Cotreatment of the mice with TCDD (233 mol/kg) plus MCDF (750 μmol/kg) resulted in partial antagonism of TCDD-induced hepatic porphyrin accumulation only in the female mice. Parallel studies in female C57BL/6 mice showed that the TCDD-induced porphyria was accompanied by the induction of hepatic microsomal aryl hydrocarbon hydroxylase (AHH) and ethoxyresorufin O-deethylase (EROD) activities and the depression of uroporphyrinogen decarboxylase (UROD). MCDF (750 μmol/kg) did not significantly affect these enzymes. In the cotreatment studies (MCDF plus TCDD), MCDF partially antagonized TCDD-induced hepatic porphyrin accumulation but did not affect the levels of hepatic AHH, EROD, or UROD. These results indicate that other factors, in addition to the induction of cytochrome P450-dependent monooxygenases and depressed UROD activity, are important in TCDD-induced porphyria in C57BL/6 female mice.

Investigations on the role of free radical processes in hexachlorobenzene-induced porphyria in mice.

Male C57Bl/10 mice were chronically fed hexachlorobenzene (HCB) (0.02% of the diet) alone or in combination with a single subcutaneous dose of iron (12.5 mg iron per mouse). After eight weeks the group of mice pretreated with the iron overload was highly sensitized to the porphyrogenic effect of HCB, as shown by liver porphyrin accumulation. A synergistic effect of iron was evident on other parameters too, such as HCB-induced hepatic damage, activation of type O of xanthine oxidase, and decreased activity of copper zinc superoxide dismutase and glutathione peroxidase(s). None of these parameters was affected by iron alone. Iron alone and in association with HCB markedly raised the level of lipid peroxides, the increase in the HCB group being smaller. The combined treatment resulted in a significant reduction of HCB's inductive effects on microsomal heme and cytochromes P-450 and b5 and on the activity of aryl hydrocarbon hydroxylase. The content of nonprotein sulfhydryl groups was reduced to the same extent in mice treated with HCB or HCB plus iron. The results suggest that reactive intermediates such as are formed by lipid peroxidation are not sufficient on their own to create the conditions for uroporphyrinogen decarboxylase impairment, as evident in the group of mice receiving iron overload alone. Conversely, HCB administration induced a specific condition of imbalance in the liver between formation and inactivation of reactive intermediates which was associated with hepatic porphyrin accumulation and was potentiated by concomitant administration of iron.

Effect of some antineoplasics on metabolic heme pathway

1. 1. The porphyrinogenic ability of several antineoplasics used in the therapy of the different cancers was evaluated. The action of cyclophosphamide, busulfan and 5-fluorouracil on the amount and nature of the accumulated hepatic porphyrins and on the activity of δ-aminolaevulinate synthase(ALA-S). were estimated at different doses and times of drug treatment in 17-day-old chick embryos. 2. 2. It was observed that cyclophosphamide produces a significant increase in the accumulation of hepatic porphyrins at different doses as well as in the activity of the ALA-S, at all the incubation times. Cyclophosphamide alters the pattern of porphyrins accumulated in the liver, where a coproporphyrin: protoporphyrin ratio higher than in the controls can be observed. 3. 3. Busulfan increased the hepatic porphyrins accumulated in the liver but to a lesser degree than cyclophosphamide. 4. 4. 5-Fluorouracil did not modify the hepatic porphyrin content when it was administered at doses up to 40 mg/embryo. 5. 5. When the embryos were injected with busulfan or 5-fluorouracil no significant differences were observed in the activity of ALA-S up to 11 hr of incubation. 6. 6. These results indicate that cyclophosphamide has a remarkable porphyrinogenic capacity in chick embryo while busulfan. notwithstancling the fact that it alters the haem pathway, it does so to a degree that does not impair the regulation of ALA-S activity. Fluorouracil seems to be non porphyrinogenic in this system, up to 40 mg/embryo.

Allylisopropylacetamide induces rat hepatic ornithine decarboxylase.

In rat liver, allylisopropylacetamide (AIA) treatment strongly induced (25-fold) the activity of rat hepatic ornithine decarboxylase (ODC). By either the oral or the subcutaneous route, AIA produced a long-lasting induction (30 to 40 hours) of hepatic ODC activity. Three analogs of AIA, propylisopropylacetamide (PIA), allobarbital, and allylbenzene, were active ODC inducers while a fourth, allylacetate, was not. Although induction of hepatic aminolevulinic acid (ALA) synthetase activity and the accumulation of hepatic porphyrins depend on the allyl moiety of AIA, this is not the case with hepatic ODC induction. Allylisopropylacetamide did not elevate serum alanine aminotransferase (SGPT) nor did it cause DNA damage, as measured by the alkaline elution assay. Thus, hepatic cell death is not a likely explanation of AIA's long-lasting induction of ODC. As AIA does not belong to any of the common categories of ODC inducers, it may be the chemical prototype of a new class of hepatic ODC inducers.

Effect of parathion, malathion, endosulfan and chlordane on porphyrin accumulation and alasynthetase in chick embryo liver

In the present study we investigated the porphyrinogenic ability of 4 pesticides: chlordane, endosulfan, parathion and malathion, all of which are widely used in agriculture. In order to determine whether they affect the heme biosynthetic pathway we studied, in 17-day old chick embryo liver “in ovo”, their effects on the amount of porphyrins and on the activity of δ-ALA synthetase (ALA-S), the first and rate limiting enzyme of this pathway. All of them induced hepatic porphyrin accumulation to a different extent compared with dimethylsulfoxide (DMSO) controls. Parathion as well as endosulfan promoted remarkable increases, chlordane raised porphyrin level in a lower degree and malathion slightly modified it. However, the accumulation observed with malathion was markedly enhanced if the period of incubation was extended to 48 h and, even more, if a second dose was injected during these 48 h. When ALA-S activity was analyzed in the chick embryos treated with parathion or endosulfan no alteration could be found in spite of producing noticeable accumulation of porphyrins. In contrast, chlordane promoted a statistically significant elevation of ALA-S as well as malathion which produced the highest induction observed. These results show that not only organochlorinated but also organophosphorous pesticides affect heme metabolism and that induction of ALA-S and porphyrin accumulation are not parallel.

Porphyrinogen carboxy-lyase from chick embryo liver— in vivo effect of heptachlor and lindane

1. 1. Heptachlor and lindane are widespread chlorinated insecticides. But porphyrinogen carboxy-lyase seems to be a key enzyme in the porphyria produced by chlorinated compounds in mammals. It was then considered interesting to perform the present investigation, the purpose of which was to study: (a) the porphyrinogenic ability of heptachlor and lindane when they were administered in vivo to whole chick embryo and (b) the in vivo effect of heptachlor and lindane on porphyrinogen carboxy-lyase activity. 2. 2. The results obtained showed that both compounds produce a significant accumulation of hepatic porphyrins, that produced by lindane being greater than that produced by heptachlor. 3. 3. The effect of heptachlor and lindane administration on porphyrinogen carboxy-lyase was studied in embryos from New Hampshire and Leghorn strains and in New Hampshire 1-day old chicks. The influence of phenobarbital and SKF-525 A pretreatment on the response of porphyrinogen carboxy-lyase to these drugs was also studied in New Hampshire chick embryos. The results indicate that the drugs do not produce decreased enzyme activity in the New Hampshire embryos, while a decrease of enzyme activity in the White Leghorn embryos and in the New Hampshire new-born chicks was observed. Results from pretreatment assays suggest that the decrease of porphyrinogen carboxy-lyase activity seems to be associated with the formation of a drug metabolite. 4. 4. It is concluded that heptachlor and lindane are porphyrinogenic drugs and that the response of decarboxylating enzyme to them seems to be dependent on: (a) the strain, (b) the development of the animal and (c) the drug metabolization system; some of the lindane and heptachlor metabolites being responsible for hepatic porphyrinogen carboxy-lyase decrease.

Different patterns of hepatic microsomal enzyme activity produced by administration of pure hexachlorobiphenyl isomers and hexachlorobenzene

Three hexachlorobiphenyl isomers, 2,2′,4,4′,5,5′-hexachlorobiphenyl (I), 2,2′,3,3′,4,4′-hexachlorobiphenyl (II) and 2,2′,3,4,4′,5′-hexachlorobiphenyl (III), have been administered to rats and the effects of these three compounds upon hepatic microsomal drug metabolism and upon hepatic porphyrins have been studied. Comparisons have been made with hexachlorobenzene and a commercial polychlorinated biphenyl mixture, Aroclor 1254. From measurements of activities of microsomal drug oxidations in vitro, the durations of pharmacological actions of certain drugs in vivo and spectral shifts associated with cytochrome P-450 it is shown that the three pure hexachlorobiphenyl isomers initially produce changes in hepatic microsomal activity which resemble those seen after treatment with phenobarbitone (PB). In contrast, following chronic feeding of the isomers, compounds II and III but not I produce a pattern of hepatic microsomal enzyme activity which shows some characteristics of the 3-methylcholanthrene (3-MC) and some characteristics of the phenobarbitone classes of inducer. Also, compounds II and III, but not I, cause accumulation in the liver of porphyrins containing either seven or eight carboxyl groups. These two responses are similar to those observed following hexachlorobenzene treatment and suggest that a relationship may exist between the mixed pattern of enzyme induction and the onset of hepatic porphyrin accumulation.

Toxicological assessment of hexachlorobiphenyl isomers and 2,3,7,8-tetrachlorodibenzofuran in chicks: II. Effects on drug metabolism and porphyrin accumulation

Pure hexachlorobiphenyl (HCB) isomers induce a number of changes in parameters of drug metabolism in the chick including changes in cytochrome P-450, liver weight, and p-nitrophenol glucuronyl transferase, but not in testosterone glucuronyl transferase activity. The most active inducers of drug metabolism were 2,3,4,2′,3′,4′-HCB and 2,4,6,2′,4′,6′-HCB, while 2,4,5,2′,4′,5′-HCB produced intermediate effects and 2,3,6,2′,3′,6′-HCB was a poor inducer. All HCBs caused uroporphyrin accumulation and increased δ-aminolevulinic acid (ALA) synthetase activity, but only 3,4,5,3′,4′,5′-HCB, 2,3,4,2′,3′,4′-HCB, and 2,4,5,2′,4′,5′-HCB produced gross accumulation of hepatic porphyrins. Tissue HCB concentrations correlated well with hepatic effects. 2,3,7,8-Tetrachlorodibenzofuran (TCDF), a contaminant of commercial polychlorinated biphenyl (PCB) mixtures, had no effects on hepatic ALA synthetase activity, porphyrin accumulation, or glucuronyl transferase. TCDF did produce a slight increase in cytochrome P-450, but the increase was smaller than that produced by HCBs. Also, the cytochrome induced by TCDF appeared qualitatively different from the cytochrome induced by TCBs, differing by a shift in the peak of the CO-difference spectrum from 450 to 449 nm and by the ratio of absorbance at 455 to 430 nm of the ethyl isocyanide difference spectrum.
